氟铃脲对斜纹夜蛾酚氧化酶活性的影响   总被引:2,自引:0,他引:2  
【目的】探讨氟铃脲对斜纹夜蛾Spodoptera litura酚氧化酶活性的影响.【方法】应用酶动力学法,在确定斜纹夜蛾酚氧化酶最适反应条件基础上,测定氟铃脲与酚氧化酶直接作用及氟铃脲处理幼虫后酚氧化酶的活性.【结果】以邻苯二酚为底物,斜纹夜蛾酚氧化酶的最适pH 6.5,最适温度30℃.氟铃脲没有和酚氧化酶结构中的铜离子结合,而是直接与酶蛋白发生作用.氟铃脲浓度低于1 153 mg/L对离体酚氧化酶有激活作用,在138 mg/L激活作用最大,浓度高于1 153 mg/L则产生抑制作用.用46 mg/L和92 mg/L亚致死剂量氟铃脲连续处理5龄幼虫不同时间后,酚氧化酶活性均显著提高,且高浓度的激活作用大于低浓度的激活作用.进一步测定药剂处理幼虫24,48和72 h的血淋巴、表皮和头部酚氧化酶活性,随着氟铃脲处理时间延长,两个剂量的氟铃脲对血淋巴、表皮及头部酚氧化酶的激活作用逐渐增大,氟铃脲对血淋巴酚氧化酶的激活作用最大,表皮次之,头部最低.氟铃脲处理幼虫后,其预蛹和蛹酚氧化酶的活性也显著提高,表现出一定的后效应.【结论】一定浓度范围内,氟铃脲对斜纹夜蛾酚氧化酶具有激活作用.  相似文献

采用40%硫酸铵沉淀、Blue Sepharose CL-6B亲和层析和Phenyl Sepharose CL-4B疏水层析等方法,从亚洲玉米螟Ostrinia furnacalis (Guenée) 幼虫血清中分离纯化了酚氧化酶原。酚氧化酶原全酶相对分子量约为158 kD,亚基相对分子量约为80 kD和78 kD。酚氧化酶原为糖蛋白,该酶原易被0.1 mmol/L CPC (氯代十六烷基吡啶)、 50%甲醇、 1 mg/mL昆布多糖和1 mg/mL胰蛋白酶激活。该酶反应的最适pH为7.0,最适温度为25~30℃,Ca2+和Mg2+可增强该酶的活性。  相似文献

利用热带假丝酵母由烷烃生产二元酸时,二元酸面临被β氧化降解的代谢途径。酰基辅酶A氧化酶是二元酸β氧化的限速酶。以热带假丝酵母1230菌株为材料,经硫酸铵分级沉淀、阴离子交换柱层析、BlueSepharose亲和柱层析,得到电泳均一的酰基辅酶A氧化酶。该酶有两种亚基,分子量分别为74kD和78kD。酶作用最适pH和最适温度分别为80和50℃。金属离子Ag+、Pb2+完全抑制酶活性,Ba2+、Mg2+、Ca2+对酶活性有明显抑制作用。丙烯酸是酶的反竞争性抑制剂,Ki为0633mmol/L,维生素C是竞争性抑制剂,Ki为2.01×10-3mmol/L。  相似文献

海洋弧菌褐藻胶裂解酶的分离纯化及性质   总被引:3,自引:0,他引:3  
从海带糜烂物中分离到一株高产胞外褐藻胶裂解酶的海洋弧菌 (Vibriosp .QY10 1) ,利用硫酸铵沉淀、离子交换层析、凝胶过滤层析等方法从发酵液中分离纯化了褐藻胶裂解酶 (alginatelyase)。SDS PAGE电泳结果表明 ,该酶分子量为 39kD。酶反应最适pH为 7.5 ,最适反应温度为 30℃。Na 、Ca2 、Mn2 对酶活性有促进作用 ,Fe2 、Ni2 以及EDTA对酶活性有抑制作用。酶的底物专一性初步分析结果表明 ,该酶具有降解多聚古罗糖醛酸[poly(G) ]及多聚甘露糖醛酸 [poly(M) ]的活性。  相似文献

通过硫酸铵分级沉淀、疏水层析及阴离子交换层析等三步 ,有效地从一菌株NO .2 2 6 2中纯化了N 氨甲酰基 D 氨基酸酰胺水解酶。结果表明 ,酶活性回收约 2 0 %,纯化了 8 4倍。天然PAGE与SDS PAGE分析表明 ,该酶分子为同源四聚体 ,单体分子量约为 3 5kD。酶催化反应的最适pH为 7 7~ 8 0 ,最适温度为 45℃。以N 氨甲酰 DL 丙氨酸为底物时 ,Km =1 3×1 0 - 3 mol L ,Vmax=0 .3 3mol min。二价金属离子Ni2 + 有激活作用 ,Zn2 + 有明显的抑制作用 ,而Co2 + 对酶活无影响。该酶N 末端 8个氨基酸残基依次为TRQKILAF。  相似文献

本研究首次发现Monodictyx asperospera(Cooke&Massee)Ellis具有较好的产漆酶能力.粗酶液经硫酸铵盐析、DEAE-纤维素层析及丙烯葡聚糖凝胶S-300层析纯化,纯化倍数为8.1,回收率为5.7%.漆酶分子量约为77kD,最适反应温度为55℃,最适反应pH6.0,以丁香醛连氮为底物时Km为0.163mm0l/L,Vmax为0.194 mmol(L·min),含糖量为18.14%,Cu2+对漆酶有明显抑制作用.  相似文献

研究了中华卵索线虫Ovomermis sinensis感染棉铃虫Helicoverpa armigera幼虫后宿主体内酚氧化酶活性的变化。研究结果表明,在感染后的第1天,中华卵索线虫的侵入引起酚氧化酶活性的增加,感染组酶活性是同期对照组的1.12倍; 但在随后的寄生期间,中华卵索线虫抑制了宿主的酚氧化酶活性,其中以第5天的抑制最为强烈: 同期对照组酶活性是感染组的1.52倍。对酚氧化酶进行了初步的分离纯化,纯化倍数为41.5倍,酶得率为12.7%,比活力为4 030.6 U/mg。  相似文献

黑木耳漆酶研究可为漆酶的进一步分离纯化、基因克隆表达和大规模生产应用奠定基础。对黑木耳"黑29"菌株漆酶粗酶液进行硫酸铵分级沉淀后,通过Native SDS-PAGE电泳检测,存在3种漆酶LacA、LacB、LacC,分子量分别为60,34,19 kD。经硫酸铵分级沉淀和DEAE-Sephacel柱层析技术分离得一纯化成分LacC,纯化倍数7.60,酶活性回收4.28%。对LacC的pH、温度、金属离子和Km值等部分酶学性质进行研究发现,该酶氧化ABTS的Km值为1.18×10-6mol/L,催化氧化底物ABTS的最适pH为3.8,在pH 3.0~4.6表现出较强的稳定性;最适反应温度为55℃,低于50℃时有较好的热稳定性;金属离子Ag+对漆酶有激活作用,而Fe3+、Mn2+、Co2+则有抑制作用。  相似文献

对毛木耳AuriculariapolytrichaAP4的粗酶液进行PAGE电泳后发现含有三种漆酶同工酶,并且通过运用NativeSDS-PAGE获得三种漆酶的分子量大小分别约为:LacA(110kD);LacB(84kD);LacC(65kD)。对漆酶粗酶液通过硫酸铵分级沉淀和离子交换柱层析进行纯化,用SDS-PAGE证明获得纯化的单一漆酶LacB。LacB漆酶的反应的最适温度为30℃,最适pH为3.0。此酶氧化ABTS的Km值为6.64×10-mmol/L,金属离子对酶活的影响很大,其中5Ca2+,Mg2+,Zn2+,Na2+,Ag2+对漆酶LacB有明显的激活作用;Co2+,Hg2+,Fe3+,Fe2+,Ba2+等对酶活有明显的抑制作用。LacB和其它真菌漆酶一样具有底物专一性不强的特点,并且LacB对RB亮兰染料有很好的脱色作用。  相似文献

It has now been over twenty years since a novel herpesviral genome was identified in Kaposi's sarcoma biopsies. Since then, the cumulative research effort by molecular biologists, virologists, clinicians, and epidemiologists alike has led to the extensive characterization of this tumor virus, Kaposi's sarcoma-associated herpesvirus(KSHV; also known as human herpesvirus 8(HHV-8)), and its associated diseases. Here we review the current knowledge of KSHV biology and pathogenesis, with a particular emphasis on new and exciting advances in the field of epigenetics. We also discuss the development and practicality of various cell culture and animal model systems to study KSHV replication and pathogenesis.  相似文献

Comprises species occurring mostly in subtidal habitats in tropical, subtropical and warm-temperate areas of the world. An analysis of the type species, V. spiralis (Sonder) Lamouroux ex J. Agardh, a species from Australia, establishes basic characters for distinguishing species in the genus. These characters are (1) branching patterns of thalli, (2) flat blades that may be spiralled on their axis, (3) width of the blade, (4) primary or secondary derivation of sterile and fertile branchlets and (5) position of sterile and fertile branchlets on the thalli. Application of the latter two characters provides an important basic method for separation of species into three major groups. Osmundaria , a genus known only in southern Australia, was studied in relation to Vidalia , and its separation from the Vidalia assemblage is not accepted. Species of Vidalia therefore are transferred to the older genus name, Osmundaria. Two new species, Osmundaria papenfussii and Osmundaria oliveae are described from Natal. Confusion in the usage of the epithet, Vidalia fimbriala Brown ex Turner has been clarified, and Vidalia gregaria Falkenberg, described as an epiphyte on Osmundaria pro/ifera Lamouroux, is revealed to be young branches of the host, Osmundaria prolifera.  相似文献

Fifteen chromosome counts of six Artemisia taxa and one species of each of the genera Brachanthemum, Hippolytia, Kaschgaria, Lepidolopsis and Turaniphytum are reported from Kazakhstan. Three of them are new reports, two are not consistent with previous counts and the remainder are confirmations of very scarce (one to four) earlier records. All the populations studied have the same basic chromosome number, x = 9, with ploidy levels ranging from 2x to 6x. Some correlations between ploidy level, morphological characters and distribution are noted.  相似文献

采用原位分子杂交方法检测HCV RNA及HBV X基因;采用免疫组织化学方法研究HCV核心抗原,非结构区C33c抗原及HBxAg在肝细胞肝癌中的定位及分布.结果表明(1)HCV RNA、HBV X基因在肝细胞肝癌组织检出率分别为40%(55/136)和82%(112/136).HCV RNA定位于癌细胞的胞浆内,阳性细胞呈散在、灶状及弥漫分布三种形式;HBV X基因在肝癌细胞中的分布呈胞浆型、核型及核浆型,阳性细胞也呈上述三种分布形式;(2)HCV C33c抗原、核心抗原在肝细胞肝癌中的阳性率为81%(133/164)及86%(141/164).C33c抗原定位于癌细胞及肝细胞的胞浆内;核心抗原既定位于癌细胞核中,又可定位于胞浆中.C33c抗原阳性细胞以灶状分布为主;而核心抗原阳性细  相似文献

For a plant selection model with frequency-independent viabilities, fertilities and selfing rates, it is shown that apart from global fixation, for certain parameter combinations a protected polymorphism and facultative fixation (either allele may become fixed according to initial frequencies) may both occur. Facultative fixation requires different selling rates for the dominant and recessive type. Protection of the polymorphism requires resource allocation for male and female function. In this connection the problem of purely genetically caused population extinction is discussed.
For general frequency dependence and regular segregation, the chances for establishment of a completely recessive gene are compared to those of a completely dominant gene. It is proven that the process of establishment of the recessive gene, despite a fitness advantage, may be considerably endangered by drift effects if random mating prevails. The recessive gene may reach the same effectivity in establishment as a dominant gene, only if the recessive homozygote mates exclusively with its own type during the period of establishment.  相似文献
