The effects of long term salinity on some morpho-physiological characteristics were studied in strawberry Kurdistan and Queen elisa cultivars. Vegetative and biochemical traits were measured in strawberry cultivars subjected to three levels of salinity including 0, 40 and 80 mM at 20, 40 and 60th days after NaCl addition. Results showed that in both cultivars the dry weight of plant organs decreased in response to NaCl, except of crown weight in cv. Kurdistan. Root to shoot ratio increased due to a greater reduction in above ground biomass under salinity. Strawberry cultivars tended to decrease their stomatal conductance, RWC, proline, soluble carbohydrates and proteins during the different evaluation periods. Compared to the 20th day, peroxidase activity decreased at 80 mM during 40 and 60 days in cv. Queen elisa. On the contrary, ascorbate peroxidase activity elevated until the 40th day and decreased afterwards, in addition application of 40 and 80 mM NaCl increased the ascorbate peroxidase activity of both studied cultivars. Catalase activity increased from 20th until 60th days in cv. Queen elisa, while showed increase in cv. Kurdistan until day 40 and then decreased again at day 60. Application of 40 and 80 mM NaCl resulted in an increase in peroxidase, ascorbate peroxidase and catalase activities of both cultivars. The Queen elisa cv. showed lower tolerance index (45.88%) compared with cv. Kurdistan (67.97%). Finally, higher salinity resistance of cv. Kurdistan is probably associated with its ability to maintain higher RWC and higher activity of antioxidant enzymes.  相似文献

Intra-specific variations in nonstructural carbohydrates and free proline were determined in leaves, apices, roots, and maturing seeds of two salt-tolerant cultivars (CR and Kharchia-65) and one salt-sensitive cv. Ghods of spring wheat (Triticum aestivum L.) grown in sand culture at various levels of salinity (0, 100, 200, and 300 mM NaCl and CaCl2 at 5 : 1 molar ratio) under controlled environmental conditions. The levels of leaf, apex, and root ethanol-soluble carbohydrates, fructans, starch, and proline increased in line with elevating level of salinity in all three cultivars under investigation. The contents of proline, soluble and insoluble carbohydrates in the apex increased to levels exceeding those in the leaves and roots. Soluble carbohydrate content of salt-sensitive cv. Ghods was higher in the leaves, apices, and roots and lower in the maturing seeds than in the other cultivars at all levels of salinity except at 300 mM. The results show considerable variation in the amount of soluble, insoluble sugars, and proline among plant tissues and wheat genotypes in response to salinity. Higher soluble carbohydrates and fructan in leaves, roots and maturing seeds of stressed plants indicate that their accumulation may help plant to tolerate salinity. Salt-sensitive cv. Ghods accumulated less soluble sugars in the maturing seeds and higher soluble sugars in the apices, which might be used as an indicator in screening wheat genotypes for salinity tolerance.  相似文献

Proline content, ion accumulation, cell wall and soluble peroxidase activities were determined in control and salt-treated calli (150 nM NaCl) and whole plants (30 mM NaCl) of two rice cultivars (salt sensitive cv. IKP and salt tolerant cv. Aiwu). Under salinity, the highest accumulation of Na+, Cl? and proline occurred in calli, roots and younger leaves of cv. IKP, coupled with the highest decrease in K+ content; accumulations of Na+ and Cl? were restricted to older leaves in cv. Aiwu. Relative growth rates of calli and roots or shoots from both cultivars were not linked to peroxidase activities. High concentrations (1 M) of exogenously applied glycerol did not inhibitin vitro activities of soluble peroxidase extracted from control and salt-treated calli or plants. Conversely, 35–55% (in cv. IKP) or 60–80% (in cv. Aiwu) of soluble peroxidase activities were found in presence of isosmotic proline concentration. There were no differences between proline and glycerol effects onin vitro cell wall peroxidase activities.  相似文献

Mungbean (Vigna radiata L. Wilczek cv. Sujata and cv. K851) seedlings were grown in paper towelins in dark under 0, 0.5, 1, 2 and 3 % (m/v) NaCl salinity. Germination percentage, shoot and root length, fresh mass of both cultivars decreased with salinity. Total soluble saccharides and proline accumulated in the root and shoot of salt stressed seedlings. The proline accumulation in the root was four to five times higher than that of the shoot of NaCl treated etiolated mungbean seedlings.  相似文献

The present study pertains to the effect of different concentration of NaCl on the contents of proteins, free amino acids, proline and glycine betaine in leaves, stems and roots of Ipomoea pes-caprae. The protein content of the tissues increased in response to salinity upto 200 mM NaCl; the free amino acids content showed a reversal trend. The proline and glycine betaine contents increased with increasing salinity upto 500 mM NaCl. The accumulation of proline and glycine betaine might play a role in the alleviation of salt stress. Effects of NaCl on growth in vitro and contents of sugars, free proline and proteins in the seedlings and leaf explants of Nicotiana tabacum cv. Virginia were investigated. The fresh and dry mass of the seedlings decreased under salinity. These growth parameters in leaf explants decreased at 50 mM NaCl and increased up to 150 mM NaCl and then decreased at higher level of salinity. Free proline content in both seedlings and leaf explants increased and polysaccharide content decreased continuously with increasing of NaCl concentration. Reducing sugars, oligosaccharides, soluble sugars and total sugars contents in both seedlings and leaf explants decreased up to 150 mM NaCl and then increased at higher concentrations of NaCl.  相似文献

The physiological response ofScenedesmus obliquus to salinity (NaCl concentration of 40, 80, 120, 60 and 200 mM) for 7 d (long-term experiments) or 2 h (short-term experiments) was followed. Cell number, dry matter and the content of photosynthetic pigments decreased with the rise of NaCl concentration. However, the photosynthetic O2 evolution mostly increased with the increase of NaCl concentration up to 80 mM, and respiration (dark O2 uptake) was markedly promoted. Photosynthesis/respiration ratio went in concomitance with the cell number, dry matter or chlorophyll content. Contents of soluble saccharides and soluble proteins increased with the rise of salinization, while the content of insoluble and total saccharides or proteins decreased. Proline content increased greatly with salinization, whereas of other free amino acids were mostly reduced, especially at higher salinities. Similarly, the lipoid contents of salinizedScenedesmus obliquus were obviously higher than those of the control cultures.  相似文献

The interactive effects of salinity stress (40, 80, 120 and 160 mM NaCl) and ascorbic acid (0.6 mM), thiamin (0.3 mM) or sodium salicylate (0.6 mM) were studied in wheat (Triticum aestivum L.). The contents of cellulose, lignin of either shoots or roots, pectin of root and soluble sugars of shoots were lowered with the rise of NaCl concentration. On the other hand, the contents of hemicellulose and soluble sugars of roots, starch and soluble proteins of shoots, proline of either shoots or roots, and amino acids of roots were raised. Also, increasing NaCl concentration in the culture media increased Na+ and Ca2+ accumulation and gradually lowered K+ and Mg2+ concentration in different organs of wheat plant. Grain soaking in ascorbic acid, thiamin or sodium salicylate could counteract the adverse effects of NaCl salinity on the seedlings of wheat plant by suppression of salt stress induced accumulation of proline.  相似文献

Salt stress-induced changes in antioxidant enzymes, lipid peroxidation, proline and glycine betaine contents, and proline-metabolizing enzymes were examined in the leaves of two mulberry cultivars (Local and Sujanpuri). With increasing salinity up to 150 mM NaCl, superoxide dismutase, catalase, ascor-bate peroxidase, guaiacol peroxidase, glutathione reductase, and monodehydroascorbate reductase activities were increased in both cultivars as compared to control, but more pronounced increase was observed in cv. Local. Salt stress enhanced the rate of lipid peroxidation (as indicated by increasing MDA content) in both cultivars. Under NaCl stress, cv. Local showed less change in the MDA content than cv. Sujanpuri. Salt stress resulted in a significant accumulation of free proline in mulberry leaves, and more accumulation was detected in cv. Local than cv. Sujanpuri. The leaves of cv. Local showed 9-fold accumulation of glycine betaine in comparision with cv. Sujanpuri after 20 days at 150 mM NaCl. A decrease in proline oxidase activity and an increase in γ-glutamyl kinase activity were observed with increasing NaClconcentration. The relative water content and electrolyte leakage also decreased after increasing the NaCl concentration, but a decrease was more pronounced in cv. Sujanpuri than in cv. Local. The results indicate that oxidative stress may play an important role in salt-stressed mulberry plants and cv. Local have more efficient antioxidant characteristics, which could provide for a better protection against oxidative stress.  相似文献

During the germination, wheat and sorghum plants tolerated salinity up to 10 mM NaCl. Seedling growth and dry matter production remained more or less unchanged up to 10 mM NaCl in case of wheat and up to 5 mM NaCl in case of sorghum. The water content of test plant species exhibited nearly constant values irrespective of the salinity level applied. The proline and carbohydrate content increased with elevating NaCl, while free amino acids content decreased. Irrigating of seedlings with tryptophan had non significant effect at all salinity levels used.  相似文献

Seeds of three Saudi pearl millet cultivars (Pennisetum spicatum) from three regions (Madinah, Khulais and Jaizan) were inoculated with arbuscular mycorrhizal fungus Glomus mosseae obtained from the Agriculture Research Center of Giza, Egypt to enhance their salt tolerance. Five different NaCl concentrations (0, 30, 60, 90, and 120 mM) were used for treating cultivars with and without mycorrhiza. Growth rates, chlorophyll content, chlorophyll fluorescence (Fv/Fm), proline content and gas exchange were measured to determine the effect of salinity on these cultivars. The results indicated that compared to cultivars without mycorrhiza, all cultivars with mycorrhiza had enhanced growth and physiological parameters including shoot and root length, area and number of leaves, fresh and dry weights of shoots and roots, chlorophyll contents and gas exchanges at 0 and 30 mM of salinity. In addition, the measurements of the different growth rates showed higher growth performance of the cultivars from Madinah and Khulais than the cultivar from Jaizan. However, all cultivars with and without mycorrhiza showed significant reductions in growth rates, chlorophyll contents and gas exchanges at a salinity of 60 mM than those grown at 0 and 30 mM. Moreover, the values of Fv/Fm were significantly reduced in all cultivars with and without mycorrhiza grown at 60 mM than in those grown at 0 mM and 30 mM. Proline contents indicated a progressive increase with the elevation of NaCl concentration stress. The proline contents in the mycorrhiza-inoculated cultivars were significantly higher than those in the non-inoculated cultivars. On the other hand, all cultivars with and without mycorrhiza underwent senescence within four weeks of growth at salinity concentrations of 90 mM and 120 mM. Therefore, relatively low salinity must be maintained to achieve high growth rates and gas exchanges of these inoculated cultivars.  相似文献

Changes in the antioxidant defense system and proline accumulation were examined at different growth stages (vegetative, boot and reproductive) in plants of two hexaploid spring wheat cultivars (S-24, salt tolerant; MH-97, salt sensitive), grown in hydroponics and salinity-affected with 0, 50, 100 and 150 mM of NaCl. Salt stress provoked a marked decline in plant dry mass and ascorbic acid contents, and increased proline, total soluble proteins and H2O2 contents in both wheat cultivars at all growth stages. However, higher accumulation of proline and H2O2 contents was recorded at the vegetative and boot stages, respectively, in both wheat cultivars. Salt stress caused a consistent rise in the activities of some key antioxidant enzymes (CAT, SOD, POD, and APX) at all growth stages only in the salt tolerant cultivar S-24, whereas such pattern of enhanced activities of enzymatic antioxidants in cv. MH-97 was found only at the vegetative stage under saline regimes. Maximum activities of various enzymatic antioxidants were observed at the vegetative stage in both wheat cultivars under varying external salt treatments. The results showed that high salinity tolerance of cv. S-24, as manifested by lower decrease in its dry matter under salt stress, was associated with higher activities of antioxidant enzymes, increased accumulation of proline, and lower levels of H2O2, as compared with cv. MH-97 at all growth stages under saline regimes.  相似文献

Stress tolerance of two Egyptian cotton cultivars (Gossypium barbadense L.) (Giza 45 and Giza 86) exposed to various doses (40, 80, 160, and 320 min) of artificial ultraviolet-A (UV-A) radiation (366 nm) was investigated. Seed germination of cv. Giza 86 was promoted at 40 min, progressively inhibited at 80 and 160 min, and completely suppressed at 320 min irradiation. However, seed germination of cv. Giza 45 was not promoted but inhibited by UV-A light and stopped at the dose of 160 min. In contrast to seed germination, seedling growth of cv. Giza 86 was negatively stressed even at 40 min-dose. UV-A radiation reduced leaf carbohydrate content and shoot growth of both cultivars, but the response was comparatively higher in cv. Giza 45. UV-A radiation decreased chlorophyll (Chl) and carotenoid contents in parallel with an increase in the Chl a/b ratio, diminished Hill reaction activity, and quenched Chl a fluorescence independent of the presence of 3-(3,4-dichlorophenyl)-1,1-dimethylurea, suggesting an inhibitory effect on both the water-splitting system and electron transport from the primary to the secondary acceptors of photosystem II (PSII) (acceptor side). UV-A radiation also decreased the ratio of unsaturated to saturated fatty acids in thylakoid membranes, thus indicating that the inhibition of PSII activity was followed by lipid peroxidation and changes in thylakoid membrane fluidity. These changes reflect the disturbance of structure, composition, and functioning of the photosynthetic apparatus, as well as the sensitivity of PSII to UV-A stress. Both cultivars developed adaptive mechanisms for damage alleviation involving the accumulation of flavonoids, total lipids, and total soluble proteins, as well as development of smaller and thicker leaf blades. Since cv. Giza 86 showed comparatively higher level of adaptation, it tolerates UV-A stress more successful than cv. Giza 45.  相似文献

Soybean (Glycine max) cv. Clark tolerated NaCl salinity up to osmotic potential in soil -1800 kPa, cv. Forest to -1500 kPa and cv. Kint to -1200 kPa. The tolerance of cvs. Clark and Forest was found to be related to the accumulation of soluble proteins, amino acids, proline, K and Ca. The sensitivity of cv. Kint was found to be associated with decrease in water, saccharide, protein, K and Ca contents. The leaf osmotic potential decreased in cvs. Clark and Forest while remained unchanged in cv. Kint. Effect of grain soaking presowing in 1 mM salicylic acid (SA) and NaCl (0, 50, 100, 150 and 200 mM) on barley (Hordeum vulgare cv Gerbel) was studied. Increasing of NaCl level reduced the germination percentage, the growth parameters (fresh and dry weight), potassium, calcium, phosphorus and insoluble sugars content in both shoots and roots of 15-day old seedlings. Leaf relative water content (RWC) and the photosynthetic pigments (Chl a, b and carotenoids) contents also decreased with increasing NaCl concentration. On the other hand, Na, soluble sugars, soluble proteins, free amino acids including proline content and lipid peroxidation level and peroxidase activity were increased in the two plant organs with increasing of NaCl level. Electrolyte leakage from plant leaves was found to increase with salinity level. SA-pretreatment increased the RWC, fresh and dry weights, water, photosynthetic pigments, insolube saccharides, phosphorus content and peroxidase activity in the stressed seedlings. On the contrary, Na+, soluble proteins content, lipid peroxidation level, electrolyte leakage were markedly reduced under salt stress with SA than without. Under stress conditions, SA-pretreated plants exhibited less Ca2+ and more accumulation of K+, and soluble sugars in roots at the expense of these contents in the plant shoots. Exogenous application (Grain soaking presowing) of SA appeared to induce preadaptive response to salt stress leading to promoting protective reactions to the photosynthetic pigments and maintain the membranes integrity in barley plants, which reflected in improving the plant growth.  相似文献

Two contrasting maize (Zea mays L.) cultivars, i.e., Giza 2 (salt tolerant) and Trihybrid 321 (salt sensitive), were grown hydroponically to study NaCl effect (100 mM) on root plasma membrane (PM) lipid and protein alterations. The PM total sterols of Trihybrid 321 were decreased while that of Giza 2 was increased in response to salt. Salt imposition had no significant effect on PM total glycolipids and proteins of both cultivars. The PM total phospholipids were increased in Trihybrid 321 but it did not change significantly in Giza 2 after salinity stress. Molecular percentage of PM phospholipids and fatty acids of both cultivars was different in absence (0 mM) and presence (100 mM) of salt. The most abundant phospholipids in untreated Trihybrid 321 PM were phosphatidylglycerol (PG), phosphatidylcholine (PC), phosphatidylethanolamine (PE) and phosphatidylserine (PS), which changed into PG, PS, phosphatidylinositol (PI) and PC after salt treatment. However, the dominant phospholipids of the control PM of Giza 2 were PC, PE, PS and PG, which changed into PG, PE, PS and diphosphatidylglycerol (DPG) after salt imposition. Over 60% of the total fatty acids were saturated in control and salinized PM of both cultivars, which was increased after salt stress. The predominant fatty acid in the control and salinized PM of Trihybrid 321 was C18:1 and C17:0, respectively. However, in control and treated PM of Giza 2, the predominant fatty acid was C17:0 and C20:0, respectively. Qualitative and quantitative differences in PM protein patterns were found in both cultivars with and without salt. PM lipid changes enhanced membrane integrity, reflected in different ion accumulation (Mansour et al. 2005), and hence salt tolerance of Giza 2.  相似文献

Salinity tolerance of sugar beet (Beta vulgaris L.) cultivars in terms of growth, proline and soluble sugars concentrations, and Na+/K+ and Na+/Ca2+ ratios were analyzed in this study. Three-week-old seedlings of three sugar beet cultivars, ‘Gantang7’, ‘SD13829’, and ‘ST21916’, differing in salinity tolerance, were treated with 0, 50, 100, and 200 mM NaCl. Plant shoots and roots were harvested at 7 days after treatment and subjected to analysis. Low concentration of NaCl (50 mM) enhanced fresh and dry weights of shoot and root in ‘Gantang7’, whereas high one (200 mM) reduced growth in all cultivars and the less reduction was observed in ‘ST21916’. Shoot proline was strongly induced by salinity stress in both ‘Gantang7’ and ‘SD13829’, while it remained unchanged in ‘ST21916’. The addition of 50 mM NaCl significantly increased shoot soluble sugars concentrations in ‘Gantang7’ while it had no significant effects in the other two cultivars. ‘Gantang7’ also showed a higher level of root soluble sugars concentration as compared to the other two cultivars. At 50 mM NaCl, the lower shoot Na+ concentration, and the higher shoot K+ and root Ca2+ concentration in ‘Gantang7’ resulted in the lower shoot Na+/K+ and root Na+/Ca2+ ratio. However, ‘SD13829’ maintained a lower Na+/K+ ratio in both shoot and root when subjected to 200 mM NaCl treatment. According to comprehensive evaluation on salinity tolerance, it is clear that ‘Gantang7’ is more tolerant to salinity than the other two cultivars. Therefore, it is suggested that ‘Gantang7’ should be more suitable for cultivating in the arid and semi-arid irrigated regions.  相似文献

The present study was carried out to examine the effects of seed soaking in 1 mM ascorbic acid (AA) or 1 mM proline on the growth, content of photosynthetic pigments and proline, relative water content, electrolyte leakage, antioxidant enzymes and leaf anatomy of Hordeum vulgare L. Giza 124 seedlings grown in greenhouse under 100 or 200 mM NaCl. The plants exposed to the NaCl stress exhibited a significant reduction in growth, relative water content, leaf photosynthetic pigments, soluble sugars, as well as alterations in leaf anatomy. However, the treatment with AA or proline ameliorated the stress generated by NaCl and improved the above mentioned parameters. NaCl increased electrolyte leakage, proline content, and activities of antioxidant enzymes (SOD, CAT, and POX). The antioxidant enzymes and leaf anatomy exhibited considerable changes in response to AA or proline application in the absence or presence of NaCl.  相似文献

The lengths of roots and shoots, fresh and dry matter yield, and the contents of insoluble saccharides and free amino acids were reduced with the rise in NaCl concentration. However, under combination of NaCl with Ca2+ ions, these parameters generally raised. Contents of soluble saccharides, proline and quaternary ammonium compounds increased with increasing NaCl concentration, but under addition of CaCl2 or CaSO4, contents of these compounds were decreased. Low concentrations of NaCl stimulated soluble proteins, production, but higher concentrations decreased the content of soluble proteins. Addition of Ca2+ in the media did not improve the soluble protein production. Insoluble proteins content was increased with the rise of salinity level, but these effects were more pronounced with NaCl and CaCl2 or CaSO4 than with NaCl only.  相似文献
