The effect of combinations of different concentrations of copper (Cu) and mercury (Hg) ions with different constant temperatures (T) onPhaseolus vulgaris plants was studied. Criteria investigated included shoot and root length, dry mass, chlorophyll content (Chl) and stability index (CSI), and contents of soluble (SS) and hydrolysable saccharides (HS), soluble proteins (SP) and total free amino acid (AA). Each of the factors (T, Cu and Hg) significantly affected the parameters tested with exception of T effect on Chlb content as well as on Chla/b ratio. Bifactorial interactions (T×Cu) or (T×Hg) were also significant, except the interaction (T×Hg) in shoot elongation, Chlb content and Chla/b ratio. Statistical treatment of the data lead to three findings: (1) temperature was dominant in affecting CSIa, shoot AA and root SS, (2) Cu and Hg had the predominant effect on growth parameters and Chl content, and (3) interactions (T×Cu and T×Hg) were dominant in affecting CSIb, shoot SP, and root HS.  相似文献

Increasing soil acidity (from pH 6.5 to pH 2.0) decreased chlorophyll (Chl)a andb contents, dry matter accumulation by plants and the transpiration coefficient. Chl stability to heat significantly increased with increased soil acidity. The Chla/b ratio was increased significantly at pH 5 and 4 and decreased at pH 3 and 2. SprayingSorghum shoots with kinetin solutions counteracted the above adverse effects on Chl content and dry matter accumulation. Kinetin-treated plants showed a lower transpiration coefficient than the untreated plants.  相似文献

Patchouli (Pogostemon cablin Benth.) plants grown under shade (LI) showed an increased height due to internodal elongation, leaf area, leaf area index, and chlorophyll (Chl)b, Chl (a+b) and carotenoid (Car) contents compared to the plants grown in sunlight (HI). The number of branches and green leaves decreased under LI treatment, with a marginal variation in the patchouli oil yield. A comparison between the relative contents of photosynthetic pigments indicated that Chlb and Car accumuled preferentially over Chla in the LI grown plants.  相似文献

We use femtosecond transient absorption spectroscopy to study chlorophyll (Chl)-Chl energy transfer in the peridinin-chlorophyll protein (PCP) reconstituted with mixtures of either chlorophyll b (Chlb) and Chld or Chla and bacteriochlorophyll a (BChla). Analysis of absorption and transient absorption spectra demonstrated that reconstitution with chlorophyll mixtures produces a significant fraction of PCP complexes that contains a different Chl in each domain of the PCP monomer. The data also suggest that binding affinity of Chla is less than that of the other three Chl species. By exciting the Chl species lying at higher energy, we obtained energy transfer times of 40 ± 5 ps (Chlb-Chld) and 59 ± 3 ps (Chla-BChla). The experimental values match those obtained from the Förster equation, 36 and 50 ps, respectively, showing that energy transfer proceeds via the Förster mechanism. Excitation of peridinin in the PCP complex reconstituted with Chla/BChla mixture provided time constants of 2.6 and 0.4 ps for the peridinin-Chla and peridinin-BChla energy transfer, matching those obtained from studies of PCP complexes reconstituted with single chlorophyll species.  相似文献

Effects of zinc (12–180 μM) alone and in mixtures with 12 μM Cd on metal accumulation, dry masses of roots and shoots, root respiration rate, variable to maximum fluorescence ratio (FV/FM), and content of photosynthetic pigments were studied in hydroponically cultivated chamomile (Matricaria recutita) plants. The content of Zn in roots and shoots increased with the increasing external Zn concentration and its accumulation in the roots was higher than that in the shoots. While at lower Zn concentrations (12 and 60 μM) the presence of 12 μM Cd decreased Zn accumulation in the roots, treatment with 120 and 180 μM Zn together with 12 μM Cd caused enhancement of Zn content in the root. Presence of Zn (12–120 μM) decreased Cd accumulation in roots. On the other hand, Cd content in the shoots of plants treated with Zn + Cd exceeded that in the plants treated only with 12 μM Cd. Only higher Zn concentrations (120 and 180 μM) and Zn + Cd mixtures negatively influenced dry mass, chlorophyll (Chl) and carotenoid content, FV/FM and root respiration rate. Pollution of the root environment with excess of Pb retarded shoot growth, decreased chlorophyll (Chl) content and reduced Chl stability (CSI) to heat. Plants growing in Pb polluted soil accumulated much more free amino acids and less soluble sugars than the control plants. Stability of leaf membranes to heat (51 °C) or dehydration stresses (40% polyethylene glycol, 6000) decreased in response to Pb pollution where the membranes of leaf discs excised from Pb-treated plants were damaged more than those taken from plants growing in Pb free soil. Supplying kinetin ameliorated the deleterious effects of Pb pollution on the parameters tested. Kinetin-treated plants had higher Chl, soluble sugars content and produced more biomass in their shoots. Also, kinetin increased leaf membrane stability especially in Pb-treated plants, effectively protected chlorophyll degradation by heat and increased Chl a and b stability index; the most effective concentration was 10 mg L–1. The effects of Pb and kinetin as well as their interaction (Pb × Kin) on the parameters tested were statistically significant. Applied kinetin had a dominant role (as indicated by 2) in affecting shoot growth, soluble sugars, Chl a and b contents, stability of leaf membranes to dehydration stress as well as the Chl stability index. Pb had a dominant role on total free amino acids (TAA) and leaf relative water content (RWC). The interaction between Kin × Pb influenced the stability of leaf membranes to heat stress in a major way.  相似文献

The effects of cadmium (Cd) supply level in nutrient solution (0, 12.5, 25, 50, 100, 200, 400, and 800 μM) on growth, Cd accumulation ability, and the related physiological indices of maize (Zea mays L.) seedlings were studied under hydroponic conditions. The results showed that the increments in the shoot height and biomass were stimulated at relatively low external Cd supply levels (<100 μM), while they were inhibited at Cd supply levels over 200 μM. Cd accumulation ability of the maize seedlings also showed the similar stimulation/inhibition pattern as shoot growth, and the Cd contents in the shoots and roots reached the peaks (389.5 and 505.5 mg/kg dry wt, respectively) at 50 μM Cd. The contents of chlorophyll a, chlorophyll b, and carotenoids in the maize leaf blades decreased with increasing external Cd supply level. At the highest Cd supply level (800 μM), the contents of chlorophyll a, chlorophyll b, and carotenoids in the leaf blade were only 38.9, 46.0, and 29.7% of the control plants, respectively. Moreover, chlorophyll b was more sensitive to the Cd stress than chlorophyll a. The increased proline content in the leaf blade of maize seedlings resulted from external Cd stress indicates that maize can adapt to the adversity menace via changing the content of proline.  相似文献

Excitation energy transfer in the light-harvesting complex II of higher plants is modeled using excitonic couplings and local transition energies determined from structure-based calculations recently (Müh et al., 2010). A theory is introduced that implicitly takes into account protein induced dynamic localization effects of the exciton wavefunction between weakly coupled optical and vibronic transitions of different pigments. Linear and non-linear optical spectra are calculated and compared with experimental data reaching qualitative agreement. High-frequency intramolecular vibrational degrees of freedom are found important for ultrafast subpicosecond excitation energy transfer between chlorophyll (Chl) b and Chla, since they allow for fast dissipation of the excess energy. The slower ps component of this transfer is due to the monomeric excited state of Chlb 605. The majority of exciton relaxation in the Chla spectral region is characterized by slow ps exciton equilibration between the Chla domains within one layer and between the lumenal and stromal layers in the 10-20 ps time range. Subpicosecond exciton relaxation in the Chla region is only found within the terminal emitter domain (Chls a 610/611/612) and within the Chla 613/614 dimer. Deviations between measured and calculated exciton state life times are obtained for the intermediate spectral region between the main absorbance bands of Chla and Chlb that indicate that besides Chlb 608 another pigment should absorb there. Possible candidates, so far not identified by structure-based calculations, but by fitting of optical spectra and mutagenesis studies, are discussed. Additional mutagenesis studies are suggested to resolve this issue.  相似文献

The in-chlorophyll centre waveband (ICCW) (640–680 nm) is the specific chlorophyll (Chl) absorption band, but the reflectance in this band has not been used as an optimal index for non-destructive determination of plant Chl content in recent decades. This study develops a new spectral index based solely on the ICCW for robust retrieval of leaf Chl content for the first time. A glasshouse experiment for solution-culture of one chlorophyll-deficient rice mutant and six wild types of rice genotypes was conducted, and the leaf reflectance (400–900 nm) was measured with a high spectral resolution (1 nm) spectrophotometer and the contents of chlorophyll a (Chla), chlorophyll b (Chlb) and chlorophyll a+b (Chlt) of the rice leaves were determined. It was found that the reflectance curves from 640 nm to 674 nm and from 675 nm to 680 nm of the low-chlorophyll mutant leaf were drastically steeper than that of the wild types in the ICCW. The new index based on the reflectance variation within ICCW, the difference of the first derivative sum within the ICCW (DFDS_ICCW), was highly sensitive (r = −0.77, n = 93, P<0.01) to Chlt while the mean reflectance (R_ICCW) in the ICCW became insensitive (r = −0.12, n = 93, P>0.05) to Chlt when the leaf Chlt was higher than 200 mg/m2. The best equations of R-ICCW and DFDS_ICCW yielded an RMSE of 78.7, 32.9 and 107.3 mg/m2, and an RMSE of 37.4, 16.0 and 45.3 mg/m−2, respectively, for predicting Chla, Chlb and Chlt. The new index could rank in the top 10 for prediction of Chla and Chlt as compared with the 55 existing indices. Additionally, most of the 55 existing Chl-related VIs performed robustly or strongly in simultaneous prediction of leaf Chla, Chlb and Chlt.  相似文献

Eight chlorophyll-protein complexes were isolated from thylakoid membranes of a Codium species, a marine green alga, by mild SDS-polyacrylamide gel electrophoresis. CP 1a1, CP 1a2, CP 1a3 and CP 1a4 were partially dissociated Photosystem (PS) I complexes, which in addition to the core reaction centre complex, CP 1, possessed PS I light-harvesting complexes containing chlorophyll (Chl) a, Chl b and siphonaxanthin. LHCP1 and LHCP3 are orange-brown green chlorophyll ab-proteins (Chl aChl b ratios of 0.66) that contain siphonaxanthin and its esterified form, siphonein. CP a and CP 1, the core reaction centre complexes of PS II and PS I, respectively, had similar spectral properties to those isolated from other algae or higher plants. These P-680- or P-700-Chl a-proteins are universally distributed among algae and terrestrial plants; they appear to be highly conserved and have undergone little evolutionary adaptation. Siphonaxanthin and siphonein which are present in the Codium light-harvesting complexes of PS II and PS I are responsible for enhanced absorption in the green region (518 and 538 nm). Efficient energy transfer from both xanthophylls and Chl b to only Chl a in Codium light-harvesting complexes, which have identical fluorescence emission spectra at 77 K to those of the lutein-Chl ab-proteins (Chl aChl b ratios of 1.2) of most green algae and all higher plants, proved that the molecular arrangement of these light-harvesting pigments was maintained in the isolated Codium complexes. The siphonaxanthin-Chl ab-proteins allow enhanced absorption of blue-green and green light, the predominant light available in deep ocean waters or shaded subtidal marine habitats. Since there is a variable distribution of lutein, siphonaxanthin and siphonein in marine green algae and siphonaxanthin is found in very ancient algae, these novel siphonein-siphonaxanthin-Chl ab-proteins may be ancient light-harvesting complexes which were evolved in deep water algae.  相似文献

Effect of grain soaking presowing in 1 mM salicylic acid (SA) and NaCl (0, 50, 100, 150 and 200 mM) on barley (Hordeum vulgare cv Gerbel) was studied. Increasing of NaCl level reduced the germination percentage, the growth parameters (fresh and dry weight), potassium, calcium, phosphorus and insoluble sugars content in both shoots and roots of 15-day old seedlings. Leaf relative water content (RWC) and the photosynthetic pigments (Chl a, b and carotenoids) contents also decreased with increasing NaCl concentration. On the other hand, Na, soluble sugars, soluble proteins, free amino acids including proline content and lipid peroxidation level and peroxidase activity were increased in the two plant organs with increasing of NaCl level. Electrolyte leakage from plant leaves was found to increase with salinity level. SA-pretreatment increased the RWC, fresh and dry weights, water, photosynthetic pigments, insolube saccharides, phosphorus content and peroxidase activity in the stressed seedlings. On the contrary, Na+, soluble proteins content, lipid peroxidation level, electrolyte leakage were markedly reduced under salt stress with SA than without. Under stress conditions, SA-pretreated plants exhibited less Ca2+ and more accumulation of K+, and soluble sugars in roots at the expense of these contents in the plant shoots. Exogenous application (Grain soaking presowing) of SA appeared to induce preadaptive response to salt stress leading to promoting protective reactions to the photosynthetic pigments and maintain the membranes integrity in barley plants, which reflected in improving the plant growth.  相似文献

In three separate experiments, the effectiveness of a SPAD-502 portable chlorophyll (Chl) meter was evaluated for estimating Chl content in leaves of Eugenia uniflora seedlings in different light environments and subjected to soil flooding. In the first experiment, plants were grown in partial or full sunlight. In the second experiment plants were grown in full sunlight for six months and then transferred to partial sunlight or kept in full sunlight. In the third experiment plants were grown in a shade house (40% of full sunlight) for six months and then transferred to partial shade (25–30% of full sunlight) or full sunlight. In each experiment, plants in each light environment were either flooded or not flooded. Non-linear regression models were used to relate SPAD values to leaf Chl content using a combination of the data obtained from all three experiments. There were no significant effects of flooding treatments or interactions between light and flooding treatments on any variable analyzed. Light environment significantly affected SPAD values, chlorophyll a (Chl a), chlorophyll b (Chl b), and total chlorophyll [Chl (a+b)] contents in Experiment I (p≤0.01) and Experiment III (p≤0.05). The relationships between SPAD values and Chl contents were very similar among the three experiments and did not appear to be influenced by light or flooding treatments. There were high positive exponential relationships between SPAD values and Chl (a+b), Chl a, and Chl b contents.  相似文献

The relationship between the accumulation of Chl and the apoproteinsof the light-harvesting Chl a/b-protein complex of PS II (LHCII)during the greening of cucumber cotyledons was studied. LHCIIapoproteins were not detected in etiolated cotyledons. Uponillumination, Chl a was formed as a result of photoconversionof protochlorophyllide (Pchlide) which had accumulated in thedark. During the lag period that preceded the accumulation ofChl, a small amount of LHCII apoproteins appeared. The amountof LHCII apoproteins increased with increases in levels of Chlb, though somewhat more rapidly during the first 10 h of greening.Treatment with benzyladenine (BA) or levulinic acid (LA) wasused to vary the supply of Chl a for apoproteins by promotingor inhibiting the synthesis of Chl a, respectively. LA decreasedbut BA increased the rate of accumulation of Chl b and LHCIIapoproteins. Only small amounts of Chl b and LHCII apoproteinswere formed under intermittent illumination. However, in thepresence of chloramphenicol (CAP), which inhibits the synthesisof plastome-coded proteins including apoproteins of the P700-Chla-protein complex (CP1) and a Chl a-protein complex of PS II(CPa), we observed the accumulation of Chl b and LHCII apoproteins,both of which are of nuclear origin. During incubation in thedark after intermittent exposure to light, CAP alone allowedneither destruction nor accumulation of Chl b and LHCII apoproteins,but it did enhance the effect of CaCl2 in inducing both Chlb and these apoproteins. These results can be explained by assumingthat apoproteins of CP1 and CPa have a higher affinity for Chla than do LHCII apoproteins. When the availability of Chl ais limited, these apoproteins compete with one another for Chla, with the resultant preferential formation of CP1 and CPa.However, when the supply of Chl a becomes large enough for saturationof apoproteins of CP1 and CPa, some of the Chl a is incorporatedinto LHCII apoproteins either directly or after conversion toChl b. Enhanced ultraviolet-B (UVB) radiation and water deficit affect plant growth and development. We determined the effects of UVB and drought stress on growth parameters and chemical attributes of two ecotypes (alpine and prairie) of Stellaria longipes under controlled-environment conditions. Clonal ramets of these ecotypes were grown under three UVB levels (0, 5, and 10 kJ m−2 d−1) and exposed to two watering regimes (well watered and drought stressed) for 21 days. Compared to the alpine, the prairie ecotype was taller, had higher number of nodes, and greater leaf area and specific leaf weight (leaf dry weight: leaf area), which resulted in increased dry matter in this ecotype. Overall, ‘prairie’ was higher in total chlorophyll (Chl), but lower in Chla:b ratio, flavonoids, and ethylene, than ‘alpine’. In both ecotypes, UVB and drought stress reduced growth and dry matter, whereas UVB increased carotenoids and flavonoids. Drought stress decreased ethylene evolution. These characteristics were also determined in plants growing in the field. In the field-growing plants, ‘prairie’ had higher growth and dry matter, but lower Chla:b ratio and flavonoids, than ‘alpine’. The two ecotypes responded differentially to UVB and watering regime, as ‘prairie’ appeared to be more sensitive to UVB and drought stress than ‘alpine’.  相似文献

This study was aimed to assess physiological responses of melon (Cucumis melo L.) cultivars to salinity stress under field conditions. Seventeen melon cultivars including 16 widely distributed native and one exotic (‘Galia’) were subjected to 2-year (2014–2015) field salinity stress. Leaf relative water content (RWC), membrane stability index (MSI), pigments [chlorophyll a, b, total chlorophyll (TChl), carotenoid (Car) and their ratios], malondialdehyde (MDA), H2O2 content, proline content (Pro), total soluble sugar content (TSC), salinity tolerance and susceptibility indices as well as yield were evaluated. The results of combined analysis of variance showed significant genotypic variation for all the traits and significant effect of salinity stress on all the traits with the exception of Chla/Chlb and TChl/Car ratios. Overall, field salinity stress caused an increase in leaf MDA, H2O2, Chla, Chlb, TChl, Car, Pro and TSC and caused a reduction in leaf MSI and RWC as well as yield. The results of correlation coefficients showed that accumulation of osmolytes (proline and TSC) led to an increase in RWC and a decrease in MDA contents. In addition, the results of multiple regression analysis showed that leaf MDA, TSC, MSI and Chla contents were the most important predictors of yield justifying 72% total variation of yield under saline conditions. These results may highlight a dynamic interplay among biomarkers for lipid peroxidation (MDA), sugar osmolytes (TSC) and photosynthetic pigment (Chla) to maintain cell viability and cell wall integrity under salinity stress conditions in melon.  相似文献

In order to study chloroplast biogenesis, we chose natural variegated Epipremnum aureum (golden pothos) and regenerated pale yellow, variegated and green plants from all three types of tissue explants. The percentage of three types of regenerated shoots from three different explants was very close. Regenerated plants have been maintained for a year and show no sign of a colour switch. By comparing their protein profiles, two major differences between pale yellow and green plants were observed at the 15 and 40 to 50 kDa proteins. Moreover, pale yellow plants had unexpected high molecular mass proteins (greater than 60 kDa). Both variegated and green plants had more chlorophyll (Chl) a than Chl b, the ratios were about 1.46 and 1.93, respectively. In contrast, the pale yellow plants not only had less total Chl, but also the reduction of Chl a was much greater than Chl b, resulting in a higher content of Chl b than Chl a. Microscopic analysis revealed that pale yellow plants contained predominantly undeveloped chloroplasts with low Chl contents, even though their mesophyll cells were similar to green and variegated plants. PCR amplification of chloroplast DNA with 14 universal chloroplast primers did not reveal any difference among these regenerated plants.  相似文献

Salicylic acid (SA) and nitric oxide (NO) are reported to alleviate the damaging effects of stress in plants rather similarly when applied at appropriate low concentrations. An experiment was therefore conducted to study the impact of SA, sodium nitroprusside (SNP; as NO donor), and methylene blue (MB; as a guanylate cyclase inhibitor) on wheat seedling performance under osmotic stress. Osmotic stress significantly reduced shoot fresh weight (SFW), chlorophyll contents (Chla, Chlb, total Chl), and membrane stability index (MSI) and also increased malondialdehyde (MDA) level, lipoxygenase (LOX) activity, and hydrogen peroxide production. Moreover, enzymatic antioxidant activities including superoxide dismutase, guaiacol peroxidase, and glutathione reductase activity were enhanced under osmotic stress. On the contrary, SA or SNP pretreatment reduced the damaging effects of osmotic stress by further enhancing the antioxidant activities that led to increased SFW, Chl, and MSI and reduced MDA level and LOX activity. However, pretreatment of plants with MB reversed or reduced the protective effects of SA and SNP suggesting that the protective effects were likely attributed to NO signaling. Therefore, NO may act as downstream of SA signaling in reduction of induced oxidative damage in wheat seedlings.  相似文献

The chlorophyll (Chl) a-b light harvesting complex II (LHC II)contains more than 80% of the light-harvesting pigments of photosystemII (PS II) in chloroplasts. The supramolecular assembly andfunction of this auxiliary antenna system was investigated inChi b-deficient and Chi b-less mutant chloroplasts from soybeanand barley plants, and in their wild-type counterparts. Fourdistinct LHC II polypeptides were resolved by SDS-PAGE (subunitsa, b, c and d), having apparent molecular masses of 29, 28,27.2 and 26.8 kDa, respectively. The analysis of LHC II subunitcomposition in different developmental stages of the PS II unitin soybean (3>Chla/Chlbb>6), indicated the associationof specific subunits with the LHC H-inner and LHC II-peripheralin the chloroplast. The amount of subunit a in PS II was constantover a broad range of Chl a/Chl b ratios, suggesting that thissubunit is closely associated with the PS II-core complex. Subunitd also appeared to be constant over a wide range of Chl a/Chlb ratios, suggesting close association with the LHC II-inner.The PS II content in subunits b and c increased with the PSII antenna development in soybean but the ratio of b/c remainedconstant in all developmental stages and equal to 2 :1. Subunita was present in the Chl b-less chlorina f2 mutant of barleygrown under continuous illumination but was absent under intermittentillumination. The results suggest that each subunit binds 13-15Chl molecules. A working hypothesis is presented on the PS IIantenna development and LHC II subunit composition in soybeanchloroplasts. Direct effects and after-effects of soil drought for 7 and 14 d were examined on seedling dry matter, leaf water potential (ψ), leaf injury index (LI), and chlorophyll (Chl) content of drought (D) resistant and sensitive triticale and maize genotypes. D caused higher decrease in number of developed leaves and dry matter of shoots and roots in the sensitive genotypes than in the resistant ones. Soil D caused lower decrease of ψ in the triticale than maize leaves. Influence of D on the Chl b content was considerably lower than on the Chl a content. In triticale the most harmful D impact was observed for physiologically younger leaves, in maize for the older ones. A period of 7-d-long recovery was too short for a complete removal of an adverse influence of D.  相似文献

With the use of low temperature spectrofluorometry and matrix calculations it was demonstrated that the chlorophyll a pool of higher plants is made up of four different chlorophyll a chromophores. The latter were segregated by high pressure liquid chromatography on a silica column. They were designated Chl a (E432 F664), Chl a (E436 F670), Chl a (E443 F672) and Chl a (E446 F674), where E refers to the Soret excitation maximum and F to the fluorescence emission maximum at 77 K in ether. Likewise the Chl b pool was shown to consist of at least four different Chl b chromophores which were designated: Chl b (E465), Chl b (E470), Chl b (E475) and Chl b (E485). It was proposed that the various chlorophyll chromophores differed by the degree of oxidation of their side chains at the 2 and 4 positions of the macrocycle. It was also suggested that the chemical modifications at the 2 and 4 positions of the macrocycle may play an important role in positioning the different chlorophyll chromophores in the thylakoid membranes.  相似文献
