The fatal autosomal recessive disease cystic fibrosis (CF) is caused by mutations in the gene which encodes the cystic fibrosis transmembrane conductance regulator (CFTR). Many of these disease-causing mutations, including the deletion of F508 (F508) which accounts for approximately 70% of the disease alleles, occur in one of the two consensus nucleotide binding sequences. Peptide studies have directly demonstrated that the N-terminal nucleotide binding sequences bind adenine nucleotides. Structurally, circular dichroism spectropolarimetry indicates that this region of CFTR assumes a -stranded structure in solution. The F508 mutation causes a diminution in the amount of -stranded structure and a concomitant increase in the amount of random coil structure present, indicating that either the mutant peptide has a different native structure or that the conformational equilibrium is shifted toward a more disordered form. Furthermore, the mutant peptide is more sensitive to denaturation, indicating that F508 is a stability, or protein-folding mutant. Here we review these results and discuss their implications for interpreting the behavior of F508in situ and for the rational design of new CF drugs.  相似文献

Summary Female freshwater eels injected with estradiol-17 (E2) for 15–78 days appear paler and secrete more mucus than controls. The resulting strongly opalescent blood plasma indicates that vitellogenin synthesis occurs in the liver, which shows a significant hypertrophy, an increased vacuolization (lipid material) and glycogen depletion. Plasma sodium is lowered, but calcium levels are considerably increased. The gonosomatic index increases (0.92±0.1 to a maximum of 2.21). Oocytes are enlarged, but the incorporation of vitellogenin remains discrete. Gonadotrophs (GTH cells), small and scarcely visible in the pituitary of control eels, are hypertrophied and contain numerous glycoprotein granules after E2-administration. E2 may act on the pituitary and/or hypothalamus via a positive feedback to induce gonadotrophin (GTH) synthesis; GTH release seems to be very limited as indicated by the ovarian response. The differentiation of GTH cells in eels treated with fish pituitary extracts is most probably due to secretion of E2 by the ovary, which reacts on the pituitary. Various hypotheses are considered to explain the low GTH release.Thyrotrophs, somatotrophs and prolactin cells of the pituitary are stimulated. In the pars intermedia, MSH and PAS-positive cells appear less active. A possible antidopaminergic effect of E2 is discussed.E2 administration constitutes a simple and economic technique to induce the synthesis of GTH and will facilitate the biochemical and biological study of the latter hormone in eels.  相似文献

Dark grown Scenedesmus obliquus strain PG1 accumulated -carotene and phytoene as its major carotenoids. On illumination of dark-grown cultures in air/CO2, cyclic carotenes and xanthophylls were formed, apparently at the expense of the accumulated phytoene and -carotene. This interconversion of carotenoids was accompanied by chlorophyll synthesis. In an atmosphere of nitrogen/CO2 the light-induced changes occurred more slowly and in nitrogen alone the changes were incomplete. No massive production of cyclic carotenes from the accumulated -carotene was observed in cultures illuminated under anaerobic conditions.  相似文献

Zusammenfassung Biloxi-Soja-Bohnen (Glycine max. L. Merr) erhielten photoperiodische Cyclen, die aus 8-oder 9stündigen Lichtperioden und unterschiedlich langen Dunkelperioden bestanden. Die Dunkelperioden wurden zu verschiedenen Zeitpunkten durch halbstündiges Störlicht unterbrochen. Die Wirkung dieser unterschiedlichen Behandlungen wurde durch Messung der Blühreaktionen bestimmt.Ein Störlicht, welches in frühen Abschnitten der langen Dunkelperiode geboten wird, verhindert die Blütenbildung unabhängig von der Länge des Gesamtcyclus, dem die Pflanzen ausgesetzt werden. Diese Unterdrückung erreicht ihr Maximum mit einem Störlicht, welches 8 Std nach dem Beginn der Dunkelperiode wirkt; die Hemmung ist schwächer, wenn das Störlicht früher geboten wird. Ein Störlicht, das gegen Ende der Dunkelperiode geboten wird, reduziert die Blütenbildung ebenfalls. Dieser Effekt erreicht sein Maximum mit einem Störlicht, welches ungefähr 8 Std vor dem Ende der Dunkelperiode geboten wird, er wird mit abnehmender Länge der nach dem Störlicht folgenden Dunkelperiode kleiner. Die Wirkung von Störlicht, das zu anderen Zeiten während der Dunkelperiode geboten wird, hängt von der Länge des Gesamtcyclus ab. Bei Cycluslängen, die für Blütenbildung ungünstig sind, kann solches Störlicht blühfördernd wirken.

Glomerular visceral epithelial cells (podocytes) contain interdigitated processes that form specialized intercellular junctions, termed slit diaphragms, which provide a selective filtration barrier in the renal glomerulus. Analyses of disease-causing mutations in familial nephrotic syndromes and targeted mutagenesis in mice have revealed critical roles of several proteins in the assembly of slit diaphragms. The nephrin–podocin complex is the main constituent of slit diaphragms. However, the molecular mechanisms regulating these proteins to maintain the slit diaphragms are still largely unknown. Here, we demonstrate that the PAR3–atypical protein kinase C (aPKC)–PAR6β cell polarity proteins co-localize to the slit diaphragms with nephrin. Furthermore, selective depletion of aPKCλ in mouse podocytes results in the disassembly of slit diaphragms, a disturbance in apico-basal cell polarity, and focal segmental glomerulosclerosis (FSGS). The aPKC–PAR3 complex associates with the nephrin–podocin complex in podocytes through direct interaction between PAR3 and nephrin, and the kinase activity of aPKC is required for the appropriate distribution of nephrin and podocin in podocytes. These observations not only establish a critical function of the polarity proteins in the maintenance of slit diaphragms, but also imply their potential involvement in renal failure in FSGS.  相似文献

1. Mechanoreceptors monitoring tension in working muscles are described in the Decapoda Crustacea.

2. The receptors are associated with apodemes of muscles in the walking leg and are well‐developed in the extensor and flexor of the meropodite (Figures 1, 2).

The effect of the Razdol’naya River on the littoral and sublittoral nematode community was studied in the innermost part of the Amursky Bay. Thanks to the seasonal abundance of juveniles of macrobenthic animals, pseudomacrobenthic species prevailed in the meiobenthos littoral community. Juvenile amphipods dominated (39.5%), the percentage of nematodes was 18.4% (45 species). The nematodes dominated in the meiobenthos community (65.7%, 71 species) at the sublittoral zone. The similarity of the species structure of littoral and sublittoral nematodes reached 36.5%. Low population density (2.6±1.7 thousand ind/m2) and poor species diversity of nematodes (7 species), the dominating form being Oncholaimium paraolium, were recorded at the littoral zone, which was subjected to direct riverine influence (1‰salinity). The correlation analysis revealed no relationship between salinity and population density of nematodes at the littoral zone. At the sublittoral zone (at stations of salinity less than 2.85‰) the parameters of population density (73.9±32.3 thousand ind/m2) and species diversity (12 species) of nematodes were the lowest, Parodontophora timmica dominated. A poor expressed dependence between the density of nematode populations and salinity was revealed. The correlation between the average size of ground particles and population density of nematodes was recorded. The greatest similarity of the species structure of littoral and sublittoral nematodes (26.67%) was recorded on gravel-sand bottoms.  相似文献

Analysis of bacterial diversity in soils along the banks of the Solimões and Negro rivers, state of Amazonas, Brazil, was by partial sequencing of the genes codifying the rDNA16S region. Diversity of operational taxonomic units (OTU) and of the divergent sequences obtained were applied in comparative analysis of microbiological diversity in the two environments, based on richness estimators and OTU diversity indices. The higher OTU diversity in the Solimões was based on the higher number of parameters that evoke this. The interaction between the nucleotide sequences of bacteria inhabiting the two riverine environments indicated that the two microrganism communities are similar in composition.  相似文献

A rapid stepwise measurement for the activities of calpastatin and μ- and m-calpains was developed by using 2-stage elution at pH 8.5 and then 7.0. The activities of calpastatin, μ-calpain and m-calpain can be rapidly assayed following the separation on DEAE-Sephacel chromatography by a 2 stage elution with 90 mM NaCl (pH 8.5), and then by 200 and 300 mM NaCl in elution buffer (pH 7.0). No significant differences in the recovery of these proteinases and inhibitor was observed between stepwise gradient and linear gradient methods.  相似文献

In the Loess Plateau, China, arable cultivation of slope lands is common and associated with serious soil erosion. Planting trees or grass may control erosion, but planted species may consume more soil water and can threaten long‐term ecosystem sustainability. Natural vegetation succession is an alternative ecological solution to restore degraded land, but there is a time cost, given that the establishment of natural vegetation, adequate to prevent soil erosion, is a longer process than planting. The aims of this study were to identify the environmental factors controlling the type of vegetation established on abandoned cropland and to identify candidate species that might be sown soon after abandonment to accelerate vegetation succession and establishment of natural vegetation to prevent soil erosion. A field survey of thirty‐three 2 × 2–m plots was carried out in July 2003, recording age since abandonment, vegetation cover, and frequency of species together with major environmental and soil variables. Data were analyzed using correspondence analysis, classification tree analysis, and species response curves. Four vegetation types were identified and the data analysis confirmed the importance of time since abandonment, total P, and soil water in controlling the type of vegetation established. Among the dominant species in the three late‐successional vegetation types, the most appropriate candidates for accelerating and directing vegetation succession were King Ranch bluestem (Bothriochloa ischaemum) and Lespedeza davurica (Leguminosae). These species possess combinations of the following characteristics: tolerance of low water and nutrient availability, fibrous root system and strong lateral vegetative spread, and a persistent seed bank.  相似文献

This study examined the influence of the following growth factors and cytokines on early embryonic development: insulin-like growth factors I and II (IGF-I, IGF-II), basic fibroblast growth factor (bFGF), transforming growth factor (TGF-β), granulocyte-macrophage colony-stimulating factor (GM-CSF), and leukemia inhibitory factor (LIF). Synthetic oviduct fluid (SOF) was used as the culture medium. We studied the development of bovine embryos produced in vitro and cultured until Day 9 after fertilization. TGF-β1, bFGF, GM-CSF, and LIF used on their own significantly improved the yield of hatched blastocysts. IGF-I, bFGF, TGF-β1, GM-CSF, and LIF significantly accelerated embryonic development, especially the change from the expanded blastocyst to hatched blastocyst stages. Use of a combination of these growth factors and cytokines (GF-CYK) in SOF medium produced higher percentages of blastocysts and hatched blastocysts than did use of SOF alone (45% and 22% vs. 24% and 12%; P < 0.05) on Day 8 after in vitro fertilization and similar results to use of SOF + 10% fetal calf serum (38% and 16%, at the same stages, respectively). The averages of total cells, inner cell mass cells, and trophectoderm cells of exclusively in vitro Day-8 blastocysts for pooled GF-CYK treatments were higher than those for SOF and similar to those for fetal calf serum. The presence of these growth factors and cytokines in the embryo culture medium therefore has a combined stimulatory action on embryonic development; in particular through an increase in hatching rate and in the number of cells of both the inner cell mass and trophoblast. These results are the first to demonstrate that use of a combination of recombinant growth factors and cytokine, as IGF-I, IGF-II, bFGF, TGF-β1, LIF, and GM-CSF, produces similar results to 10% fetal calf serum for the development of in vitro-produced bovine embryos. This entirely synthetic method of embryo culture has undeniable advantages for the biosecurity of embryo transfer.  相似文献

An analysis of Cladocera has been made on sediments from the three Polish lakes Suszek, B\cedowo, and Skrzetuszewskie. The analysed sediments are gyttja from the Subboreal and Subatlantic periods, during which the lakes were subject variously to eutrophication. The eutrophication process has been analysed from the frequency of Cladocera, particularly the index species, and has been correlated with climatic and settlement changes. It was found that in Suszek lake, in which no significant human activity had taken place (palynologically documented), the eutrophication must have been slow and gradual. On the contrary, in the two remaining lakes, in which the human influence was more intense, the eutrophication process underwent intensive changes and coincided somewhat with settlement fluctuations.  相似文献

Data already examined by regression analysis were subjected to factor analysis to scrutinize the effects of environmental factors on microbial populations in the brackish waters of the Tv?rminne archipelago on the southern coast of Finland. Water samples were collected from 1.0-m depth at one point in Tv?rminne Storfj?rd, 71 times over about 2 years. Twenty-six parameters were determined on each sample, 10 of environmental and 16 of microbiological type. The correlations between the parameters were factorized using the principal axis solution, and eight factors chosen for further consideration were rotated by the varimax method. The major part of the variance (about 90% of the total communality) of the microbiological parameters was covered by five factors, interpreted as phytoplankton blooms, the periods before and after the blooms, freshwater outflows, and water temperature. Wind variables were components in the factors interpreted as freshwater outflows. Rainfall played a minor part in the total variance of the microbial community, but it washed yeasts and proteolytic bacteria from the land into the study area. The eight factors selected covered about 60 to 98% of the variance of the microbiological parameters. The highest values (above 90%) were obtained for direct counts of bacteria, for plate counts of mesophilic and polymyxin-resistant bacteria, and for the two community respiration parameters; the lowest values (60 to 75%) were obtained for H(2)S-producing and proteolytic bacteria.  相似文献

Reports have demonstrated occurrences of abnormalities in the early stages of chicken embryonic development due to the exposure to electromagnetic fields (EMFs). This article was designed to investigate the effects of sinusoidal EMF on the histopathology, apoptosis, and expressions of c-Fos and β-Catenin genes of the livers of preincubated White Leghorn chicken embryos, based on our published experiments. 300 healthy, fresh fertilized eggs were divided into control (n?=?70), sham (n?=?70), and four experimental (1-4,days 13, 14, 5, and 19, n?=?40) groups. Experimental eggs were exposed to the most effective intensity in a coil with 7.32?mT density, and sham groups were also located in the same coil with no exposure, both for 24?h before incubation. Control, sham, and experimental groups were then incubated in an incubator (37°C, humidity 60%) for 13,14,15, and 19 days. Livers of 13-15 and 19 day-old chicken embryos were removed by C-section and fixed in formalin (10%), stained with Hematoxylin-Eosin and TUNEL for histopathological and apoptosis studies. Others were used for investigating c-Fos and β-Catenin expressions, using RT-PCR. Results showed extensive hemorrhages all over the chicken embryos' bodies and livers, more lymphoid tissues, disturbed parenchymal tissues, sinusoid denaturation, vesiculizad cytoplasm, an increase in the number of apoptotic cells, and a decrease on the levels of expressions of c-Fos and β-Catenin genes in experimental groups of 1-4, comparing control and sham groups.  相似文献

The lytic effect of the expression of the cloned geneE of bacteriophage X174 inEscherichia coli is considerably amplified by a mutation in thefadR gene, which primarily affects the regulation of fatty acid degradation. In contrast, reduction of the fluidity of the cell membranes by use of thefabB andfadE mutations, which interfere with the synthesis and the oxidation of unsaturated fatty acids, severely inhibits the action of the X174 lysis gene product. A chain-forming mutant carrying a pleiotropic mutation in theenvC locus is also refractory to the X174 lysis protein. As shown by reversion and complementation of theenvC mutatation, a defect in at least one additional gene (rle) is involved in the generation of this refractoriness.  相似文献

Summary Molecular mechanisms of thermoinactivation of the thermostable -amylase of Bacillus caldovelox were examined. Monomolecular conformational processes were found to be the major causes of thermoinactivation at both pH 4.5 and 8.0. The enzyme possessed considerable additional thermostability at pH 8.0, with half-lives of 0.75 and 7.0 min at 90° C and pH 4.5 and 8.0, respectively. The amino acid composition was examined with respect to the underlying thermostability exhibited by this enzyme. The inherent thermostability exhibited may be due to the high proline content (4.47 mol%), but more likely due to the high content of residues forming hydrophobic bonds (60.89 mol%) allied to a low content of residues responsible for ionic interactions (28.34 mol%). A study of the environmental radioactivity in the Kopa?ki Rit Nature Park, Croatia, is presented. This wildlife reserve is part of the Middle Danube River Basin, and it is exposed to various pollutants due to a number of human activities in the surroundings, where there is a nuclear power plant and also urban centres and areas of intense agricultural production. Results for the activity concentrations of soil and surface water samples do not indicate any elevated radioactivity level, which is confirmed by on-site measurements of ambient dose rate equivalent. An assessment of the radiological load on the local biota, carried out using the ERICA tool, implies an overall low radiological risk even if conservative values of the risk quotient are used. Therefore, human activities do not have a significant effect on the radiological load on the Kopa?ki Rit area. A similar conclusion might be made with regard to numerous similar environments in the Middle Danube River Basin.  相似文献
