Cobalt(II) complexes with the non-steroidal anti-inflammatory drug naproxen in the presence or absence of nitrogen-donor heterocyclic ligands (pyridine, 2,2′-bipyridine or 1,10-phenanthroline) have been synthesized and characterized with physicochemical and spectroscopic techniques. The deprotonated naproxen acts as monodentate ligand coordinated to Co(II) ion through a carboxylato oxygen. The crystal structure of [bis(aqua)bis(naproxenato)bis(pyridine)cobalt(II)], 2 has been determined by X-ray crystallography. The EPR spectrum of complex 2 in frozen solution reveals that it retains its structure. UV study of the interaction of the complexes with calf-thymus DNA (CT DNA) has shown that the complexes can bind to CT DNA and [(2,2′-bipyridine)bis(methanol)bis(naproxenato)cobalt(II)] exhibits the highest binding constant to CT DNA. The cyclic voltammograms of the complexes recorded in DMSO solution and in the presence of CT DNA in 1/2 DMSO/buffer (containing 150 mM NaCl and 15 mM trisodium citrate at pH 7.0) solution have shown that they can bind to CT DNA by the intercalative binding mode which has also been verified by DNA solution viscosity measurements. Competitive study with ethidium bromide (EB) has shown that the complexes can displace the DNA-bound EB indicating that they bind to DNA in strong competition with EB. Naproxen and its cobalt(II) complexes exhibit good binding propensity to human or bovine serum albumin proteins having relatively high binding constant values. The antioxidant activity of the compounds has been evaluated indicating their high scavenging activity against hydroxyl free radicals and superoxide radicals.  相似文献

The biological activity of the complexes has been evaluated by examining their ability to bind to calf-thymus DNA (CT DNA) with UV and fluorescence spectroscopies. UV studies of the interaction of the complexes with DNA have shown that they can bind to CT DNA and the DNA-binding constants have been calculated. Competitive studies with ethidium bromide (EB) have shown that complex 3 exhibits the ability to displace the DNA-bound EB indicating that it binds to DNA in strong competition with EB.  相似文献

The quinolones and their Ni(II) complexes have been tested for their antioxidant and free radical scavenging activity. The deprotonated diflunisal ligands are coordinated to Cu(II) ion through carboxylato oxygen atoms. The crystal structures of [tetrakis(diflunisal)bis(N,N-dimethylformamide)dicopper(II)] 1 and [bis(diflunisal)bis(pyridine)copper(II)], 2 have been determined by X-ray crystallography and are the first reported crystal structures of diflunisal complexes. UV study of the interaction of the complexes with calf-thymus DNA (CT DNA) suggests binding of the complexes to CT DNA with the dinuclear [tetrakis(diflunisal)bis(N,N-dimethylformamide)dicopper(II)] compound exhibiting the highest binding constant, Kb. Intercalative binding mode may also be concluded using cyclic voltammetry and solution viscosity measurements of the complexes in the presence of CT DNA. Competitive studies with ethidium bromide (EB) indicate that the complexes can displace the DNA-bound EB suggesting competition with EB. Diflunisal and its complexes exhibit good binding propensity to human or bovine serum albumin protein showing relatively high binding constant values.  相似文献

The biological properties of the previously reported complexes [Cu2(naproxenato)4(H2O)2], [Cu2(diclofenac)4(H2O)2] and [Cu(naproxenato)2(pyridine)2(H2O)] have been also evaluated. The dinuclear complexes exhibit similar affinity for CT DNA as the 2,2′-bipyridine or 1,10-phenanthroline containing complexes. The pyridine containing complexes exhibit the lowest affinity for CT DNA and the lowest ability to displace EB from its EB-DNA complex.  相似文献

The [RhCl3(N-N)(DMSO)] complexes, the N-N being 2,2′-bipyridine (1), 1,10-phenanthroline (2), 4,7-diphenyl-1,10-phenanthroline (3), 4,4′-dimethyl-2,2′-bipyridine (4) and 1,10-phenanthroline-5,6-dione (5), have been synthesized and characterized with spectroscopic methods. The compounds 2-5 adopt mer- and complex 1fac-structure. The molecular and electronic structure studies of mer- and fac-complexes with bpy and phen ligands at the DFT B3LYP level with 3-21G∗∗ basis set showed that mer-isomers are more stable. The cytostatic activity of the [RhCl3(N-N)(DMSO)] complexes against Caco-2 and A549 tumor cells have been studied. Their antibacterial activity have also been investigated. It has been found that the very promising biological activity show complexes 2, 3 and 4.  相似文献

A series of mixed ligand Ru(II) complexes of 5,6-dimethyl-1,10-phenanthroline (5,6-dmp) as primary ligand and 1,10-phenanthroline (phen), 2,2′-bipyridine (bpy), pyridine (py) and NH3 as co-ligands have been prepared and characterized by X-ray crystallography, elemental analysis and 1H NMR and electronic absorption spectroscopy. The X-ray crystal structure of the complex [Ru(phen)2(bpy)]Cl2 reveals a distorted octahedral coordination geometry for the RuN6 coordination sphere. The DNA binding constants obtained from the absorption spectral titrations decrease in the order, tris(5,6-dmp)Ru(II) > bis(5,6-dmp)Ru(II) > mono(5,6-dmp)Ru(II), which is consistent with the trend in apparent emission enhancement of the complexes on binding to DNA. These observations reveal that the DNA binding affinity of the complexes depend upon the number of 5,6-dmp ligands and hence the hydrophobic interaction of 5,6-dimethyl groups on the DNA surface, which is critical in determining the DNA binding affinity and the solvent accessibility of the exciplex. Among the bis(5,6-dmp)Ru(II) complexes, those with monodentate py (4) or NH3 (5) co-ligands show DNA binding affinities slightly higher than the bpy and phen analogues. This reveals that they interact with DNA through the co-ligands while both the 5,6-dmp ligands interact with the exterior of the DNA surface. All these observations are supported by thermal denaturation and viscosity measurements. Two DNA binding modes - surface/electrostatic and strong hydrophobic/partial intercalative DNA interaction - are suggested for the mixed ligand complexes on the basis of time-resolved emission measurements. Interestingly, the 5,6-dmp ligands promote aggregation of the complexes on the DNA helix as a helical nanotemplate, as evidenced by induced CD signals in the UV region. The ionic strength variation experiments and competitive DNA binding studies on bis(5,6-dmp)Ru(II) complexes reveal that EthBr and the partially intercalated and kinetically inert [Ru(phen)2(dppz)]2+ (dppz = dipyrido[3,2-a:2′,3′-c]phenazine) complexes revert the CD signals induced by exciton coupling of the DNA-bound complexes with the free complexes in solution.  相似文献

New mixed polypyridyl {NMIP = 2′-(2″-nitro-3″,4″-methylenedioxyphenyl)imidazo-[4′,5′-f][1,10]-phenanthroline, dmb = 4,4′-dimethyl-2,2′-bipyridine, bpy = 2,2′-bipyridine} ruthenium(II) complexes [Ru(dmb)2(NMIP)]2+ (1) and [Ru(bpy)2(NMIP)]2+ (2) have been synthesized and characterized. The binding of these complexes to calf thymus DNA (CT-DNA) has been investigated with spectroscopic methods, viscosity and electrophoresis measurements. The experimental results indicate that both complexes could bind to DNA via partial intercalation from the minor/major groove. In addition, both complexes have been found to promote the single-stranded cleavage of plasmid pBR 322 DNA upon irradiation. Under comparable experimental conditions compared with [Ru(phen)2(NMIP)]2+, during the course of the dialysis at intervals of time, the CD signals of both complexes started from none, increased to the maximum magnitude, then no longer changed, and the activity of effective DNA cleavage dependence upon concentration degree lies in the following order: [Ru(phen)2NMIP]2+ > complex 2 > complex 1.  相似文献

Two novel Ru(II) complexes [Ru(bpy)2(MCMIP)]2+ (1) and [Ru(phen)2(MCMIP)]2+ (2) (bpy = 2,2′-bipyridine; phen = 1,10-phenanthroline; MCMIP = 2-(6-methyl-3-chromonyl)imidazo[4,5-f][1,10]-phenanthroline) have been synthesized and characterized by elemental analysis, mass spectra and 1H NMR. The DNA-binding properties of the complexes were investigated by absorption, emission, melting temperature and viscosity measurements. Experimental results indicate that the two complexes can intercalate into DNA base pairs. Upon irradiation at 365 nm, two Ru(II) complexes were found to promote the cleavage of plasmid pBR 322 DNA from supercoiled form I to nicked form II, and the mechanisms for DNA cleavage by the complexes were also investigated.  相似文献

The dinuclear complexes [Pd2(L)2(bipy)2] (1), [Pd2(L)2(phen)2] (2), [Pt2(L)2(bipy)2] (3) and [Pt2(L)2(phen)2] (4), where bipy = 2,2′-bipyridine, phen = 1,10-phenanthroline and L = 2,2′-azanediyldibenzoic dianion) dibridged by H2L ligands have been synthesized and characterized. The binding of the complexes with fish sperm DNA (FS-DNA) were investigated by fluorescence spectroscopy. The results indicate that the four complexes bound to DNA with different binding affinity, in the order complex 4 > complex 3 > complex 2 > complex 1, and the complex 3 binds to DNA in both coordination and intercalative mode. Gel electrophoresis assay demonstrates the ability of the complexes to cleave the pBR 322 plasmid DNA. The cytotoxic activity of the complexes was tested against four different cancer cell lines. The four complexes exhibited cytotoxic specificity and significant cancer cell inhibitory rate.  相似文献

Four related ruthenium(III) complexes, with the formula mer-[RuCl3(dmso)(N−N)] (dmso = dimethyl sulfoxide; N−N = 2,2′-bipyridine (1), 1,10-phenantroline (2), dipyrido[3,2-f:2′,3′-h]quinoxaline (3) and dipyrido[3,2-a:2′,3′-c]phenazine (4)), have been reported. Complexes 3 and 4 are newly synthesized and characterized by X-ray diffraction. The hydrolysis process of 1-4 has been studied by UV-vis measurement, and it has been found that the extension of the N−N ligands can increase the stability of the complexes. The binding of these complexes with DNA has been investigated by plasmid cleavage assay, competitive binding with ethidium bromide (EB), DNA melting experiments and viscosity measurements. The DNA binding affinity is increased with the extension of the planar area of the N−N ligands, and complex 4 shows an intercalative mode of interaction with DNA. The in vitro anticancer activities of these compounds are moderate on the five human cancer cell lines screened.  相似文献

A novel polypyridyl ligand CNPFIP (CNPFIP = 2-(5(4-chloro-2-nitrophenyl)furan-2-yl)-1H-imidazo[4,5f][1,10]phenanthroline) and its mononuclear Ru(II) polypyridyl complexes of [Ru(phen)2CNPFIP]2+(1) (phen = 1,10-phenanthroline), [Ru(bpy)2CNPFIP]2+(2) (bpy = 2,2′-bipyridine), and [Ru(dmb)2CNPFIP]2+(3) (dmb = 4,4′-dimethyl-2,2′-bipyridine) have been synthesized successfully and characterized thoroughly by elemental analysis, UV/Vis, IR, NMR, and ESI-MS. The interaction of the Ru(II) complexes with calf thymus DNA (CT-DNA) was investigated by absorption titration, fluorescence, viscosity measurements. The experimental results suggest that three complexes bind to CT-DNA through an intercalative mode and the DNA-binding affinity of complex 1 is greater than that of complexes 2 and 3. The photocleavage of plasmid pBR322 DNA by ruthenium complexes 1, 2, and 3 was investigated. We have also tested three complexes for their antimicrobial activity against Escherichia coli (Gram-negative) and Staphylococcus aureus (Gram-positive) bacteria. The in vitro cytotoxicity of these complexes was evaluated by MTT assay, and complex 1 shows higher cytotoxicity than 2 and 3 on HeLa cells. The induced apoptosis and cell cycle arrest of HeLa cells were investigated by flow cytometry for 24 h. The molecular docking of ruthenium complexes 1, 2, and 3 with the active site pocket residues of human DNA TOP1 was performed using LibDock.  相似文献

Two bis-heteroleptic Ru(II) complexes [Ru(bpy)2(pcip)]2+ (1, bpy = 2,2′-bipyridine, pcip = 2-[4-phenylcarboxy]-1H-imidazol[4,5-f][1,10]phenanthroline) and [Ru(phen)2(pcip)]2+ (2, phen = 1,10-phenanthroline), bearing highly conjugated diimine ligands, were prepared and isolated as their PF6 salts. The bpy-derivative 1 showed better photophysical properties (emission quantum yield, lifetime of the emitting state, and the radiative decay rate constant) than the phen-compound 2. These results followed by theoretical calculations at DFT level established a comprehensive understanding between the structural parameters and the photophysical properties, as well as of the influence of π conjugation and the symmetry of the molecules on spectroscopic characteristics. These results provide fundamental photophysical data for selecting ancillary ligands in the design and improvement of Ru-based light-harvesting complexes.  相似文献

Four new Cu(II) complexes [Cu(pzda)(2,2′-bpy)(H2O)] · 2.5H2O (1), [Cu(pzda)(phen)(H2O)] · H2O (2), [Cu(pzda)(4,4′-bpy)] · H2O (3) and [Cu(pzda)(bpe)0.5(H2O)] (4) were synthesized by hydrothermal reactions of copper salt (acetate or sulphate) with pyrazine-2,6-dicarboxylic acid (H2pzda), and 2,2′-bipyridine (2,2′-bpy), 1,10-phenanthroline (phen), 4,4′-bipyridine (4,4′-bpy) or 1,2-bis(4-pyridyl)-ethane (bpe), respectively. For 1 and 2, they are both monomeric entities which are further assembled into 3D supramolecular networks by hydrogen bonds and π-π stacking interactions. Complex 3 has a 2D metal-organic framework which is connected into 3D supramolecular network by hydrogen bonds. However, for 4, the bpe ligand bridges two Cu(II) ions into binuclear unit, and then the binuclear molecules are assembled into 3D supramolecular network by hydrogen bonds between the coordination water molecule and the carboxylate oxygen atoms. The thermal decomposition mechanism of complexes 1 and 2 cooperated with powder XRD at different temperatures is discussed. The results reveal that once liberation of water molecules takes place the supramolecular network of 1 and 2 collapses.  相似文献

Six antimony adducts with N-donor neutral ligands (1,10-phenanthroline, 4,4′-bipyridine) have been obtained following the reaction of antimony halides with phenanthroline and 4,4′-bipyridine. By changing the solvent and stoichiometry, we obtained six different complexes, Sb(phen)Cl3 (1), Sb(phen)Br3 (2), Sb2(phen)4Br8 (3) and Sb(bpy)Cl3 (4), Sb(bpy)2Cl3 (5), Sb(bpyH · bpyH2)Br6 (6) (where phen = 1,10-phenanthroline, bpy = 4,4′-bipyridine). All the complexes have been characterized via elemental analysis, FT-IR and NMR (1H, 13C) spectroscopy. The crystal structures of complexes 2, 3 and 6 have been determined by X-ray single crystal diffraction.The structural analysis show that the coordination sphere around antimony atom in complex 2 is a distorted square pyramid, coordinated by three bromine atoms and two nitrogen atoms from phen. In complex 3, the central antimony atom is six-coordinated through four bromine atoms and two nitrogen atoms forming a distorted octahedral geometry. Besides that, there are also uncoordinated 1,10-phenanthroline bonded by hydrogen bonds and π-π stacking interactions, which is rarely observed in previous reports. The crystal structure of complex 6 consists of bpyH · bpyH2 trications and hexabromoantimonate trianions. The antimony atom in the anion has a distorted octahedral environment. Additionally, all complexes present a 3D framework built up by N-H?Br, C-H?Br and C-H?Cl weak hydrogen bonds interactions.  相似文献

A series of coordination polymers have been prepared by the combination of flexible ligand 1,1′-biphenyl-2,2′-dicarboxylic acid (H2dpa) and different types of nitrogen-containing ligands, with various metal ions such as Co(II), Zn(II) and Cd(II). The single-crystal structure analyses reveal that the above complexes possess different structure features with the introduction of different nitrogen-containing ligands. When auxiliary linear ligand 4,4′-bipyridine (4,4′-bpy) is introduced, two-dimensional layered complex, [Co2(dpa)2(4,4′-bpy)2(H2O)]n (1) is formed. Whereas if chelating ligand, 1,10-phenanthroline (1,10′-phen) and 2,2′-bipyridine (2,2′-bpy) are introduced, one-dimensional complex [Zn(dpa)(1,10′-phen)]n (2) and discrete complexes [Co2(dpa)2(2,2′-bpy)2(H2O)2] (3), [Co3(dpa)3(1,10′-phen)6(H2O)2] (4), [Cd(dpa)(1,10′-phen)2][(H2dpa)2(H2O)2] (5) are synthesized. To our interest, 1 and 2 crystallize in homochiral spacegroup. Furthermore, the magnetic property of complex 1 and the fluorescent properties of complexes 2 and 5 are studied.  相似文献
