Lateral Bud Dormancy in the Blackcurrant Ribes nigrum (L.) 总被引:1,自引:0,他引:1
Previous investigations have shown that the inhibition of lateralbuds in blackcurrants soon after their formation is due to apicaldominance and this is followed by leaf inhibition. The presentstudies have revealed that bud scales are the main cause ofinhibition even before leaf fall. The inhibitor level assayedby the wheat coleoptile test shows large seasonal changes; itis maximal in early autumn and minimal in spring. Inhibitor levels are reduced by low-temperature treatment inparallel with the promotion of dormancy breaking; temperatureswell below freezing, applied for very short times, almost certainlyact through the killing of bud scales in the same way as doesrapid high-temperature treatment. Other treatments causing budbreak in the presence of high inhibitor levels are: continuousillumination, ethylene vapour, and application of gibberellicacid. It is suggested that bud inhibition in the blackcurrant is dueto the formation of an inhibitor produced by the leaves andaccumulated in the bud scales. The inhibitor (probably abscisicacid) may then interact with internal gibberellin levels, summerand winter dormancy differing only quantitatively. 相似文献

Ribes nigrum germplasm was screened for random amplified polymorphic DNA (RAPD) markers. Fiftyfour markers were identified which generated individual fingerprints for each of 21 cultivars. Genetic variation within R. nigrum germplasm, as detected by RAPDs, demonstrated that the genetic basis for improvement of blackcurrant is narrower than would be expected by the analysis of parentage. 相似文献

Overwintering buds of blackcurrant cultivars 'Ben Lomond' and'Ben More' were examined by differential thermal analysis (DTA).Photographic evidence relates the first (primary) exotherm tothe freezing of water in the basal pith and bud scales. Thenumber of secondary exotherms either matched, or was fewer than,the number of floral racemes within the bud. The first genetic linkage map of blackcurrant, published by Brennan et al. (Euphytica 161:19–34, 2008), identified regions where quantitative trait loci (QTLs) for some important traits were located. The analysis was complicated by the fact that the mapping population was found to contain two subgroups, with segregation ratios consistent with these being crossed and selfed offspring. The QTL analysis was based on the trait mean over 3 years and focused on the crossed offspring. Here we proposed a mixed model multi-environment approach for this population. The 3 years are considered as three separate environments, the data from both the selfed and crossed offspring are combined and different residual terms are explored to model the correlation between the years. This permits tests for interactions between the QTLs, the year and the type of offspring (selfed or crossed). This is applied to re-analyse two important traits, anthocyanin concentration and budbreak. Several additional QTLs were identified, some affecting the trait in both the selfed and crossed offspring, others in just one. 相似文献

Heterozygosity in three cultivars of the blackcurrant (Ribes nigrum L.) was estimated. A selfed population of each cultivar was screened for Random Amplified Polymorphic DNA markers (RAPDs) and heterozygous loci were identified by band segregation in contrast with the non-segregation of homozygous loci. On average, 21% of the loci scored in each cultivar were heterozygous. The implications for mapping studies in Ribes nigrum are discussed. 相似文献

Free and bound abscisic acid (ABA) levels in blackcurrant andbeech buds have been determined by an extraction procedure involvingseveral thin-layer chromatography purification stages and finalbioassay in the wheat coleoptile straight-growth test. In bothspecies the highest level of free ABA occurs in the autumn atabout the time of onset of winter dormancy. The free ABA contentthen declines throughout the winter months reaching its lowestvalue just before bud burst. These results strengthen the viewthat free ABA plays an important role in the induction and maintenanceof winter dormancy. The bound ABA level increases in both species throughout theautumn and winter until halfway through the bud swelling phaseand then declines. It is proposed that the bound ABA accumulatesin the buds because it cannot be translocated or further metabolizedduring the winter months. The ratios of free to bound ABA were plotted and the resultingcurves show a distinctive annual cycle with the highest freeABA/ bound ABA ratio (F/B) occurring in the autumn and the highestbound ABA/ free ABA ratio (B/F) coinciding with bud burst. Agradual increase in the B/F ratio during bud swelling may indicatea feed-back reaction between free and bound ABA which maintainsthe free ABA at a sufficiently high level to put a gentle brakeon growth. At bud burst the B/F ratio falls very sharply anda period of very rapid growth ensues. 相似文献

The fruit juice of black currant was found to contain a polysaccharide-rich substance, which was designated cassis polysaccharide (CAPS), with macrophage-stimulating activity. Especially, its interleukin (IL)-1beta-inducing activity was remarkably high, compared with other fruit juice preparations. CAPS was found to consist of rhamnose, mannose, arabinose, galactose, xylose, and glucose in a molar ratio of 11.3:0.9:54.1:29.8:2.0:1.9. CAPS turned out to be partitioned into a soluble component (CAPS-l.m.) and a precipitable component (CAPS-h.m.) with mean MWs of 80,000 and 600,000 respectively in 45% (v/v) ethanol solution. At least in vitro, CAPS-l.m. rather than CAPS-h.m. appeared to play an important role in macrophage activation. Oral administration of black currant juice and CAPS to Ehrlich carcinoma-bearing mice retarded the growth of the solid tumor by 45% and 51% respectively. CAPS administration had a stimulatory effect on the release of IL-2, IL-10, interferon-gamma, and IL-4 from splenocytes in comparison with PBS treatment in tumor-bearing mice. The IL-4 level was, however, still lower than that exhibited by a group of normal mice. CAPS showed a certain cytotoxicity directly against tumor cells. 相似文献

The floral volatiles of blackcurrant (Ribes nigrum) were collected in situ using the headspace technique. Eleven compounds were identified, including monoterpenes hydrocarbons and monoterpene ethers. The fragrance was emitted in a rhythmic manner, the maximum being in the middle of the photoperiod. Emission at 20°C was significantly higher than at 10°C. The rhythmic nature of emission seemed to cease under conditions of constant light. However, a slight rise in emission during the second period in continuous light may indicate a circadian control of emission. The significance of flower volatiles in relation to pollination is discussed. 相似文献

In this report, we show that the non-conjugated octadecatetraenoic acid found in the oil of the seeds from Ribes nigrum is identical to the C18-polyunsaturated fatty acid previously isolated in a number of fish oils and seed oils. Evidence obtained from mass spectral data of its triazolopyridine derivative clearly indicates the presence of methylene-interrupted double bonds. Comparison with authentic material prepared by chemical synthesis provides further confirmation of the (all-cis)-6,9,12,15-octadecatetraenoic acid structure. The (all-cis)-4,8,12,15-structural arrangement erroneously attributed to this acid in several literature reports is thus definitely ruled out. 相似文献

Two novel nitrile-containing compounds, nigrumin-5-p-coumarate and nigrumin-5-ferulate, together with six known flavonoids, were isolated from the seed of black currant (Ribes nigrum). The chemical structures of nigrumin-5-p-coumarate and 5-ferulate were elucidated using NMR spectroscopy as 2-trans-p-coumaroyloxymethyl-4-beta-D-glucopyranosyloxy-2(E)-butenenitrile and 2-trans-feruloyloxymethyl-4-beta-D-glucopyranosyloxy-2(E)-butenenitrile, respectively. 相似文献

A genotyping by sequencing (GbS) approach is reported in blackcurrant (Ribes nigrum L.) using a de novo read assembly method developed because of the current absence of a reference genome sequence for this species. A new approach to single nucleotide polymorphism (SNP) genotype calling is described, where individual genotypes for a large number of SNPs were characterised from the GbS counts using a novel method based on a functional regression of major and minor allele read counts. The high-quality GbS SNPs were combined with SNPs and simple sequence repeats generated from other technologies to develop a linkage map with increased marker density and improved genome coverage, containing up to 204 SNPs on each linkage group. SNPs of lower quality were then located on the map using quantitative trait locus (QTL) interval mapping of the proportion of the major allele. Two QTL each for 100-berry weight and Brix scores, measured over three years, were identified using the map. The use of this approach to identify and map a significant number of novel SNPs in a woody species with hitherto limited genomic resources may have generic application to other under-resourced and minor species in the development of cost-effective and efficient high-density genetic maps. 相似文献

The terpenoid fractions of bud extracts from blackcurrant genotypes showing resistance or susceptibility to gall mite (Cecidophyopsis ribis) infestation were examined and compared using gas chromatography. By the use of discriminant analysis, and the application of the derived discriminator to the data, resistance status was correctly predicted in 88% of the genotypes sampled. A polysaccharide-rich substance isolated from black currant, named cassis polysaccharide (CAPS), was partially digested with β-galactosidase from Aspergillus oryzae and its immunostimulatory activity was investigated. The in vitro cytokine-inducing effect of CAPS on RAW264 cells was gradually decreased along with lowering of the average MW of CAPS. In vivo, partially digested CAPS with a mean MW of approximately 20,000 showed the most potent antitumor activity against Ehrlich carcinoma in mice. 相似文献

Ribosomal DNA from Cecidophyopsis mites from different Ribes species was amplified using the polymerase chain reaction and the products digested using restriction enzymes. After separating the DNA fragments on gels, it was possible to identify specimens of mites obtained from field samples by comparing the profiles of their DNA banding patterns with those of known Cecidophyopsis species. Using this analysis, a non-gall forming mite found infesting blackcurrant buds in New Zealand was identified as the gooseberry mite (C. grossulariae). On wild red currant (Ribes spicatum) from Finland showing two sizes of galled buds, the red currant gall mite (C. selachodon) was identified in the larger galls located at the tips of branches and a distinct mite in the smaller galls located on the lower parts of the branches. A mite with a DNA banding profile indistinguishable from this latter mite from R. spicatum was also identified in galled buds of blackcurrant genotypes growing in Finland, including those containing the blackcurrant gall mite (C. n'ftw)-resistance genes P or Ce. The DNA banding profile of this mite resembled most closely that of C. ribis , but was distinct from it. The occurrence of C. grossulariae and this distinct Cecidophyopsis mite on blackcurrant has implications for the genetic control of Cecidophyopsis mites and possibly for the spread of the reversion disease agent in this crop. 相似文献
