Brief treatment of rat liver plasma membranes with phospholipase C of Clostridium welchii increased both the ratio of saturated to unsaturated fatty acids and the ratio of cholesterol to phospholipids. Using 5-doxylstearic acid spin probes two breaks at 29 and 19.6 °C could be observed in the order parameter, SA, vs temperature curve for untreated membranes. Upon phospholipase C digestion the lower phase transition temperature was shifted to 23 °C, while the higher phase transition temperature could not be detected up to 40 °C. The order parameter, SA, was consistently higher at all temperatures in the phospholipase C-treated membranes. As phospholipase C is known to attack the outer lamella, these results can be interpreted as indicating an increase in ordering (i.e., decrease in fluidity) of the outer membrane lamella. On the other hand, an increase in basal activity of adenylate cyclase of the treated membranes was observed with an apparent reduction of the activation energies both below and above the break (at 20 °C) in the Arrhenius plot of enzyme activity. Phospholipase C treatment did not affect the temperature of the break in Arrhenius kinetics of the enzyme. The results are discussed in terms of the role of the ordering state of membrane lipids in adenylate cyclase activity.  相似文献

Delipidation of beef heart electron transport particles with phospholipase A2 has been examined. When the particles were treated with the lipase and subjected to a low bovine serum albumin wash, ATPase activity was unaffected as was the lipid/protein ratio of the particles. However, energisation by ATP/Mg2+ was abolished. Furthermore, unsaturated but not saturated fatty acids discharged the steady-state ATP-driven membrane potential of control samples. When the phospholipase A2 hydrolysis products were removed, inhibition of energy-linked reactions in the lipid-depleted particles was still observed and was interpreted in terms of non-specific leaks in the vesicle membranes, and ‘specific’ leaks through impaired H+-ATPase complexes. ATPase activity was less susceptible to delipidation than energisation but was, nevertheless, strongly inhibited at 50 percent lipid depletion.

Spin label studies indicated a decrease in the fluidity of particle membranes accompanying delipidation. Moreover, the discontinuity seen in Arrhenius plots of ATPase activity was shifted from 17°C (control) to 22°C at 50 percent phospholipid depletion. The data are consistent with a release of unsaturated fatty acids by phospholipase A2 rendering the transport particles both leakier and the membranes less fluid than controls.  相似文献

Laser light scattering has been used to investigate the thermal pretransition of dipalmitoylglycerophosphocholine vesicles with variable radius as obtained by the mild sonication method. Intensity changes in 90° scattered light are observed at the pretransition for larger vesicles and actually increase with increasing vesicle size, reaching a constant value.This constant value is in good agreement with the value calculated from the refractive index data.The intensity ratio of scattered light at temperatures of 30°C and 40°C (I40/I30) approaches unity at a radius of small single-bilayer vesicle. This result is interpreted as no pretransition for small vesicles in agreement with the calorimetric results. An expression of the particle scattering factor is also presented for multilayered shells composed of anisotropic elements. It is shown numerically, using this expression, that changes in the lipid layer thickness and the tilting angles at the pretransition have no effects on the scattering factor. Therefore it is concluded that the intensity changes in scattered light reflect the changes in the refractive index of the vesicle originating in the polar head groups.  相似文献

The pldA gene encoding membrane-bound phospholipase A1 of Yersinia pseudotuberculosis was cloned and expressed in Escherichia coli cells. Recombinant phospholipase A1 (rPldA) was isolated from inclusion bodies dissolved in 8 M urea by two-stage chromatography (ion-exchange and gel-filtration chromatography) as an inactive monomer. The molecular mass of the rPldA determined by MALDI-TOF MS was 31.7 ± 0.4 kDa. The highly purified rPldA was refolded by 10-fold dilution with buffer containing 10 mM Triton X-100 and subsequent incubation at room temperature for 16 h. The refolded rPldA hydrolyzed 1,2-dioleoyl-sn-glycero-3-phosphatidylcholine in the presence of calcium ions. The enzyme exhibited maximal activity at 37°C and nearly 40% of maximal activity at 15°C. The phospholipase A1 was active over a wide range of pH from 4 to 11, exhibiting maximal activity at pH 10. Spatial structure models of the monomer and the dimer of Y. pseudotuberculosis phospholipase A1 were constructed, and functionally important amino acid residues of the enzyme were determined. Structural differences between phospholipases A1 from Y. pseudotuberculosis and E. coli, which can affect the functional activity of the enzyme, were revealed.  相似文献

Abstract

S-PLI, an inhibitor of phospholipase C (PLC) produced by Strepromyces sp. strain No. 6288, was purified from the culture filtrate by salting-out with solid ammonium sulfate, column chromatography on CM-cellulose and gel filtration on Sephadex G-75. The molecular weight of S-PLI was estimated to be 65,000 by SDS-polyacrylamide gel electrophoresis. The inhibitor was found to be a glycoprotein with a composition of 609 amino acids and 19 glucose residues having an isoelectric point at 7.8. S-PLI was stable from pH 3 to 10 at 37°C and up to 40° at pH 6.0. The inhibitory activity showed pH-and temperature-dependence with a maximum around pH 7.0 at 50°C. S-PLI inhibited phospholipase C in a competitive manner (Ki value; 9.5 × 10-6 mM), but did not inhibit S-Hemolysin, phospholipase A2, phospholipase B, phospholipase D and phosphatases. S-PLI is the first reported example of a glycoproteinaceous inhibitor of microbial origin which is able to specifically inhibit phospholipase C.  相似文献

Properties of the aqueous dispersions of n-octadecylphosphocholine are examined by differential scanning calorimetry, fluorescence depolarization, light scattering, 31P-NMR, pig pancreatic phospholipase A2 binding, and X-ray diffraction. On heating, these dispersions exhibit a sharp lamellar to micelle transition at 20.5°C. The lamellar phase consists of frozen (gel-state) alkyl chains which do not bind phospholipase A2. The kinetics of the transition are asymmetric: the micelle to lamellar transition is very slow and the lamellar to micelle transition is fast. It is suggested that the lamellar phase is a frozen chain bilayer in which the chains interdigitate.  相似文献

A novel microbial phospholipase C (PLC) from Bacillus licheniformis MTCC 7445 was purified to homogeneity by ammonium sulphate fractionation, dialysis, anion exchange chromatography and gel exclusion chromatography. The bacteria growing on vegetable oils secreted significantly high amount of PLC. The enzyme was purified to 23.4-fold with 46% recovery and specific activity 398 U/mg. It exhibited optimum activity at 70°C and pH 10.0. Using diphosphatidylglycerol as substrate the PLC of B. licheniformis MTCC 7445 had a V max and K m of 0.68 mM/min and 32 mM, respectively. It hydrolyzed phosphatidylinositol and phosphatidylserine as well as phosphatidylcholine but not other glycerophospholipids. Its activity was enhanced by 113% with Mn2+ and 110% with Mg2+. During degumming of vegetable oils with this enzyme preparation, the phosphorus content of the oil became lower than 4 mg/kg after 5 h of enzyme treatment at 40°C. The novel PLC from B. licheniformis MTCC 7445 is potentially useful for the refining of high quality oils with 95% removal of phospholipids with attractive yield.  相似文献

Isotopic exchange kinetics at equilibrium for E. coli native aspartate transcarbamylase at pH 7.8, 30 °C, are consistent with an ordered BiBi substrate binding mechanism. Carbamyl phosphate binds before l-Asp, and carbamyl-aspartate is released before inorganic phosphate. The rate of [14C]Asp C-Asp exchange is much faster than [32P]carbamyl phosphate Pi exchange. Phosphate, and perhaps carbamyl phosphate, appears to bind at a separate modifier site and prevent dissociation of active-site bound Pi or carbamyl phosphate. Initial velocity studies in the range of 0–40 °C reveal a biphasic Arrhenius plot for native enzyme: Ea (>15 °C) = 6.3 kcal/ mole and Ea (<15 °C) = 22.1 kcal/mole. Catalytic subunits show a monophasic plot with Ea ? 20.2 kcal/mole. This, with other data, suggests that with native enzyme a conformational change accompanying aspartate association contributes significantly to rate limitation at t > 15 °C, but that catalytic steps become definitively slower below 15 °C. Model kinetics are derived to show that this change in mechanism at low temperature can force an ordered substrate binding system to produce exchange-rate patterns consistent with a random binding system with all exchange rates equal. The nonlinear Arrhenius plot also has important consequences for current theories of catalytic and regulatory mechanisms for this enzyme.  相似文献

Land surface temperature (LST) is crucial in surface energy balance, urban climatology, intensifying global change, ecological and environmental concerns. The present study examined the LST trends and spatio-temporal variation over India from 2002 to 2022. This includes comparison of LST for the summer and winter seasons over two decades. Secondly, the present study examined the LULC category wise LST variability during the day and night-time using MODIS (The Terra Moderate Resolution Imaging Spectroradiometer) derived products. This study explored the feasibility of cloud computing for big data analysis for LST distribution in seven landuse categories over India, providing a conceptual response to global warming. Results showed the existence of spatial LST variation due to changes in land-use patterns and MODIS derived vegetation indices- NDVI. Daytime LST for the summer and winter seasons of 2002 was found to be 45.17 °C and 39.13 °C, respectively. Outcomes illustrate declining trends in range (LSTmin-LSTmax) for winter seasons, initially, it was observed as 56.29 °C for 2002 while later on it was observed to be 20.21 °C and 20.87 °C for 2021 and 2022 respectively. The LSTmin (summer) has shown an increasing trend towards upper LST values, from −3.01 °C to 12.21 °C from 2002 TO 2022. LSTmin_winter has shown a rising trend towards the upper LST values from −17.16 °C to 9.15 °C in 2022. The maximum LST for the DRs was observed to be 61.56 °C, followed by UR as 56.24 °C. The findings demonstrate that daytime LSTmin and LSTmax are found to be 19.50 °C to 56.24 °C for UR, 29.5 °C to 61.56 °C for DR, 19.24 °C to 54.08 °C for SAR, −21.1 °C to 0.05 °C for SCR and 15 °C to 32.18 °C for FHR. NDVI-LST (daytime, nighttime and diurnal temperature range) feature space generates an obtuse triangle and depicts a negative correlation of vegetation for a few LULC categories. The outcomes indicated that desert and snow regions have highest LSTmax followed by urban and semiarid regions during daytime. During the nighttime, desert and urban regions have the highest temperature followed by semi-arid and forest regions. The outcomes support the efficacy of earth observation datasets and help to facilitate a better understanding of LULC and its impact on regional climate.  相似文献

  • 2.2. In non-panting Acc birds, brain temperature gradually increased from 40.1 ± 0.4°C at 30°C to 41.2 ± 0.4°C at 60°C Ta. A mean body temperature (Tb) of 41.2 ± 0.2°C was measured at Ta up to 50°C; an increase of 1.1°C was observed at 60°C (Tb 42.2 ±0.6°C).
  • 3.3. In Acc panting birds exposed for 2 hr to 60°C, Thy was 41.9 ± 0.8°C and Ts was somewhat (but insignificantly) higher, i.e., 42.2 ± 0.7°C. It looks as if both values were increased as a result of a slight hyperthermia that developed (Tb = 43.5 ± 0.9°C).

Development of the wild type and two temperature-sensitive mutants of the multicellular green alga Ulva mutabilis is compared. The mutants develop normal phenotypes at 22°C and abnormal phenotypes at 15°C. Normal development starts by formation of a filament consisting of a row of cells. The growth rate, the generation times, and the cell length at division change in a coordinated manner according to the positions of the cells within the filament. In the mutant cs2 transfer to 15°C inhibits all cytoplasmic divisions during early development. In the mutant cs6 the first three divisions proceed normally. Then cytoplasmic division is blocked in the most distal cells, while the proximal cells continue to divide according to a branched pattern. In the cs2 mutant cell determination seems to occur at 15°C, while the differentiation of the determined cells can only occur at 22°C. In the mutant cs6 the cells are not determined at 15°C. The cs6+ gene, as well as the previously described Slender-like genes, presumably has a short period of activity and is concerned with more fundamental epigenetic processes than the cs2+-gene and the previously described precocious-like genes, which seem to have more prolonged periods of activity.  相似文献

Much interest exists in the extent to which constant versus fluctuating temperatures affect thermal performance traits and their phenotypic plasticity. Theory suggests that effects should vary with temperature, being especially pronounced at more extreme low (because of thermal respite) and high (because of Jensen's inequality) temperatures. Here we tested this idea by examining the effects of constant temperatures (10 to 30 °C in 5 °C increments) and fluctuating temperatures (means equal to the constant temperatures, but with fluctuations of ±5 °C) temperatures on the adult (F2) phenotypic plasticity of three thermal performance traits – critical thermal minimum (CTmin), critical thermal maximum (CTmax), and upper lethal temperature (ULT50) in ten species of springtails (Collembola) from three families (Isotomidae 7 spp.; Entomobryidae 2 spp.; Onychiuridae 1 sp.). The lowest mean CTmin value recorded here was -3.56 ± 1.0 °C for Paristoma notabilis and the highest mean CTmax was 43.1 ± 0.8 °C for Hemisotoma thermophila. The Acclimation Response Ratio for CTmin was on average 0.12 °C/°C (range: 0.04 to 0.21 °C/°C), but was much lower for CTmax (mean: 0.017 °C/°C, range: -0.015 to 0.047 °C/°C) and lower also for ULT50 (mean: 0.05 °C/°C, range: -0.007 to 0.14 °C/°C). Fluctuating versus constant temperatures typically had little effect on adult phenotypic plasticity, with effect sizes either no different from zero, or inconsistent in the direction of difference. Previous work assessing adult phenotypic plasticity of these thermal performance traits across a range of constant temperatures can thus be applied to a broader range of circumstances in springtails.  相似文献

The complex permittivity of sonicated aqueous solutions of purified dimyristoylphosphatidylcholine has been measured as a function of frequency between 3 kHz and 40 GHz. The dielectric spectrum of the samples shows two dispersion/absorption regions, one centered at about 80 MHz the other at about 20.GHz (30°C). Otherwise than in previous studies no additional dispersion/absorption process has been found at frequencies below 10 MHz.The complex dielectric spectrum of the samples is discussed with respect to the dynamical state of solvent water in solutions of single-bilayer vesicles. The main relaxation time of the solvent water, τ1 ((2πτ1)?1 ≈ 20 GHz), is smaller than that of pure water, τW, at the same temperature. This effect results from the action of internal depolarizing fields which obviously overcompensate and enhancement of τ1 due to specific solute/solvent interactions (hydration) as had been previously found with micellar solutions of lysolecithins.It cannot be excluded, that some solvent water shows unusual dynamical behaviour. If there exists a substantial amount of such motionally perturbed water, however, it must be characterized by a relaxation time close to that of the phosphorylcholine zwitterions, τ2 ((2πτ2)?1 ≈ 80 MHz).  相似文献

Environmental conditions play a major role in shaping reptilian embryonic development, but studies addressing the impact of interactions between chronic and acute environmental stressors on embryonic systems are lacking. In the present study, we investigated thermal dependence of cholinergic and adrenergic cardiovascular tone in embryonic American alligators (Alligator mississippiensis) and assessed possible phenotypic plasticity in a chronic hypoxic incubation treatment. We compared changes in heart rate (f H) and mean arterial blood pressure (P M) for chronically hypoxic and normoxic-incubated embryos after cholinergic and adrenergic blockade following three different acute temperature treatments: (1) 30 °C (control incubation temperature), (2) acute, progressive decrease 30–24 °C then held at 24 °C, and (3) acute, progressive increase 30–36 °C then held at 36 °C. f H progressively fell in response to decreasing temperature and rose in response to increasing temperature. P M did not significantly change with decreasing temperature, but was lowered significantly with increasing acute temperature in the normoxic group at 90 % of development only. Propranolol administration (β adrenergic antagonist) produced a significant f H decrease at 24, 30, and 36 °C that was similar at all temperatures for all groups. For normoxic-incubated embryos at 90 % of development, atropine administration (cholinergic antagonist) significantly increased f H in both 24 and 36 °C treatments, but not in the 30 °C control treatment. This atropine response at 24 and 36 °C demonstrated acute thermally dependent cholinergic tone on f H late in development for normoxic-incubated, but not chronically hypoxic-incubated embryos. Collectively, data indicated that cardiovascular control mechanisms in embryonic alligators may be activated by thermal extremes, and the maturation of control mechanisms was delayed by chronic hypoxia.  相似文献

The action of phospholipase C (Bacillus cereus) toward mixed micelles of phosphatidylcholine and the nonionic surfactant Triton X-100 is analyzed according to the “surfaceas-cofactor” kinetic scheme recently proposed for characterizing the action of lipolytic enzymes [Deems, R. A., Eaton, B. R., and Dennis, E. A. (1975) J. Biol. Chem.250, 9013–9020]. According to this scheme, the enzyme first associates with the surface or mixed micelles, where the dissociation constant is KsA. The enzyme, now part of the mixed micelle surface, then binds the substrate phospholipid molecule in its active site and this binding is related to the Michaelis constant, KmB. The surface, or mixed micelles in this scheme, behaves kinetically as a cofactor in that, under initial rate conditions, the surface properties of the mixed micelles are virtually unchanged after catalysis. For phospholipase C with egg phosphatidylcholine as substrate, it was found that at pH 6.4 (the pH optimum for the enzyme) and 40 °C, V is about 2 × 103 μmol min?1 (mg of protein)?1. KsA is about 2 mm and KmB is 1 to 2 × 10?10 mol cm?2. The kinetic constants for phospholipase C are compared with those previously reported for phospholipase A2 and the membrane-bound enzyme phosphatidylserine decarboxylase determined under similar conditions.  相似文献

The effect of various physico-chemical factors on production of intra- and extracellular phospholipase A1 bySalmonella newport was investigated. Maximum intracellular enzyme levels were observed when cells were grown in brain heart infusion broth, after 12 h of incubation at 37°C. Highest level of extracellular phospholipase A1, however, was seen in synthetic medium (pH 7.0) after 24 h of incubation at 37°C. Agitation during incubation had no effect on the intracellular enzyme synthesis but enhanced extracellular enzyme levels. Addition of surfactants to the growth media significantly decreased both intra- and extracellular phospholipase A1 production.  相似文献

Organisms inhabiting small water bodies frequently encounter radical, short-term fluctuations in temperature. The population of Brachionus plicatilis that was used in this study encounters temperature changes as great as 12°C in 24 hours. In this paper we describe the metabolic response to temperature change in this eurythermal rotifer population. Metabolism was determined over the environmental range (15°C to 32°C) in intervals of 2°C. Utilization of such narrow temperature intervals allowed us to approximate the instantaneous Q10, which we define as the Q10 over an infinitely small temperature interval. Our results show the existence of a plateau in the curve of respiration against temperature (Q10 > 1) over the range 20–28°C. The plateau is bounded on either side by temperature ranges over which metabolism is temperature sensitive (Q10 values from 3.4 to 4.8). It is significant that the plateau occurs over the environmental temperature range for the major portion of the growing season. This population is thus programmed to hold a constant (preferred?) metabolic rate in spite of diel temperature fluctuations when the environment is otherwise favorable.  相似文献

Temperature responses of the cockroach, Blaberus craniifer, to rapid changes of ambient temperature (Ta) have been studied. In static conditions at Ta = 27°C the body-to-ambient temperature difference was only 0.10 ± 0.07°C. Two test situations were used, either a ramp increase of Ta from 27 to 31°C (0.1°C/min) or “step” changes from 27 to 28°C and back (0.5°C/min). In both cases body temperature closely followed Newtonian model, the body time constants measured in various conditions being very similar: 543 ± 99 sec in ramp tests, 550 ± 68 sec and 542 ± 124 sec in rising and falling step tests respectively. It is concluded that in spite of evident differences between the cockroach and an inert solid, the Newtonian model adequately represents the thermal responses of this insect to moderate changes in ambient temperature.  相似文献

At 37 °C, when samples of blood, washed erythrocytes, or isolated hemoglobin from individuals with sickle cell disease are deoxygenated, the transverse water proton relaxation time is sharply decreased. In similar samples from normal adults homozygous for hemoglobin A, only a slight decrease in t2 is observed upon deoxygenation at 37 °C. In samples containing deoxyhemoglobin S the value of t2 increases as the temperature is decreased from 37 °C to 4 °C, in contrast to samples containing oxyhemoglobin S, oxyhemoglobin A, or deoxyhemoglobin A where t2 decreases as the temperature decreases. It is suggested that this decrease in t2 observed in samples of deoxyhemoglobin S at 37 °C is the result of an increase in the amount of preferentially oriented water at macromolecular interfaces which occurs under conditions known to produce deoxyhemoglobin S gelation. Conditions which reverse deoxyhemoglobin S gelation such as lowering the temperature to 4 °C decrease the amount of preferentially oriented water which results in an increase in the value of t2. Thus, measurement of the transverse water proton relaxation time can be used to monitor the gelation of deoxyhemoglobin S inside the erythrocyte.  相似文献

The kinetics of the photoreduction of C-550, the photooxidation of cytochrome b559 and the fluorescence yield changes during irradiation of chloroplasts at ?196 °C were measured and compared. The photoreduction of C-550 proceeded more rapidly than the photooxidation of cytochrome b559 and the fluorescence yield increase followed the cytochrome b559 oxidation. These results suggest that fluorescence yield under these conditions indicates the dark reduction of the primary electron donor to Photosystem II, P680+, by cytochrome b559 rather than the photoreduction of the primary electron acceptor.The photoreduction of C-550 showed little if any temperature dependence over the range of ?196 to ?100 °C. The amount of cytochrome b559 photooxidized was sensitive to temperature decreasing from the maximal change at temperatures between ?196 to ?160 °C to no change at ?100 °C. To the extent that the reaction occurred at temperatures between ?160 and ?100 °C the rate was largely independent of temperature. The rate of the fluorescence increase was dependent on temperature over this range being 3–4 times more rapid at ?100 than at ?160 °C. At ?100 °C the light-induced fluorescence increase and the photoreduction of C-550 show similar kinetics. The temperature dependence of the fluorescence induction curve is attributed to the temperature dependence of the dark reduction of P680+.The intensity dependence of the photoreduction of C-550 and of the photooxidation of cytochrome b559 are linear at low intensities (below 200 μW/cm2) but fall off at higher intensities. The failure of reciprocity in the photoreduction of C-550 at the higher intensities is not explained by the simple model proposed for the Photosystem II reaction centers.  相似文献
