Changes in fresh matter content, protein content, chlorophyl content, ribonuclease activity, and glucose-6-phosphate dehydrogenase activity, associated with potato Y-virus multiplication (common strain, PVY ordinary) were studied in discs cut from potato leaves. The results obtained showed that marked decreases in disc fresh matter, in protein content, and in chlorophyll content occurred during a 5-day-long cultivation period. The activity of glucose-6-phosphate dehydrogenase, that is of the rate limiting enzyme of the pentose phosphate pathway, and the activity of ribonucleases which characterize the rate and intensity of host RNA degradation were markedly enhanced in this period. The fact that activity curves of both these enzymes were in linear relationship with the PVY reproduction curve indicates that not only nucleotides produced in the reactions of the oxidative pentose phosphate pathway but also nucleotides released in the process of host RNA degradation were the main source of nucleotides necessary for PVY-RNA biosynthesis, in spite of a high photosynthetic rate.  相似文献

It has been suggested by some authors that during amphibian development, due to the higher glucose-6-phosphate dehydrogenase (EC 1.1.1.49) activity compared to that of 6-phosphogluconate dehydrogenase (EC 1.1.1.43), 6-phosphogluconate could accumulate in the embryo tissues and regulate the channelling of glucose-6-phosphate into glycolysis. Here, on the base of the specific activities of glucose-6-phosphate dehydrogenase, 6-phosphogluconate dehydrogenase and glucose-6-phosphate isomerase (EC 5.3.1.9) found in the embryos of Bufo bufo during development, it is discussed whether 6-phosphogluconate can accumulate and play a regulative role on glucose-6-phosphate metabolism in the anuran embryo.  相似文献

The contents of NADP+, NADPH, changes in the activity of glucose-6-phosphate dehydrogenase and some questions relating to its regulation in leaf tissues of tobacco plants infected with PVY were studied. The content of NADP+ and the total sum of pyridine nucleotides decreased after inoculation to 15 % and 30 %, respectively, whereas the content of NADPH increased up to the threefold control value. The contents of NADP+, NADPH and Σ(NADPH + NADP+) linearly correlated with the reproduction curve of PVY. The value of the reduction charge RC and the value of the ratio NADPH/NADP+ sharply increased after inoculation up to tenfold and eighteenfold values, respectively, of the healthy control. The activity of glucose-6-phosphate dehydrogenase was markedly increased in virus-infected tissues during the entire experimental period both in crude homogenate and after its partial purification when compared with the values found in healthy control plants. The time courses of the activity curves of both crude and partially purified enzymes were coincident and correlated with the reproduction curve of PVY. The results indicate the involvement of coarse regulation of the activity of the enzyme by its content without the involvement of fine regulation by the ratio NADPH/NADP+ and RC.  相似文献

Activities of hexokinase and glucose-6-phosphate dehydrogenase have been measured in red blood cells from thyroidectomized, triiodothyronine treated and hyperthyroid rats. After thyroidectomy, significant decrease in the activities of hexokinase and glucose-6-phosphate dehydrogenase was observed as compared to controls. The effects were reversed with triiodothyronine administration to the thyroidectomized rats. Hyperthyroidism increased both enzymes. The observations further confirm the hypermetabolic effects of thyroid hormones on the cellular metabolism.  相似文献

Cancer cells consume large quantities of glucose and primarily use glycolysis for ATP production, even in the presence of adequate oxygen. This metabolic signature (aerobic glycolysis or the Warburg effect) enables cancer cells to direct glucose to biosynthesis, supporting their rapid growth and proliferation. However, both causes of the Warburg effect and its connection to biosynthesis are not well understood. Here we show that the tumour suppressor p53, the most frequently mutated gene in human tumours, inhibits the pentose phosphate pathway (PPP). Through the PPP, p53 suppresses glucose consumption, NADPH production and biosynthesis. The p53 protein binds to glucose-6-phosphate dehydrogenase (G6PD), the first and rate-limiting enzyme of the PPP, and prevents the formation of the active dimer. Tumour-associated p53 mutants lack the G6PD-inhibitory activity. Therefore, enhanced PPP glucose flux due to p53 inactivation may increase glucose consumption and direct glucose towards biosynthesis in tumour cells.  相似文献

Changes in glucose-6-phosphate dehydrogenase (G6P DH; EC 1.1.1.49) activity caused by infection of tobacco ( Nicotiana tabacum L.) leaves with potato virus Y (PVY), cucumber mosaic virus, potato virus X, tobacco rattle virus and turnip mosaic virus, the subcellular localisation of G6P DH isozymes in mesophyll protoplasts derived from healthy and PVY-infected tobacco leaves, as well as G6P DH control and the relationship of its isozymes with the degree of tobacco resistance to PVY multiplication, were studied. The activities of G6P DH were markedly increased in locally and systemically infected leaves and the time courses of the activity linearly correlated with those of virus multiplication. In leaves infected with PVY, the activity time courses of the crude and the partially purified G6P DH were coincident. This probably indicates the involvement of coarse regulation of the enzyme. PVY content linearly correlated with enhanced G6P DH activity in leaf discs derived from susceptible, tolerant and resistant cultivars of tobacco. The increased activity of the enzyme in infected protoplasts and plant tissues was predominantly caused by the increased activity of chloroplastic isozymes. This was confirmed by the specific staining of isozymes after electrophoretic separation of chloroplastic proteins of tobacco leaves. These findings enable the degree of resistance to virus multiplication to be quantified for the use of gene manipulation and breeding.  相似文献

Histochemistry and cytochemistry of glucose-6-phosphate dehydrogenase has found many applications in biomedical research. However, up to several years ago, the methods used often appeared to be unreliable because many artefacts occurred during processing and staining of tissue sections or cells. The development of histochemical methods preventing loss or redistribution of the enzyme by using either polyvinyl alcohol as a stabilizer or a semipermeable membrane interposed between tissue section and incubation medium, has lead to progress in the topochemical localization of glucose-6-phosphate dehydrogenase. Optimization of incubation conditions has further increased the precision of histochemical methods. Precise cytochemical methods have been developed either by the use of a polyacrylamide carrier in which individual cells have been incorporated before staining or by including polyvinyl alcohol in the incubation medium. In the present text, these methods for the histochemical and cytochemical localization of glucose-6-phosphate dehydrogenase for light microscopical and electron microscopical purposes are extensively discussed along with immunocytochemical techniques. Moreover, the validity of the staining methods is considered both for the localization of glucose-6-phosphate dehydrogenase activity in cells and tissues and for cytophotometric analysis. Finally, many applications of the methods are reviewed in the fields of functional heterogeneity of tissues, early diagnosis of carcinoma, effects of xenobiotics on cellular metabolism, diagnosis of inherited glucose-6-phosphate dehydrogenase deficiency, analysis of steroid-production in reproductive organs, and quality control of oocytes of mammals. It is concluded that the use of histochemistry and cytochemistry of glucose-6-phosphate dehydrogenase is of highly significant value in the study of diseased tissues. In many cases, the first pathological change is an increase in glucose-6-phosphate dehydrogenase activity and detection of these early changes in a few cells by histochemical means only, enables prediction of other subsequent abnormal metabolic events. Analysis of glucose-6-phosphate dehydrogenase deficiency in erythrocytes has been improved as well by the development of cytochemical tools. Heterozygous deficiency can now be detected in a reliable way. Cell biological studies of development or maturation of various tissues or cells have profited from the use of histochemistry and cytochemistry of glucose-6-phosphate dehydrogenase activity.(ABSTRACT TRUNCATED AT 400 WORDS)  相似文献

Summary Characterization of partially purified eryrhrocyte G-6-PD from 50 enzymedeficient males in 45 unrelated Thai families revealed 6 enzyme variants. Thirty-five subjects in 31 families had G-6-PD variant with normal electrophoretic mobility, slightly low Km G-6-P, normal substrate-analog utilization, normal pH-optimum curve, and slightly increased heat stability. This enzyme variant is called G-6-PD Mahidol.Six subjects had enzyme with fast electrophoretic mobility (106–108% of normal), low Km G-6-P, slightly increased substrate-analog utilization, biphasic pH-optimum curve, and slightly low to normal heat stability. This variant was identical to G-6-PD Canton.Five subjects had G-6-PD with fast electrophoretic mobility (103–106% of normal), low Km G-6-P, very high substrate-analog utilization except for DPN which it did not use as cofactor, markedly biphasic pH-optimum curve and very low heat stability. This variant is called G-6-PD Union (Thai).Two brothers had G-6-PD with normal electrophoretic mobility, low Km G-6-P, slightly increased substrate-analog utilization, biphasic pH-optimum curve and low heat stability. This variant is designated G-6-PD Siriraj.G-6-PD from one patient had slightly fast electrophoretic mobility, increased substrateanalog utilization, especially of DPN, and very low thermal stability. It is called G-6-PD Kan.One subject had G-6-PD with normal electrophoretic mobility, Km G-6-P, pH-optimum curve and heat stability, and increased substrate-analog utilization. This G-6-PD variant is named G-6-PD Anant.G-6-PD Mahidol is far more common than any other known variants in Thailand.
Zusammenfassung Eine Charakterisierung von teilweise gereinigtem Erythrocyten-G-6-PD von 50 Männern mit Enzym-Defekt aus 45 nicht miteinander verwandten Thai-Familien ergab 6 Enzym-Varianten. 35 Personen in 31 Familien hatten eine G-6-PD-Variante mit normaler elektrophoretischer Wanderungsgeschwindigkeit, einen leicht verminderten G-6-P-Km-Wert, einer normalen Substratanalog-Verwertung, einer normalen pH-Optimum-Kurve und einer leicht erhöhten Hitze-Stabilität. Diese Enzym-Variante wurde G-6-PD Mahidol genannt.Sechs Personen hatten ein Enzym mit rascher elektrophoretischer Wanderung (106–108% der Norm), niedrigem Km für G-6-P, leicht erhöhter Substrat-Verwertung, einer biphasischen pH-Optimum-Kurve und normaler bis leicht erniedrigter Hitzestabilität. Diese Variante ist identisch mit G-6-PD Canton.Fünt Personen hatten G-6-PD mit rascher elektrophoretischer Wanderung (103–106%), niedrigem Km G-6-P, sehr hoher Substratanalog-Verwertung—mit Ausnahme von DPN, das nicht als Cofactor wirkte—, einer stark biphasischen pH-Optimum-Kurve und sehr geringer Hitze-Stabilität. The changes in the activity of glucose-6-phosphate dehydrogenase (G6PDH) (EC 1.1.1.49) and 6-phosphogluconate dehydrogenase (6PGDH) (EC 1.1.1.44) in leaf tissues and the subcellular localisation of their isozymes in protoplasts derived from healthy and potato virus Y (PVY) infected plants of Nicotiana tabacum L. cv. Samsun were determined. The activities of G6PDH and 6PGDH were markedly increased in virus-infected leaves during the acute phase of infection both in crude homogenate and partial purificate (when compared with the values found in healthy control plants) and correlated with the multiplication curve of PVY. Intact chloroplasts and soluble cytosolic proteins were obtained from whole plants upon the culmination of the multiplication curve of PVY and upon the enhancement of the activity of both dehydrogenases by means of differential centrifugation of broken protoplasts. The chloroplastic fraction from infected protoplasts (based on chlorophyll content or NADP+-triosephosphate dehydrogenase activity) showed an enhanced activity of G6PDH (1.81 times that of healthy protoplasts), and 6PGDH (1.77 times). Cytosol from infected protoplasts (based on phosphoenolpyruvate carboxylase activity) contained only slightly enhanced activities of G6PDH and 6PGDH (only 1.26 and 1.16 times, respectively).  相似文献

Studies were carried out on glucose-6-phosphate dehydrogenase (G6P-DH) during the differentiation of rabbit bone marrow erythroid cells. It was found that G6P-DH, although displaying a 7-fold activity decrease, did not change the relative amounts of its three dimeric forms.Using homogeneous enzyme preparations, we observed that from dividing to non-dividing erythroblasts the following properties remained constant: V max dependence on pH and temperature, Km for G6P dependence on pH, heat stability, 2-deoxy glucose-6-phosphate utilization, molecular weight, while the Km for NADP significantly increased in non-dividing erythroblasts. These results indicate that no shift towards the oxidized form of the enzyme and no substantial modifications of the protein take place during cell differentiation.  相似文献
