Animals have evolved adhesive structures on their legs to cling to the substrate during locomotion. Here we characterise the ultrastructure and mechanical properties of adhesive pads in Carausius morosus (Phasmatodea) using atomic force microscopy (AFM) as well as transmission and scanning electron microscopy (TEM, SEM). The smooth adhesive arolium has a soft cuticle consisting of principal rods, which branch into finer fibres near the surface. Indentation experiments showed that the pad material consists of distinct layers with different mechanical properties. The 100–300 nm thick outermost layer consisting of the cuticulin envelope and the epicuticle is extremely soft and resilient (mean effective Young’s modulus 12 kPa), while the subjacent procuticle is a much stiffer material (mean effective Young’s modulus 625 kPa). AFM contact mode imaging revealed that the cuticle is mechanically anisotropic, which can be explained by its fibrillar inner structure. We propose that the described layered structure of smooth adhesive pads, consisting of materials decreasing in stiffness towards the outer surface, represents a superior design to conform and adhere to substrates with roughnesses at different length scales. This design principle could be easily implemented in technical adhesives, and thus has a potential to inspire biomimetic applications.  相似文献

We investigated insects Carausius morosus walking whilst hanging upside down along a narrow 3 mm horizontal beam. At the end of the beam, the animal takes a 180° turn. This is a difficult situation because substrate area is small and moves relative to the body during the turn. We investigated how leg movements are organised during this turn. A non-contact of either front leg appears to indicate the end of the beam. However, a turn can only begin if the hind legs stand in an appropriate position relative to each other; the outer hind leg must not be placed posterior to the inner hind leg. When starting the turn, both front legs are lifted and usually held in a relatively stable position and then the inner middle leg performs a swing-and-search movement: The leg begins a swing, which is continued by a searching movement to the side and to the rear, and eventually grasps the beam. At the same time the body is turned usually being supported by the outer middle leg and both hind legs. Then front legs followed by the outer middle leg reach the beam. A scheme describing the turns based on a few simple behavioural elements is proposed.  相似文献

Adhesive organs on the legs of arthropods and vertebrates are strongly direction dependent, making contact only when pulled towards the body but detaching when pushed away from it. Here we show that the two types of attachment pads found in cockroaches (Nauphoeta cinerea), tarsal euplantulae and pretarsal arolium, serve fundamentally different functions. Video recordings of vertical climbing revealed that euplantulae are almost exclusively engaged with the substrate when legs are pushing, whereas arolia make contact when pulling. Thus, upward-climbing cockroaches used front leg arolia and hind leg euplantulae, whereas hind leg arolia and front leg euplantulae were engaged during downward climbing. Single-leg friction force measurements showed that the arolium and euplantulae have an opposite direction dependence. Euplantulae achieved maximum friction when pushed distally, whereas arolium forces were maximal during proximal pulls. This direction dependence was not explained by the variation of shear stress but by different contact areas during pushing or pulling. The changes in contact area result from the arrangement of the flexible tarsal chain, tending to detach the arolium when pushing and to peel off euplantulae when in tension. Our results suggest that the euplantulae in cockroaches are not adhesive organs but 'friction pads', mainly providing the necessary traction during locomotion.  相似文献

In order to understand the attachment mechanism of flies, it is important to clarify the question of how the adhesive pad (pulvillus) builds and breaks the contact with the substrate. By using normal and high-speed video recordings, the present study revealed that pulvilli are positioned on the surface in a particular way. The pulvilli are apparently loaded or pressed upon the substrate after leg contact, as evidenced by splaying of the claws. Detachment of pulvilli from the substrate may be achieved in four different modes depending on the leg (fore-, mid- or hindleg): pulling, shifting, twisting, and lifting. Lifting is the only detachment mode depending on the claws' action. Kinematics of the tarsal chain is studied in leg preparations, in which the tendon of the claw flexor muscle was pulled by tweezers and video recorded. The morphological background of tarsal movements during attachment and detachment is studied by scanning electron microscopy, fluorescent microscopy, and bright field light microscopy followed by serial semithin sectioning of pretarsal structures. Several resilin-bearing springs are involved in the recoil of the tarsal segments to their initial position, when the tendon is released after pull.  相似文献

Summary Carausius morosus starved and deprived of water lose about 30% of their body weight in 4 days, mainly due to water loss. Isolated inferior tubules from starved dehydrated insects secrete urine at 0.041 nl·mm–1·min–1 compared with 0.118 nl·mm–1·min–1 in those from fed hydrated insects. This difference is due partly to the level of a diuretic (and perhaps also an antidiuretic) hormone in the haemolymph acting directly on the urine-secreting mechanism and partly to changes in the intrinsic capacity of the tubule cells for urine secretion. This latter change is accompanied by structural changes in the tubules. During starvation and dehydration the lumen becomes packed with white granules, the height of the type 1 cells is reduced, their basal infoldings and brush border become shorter and their mitochondrial volume is reduced.  相似文献

During embryonic development, insect vitellins (Vt) are degraded by limited proteolysis to yield a number of lower-molecular weight polypeptides. The aim of the present study was to identify these polypeptides in the embryo and to verify how they relate to Vt polypeptides deposited in the oocyte during vitellogenesis. To this end a panel of poly- and monoclonal antibodies (Pab, Mab) was raised against Vt polypeptides and employed by immunoelectrophoresis and immunoblotting on embryos belonging to different developmental stages. Through this approach three major staining patterns were observed. First, Mab 4 reacts with both polypeptides B1 and E20, suggesting that polypeptide B1 is gradually trimmed to yield polypeptide E20 in late embryos. Second, Mab 12 is specific for polypeptide A3 which is retained unchanged throughout embryogenesis. Third, Pab anti-A2 and Mab 13 show that polypeptide A2 is processed to yield polypeptide E9 through limited proteolysis. In conclusion, the staining patterns reported in this study show that Vt polypeptides in developing embryos of the stick insect Carausius morosus undergo at least two major processing events concerning polypeptides B1 and A2.  相似文献

Summary Histological sections of the brain, suboesophageal ganglion, and the corpora cardiaca/corpora allata complex were examined for the presence of crustacean hyperglycemic hormone-like immunoreactive material. With the use of an antiserum directed against the hyperglycemic hormone of Carcinus maenas, immunofluorescence was found in the median portion of the pars intercerebralis, and the corpora cardiaca. Extracts of corpora cardiaca were examined by radioimmunoassay for competitive binding to the antiserum; one pair of corpora cardiaca contains at least 7 pg crustacean hyperglycemic hormone-like material.  相似文献

The effect ofCyrtobagous salviniae, Samea multiplicalis, andPaulinia acuminata on the growth ofSalvinia molesta was assessed in relation to temperature in field cages at 2 sites in Brazil, a canal with running-water and a lagoon with stillwater. Each insect species reduced the growth ofS. molesta over a mean leaftemperature range of 16–30°C, causing more damage as temperature increased. There was no difference between the 3 species in response to temperature. These results and their implication to the performance of these insects as biological control agents againstS. molesta are discussed.

A budding coccoid bacterium, (CH1), a Vibrio sp. and a Pseudomonas sp. were investigated for factors governing their attachment to glass surfaces in static batch culture and laminar flow continuous culture systems. An analysis of variance showed that the three species exhibited very different responses. For CH1 attachment was dependent on cell density, incubation time and nutrient concentration. The Vibrio sp. was affected by nutrient concentration while the attachment of the Pseudomonas sp. was independent of cell density, incubation time and nutrient concentration. A comparison of attachment to hydrophilic and hydrophobic surfaces showed that attachment of the Vibrio sp. and CH1 to hydrophilic surfaces was 3 and 10 times greater respectively than to hydrophobic surfaces while Pseudomonas attached in equal numbers to both surfaces. The continuous culture system with defined flow hydrodynamics and growth conditions at steady state revealed a random sampling effect 3 times smaller than the batch culture system did. When the biofilm development of Pseudomonas sp. was followed during 46 h at different fluid shear under laminar and turbulent flow conditions, the former biofilm reached 3.3·108 cells·cm-2 and the latter 8.2·107 cells·cm-2.Non-common abbreviation NSS Nine salt solution  相似文献

Summary Electron-microscopic immunocytochemistry for the demonstration of CCK-like material in basal endocrine cells of the midgut of Aeschna cyanea, Locusta migratoria, Carausius morosus and Periplaneta americana was performed by use of the peroxidase-antiperoxidase procedure and the colloidal gold method. Immunoreactive cells appeared scattered among digestive and regenerative cells of the epithelium. Immunoreactivity was specifically detected over round to oval electron-dense granules whose size appeared rather different from species to species. Thus, the average size of 30% (d30) of the largest granules ranged from 312 nm in Periplaneta americana to 159 nm in Carausius morosus with intermediate values in Aeschna cyanea (d30=195 nm) and Locusta migratoria (d30=225 nm).  相似文献

The early in vitro kinetics of Candida albicans attachment to human buccal epithelial cells was studied with the aid of an adhesion assay and solutions of concanavalin A (Con A), a lectin which is capable of inhibiting yeast adhesion. Various saccharides and putative receptor analogues were also tested. Solutions of each single reagent were added to tubes containing aliquots of mucosal cells and germinated yeasts at the beginning of a 1-hour incubation period (time O) or at 10 minute intervals during the assay. The number of yeasts attached to 200 mucosal cells was subsequently determined microscopically. Yeast adhesion remained constant following addition of phosphate-buffered saline (PBS) at time 0 or at any time thereafter. However, addition of Con A at 0, 10 or 20 minutes of incubation decreased adhesion significantly to 38%, 45% and 63% of control values. This inhibitory effect dwindled as time of incubation prior to lectin addition increased and Con A could not inhibit adhesion significantly after twenty minutes. Results obtained with Con A using live germinated yeasts were similar to those obtained with formalin-killed C. albicans. The other reagents tested failed to decrease adhesion significantly. These included the putative receptor analogues fibronectin, N-acetyl-d-glucosamine and d-galactose, and several non-specific saccharides such as -d-methylglucopyranoside, d-ribose and d-xylose. It is suggested that in vitro attachment to human mucosal cells by C. albicans is inhibitable up to a defined point in time by a lectin with affinity for mannosecontaining surface moieties, but becomes non-reversible thereafter. We report and discuss effects of four insect growth regulators: buprofezin, fenoxycarb pyriproxyfen and chlorfluazuron, at concentrations recommended for agricultural use on six species of natural enemies of homopteran pests. Dipping in buprofezin had no appreciable effect on adult mortality, oviposition and development ofComperiella bifasciata (Howard), (Hymenoptera: Encyrtidae). When exposed to hosts treated with buprofezin, percentage mortality of adultEncyrtus infelix Embleton (Encyrtidae) was low; buprofezin had some detrimental effect on immature stages ofE. infelix when applied prior to parasitization, but not when introduced after parasitization. Buprofezin had a slight effect on the immature stages ofCryptochaetum iceryae Williston (Diptera: Cryptochaetidae), while fenoxycarb and pyriproxyfen had marked detrimental effects on parasitization and/or development of the parasitoid fly. None of the larvae ofRodolia cardinalis Mulsant (Coleoptera: Coccinellidae) developed into adults after application of buprofezin, fenoxycarb or pyriproxyfen. Buprofezin and chlorfluazuron completely prevented egg hatch ofChilocorus bipustulatus L. (Coleoptera: Coccinellidae). Buprofezin did not adversely affect egg hatch and larval development ofElatophilus hebraicus Pericart (Hemiptera: Anthocoridae); fenoxycarb or pyriproxyfen applied either before or after oviposition on pine needles caused total suppression of egg hatch.  相似文献

Abundances and interactions among biological control insects and their effects on target invasive plants were monitored within the flower heads and roots of diffuse knapweed, Centaurea diffusa, and in spotted knapweed, Centaurea stoebe, along the Colorado Front Range. Flower weevils, (Larinus species) and root-feeders (Cyphocleonus achates and Sphenoptera jugoslavica) were released on knapweed that already supported biological control gall flies (Urophora species). At a single monitoring site, seed production by C. diffusa declined from 4400 seeds m−2 in 1997 to zero seeds m−2 on the monitoring sites in 2006, while the flowering stem density of C. diffusa declined from a peak of almost 30 stems m−2 in 2000 to zero stems m−2 in 2006. The average abundance of Urophora and Larinus in flower heads fluctuated independently during the 2001–2006 interval, while the relative abundance of C. achates and S. jugoslavica in roots exhibited a weak inverse relationship that appeared driven by climate effects. The relative abundance of insects on a population of C. stoebe was monitored for five years as Larinus species and C. achates became established on spotted knapweed that already supported Urophora species. Spotted knapweed seed production on our monitoring site declined from 4600 seeds m−2 in 2003 to zero seeds m−2 in 2006. Unlike C. diffusa, substantial numbers of rosettes of C. stoebe remained present. Larinus consumed almost all Urophora encountered in C. diffusa, and consumed about 40% of the Urophora in co-infested flower heads of C. stoebe (ca. 10–15% of the total Urophora population). No negative correlations between the relative densities of flower head and root-feeding insects were observed. The effects of these insects on target plants have produced results consistent with the ‘cumulative stress hypothesis’ for biological control of Centaurea species.  相似文献

Transformation of vinca cells was performed by the co-cultivation of cell-wall regenerated vinca protoplasts withAgrobacterium tumefaciens. Using thisin vitro and single cell system, attachment of the bacteria to the surface of vinca cells was observed by scanning electron microscopy (SEM). Figures of the bacteria polarly binding to the plant cell wall were often observed. AsEscherichia coli does not attach to the plant cells at all, the observed attachment ofA. tumefaciens is suggested as a characteristic feature in crown gall induction. Even though no evidence of transformation was obtained by the co-cultivation methods, a similar attachment was observed in the cell-wall regenerated protoplasts of rice. The bacteria also attached to the surface of isolated mesophyll cells of asparagus and root hairs of rice. From these observation, we concluded that the attachment is not the limiting step of crown gall induction byA. tumefaciens in monocotyledonous plants. Extracellular fibrils like pili were observed with a few strains of A.tumefaciens for the first time. These fibrils were observed regardless of their ability of attachment and infectivity.  相似文献

Summary Receptor cell axons from the antennal flagellum terminate in the glomeruli of the ipsilateral deutocerebrum in Periplaneta americana and Locusta migratoria. Processes from several groups of deutocerebral neurons also enter the glomeruli and terminate in characteristic branching patterns. There, they contact the antennal axons. Connections are both convergent and divergent. Not only do single central neurons collect the inputs from many receptor cells, but receptor axons were often observed to branch and terminate at more than one deutocerebral neuron. The axons from a portion of the neurons go to form the deutocerebral bundle of the tractus olfactorioglobularis. These axons of the bundle terminate in the ipsilateral calyx of the corpus pedunculatum and in the lateral lobus protocerebri. The processes of the majority of the deutocerebral neurons stay within the deutocerebrum itself and may serve as local interneurons. Part of some antennal fibers terminate in the lobus dorsalis. The lobus glomeratus receives inputs from the maxillary palps and also from processes of deutocerebral neurons.Electron microscopy of synaptic connections and anatomical experiments reveal a complicated pattern of connections between receptor axons and higher order neurons as well as between higher order neurons themselves within the glomeruli.The ratio of the number of antennal fibers to that of relay fibers could easily lead to the interpretation, that the deutocerebrum merely serves as a device for reducing the number of transmission channels. However, coupled with physiological data, anatomical details such as conand divergence of input and interconnections between input channels suggest rather a filtering system and a highly complicated integrative network.  相似文献

Larvae of Deliaradicum Linnaeus (Diptera: Anthomyiidae) wereexposed to Steinernema feltiae Filipjev(Rhabditida) to study the ability of thenematodes to fulfill their life cycle (recycle)in this host. In addition, the recycling ofSteinernema and Heterorhabditis(Rhabditida) species was studied in larvae ofthe following insects: Meligethes spp.Stephens (Coleoptera: Nitidulidae),Dasyneura brassicae Winnertz (Diptera:Cecidomyidae), Ceutorrhynchus assimilisPaykull, C. pallidactylus Marsham (Coleoptera: Curculionidae) and Mamestrabrassicae Linnaeus (Lepidoptera: Noctuidae).All larval instars of D. radicum werestudied (larvae live and pupate in soil)whereas the final instars of the remaininginsects were studied (larvae only pupate in soil).On average, the following numbers of infectivejuveniles were produced in the different hosts:1,200–1,400 in C. assimilis; 700–1,300 inMeligethes spp. and 47,000 in M.brassicae. In D. radicum, around 400infective juveniles were produced on average inthe smallest larvae and around 3,500 in thelargest larvae. The highest number of nematodesrecorded in a D. radicum larva (thirdinstar) was 9,500 infective juveniles. Thenumber of nematodes produced in D.radicum cadavers was positively related to thesize of the insect host, but large variationwas observed.  相似文献

The effects of cultivation pH and agitation rate on growth and extracellular xylanase production by Aspergillus oryzae NRRL 3485 were investigated in bioreactor cultures using spent sulphite liquor (SSL) and oats spelts xylan as respective carbon substrates. Xylanase production by this fungus was greatly affected by the culture pH, with pH 7.5 resulting in a high extracellular xylanase activity in the SSL-based medium as well as in a complex medium with xylan as carbon substrate. This effect, therefore, was not solely due to growth inhibition at the lower pH values by the acetic acid in the SSL. The xylanase activity in the SSL medium peaked at 199 U ml(-1) at pH 7.5 with a corresponding maximum specific growth rate of 0.39 h(-1). By contrast, the maximum extracellular beta-xylosidase activity pf 0.36 U ml(-1) was recorded at pH 4.0. Three low molecular weight xylanase isozymes were secreted at all pH values within the range of pH 4-8, whereas cellulase activity on both carbon substrates was negligible. Impeller tip velocities within the range of 1.56-3.12 m s(-1) had no marked effect, either on the xylanase activity, or on the maximum volumetric rate of xylanase production. These results also demonstrated that SSL constituted a suitable carbon feedstock as well as inducer for xylanase production in aerobic submerged culture by this strain of A. oryzae.  相似文献

Host diet affects the defensive efficacy of Neodiprion sawflies. In laboratory assays with wood ants (Formica obscuripes), secretions from larvae reared on Pinus banksiana were the most repellent, while those from P. resinosa feeders were the least so. This was explained diterpene resin acid content, but not total monoterpene content. The terpene content of regurgitant generally reflected dietary concentrations. Compounds were sequestered nonselectively by larvae. Host-based differences in defense persisted at the behavioral level. P. banksiana feeders regurgitated greater volumes of fluid and were less likely to be disabled or killed by ants in one-on-one interactions than were larvae fed on P. resinosa. The defensive advantages of host diet conflicted with developmental requirements. N. sertifer reared on P. banksiana (the best diet for defense in all cases) had lower cocoon weights (a correlate of fecundity) than those reared on other diets, and had prolonged larval development compared to insects fed P. sylvestris. No such tradeoff was detected in N. lecontei. Larvae of both species strongly preferred P. banksiana over P. resinosa in feeding choice assays.  相似文献

The evolution of larval head morphology in holometabolous insects is characterized by reduction of antennal appendages and the visual system components. Little insight has been gained into molecular developmental changes underlying this morphological diversification. Here we compare the expression of the segment polarity gene wingless (wg) in the pregnathal head of fruit fly, flour beetle and grasshopper embryos. We provide evidence that wg activity contributes to segment border formation, and, subsequently, the separation of the visual system and protocerebrum anlagen in the anterior procephalon. In directly developing insects like grasshopper, seven expression domains are formed during this process. The activation of four of these, which correspond to polar expression pairs in the optic lobe anlagen and the protocerebral ectoderm, has shifted to postembryonic stages in flour beetle and Drosophila. The remaining three domains map to the protocerebral neuroectoderm, and form by disintegration of a large precursor domain in flour beetle and grasshopper. In Drosophila, the precursor domain remains intact, constituting the previously described “head blob”. These data document major changes in the expression of an early patterning gene correlated with the dramatic evolution of embryonic visual system development in the Holometabola.  相似文献

The relationship between oviposition preference and growth, survival, and reproduction of offspring is the crux of the problem in the evolution of host associations between phytophagous insects and plants. Observed relationships between oviposition preference and performance of offspring range from good to poor. At least four hypotheses have been suggested to explain observed use of particular host plants that may not result in the fastest growth rates or greatest pupal masses: time, patch dynamics, parasite versus grazer lifestyles, and enemy-free space. Our current understanding of these relationships, however, is hampered by an almost complete lack of data on how preference and performance are related genetically. These data are needed to understand the origins of covariance between preference and performance and constraints on the evolution of host associations.  相似文献
