Summary We report a girl with a de novo duplication of the distal part of the long arm of chromosome 3 and review the literature. Our patient had the facial characteristics and many other anomalies of the partial 3q duplication syndrome. As a hitherto undescribed symptom in partial 3q trisomy syndrome, she had microphthalmia. The karyotype of this girl was interpreted as an inverse duplication of the terminal portion of chromosome 3: 46,XX,inv dup (3)(pter-q28::q28–q25::q28-qter). Quantitative hybridisation studies with 3p and 3q probes gave a consistent 32 ratio of the relative intensities of the q bands in relation to the p bands between patient and control. This confirmed the presence of a 3q duplication and delineated the location of D3S5 to 3q25–3q28.  相似文献

Summary A male patient with an interstitial deletion 13q14q31 is described. Our necropsy findings included a left retinoblastoma and several gross internal malformations. In this paper we reaffirm that band 13q14 is involved in cases of retinoblastoma and we propose, after studying accompanying cases of total or partial long arm trisomies 13, that the loss of specific 13q bands, from 13q14 to 13q31 is responsible for the congenital defects we are describing.  相似文献

We present prenatal diagnosis and array comparative genomic hybridization characterization of 3q26.31–q29 duplication and 9q34.3 microdeletion in a fetus with omphalocele, ventricular septal defect, increased nuchal translucency, abnormal first-trimester maternal screening and facial dysmorphism with distinct features of the 3q duplication syndrome and Kleefstra syndrome. The 26.61-Mb duplication of 3q26.31–q29 encompasses EPHB3, CLDN1 and CLDN16, and the 972-kb deletion of 9q34.3 encompasses EHMT1. We review the literature of partial trisomy 3q associated with omphalocele and discuss the genotype–phenotype correlation in this case.  相似文献

Summary An inv ins(7;2)(q21.2;q3105q24.2) was found to segregate through four generations of a family. Adjacent-1 segregation aneusomies were ascertained in five patients: three monosomics and two trisomics; and the corresponding syndromes were delineated. The comparative analysis between these and other previously described 2q aneusomic individuals led to the conclusion that a large cleft between first and second toes is a constant feature in monosomy 2q24q31. No other trait could plausible be mapped. Risks of 7.9 to 31.9% for aneusomic children and of 26.3% for abortion were estimated in the present family.  相似文献

The quantum chemical and molecular dynamics studies have been performed to infer the structural changes of all-trans and all-cis forms of cyclo[(1R,3S)-3-aminocyclohexanecarboxylicacid(γ-Acc)-α-Glycine(Gly)]3 hexapeptide. The backbone conformations of the above peptide have been analyzed using the valence and peptide deformation angles applying B3LYP/6–311G** level of theory. The conformational preference of the backbone of all-trans and all-cis cyclo[(1R,3S)-γ-Acc-Gly]3 hexapeptides is found to depend on the puckering of cyclohexane rings. The non-uniform distribution of water inside the cavity is observed, where sometimes water molecules formed a chain like conformation through hydrogen bond networks while traversing the pore of all-cis cyclo[(1R,3S)-γ-Acc-Gly]3 peptide. Larger relaxation times of the order of a hundred to two hundred pico seconds for active site…water hydrogen bond interactions were noticed. The hydrophobic nature of the cavity of all-trans cyclo[(1R,3S)-γ-Acc-Gly]3 due to the presence of cyclohexane moiety has been analyzed. Further this investigation emphasized on the non-transport of molecules through the pore of all-trans cyclo[(1R,3S)-γ-Acc-Gly]3 peptide due to the obstruction produced by cyclohexane groups.  相似文献

A particulate enzyme preparation from Phaseolus aureus (mung bean) seedlings catalyzed the synthesis of a water insoluble β-1,3-glucan from UDP-α-d-glucose (UDPG) at high concentrations (0.4~20 mm) and an alkaline insoluble β-1,3 and β-1,4-mixed glucan from UDPG at a low concentration (8.5 µm).

Furthermore, the two kinds of β-glucan synthetases which were investigated with two reaction systems at high and low concentrations of UDPG had different properties in optimal pH, stability of enzyme activity, and metallic ion requirement.  相似文献

Different subtypes of opioid receptors (OR) were activated in rats in vivo to study the activation effect on the heart’s resistance to ischemia and reperfusion. It has been established that administration of deltorphin II, a selective δ2-OR agonist, lowered the infarct size/area at risk index (IS/AAR) by 23%. Naltrexone, naloxone methiodide (an OR inhibitor not penetrating the blood-brain barrier (BBB)), and naltriben (δ2-antagonist) eliminated the cardioprotective effect of deltorphin II, while BNTX (a δ1-antagonist) produced no effect on the cardioprotective action of the δ2-agonist. The infarct-reducing effect of deltorphin II was eliminated by administration of chelerythrine (a protein kinase C (PKC) inhibitor), glibenclamide (a KATP-channels inhibitor), and 5-hydroxydecanoate (a mitochondrial KATP-channel blocker). Administration of other opioids did not reduce the IS/AAR index. It has been established that all the deltorphins manifest antiarrhythmic potency. Other opioids do not produce any effect on the incidence of arrhythmia occurrences. The antiarrhythmic effect of deltorphin II was eliminated by preliminary administration of naltrexone, naloxone methiodide, and naltriben, but BNTX did not affect the δ2-agonist’s anti-arrhythmic effect. The preliminary administration of chelerythrine, a PKC inhibitor, eliminated the δ2 agonist’s antiarrhythmic action. However, glibenclamide and 5-hydroxydecanoate did not alter the antiarrhythmic effect by deltorphin II. Therefore, activation of the peripheral δ2-ORs reduces the infarct size and prevents the onset of arrhythmias. The antiarrhythmic effect of the δ2-OR stimulation is mediated by activating PKC and opening the mitochondrial KATP-channels. PKC participates in the antiarrhythmic effect of the δ2-OR activation, but this effect does not depend on the condition of KATP-channels.  相似文献

Summary Deletion 3q273qter in an infant is described. A chromosomal abnormality was suspected because of minor facial dysmorphism and closed parietal meningocele. On the first day of life, a large exudative inflammation appeared on the skin of her back, which completely resolved after 1 week. Biopsy showed dilated sweat gland openings resembling miliaria rubra, which has not been previously reported in this age group. It is unclear if the skin change was due to the chromosomal abnormality. The meningocele was repaired at age 8 months. At age 20 months, slight neurodevelopmental delay was evident, the main features being hypertonicity and inability to walk without support. The patient has two healthy sisters, and prometaphase chromosome studies in both parents were normal. This infant represents the first example of del3q273qter and the first reported association of meningocele with an abnormality of chromosome 3.  相似文献

Three novel oligosaccharides of human infant faeces have been fully characterised by methylation analysis and 500/600 MHz 1H NMR spectroscopy including DQF-COSY, TQF-COSY, TOCSY and ROESY experiments. The oligosaccharides were shown to be lactose-based structures two of which were substituted at C-6 of Gal with either the Lex trisaccharide, Gal(β1–4)[Fuc(α1–3)]GlcNAc(β1-, or Neu5Ac(α2–6)Gal(β1–4)GlcNAc-(β1-. They differ from other free oligosaccharides previously isolated from the human by having the (1 → 6) linkage to Gal in the absence of a (1 → 3) branch. The third oligosaccharide has Neu5Ac(α2–6) linked to GlcNAc of the trisaccharide GlcNAc(β1–3)Gal(β1–4)Glc. This is a linear fragment of the disialylated tetrasaccharide sequence Neu5Ac(α2–3)Gal(β1–3)[Neu5Ac(α2–6)]GlcNAc(β1- found in the milk oligosaccharide disialyl LNT (the GlcNAc residue of the tetrasaccharide linked to lactose) and also of N-linked chains (GlcNAc linked to Man).  相似文献

Summary Gene marker analyses have been carried out in a patient with 10q(q23qter) duplication. The observed elevation of red cell glutamic oxaloacetic transaminase activity is compatible with earlier somatic cell hybridization studies that mapped the locus to this region. Hexokinase-1 activity in the red cells was normal, which is consistent with its prior assignment to the unaffected part of chromosome 10 (10pterq23).  相似文献

The three-step synthesis of new mixed P/N/N′/O-donor ligands C6H3(OH){2-NHC(O)CH2NCHC6H4PPh2}(4-CH3) 3a·HH and C6H4(OH){3-NHC(O)CH2NCHC6H4PPh2} 3b·HH, by Schiff base condensation of the 1° amines C6H3(OH){2-NHC(O)CH2NH2}(4-CH3) 2a or C6H4(OH){3-NHC(O)CH2NH2} 2b with C6H4(CHO)(2-PPh2) in refluxing EtOH, is described. Reaction of 1 equiv. of 3a·HH or 3b·HH with MCl2(cod) (M = Pt, Pd; cod = cycloocta-1,5-diene) affords the κ2-PN-chelate complexes MCl2(3a·HH) (M = Pd 4a; M = Pt 4b) and MCl2(3b·HH) (M = Pt 4c). The dichlorometal(II) complexes 4d and 4e, bearing instead a pendant 4-phenolic group, were similarly prepared (in >90% yield). Chloro-bridge cleavage of [Pd(μ-Cl)(η3-C3H5)]2 with 3a·HH or 3b·HH gave the monocationic κ2-PN-chelate complexes [Pd(η3-C3H5)(3a·HH)]Cl 5a or [Pd(η3-C3H5)(3b·HH)]Cl 5b, respectively. Elimination of cod, and single CH3 protonation, from Pt(CH3)2(cod) upon reaction with 1 equiv. of 3a·HH or 3b·HH in C7H8 at room temperature afforded the neutral complexes C6H3(OH){2-NC(O)CH2NCHC6H4PPh2Pt(CH3)}(4-CH3) 6a and C6H4(OH){3-NC(O)CH2NCHC6H4PPh2Pt(CH3)} 6b, respectively bearing a monoanionic (3a·H or 3b·H) κ3-PNN′-tridentate ligand. Amide and phenol deprotonation were readily achieved, using KOtBu as base, to give high yields of the κ4-PNN′O-tetradentate complexes C6H3(O){2-NC(O)CH2NCHC6H4PPh2Pd}(4-CH3) 7a and C6H3(O){2-NC(O)CH2NCHC6H4PPh2Pt}(4-CH3) 7b bearing the dianionic ligand 3a2−. All new compounds have been characterised by multinuclear NMR, FTIR, mass spectroscopy and microanalysis. Single crystal X-ray studies have been performed on compounds 1b·1.5CH2Cl2, 3b·HH·0.5Et2O, 6b·CHCl3 and 7b·0.5Et2O.  相似文献

Abstract

A simple procedure is described for the preparation of the title compounds 1, 8 and 9. 3′-3′ or 3′-5′ or 5′-5′ TpT was reacted with a twofold molar excess of TPS in anhydrous DMF, at room temperature, for 5 min, followed by a 1 min in situ treatment of the reaction mixture with excess 7.0 N NH4OH, at 0°C. The alkaline hydrolysis of 1, 8 and 9 proceeds without the assistance of 3′- and 5′-hydroxyl groups resulting in equimolar mixtures of thymidine (4) and thymidine 3′-phosphoramidate (6) (for the 3′-3′ isomer) or thymidine 5′-phosphoramidate (7) (for the 5′-5′ isomer) or 6 and 7 in equal quantities (for the 3′-5′ isomer).  相似文献

Polyunsaturated fatty acids, like arachidonic acid, can bind proteins and affect their function. The 14-3-3 proteins bind phosphorylated sites on a diverse array of client proteins and, in this way, are involved in many intracellular signaling pathways. In this study, we used a novel approach to discover that 14-3-3ζ is able to directly bind arachidonic acid. Furthermore, arachidonic acid, at physiological concentrations, reduced the binding of 14-3-3ζ to phosphorylated BAD, an interaction that is important in regulating apoptosis. In addition, high concentrations of arachidonic acid caused the polymerization of 14-3-3ζ, an event observed in neurodegenerative disorders. Taken together, these results indicate that arachidonic acid directly interacts with 14-3-3ζ and that this interaction may be important in both normal and pathological cellular events. If so, then factors that mediate the release, metabolism and reacylation of arachidonic acid into membranes represent key points of regulation.  相似文献

为了观察热量限制对主动脉内皮细胞中HNF3γ及NOX4基因表达的影响, 揭示HNF3γ-NOX4-活性氧通路介导热量限制抗内皮细胞衰老的分子机制, 文章将主动脉内皮细胞分为5组:对照组、高热量组、低热量组、siRNA+低热量组、siRNA+高热量组。应用逆转录实时定量PCR(Real-time quantitative PCR, RT-qPCR)、Western blotting分析各组HNF3γ、NOX4 mRNA及蛋白水平变化, 并检测各组细胞内活性氧产量及细胞衰老程度变化。采用染色质免疫共沉淀分析HNF3γ蛋白与NOX4基因启动子区域结合情况, 萤光素酶报告基因检测HNF3γ蛋白结合后对NOX4基因启动子活性的影响。结果显示:与对照组比较, 低热量组HNF3γ mRNA和总HNF3γ蛋白表达水平、磷酸化/总HNF3γ比值显著升高(P<0.05), NOX4 mRNA和蛋白表达水平、细胞内活性氧产量及细胞衰老程度显著降低(P<0.05); 高热量组HNF3γ mRNA和总HNF3γ蛋白表达水平、磷酸化/总HNF3γ比值显著降低(P<0.05), NOX4 mRNA和蛋白表达水平、细胞内活性氧产量及细胞衰老程度显著升高(P<0.05); siRNA+低热量组及siRNA+高热量组中NOX4 mRNA和蛋白表达水平、细胞内活性氧水平及细胞衰老程度显著升高(P< 0.05)。染色质免疫共沉淀证实HNF3γ蛋白可与NOX4基因启动区域4个结合位点(-6 bp、-76 bp、-249 bp、-954 bp)结合。萤光素酶报告基因检测显示HNF3γ蛋白与NOX4启动子区域1个位点(-6 bp)、2个位点(-6、-76 bp)、3个位点(-6、-76、-249 bp)、4个位点(-6、-76、-249、-954 bp)结合, 可使NOX4启动子活性分别降低至对照组的80.15±4.64%、40.02.±2.15%、16.46±2.24%、12.13±1.46%, P<0.05。上述结果提示热量限制可上调HNF3γ基因表达, 增强HNF3γ蛋白活性, 促进HNF3γ蛋白同NOX4基因启动子区域结合, 抑制NOX4基因表达, 进而减少细胞内活性氧产生而延缓动脉内皮细胞衰老。  相似文献

本文报道一例t(3; 22) (p21; q13)平衡 相互易位的家系。先证者,男性,一岁半,淋巴 细胞及皮肤成纤维细胞G带分析结果:核型均 为46, XY, t(3; 22)(p21;q13)或46, XY,t(3; 22)(3gter” 3p21::22813” 22gter;2 2 pter” 22gl3::3p21” 3 pter );先证者母亲(图1)与 外祖母核型均为46, XX, t(3; 22)(p21; q13) 或46, XX, t(3; 22)(3gter、3p21::22813一 22gter; 22pter~ 22813::3p21一3pter)。经银 染与G带复合显示技术,先证者及母亲的22der 可见清晰的AgNOR区。先证者的父亲与舅父 G带分析核型正常。在此情况下,有生育正常 婴儿的可能,但必须作产前诊断。  相似文献

Abstract

The 5′-O-(4,4′-dimethoxytrityl) and 5′-O-(tert-butyldimethylsilyl) derivatives of 2′-,3′-O-thiocarbonyl-6-azauridine and 2′,3′-O-thiocarbonyl-5-chlorouridine were synthesized from the parent nucleosides by reaction with 4, 4′-dimethoxytrityl chloride and tert-butyldimethylsilyl chloride, respectively, followed by treatment with 1,1′-thiocarbonyldiimidazole. Introduction of a 2′-,3′-double bond into the sugar ring by reaction of the 5′-protected 2′-,3′-O-thionocarbonates with 1, 3-dimethyl-2-phenyl-1, 3, 2-diazaphospholidiine was unsuccessful, but could be accomplished satisfactorily with trimethyl phosphite. Reactions were generally more successful with the 5′-silylated than with the 5′-tritylated nucleosides. Formation of 2′-,3′-O-thiocarbonyl derivatives proceeded in higher yield with 5′-protected 6-azauridines than with the corresponding 5-chlorouridines because of the propensity of the latter to form 2,2′-anhydro derivatives. In the reaction of 5′-O-(tert-butyldimethylsilyl)-2′-,3′-O-thiocarbonyl-6-azauridine with trimethyl phosphite, introduction of the double bond was accompanied by N3-methylation. However this side reaction was not a problem with 5′-O-(tert-butyldimethylsilyl)-2′-, 3′-O-thioarbonyl-5-chlorouridine. Treatment of 5′-O-(tert-butyldimethylsilyl)-2′-, 3′-didehydro-2′-,3′-dideoxy-6-azauridine with tetrabutylammonium fluoride followed by hydrogenation afforded 2′-,3′-dideoxy-6-azauridine. Deprotection of 5′-O-(tert-butyldimethylsilyl)-2′-, 3′-didehydro-2′-,3′-dideoxy-5-chlorouridine yielded 2′-,3′-didehydro-2′-,3′-dide-oxy-5-chlorouridine.  相似文献
