【目的】为了研究青草沙水库中土著微生物对藻类生长的抑制作用,从水库水体中筛选出对藻类有抑制作用的细菌并研究其对铜绿微囊藻的抑制效果。【方法】通过对水库水体中的细菌进行划线分离和筛选,挑选出一株对铜绿微囊藻生长有较好抑制作用的菌株CL。考察其对铜绿微囊藻的抑制效果及不同培养时间和菌液浓度对抑藻效果的影响,并对菌株进行16S rDNA序列分析。【结果】实验菌液浓度为4.5×108CFU/mL 8.4×108CFU/mL时,细菌对铜绿微囊藻的抑制率可达45.4%。抑藻效果随培养时间先增后降,在静置培养第6天抑藻效果达到最大。该菌经过16S rDNA序列分析,属于黄单胞菌科的寡养单胞菌。【结论】从青草沙水库中筛选出了对铜绿微囊藻有抑制作用的土著细菌寡养单胞菌,对青草沙水体铜绿微囊藻的控制具有一定的潜在应用价值。  相似文献

文章开展了25℃下两个品系铜绿微囊藻(有毒与无毒)不同浓度对萼花臂尾轮虫生活史的影响研究, 及在5个温度下不同浓度有毒铜绿微囊藻对萼花臂尾轮虫生活史影响的研究。结果表明铜绿微囊藻毒性、浓度及二者交互作用对轮虫生活史参数净生殖率(R0; F=31.83, P<0.01; F=30.36, P<0.01; F=13.51, P<0.01)、内禀增长率(rm; F=34.67, P<0.01; F=18.73, P<0.01; F=12.99, P<0.01)均有显著影响; 温度、铜绿微囊藻浓度及二者交互作用对轮虫生活史参数净生殖率、内禀增长率也均有显著影响。无毒铜绿微藻在低浓度(1×104 cells/mL)下对轮虫种群有促进作用, 可作为轮虫食物来源, 但缺乏脂肪酸等营养物质, 食物质量比蛋白核小球藻低; 在高浓度(1×105和5×105 cells/mL)下轮虫摄食无毒铜绿微囊藻机率变大, 整体食物品质下降, 对轮虫有抑制作用。有毒铜绿微囊藻对轮虫种群的抑制作用更加明显, 微囊藻浓度升高, 净生殖率和内禀增长率显著下降。研究结果还表明30℃和35℃高温下轮虫生长繁殖变快, 世代时间缩短。在高温(30℃和35℃)环境下, 铜绿微囊藻浓度升高对轮虫抑制作用更加明显。  相似文献

不同光照度和相同的最适营养、温度(25±1)℃以及光周期(12h/12 d)培养铜绿微囊藻、多变鱼腥藻和斜生栅藻的实验结果表明,在15 000 lx和21 000~25 000 lx的光照度下,3种藻的比增长率和现存量最高.15 000 lx是铜绿微囊藻和多变鱼腥藻生长的最适光照度,21 000~25 000 lx是斜生栅藻生长的最适光照度范围.  相似文献

通过10种中草药水提液对铜绿微囊藻抑制效应的实验,筛选出五倍子具有较强的抑藻效果。结果显示:五倍子水提液抑制铜绿微囊藻的最大比生长率为-0.834 d-1,对叶绿素a的抑制率显著,且持续时间长,低浓度组50 mg·L-1处理3 h抑制率超过50%,高浓度组500和1 000 mg·L-1处理24 h后抑制率均超过90%。证明五倍子抑藻作用具迅速、持续稳定的特点,对治理水华的发生有较大的应用前景。  相似文献

沉水植物分布格局对湖泊水环境N、P因子影响   总被引:16,自引:4,他引:16  
利用实地调查数据模拟保安湖沉水植物分布及水环境生态因子场。应用GIS空间分析功能 ,分别空间选取四种优势沉水植物 (金鱼藻CeratophyllumdemersumL .,穗状狐尾藻MyriophyllumspicatumL .,微齿眼子菜PotamogetonmaackianusA .Benn .,及苦草VallisneriaspiralisL .)的分布水域及无沉水植物分布水域的局部生态因子场。根据得到的局部因子场特征 ,比较分析不同水生植物分布格局对水环境中N、P因子的影响。结果显示四种沉水植物的分布对水环境中N、P因子均有显著影响 ,但效果和强度有所差异。四种植物各自分布水域内的N、P因子的平均浓度分别为 ,总氮 (TN) :穗状狐尾藻 (0 .774mg·L- 1 ) >苦草 (0 .71 4mg·L- 1 ) >金鱼藻 (0 .70 1mg·L- 1 ) >微齿眼子菜(0 .695mg·L- 1 ) ;总磷 (TP)平均含量 :穗状狐尾藻 (0 .1 2 3mg·L- 1 ) >微齿眼子菜 (0 .1 1 8mg·L- 1 ) >金鱼藻 (0 .1 0 7mg·L- 1 ) >苦草 (0 .0 79mg·L- 1 )。结果同时表明金鱼藻、微齿眼子菜和苦草对水中TN含量无显著影响 ,而穗状狐尾藻则明显可以提高水中TN水平。四种沉水植物均能有效吸收水中的P ,从而降低水中的TP含量。综合比较发现 ,穗状狐尾藻分布可以加重水体的营养程度  相似文献

【目的】评价白腐真菌Irpex lacteus XX-5、Trichaptum abietinum 1302BG、Ceriporia lacerata P2、Bjerkandera adusta XX-2处理铜绿微囊藻废水的应用潜力。【方法】采用分批次实验研究pH、温度、铜绿微囊藻浓度、金属离子、氮源、磷源对白腐真菌I. lacteus XX-5、T. abietinum 1302BG、C. lacerata P2、B. adusta XX-2溶解铜绿微囊藻的影响。【结果】在不同外源条件下,4种白腐真菌对铜绿微囊藻的抑制效果明显,均达60%以上。菌株C. lacerata P2和B. adusta XX-2受外源条件的影响很小,菌株C. lacerata P2的抑制率达70%以上,菌株B. adusta XX-2的抑制率达60%以上;菌株T. abietinum 1302BG、I. lacteus XX-5在不同外源条件下抑制率均会发生相应的变化,但抑藻率均可达60%以上。【结论】研究所使用的4种白腐真菌对抑制铜绿微囊藻具有较好的应用潜力,尤其是菌株C. lacerata P2和B. adusta XX-2。  相似文献

本文研究不同浓度B9对紫叶酢浆草(OxalisviolaceaL.)试管苗丛生芽增殖与保苗的影响。以MS+6-BA0.5mg·L-1(单位下同)+NAA0.1+30g·L-1蔗糖+7.5g·L-1琼脂为基本培养基,分别添加0、5、10、20、40mg·L-1的B9,pH5.8。每处理接种20瓶,每瓶接种3株苗(剪取约3cm高、带3个小芽的无菌丛生芽),于35d后随机抽取10瓶统计丛生苗发生率,120d后将剩余10瓶统计成活率。培养温度为(23±2)℃,光强约为15μmol·m-2·s-1,光照时间14h·d-1。得到如下结果(表1和2):1.在附加不同浓度B9培养基中的试管苗生长均受到不同程度的抑制,且株高随B9浓度的增加而…  相似文献

对北京密云水库集水区人工油松水源保护林降水化学性质研究表明 ,在研究区大气降水化学元素含量在观测时段内变幅较大 ;测定时段内各元素加权平均值之和为 12 0 0 1mg·L-1,大气降水中各元素含量按大小排序为Ca >N >K >Mg >Na >P >Fe >Zn >Cu >Mn ;大气降水经过林冠层后 ,林内穿透降水和树干茎流中各化学元素含量变化不一 ,但多数元素含量增加且总量呈增加趋势 ,林内穿透降水化学元素加权平均值之和为2 0 614mg·L-1,树干茎流为 73 3 2 4mg·L-1,比林外降水分别高出 8 613mg·L-1和 61 3 2 3mg·L-1,引起树干茎流和穿透降水中化学元素总量增加的主要原因是K含量增加所致 ,K在林内穿透降水和树干茎流中的含量约是大气降水含量的 3倍和 2 5倍 .  相似文献

为快速高效筛选L-精氨酸高产突变株,建立一种缺陷菌株平板显色法并采用低能N+离子束对L-精氨酸生产用菌株钝齿棒杆菌SYPA5-5进行诱变处理,通过上述平板显色法筛选获得高产突变株.对突变株进行摇瓶发酵实验,最终选育出一株L-精氨酸产量较高且产酸性能比较稳定的突变菌株钝齿棒杆菌SYPA5-5-36.该菌株摇瓶发酵L-精氨酸产量可达35.85 g/L,比出发菌株提高了19.5%.因此,缺陷型菌株平板显色法可以用于快速、高效筛选高产L-精氨酸突变株.  相似文献

为了解氮浓度对生物操纵和草-藻竞争的影响, 选取铜绿微囊藻、大型溞和金鱼藻分别作为浮游植物、浮游动物和沉水植物的代表, 在温度25℃, 光强2600 lx, 光暗比14h﹕10h, 磷浓度1.5 mg/L时, 研究5种氮浓度(0.5、2、4、8和16 mg/L, 用KNO3溶液配制)下, 溞-藻, 草-藻和溞-草-藻共培养时各自的增长率和培养液中氮磷削减率的变化。结果表明: 在单独培养铜绿微囊藻时, 氮浓度控制在1.97 mg/L以下, 可有效降低培养液中藻的增长率。在溞-藻共培养时, 大型溞有效控藻的氮浓度范围为0.5—4 mg/L; 在草-藻共培养时, 有效控藻的氮浓度范围为0.5—2 mg/L, 对应氮浓度下(0.5和2 mg/L), 实验末期铜绿微囊藻细胞密度分别是溞-藻共培养的23.89%和21.51%, 控藻效果更好; 在溞-草-藻三者共培养时, 有效控藻的氮浓度范围为0.5—16 mg/L, 且氮浓度为0.5—4 mg/L时, 大型溞和金鱼藻的增长率均显著大于铜绿微囊藻, 铜绿微囊藻的增长率均为负值, 控藻效果最好。大型沉水植物的加入, 可以有效提高生物操纵的控藻效果, 减少水中氮磷含量, 长期有效地改善水质。  相似文献

Arctic charr strain crosses: effects on growth and sexual maturity   总被引:1,自引:0,他引:1  
Three Arctic charr strains and their crosses were evaluated with respect to growth and sexual maturation frequency. No significant heterosis was observed for growth. One out of five crosses was heavier, but not significantly so, than the best performing parental strain at ages 1 + and 2. The five other crosses did not perform better than the best performing parental strain at any age. Cross-breeding was not recommended as an effective method to improve growth performance in Arctic charr. Growth performance of reciprocal crosses differed significantly in all strain combinations: crosses resembled more the male parent strain than the female parent strain. ANOVA for male maturation frequency showed a highly significant effect of the male parent strain and a weakly significant effect of female parent strain. In the Ottsjö strain mature males were significantly larger than immature fish and females. Such size difference was also observed in the offspring when sires of the Ottsjö strain were crossed with dams of the faster growing Honavan strain or with dams of the Rensjö strain. When Ottsjö dams were crossed to sires of the other strains no significant size difference between mature males and other fish was found.  相似文献

研究虫酰肼对甜菜夜蛾汰选种群生长发育及生殖的影响。结果显示:经虫酰肼处理后,甜菜夜蛾SS种群和RR种群卵的孵化抑制中浓度分别为179.38和160.71mg/kg,孵化幼虫的存活抑制中浓度分别为18.64和30.53mg/kg,虫酰肼对甜菜夜蛾卵的孵化率影响较小,而对孵化后幼虫的存活率影响较大。处理初孵幼虫6d后,2种群幼虫存活率及化蛹率随处理浓度的增加而降低,幼虫历期最高饲毒浓度与最低饲毒浓度相比分别延长了2.71d和6.19d,同一浓度下,RR种群幼虫历期比SS种群缩短了0.94~2.59d。处理甜菜夜蛾3龄幼虫12h后,随虫酰肼浓度的增加,2种群化蛹率逐渐降低,雌雄蛹重均有递减的趋势,雌蛹比例随浓度的增加变化较小,但SS种群雌蛹比例≤50.00%,而RR种群雌蛹比例≥50.00%,雌雄成虫羽化率总体上依次递减,处理浓度大于1.00mg/kg时虫酰肼对甜菜夜蛾成虫羽化的影响较大。  相似文献

The effect of certain nutrients on the growth and production of the Bacillus intermedius subtilisin-like serine proteinase by the recombinant strain Bacillus subtilis AJ73(pCS9) was studied. Glucose was found to inhibit the synthesis of proteinase in the early (28 h of growth) but not in the late stationary phase (48 h of growth). The inhibitory effect of the other mono-and disaccharides studied was less pronounced. Casamino acids added to the medium at concentrations of 0.1–1% as an additional carbon and nitrogen source stimulated enzyme biosynthesis. Individual amino acids (cysteine, asparagine, glutamine, tryptophan, histidine, and glutamate) also stimulated enzyme biosynthesis in the early stationary phase by 25–30%, whereas other amino acids (valine, leucine, alanine, and aspartate) were ineffective or even slightly inhibitory to enzyme production. The stimulatory effect of the first group of amino acids on the synthesis of proteinase in the late stationary phase was negligible. In contrast, the bivalent ions Ca2+, Mg2+, and Mn2+ stimulated biosynthesis of proteinase in the late stationary phase (by 20–60%) and not in the early stationary phase. The data indicate that there are differences in the biosyntheses of proteinase by the recombinant B. subtilis strain during the early and late periods of the stationary phases.  相似文献

The cylindrical shape of soft-bodied invertebrates is well suited to functions in skeletal support and locomotion, but may result in a previously unrecognized cost—large non-uniformities in muscle strain and strain rate among the circular muscle fibres of the body wall. We investigated such gradients of strain and strain rate in the mantle of eight long-finned squid Doryteuthis pealeii and two oval squid Sepioteuthis lessoniana. Transmural gradients of circumferential strain were present during all jets (n = 312); i.e. for a given change in the circumference of the outer surface of the mantle, the inner surface experienced a greater proportional change. The magnitude of the difference increased with the amplitude of the mantle movement, with circular muscle fibres at the inner surface of the mantle experiencing a total range of strains up to 1.45 times greater than fibres at the outer surface during vigorous jets. Differences in strain rate between the circular fibres near the inner versus the outer surface of the mantle were also present in all jets, with the greatest differences occurring during vigorous jetting. The transmural gradients of circumferential strain and strain rate we describe probably apply not only to squids and other coleoid cephalopods, but also to diverse soft-bodied invertebrates with hollow cylindrical or conical bodies and muscular organs.  相似文献

Although the physiological strain index (PhSI) is universal and comprehensive, its restrictions are recognized in terms of invasive on-site measurements and the requirement of accurate instruments. The perceptual strain index (PeSI) has been proposed as a user-friendly and practical indicator for heat strain. However, the application of this index in assessing the heat strain of construction workers has yet to be examined and documented. This study aims to ascertain the reliability and applicability of PeSI in an experimental setting that simulates a stressful working environment (i.e., environment, work uniform, and work pace) experienced by construction workers. Ten males and two females performed intermittent exercise on a treadmill while wearing a summer work uniform at 34.5 °C and 75% relative humidity in a climatic chamber. Physiological parameters (core temperature, heart rate) and perceptual variables (thermal sensation, perceived exertion) were collated synchronously at 3 min intervals. The results of two-way repeated measures analysis of variance (clothing×time) revealed that the PeSI was useful in differentiating the heat strain levels between different work uniforms. Not only did the PeSI change in the same general manner with the PhSI, but it was also powerful in reflecting different levels of physiological strain. Thus, the PeSI offers considerable promise for heat strain assessment under simulated working conditions.  相似文献

【目的】通过优化生防菌CNY-04的培养条件,提高其对灰霉病菌的抑菌效果。【方法】在单因素试验的基础上,利用响应面法(Response surface methodology)对灰霉病生防菌CNY-04培养条件进行整体优化,并测其生长曲线。【结果】生防菌CNY-04最优培养条件为牛肉膏0.5%、蛋白胨2.0%、酵母膏0.1%、葡萄糖0.5%、时间48 h、接种量4%、温度32°C、pH 8.0、装液量75 mL/250 mL和转速150 r/min,在此培养条件下生防菌CNY-04的OD600为2.907,与模型预测值相符,对灰葡萄孢菌的抑菌圈直径为44.5 mm,较优化前提高了30.9%。【结论】从整体上确定了生防菌CNY-04的最优培养条件,为该菌扩大化生产提供理论基础。  相似文献

Hypotheses suggest that structural integrity of vertebrate bones is maintained by controlling bone strain magnitude via adaptive modelling in response to mechanical stimuli. Increased tissue-level strain magnitude and rate have both been identified as potent stimuli leading to increased bone formation. Mechanotransduction models hypothesize that osteocytes sense bone deformation by detecting fluid flow-induced drag in the bone''s lacunar–canalicular porosity. This model suggests that the osteocyte''s intracellular response depends on fluid-flow rate, a product of bone strain rate and gradient, but does not provide a mechanism for detection of strain magnitude. Such a mechanism is necessary for bone modelling to adapt to loads, because strain magnitude is an important determinant of skeletal fracture. Using strain gauge data from the limb bones of amphibians, reptiles, birds and mammals, we identified strong correlations between strain rate and magnitude across clades employing diverse locomotor styles and degrees of rhythmicity. The breadth of our sample suggests that this pattern is likely to be a common feature of tetrapod bone loading. Moreover, finding that bone strain magnitude is encoded in strain rate at the tissue level is consistent with the hypothesis that it might be encoded in fluid-flow rate at the cellular level, facilitating bone adaptation via mechanotransduction.  相似文献

Abstract

Sporting helmets with linear attenuating strategies are proficient at reducing the risk of traumatic brain injury. However, the continued high incidence of concussion in American football, has led researchers to investigate novel helmet liner strategies. These strategies typically supplement existing technologies by adding or integrating head-helmet decoupling mechanisms. Decoupling strategies aim to redirect or redistribute impact force around the head, reducing impact energy transferred to the brain. This results in decreased brain tissue strain, which is beneficial in injury risk reduction due to the link between tissue strain and concussive injury.

The purpose of this study was to mathematically demonstrate the effect of ten cases, representing theoretical redirection and redistribution helmet liner strategies, on brain tissue strain resulting from impacts to the head. The kinematic response data from twenty head impacts collected in the laboratory was mathematically modified to represent the altered response of the ten different cases and used as input parameters to determine the effect on maximum principal strain (MPS) values, calculated using finite element modeling. The results showed that a reduced dominant coordinate component (contributes the greatest to resultant) of rotational acceleration decreased maximum principal strain in American football helmets. The study theoretically demonstrates that liner strategies, if applied correctly, can influence brain motion, reduce brain tissue strain, and could decrease injury risk in an American football helmet.  相似文献

Experimental evolution studies with microorganisms such as bacteria and yeast have been an increasingly important and powerful tool to draw long‐term inferences of how microbes interact. However, while several strains of the same species often exist in natural environments, many ecology and evolution studies in microbes are typically performed with isogenic populations of bacteria or yeast. In the present study, we firstly perform a genotypic and phenotypic characterization of two laboratory and eight natural strains of the yeast Schizosaccharomyces pombe. We then propagated, in a rich resource environment, yeast communities of 2, 3, 4, and 5 strains for hundreds of generations and asked which fitness‐related phenotypes—maximum growth rate or relative competitive fitness—would better predict the outcome of a focal strain during the propagations. While the strain''s growth rates would wrongly predict long‐term coexistence, pairwise competitive fitness with a focal strain qualitatively predicted the success or extinction of the focal strain by a simple multigenotype population genetics model, given the initial community composition. Interestingly, we have also measured the competitive fitness of the ancestral and evolved communities by the end of the experiment (≈370 generations) and observed frequent maladaptation to the abiotic environment in communities with more than three members. Overall, our results aid establishing pairwise competitive fitness as good qualitative measurement of long‐term community composition but also reveal a complex adaptive scenario when trying to predict the evolutionary outcome of those communities.  相似文献

采用三亲本杂交法将发光酶luxAB基因转移进棉花根际促生菌芽胞杆菌K2116菌株中,获得标记菌株K2116L.标记菌株连续传代15次均未发生质粒丢失现象,表明标记菌株具有较好的遗传稳定性;K2116L菌株的生长及其释钾能力未受到标记质粒的影响.K2116L菌株在灭菌和非灭菌的黄褐土、黄潮土和红壤3种土壤中均能长期存活;在灭菌土壤中的数量稍高于非灭菌土壤;在3种土壤中的数量依次为:黄褐土>黄潮土>红壤;在不同土壤中,K2116L菌株具有与土著菌株进行空间和营养竞争的能力.采用根盒试验追踪标记菌株在棉花根部的定殖动态,棉花播种12d时标记菌株在0~2、2~4 cm深度根际土壤定殖密度达到最大;18 d时在4 cm以下的深度达到最高水平.棉花播种18 d时标记菌株在所有深度的根表均达到最高定殖水平,0~2 cm根段定殖密度为1.76×106cfu·g-1,8cm以下根段达到1.6×105cfu·g-1.补充营养后,根际和根表标记的菌株数量均有明显上升.试验数据显示,随着根的生长标记菌株不断向根尖方向扩散.  相似文献
