Leaves of pea and broad bean plants were incubated with acetate-[14C] at temperatures varying from 7 to 34°. No significant difference was observed in the distribution of radioactivity between phosphatidylcholine and the galactosylglycerides in pea with different temperatures. However, increasing temperatures increased the labelling of phosphatidylcholine in broad bean leaves, at the expense of polar lipids other than the galactosylglycerides. The incubation temperature had no significant effect on the pattern of labelling of the fatty acids of the major leaf lipids. A correlation was seen in the specific radioactivity of oleate and linoleate in phosphatidylcholine and, especially, in the galactosylglycerides. The data emphasise the rapid equilibration of oleate and linoleate (which probably occurs by transacylation) between the two galactosylglycerides and phosphatidylcholine in leaf tissues.  相似文献

Neutral lipids of ambrosial fungi, nutritionally important to development of Xyleborus ferrugineus beetle, were analyzed for fatty acid composition. Predominant components were 16:0, 18:0, 18:1 and 18:2 in all three mutualistic fungi, Fusarium solani, Cephalosporium sp. and Graphium sp. F. solani had nearly twice the total content of the other two species; age of mycelia did not significantly affect the qualitative composition and total yield.  相似文献

[1-14C]-Acetate incorporation into total and polar lipids was studied in the growing pollen tubes of Crotalaria juncea. Ungerminated pollen had phosphatidyl inositol, phosphatidyl serine, phosphatidyl choline, phosphatidyl ethanolamine, phosphatidyl glycerol, monogalactosyl diglyceride, digalactosyl diglyceride, sulpholipid and steryl glycosides. In the growing pollen tubes considerable [1-14C]-acetate incorporation was observed into the individual polar lipids. The exogenous carbon source significantly influenced lipid biosynthesis. Boric acid (20mg/l.) promoted both pollen tube growth and acetate incorporation into phospholipids. In comparison to 5′-adenosine monophosphate, cyclic-3′,5′-adenosine monophosphate (cAMP) promoted tube growth and also enhanced phospho-and glycolipid biosynthesis. The regulation of membrane component biosynthesis by cAMP is suggested.  相似文献

Polar lipids were extracted from the leaves of Passiflora species which varied in their resistance to chilling injury. The fatty acid compositions of the 8 major polar lipid classes from P. caerulea (chill-resistant) were generally similar to those of the corresponding lipids from P. flavicarpa (chill-sensitive). Using ESR spectroscopy, the motion of spin-labelled molecules was measured in phospholipids isolated from a range of Passiflora species. The temperature dependence of the motion of the spin labels showed a change at 1° for lipids of the most chill-resistant species and at 9° for the lipids of the most chill-sensitive species. Lipids from other species showed changes at intermediate temperatures, and the greater the chilling sensitivity of the species, the higher was the temperature of the change. It is concluded that pronounced differences in chilling sensitivity of the Passiflora species are correlated with physical differences in their membrane lipids; however, the degree of unsaturation of the lipids is not a reliable guide to chilling sensitivity.  相似文献

The lipid classes, fatty acid methyl esters and the sterols of oilpalm pollen were analysed. The neutral lipid fraction consisted of triglycerides, esterified and free sterols and trace amounts of hydrocarbons. Monogalactosyl and digalactosyl diglycerides, phosphatidyl choline, phosphatidyl inositol and phosphatidyl ethanolamine represented the polar lipids. The major fatty acids were linoleic, palmitic and linolenic acids together with small to trace amounts of oleic, stearic, arachidic, myristic, lauric, palmitoleic and margaric acids. Unsaturated fatty acids predominated over saturated ones in the ratio of 3:2. The 4-desmethyl sterols were the major phytosterols in the free form but they constituted a lower proportion of the sterols in the esterified state. 28-Isofucosterol was isolated and characterized as the principal sterol.  相似文献

The lipids of the brown alga Fucus serratus were isolated, identified and quantified. The major acyl lipids were the three glycosylglycerides, diacylgalactosylglycerol, diacyldigalactosylglycerol and diacylsulphoquinovosylglycerol. These represent over 70% of the total acyl lipids. The fatty acid compositions of the major lipids were examined and most showed rather distinctive fatty acid contents. For example, diacylgalactosylglycerol was enriched in n-3 polyunsaturated fatty acids while phosphatidylcholine and phosphatidylethanolamine had very high levels of arachidonate. Phosphatidylglycerol contained the unusual trans-Δ3-hexadecenoic acid. The labelling of lipids and fatty acids from [14C]acetate was examined and the distribution of label between individual components as a function of the incubation period and in algae collected at different times of the year is reported. Algae collected in the winter incorporated much more radioactivity into non-esterified fatty acids when compared to algae collected in the summer. All algae could label myristate, palmitate, stearate and oleate at high rates. Longer incubation times allowed the labelling of polyunsaturated fatty acids such as linoleic acid.  相似文献

Extraction of light and dark grown cells of Neurospora crassa with chloroform-methanol gave a free lipid extract in which the relative amounts and compositions of sterols, fatty acid and carotenoid fractions were determined. Further extraction of the cells with methanolic potassium hydroxide solution liberated a bound lipid fraction from the cells. The levels of the bound lipid fraction were much lower than those of the free lipids but analysis showed that the composition was similar to that of the free lipids.  相似文献

Linoleic acid (LA, C18:2) and α-linolenic acid (ALA, C18:3) are polyunsaturated fatty acids (PUFAs) and major storage compounds in plant seed oils. Microsomal ω-6 and ω-3 fatty acid (FA) desaturases catalyze the synthesis of seed oil LA and ALA, respectively. Jatropha curcas L. seed oils contain large proportions of LA, but very little ALA. In this study, two microsomal desaturase genes, named JcFAD2 and JcFAD3, were isolated from J. curcas. Both deduced amino acid sequences possessed eight histidines shown to be essential for desaturases activity, and contained motif in the C-terminal for endoplasmic reticulum localization. Heterologous expression in Saccharomyces cerevisiae and Arabidopsis thaliana confirmed that the isolated JcFAD2 and JcFAD3 proteins could catalyze LA and ALA synthesis, respectively. The results indicate that JcFAD2 and JcFAD3 are functional in controlling PUFA contents of seed oils and could be exploited in the genetic engineering of J. curcas, and potentially other plants.  相似文献

The neutral lipids of white clover leaves and stems have been separated into wax esters, free fatty acids, free fatty alcohols, free sterols, triglycerides and hydrocarbons. The wax esters were mainly of C18 di- and tri-unsaturated fatty acids and C30 fatty alcohol. Linolenic acid was the predominant free fatty acid and triacontanol was the principal free fatty alcohol. Addition to Bacillus acidocaldarius of acids which can act as primers for fatty acid synthesis promote the synthesis of corresponding fatty acids competitively. The effective acids are n?C5 to -?7 (not C4 or C8), iso- and anteiso-C, and ?C, (not C4), and a range of cyclic acids from cyclobutylacetic and cyclopentanecarboxylic to cycloheptylacetic. New non-natural ω-cyclobutyl-, ω-cyclopentyl-, and ω-cycloheptyl-fatty acids are obtainable. The range of acceptable primers and the range of fatty acids produced therefrom indicate, respectively, the substrate specificities of the transacylase which introduces acyl species into fatty acids synthesis and the one which removes them. The specificity of the primer transacylase may be similar to that in some rumen anaerobes.  相似文献

Fatty acid analyses of phosphoglycerides from Chlamys islandica (Muller) and Ctenodiscus crispants (Retzius) show several interesting features. Major phospholipids from the ovaries and muscles of Chlamys have high levels of (n-3) polyunsaturated fatty acids (PUFA). The phosphatidylinositol (PtdIns) from these tissues contains, however, large amounts of arachidonic acid, 20:4(n?6), so that the (n?3)(n?6) ratio of PtdIns is considerably lower than that of the other phospholipids. In contrast, the major phospholipids from the ovaries of Ctenodiscus contain approximately equal amounts of (n?6), mostly 20:4(n?6), and (n?3)PUFA. The 20:4(n?6) is mostly in phosphatidylethanolamine (PtdEtn) and PtdIns and the (n?3)(n?6) ratio is again lowest in PtdIns. The PtdIns in both species is also rich in 18:0 and consequently resembles PtdIns from elasmobranchs, marine teleosts, and terrestrial mammals. The results are discussed in relation to dietary origins of PUFA in benthic marine animals and to metabolic functions of PtdIns.  相似文献

FGF21 is a unique member of the fibroblast growth factors (FGFs) and a novel hormone that regulates glucose, lipid, and energy homeostasis. The beneficial effects of FGF21 reported thus far have mostly been from chronic treatments. In order to better understand the mechanism for FGF21 action, we evaluated the acute effects of FGF21 in vivo and in vitro. Here we report that a single injection of FGF21 acutely reduced plasma free fatty acid levels similar to its acute effects on plasma glucose in ob/ob mice. In vitro, FGF21 inhibited lipolysis in adipocytes during a short treatment and reduced total lipase activity. These results demonstrate the potential importance of adipocyte lipolysis to the observed acute improvements in plasma parameters.  相似文献

人工授粉作为一项关键性技术在生态研究与农业生产领域广泛应用,但是授粉过程中的工具有效性以及花粉保存尚未受到研究者的关注。以植物野慈姑为试验材料,研究了人工授粉方式和花粉保存条件对繁殖的影响。两年的结果表明,相较于直接接触式授粉,借助毛刷工具授粉显著降低了野慈姑的座果概率和种子数。花粉经过半小时野外高温保存后,其授粉的座果概率显著高于对照组,但单果种子数和种子大小保持稳定。综上,在野慈姑的人工授粉中应避免使用毛刷等工具,离体花粉可置于野外环境适当保存。在相关应用中,研究人员应当警惕工具授粉造成的负面影响,注意比较工具与其他授粉方式的等效性,保证实验的科学性。  相似文献

The predominating flavour compounds in the fruit pulp of Durio zibethinus were hydrogen sulfide, ethyl hydrodisulfide and several dialkyl polysulfides, particularly (C2H5)2Sn, where n = 2 or 3. Ethyl acetate, 1,1-diethoxyethane and ethyl 2-methylbutanoate contribute to an additional fruity odour note. Hydrodisulfides are probably the precursors of the dialkyl sulfides. In the pericarp and seed no volatile sulfur compounds could be detected. The fatty acid composition of the lipids in pericarp, pulp and seed depended on the origin and/or harvest season of the fruit. The main components were oleic and palmitic acids or arachidic acid together with appreciable quantities of palmitoleic, stearic, linoleic and linolenic acids.  相似文献

Phytic acid has been detected in the anthers of young flower buds of Petunia hybrida, the amount increasing slowly as the flower develops until anther dehydration, when there was a more rapid increase in phytic acid content. In mature pollen, the phytic acid content was found to be 2.0 % by weight, of which 90 % was water soluble, while free myo-inositol was a relatively low 0.06 % by weight. Breakdown of phytic acid was initiated soon after pollen germination began, and its degradation products, myo-inositol and inorganic phosphate, were rapidly mobilized for phospholipid and pectin biosynthesis. Both are in high demand during pollen tube elongation. Utilization of myo-[2-3H]inositol for phospholipid biosynthesis was about five times that for pectin synthesis during the first few hours of pollen germination. The label in the phospholipid was identified as the myo-inositol moiety of phosphaltidylinositol, while the pectin material contained predominantly labelled arabinose, with smaller amounts of label in galacturonic acid, glucose and xylose. A chase experiment showed that the myo-inositol moiety of phosphatidylinositol was subject to a relatively rapid turnover, while the label in pectin was not. Labelling germinating pollen with [32P]orthophosphate gave label in phosphatidic acid, phosphatidylinositol, phosphatidylethanolamine and phosphatidylcholine of the phospholipids. Phosphatidylinositol contained 30 % of this label initially, a proportion which declined to 10 % over longer periods of germination.  相似文献
