目的 剖析闵行区公立医疗机构政府补偿机制的要素与特点,为深化公立医疗机构改革提供决策参考。方法 收集和梳理政策文件,分析改革前后,政府补偿范围、补偿方式和补偿水平的变化,总结补偿机制特点。结果 改革后医疗服务和公共卫生服务经费按绩效考核结果进行补偿,基本建设和设备购置经费、政府指令性任务、学科发展、人员培养和科研教育经费的补偿标准和办法得到了明确。结论 闵行区公立医疗机构政府补偿机制以公益性为目标,以绩效考核为依据,运用了信息化工具,考虑了机构类型的差异,有利于引导医疗机构持续改进绩效,实现公益性目标。  相似文献

目的 验证公立医院绩效考核“PATH优化模型”指标体系的可行性。方法 按照指标要求进行数据采集与测算,根据实证医院实际情况确定指标值,进行模型模拟,测算结果,论证模型的可操作性。 结果 公立医院绩效考核“PATH优化模型”在稳定的评价维度下,指标设计具有一定的灵活性,医院可根据阶段性目标的达成情况对指标进行动态调整;在规范化管理基础较好、绩效管理信息系统建设完善的医院,指标数据的收集可实现准确、便捷。结论 公立医院绩效考核“PATH优化模型”具有较好的可行性。  相似文献

Accelerated terrestrial ecosystem carbon turnover and its drivers   总被引:1,自引:0,他引:1  
The terrestrial carbon cycle has been strongly influenced by human‐induced CO2 increase, climate change, and land use change since the industrial revolution. These changes alter the carbon balance of ecosystems through changes in vegetation productivity and ecosystem carbon turnover time (τeco). Even though numerous studies have drawn an increasingly clear picture of global vegetation productivity changes, global changes in τeco are still unknown. In this study, we analyzed the changes of τeco between the 1860s and the 2000s and their drivers, based on theory of dynamic carbon cycle in non‐steady state and process‐based ecosystem model. Results indicate that τeco has been reduced (i.e., carbon turnover has accelerated) by 13.5% from the 1860s (74 years) to the 2000s (64 years), with reductions of 1 year of carbon residence times in vegetation (rveg) and of 9 years in soil (rsoil). Additionally, the acceleration of τeco was examined at biome scale and grid scale. Among different driving processes, land use change and climate change were found to be the major drivers of turnover acceleration. These findings imply that carbon fixed by plant photosynthesis is being lost from ecosystems to the atmosphere more quickly over time, with important implications for the climate‐carbon cycle feedbacks.  相似文献

The Biotechnology Certificate Program (BCP) at California State University, Hayward was initiated in 1986 in response to industry demands for qualified employees in the molecular life sciences. This 9-month post-baccalaureate program includes laboratory courses in recombinant DNA techniques, protein chemistry, PCR, DNA sequencing, animal cell culture as well as two lecture courses in molecular biology. Rigorous selection at both entry and exit stages of the program ensures knowledgeable graduates with a greater than 90% employment placement. Corporate participation has been a cornerstone of the BCP and we anticipate continued cooperation in the future as the program evolves to meet the expanding needs of the biotechnology industry. 目的 分析河北省二甲、三甲医院卫生技术人员参加继续医学教育项目学习的状况和经费负担特点。方法 采用分层抽样方法对45所二甲、三甲医院6 013名卫生技术人员问卷调查和描述性研究方法分析。结果 三甲医院人员各类项目参加率均高于二甲医院。两级医院人员参加率高的项目有收益率低,参加率与有收益率背离。教育费用负担有单位全部负担、单位和个人共同负担和个人全部负担3种形式。二甲医院3种负担的课程次数构成比分别为22.23%、16.03%、61.74%,单位全部负担和个人全部负担是主要的负担形式;三甲医院为19.50%、29.56%、50.94%,单位和个人共同负担是主要的负担形式。结论 投入不足和制度落实不到位是二甲医院滞后的主要原因,单位和个人共同负担是较为合理的负担形式。  相似文献

Recent outbreaks of cryptosporidiosis from contaminated water supplies have led to a need for the detection of Cryptosporidium oocysts from various hosts and contaminating sources. The presence of nonpathogenic species or strains of Cryptosporidium is important for diagnostic purposes as there is a potential for false-positive detection of pathogenic parasites. The present review focuses on phenotypic differences and recent advances in genotypic analyses of the genus Cryptosporidium with an emphasis on detecting various isolates and identifying differences in Cryptosporidium parvum and other species in this genus. The information currently available demonstrates important patterns in DNA sequences of Cryptosporidium, and our understanding of macro- and microevolutionary patterns has increased in recent years. However, current knowledge of Cryptosporidium genetic diversity is far from complete, and the large amount of both phenotypic and genotypic data has led to problems in our understanding of the systematics of this genus. The increasing number of microbreweries and brewpubs and the popularity of homebrewing present a heady opportunity for undergraduate projects that blend together theory, practice and job opportunities in the realm of fermentation science. However, a perception of prohibitive regulation on campus and other concerns has led many instructors to abandon that most practical demonstration of fermentation, the production of ethanol by Saccharomyces cerevisiae. We present our success in introducing fermentation using homebrewing in a microbiology laboratory. Virtually all fundamental topics of microbiology are remarkably easy to demonstrate with this project: growth and replication, physiology, limitation of growth by sterilization and other methods, competition, energy conservation and utilization, consequences and uses of mutations, and genetic engineering are among the topics which dovetail with this activity. Further, this activity also represents a natural introduction to a number of industrial topics: issues of scaling-up, pilot tests and environmental conditions. Abstract

Algorithms of secondary structure prediction have undergone the developments of nearly 30 years. However, the problem of how to appropriately evaluate and compare algorithms has not yet completely solved. A graphic method to evaluate algorithms of secondary structure prediction has been proposed here. Traditionally, the performance of an algorithm is evaluated by a number, i.e., accuracy of various definitions. Instead of a number, we use a graph to completely evaluate an algorithm, in which the mapping points are distributed in a three-dimensional space. Each point represents the predictive result of the secondary structure of a protein. Because the distribution of mapping points in the 3D space generally contains more information than a number or a set of numbers, it is expected that algorithms may be evaluated and compared by the proposed graphic method more objectively. Based on the point distribution, six evaluation parameters are proposed, which describe the overall performance of the algorithm evaluated. Furthermore, the graphic method is simple and intuitive. As an example of application, two advanced algorithms, i.e., the PHD and NNpredict methods, are evaluated and compared. It is shown that there is still much room for further improvement for both algorithms. It is pointed out that the accuracy for predicting either the α-helix or β-strand in proteins with higher α-helix or β-strand content, respectively, should be greatly improved for both algorithms.  相似文献

A concentric cylinder reactor (CCR) is described that enables the steady-state kinetics of microbial biofilms to be evaluated under conditions of constant nutrient flow and variable shear-stress. The reactor has been used to evaluate the influence of fluid dynamic shear on the extent and mode of detachment of bacteria from biofilms. Using a food factory isolate of Pseudomonas aeruginosa, a general increase in the overall growth rate and detachment of the biofilms (cfu cm−2 min−1) with time was shown for each biofilm, regardless of the prevailing shear. As the shear rate was increased beyond 0.123 ms−1, populations tended toward a pseudo steady-state. Sudden changes in shear force, however, caused dramatic changes in the productivity of steady-state populations. The CCR provides an effective means of testing disinfectant activity, particularly for clean-in-place situations, and allows for an examination of the residual effects of a cleansing programme on a treated surface for three different chemical classes of disinfectant. Utilisation of the CCR would, therefore, provide enhanced ability to determine the efficacy and efficiency of chemical products for use in sanitation protocols. Background  

Pandemic Vibrio parahaemolyticus has undergone rapid changes in both K- and O-antigens, making detection of outbreaks more difficult. In order to understand these rapid changes, the genetic regions encoding these antigens must be examined. In Vibrio cholerae and Vibrio vulnificus, both O-antigen and capsular polysaccharides are encoded in a single region on the large chromosome; a similar arrangement in pandemic V. parahaemolyticus would help explain the rapid serotype changes. However, previous reports on "capsule" genes are controversial. Therefore, we set out to clarify and characterize these regions in pandemic V. parahaemolyticus O3:K6 by gene deletion using a chitin based transformation strategy.  相似文献

It is now possible to obtain sequence information from gel-separated proteins by mass spectrometry at levels too low for conventional approaches. Usually this tandem mass spectrometric data are used for database searches with the aim of identifying the corresponding gene. Recently it has been shown that long and accurate amino acid sequences can be obtained which are sufficient for PCR-based strategies to clone the corresponding gene [Wilm et al. (1996), Nature 379, 466–469]. More than eight proteins have now been cloned based on that method. In many more cases the sequence information identified homologous proteins. Issues involved in cloning by mass spectrometric sequence information are discussed, as are two case studies. These results clearly establish mass spectrometry as a viable tool not only for the database identification of proteins, but also for the de novo sequencing of gel-separated proteins at the low-picomole to femtomole level.  相似文献

ABSTRACT

Developing students’ systems thinking is an often-posed demand in education for sustainable development (ESD) and science literacy. Several studies have shown that systems thinking can be fostered in students of different education levels. Therefore, science teachers who are required to teach ESD-relevant topics should be proficient in systems thinking and be able to transfer that knowledge effectively to their students. The research project SysThema (Systems Thinking in Ecological and Multidimensional Areas) investigated the effect of three courses designed to foster systems thinking in student teachers of biology and geography. Courses varied in their proportions of technical fundamentals of system science and didactical content for teaching systems thinking. To conceptualise systems thinking, a heuristic structural competence model for systems thinking was developed. This model served as the basis for a test in evaluating the courses in a quasi-experimental intervention study that employed a pre-, post- and follow-up test control group design. After the completion of the courses, a high effect of fostering systems thinking was found in all treatment groups compared to the control group.  相似文献

Background: Some 3% of the area of Tyrol is covered by glaciers. Since the end of the Little Ice Age, these glaciers have shrunk by about 50%, providing open ground for succession of biota. As the exposed bare ground becomes available for colonisation, pedogenesis, and primary succession, glaciological data provide important information for the analysis of chronosequences of biological succession.

Aims: This paper summarises long-term surveillance studies and provides information on annual glacier changes with a focus on the three Tyrolean LTER sites Hintereisferner, Kesselwandferner and Jamtalferner, together with two additional glaciers.

Methods: Annual changes in the length of 54 glaciers were recorded, with the first measurements made in 1891. In addition, the long-term mass balance data of five glaciers are presented. Two of these glaciers have been surveyed since 1953.

Results: Between 1901 and 2008, the temperature during the ablation season (May to September) increased by 1.7 °C (measured at one site in Vent). In the same period, winter (October to April) accumulation showed no significant trends. Measurements of length changes showed a general glacier retreat, with limited advances in the 1920s and 1980s. The mass balance measurements indicated mass losses since the 1980s at increasing rates, especially after 2000. The reaction of glaciers to the climate signal differed in timing and magnitude according to their topographic properties.

Conclusions: Although the Tyrolean glaciers have generally retreated since the end of the Little Ice Age, the magnitude and pace of advance or recession differs from glacier to glacier. This is important to consider in studies on succession of biological communities in glacier forelands  相似文献

ABSTRACT

Proteome—the protein complement of a genome—has become the protein renaissance and a key research tool in the post-genomic era. The basic technology involves the routine usage of gel electrophoresis and spectrometry procedures for deciphering the primary protein sequence/structure as well as knowing certain unique post-translational modifications that a particular protein has undergone to perform a specific function in the cell. However, the recent advancements in protein analysis have ushered this science to provide deeper, bigger and more valuable perspectives regarding performance of subtle protein-protein interactions. Applications of this branch of molecular biology are as vast as the subject is and include clinical diagnostics, pharmaceutical and biotechnological industries. The 21st century hails the use of products, procedures and advancements of this science as finer touches required for the grooming of fast-paced technology.  相似文献

The dragonfly Hemicordulia tau has five spectral classes of photoreceptor which drive five lamina monopolars, m1-m5. The monopolars encode spectral information. Here, spectral coding by m2, m4 and m5 are described. m2 is the most sensitive to dim light. m4 and m5 are less sensitive than m2, and so we call them photopic cells. The effects of selective adaptation of receptor inputs to m4 and m5 are unusual. For example, in m4 adaptation of the green receptor suppresses the UV input, but green sensitivity is unaffected, while green adaptation of m5 increases its green sensitivity. In m5 the dark adapted spectral tuning resembles the 520 nm receptor, but on adaptation to 430 nm light this narrows markedly. Adaptation either of green or of UV receptor input to m2 and m4 modifies the time course of responses both to green and to UV, implying that changes in the time courses of monopolar responses with adaptation state do not directly reflect receptor responses (Fig. 2). The class consisted of senior molecular biology majors who had previously taken cell biology, microbiology, biochemistry and molecular biology lecture courses but who had little or no previous lab experience. These students were asked to design and create an expression vector and purify the expressed protein. Abstract

The public concern about electromagnetic field (EMF) exposure varies due to different reasons. A part of them are connected with the better and higher quality of information that people receive from science, media, Internet, social networks, industry, but others are based on good communication programs performed by the responsible institutions, administration and persons. Especially, in Bulgaria, public concern follows interesting changes, some of them in correlation with the European processes of concern, but others following the economic and political processes in the country. Here, we analyze the dynamics of the public concern over the last 10 years. Our explanation of the decrease of the people’s complaints against EMF exposure from base stations for mobile communication is as a result of our risk communication program that is in implementation for >10 years.  相似文献

ABSTRACT

Capsule: The distribution range of the European Roller Coracias garrulus has undergone large changes over geological times, but although the species is warm-adapted, the human induced climate change is predicted to affect negatively the range of the currently large populations.

Aim: Information on species-specific vulnerability to climate change is crucial not only for designing interventions and setting conservation goals, but also to inform conservation decision-making. Our goal was to map climate suitability for the European Roller in the Western Palaearctic under current climate, and for past (last glacial maximum and mid-Holocene) and future (2050 and 2070) climate scenarios.

Methods: We used MaxEnt for species distribution modelling based on the reconstructed distribution map of the species.

Results: Our results suggest that during glacial periods Rollers persisted in small southern refugia, and then spread and colonized northern latitudes during the mid-Holocene. In the future, our models forecast a shift in climatically suitable range towards northern latitudes and an overall small range contraction (4.5–5.5%). Warmer temperatures will increase climate suitability in northern countries where the species is currently declining or became locally extinct. On the other hand, wide suitable areas under current climatic conditions are predicted to become unsuitable in the future (35–38% by 2050 and 2070, respectively), significantly impacting large populations such as those in Romania, Spain, Bulgaria and Hungary. French and Italian populations are identified to be future key populations for Roller conservation.

Conclusions: Our findings suggest that future climate changes will likely amplify the impacts of existing threats on the majority of large European Roller populations in Europe.  相似文献

This collaborative laboratory exercise integrates two upper division laboratory courses (Developmental Biology and Neurobiology) offered to biology majors at Wake Forest University. The laboratory exercise involves the use of the Xenopus oocyte expression system to study the function of specific membrane receptors and ligand-activated channels. cDNA or mRNA for receptor proteins is injected into Xenopus oocytes. The oocytes are assayed for expression of receptor proteins and two-electrode voltage clamping is done to determine whether the expressed proteins are functional in the oocyte system. This series of laboratory exercises is innovative in its interdisciplinary and collaborative approach to undergraduate teaching, and in its use of sophisticated molecular biological and physiological techniques in the undergraduate teaching laboratory. Students learn first-hand how these techniques have been used to achieve a new level of understanding of both development and neurobiology. Creatine kinase (ATP: creatine N-phosphotransferase, EC 2.7.3.2) was completely denatured using 3 M guanidine hydrochloride for 2 h as in previous studies [Yao et al. (1982), Sci. Sin. 25B, 1296–1302; Yao et al. (1984), Biochemistry 23, 2740–2744; Yao et al. (1982), Sci. Sin. 25B, 1186–1193]. Under suitable conditions, about 60–70% of the activity can be recovered in the presence of different Mg2+ concentrations. Both the reactivation and the refolding processes follow two-phase courses after dilution in the proper solutions. A comparison of the rate constants for the refolding of unfolded creatine kinase with those for the recovery of its catalytic activity at various Mg2+ concentrations shows that these are not synchronized. The reactivity of guanidine hydrochloride-denatured creatine kinase can be inhibited by Mg2+; however, the rates of reactivation are independent of the Mg2+ concentration. In addition, Mg2+ affects the fluorescence intensity, but the rate constants of refolding are independent of Mg2+ concentration. Although the reactivation of GdHCl-denatured creatine kinase is complete about 3 h after dilution with reactivation solutions, the conformational changes during refolding occur in a much slower reaction. Mg2+ can induce complex changes in the relative fluorescence intensity during refolding over a broad range of concentrations.  相似文献
