Cytokinins from leaf laminae, buds, petioles, stems, roots,and root exudate of mature vegetative plants of Xanthium strumariumL. were extracted, fractionated, and partially characterizedby means of column chromatography with Sephadex LH20. Two peaksof cytokinin activity with elution volumes corresponding tozeatin and zeatin riboside were detected, in varying concentrations,in all plant parts. A third cytokinin, detected only in petiolesand in expanding and mature leaves, eluted off the Sephadexcolumn before zeatin riboside. This cytokinin (peak ‘a’)was converted to zeatin or to a zeatin-like cytokinin followingboth acid hydrolysis and treatment with ß-glucosidase.Peak ‘a’ was not detected in buds or in the youngestdeveloping leaves but was the predominant cytokinin presentin half-expanded and more mature leaves. By contrast, the zeatinriboside-like peak (peak ‘b’) constituted the majorcytokinin in root exudate, apical buds, and the youngest developingleaves, while not greatly contributing to the cytokinin contentof mature leaves. The detopped root system was shown to be capable of cytokininproduction. The distribution of cyrtokinins in the plant isdiscussed in relation to their probable origin in the root system.  相似文献

The effects of the foliar application of phytocidal concentrationsof 2-methyl-4-chlorophenoxyacetic acid (MCPA) on change in totaldry weight, and in ‘available carbohydrate’ (starch,‘total’ and ‘reducing’ sugars), totalnitrogen, phosphorus, potassium, calcium, and magnesium of ‘tops’and roots of tomato plants have been followed over a periodof 14 days following spraying. There were two main treatments—‘nutrient’(nutrient supply to roots continued after spraying) and ‘water’(distilled water only supplied to roots after spraying) and‘water’ (distilled water only supplied to rootsafter spraying)—the sub-treatments consisting of ‘MCPA’versus ‘no-MCPA’ for each of the main treatments.Twelve different times of sampling were used. In analysing the present data, the quantity ‘residualdry weight’ (total dry weight less ‘available carbohydrate’),which was originally introduced by Mason and Maskell as a basisof reference for analyses of plant organs in short-period experimentsnot involving appreciable growth, has been used as an estimateof the permanent structure of plant growth. This new use ofthe ‘residual dry weight’ basis has brought outimportant features which were obscured when the data were leftin their primary form (as percentages of total dry weight oramounts per plant). Growth, as measured by increase in ‘residual dry weight’,was greatly inhibited by 2-methyl-4-chlorophenoxyacetic acidshortly after spraying, in both the presence and the absenceof nutrient. In the presence of 2-methyl-4-chlorophenoxyacetic acid, netassimilation rate (estimated as rate of increase in total dryweight per gram ‘residual dry weight’ of the ‘tops’)was greatly diminished while uptake of total nitrogen and ofP2O5 (estimated as increase in total nitrogen or of P2O5 ofthe whole plant per day per 1 g. ‘residual dry weight’of the roots) appeared to undergo a similar but much smallerdiminution. It seemed probable, however, that in the presenceof MCPA a larger proportion of the carbohydrate actually formedwas utilized for synthesis of aminoacids and protein. In the plant as a whole there was no evidence of actual depletionof ‘available carbohydrate’ as a result of MCPAtreatment, this fraction showing a steady increase in all treatmentsthroughout the experiment. The rate of increase was, however,much reduced by MCPA treatment. The ‘tops’ presentedmuch the same picture as the whole plant, but for the rootsthe situation was quite different. While the roots of the ‘no-MCPA’plants and also of the ‘MCPA-water’ plants showeda steady increase in available carbohydrate, those of the ‘MCPA-nutrient’plants rose only very slightly (from the initial value of 8mg. per plant to about 10 mg.) during the first 2 days, andthen in the next 2 days declined to a value (about 6 mg.) belowthe initial and remained at this low level for the rest of theexperiment. It is suggested that the phytocidal effect of 2-methyl-4-chlorophenoxyaceticacid in the presence of nutrient may be due to depletion ofthe ‘available carbohydrate’ supplies in the roots,which is shown to be brought about, in part, by reduced transportfrom the tops, and partly by the relatively greater utilizationof the carbohydrate present. These results offer an explanationfor the facts that plants showing vigorous growth are more easilykilled by MCPA and that perennial plants, particularly thosewith storage tissues in their roots, are more resistant. Further,they suggest the useful practical application that MCPA treatmentshould be given when the carbohydrate reserves of the rootsare at a minimum. For perennial plants, conditions might beexpected to be optimal for the application of MCPA in late spring,at a time when the first ‘flush’ of growth is slowingdown and before any appreciable new reserves of carbohydratehave been accumulated. It was also shown that 2-methyl-4-chlorophenoxyacetic acid preventedthe net synthesis of starch, but still permitted an appreciablenet formation of sucrose. 2-methyl-4-chlorophenoxyacetic acid appeared to have no effecton the uptake of potassium, calcium, or of magnesium. The lackof effect on potassium is contrasted with the previous observationby Rhodes, Templeman, and Thruston (1950) that sub-lethal concentrationsof MCPA, applied over a relatively long period to the rootsof tomato plants, specifically depressed the uptake of potassium.  相似文献

Experiments are described which indicate that the annual vernalizationrequirement of the basal shoots of the Chrysanthemum is dueto annual devernalization of these shoots as the main axis growsup and flowers. Plants sprayed with varying concentrations ofmaleic hydrazide were arrested in their growth for considerableperiods, but this enforced ‘dormancy’ did not affecttheir vernalization status. This makes it appear unlikely thatmere suppression of growth through apical dominance of the mainshoot is the cause of this de vernalization of basal shoots.Fully or partly vernalized plants heated to 40° C. for upto 30 hours did not become dc-vernalized. Heat treatment at35° C. for as long as 30 days also failed to achieve completedc-vernalization, but here flowering was delayed by periodsequivalent to the time spent at high temperature. However, atthe end of the heat treatment progress towards flowering wasresumed at the normal rate. Complete dc-vernalization can bebrought about by prolonged exposure to low intensity illumination.This treatment appears to be effective right up to the stagewhen the first morphological changes leading to inflorescenceformation take place. These results are discussed in relationto similar experiments on the de-vernalization of rye and Hyoscyamusniger.  相似文献

The activity of phenylalanine ammonia-lyase in the seedlingsof Pharbitis nil strains ‘Violet’ and ‘Kidachi’during the non-photoperiodic induction of flowering increasedin close association with the accumulation of phenylpropanoidsand the induction of flower buds. This observation suggestedthat the increased activity of phenyalanine ammonia-lyase causedthe accumulation of the phenylpropanoids and that the activationof the biosynthesis of phenylpropanoids might be involved inthe non-photoperiodic induction of flowering in the seedlingsof ‘Violet’ and ‘Kidachi’. This review compares new developmental models on flowering andother vascular plants with evolutionary hypotheses formulatedby Agnes Arber (1879–1960) and like-minded botanists.Special emphasis is laid on philosophical basics such as perspectivism,pluralism about evolutionary modelling, continuum way of thinking,and fuzzy logic. Arber's perspective is best labelled as F uzzyA rberian M orphology (FAM Approach). Its proponents (‘FAMmers’)treat structural categories (e.g. ‘roots’, ‘shoots’,‘stems’, ‘leaves’, ‘stipules’)in vascular plants as concepts with fuzzy borderlines allowingintermediates (including transitional forms, developmental mosaics).The FAM Approach complements Cla ssical Plant M orphology (ClaMApproach), which is the traditional approach in botany. ClaMproponents (‘ClaMmers’) postulate that the structuralcategories of vascular plants are regarded as concepts withclear-cut borderlines and without intermediates. However, duringthe evolution of vascular plants, the root-shoot distinctionand the stem-leaf distinction have become blurred several timesdue to developmental changes, resulting in organs with uniquecombinations of features. This happened, for example, in thebladderworts (Utricularia, Lentibulariaceae). When focusingon the ‘leaf’, the FAM Approach is identical toArber's ‘partial-shoot theory of the leaf’ and Sinha's‘leaf shoot continuum model’. A compound leaf canrepeat the developmental pathway of the whole shoot, at leastto some degree. For example, compound leaves of Chisocheton(Meliaceae)with indeterminate apical growth and three-dimensional branchingmay be seen as developmental mosaics sharing some growth processeswith whole shoots! We focus here on the FAM Approach becausethis perspective is especially promising for developmental geneticistsstudying flowering and other vascular plants. Investigations revealed that the anatomy of the primary radicularroot of yam bean (Pachyrhizus erosus L.) was typically dicotyledonousexcept that the xylem was not completely developed centripetally.Most of the roots had tetrarch xylem, although a few triarchand pentarch roots were also observed. In both tuberous andnon-tuberous roots, secondary thickening occurred by the formationof the meristematic vascular cambium which formed secondarytissues in a normal fashion. Subsequently, tuberization wasinitiated in the secondary xylem by the development of anomalous‘secondary’ cambia from parenchyma cells surroundingvessel elements. Anomalous ‘secondary’ cambia alsodeveloped from parenchyma cells not associated with vessels.Subsequently, anomalous ‘tertiary’ cambia differentiatedfrom tissues produced by the anomalous ‘secondary’cambia. Activities of these anomalous cambia resulted in theproduction of parenchyma storage cells and were chiefly responsiblefor the growth of the mature tuber. The partition of 14C labelled current assimilates to root insimulated swards of Lolium perenne cv. S24 was measured duringthe transition from vegetative growth in autumn to reproductivegrowth in spring under close to natural conditions of lightand temperature. Assimilate partitioning was also measured in‘established’ swards cut three times during thegrowing season and in vegetative ‘seedling’ swardsgrowing in autumn and in spring. All measurements were madewhen the swards had achieved more than 90 per cent light interception,and all swards were abundantly supplied with water and mineralnutrients. During autumn there was a gradual decrease in the proportionof assimilates partitioned to the roots in both the ‘established’and the ‘seedling’ swards. In the established swards,partition to roots was low over winter, increased during earlyspring, but decreased dramatically, later in the spring, whenstem elongation began. In contrast, in the unvernalized vegatativeseedling swards in spring, partition to roots remained high. The seasonal pattern of assimilate partitioning is consideredin relation to changes in the natural environment and the rateat which the crop fixed carbon in photosynthesis. A decreasein the proportion of assimilates partitioned to roots duringlate spring was significant in increasing the production ofshoot at that time but seasonal differences in partition contributedvery little to the marked differences in shoot growth betweenthe spring and autumn crop. Floral buds of the ‘False Horn’ plantain clonesMusa (AAB) ‘Harton Verde’, ‘Harton Negra’,and ‘Currare’ terminate in a large single floralstructure. The apices of these floral buds are here designatedas determinate since they have lost the ability to produce additionalfloral initials or buds. Terminal peduncle segments can be culturedin a modified Murashige and Skoog (1962) medium supplementedwith N6-benzyl-aminopurine (5 mg I–1). Under these conditions,this apparent inability to yield buds can be overcome as vegetativeshoot clusters form in the axils of the bracts. Rooted plantletsare obtainable by treating shoots with naphthaleneacetic acid(1 mg I–1) and activated charcoal (0.025%). The adventitiousorigin of the shoots has been established. The flowering requirements of six European varieties of Loliumperenne L. were studied in controlled environments. In experimentson primary induction, flowering was recorded after transferto long days (LD) in a greenhouse at 12–24°C. In experimentson secondary induction, primary induction was first accomplishedat 6°C/10 h daylength for 12 weeks. When evaluated by the50% heading criterion, the requirement for duration of primaryinduction at 6°C/8 h daylength was <3 weeks in Mediterranean,5–6 weeks in Central European and 7–8 weeks in Scandinavianvarieties. While ‘Veyo’ (Italy) flowered profuselyregardless of temperature or daylength during primary induction,critical temperatures for primary induction in SD and LD were15 and 11°C in ‘Baca’ (Czech Republic) and 11and 7°C in ‘Falster’ (Denmark). The criticalphotoperiod for secondary induction at 15°C ranged from12 h in ‘Veyo’ and 14 h in ‘Baca’ to16.5 h in ‘Falster’ and 17.5 in ‘Kleppe’(Norway). The critical number of LD cycles varied correspondingly.While the Central and North European varieties required fewerLD cycles for 50% heading at 18 than at 12°C, ‘Veyo’showed the opposite response. It is concluded that the requirementsfor both primary and secondary induction of Lolium perenne increasewith increasing latitude of origin of the germplasm. In oneexperiment, 39–87% of the inflorescences came from tillersthat were not visible on transfer from primary to secondaryinduction, thus it is also concluded that there is no juvenilestage in tillers of Lolium perenne. Despite the high probability that sensitive post-emergent seedlingswill be exposed to suboptimal temperatures which significantlyaffect subsequent development and yield, growers of long-season,indeterminate crops must rely upon empirically determined ‘earliestplanting dates’ Most ‘chilling-stress’ studieshave dealt, not with photosynthetic seedlings, but rather withthe effect of low, non-freezing temperatures (< 10 ?) upongerminating seeds and pre-emergent seedlings. A photosyntheticseedling growth system was used to monitor the effects of temperaturesfrom 10 to 35? upon the roots and aerial portions of chilling-sensitivecotton and relatively chilling-resistant sunflower oil-seedlings.Length measurements of cotton seedlings were too variable tobe reliable indicators of temperature treatment effects, butroot and shoot fresh weights could be used as non-destructivemeasurements of differences in root and shoot temperature responsesin both cotton and sunflower seedlings. The fresh weights wereindicative of the relative water status of seedling roots andshoots, including that of seedlings returned to 30? after exposureto other temperatures. Simulated swards of each of two selection lines of Lolium perennecv. S23 with ‘fast’ and ‘slow’ ratesof ‘mature tissue’ respiration were establishedin growth rooms at 20/15 °C day/night temperatures and studiedover four successive regrowth periods of 46, 30, 26 and 53 daysduration. The ‘slow’ line outyielded the ‘fast’,both in harvestable shoot (above a 5 cm cut) and in root andstubble. Its advantage increased over successive regrowth periodsto 23 per cent (total biomass). Gas analysis measurements onthe entire communities (including roots), during the final regrowthperiod, showed that the ‘slow’ line had a 22–34per cent lower rate of dark respiration per unit dry weight.This enabled it to maintain its greater mass of tissue for thesame cost in terms of CO2 efflux per unit ground area. Halfthe extra dry weight produced by the ‘slow’ line,relative to the ‘fast’, could be attributed to itsmore economic use of carbon. The rest could be traced to a 25per cent greater tiller number which enabled the ‘slow’line to expand leaf area faster (though not at a greater rateper tiller), intercept more light and fix more carbon, earlyin the regrowth period. Controlled environments were used to establish the base-linegrowth and to evaluate the photoperiodic response of two Tossajute (Corchorus olitorius) cultivars. Fresh and dry weightsof shoots and roots, leaf areas and stem lengths were much greaterat long day (LD) 12·5, 11·5 than LD 11·5,12·5 photoperiods for both cultivars. Root lengths weregreater at LD 12·5, 11·5 initially but were longestat LD 11·5, 12·5 at 35 days from emergence. Meanrelative growth rates (MRGR) were similar irrespective of photoperiodor cultivar but declined with plant age. The superior growthof cultivar ‘Yaya’ at LD 12·5, 11·5was due to more growth in the first 7 days, while, for cultivar‘Angbadu’ it was a combination of growth for 7 days,and higher MRGR up to 21 days. Horseradish roots reach a stage of ‘ripeness to regenerate’at the age of 1–2 months, when secondary thickening hasbegun, a phellogen has formed, and the primary cortex has beenshed; provided that they are then detached from the parent plant. 2. New primordia arise (as is already known) only in associationwith the scars of lateral roots; but they are produced, notas previously believed from a callus, but individually as organizedmeristems whose cells are derived only from the phellogen ofthe main root. 3. Background and Aims: Plant growth responses to the rare earth elements lanthanum(La) and cerium (Ce) have been reported, but little is knownabout the effects of these two elements on plant mineral nutrition. Methods: Corn (Zea mays ‘Hycorn 82’) and mungbean (Vignaradiata ‘Berken’) were grown in continuous flowingnutrient solutions containing 0, 0·2, 1·0 and5·0 µM La or Ce. At harvest plants were dividedinto roots and shoots, dried, weighed and analysed for macro-and micronutrients, as well as for La and Ce. Key Results: La and Ce did not increase the growth of corn or mungbean. Thedry weight of corn shoots was decreased by 32 % in the presenceof 5·0 µM Ce; the other La and Ce concentrationshad no effect. La and Ce concentrations of 0·9 and 5·0µM decreased the shoot dry weight of mungbean by 75 or95 %, the two elements having closely similar effects. Decreasesin the uptake of Ca, Na, Zn and Mn by corn were observed withincreases in solution La and Ce. For mungbean, the uptake ratesof all measured elements decreased with increases in solutionLa and Ce. The concentrations of La and Ce in the roots of bothspecies were higher than in the shoots and increased stronglywith increasing concentrations of La or Ce in solution. TheLa and Ce concentrations in mungbean shoots were always higherthan in corn shoots. Conclusions: La and Ce did not enhance the growth of corn or mungbean, butdecreased the growth, root function and consequently the nutritionalstatus of mungbean at concentrations >0·2 µMin solution. It is concluded that if La or Ce have positiveeffects on corn and mungbean growth, they can only occur atsolution concentrations below 0·2 µM.  相似文献

Continuous flooding of the soil (‘flooded’ treatment)gave best growth of IR-8 variety of rice whereas soil drainedfor 4 weeks and then flooded for 8 (‘drained and flooded’treatment) resulted in poorest growth and chlorotic plants.Plants grown in the continuously drained soil (‘drained’treatment) and those in the soil flooded for 4 weeks and thendrained for 8 (‘flooded and drained’ treatment)showed intermediate growth. There were no differences in therelative water content of plants growing in the various treatments.Analyses of plant tissues showed that a consideration of therelative concentration of Fe, Mn, and P in the shoots is mostclosely related to the performance of rice under various culturalconditions. An increase in the concentration of Fe in the planttissues following flooding was correlated with the best growth(‘flooded’ treatment) unless it was accompaniedby high level of Mn (as in the ‘drained and flooded’treatment) which may have proved toxic, e.g. by interferencewith Fe metabolism as was evidenced by chlorosis. Measurementsof oxidation-reduction potentials, oxygen diffusion rates, andthe concentration of exchangeable and soluble Fe and Mn in thesoils have shown that the ‘drained and flooded’treatment caused the most extreme reducing conditions. Floodingaccompanied by the development of extreme reducing conditionsled to a greater accumulation of Mn in the shoots (‘drainedand flooded’ treatment) whereas flooding accompanied bythe maintenance of oxidizing conditions (‘flooded’treatment) resulted in a lower uptake of Mn. Growth of riceplants for 4 weeks in the drained soil did not fit them forthe reduced conditions which developed during subsequent flooding(‘drained and flooded’ treatment).  相似文献

The effectsof flooding on the uptake and leakage of ‘non-electrolytes’by roots.—J. exp. Bot. 36: 1705–1715. The object of this work was to examine some of the early effectsof flooding on roots. A hydroponic system was developed thatgave good control over watering, degree of oxygenation of thebathing medium and allowed measurement of short term changesin the composition of the bathing medium. Excised roots, floodedfor 24 h, were shown to take up less [3H) ß-alaninethan non-flooded roots and also leaked more [3H] ß-alanineinto a distilled water bathing medium. Further, flooded excisedroots lost more protein to the bathing medium, with ‘young’(5–7 d) roots showing greater losses than ‘old’(11–14 d) roots. However, young roots had more proteinin the tissue even after greater loss. Young roots remainedhealthier and lost less fresh weight than old roots. Two lines of Grand Rapids lettuce were isolated by self-pollinationof single plants. Seeds of one line, designated ‘A’,germinate in the dark at 25 °C. Seeds of the second line,designated ‘B’ germinate in a manner typical oftheir variety. The lines differ in sensitivity to red (R) andfar-red (FR) light, in the ability of their seeds to overcomeresistance of an external osmotically active medium, in theresponse of mature seeds to abscisic acid, and in the responseof young seeds to gibberellin and benzyl adenine. ‘A’seeds germinate faster than ‘B’ seeds in the darkor after R or FR irradiation.  相似文献

The influence of different sugars on shoot multiplication invitro and on the activity of invertase was studied with sourcherry (Prunus cerasus L.) cultures. The sugars sucrose, glucoseand fructose, and the sugar alcohol sorbitol, were investigatedat a wide range of concentrations. The optimum concentrationsof all carbon sources were 2% and 3% (w/v). Sucrose and glucosefavoured a similar rate of proliferation. However, in the presenceof fructose, proliferation was lowest but was coupled with thehighest frequency of formation of long shoots. The highest activityof total invertase was for tissues growing on a sucrose-containingmedium while, in a sugar-free medium, invertase activity wasmainly found in the ‘salt extracted’ fraction. Inthe remaining treatments, ‘soluble invertase’ dominated.For each sugar investigated, the activity of both forms of invertasewas significantly higher at 2% than at 3% (w/v) sugar. A comparison was made of effects of ‘low’ and ‘high’soil temperature (LST and HST, about 9 and 21·5 °Crespectively) on shoot growth of Norwegian and Portuguese populationsof Dactylis glomerata. In experiments lasting 8 short days (SD,8 h photoperiods) LST decreased leaf extension more markedlyin the Portuguese population. No differential effect of LSTon leaf growth was recorded in experiments lasting 20 or 21SD or in experiments of 8 d duration in long days (LD, 16 hphotoperiods). Since the meristem and region of cell extensionis close to the soil surface LST could directly influence bothroot and shoot growth. The application of gibberellic acid enhanced leaf extension,particularly in plants grown at HST. 6-Benzylaminopurine tendedto decrease leaf length. Extractable gibberellin levels wererelatively low after 8 SD in shoots of both races grown at LST.Cytokinin levels increased at LST, more so in shoots of Portugueseplants which sustained the greater reduction in leaf extension. The data suggest that LST may reduce the production of endogenousgibberellins important for leaf growth in Dactylis glomerata.Cytokinins are probably necessary for growth processes but theirlevel may reflect, rather than direct, the rate of leaf growth.  相似文献
