Stems from 198 species representing 165 genera of 46 monocotyledonous families were examined for the presence of fructose oligosaccharides. Aside from occurring in the subfamily Festucoideae of the Poaceae, in Cyperaceae and in Zostera marina of the Zosteraceae, the sugars were found exclusively in some closely related liliaceous families. These are the Agavaceae, Alstromeriaceae, Amaryllidaceae, Cyanastraceae, Haemodoraceae, Iridaceae, Liliaceae, Ruscaceae and Xanthorrhoeaceae. On the other hand, the fructosides were not detected in representatives of certain families whose placement in the order Liliales is controversial or dubious. These are the Dioscoreaceae, Philesiaceae, Philydraceae, Pontederiaceae, Smilacaceae, Stemonaceae, Taccaceae and Velloziaceae. These findings support the assertion of some systematists that these latter families should perhaps not be placed in the order.  相似文献

Besides the non-reducing inulin series of gluco-frueto oligosaccharides which are common in Asteraceae, two series of reducing fructofuranosyl-only oligosaccharides occur in overwintering, forced, and sprouting chicory roots. Such inulo-n-oses could be produced by the action of an endo-inulinase on gluco-fructo oligosaccharides. We show here that similar series of inulo-n-oses as found in vivo can be produced by incubating fructose and inulin with a purified fructamfructan tructosyl transferase (FFT; EC 2.4.1.100) from chicory roots ( Cichorium intybus L.). Therefore fructose (but not glucose) can act as an acceptor for FFT. Since the inulo-n-oses only appear in vivo once the endogenous fructose concentration increases, we conclude that they are not the result of a putative endo-inulinase but of FFT activity.  相似文献

1. Arthrobacter ureafaciens inulinase II which converts inulin to di-D-fructofuranose 1,2' : 2,3' dianhydride (difructose anhydride III) leaving a small amount of oligosaccharides, was investigated in order to characterize its mode of action. 2. After the enzymatic reaction on the glucose-terminated inulin molecules had been completed, the oligosaccharides left in the enzyme digest were isolated, and identified to be the fructose-glucose oligosaccharides; O-beta-D-fructofuranosyl-(2 leads to 1)-O-beta-D-fructofuranosyl alpha-D-glucopyranoside (1-kestose), O-beta-D-fructofuranosyl-[(2 leads to 1)-O-beta-D-fructofuranosyl]2 alpha-D-glucopyranoside and O-beta-D-fructofuranosyl-[(2 leads to 1)-O-beta-D-fructofuranosyl]3 alpha-D-glucopyranoside. The difructose anhydride formation from the three fructose-glucose oligosaccharides in the separate reaction system with an increased substrate concentration was observed only with the latter two substrates, but not with the first one. 3. The difructose anhydride formation with several (2 leads to 1)-beta-linked fructose oligosaccharides and bacterial (2 leads to 6)-beta-fructans was examined. The (2 leads to 1)-beta-linked fructose oligosaccharides were effective as substrates for the enzyme with the exception of inulobiose, but the (2 leads to 6)-beta-fructans remained unaffected. 4. It was concluded that the enzyme attacks (2 leads to 1)-beta-linked fructan molecules from the nonreducing fructose ends and requires the presence of at least two adjacent (2 leads to 1)-beta-fructofuranosyl linkages.  相似文献

Evidence is presented that fructan accumulation in leaves of Lolium temulentum plants grown at 5° proceeded via the synthesis of trisaccharide intermediates. Studies on the oligosaccharide components of this tissue indicated that the major intermediate was probably 1F-fructosyl sucrose (isokestose) but that two distinct series of oligofructosides could be isolated. One of these had chromatographic properties identical to the 1,2-linked inulin series from Helianthus tuberosus. The relationship of this synthetic pattern to the structure of grass fructans and their accumulation in other species is discussed.  相似文献

The freezing resistance of the grass species Phleum pratense L. (timothy) and Phalaris arundinaces L. increases significantly after cold hardening. The content and composition of soluble carbohydrates were determined in the plants after short day treatment, cold hardening and dehardening. The amounts of mono-, di- and trisaccharides were reduced during the short day treatment, increased during cold hardening and decreased again during dehardening. The total amounts of soluble carbohydrates (mono-, di-, tri- and polysaccharides) were the same in hardened and dehardened plants, indicating that during hardening soluble polysaccharides (fructose polymers, fructans) were converted to mono- and oligosaccharides. Sucrose increased most after hardening conditions and, in P. arundinacea , a significant increase in 1-F-fructosylsucrose (isokestose) was also observed.
Invertase (β-fructofuranosidase. EC 3.2.1.26) activity increased following cold hardening and decreased following dehardening, while the α-galactosidase (EC 3.2.1.22) activity seemed to increase after dehardening. The glycosidases are probably involved in the mobilisation of polysaccharides during cold hardening.  相似文献

A total of 389 birds of 32 species representing 14 families from Tchad were examined for blood parasites. Eighty-nine (22.9%) harbored infections of Haemoproteus (64%), Plasmodium (12.4%), Trypanosoma (1.1%), Atoxoplasma (=Lankesterella) (5.6%), and microfilaria (28.1%). Species of Leucocytozoon were not observed in the present study. The occurrence of the different genera differed markedly between bird families; members of the Ploceidae comprised 85% of the infected birds.  相似文献

AIMS: To characterize the fructose polymer degrading enzymes of rumen bacterium Treponema saccharophilum strain S. METHODS AND RESULTS: Conventional methods were used to examine bacterial growth and enzyme activities. Electrophoretic zymogram under native conditions, and thin layer chromatography, were applied to identify and characterize the enzymes. Treponema saccharophilum utilized Timothy grass fructan, inulin and sucrose but not free fructose. Timothy grass fructan was degraded at a significantly higher rate than sucrose and inulin. Two fructanolytic enzymes were found in the soluble, and one in the membrane fraction of bacterial cell extract. The first degraded each mentioned carbohydrate to monosaccharides. The second released oligosaccharides only from Timothy grass fructan. CONCLUSIONS: The bacterium T. saccharophilum strain S is capable of synthesizing non-specific beta-fructofuranosidases and 2,6-beta-D-fructan fructanohydrolase. The enzymes are of constitutive character. SIGNIFICANCE AND IMPACT OF THE STUDY: It has been stated for the first time that the 2,6-beta-D-fructan fructanohydrolase is synthesized by the rumen bacterium T. saccharophilum. This organism appears to be responsible for grass fructan degradation in the rumen.  相似文献

Soderstrom , Thomas R. (Smithsonian Institution, Washington, D.C.) The isocitric acid content of crassulacean plants and a few succulent species from other families. Amer. Jour. Bot. 49(8): 850–855. 1962.—The leaves of 58 species of plants were assayed for isocitric acid content by the highly specific enzymatic method of Grafflin and Ochoa (1950). These species included 39 from the family Urassulaceae in addition to 19 species of succulents representing 8 families and 12 genera. With but one exception (Echeveria derenbergii), isocitric acid was found in relatively large concentrations in the leaves of all crassulacean species examined, and was recorded for the first time in 5 genera of this family. Isocitric acid was found in 2 genera of the Liliaceae and in 1 genus of the Piperaceae, families in which it previously had not been known to occur. The presence of substantial concentrations of isocitric acid in the leaves appears to be characteristic of the family Crassulaceae and of certain genera in other widely unrelated families. There is no indication, however, that the concentrations of this acid in the leaves of species and genera of the Crassulaceae is correlated with their affinities based on morphology.  相似文献

Chloroplast trnL/F and nuclear ribosomal ITS and ETS sequence data were used to analyze phylogenetic relationships among members of tribe Mimuleae (Scrophulariaceae) and other closely related families in Lamiales. The results of these analyses led to the following conclusions. (1) The Australian genera Glossostigma and Peplidium and the taxonomically isolated Phryma join four genera of tribe Mimuleae to form a well-supported clade that is distinct from other families in the Lamiales. We refer to that clade as the subfamily Phrymoideae. (2) The genera Mazus and Lancea (tribe Mimuleae) together form a well-supported clade that we recognize as the subfamily Mazoideae. Mazoideae is weakly supported as sister to Phrymoideae. We assign Mazoideae and Phrymoideae to a redefined family Phrymaceae. (3) Mimulus is not monophyletic, because members of at least six other genera have been derived from within it. In light of the molecular evidence, it is clear that species of Phrymaceae (about 190 species) have undergone two geographically distinct radiations; one in western North America (about 130 species) and another in Australia (about 30 species). Phylogenetic interpretations of morphological evolution and biogeographical patterns are discussed.  相似文献

This article highlights the research work carried out in the production of inulinases from various inulin substrates using strains of bacteria, yeast and fungi. Inulin is one of the numerous polysaccharides of plant origin that contains glucose or fructose. It is used as a substrate in industrial fermentation processes and in food industries due to its relatively cheap and abundant source for the microbiological production of high-fructose syrups, ethanol and acetone–butanol. The various oligosaccharides derived from inulin also find their application in the medical and dietary sector. The inulinase acts on the β-(2,1)-D-fructoside links in inulin releasing D-fructose. Hence, this article illustrates the capability of various microbes in hydrolyzing the carbon at its optimum nutrient concentration and operating condition towards inulinase production.  相似文献

本文在2015年发表的爬行动物名录及同年《中国两栖类信息系统》发布的两栖动物名录的基础上, 通过整理新发表的分类学研究及先前名录遗漏的部分早期文献, 更新了截至2019年底中国现生本土两栖、爬行动物物种名录。2015-2019年间, 中国两栖动物新记录1科, 新描述2属, 恢复1属有效性, 新记录1属, 新描述或恢复有效种74种, 新增国家纪录18种; 另6属、8种的有效性未得到近年研究证据支持(在此视为次定同物异名而未做收录, 后同)。同期, 中国爬行动物新恢复5科, 新描述1亚科, 新描述1属, 恢复3属有效性, 新记录3属, 新描述、恢复或提升有效种43个, 新增国家纪录10种; 另有5属、4种的有效性未得到近年研究证据支持, 并移除1属、4种在我国的分布纪录。此外, 通过整理2015年前文献, 爬行动物增补3属, 提升3亚种至种级地位, 增补国家新纪录3种, 另有3属、2种的有效性未得到近年研究证据支持, 同时移除1种在我国的分布纪录。综上, 截至2019年底, 我国共记录现生本土两栖动物3目13科62属515种(蚓螈目1科1属1种, 有尾目3科14属82种, 无尾目9科47属431种), 爬行动物3目35科135属511种(鳄形目1科1属1种, 龟鳖目6科18属34种, 有鳞目蛇亚目18科73属265种、蜥蜴亚目10科43属211种)。此外, 本文还对先前名录中部分爬行动物的中文名提出了修改建议, 建议恢复部分物种的惯用中文名。2015-2019年, 新物种及新纪录已知物种数量占现两栖、爬行动物物种总数的17.1%和10.2%。近年来, 我国发表的两栖、爬行动物新物种和已知物种的新纪录数量持续增加, 分类体系也在研究中不断完善, 建议今后及时地进行阶段性总结, 同时对存在的问题提出讨论, 以推动中国两栖、爬行动物分类学研究工作的进一步开展。  相似文献

Bifidobacteria are a minor fraction of the human colon microbiota with interesting properties for carbohydrate degradation. Monosaccharides such as glucose and fructose are degraded through the bifid shunt, a dedicated pathway involving phosphoketolase activity. Its stoechiometry learns that three moles of acetate and two moles of lactate are produced per two moles of glucose or fructose that are degraded. However, deviations from this 3 : 2 ratio occur, depending on the rate of substrate consumption. Slower growth rates favour the production of acetate and pyruvate catabolites (such as formate) at the cost of lactate. Interestingly, bifidobacteria are capable to degrade inulin‐type fructans (ITF) (oligofructose and inulin) and arabinoxylan‐oligosaccharides (AXOS). Beta‐fructofuranosidase activity enables bifidobacteria to degrade ITF. However, this property is strain‐dependent. Some strains consume both fructose and oligofructose, with different preferences and degradation rates. Small oligosaccharides (degree of polymerization or DP of 2–7) are taken up, in a sequential order, indicating intracellular degradation and as such giving these bacteria a competitive advantage towards other inulin‐type fructan degraders such as lactobacilli, bacteroides and roseburias. Other strains consume long fractions of oligofructose and inulin. Exceptionally, oligosaccharides with a DP of up to 20 (long‐chain inulin) are consumed by specific strains. Also, the degradation of AXOS by α‐arabinofuranosidase and β‐xylosidase is strain‐dependent. Particular strains consume the arabinose substituents, whether or not together with a consumption of the xylose backbones of AXOS, either up to xylotetraose or higher and either extra‐ or intracellularly. The production of high amounts of acetate that accompanies inulin‐type fructan degradation by bifidobacteria cross‐feeds other colon bacteria involved in the production of butyrate. However, bifidobacterial strain‐dependent differences in prebiotic degradation indicate the existence of niche‐specific adaptations and hence mechanisms to avoid competition among each other and to favour coexistence with other colon bacteria.  相似文献

Halobacterium marismortui is an obligatorily halophilic species isolated from the Dead Sea. When inulin, fructose or glycerol are added to suspensions of bacteria, the amounts of these substances recovered from centrifuges pellets are more than could have been present in the extracellular space. Thus a certain amount becomes associated with the bacteria, though not enough to equilibrate with all the cell water. The inulin or fructose concentration found after uptake of these substances was correlated with the cell sodium concentration. It is argued that inulin, fructose or glycerol is unlikely to be adsorbed on the outside of the bacteria and more probably crosses the plasma membrane. A possible scheme for explaining the data is presented.  相似文献

Analysis of the neutral urinary oligosaccharides in bovine, feline and human mannosidosis by thin-layer and gel-permeation chromatography has shown that the patterns of stored oligosaccharides in the three species are different. In bovine and feline mannosidosis the most abundant urinary oligosaccharide is also the most abundant in the tissues of each species. The predominant oligosaccharides were purified by a combination of gel-filtration, ion-exchange and thin-layer chromatography and shown to contain only mannose and N-acetylglucosamine by g.l.c. and g.l.c.--mass spectrometry. The probable composition and size of each oligosaccharide were predicted from its chromatographic properties, sugar composition and the known structure of asparagine-linked oligosaccharides. The bovine and feline oligosaccharides belonged to a homologous series of general composition Mann (GlcNAc)2, whereas the human oligosaccharides belong to a different series, MannGlcNAc. These structures suggest that lysosomal endohexosaminidase is not present in bovine and feline tissues. The predominant feline storage product, Man3(GlcNAc)2, was the expected storage product from the catabolism of complex asparagine-linked glycans. In contrast, the predominant bovine oligosaccharide, Man2(GlcNAc)2, probably lacks one of the alpha-linked mannose residues in the core region. A similar situation occurs in human mannosidosis. It is predicted that in these species either that the residual mutant alpha-D-mannosidase retains activity towards one of the core alpha-linked mannose residues or that another form of lysosomal alpha-D-mannosidase that is unaffected in these disorders occurs. It is concluded that the differences in storage products are due to differences in the catabolic pathways of glycoproteins among the species.  相似文献

The content and composition of fructans in tuberous roots ofViguiera discolor Baker (Asteraceae), a perennial herbaceousspecies native to the Brazilian cerrado, are analysed accordingto season and in sequence from late dormancy (winter) to thebeginning of the next dormant period (autumn). The general patternof fructose found in decreasing concentrations of ethanol extractsis similar at each phenological phase. Hot water soluble fructans(high molecular mass inulin) are predominant throughout, particularlyin the dormant period, and account for more than half the totalfructan. Lower molecular mass components, especially free fructose,are present in higher amounts during the sprouting phase; thiswould indicate that hydrolysis occurs in association with thebreaking of dormancy. The absence of oligosaccharides of intermediatesize during the flowering/fruiting phase suggests they mightbe metabolized in the course of this process. The total concentrationof fructans and the mean molecular mass of inulin remain highin spite of these variations. (Received July 15, 1992; Accepted May 10, 1993)  相似文献

Blood smears from 259 birds of 12 species, representing four families of raptors, from New Jersey, Pennsylvania, Delaware, and Virginia were examined for blood parasites. Infected birds constituted 59.1% of the total. Birds were infected with one or more of the following genera of protozoa: Leucocytozoon (43.2%); Haemoproteus (21.6%); Plasmodium (1.2%); and Trypanosoma (1.2%). Blood culture of 142 raptors of 11 species for Trypanosoma revealed a prevalence of 41.5%. Plasmodium circumflexum is reported for the first time in Accipiter striatus, and Trypanosoma sp. in Buteo jamaicensis.  相似文献

Summary The utilization of some mono- and oligosaccharides by the members of Choanephoraceae has been studied in detail. The filtrate was analysed by using circular paper chromatography. Amongst the seven monosaccharides tested, viz., glucose, galactose, fructose, mannose, xylose, sorbose and rhamnose, the first five were completely utilized within the specified period, while sorbose and rhamnose remained in the medium throughout the incubation period. A mixture of glucose, galactose and fructose was found to support better growth of all the present species, than that when these sugars were supplied singly. Out of the four oligosaccharides tested, only maltose could be hydrolysed, and it was completely consumed within the specified period. The other three oligosaccharides, viz., sucrose, lactose and raffinose were not hydrolysed and they remained in the medium throughout the incubation period.  相似文献

Mygalomorph spiders, which include the tarantulas, trapdoor spiders, and their kin, represent one of three main spider lineages. Mygalomorphs are currently classified into 15 families, comprising roughly 2500 species and 300 genera. The few published phylogenies of mygalomorph relationships are based exclusively on morphological data and reveal areas of both conflict and congruence, suggesting the need for additional phylogenetic research utilizing new character systems. As part of a larger combined evidence study of global mygalomorph relationships, we have gathered approximately 3.7 kb of rRNA data (18S and 28S) for a sample of 80 genera, representing all 15 mygalomorph families. Taxon sampling was particularly intensive across families that are questionable in composition-Cyrtaucheniidae and Nemesiidae. The following primary results are supported by both Bayesian and parsimony analyses of combined matrices representing multiple 28S alignments: (1) the Atypoidea, a clade that includes the families Atypidae, Antrodiaetidae, and Mecicobothriidae, is recovered as a basal lineage sister to all other mygalomorphs, (2) diplurids and hexathelids form a paraphyletic grade at the base of the non-atypoid clade, but neither family is monophyletic in any of our analyses, (3) a clade consisting of all sampled nemesiids, Microstigmata and the cyrtaucheniid genera Kiama, Acontius, and Fufius is consistently recovered, (4) other sampled cyrtaucheniids are fragmented across three separate clades, including a monophyletic North American Euctenizinae and a South African clade, (5) of the Domiothelina, only idiopids are consistently recovered as monophyletic; ctenizids are polyphyletic and migids are only weakly supported. The Domiothelina is not monophyletic. The molecular results we present are consistent with more recent hypotheses of mygalomorph relationship; however, additional work remains before mygalomorph classification can be formally reassessed with confidence-increased taxonomic sampling and the inclusion of additional character systems (more genes and morphology) are required.  相似文献
