A simplified system, consisting of NADPH, Fe3+-ADP, EDTA, liposomes, NADPH-cytochrome c reductase and Tris - HCl buffer (pH 6.8), has been employed in studies of the generation of singlet oxygen in NADPH-dependent microsomal lipid peroxidation. The light emitted by the system involves 1deltag type molecular oxygen identifiable by its characteristic emission spectrum and its behavior with beta-carotene. The generation of another excited species (a compound in the triplet state) could be demonstrated in this system by changes of light intensity and emission spectra which arise from photosensitizer (9,10-dibromoanthracene sulfonate, eosin, Rose-Bengal)-mediated energy transfers. Chemiluminescence in the visible region was markedly quenched by various radical trappers and by an inhibitor of NADPH-cytochrome c reductase, but not by superoxide dismutase. During the early stage of lipid peroxidation, the intensity of chemiluminescence was proportional to the square of the concentration of lipid peroxide. These characteristics suggest that singlet oxygen and a compound in the triplet state (probably a carbonyl compound) are generated by a self-reaction of lipid peroxy radicals.  相似文献

Single isolates of bacterial endophytes, isolated from the nematode antagonistic plant species African (Tagetes erecta L.) and French (T. patula L.) marigold, were introduced into potatoes (Solanum tuberosum L.). Several bacterial species possessed activity against root-lesion nematodes (Pratylenchus penetrans) in soils around the root zone of potatoes, namely: Microbacterium esteraromaticum, Tsukamurella paurometabolum, isolate TP6, Pseudomonas chlororaphis, Kocuria varians and K. kristinae. Of these, M. esteraromaticum and K. varians depressed the population densities of root-lesion nematodes without incurring any yield penalty (tuber wet weight). No significant differences were found in the total numbers of P. penetrans nematodes, rhabditid nematodes or `other' parasitic nematode species within the root tissues of bacterized potato plants compared to the unbacterized check. Overall, tuber fresh weights and tuber number were equal to or significantly lower (P\le0.05) in bacterized plants than their unbacterized counterpart. We conclude that endoroot bacteria from Tagetes spp. can play a role in nematode suppression through the attenuation of nematode proliferation. We propose that these nematode control properties are capable of transfer to other crops in a rotation as a beneficial `residual' microflora – a form of beneficial microbial allelopathy.  相似文献

This paper reviews the results obtained in studies of the extracellular polysaccharides, lipopolysaccharide-protein complexes, polysaccharide-lipid complexes, lipopolysaccharides, and O-specific polysaccharides from bacteria of the genus Azospirillum. On the basis of present knowledge, the possible roles of the extracellular polysaccharides and polysaccharide-containing complexes of azospirilla in interaction with the roots of plants are discussed. Some pieces of evidence are considered in light of the lectin hypothesis originally proposed for the legume-Rhizobium symbiosis. In the context of these views of Azospirillum-cereal associative pairs, a key process at the early stages of the interaction is the specific reaction of cereal root lectins with the extracellular polysaccharide components, containing N-acetyl-d-glucosamine as part of their structure.  相似文献

It has been studied whether 2,5-diphenylfuran is a specific singlet oxygen trap in aqueous systems. With certain 1O2 generating systems (Rose Bengal photooxygenation and NaOClH2O2 systems) and·OH generating systems (Fenton's reagent and acetaldehyde-xanthine oxidase system), diphenylfuran was chiefly converted in all cases to cis-dibenzoyl-ethylene, but not to trans-dibenzoylethylene. Low but detectable conversion of diphenylfuran to a hydroperoxide, probably a distinct 1O2-derived reaction in aqueous media, was found only in the Rose Bengal photooxygenation system.  相似文献

In contrast to expectations that unsaturated fatty acids contribute to oxidative stress by providing a source of lipid peroxides, we demonstrated the protective effect of double bonds in lipids on oxidative damage to membrane proteins. Photodynamic inactivation of gramicidin channels was decreased in unsaturated lipid compared to saturated lipid bilayers. By estimating photosensitizer (boronated chlorine e6 amide) binding to the membrane with the current relaxation technique, the decrease in gramicidin photoinactivation was attributed to singlet oxygen scavenging by double bonds in lipids rather than to the reduction in photosensitizer binding. Gramicidin protection by unsaturated lipids was also observed upon induction of oxidative stress with tert-butyl hydroperoxide.  相似文献

Phenolic molecules are widely present in the environment and some of them are well known carcinogens. Some phenolic molecules are also genotoxic but the mechanisms involved in this process are not fully understood. We have studied the induction of chromosomal aberrations by phenol, catechol and pyrogallol in V79 cells at different pH values (6.0, 7.4 and 8.0). At the same pH values, the production of hydroxyl radicals was assessed by measuring the degradation of deoxyribose. Apart from phenol, which only induces a non-significant increase in chromosomal aberration in this experimental system, catechol and pyrogallol showed clear clastogenic effect in a pH-dependent way. Experiments carried out at pH 7.4 in the presence of S9 Mix, SOD, catalase and catalase + SOD suggest that the formation of reactive oxygen species is not the main mechanism involved in the genotoxicity of catechol. However, concerning pyrogallol, our results suggest that its genotoxicity is almost exclusively mediated by reactive oxygen species. Taken together, these results suggest that, in spite of the structural similarity between the different molecules studied, the mechanisms of genotoxicity of these molecules could be considerably different. The existence of several mechanisms of genotoxicity, partially shared by this class of compounds, could explain the synergistic effects observed between these compounds in several genotoxicity test systems. Accurate knowledge of their mechanisms of genotoxicity could improve considerably the assessment of their relevance to human health, since these compounds, once absorbed, are subject to a wide range of pH values in vivo.  相似文献

In the presence of NADPH, cytochrome c stimulates approximately a 200-fold increase in the production of singlet oxygen by the bovine adrenodoxin reductase-adrenodoxin system. The formation of singlet oxygen, which was monitored by the attending chemiluminescence, was markedly inhibited by the addition of superoxide dismutase or 1,4-diazabicyclo[2.2.2]octane. The adrenal system, in the presence of cytochrome c, peroxidized adrenal mitochondrial lipids, as indicated by the formation of malondialdehyde. This oxidation is also inhibited by the addition of dismutase and 1,4-diazabicyclo[2.2.2]octane.  相似文献

A new benzofuran derivative (1) was isolated from the roots of Petasites hybridus and its structure was determined as 1-(6-hydroxy-2-isopropenyl-1-benzofuran-5-yl)-1-ethanone by spectroscopic data and X-ray crystallographic analysis. The compound 1 showed moderate inhibitory activity on human breast cancer MCF-7 cells proliferation in vitro with an IC50 value of 48 μmol/L. The interaction of bovine serum albumin (BSA) and DNA with compound 1 were studied in an aqueous solution under physiological conditions by UV–vis spectroscopy.  相似文献

The distribution of ommatidial diameters and interommatidial angles, as determined by measuring the angles between the optic axes of adjacent ommatidia, are mapped across the surface of the compound eyes of a variety of species selected for different adult behaviors, developmental histories, and taxonomic positions. The size of the visual fields, prey capture foveas, foveas composed of large dorsal ommatidia, and other specializations in the numbers of ommatidia that view various directions in the visual field are discussed in relation to adult behavior. Advanced species have less resemblance between their larval and adult eyes than primitive species. In contrast to their larvae, adults increase the monocular resolution of each eye at the expense of binocular vision. Most species have foveas which view in approximately the anterior direction, instead of in a region of binocular overlap, and many species have foveal bands which view along the horizon. Some advanced perching species, which approach their prey and other odonates from below, have an additional vertical foveal band that views along a vertical plane from the anterior direction to a more dorsal direction. The most unusual foveal band is seen in active flying species. The large dorsal ommatidia of the migratory Anax junius, which cover approximately one third of the eye surface, view a narrow region of the visual field that extends along a plane from the most lateral direction of one eye to a dorsal direction, and continues without interruption to the most lateral direction of the other eye.  相似文献

Butylated hydroxyanisole (BHA) was found to induce chromosome aberrations in Chinese hamster ovary (CHO) cells in the presence of Aroclor-induced rat-liver S9. The effects were more marked when washed microsomes were employed and chromosome damage was considerably reduced in the presence of catalase, suggesting that hydrogen peroxide was involved. Stimulation of H2O2 production by BHA in S9 or microsome incubation mixtures was demonstrated using the catalase-mediated production of formaldehyde from methanol. One of the major microsomal metabolites of BHA, tert.-butyl hydroquinone (t-BHQ), which autoxidises in solution producing H2O2 also induced extensive catalase-sensitive chromosome damage in the absence of metabolic activation. These observations suggest that extracellular generation of reactive oxygen species may be implicated in the mechanism of BHA clastogenicity in vitro. However, chromosome damage was not completely abolished by catalase and the end product of t-BHQ oxidation, tert.-butyl quinone, was also weakly clastogenic, suggesting that intracellular effects of quinone metabolites may also be involved in the clastogenicity of BHA.  相似文献
