The taxonomic relationship between two microcyclic rust species, Puccinia melampodii and Puccinia xanthii, recorded from a number of Asteraceae hosts, was explored by comparing teliospore morphology, host specificity and sequence data. Teliospore morphology varied between and within individual rust accessions but, in general, teliospores of P. xanthii were larger than those of P. melampodii. Field observations and cross-inoculation greenhouse studies showed that P. melampodii accessions were highly specific to their original host species and unable to establish compatible interactions with related Asteraceae species. Though small amounts of genetic variation were detected between P. melampodii and P. xanthii accessions, the ITS and TEF phylogenetic analyses comprising several other rust species grouped these accessions within a well supported clade. Our data indicate that despite differences between P. xanthii and P. melampodii accessions these rusts cannot readily be separated at the species level, supporting earlier taxonomic conclusions and the recent proposal to group them into a morphospecies. Based on host-specificity data, the name Puccinia xanthii var. parthenii-hysterophorae is proposed to accommodate all Mesoamerican records of P. melampodii associated with the host Parthenium hysterophorus.  相似文献

The suspension-feeding slippersnail Crepidula convexa is commonly associated with hermit crabs (Pagurus longicarpus) living in periwinkle shells (Littorina littorea) at our study site in Nahant, MA, USA. In 15 field surveys conducted at Nahant in 2000, 2001 and 2003, we found that (1) more than 61.8% of individuals of C. convexa resided on shells occupied by hermit crabs, as opposed to the shells of live periwinkles, empty periwinkle shells or other solid substrates; (2) an average of 8.3% of hermit crabs carried at least one individual of C. convexa; and (3) 39.1-75.0% of hermit crabs carrying C. convexa were carrying “large” individuals (snails with wet weight >10% of the weight of the periwinkle shells they occupied). However, it is unlikely that individuals of C. convexa seek out shells occupied by hermit crabs to colonize, and they showed no preference for empty periwinkle shells over other solid substrates in the laboratory. Moreover, in the laboratory the hermit crabs preferentially occupied intact shells bearing individuals of C. convexa only when the alternatives were shells that had been drilled by naticid snails. Thus, neither party preferentially associates with the other: rather, extensive predation by naticid snails on periwinkles at Nahant appears to limit the availability of suitable shells for the hermit crabs, forcing them to inhabit shells bearing “large” individuals of C. convexa. Individuals of C. convexa may benefit from this inadvertent association with hermit crabs: by facilitating snail dispersal, transport by hermit crabs should reduce the potential for inbreeding, an important consideration for a species that lacks free-living larvae in its life history.  相似文献

Looking back over the century long research career of the fruit fly, Drosophila melanogaster has frequently been in the scientific spotlight with respect to fundamental discoveries in biology. The last decade witnessed the increasing importance of the fly as a human disease model but studies on energy homeostasis and lipometabolism remain in their infancy. This perspective, addressing readers largely unfamiliar with the Drosophila model system, aims to highlight the starting points for which the fly could be employed to gain a deeper understanding of lipotoxicity and possibly contribute to strategies for the identification of novel drug targets relevant to type 2 diabetes mellitus and the metabolic syndrome.  相似文献

The increase of infections due to non-Candida albicans species made it very necessary to conduct adequate characterization to be able to identify the species of Candida isolated from traditional fermented foods. In this study, based on their hue on Candida Chromogenic Agar medium, a total of 136 yeast strains were isolated from tchapalo and bangui. Molecular identification based on PCR-RFLP of internal transcribed spacers of rDNA (ITS) and sequencing of the ITS and the D1/D2 regions allowed us to assign these isolates to seven species: Candida tropicalis, Candida inconspicua, Candida rugosa, Saccharomyces cerevisiae, Kluyveromyces marxianus, Hanseniaspora guilliermondii, Trichosporon asahii. With the respect to each beverage, six species were found among with four species are regarded as opportunistic pathogens. From these, C. tropicalis, C. inconspicua and K. marxianus were the most commonly encountered. The enzyme activities of the potential pathogens assessed using API ZYM system showed that almost strains had esterase, esterase lipase, valine and cystine arylamidase, alpha chymotrypsin, alkaline phosphatase and naphthol phosphohydrolase activities. The activity of α-glucosidase was found only in C. tropicalis and C. inconspicua strains isolated from tchapalo while β-glucosidase activity was found in all strains from tchapalo and only in C. inconspicua isolated from bangui.  相似文献

Aquatic Batrachium Ranunculus species are a key component of river macrophyte communities selected for protection under European Union legislation. The group's simplified morphology and variable taxonomic interpretation often makes identification to species level very difficult. A genetic approach was trialled as an alternative, more reliable, means of identification. DNA barcoding using four markers (chloroplast and nuclear) was tested. The chloroplast sequence trnH-psbA worked best and allowed identification of three out of five species while nuclear sequences supported the identification of two hybrids. This method is amenable to simplification through techniques such as PCR-RFLP or RT-PCR. It has the potential to provide easy, rapid and inexpensive identification of Batrachium Ranunculus species.  相似文献

The presence of two distinct types of adipose tissue, which have opposing functions, has been known for decades. White adipose tissue (WAT) is the main tissue of energy storage, while brown adipose tissue (BAT) dissipates energy as heat and is required for non-shivering thermoregulation. In the last few years, a third type of adipocyte was identified, termed the brite (“brown and white”) or beige adipocyte. Their physiological control and role, however, are not fully clarified. Brite/beige adipocytes have a positive impact on systemic metabolism that is generally explained by the thermogenesis of brite/beige adipocytes; although thermogenesis has not been directly measured but is mostly inferred by gene expression data of typical thermogenic genes such as uncoupling protein 1 (UCP1). Here we critically review functional evidence for the thermogenic potential of brite/beige adipocytes, leading to the conclusion that direct measurements of brite/beige adipocyte bioenergetics, beyond gene regulation, are pivotal to quantify their thermogenic potential. In particular, we exemplified that the massive induction of UCP1 mRNA during the browning of isolated subcutaneous adipocytes in vitro is not reflected in significant alterations of cellular bioenergetics. Herein, we demonstrate that increases in mitochondrial respiration in response to beta-adrenergic stimulus can be independent of UCP1. Using HEK293 cells expressing UCP1, we show how to directly assess UCP1 function by adequate activation in intact cells. Finally, we provide a guide on the interpretation of UCP1 activity and the pitfalls by solely using respiration measurements. The functional analysis of beige adipocyte bioenergetics will assist to delineate the impact of browning on thermogenesis, possibly elucidating additional physiological roles and its contribution to systemic metabolism, highlighting possible avenues for future research. They unravel the integral adaptive and active nature of the organism behavior in the environment. Otoliths are dense structures in the ears of fishes that function in hearing and gravity perception. Otolith (sagitta) diameters, as percentages of standard length (% SL), are calculated for 247 marine fish species in 147 families and compared by taxonomic group (usually order), habitat and presence or absence of luminescence. Otolith sizes range from 0.4-31.4 mm and 0.08-11.2% SL. The eel and spiny eel orders Anguilliformes and Notacanthiformes have small to very small otoliths, as do the triggerfish order Tetraodontiformes, pipefish order Gasterosteiformes, billfish suborder Scombroidei and many of the dragonfish order Stomiiformes. The soldierfish order Beryciformes has moderate to very large otoliths. The perch order Perciformes has a wide range of otolith sizes but most have small to moderate otoliths 2-5% SL. Only 16 out of the 247 species have the relatively largest otoliths, over 7% SL. Seven out of these 16 species are also luminous from a variety of habitats. Luminous species have slightly to much larger otoliths than non-luminous species in the same family Both beryciforms and luminous fishes live in low-light environments, where acute colour vision is probably impossible. Most fishes of the epipelagic surface waters have very small otoliths, perhaps due to background noise and/or excessive movement of heavy otoliths in rough seas. Bathypelagic species usually have small otoliths and regressed or absent swimbladders. Other habitats have species with a range of otolith sizes. While the relationship between hearing ability and otolith length is unknown, at least some groups with modified swim-bladders have larger otoliths, which may be associated with more acute hearing.  相似文献

Members of the Paracoccidioides complex are human pathogens that infect different anatomic sites in the host. The ability of Paracoccidioides spp. to infect host niches is putatively supported by a wide range of virulence factors, as well as fitness attributes that may comprise the transition from mycelia/conidia to yeast cells, response to deprivation of micronutrients in the host, expression of adhesins on the cell surface, response to oxidative and nitrosative stresses, as well as the secretion of hydrolytic enzymes in the host tissue. Our understanding of how those molecules can contribute to the infection establishment has been increasing significantly, through the utilization of several models, including in vitro, ex vivo and in vivo infection in animal models. In this review we present an update of our understanding on the strategies used by the pathogen to establish infection. Our results were obtained through a comparative proteomic analysis of Paracoccidioides spp. in models of infection.  相似文献

Spatialassociations among overstory and understory species tend to increase ongradients from wet to dry climates. This shift in the strength of spatialassociations has usually been attributed to shared abiotic requirements betweencanopy species and understory assemblages within communities and/or to anincrease in habitat heterogeneity in dry climates and therefore higher betadiversity. On another hand, more important positive effects of tree canopies onunderstory species in drier climates may also explain stronger associations andhigher beta diversity. We examined these three hypotheses along a strongrainshadow gradient that occurs from the wet external Alps to the dry innerAlpsby analyzing with correspondence analysis and canonical correspondence analysisthe species composition of 290 relevés of forests dominated to differentdegrees by Abies alba and Piceaabies.We found important differences in climatic requirements forAbies and Picea, withAbies occurring in warmer and drier habitats thanPicea. The understory species associated with these twospecies showed similar correlations with temperature but not with moisture,withunderstory species of Picea-communities having strongerxeric affinities than understory species ofAbies-communities. We found no significant associationsbetween canopy species and understory composition in the external Alps despitethe fact that Abies and Piceaoccurredin substantially different environments. In contrast,Abiesand Picea occurred in more similar environments in theinner Alps, but the understory assemblages associated with eitherAbies or Picea were significantlydifferent. This increase in canopy-understory associations was in partdetermined by strong differences in moisture between southern and northernaspects in the inner Alps, which affected both canopy and understory speciesdistributions. However, differences between the canopy effects ofPicea and Abies also appeared tocontribute to stronger associations between canopy and understory species, andconsequently to increase beta diversity. This pattern only occurred on southernaspects of the inner Alps but was highly significant. Our results suggest thatspecies distributions may be continuous on the wet ends of moisture gradientsbut discrete on dry ends. Relatively discrete communities at stressful ends ofgradients appear to develop as a result of both habitat differentiation and thepositive effects of overstory species.  相似文献

An important goal in evolutionary ecology is to understand how and why coexisting closely related species partition habitat among themselves. Although studies of interspecific interactions typically focus on males, interactions between females may also play an important role in shaping habitat use within multi‐species communities. The green anole (Anolis carolinensis) exhibits a wide range of habitat use in south‐eastern Louisiana, but its observed habitat use is restricted and altered in areas where it occurs with the introduced Anolis sagrei. We staged interactions between these two species in the laboratory to test the hypothesis that A. sagrei dominate A. carolinensis in contests over shared habitat. We examined whether species identity, bite force, dewlap size, and body size affected the outcome of interspecific interactions between both males and females, and tested the prediction that bite force and size would be the most important determinants of interaction outcomes in both sexes. In male interspecific interactions, we found that individuals with relatively larger dewlaps tended to score higher on aggressive behaviours regardless of species identity, and that interactions consisted of signalling and rarely escalated to physical combat. However, we found that A. sagrei females achieved higher aggressive scores than A. carolinensis females in almost all cases, lending support to the notion that female interspecific behaviour is probably more important than male behaviour in driving changes in habitat use. © 2013 The Linnean Society of London, Biological Journal of the Linnean Society, 2013, 110 , 843–851.  相似文献

Speciation with gene flow may be more common than generally thought, which makes detailed understanding of the extent and pattern of genetic divergence between geographically isolated populations useful. Species of the Drosophila simulans complex provide a good model for speciation and evolutionary studies, and hence understanding their population genetic structure will increase our understanding of the context in which speciation has occurred. Here, we describe genetic diversity and genetic differentiation of two distant populations of D. mauritiana (Mauritius and Rodrigues Islands) at mitochondrial and nuclear loci. We surveyed the two populations for their mitochondrial haplotypes, eight nuclear genes and 18 microsatellite loci. A new mitochondrial type is fixed in the Rodrigues population of D. mauritiana. The two populations are highly differentiated, their divergence appears relatively ancient (100,000 years) compared to the origin of the species, around 0.25MYA, and they exhibit very limited gene flow. However, they have similar levels of divergence from their sibling, D. simulans. Both nuclear genes and microsatellites revealed contrasting demographic histories between the two populations, expansion for the Mauritius population and stable population size for the Rodrigues Island population. The discovery of pronounced geographic structure within D. mauritiana combined to genetic structuring and low gene flow between the two island populations illuminates the evolutionary history of the species and clearly merits further attention in the broad context of speciation.  相似文献
