Summary The effects of metopirone (SU-4885) and aldactone (SC-9420) on the histology and histochemistry of the interrenal tissue of male common frogs, Rana temporaria, have been studied. From the changes in nuclear and cellular size, mitotic rate, and lipid and cholesterol content it has been concluded that in intact animals the administration of metopirone as well as of aldactone resulted in an increase of the interrenal secretory activity. The effects were very similar to that of ACTH. Ablation of the pars distalis prevented the interrenal activation by metopirone. The effect of aldactone was markedly reduced in operated frogs, but some stimulation remained. It is suggested that both substances act by way of enhancing pituitary ACTH ouput. The dependence of the secretion of corticosteroids on pituitary ACTH is discussed.I wish to thank Dr. W. The purpose of this investigation was to measure quantitatively nuclear volumes in outer fasciculata cells in adrenal glands of rats subjected to chronic stimulation of steroidogenesis by ACTH, or to chronic inhibition of functional activity by hypophysectomy or by injection of the steroidogenic inhibitos, U-8113 (p-aminophenyl butanone) and SU-4885 (metopirone). Nuclear volumes, after a recovery period from those effects, were also measured. Chronic daily administrations of ACTH for 7, 14, and 30 days led to a progressive, statistically significant increase in nuclear volumes of fasciculata cells. These changes were found to have been reversed 14 days after discontinuation of hormone treatment. Hypophysectomy resulted in a significant decrease in nuclear volumes. The steroidogenic inhibitors SU-4885 and U-8113 had a biphasic effect on the nuclear volumes with an early decrease and subsequent increase to normal values. Since a decrease in nuclear volumes in the adrenal glands of inhibitor-treated subjects occurred in the presence of cellular hypertrophy and a significant increase in adrenal weights, it is concluded that changes in nuclear volumes can be positively correlated with the secretory activity of the cell rather than with the sizes of cells or glands.  相似文献

The interrenal and pituitary cells showed increase in nuclear diameter eight days after treatment with ACTH, salicylate or salicylate + ACTH. After ACTH treatment, however, the cytoplasm in the interrenal cells was granular. Similarly, the lead-haematoxylin positive (PbH + ve) cells in the rostral pars distalis also showed partial depletion of stainable granules after ACTH treatment. Salicylate or salicylate ACTH caused depletion of cytoplasmic material in both the interrenal as well as PbH + ve cells.  相似文献

The effects of chronic administration of ACTH and dexamethasone on the morphology of mitochondria in zona glomerulosa cells of the rat adrenal cortex were investigated by stereological techniques. It was found that the volume of the mitochondrial compartment as well as the surface of the outer and inner mitochondrial membranes were significantly increased or decreased in relation to the number of days of ACTH- or dexamethasone-treatment. In ACTH-administered rats, the average volume of individual mitochondria decreased significantly up to the 6th day of treatment and then showed a conspicuous increase from the 6th to the 15th day, whereas in dexamethasone administered animals this parameter, after a small increase during the first 6 days of treatment, displayed a significant decrease. The number of mitochondria per cell showed a dramatic increase during the first 6 days of treatment with ACTH and continued to increase, but only slightly, with the subsequent treatment. In contrast, this parameter showed a parabolic decrease as a function of the duration of treatment in animals receiving dexamethasone. In the light of evidence showing that dexamethasone blocks ACTH-release, these findings are discussed and interpreted to indicate that ACTH is involved in the maintenance and stimulation of the growth and proliferative activity of mitochondria in rat adrenal zona glomerulosa.  相似文献

The mammalian ACTH (20 I.U.) injected in Heteropneustes fossilis (Bloch) caused hyperplasia of the adrenocortical tissue. The interrenal as well as chrome cells became vacuolated. The interrenal nuclear size increased by 15% as an average than the control fishes. The glycogen granules were found to be absent from the interrenal cells. The effect of bilateral gonadectomy in both sexes has been marked in the gradual decrease of the interrenal nuclear diameter which 12 weeks after the operation, recorded nearly 25% reduction in size. The glycogen content was reduced considerably. Removal of gonads during maturation period bringing inhibition of hyperplasia of the gland at the time of spawning possibly indicates action of gonadal steroids on the physiological alterations of the interrenal cells.  相似文献

The effects of experimentally elevated plasma thyroxine levels on the subsequent response of interrenals of coho salmon (Oncorhynchus kisutch) to adrenocorticotropic hormone (ACTH) in vitro were examined. Animals were treated with thyroxine by immersion (200 micrograms/L) for 3 days, which resulted in physiological elevations in circulating thyroxine. In animals treated before the parr-smolt transformation was completed (early smolts), thyroxine had no effect on plasma cortisol levels but significantly enhanced the sensitivity of the interrenal to ACTH in vitro. In animals treated after the period of smoltification (postsmolts), plasma cortisol levels were significantly higher than those of controls; both experimental and control animals had plasma cortisol levels higher than normally observed at this stage of development. The response of the interrenals of thyroxine-treated postsmolts to ACTH in vitro was significantly lower than that of controls. Results from the experiments using early smolts are in agreement with studies in other vertebrates showing that thyroid hormones are involved in maintaining the normal functioning of corticosteroidogenic tissue and suggest that thyroid hormones may be involved in the activation of the interrenal that occurs during smoltification. The results obtained using postsmolts are more difficult to interpret because of the possibility that these animals were physiologically stressed by the treatment. Increased ACTH release in vivo resulting from stress may have led to a depression of interrenal sensitivity to ACTH in vitro and may have masked a refractoriness of the pituitary-interrenal axis to thyroxine.  相似文献

The effects of gonadectomy on the cytological structure of the hypophysis and interrenal tissue of Clarias batrachus have been investigated. Forty days after gonadectomy remarkable hypertrophy of certain cyanophils, identified later as gonadotrops, has been observed followed by hyperplasia. The somatotrops also seem to undergo similar changes, and as a result the proximal pars distalis (PPD) increases enormously and encroaches upon the pars intermedia (PI). The gonadotrops undergo a gradual degranulation and become devoid of stainable cytoplasm by 90 days. No signet ring cells have been noticed at any stage. The cyanophils in the ventral region of the PPD, identified as thyrotrops, also show degranulation, but to a comparatively limited degree. The ACTH cells undergo gradual hypertrophy and degranulation. A progressive increase is noticed in the nuclear diameter. The interrenal cells are activated as indicated by the enhanced nuclear indices. It is suggested that a fall in gonadal steroids may result in ACTH release which in turn activates the interrenal cells.  相似文献

Exogenous ACTH1-24 promotes adrenal maturation in fetal sheep, and this effect appears to be modulated in part by cortisol (Challis et al. 1985). We have examined whether similar changes in adrenal metabolism of progesterone occur with ACTH-induced labour as at spontaneous term and whether the site of cortisol modulation is on adrenal steroidogenesis or at the level of cAMP generation. Chronically catheterized fetal sheep were infused in utero for 100 h between days 127 and 131 of pregnancy with P-ACTH, P-ACTH + metopirone, P-ACTH + metopirone + cortisol, or saline. After 100 h the metabolism of [3H]progesterone was measured in adrenal homogenates. Similar incubations were performed with adrenal tissue from fetal sheep at day 130 of pregnancy and at spontaneous labour. In the treatment groups of sheep, cAMP output by dispersed adrenal cells in response to ACTH added in vitro was also determined. Similar qualitative patterns of [3H]progesterone metabolism were found in adrenal homogenates after in vivo ACTH or at term. At both times there was an increase in cortisol and in total 17 alpha-hydroxycorticosteroid accumulation and also evidence for increased activity of 11 beta-hydroxylase enzyme. The formation of total 17 alpha-hydroxycorticosteroids was not affected significantly by concurrent treatment in vivo with metopirone +/- cortisol. The accumulation of cAMP in vitro was increased after in vivo ACTH, attenuated after ACTH + metopirone, but statistical significance over controls was restored after ACTH + metopirone + cortisol treatment. We conclude that ACTH-induced labour and spontaneous parturition in sheep is associated with qualitatively similar changes in progesterone metabolism by the fetal adrenal gland.(ABSTRACT TRUNCATED AT 250 WORDS)  相似文献

Gravimetric and histologic modifications in the pigeon were studied following chronic therapy with ACTH and metopirone (SU 4885) for a period of 15 days. The organs studied were proventriculus, duodenum, heart, kidney, salivary gland, pancreas, liver, uropygial gland, thymus, spleen, bursa fabricii, testis, ovary, islets of Langerhans, adenohypophysis, thyroid and interrenal and chromaffin tissue of the adrenal gland. Induced states of hyper- and hypoadrenocorticalism elicited pathomorphic changes in endocrine and reproductive systems and some other organs of the pigeon. There were many differences and similarities in the nature of response of some organs following the two experimental conditions. Many of these cellular interactions might have resulted from alteration of interrenal function in the pigeon.  相似文献

1. The ACTH cell type has been demonstrated for the first time in the hypophysis of the squirrels. These are large cells with homogenous cytoplasm and are more abundant in the anteromedian zone and in the peripheral portion than elsewhere. 2. A great hypertrophy and hyperplasia of the pituitary corticotrophs were observed in the experimental group at all the intervals. 3. Metopirone treated animals showed a moderate hypertrophy and degranulation of pituitary thyrotrophs. 4. By effect of metopirone, a marked regression of pituitary interstitiotrophs and total degranulation took place by 30 days treatment. The folliculotrophs did not show alterations.  相似文献

Studies have been made of the effect of injections of hypothalamic nonapeptide neurohormone, arginine vasotocin, on functional condition of the interrenal gland in mature frogs. In unoperated, sham-operated and in animals 10 days after hypophysectomy, single and especially three subsequent injections of arginine vasotocin (5 x 10(-9) M per 1 kg of the body weight) result in evident activation of glandular cells of the interrenal gland which is manifested in the increase of the volume of their nuclei and cytoplasmic area, as well as in the dilatation of the blood vessels. Activation of the interrenal gland in hypophysectomized frogs, which lack endogenous ACTH, indicate the direct para-adenohypophyseal influences of nonapeptide hypothalamic hormones on the activity of glandular cells in the peripheral endocrine glands, in particular, the interrenal gland of the grass frog.  相似文献

Ultrastructural morphometry (point-counting method) was used to evaluate stereological parameters of rough endoplasmic reticulum (RER), Golgi complex, secretory granules, mitochondria and lysosomes of Rana ridibunda pituitary corticotropic (ACTH) cells in animals subjected to constant environmental conditions (CEC) in relation to temperature and photoperiod. During this experiment, corticotropic cells are particularly sensitive to the effects of changes in environmental conditions in such a way that they course through a phase of RER and Golgi complex hypertrophy after 7 days on CEC and a phase of enhancement in the amount of secretory granules (the 11th day). After 15 days on CEC, the cells seemingly recuperate almost all the control evaluate parameters. These results strongly suggest that, in frog kept in captivity, ACTH cells play an important role in the acclimation process to new constant environmental conditions.  相似文献

Relationships between prolactin and adrenal secretion were studied in the adult male rat by different experimental approaches. Administration of a long acting 1-24 ACTH preparation during 11 days induced a significant decrease in plasma prolactin levels. Adrenalectomy on the contrary resulted in an increase of prolactin levels that were not affected by ACTH treatment. Dexamethasone administration to intact or adrenalectomized animals resulted in a significant reduction of plasma prolactin in both cases. In order to elucidate if the inhibitory effect of the adrenal stimulation on prolactin was mediated through the blockade of endogenous ACTH, stimulation of hypothalamic-pituitary-adrenal axis with chronic intermittent immobilization stress was performed. Stress induced a significant elevation in plasma corticosterone levels, together with a decrease in prolactin values. These data indicated that the inhibitory role of ACTH and stress on prolactin secretion was mediated through the adrenal glucocorticoid stimulation.  相似文献

Summary In nine cell types of the adenohypophysis in untreated adult rainbow trout, histologically different activity phases, seasonal changes in activity, and the relation between certain cell types and the interrenal gland, thyroid or gonads were investigated by light and, occasionally, by electron microscopy. Special attention was given to the effect of social rank on the synthetic activity in adenohypophysial cells of trout kept in small groups in which a social hierarchy with one (light) dominant and several (dark) submissives is established.Cell types in the rostral pars distalis were azocarminophil (I) or amphiphil (II). Proximal pars distalis cell types were slightly basophil (IV), orangeophil (V), strongly basophil (VI) or chromophobe (VII). In the pars intermedia, cell types were amphiphil (VIII) or very slightly basophil (IX). Type III was a non-secretory supporting (?) cell.Histologically different activity phases abounded in type IV cells, which mainly occurred in the proximal pars distalis but were also found dispersed in the rostral pars distalis, the pars intermedia and the neurohypophysis.Influences of social rank were pronounced in type IV cells. Phases with a high synthetic activity were exclusively found in submissive animals, phases with a low synthetic activity occurred in dominants. As a positive relation existed between type IV cell activity and the social rank dependent activity of the interrenal gland, it was suggested that type IV cells produce ACTH.In (dominant) male trout treated with DOCG or ACTH, colloid-containing type IV cell phases, reflecting accumulation of the secretory product, were found. This supported the earlier suggestion that ACTH in the trout is produced in the basophil type IV cells and not, as reported in the literature, in cells comparable to type II.The author is greatly indebted to Miss M.C. Oncofetal aspects of ACTH and pro-opiomelanocortin (POMC)-derived peptides were studied immunohistochemically at the light and electron microscopic level in human fetal pituitary glands, pituitary adenomas, and small-cell carcinoma of the lung. ACTH, beta-endorphin, and gamma-MSH were localized in the same cells of both fetal and adult pituitary, as well as in the above-mentioned neoplastic tissues. However, alpha-MSH was observed only in the early fetal pituitary, its concentration decreasing with advancing gestational age. The adult pituitary contained only a few alpha-MSH-positive cells. By immunoelectron microscopy, ACTH in the adult pituitary was localized exclusively in the secretory granules. In fetal pituitary at 9 weeks' gestation, ACTH was localized in the perinuclear spaces (PNS), cisternae of rough endoplasmic reticulum (RER), Golgi saccules, and secretory granules. The staining pattern of ACTH in these organelles varied from cell to cell. In fetal pituitaries of greater gestational ages, ACTH was localized in secretory granules. The pituitary adenomas mimicked the staining characteristics of the adult pituitary, i.e., negative or only very occasional alpha-MSH staining and localization of ACTH in the secretory granules. The ectopic ACTH-producing tumors showed a staining pattern similar to that of the early fetal pituitary, i.e., positive staining for alpha-MSH and the presence of ACTH in PNS and cisternae of RER.  相似文献

Dispersion of cisternae of rough endoplasmic reticulum (RER) in aging rats has been determined quantitatively for mitral cells of the olfactory bulb and Purkinje cells of the cerebellum using a recently published morphometric technique (Cruz Orive, '76). In both cell types dispersion of cisternae occurred throughout the period studied and in a linear fashion. The observed RER dispersion cannot be attributed to a decrease in the total amount of RER, since the total amount of RER was constant in Purkinje cells and increased in mitral cells during the age studied.  相似文献

Influence of Hydrocortisone on Chick Embryo Retina Development   总被引:1,自引:1,他引:0  
Treatment of chick embryos in ovo with hydrocortisone-21-phosphate (a single dose of 150 micrograms) caused a marked reduction of retinal thymidine kinase activity 24 h later. The inhibitory effect was highest (65-70%) in 8-10-day-old embryos and declined with age, disappearing after day 15. It was accompanied by a reduction in thickness of the retinal layers. Adrenocorticotropic hormone (ACTH) treatment (10 micrograms daily for 2 days) also produced an age-dependent inhibitory effect on retinal thymidine kinase, whereas treatment with a single dose of 200 micrograms of metopirone, a compound that prevents the 11 beta-hydroxylation of steroid molecules in the adrenal glands, impeded the decrease in thymidine kinase activity that normally occurs in chick embryo retina after day 9 of development. In addition, metopirone prevented the inhibition exerted by ACTH on thymidine kinase activity but had no effect on the action of hydrocortisone.  相似文献

Summary Adrenals, pituitary glands, thyroids, testes and kidneys of metopirone-treated and control male lizards were studied by light microscopy. Pituitary-interrenal axis: A great hypertrophy and hyperplasia of the interrenal cells and the pituitary corticotrophs were found in the experimental group. Pituitary-thyroid axis: Metopirone-treated animals showed a moderate hypertrophy and degranulation of pituitary thyrotrophs, and signs of uneven hyperactivity of the thyroid. Pituitary-testicular axis: By effect of metopirone, a marked regression of pituitary interstitiotrophs and a virtual disappearance of the testicular interstitial gland took place. The second portion of the kidney sexual segments appeared degranulated as in castrated animals. The pituitary folliculotrophs and the seminiferous epithelium of the testis did not show alterations.  相似文献

Summary The ultrastructure of the interrenal (adrenocortical) cells of trout (Salmo fario L.) was studied after dexamethasone treatment. A procedure for identifying and isolating interrenal tissue fragments from the surrounding head kidney tissue prior to their preparation for electron microscopy is described. The peripheral plasma cortisol concentrations were measured in order to evaluate the steroidogenic activity of this tissue.The interrenal cells of control animals contain numerous mitochondria with tubular cristae, and a well developed and highly organized smooth endoplasmic reticulum (SER). The scarcity, or absence, of lipid droplets contrasts markedly with the abundance of SER.Treatment with dexamethasone results in a decrease steroidogenic activity of the interrenal cells, as indicated by the fall in plasma cortisol concentrations. The interrenal cells are small, but still contain numerous mitochondria. The SER is poorly developed, but masses of densely intermeshed smooth cisternae subsist. Lipid droplets do not accumulate in these cells; this peculiarity is discussed in connection with the virtual absence of liposomes in teleost interrenal cells.  相似文献
