We studied the effect of herbicide and nitrogen supply on photosynthesis in Perilla frutescens L. Britt. Plants were exposed to combined treatment of urea and herbicide, fenoxaprop-P-ethyl (FPE), in various concentrations. FPE reduced significantly chlorophyll (Chl) content, photosynthetic rate, and stomatal conductance, but increased significantly intercellular CO2 concentration; thus, FPE inhibited significantly the photosynthetic capacity. In addition, FPE also decreased significantly the PSII photochemical efficiency, effective quantum yield of photochemical energy conversion in PSII, PSII potential activity, and photochemical quenching of variable Chl fluorescence. It also decreased nonphotochemical quenching. It indicated that FPE impaired PSII and blocked the electron transport in light reaction. The urea treatment at moderate concentration (1–4 g L?1) could antagonize the negative effect of FPE, while the high urea concentration (8 g L?1) aggravated this effect. The treatment with urea (4 g L?1) and then with FPE (1.33 mL L?1) enhanced Chl content index, photosynthetic rate, and stomatal conductance by 12.5, 36.1, and 28.5% compared to FPE treatment alone. Thus, we suggested to treat plants first with urea (4 g L?1) and then by FPE (1.33 mL L?1) as the best and the safest method to balance the fertilization and weeding.  相似文献

The response of tomato plants to various chilling treatments was studied using two approaches for the measurement of photosynthetic activity. One involved the use of a portable fluorometer for the measurement of in-vivo chlorophyll fluorescence, while the other employed a newly introduced photoacoustic system which allowed changes in oxygen evolution to be followed in a leaf disc. A strong correlation was found between results obtained by each system and those obtained by a conventional open gas-exchange system for the determination of CO2 uptake. Both systems of measurements could readily distinguish between the effects of chilling in the dark (at 3° C for 18 h) and chilling at high photon flux density (2000 mol m-2 s-1 for 5h at 5° C). Chilling in the dark had practically no effect on the quantum yield of oxygen evolution, chlorophyll fluorescence or CO2 uptake, while chilling at excessively high photon flux density resulted in a sharp reduction (50–70%) in the quantum yields obtained. Brassinosteroids (BRs), an important class of plant steroidal hormones, play a significant role in the amelioration of various biotic and abiotic stresses. 24-epibrassinolide (EBR), an active brassinosteroid, was applied exogenously in different concentrations to characterize a role of BRs in tolerance of melon (Cucumis melo L.) to high temperature (HT) stress and to investigate photosynthetic performance of HT-stressed, Honglvzaocui (HT-tolerant) and Baiyuxiang (HTsensitive), melon variety. Under HT, Honglvzaocui showed higher biomass accumulation and a lower index of heat injury compared with the Baiyuxiang. The exogenous application of 1.0 mg L?1 EBR, the most effective concentration, alleviated dramatically the growth suppression caused by HT in both ecotypes. Similarly, EBR pretreatment of HTstressed plants attenuated the decrease in relative chlorophyll content, net photosynthetic rate, stomatal conductance, stomatal limitation, and water-use efficiency (WUE), as well as the maximal quantum yield of PSII photochemistry (Fv/Fm), the efficiency of excitation capture of open PSII center, the effective quantum yield of PSII photochemistry (ΦPSII), photochemical quenching coefficient, and the photon activity distribution coefficients of PSI (α). EBR pretreatment further inhibited the increase in intracellular CO2 concentration, leaf transpiration rate, minimal fluorescence of dark-adapted state, nonphotochemical quenching, thermal dissipation, and photon activity distribution coefficients of PSII. Results obtained here demonstrated that EBR could alleviate the detrimental effects of HT on the plant growth by improving photosynthesis in leaves, mainly reflected as up-regulation of photosynthetic pigment contents and photochemical activity associated with PSI.  相似文献

Photosystem (PS) II is the multisubunit complex which uses light energy to split water, providing the reducing equivalents needed for photosynthesis. The complex is susceptible to damage from environmental stresses such as excess excitation energy and high temperature. This research investigated the in vivo photosynthetic consequences of impairments to PSII in Arabidopsis thaliana (ecotype Columbia) expressing an antisense construct to the PsbO proteins of PSII. Transgenic lines were obtained with between 25 and 60% of wild-type (WT) total PsbO protein content, with the PsbO1 isoform being more strongly reduced than PsbO2. These changes coincided with a decrease in functional PSII content. Low PsbO (less than 50% WT) plants grew more slowly and had lower chlorophyll content per leaf area. There was no change in content per unit area of cytochrome b(6)f, ATP synthase, or Rubisco, whereas PSI decreased in proportion to the reduction in chlorophyll content. The irradiance response of photosynthetic oxygen evolution showed that low PsbO plants had a reduced quantum yield, but matched the oxygen evolution rates of WT plants at saturating irradiance. It is suggested that these plants had a smaller pool of PSII centres, which are inefficiently connected to antenna pigments resulting in reduced photochemical efficiency.  相似文献

It is known, that the multi-subunit complex of photosystem II (PSII) and some of its single proteins exhibit carbonic anhydrase activity. Previously, we have shown that PSII depletion of HCO3?/CO2 as well as the suppression of carbonic anhydrase activity of PSII by a known inhibitor of α?carbonic anhydrases, acetazolamide (AZM), was accompanied by a decrease of electron transport rate on the PSII donor side. It was concluded that carbonic anhydrase activity was required for maximum photosynthetic activity of PSII but it was not excluded that AZM may have two independent mechanisms of action on PSII: specific and nonspecific. To investigate directly the specific influence of carbonic anhydrase inhibition on the photosynthetic activity in PSII we used another known inhibitor of α?carbonic anhydrase, trifluoromethanesulfonamide (TFMSA), which molecular structure and physicochemical properties are quite different from those of AZM. In this work, we show for the first time that TFMSA inhibits PSII carbonic anhydrase activity and decreases rates of both the photo-induced changes of chlorophyll fluorescence yield and the photosynthetic oxygen evolution. The inhibitory effect of TFMSA on PSII photosynthetic activity was revealed only in the medium depleted of HCO3?/CO2. Addition of exogenous HCO3? or PSII electron donors led to disappearance of the TFMSA inhibitory effect on the electron transport in PSII, indicating that TFMSA inhibition site was located on the PSII donor side. These results show the specificity of TFMSA action on carbonic anhydrase and photosynthetic activities of PSII. In this work, we discuss the necessity of carbonic anhydrase activity for the maximum effectiveness of electron transport on the donor side of PSII.  相似文献

The crop sensitivity to ozone (O3) is affected by the timing of the O3 exposure, by the O3 concentration, and by the crop age. To determine the physiological response to the acute ozone stress, tomato plants were exposed to O3 at two growth stages. In Experiment I (Exp. I), O3 (500 μg m?3) was applied to 30-d-old plants (PL30). In Experiment II (Exp. II), three O3 concentrations (200, 350, and 500 μg m?3) were applied to 51-d-old plants (PL51). The time of the treatment was 4 h (7:30–11:30 h). Photosynthesis and chlorophyll fluorescence measurements were done 4 times (before the exposure; 20 min, 20 h, and 2–3 weeks after the end of the treatment) using a LI-COR 6400 photosynthesis meter. The stomatal pore area and stomatal conductance were reduced as the O3 concentration increased. Ozone induced the decrease in the photosynthetic parameters of tomato regardless of the plant age. Both the photosystem (PS) II operating efficiency and the maximum quantum efficiency of PSII photochemistry declined under the ozone stress suggesting that the PSII activity was inhibited by O3. The impaired PSII contributed to the reduced photosynthetic rate. The greater decline of photosynthetic parameters was found in the PL30 compared with the PL51. It proved the age-dependent ozone sensitivity of tomato, where the younger plants were more vulnerable. Ozone caused the degradation of photosynthetic apparatus, which affected the photosynthesis of tomato plants depending on the growth stage and the O3 concentration.  相似文献

Three greenhouse- and four field-ecotype varieties of domestic tomato (Lycopersicon esculentum) were compared for the sensitivity of their photosynthetic apparatus to chilling under low light intensity. After chilling at 12/7 degrees C under 100 micromolm(-2)s(-1) of light for 10 days, they were allowed to recover at 25/18 degrees C and 600 micromolm(-2)s(-1) of light for 10 days. For both pre-chilling and recovered plants, greenhouse-ecotype varieties did not necessarily show higher net CO(2) assimilation rate (A), quantum yield of electron transport at PSII (Phi(PSII)) and photochemical quenching (q(P)) than field-ecotype varieties. However for the post-chilling period, greenhouse-ecotype varieties, exhibited higher A, and Phi(PSII) values than field-ecotype varieties. The difference in Phi(PSII) was found to be largely due to q(P). The absence of ecotypic differences in pre-chilling plants indicates that the trait was not expressed constitutively, but relied mainly on adaptation/acclimation mechanisms. Greenhouse-ecotype varieties were able to adapt to low temperature and low light more quickly, and then exhibited higher A, Phi(PSII), q(P) values and greater re-growth capacity after chilling than field-ecotype varieties. Plant re-growth capacity after chilling was highly correlated with Phi(PSII) and q(P) measured in chilled plants, suggesting the usefulness of Phi(PSII) and q(P) measured at low temperature after defined chilling stresses as screening indexes for chilling tolerance in breeding programs.  相似文献

The after effects of a short exposure of intact barley leaves to moderately elevated temperature (40°C, 5 min) on the induction transients and the irradiance dependencies of photosynthesis and chlorophyll fluorescence are presented. This mild heat treatment strongly reduced the oscillations in the rate of photosynthesis and in the yield of chlorophyll fluorescence. However, only a 25% irreversible inhibition of maximum photosynthetic capacity of photosystem II (PSII) measured by oxygen evolution was produced and the intrinsic quantum yield of PSII measured by the chlorophyll fluorescence ratio (Fm‐ Fo)/Fm decreased by only 15%. In contrast, the above treatment increased radiationless dissipation processes in PSII by a factor of two. In heat‐treated leaves, photosynthesis was not saturated even by strong light. Both ΔpH‐dependent quenching of excitons in PSII (including formation of zeaxanthin) and state 1/state 2 transition were found to be stimulated. Heat exposure enhanced the control of PSII activity by PSI, as evidenced by a significant increase in the quenching effect of far‐red light on the maximum yield of chlorophyll fluorescence. It was deduced that after mild heat treatment, the photosynthetic apparatus in leaves lacks the precise coordinating control of electron transport and carbon metabolism owing to the inability of PSII to support electron transport at a level adequate for carbon metabolism. This effect was not related to the small irreversible thermal damage to PSII, but was rather due to a significant increase in non‐photochemical quenching of excitation energy.  相似文献

三磷酸腺苷(ATP)不但分布在细胞内部, 而且广泛存在于动物和植物细胞的细胞外基质中。细胞外ATP (eATP)可与细胞膜表面相应的受体结合并激发细胞内的第二信使, 从而调节细胞的多种生理学功能。但目前对于eATP是否也能对植物的光合作用产生影响则研究较少。该文以菜豆(Phaseolus vulgaris)叶片为实验材料, 研究了在不同光强下eATP对菜豆叶片叶绿素荧光特性和光合放氧速率的影响。结果显示, 随着光强的增加, 叶片的光适应下最大光化学效率(Fv′/Fm′)、光系统II (PSII)实际光化学效率(Y(II))、光化学猝灭系数(qP)均呈现下降趋势, 而电子传递速率(ETR)、非光化学猝灭系数(qN)以及调节性能量耗散的量子产量(Y(NPQ))随着光强的增加呈上升趋势。与对照相比, eATP的处理可以显著提高菜豆叶片PSII的潜在最大光化学效率(Fv/Fm)、Y(II)、qP、ETR和光合放氧速率; 但eATP的处理对Fv′/Fm′、qN以及Y(NPQ)没有显著影响。AMP-PCP (β,γ-亚甲基三磷酸腺苷, eATP细胞外受体的抑制剂)的处理显著降低了Fv/FmFv′/Fm′、Y(II)、ETR和光合放氧速率, 同时也显著增加了qN以及Y(NPQ)的水平。以上结果显示, 植物eATP水平的变化对植物光合作用的光化学反应有着重要的影响。  相似文献

以"丹麦旺盛菠菜"为材料,通过UV-B和CaCl2复合处理,测定光合色素含量、Hill反应活力、叶绿素荧光、MDA含量和抗氧化酶活性等参数,探讨了CaCl2对UV-B辐射下菠菜叶片电子传递链和光合膜酶保护系统的影响。结果表明,UV-B处理下,光合色素含量、chl/car、类囊体膜上PSII潜在活性(Fv/Fo)、光化学淬灭系数(qP)、非光化学淬灭系数(qN)、PSII光量子产量(ΦPSⅡ)、原初光能转化效率(Fv/Fm),以及Hill反应活力等降低,chla/chlb和MDA含量升高;喷洒CaCl2可不同程度缓解UV-B的伤害。不同处理下,POD、SOD和CAT活性的变化呈现补偿效应。UV-B强度与菠菜叶片PSII功能受损程度呈正相关,CaCl2则主要通过提高chlb含量、类囊体膜上的光量子产量和POD活性,以缓解伤害。重度UV-B辐射下,CaCl2使chlb含量显著提高可能是导致PSII捕光效率提高的重要因素。  相似文献

The effects of chilling (CT, day/night temperatures of 12/10 °C, an irradiance of 250 μmol m?2 s?1), chilling combined with a low irradiance (CL, 12/10 °C, 80 μmol m?2 s?1), and a high temperature (HT, 42/40 °C, 250 μmol m?2 s?1) on chlorophyll content, chlorophyll fluorescence, and gas exchange were studied in two watermelon cultivars, ZJ8424 and YS01, differing in their resistance. The chlorophyll content, net photosynthetic rate (PN), stomatal conductance (gs), and transpiration rate (E) decreased substantially, whereas the intercellular CO2 concentration (ci) increased when the two watermelon cultivars were grown under these stresses. The photosynthetic parameters showed greater changes at chilling than at the high temperature, and the CL caused a more pronounced inhibition in PN compared with the CT. After 2 d exposure to the CT, YS01 had higher PN, gs, and E, but a lower ci compared with ZJ8424. The maximum efficiency of photosystem (PS) II photochemistry (Fv/Fm), effective quantum yield of PS II photochemistry (ΦPSII), photochemical quenching (qP), and electron transport rate (ETR) decreased under the CT and CL but showed only a slight drop under the HT. All these stresses significantly increased non-photochemical quenching (NPQ). The CT brought more damage to the photosynthetic apparatus of leaves compared with the CL. In addition, after returning to normal conditions (25/15 °C, 250 μmol m?2 s?1) for 3 d, the photosynthetic parameters recovered to pre-stress levels in HT treated seedlings but not in CT treated seedlings. In conclusion, the low irradiance could help to alleviate the extent of photoinhibition of PS II photochemistry caused by chilling and cv. ZJ8424 was more sensitive to the extreme temperatures than cv. YS01.  相似文献

The effects of long term exposure to suboptimal growth temperature on the photosynthetic apparatus of Dunaliella tertiolecta Butcher were investigated using carbon fixation rate versus irradiance curves and the variable fluorescence induction method. Carbon fixation rates per unite chlorophyll a at saturating (pBm) and subsaturating (αB) irradiances were 55% and 39% lower, respectively, at 12° C than at 20° C. Chlorophyll a quotas and the spectrally averaged in vivo absorption cross section normalized to chlorophyll a (a*) were not significantly different at these two temperatures. Analysis of the fluorescence kinetics revealed 1) no significant variations of the amount of PSII photoactive reaction centers per unit chlorophyll a, 2) a 14% decrease of the PSII quantum yield(+) and 3) a 29% decrease of the energy transfer efficiency between the light harvesting chlorophyll a pigment bed and the PSII reaction centers. The decrease in energy transfer efficiency between the antennae and the PSII reaction centers at 12° C was interpreted as a mechanism to avoid photoinhibition.  相似文献

The methods of chlorophyll fluorescence induction and HPLC were used to study the influence of autumnal temperature decrease on photochemical electron-transport activity of photosystem II (PSII), nonphotochemical quenching of excessive excitation energy, and the composition of pigments in the firstyear needles of Pinus sуlvestris L. trees grown naturally in Central Yakutia. In the period from the beginning of September to October 10, the chlorophyll content was reduced by half, while the Chl a/b ratio increased from 2.9 to 4.3–4.5, indicating the degradation of peripheral antenna complexes. The decrease in average daily temperature to 4.9–6.4°C led to a transient increase in the quantum yield of nonphotochemical quenching (ΔpH-dependent parameter φNPQ). These changes were accompanied by a slow accumulation of unregulated zeaxanthin fraction insensitive to illumination conditions. The further decrease in average daily temperature to near-zero levels was paralleled by a sharp increase in zeaxanthin content, while the pH-dependent quenching was replaced with the constitutive quenching (parameter φf,D) because of the supposed structural reorganization of PSII. These processes were accompanied by a fast decrease in PSII functional activity, which was mostly due to the impairment of plastoquinone photochemical reduction. Freezing temperatures (from–3.6 to–12.1) destructed the oxygen-evolving complex in PSII and completely inactivated the PSII reaction centers. It is concluded that the largest changes in the condition of photosynthetic apparatus occur at a near-zero temperature range and proceed until complete inactivation of PSII under the action of freezing temperatures.  相似文献

The characteristics of photosynthetic gas exchange, chlorophyll a fluorescence, and xanthophyll cycle pigments during flag leaf senescence of field-grown wheat plants were investigated. With senescence progressing, the light-saturated net CO2 assimilation rate expressed either on a basis of leaf area or chlorophyll decreased significantly. The apparent quantum yield of net photosynthesis decreased when expressed on a leaf area basis but increased when expressed on a chlorophyll basis. The maximal efficiency of PSII photochemistry decreased very little while actual PSII efficiency, photochemical quenching, and the efficiency of excitation capture by open PSII centers decreased considerably. At the same time, non-photochemical quenching increased significantly. A substantial decrease in the contents of violaxanthin and zeaxanthin, but a slight decrease in the content of antheraxanthin were observed. However, the de-epoxidation status of the xanthophyll cycle was positively correlated with progressive senescence. This increase was due mainly to a smaller decrease in zeaxanthin than in violaxanthin. Our results suggest that PSII apparatus remained functional, but a down-regulation of PSII occurred under the steady state of photosynthesis in senescent flag leaves. Such a down-regulation was associated with the closure of PSII centers and an enhanced xanthophyll cycle-related thermal dissipation in the PSII antennae.  相似文献

Chlorophyll fluorescence induction curves were used as a means to assess the functional condition of the photosynthetic apparatus in cells of the halotolerant green microalga Dunaliella maritima (Massjuk) (division Chlorophyta) exposed to hyperosmotic salt shock of various intensities. The shock was caused by the transfer of algal cells grown in the medium with 0.5 M NaCl to the media with elevated NaCl concentrations (1.0, 1.5, and 2.0 M). Parameters of chlorophyll fluorescence (F 0, F m, F 0′, F t′) were measured by means of a specialized pulse-amplitude-modulation fluorometer PAM 2100. In addition, the rate of photosynthetic oxygen evolution as well as the intracellular Na+ and glycerol content (the main osmolyte in this microalga) were determined. The hyperosmotic salt shock was found to elevate the intracellular Na+ content and reduce the functional activity of PSII in D. maritima. The suppression of PSII activity was evident from the decrease in the maximal quantum yield of photochemical energy conversion in PSII, the decreased rate of linear electron transport, the increased reduction of the primary acceptor QA, and the suppression of photosynthetic O2 evolution. The functional activity of PSII recovered gradually along with restoration of osmotic and ionic balance in algal cells. It is proposed that PSI ensures energy supply during cell responses of D. maritima to hyperosmotic salt shock.  相似文献

In this study we investigated the basis for the reduction in the quantum yield of carbon assimilation in maize (Zea mays L. cv. LG11) caused by chilling in high light. After chilling attached maize leaves at 5° C for 6 h at high irradiance (1000 mol photons·m–2·s–1) chlorophyll fluorescence measurements indicated a serious effect on the efficiency of photochemical conversion by photosystem II (PSII) and measurements of [14C]atrazine binding showed that the plastoquinone binding site was altered in more than half of the PSII reaction centres. Although there were no direct effects of the chilling treatment on coupling-factor activity, ATP-formation capacity was affected because the photoinhibition of PSII led to a reduced capacity to energize the thylakoid membranes. In contrast to chilling at high irradiance, no photoinhibition of PSII accompanied the 20% decrease in the quantum yield of carbon assimilation when attached maize leaves were chilled in low light (50 mol photons·m–2·s–1). Thus it is clear that photoinhibition of PSII is not the sole cause of the light-dependent, chillinduced decrease in the quantum yield of carbon assimilation. During the recovery of photosynthesis from the chilling treatment it was observed that full [14C]atrazinebinding capacity and membrane-energization capacity recovered significantly more slowly than the quantum yield of carbon assimilation. To investigate whether brassinosteroids (BRs) could be used to alleviate chill-induced inhibition of photosynthesis in cucumber (Cucumis sativus L) during chilling and subsequent recovery, the effects of exogenously applied 24-epibrassinolide (EBR) on gas exchange, chlorophyll fluorescence parameters, and antioxidant enzyme activity were studied. Cucumber plants were exposed to chilling under low light (12/8°C and 100 μmol m−2 s−1 PPFD) for 3 days and then recovered under normal temperature and high irradiance (28/18°C and 600 μmol m−2 s−1 PPFD) for 6 days. Chilling significantly decreased the net photosynthetic rate (P N) and stomatal conductance (g s), and increased rate of O2 ·− formation and H2O2 and malondialdehyde (MDA) content in cucumber leaves, but did not influence the optimal quantum yield of PSII (Fv/Fm). Chilling also decreased the effective quantum yield of PSII photochemistry (ΦPSII) and photochemical quenching (qP), but induced an increase in nonphotochemical quenching (NPQ), and the activities of superoxide dismutase (SOD) and ascorbate peroxidase (APX). High irradiance (600 μmol m−2 s−1) further aggravated the decrease in P N, g s, ΦPSII and qP, and enhanced the increase in reactive oxygen species (ROS) generation and accumulation in the first day of recovery after chilling. However, high irradiance induced a sharp decrease in Fv/Fm and NPQ, as well as the activities of SOD and APX on the first day of recovery. EBR pretreatment significantly alleviated chill-induced inhibition of photosynthesis during chilling stress and subsequent recovery period, which was mainly due to significant increases in g s, ΦPSII, qP and NPQ. EBR pretreatment also reduced ROS generation and accumulation, and increased the activities of SOD and APX during chilling and subsequent recovery. Those results suggest that EBR pretreatment alleviates the chill reduction in photosynthesis and accelerated the recovery rate mainly by increasing of the stomatal conductance, the efficiency of utilization and dissipation of leaf absorbed light, and the activity of the ROS scavenging system during chilling and subsequent recovery period.  相似文献

When exposed to light, the cells of characean algae produce intermittent regions of H+ extrusion and H+ absorption, featuring different photosynthetic activities. Methods for local measurements of outer pH, O2 content, and photochemical activity of photosystem II (PSII) were applied to examine microscopic regions of Chara coralline Klein ex Willd. internodes. The results show that the functional spatial heterogeneity of these excitable cells is controlled not only by light but also by electric excitation of the plasma membrane. Generation of a single action potential (AP) induced a reversible transition to the state with homogenous pH distribution and had different effects on photosynthesis in cell regions producing alkaline and acid zones. The effective quantum yield of PSII primary processes and the maximal chlorophyll fluorescence decreased after AP in the alkaline cell regions but were almost unaffected in the acidic cell regions. The suppression of photosynthesis after AP was also evident in the decrease of photosynthetic O2 evolution. The results provide evidence that electric signals arising at the plasmalemma are transmitted to the level of thylakoid membranes. The effects of electric excitation on fluorescence and the quantum yield of PSII photochemistry were best pronounced at low light intensities and low level of nonphotochemical quenching. The sensitivity of chlorophyll fluorescence in resting and excited cells to light intensity and protonophores indicates that the AP-induced fluorescence changes derive from the increase in pH gradient at the thylakoid membrane. The temporal elimination of alkaline zones and inhibition of photosynthesis apparently arise from parallel operational sequences that have a common initial stage. A possible role of cytosolic Ca2+ rise in the mechanism of photosynthesis suppression after electric excitation of the plasma membrane is discussed.  相似文献

Manganese (Mn) ions are essential for oxygen evolution activity in photoautotrophs. In this paper, we demonstrate the dynamic response of the photosynthetic apparatus to changes in Mn bioavailability in cyanobacteria. Cultures of the cyanobacterium Synechocystis PCC 6803 could grow on Mn concentrations as low as 100 nm without any observable effect on their physiology. Below this threshold, a decline in the photochemical activity of photosystem II (PSII) occurred, as evident by lower oxygen evolution rates, lower maximal photosynthetic yield of PSII values, and faster Q(A) reoxidation rates. In 77 K chlorophyll fluorescence spectroscopy, a peak at 682 nm was observed. After ruling out the contribution of phycobilisome and iron stress-induced IsiA proteins, this band was attributed to the accumulation of partially assembled PSII. Surprisingly, the increase in the 682-nm peak was paralleled by a decrease in the 720-nm peak, dominated by PSI fluorescence. The effect on PSI was confirmed by measurements of the P(700) photochemical activity. The loss of activity was the result of two processes: loss of PSI core proteins and changes in the organization of PSI complexes. Blue native-polyacrylamide gel electrophoresis analysis revealed a Mn limitation-dependent dissociation of PSI trimers into monomers. The sensitive range for changes in the organization of the photosynthetic apparatus overlaps with the range of Mn concentrations measured in natural environments. We suggest that the ability to manipulate PSI content and organization allows cyanobacteria to balance electron transport rates between the photosystems. At naturally occurring Mn concentrations, such a mechanism will provide important protection against light-induced damage.  相似文献
