Summary The reproductive stages of the life cycle ofAcetabularia mediterranea have been studied in Feulgen-stained material. Light-microscopical photographs of secondary nuclei, cyst formation, gamete formation, gamete release, zygote formation and early development of the germlings are presented. The time course of cytological events during gamete formation is described. Mitoses are found between 16 and 24 hours after the induction of cyst germination.The author is indebted to Mrs. S.Artelt who helped with the prepration of the specimens and the photographs.  相似文献

Summary Ultrastructural investigation of the flagellar root system ofAcetabularia gametes reveals one type of organization for both male and female gametes. There is a modified cruciate system with four microtubular bands X-2-X-2, with X=4. A prominent distal striated fiber and a small proximal striated fiber connect the flagellar bases. A striated root fiber type I underlies the microtubular root type II, and a short striated root fiber type I underlies the microtubular root type I (terminology ofMelkonian 1980 b). This specific root system has some details in common with theChlamydomonas type, and others with theUlvaphyceae and the siphonalean algaeDerbesia andBryopsis. This might indicate the phylogenetic relationships.  相似文献

Summary The normal resting potential across theAcetabularia mediterranea cell membrane is –170 mV and the resistance is about 0.1 k·cm2. The time courses of potential and resistance changes have been studied in connection with several slow dynamic properties of the membrane. These effects include spontaneous and stimulated depolarizing spikes, spontaneous repolarization after prolonged maintenance of a quasistable depolarized condition, and a hyperpolarizing response when current is applied to the cell in this depolarized state. These processes show considerable similarities to each other, which suggests that they might all be explained by changes in permeability to Cl. In normal conditions, membrane punch-through occurs with a relatively small hyperpolarizing bias.  相似文献

Summary YoungAcetabularia mediterranea cells without cap have a fatty acid composition different from other green algae currently used for biological research. They contain important quantities of palmitic and oleic acid, but are very poor in polyunsaturated fatty acids such as linoleic and linolenic acid. Summary A polarographic method using a platinum electrode permits continuous recording of the oxygen balance of a singleAcetabularia cell during several days. The amount of oxygen which is consumed by the electrode is immeasurably small. The accuracy of the method is definitely better than 0.05l O2/ml medium. The resolution in time is less than 5 min. The length of one period in the circadian rhythm of photosynthesis inAcetabularia has been found to be about 27 hours.  相似文献

An ultrastructural analysis of the gametogenetic phases in Branchiura sowerbyi, a tubificid oligochaete, has been accomplished. These phases mostly conform to the usual pattern for the family, however, some interesting peculiarities are pointed out. An ultrastructural study of life cycle stages of Pneumocystis carinii in infected rat lungs in situ was undertaken utilizing 8 different modes of fixation. Three of the fixatives employed gave good fixation of cysts and intracystic bodies, but for the trophic forms fixation was only fair. Both the trophic forms and intracystic bodies have nuclear pores. The mitochondria of the organism have cristae that appear lamellar. One of the fixation modes revealed a thin, electron-dense layer on the outer surface of the cell wall, a "fuzzy coat" that had not been described previously. This material appears to mediate tight adhesion of trophic forms with other trophic forms, cysts, and with pneumocytes of the lung alveolus.  相似文献

Summary Cells ofAcetabularia mediterranea were irradiated for three weeks with red light and were subsequently transferred to blue light of equal quantum flux density. Blue light induces protein and chlorophyll synthesis. The chloroplasts multiply intensively between the 8th and 24th hour in blue light and within that time the chloroplast number doubles.Plastid protein, thylacoid protein, nonplastidal protein and chlorophyll are strongly correlated. This means, that their relative proportions do not change under the different light regimes used (white, blue and red light). Possible mechanisms being responsible for the inhibition of chloroplast divisions in red light are discussed.The amount of thylacoid protein can not be the limiting factor for reduced photosynthesis in red light.  相似文献

Summary We have examined the pressure-wound healing response inAcetabularia acetabulum (L.) Silva (Chlorophyta). Commonly incurred in routine cell culture, these wounds induce disruption of the vacuole and translocation of the cytoplasm away from the wound site. Daily wounding of individual cells retarded cytoplasmic healing over time, but had no effect on the rate of membrane healing. The position of the wound along the cell stalk also affected the ability of the cell to heal: cells wounded near the rhizoid healed at least 1.9 times more slowly and were only half as likely to achieve reproduction as were cells wounded either near the apex or at mid-stalk. The 50% mortality of cells wounded at the rhizoid suggests the existence of a physical structure near the primary nucleus which is important to cell viability. The impact of wounding on reproductive potential and time to heal differed with the phase of cell development: juvenile and early adult cells healed 2–2.5 times more quickly but were less likely to achieve reproduction than late adult or reproductive cells. Growth at very high population densities (2.5 cells/ml) impaired the ability of the cells to heal. Growth of cells in seawater containing a range of potassium concentrations revealed that healing depends on potassium and is optimal at a concentration of — 1.51ogM potassium.Abbreviations SE standard error of the mean - LD light/dark - NEM N-ethylmaleimide  相似文献

Continuous development, the absence of a germline, flexible and reversible cellular differentiation, and the existence of haploid and diploid generations--both of which express genes--are characteristics that distinguish plants from animals. Because these differences alter the impact of mutations, animals and plants experience varied selection pressures. Despite different life-cycles, both flowering plants and multicellular animals have evolved complex sensing mechanisms that act after fertilization as 'quality checks' on reproduction, and that detect chromosome dosage and the parent of origin for specific genes. Although flowering plant embryos escape such surveillance in vitro, embryo success in the seed often depends on a healthy endosperm--a nutritive tissue that is produced by a second fertilization event in which maternal and paternal gene contributions can be monitored immediately after fertilization and throughout development.  相似文献

Summary At the onset of zoospore cleavage the centrioles ofSaprolegnia ferax reorientate, develop into kinetosomes and become associated with microtubular roots and a striate fibre. After cytoplasmic cleavage a flagellum, with a hitherto undescribed transition zone structure, develops from each kinetosome. Flagellum axonemes occur inside recently encysted primary spores. In vegetative hyphae and germinating cysts most recognizable Golgi bodies are characteristically associated with a cisternum of the endoplasmic reticulum and a mitochondrion but during sporogenesis they all lie adjacent to nuclei where they are apparently active in vesicle production. The structural details of these changes are described and their significance discussed. We wish to acknowledge the numerous helpful discussions with Dr. J. L. Gay. The high photosynthetic activity (O2 production and CO2 consumption) ofAcetabularia mediterranea Lamour. (=A. acetabulum (L.) Silva) characteristic of cells cultured in white light decreases slowly when cells are kept in continuous red light, and is less than 20% of the original activity after three weeks. Subsequent blue irradiation restores the original activity completely within 3–5 d. The polypeptide composition of the thylakoids from cells grown in either red or blue light and after transfer from red to blue light was analyzed mainly with regards to photosystem II (PSII). The P700-containing reaction-centre complex of photosystem I, CPI, showed only minor quantitative alterations as a consequence of the growth-light quality, which correlated well with the activity of photosystem I under these conditions. In PSII, no drastic changes occurred in the quantity of the reaction-centre components D1 (herbicide-binding polypeptide) and D2, as determined by immunoblots. Likewise, the proteins associated with the water-splitting apparatus did not change detectably in thylakoids from red- or blue-light-treated cells (the 16-kDa component could not be found inAcetabularia thylakoids). The level of the major light-harvesting complex was completely unaffected by the light quality. In contrast, the quantities of the chlorophyll a-protein complexes of the core antenna, CP43 and CP47 (and probably CP29), changed, with kinetics similar to those of total photosynthetic activity. We postulate that the function of the PSII antenna became increasingly impaired in the absence of blue light (i.e. in red light), while blue light had a restoring effect. The peripheral antenna, comprising the light-harvesting complexes, is probably functionally connected with the reaction-centre chlorophylls via the core antenna chlorophyll-protein complexes (CP43, CP47 and probably CP29). A deficiency of these complexes would lead to uncoupling of antenna and reaction centre in the majority of PSII complexes after long periods of red-light treatment.  相似文献

Summary The division of the primary nucleus ofAcetabularia mediterranea, andAcetabularia cliftonii was studied by light microscopical observation of living cells. Nuclear and nucleolar volumes are reduced when the caps of the cells have reached their maximum diameter. When the nucleus has reached a size of about 30–50 m in diameter, condensed chromosomes are formed which are separated by an intranuclear spindle.  相似文献
