The chlorophyll (Chl) fluorescence imaging technique was applied to cashew seedlings inoculated with the fungus Lasiodiplodia theobromae to assess any disturbances in the photosynthetic apparatus of the plants before the onset of visual symptoms. Two-month-old cashew plants were inoculated with mycelium of L. theobromae isolate Lt19 or Lt32. Dark-adapted and light-acclimated whole plants or previously labelled, single, mature leaf from each plant were evaluated weekly for Chl fluorescence parameters. From 21 to 28 days, inoculation with both isolates resulted in the significantly lower maximal photochemical quantum yield of PSII (Fv/Fm) than those for control samples, decreasing from values of 0.78 to 0.62. In contrast, the time response of the measured fluorescence transient curve from dark-acclimated plants increased in both whole plants and single mature leaves in inoculated plants compared with controls. The Fv/Fm images clearly exhibited photosynthetic perturbations 14 days after inoculation before any visual symptoms appeared. Additionally, decays in the effective quantum yield of PSII photochemistry and photochemical quenching coefficient were also observed over time. However, nonphotochemical quenching increased during the evaluation period. We conclude that Fv/Fm images are the effective way of detecting early metabolic perturbations in the photosynthetic apparatus of cashew seedlings caused by gummosis in both whole plants and single leaves and could be potentially employed in larger-scale screening systems.  相似文献

Hemiepiphytic Ficus species exhibit more conservative water use strategy and are more drought-tolerant compared with their non-hemiepiphytic congeners, but a difference in the response of photosystem I (PSI) and photosystem II (PSII) to drought stress has not been documented to date. The enhancement of non-photochemical quenching (NPQ) and cyclic electron flow (CEF) have been identified as important mechanisms that protect the photosystems under drought conditions. Using the hemiepiphytic Ficus tinctoria and the non-hemiepiphytic Ficus racemosa, we studied the water status and the electron fluxes through PSI and PSII under seasonal water stress. Our results clearly indicated that the decline in the leaf predawn water potential (ψpd), the maximum photosynthetic rate (Amax) and the predawn maximum quantum yield of PSII (Fv/Fm) were more pronounced in F. racemosa than in F. tinctoria at peak drought. The Fv/Fm of F. racemosa was reduced to 0.69, indicating net photoinhibition of PSII. Concomitantly, the maximal photo-oxidizable P700 (Pm) decreased significantly in F. racemosa but remained stable in F. tinctoria. The fraction of non-photochemical quenching [Y(NPQ)] and the ratio of effective quantum yield of PSI to PSII [Y(I)/Y(II)] increased for both Ficus species at peak drought, with a stronger increase in F. racemosa. These results indicated that the enhancement of NPQ and the activation of CEF contributed to the photoprotection of PSI and PSII for both Ficus species under seasonal drought, particularly for F. racemosa.  相似文献

The photosynthetic performances of individual leaves of a wheat (Triticum aestivum cv Bezostaya) crop were assessed daily and throughout individual days during the winter when temperature and light levels were fluctuating. Measurements of chlorophyll fluorescence induction and the maximum quantum yield of O2 evolution were made on individual leaves. Depressions in the ratio of variable to maximal fluorescence (Fv/Fm) were correlated with low temperatures and high light levels throughout the winter and during the course of individual days. Depressions in Fv/Fm observed in the field during the day were not accompanied by any significant change in the ability of photosystem II complexes to bind 3-(3,4-dichlorophenyl)-1-dimethyl urea, indicating that the depressions in Fv/Fm were not attributable to photodamage to the D1 protein of the photosystem II reaction center. Decreases in Fv/Fm were associated with increases in the rate of dissipation of excitation energy by radiationless decay processes and decreases in the quantum efficiency of CO2 assimilation, indicative of a rapidly reversible light-induced “downregulation” of photosynthesis. No major changes were observed in the maximum quantum efficiency of O2 evolution of leaves throughout periods of fluctuating temperature and light, because light-induced depressions in photosynthetic efficiency recovered within the time required to make these measurements.  相似文献

The partial shading effect on the photosynthetic apparatus of the sunflower (Helianthus annuus L.) was examined by monitoring oxygen evolution, maximum quantum yield of PSII photochemistry in dark-adapted leaves (Fv/Fm), the chlorophyll (Chl) concentrations and the Rubisco contents, and leaf mass per area (LMA) at the leaf level and by determining the concentrations of cytochrome (Cyt) f and the reaction centres of photosystem (PS) I and PSII at the thylakoid level. In this experiment, partial shading was defined as the shading of 2nd leaves with shade cloths, and the whole treatment was defined as the covering of the whole individuals with shade cloths. In the leaf level responses, oxygen evolution, LMA, Chl concentrations and Rubisco contents decreased in all shade treatments administered for six days. Fv/Fm remained constant irrespective of the shade treatments. On the other hand, in the thylakoid-level responses, the concentrations of the thylakoid components per unit Chl and the stoichiometry of the two photosystems showed no statistical difference among the shade treatments. The data obtained from the present study indicate that the partial shading affected the leaf-level responses rather than the thylakoid-level responses. The light received at the lower leaves might serve as a factor in the regulation of the leaf properties of the upper leaves due to the whole plant photosynthesis, while this factor did not have an effect at the thylakoid level.  相似文献

Shoots of two species of moss, Plagiomnium undulatum (Hedw.) Kop. and Plagiomnium affine (Funck) Kop., were subjected to freezing at various temperatures. After thawing, the activities of different photosynthetic reactions were determined in relation to the ages of the leaves. Analysis of the fast kinetics of chlorophyll-a fluorescence of individual leaves showed that young and old tissues were considerably less frost tolerant than mature ones. In principle, the pattern of freeze inactivation of photosynthetic reactions resembles that observed in higher plants. The decreases in the amplitude of Fv (variable fluorescence) and the ratio of Fv to Fm (maximum fluorescence) with increasing freezing stress reflect a progressive inactivation of photosystem II (PSII)-mediated electron transport, i.e. inhibition of photoreaction to photochemistry and-or electron donation to the photochemical reaction, and thus a decline in the potential photochemical efficiency of PSII. The insignificant change in the F0 (constant fluorescence) level during progressive decline of Fv indicates that the excitation-energy transfer between antenna pigments and from those to reaction centres of PSII was little impaired by lethal freezing stress. Sugar analyses of various stem sections showed that ontogenetic variation in the frost tolerance of leaves cannot be attributed to differences in the cellular levels of sucrose, glucose and fructose.Abbreviations and Symbols DCMU 3-(3,4-dichlorophenyl)-1,1-dimethylurea - Fm maximum fluorescence - F0 constant (initial) fluorescence - Fv variable fluorescence  相似文献

Experiments were conducted to investigate the photosynthetic activity and thermostability of photosystem II (PSII) in elm seedling (Ulmus pumila) leaves from initiation to full expansion. During leaf development, photosynthesis, measured as CO2 fixation, increased gradually and reached a maximum value when leaves were fully developed. In parallel with the increase of carbon assimilation, chlorophyll content increased. The chlorophyll a fluorescence measurements showed that the maximum quantum yield of PSII primary photochemistry (φpo), the efficiency with which the energy of trapped excitons is converted into the electron transport beyond QA (Ψo) and the quantum yield of electron transport beyond QA (φEo) increased gradually. The low light experiments confirmed these results independently. When subjected to heat stress, young leaves exhibited progressively lower φpo and maximal fluorescence (Fm) values with considerably higher minimal fluorescence (Fo) than mature leaves, demonstrating that PSII in newly initiating leaves is more sensitive to heat stress. Further analysis revealed that PSII structure in newly initiating leaves showed a robust alteration under heat stress, which was reflected by the clear K phase in the OJIP curves. Therefore, we suggest that the enhanced thermostability of PSII in the case of leaf growth might be associated with an improvement of the stability of the oxygen-evolving complex (OEC) to heat stress during leaf development.  相似文献

CO2 assimilation, xanthophyll cycle pigments and PSII efficiency were analyzed in two different ages of pumpkin leaves (Cucurbita pepo L. cv. Ambassador) exposed to 150 nmol mol-1 of ozone (5 days, 5 h day-1). Gas-exchange measurements revealed a reduction in CO2 assimilation and stomatal conductance, accompanied by an increase in the intercellular CO2 concentration both in young and in mature leaves as compared to their respective controls. In both leaves, F0 remained unchanged, while Fm and the Fv/Fm ratio decreased after O3 fumigation, indicating that ozone may induce an alteration in the capability of photosystem II (PSII) to reduce the primary acceptor QA. In the mature leaves the photochemical quenching (qp) was significantly lowered by the pollutant, but this was not the case in the young leaves where qp did not change. In both mature and young ozonated pumpkin leaves, the development of non-photochemical quenching caused a decrease in the PSII photochemical rate, as shown by the correlation between Fv/Fm and the de-epoxidation state of dark-adapted leaves. Decreases in the Fv/Fm ratio are generally attributed to damage to the PSII reaction centre, apart from the down-regulation of the capacity of PSII electron transport. While in young ozonated leaves the decrease in the Fv/Fm ratio was not associated with damage to the D1 protein, in mature ozonated pumpkin leaves, the decrease in the Fv/Fm was accompanied by a significant decline in the D1 content. In conclusion, ozone exposure induces alterations in the light reactions of photosynthesis in both young and mature leaves. However, in young leaves the engagement of the xanthophyll cycle appears to counteract ozone effects against the photosynthetic apparatus as demonstrated by the absence of damage to the D1 protein. On the other hand, the loss of D1 protein in mature fumigated leaves suggests that the activation of the xanthophyll cycle is not sufficient to prevent photoinhibition, probably because a physiological state of senescence adds to the oxidative stress.  相似文献

Regeneration capacity of submerged rice (Oryza sativa) seedlings in terms of CO2 photosynthetic rate, chlorophyll a fluorescence and chlorophyll and carbohydrate content were investigated in three Indica rice cultivars namely FR 13A, Kalaputia and IR 42 that differed in submergence tolerance. Twenty-one day old plants were completely submerged under water for 8 days. Subsequently, plants were kept under normal conditions with 5–10 cm of stagnant water above soil surface for a further period of 15 days. After complete submergence, all genotypes showed inhibition of CO2 photosynthetic rate. Submergence treatment resulted in a significant reduction of Rubisco activity. Maximal photochemical efficiency (Fv/Fm) of PS II and area above the fluorescence curve between Fo and Fm decreased more under submergence especially in susceptible cultivar IR 42. When re-aerated, the plants recovered to various degrees. The carbohydrate content of plants was found to be significantly and positively associated with submergence tolerance and regeneration growth. The tolerant cultivar (FR 13A) could survive submergence apparently because it possessed 1.9–2.0 and 3.2–3.7-fold more non-structural carbohydrate content before and after submergence compared to the susceptible cultivar (IR 42) and it had a better capability to restore its photosynthetic capacity during post-submergence periods.  相似文献

In Amazonian floodplains, plant survival is determined by adaptations and growth strategies to effectively capture sunlight and endure extended periods of waterlogging. By measuring gas exchange, quantum efficiency of photosystem 2 (PSII), and growth parameters, we investigated the combined effects of flooding gradients and light on two common evergreen floodplain tree species, the light-tolerant Cecropia latiloba and the shade-tolerant Pouteria glomerata. Individual plants were subjected to different combinations of light and flooding intensity in short-term and long-term experiments. Plants of C. latiloba lost all their leaves under total submersion treatments (plants flooded to apex and with reduced irradiance) and showed highest maximum assimilation rates (Amax) in not flooded, high light treatments (6.1 μmol CO2 m−2 s−1). Individuals of P. glomerata showed similar patterns, with Amax increasing from 1.9 μmol CO2 m−2 s−1 under total flooding to 7.1 μmol CO2 m−2 s−1 in not flooded, high light treatments. During the long-term flooding experiment, quantum efficiency of PSII (Fv/Fm) of C. latiloba was not affected by partial flooding. In contrast, in P. glomerata Fv/Fm decreased to values below 0.73 after 120 days of total flooding. Moreover, total submergence led P. glomerata to reduce significantly light saturation point (LSP), as compared to C. latiloba. For both species morphological adjustments to long-term flooding, such as the production of adventitious roots, resulted in reduced total biomass, relative growth rate (RGR) and leaf mass ratio (LMR). Growth increase in C. latiloba seemed to be more limited by low-light than by flooding. Therefore, the predominant occurrence of this species is in open areas with high light intensities and high levels of inundation. In P. glomerata flooding induced high reductions of growth and photosynthesis, whereas light was not limiting. This species is more abundant in positions where irradiance is reduced and periods of submergence are slightly modest. We could show that the physiological requirements are directly responsible for the flooding (C. latiloba) and shade (P. glomerata) tolerance of the two species, which explains their local distribution in Amazonian floodplain forests.  相似文献

Fully exposed, senescing leaves of Cornus sanguinea and Parthenocissus quinquefolia display during autumn considerable variation in both anthocyanin and chlorophyll (Chl) concentrations. They were used in this study to test the hypothesis that anthocyanins may have a photoprotective function against photosystem II (PSII) photoinhibitory damage. The hypothesis could not be confirmed with field sampled leaves since maximum photochemical efficiency (Fv/Fm) of PSII was negatively correlated to anthocyanin concentration and the possible effects of anthocyanins were also confounded by a decrease in Fv/Fm with Chl loss. However, after short-term laboratory photoinhibitory trials, the percent decrease of Fv/Fm was independent of Chl concentration. In this case, a slight alleviation of PSII damage with increasing anthocyanins was observed in P. quinquefolia, while a similar trend in C. sanguinea was not statistically significant. It is inferred that the assumed photoprotection, if addressed to PSII, may be of limited advantage and only under adverse environmental conditions.  相似文献

Sulfite treatment of pea leaf disks in light caused a significant decrease in the relative quantum yield of photosynthetic oxygen evolution and energy storage (ES) as measured by photoacoustic (PA) spectroscopy. The inhibition was concentration dependent and was less in darkness than in light, indicating light-dependent inhibitory site(s) on the photosynthetic electron transport chain. Further, in darksulfite-treated leaves, the energy storage was more affected than the relative quantum yield of oxygen evolution, suggesting that photophosphorylation and/or cyclic electron transport around PS I are sites of sulfite action in darkness. The Rfd values, the ratio of fluorescence decrease (fd) to the steady-state fluorescence (fs), decreased significantly in leaves treated with sulfite in light but were not affected in dark-treated ones, confirming the photoacoustic observations. Similarly, the ratio of variable fluorescence (Fv) to maximum fluorescence (Fm), a measure of PS II photochemical efficiency, was affected by sulfite treatment in light and not changed by treatment in darkness. An attempt was made to explain the mechanism of sulfite action on photosynthetic electron transport in light and in darkness.Abbreviations APT amplitude of photothermal signal - Aox amplitude of oxygen signal - ES energy storage - fd fluorescence decrease - fs steady-state fluorescence - Fm maximum fluorescence - Fv variable fluorescence - PA photoacoustic(s)  相似文献

The photosynthetic performances of regenerated protoplasts of Bryopsis hypnoides, which were incubated in seawater for 1, 6, 12, and 24 h, were studied using chlorophyll (Chl) fluorescence and oxygen measurements. Results showed that for the regenerated protoplasts, the pigment content, the ratios of photosynthetic rate to respiration rate, the maximal photosystem II (PSII) quantum yield (Fv/Fm), and the effective PSII quantum yield (ΦPSII) decreased gradually along with the regeneration progress, indicated that during 24 h of regeneration there was a remarkable reduction in PSII activity of those newly formed protoplasts. We assumed that during the cultivation progress the regenerated protoplasts had different photosynthetic vigor, with only some of them able to germinate and develop into mature thalli. The above results only reflected the photosynthetic features of the regenerated protoplasts at each time point as a whole, rather than the actual photosynthetic activity of individual aggregations. Further investigation suggested a relationship between the size of regenerated protoplasts and their viability. The results showed that the middle-sized group (diameter 20–60 μm) retained the largest number of protoplasts for 24 h of growth. The changes in Fv/Fm and ΦPSII of the four groups of differently sized protoplasts (i.e. < 20, 20–60, 60–100, and > 100 μm) revealed that the protoplasts 20–60 μm in diameter had the highest potential activity of the photosynthetic light energy absorption and conversion for several hours.  相似文献

The present study was undertaken to investigate the effect of Glomus mosseae on chlorophyll (Chl) content, Chl fluorescence parameters and chloroplast ultrastructure of beach plum seedlings under 2% NaCl stress. The results showed that compared to control, both Chl a and Chl b contents of NaCl + G. mosseae treatment were significantly lower during the salt stress, while Chl a/b ratio increased significantly. The increase of minimal fluorescence of darkadapted state (F0), and the decrease of maximal fluorescence of dark-adapted state (Fm) and variable fluorescence (Fv) values were inhibited. The maximum quantum yield of PSII photochemistry (Fv/Fm), the maximum energy transformation potential of PSII photochemistry (Fv/F0) and the effective quantum yield of PSII photochemistry (??PSII) increased significantly, especially the latter two variables. The values of the photochemical quenching coefficient (qP) and the nonphotochemical quenching (NPQ) were similar between G. mosseae inoculation and noninoculation. It could be concluded that G. mosseae inoculation could protect the photosystem II (PSII) of beach plum, enhance the efficiency of primary light energy conversion and improve the primitive response of photosynthesis under salinity stress. Meanwhile, G. mosseae inoculation was beneficial to maintain the integrity of thylakoid membrane and to protect the structure and function of chloroplast, which suggested that G. mosseae can alleviate the damage of NaCl stress to chloroplast.  相似文献

In this work, photosystem II (PSII) photochemistry, leaf water potential, and pigment contents of male and female Pistacia lentiscus L. were investigated during a seasonal cycle at three different, arid locations: superior semiarid, inferior semiarid, and arid. The results showed that the gender, season, and the site conditions interacted to influence the quantum yield and pigment contents in P. lentiscus. Predawn leaf water status was determined only by the site and season. The annual patterns of PSII maximum quantum efficiency (Fv/Fm) were characterized by a suboptimal activity during the winter, especially, populations with the more negative water potential exhibited a lower chlorophyll (Chl) a content and chronic photoinhibition irrespective of a gender. We also demonstrated that both photochemical or nonphotochemical mechanisms were involved to avoid the photoinhibition and both of them depended on the season. This plasticity of photosynthetic machinery was accompanied by changes in carotenoids and Chl balance. In the spring, the female Fv/Fm ratio was significantly higher than in male individuals, when the sexual dimorphism occurred during the fruiting stage, regardless of site conditions. P. lentiscus sex-ratio in Mediterranean areas, where precipitations exceeded 500 mm, was potentially female-biased. Among the fluorescence parameters investigated, nonphotochemical quenching coefficient appeared as the most useful one and a correlation was found between Chl a content and Fv/Fm. These results suggest that functional ecology studies would be possible on a large scale through light reflectance analysis.  相似文献

Arbuscular mycorrhizal fungi (AMF) can promote plant growth performance, but their effectiveness varies depending on soil nitrogen (N) availability. To clarify the effectiveness of exogenous AMF along an N-fertilization gradient (0, 2, 10, 20, and 30 mM), the impacts of exogenous Rhizophagus irregularis and N on the growth, photochemical activity, and nutritional status of Populus?×?canadensis ‘Neva’ in natural soil were evaluated in a pot experiment. The results showed that the 10 mM N level was the optimal fertilization regime with the highest promotion effect on plant growth and the maximum quantum yield of photosystem II (PSII) (Fv/Fm). Excess N (20 and 30 mM) fertilization reduced the actual quantum yield of PSII (ФPSII) and the Fv/Fm of the plants. Regardless of the N availability, inoculated plants exhibited greater Fv/Fm values than did non-inoculated plants. The biomass of inoculated plants was significantly higher compared with the control under low N levels (0 and 2 mM). Under high N levels, inoculated plants showed significant increases in ФPSII. Moreover, the nutrient imbalance of plants inoculated with exogenous R. irregularis was eased by increasing P, Fe, Mn and Cu uptake in roots and higher P, Ca, Mg, Fe, Mn and Zn concentrations in leaves. Moreover, the Fv/Fm and ФPSII exhibited positive correlations with P, Ca, Mg and Zn concentrations in leaves. In conclusion, inoculation with exogenous R. irregularis can benefit plant fitness by improving the photochemical capacity and nutrient composition of poplar under different N levels.  相似文献

The PSII photochemical activity in a terrestrial cyanobacterium Nostoc commune Vaucher ex Bornet et Flahault during rewetting was undetectable in the dark but was immediately recognized in the light. The maximum quantum yield of PSII (Fv/Fm) during rewetting in the light rose to 85% of the maximum within ~30 min and slowly reached the maximum within 6 h, while with rewetting in the darkness for 6 h and then exposure to light the recovery of Fv/Fm required only ~3 min. These results suggested that recovery of photochemical activity might depend on two processes, light dependence and light independence, and the activation of photosynthetic recovery in the initial phase was severely light dependent. The inhibitor experiments showed that the recovery of Fv/Fm was not affected by chloramphenicol (CMP), but severely inhibited by 3‐(3,4‐dichlorophenyl)‐1,1‐dimethylurea (DCMU) in the light, suggesting that the light‐dependent recovery of photochemical activity did not require de novo protein synthesis but required activation of PSII associated with electron flow to plastoquinone. Furthermore, the test indicated that the lower light intensity and the red light were of benefit to its activation of photochemical activity. In an outdoor experiment of diurnal changes of photochemical activity, our results showed that PSII photochemical activity was sensitive to light fluctuation, and the nonphotochemical quenching (NPQ) was rapidly enhanced at noon. Furthermore, the test suggested that the repair of PSII by de novo protein synthesis played an important role in the acclimation of photosynthetic apparatus to high light, and the heavily cloudy day was more beneficial for maintaining high photochemical activity.  相似文献

Treatment with the herbicide acifluorfen-sodium (AF-Na), an inhibitor of protoporphyrinogen oxidase, caused an accumulation of protoporphyrin IX (Proto IX) , light-induced necrotic spots on the cucumber cotyledon within 12-24 h, and photobleaching after 48-72 h of light exposure. Proto IX-sensitized and singlet oxygen (1O2)-mediated oxidative stress caused by AF-Na treatment impaired photosystem I (PSI), photosystem II (PSII) and whole chain electron transport reactions. As compared to controls, the Fv/Fm (variable to maximal chlorophyll a fluorescence) ratio of treated samples was reduced. The PSII electron donor NH2OH failed to restore the Fv/Fm ratio suggesting that the reduction of Fv/Fm reflects the loss of reaction center functions. This explanation is further supported by the practically near-similar loss of PSI and PSII activities. As revealed from the light saturation curve (rate of oxygen evolution as a function of light intensity), the reduction of PSII activity was both due to the reduction in the quantum yield at limiting light intensities and impairment of light-saturated electron transport. In treated cotyledons both the Q (due to recombination of QA with S2) and B (due to recombination of QB with S2/S3) band of thermoluminescence decreased by 50% suggesting a loss of active PSII reaction centers. In both the control and treated samples, the thermoluminescence yield of B band exhibited a periodicity of 4 suggesting normal functioning of the S states in centers that were still active. The low temperature (77 K) fluorescence emission spectra revealed that the F695 band (that originates in CP-47) increased probably due to reduced energy transfer from the CP47 to the reaction center. These demonstrated an overall damage to the PSI and PSII reaction centers by 1O2 produced in response to photosensitization reaction of protoporphyrin IX in AF-Na-treated cucumber seedlings.  相似文献

Cytokinins (CKs) are one of the main regulators of in vitro growth and development and might affect the developmental state and function of the photosynthetic apparatus of in vitro shoots. Effects of different cytokinin regimes including different types of aromatic cytokinins, such as benzyl-adenine, benzyl-adenine riboside and 3-hydroxy-benzyladenine alone or in combination were studied on the capacity of the photosynthetic apparatus and the pigment content of in vitro apple leaves after 3 weeks of culture. We found that the type of cytokinins affected both chlorophyll a and b contents and its ratio. Chlorophyll content of in vitro apple leaves was the highest when benzyl-adenine was applied as a single source of cytokinin in the medium (1846–2176 μg/1 g fresh weight (FW) of the leaf). Increasing the concentration of benzyl-adenine riboside significantly decreased the chlorophyll content of the leaves (from 1923 to 1183 μg/1 g FW). The highest chl a/chl b ratio was detected after application of meta-topolin (TOP) at concentrations of 2.0 and 6.0 μM (2.706 and 2.804). Chlorophyll fluorescence was measured both in dark-adapted (Fv/Fm test) and in light-adapted leaf samples (Yield test; Y(II)). The maximum quantum yield and efficiency of leaves depended on the cytokinin source of the medium varied between 0.683 and 0.861 (Fv/Fm) indicating a well-developed and functional photosynthetic apparatus. Our results indicate that the type and concentration of aromatic cytokinins applied in the medium affect the chlorophyll content of the leaves in in vitro apple shoots. Performance of the photosynthetic apparatus measured by chlorophyll fluorescence in the leaves was also modified by the cytokinin supply. This is the first ever study on the relationship between the cytokinin supply and the functionability of photosystem II in plant tissue culture and our findings might help to increase plantlet survival after transfer to ex vitro conditions.  相似文献

Summary We have investigated the diurnal response of photosynthesis and variable photosystem II (PSII) chlorophyll fluorescence at 77 K for thalli of the chlorophyte macroalga, Ulva rotundata, grown in outdoor culture and transplanted to an intertidal sand flat in different seasons. The physiological response in summer indicated synergistic effects of high PFD and aerial exposure, the latter probably attributable to temperature, which usually increased by 8 to 10° C during midday emersion. Except at extreme emersed temperatures in summer (38° C), the light-saturated photosynthesis rate (Pm) did not decline at midday. In contrast, light-limited quantum yield of photosynthetic O2 exchange () and the ratio of variable to maximum fluorescence yield (Fv/Fm) reversibly declined during midday low tides in all seasons. Shade-grown thalli exhibited a fluorescence response suggestive of greater photodamage to PSII, whereas sun-grown thalli had greater photoprotective capacity. The fluorescence decline was smaller when high tide occurred at midday, and was delayed during morning cloudiness. These results suggest that the diurnal response to PFD in this shallow water species is modified by tidal and meteorological factors. U. rotundata has a great capacity for photoprotection which allows it to tolerate and even thrive in the harsh intertidal environment.Abbreviations Fo instantaneous yield of chlorophyll fluorescence - Fm maximum yield of fluorescence - Fv variable yield (Fm–Fo) of fluorescence - PFD photon flux density (400–700 nm) - Pm light-saturated rate of photosynthesis - PSH photosystem II - QA electron acceptor of PSII - light-limited quantum yield of photosynthesis  相似文献

We studied the developmental changes in photosynthetic and respiration rates and thermal dissipation processes connected with chloroplasts and mitochondria activity in etiolated wheat (Triticum aestivum L., var. Irgina) seedlings during the greening process. Etioplasts gradually developed into mature chloroplasts under continuous light [190 μmol(photon) m?2 s?1] for 48 h in 5-day-dark-grown seedlings. The net photosynthetic rate of irradiated leaves became positive after 6 h of illumination and increased further. The first two hours of de-etiolation were characterized by low values of maximum (Fv/Fm) and actual photochemical efficiency of photosystem II (PSII) and by a coefficient of photochemical quenching in leaves. Fv/Fm reached 0.8 by the end of 24 h-light period. During greening, energy-dependent component of nonphotochemical quenching of chlorophyll fluorescence, violaxanthin cycle (VXC) operation, and lipoperoxidation activity changed in a similar way. Values of these parameters were the highest at the later phase of de-etiolation (4–12 h of illumination). The respiration rate increased significantly after 2 h of greening and it was the highest after 4–6 h of illumination. It was caused by an increase in alternative respiration (AP) capacity. The strong, positive linear correlation was revealed between AP capacity and heat production in greening tissues. These results indicated that VXC in chloroplasts and AP in mitochondria were intensified as energy-dissipating systems at the later stage of greening (after 4 h), when most of prolamellar bodies converted into thylakoids, and they showed the greatest activity until the photosynthetic machinery was almost completely developed.  相似文献
