Colonization of the roots of beans, alfalfa, onions, red clover, corn, and four barley cultivars (Bonanza, Klondike, Gateway 63, and Olli) by Glomus dimorphicum Boyetchko and Tewari, a vesicular-arbuscular mycorrhizal fungus isolated from a barley field in Alberta, Canada, was studied under greenhouse conditions. Infection levels were low in all four barley cultivars but were higher in beans, alfalfa, and onions and were highest in red clover and corn roots. The infection patterns of G. dimorphicum varied among all the hosts. Coiling of intracellular hyphae occurred in corn, alfalfa, and red clover roots. Appreciable numbers of intraradical vesicles were found only in red clover and bean roots, while arbuscules formed in all hosts except barley. It was concluded that the pattern of root colonization by G. dimorphicum is influenced by the host genome and that the fungal morphology in the roots is variable and, thus, not diagnostic for the mycorrhizal species.  相似文献

Tobacco (Nicotiana tabacum L.) plants were grown with and without the arbuscular mycorrhizal fungus, Glomus intraradices Schenk & Smith. High-performance liquid chromatographic analyses of methanolic extracts from mycorrhizal and non-mycorrhizal tobacco roots revealed marked fungus-induced changes in the patterns of UV-detectable products. The UV spectra of these products, obtained from an HPLC photodiode array detector, indicated the presence of several blumenol derivatives. The most predominant compound among these derivatives was spectroscopically identified as 13-hydroxyblumenol C 9-O-gentiobioside (“nicoblumin”), i.e. the 9-O-(6′-O-β-glucopyranosyl)-β-glucopyranoside of 13-hydroxy-6-(3-hydroxybutyl)-1,1,5-trimethyl-4-cyclohexen-3-one, a new natural product.  Fungal enzyme activities were quantified in an interaction study between the fungus Glomus intraradices and the pea pathogen Aphanomyces euteiches. Fungal and host enzymes were separated by polyacrylamide gel electrophoresis and the activity of A. euteiches–specific glucose-6-phosphate dehydrogenase (Gd), phosphoglucomutase and peptidase (PEP) enzymes were quantified by densitometry. The activity of A. euteiches–specific enzymes increased until 14 days after inoculation with A. euteiches, and then decreased. The plants preinoculated with G. intraradices showed no symptoms of severe root rot even though the pathogen was present and active in these plants. Thus, plants preinoculated with G. intraradices were more tolerant of infection with A. euteiches than non-mycorrhizal plants. This effect was evident even though the A. euteiches infection levels of mycorrhizal and non-mycorrhizal plants were the same. A. euteiches enzyme activities in the mycorrhizal plants were different to those in non-mycorrhizal plants. The peaks of PEP and Gd enzyme activity of A. euteiches were lower and the development of A. euteiches PEP activity was later in the mycorrhizal plants than in the non-mycorrhizal plants. Growth of the mung bean (Vigna radiata L.) epicotyl was retarded by white (400–700 nm) light, especially by monochromatic red (660 nm) light. Growth promoting effects of brassinolide were observed under those light conditions that retarded growth, but were not evident in the dark or under far-red light. * The influence of carbohydrate availability to mycorrhizal roots on uptake, metabolism and translocation of phosphate (P) by the fungus was examined in axenic cultures of transformed carrot (Daucus carota) roots in symbiosis with Glomus intraradices. * 14C-labelled carbohydrates, 33P-phosphate and energy dispersive X-ray microanalysis were used to follow the uptake and transfer of C and P in the arbuscular mycorrhizal (AM) symbiosis. * The uptake of P by the extraradical mycelium (ERM) and its translocation to the mycorrhizal roots was stimulated and the metabolic and spatial distribution of P within the fungus were altered in response to increased carbohydrate availability. Sucrose supply resulted in a decrease of polyphosphates and an increased incorporation into phospholipids and other growth-related P pools and also caused elevated cytoplasmic P levels in the intraradical mycelium (IRM) within the root and higher cytoplasmic P levels in the root cortex. * These findings indicate that the uptake of P by the fungus and its transfer to the host is also stimulated by the transfer of carbon from plant to fungus across the mycorrhizal interface.  相似文献

The direct impact of fenpropimorph on the sterol biosynthesis pathway of Glomus intraradices when extraradical mycelia alone are in contact with the fungicide was investigated using monoxenic cultures. Bi-compartmental Petri plates allowed culture of mycorrhizal chicory roots in a compartment without fenpropimorph and exposure of extraradical hyphae to the presence of increasing concentrations of fenpropimorph (0, 0.02, 0.2, 2, 20 mg l−1). In the fungal compartment, sporulation, hyphal growth, and fungal biomass were already reduced at the lowest fungicide concentration. A decrease in total sterols, in addition to an increase in the amount of squalene and no accumulation of abnormal sterols, suggests that the sterol pathway is severely slowed down or that squalene epoxidase was inhibited by fenpropimorph in G. intraradices. In the root compartment, neither extraradical and intraradical development of the arbuscular mycorrhizal (AM) fungus nor root growth was affected when they were not in direct contact with the fungicide; only hyphal length was significantly affected at 2 mg l−1 of fenpropimorph. Our results clearly demonstrate a direct impact of fenpropimorph on the AM fungus by a perturbation of its sterol metabolism.  相似文献

Phosphate-solubilizing bacteria (PSB) exhibited a high efficiency to improve plant growth and nutrition in the presence of Bayovar rock phosphate when sand-vermiculive was used as a culture medium. Treatments with dual inoculum (PSB plus mycorrhiza) significantly (P≤0.05) increased alfalfa growth. Bacteria-microbial fungi interactions resulted in a greater utilization of the rock phosphate added to the rooting medium. Although Bayovar rock phosphateper se can be considered an inert substrate because it did not stimulate plant growth, metabolites released by PSB were able to transform the rock into available forms which could be utilized by alfalfa plants.Glomus fasciculatum was the most efficient mycorrhizal endophyte under the experimental conditions employed.  相似文献

The activity of cysteine endopeptidase (EP) in the cotyledons of mung bean seeds increased with time after germination. When cotyledons were excised from the embryonic axis in the course of seedling growth, the activity of EP in the excised cotyledon markedly dropped during the following incubation of 1 d. However, the level of EP protein in excised cotyledons, as examined by immunoblotting, was similar to that in axis-attached cotyledons at the corresponding stage. Thus, it seems that the low activity of EP in excised cotyledons is not due to a decrease in the content of EP protein, but due to a loss of the activity of existing EP. Treatment of attached cotyledons with polyamines (PAs; putrescine and spermidine [Spd]) resulted in a decrease in EP activity, while the same PA-treatment brought about little alteration in the level of EP protein. This indicates that PAs somehow produce an inhibitory effect on the activity of EP. Axis-removal resulted in an accumulation of Spd in the cotyledon. The possibility is suggested that PA, especially Spd, is involved in the inhibition of EP activity in excised mung bean cotyledons.  相似文献

Relations between cytokinin concentrations and effects of P and vesicular-arbuscular mycorrhizal (VAM) infection were investigated in Plantago major L. ssp. pleiosperma Pilger. Both mycorrhizal infection by Glomus fasciculatum (Thaxt. sensu Gerdemann) Gerdemann and Trappe and P addition increased the shoot to root ratio, specific leaf area (SLA), and P concentrations of shoot and roots, and decreased the percentage of dry matter in the shoot during the experiment. In general, P concentration in the shoot and roots of each treatment correlated positively with the shoot to root ratio and specific leaf area, and negatively with the percentage of dry matter in the shoot. Cytokinin concentrations in the tissue of shoots and roots were determined using an enzyme-linked immunosorbent assay. Concentrations of zeatin and zeatin-ribosides in the free base and nucleotide fractions had increased more after P addition than in the case of mycorrhizal infection in both shoot and roots, whereas the P concentrations had increased less. It is suggested that zeatin and zeatin-ribosides are not the primary growth-substances involved in mediating VAM effects.  相似文献

Phytostabilization strategies may be suitable to reduce the dispersion of uranium (U) and the overall environmental risks of U-contaminated soils. The role of Glomus intraradices, an arbuscular mycorrhizal (AM) fungus, in such phytostabilization of U was investigated with a compartmented plant cultivation system facilitating the specific measurement of U uptake by roots, AM roots and extraradical hyphae of AM fungi and the measurement of U partitioning between root and shoot. A soil-filled plastic pot constituted the main root compartment (CA) which contained a plastic vial filled with U-contaminated soil amended with 0, 50 or 200 mg KH2PO4−P kg–1soil (CB). The vial was sealed by coarse or fine nylon mesh, permitting the penetration of both roots and hyphae or of just hyphae. Medicago truncatula plants grown in CA were inoculated with G. intraradices or remained uninoculated. Dry weight of shoots and roots in CA was significantly increased by G. intraradices, but was unaffected by mesh size or by P application in CB. The P amendments decreased root colonization in CB, and increased P content and dry weight of those roots. Glomus intraradices increased root U concentration and content in CA, but decreased shoot U concentrations. Root U concentrations and contents were significantly higher when only hyphae could access U inside CB than when roots could also directly access this U pool. The proportion of plant U content partitioned to shoots was decreased by root exclusion from CB and by mycorrhizas (M) in the order: no M, roots in CB > no M, no roots in CB > M, roots in CB > M, no roots in CB. Such mycorrhiza-induced retention of U in plant roots may contribute to the phytostabilization of U contaminated environments.  相似文献

We studied the effects of temperature, carbon dioxide and abscisic acid on mung bean (Vigna radiata). Plants were grown under 26/22°C or 32/28°C (16?h?light/8?h?dark) at 400 or 700?μmol?mol?1 CO2 and received ABA application of 0 or 100?μl (10?μg) every other day for three weeks, after eight days of initial growth, in growth chambers. We measured 24 parameters. As individual factors, in 16 cases temperature; in 8 cases CO2; in 9 cases ABA; and as interactive factors, in 4 cases, each of temperature?×?CO2, and CO2?×?ABA; and in 2 cases, temperature?×?ABA were significant. Higher temperatures increased growth, aboveground biomass, growth indices, photochemical quenching (qP) and nitrogen balance index (NBI). Elevated CO2 increased growth and aboveground biomass. ABA decreased growth, belowground biomass, qP and flavonoids; increased shoot/root mass ratio, chlorophyll and NBI; and had little role in regulating temperature–CO2 effects.

Two isolates of Paenibacillus validus (DSM ID617 and ID618) stimulated growth of the arbuscular mycorrhizal fungus Glomus intraradices Sy167 up to the formation of fertile spores, which recolonize carrot roots. Thus, the fungus was capable of completing its life cycle in the absence of plant roots, but relied instead on the simultaneous growth of bacteria. The supernatant of a mixed batch culture of the two P. validus isolates contained raffinose and another, unidentified trisaccharide. Among the oligosaccharides tested, raffinose was most effective in stimulating hyphal mass formation on plates but could not promote growth to produce fertile spores. A suppressive subtractive hybridization library followed by reverse Northern analyses indicated that several genes with products involved in signal transduction are differentially expressed in G. intraradices SY 167 when grown in coculture with P. validus (DSM 3037). The present investigation, while likely representing a significant step forward in understanding the arbuscular mycorrhizal fungus symbioses, also confirms that its optimal establishing and functioning might rely on many, as yet unidentified factors.  相似文献

Two experiments were carried out to study physiological effects of vesicular-arbuseular mycorrhizal infection on Plantago major L., ssp. pleiosperma (Pilger). In the first experiment, infection by the Glomus fasciculatum (Thaxt. sensu Gerdemann) Gerdemann and Trappe increased growth, shoot to root ratio, P concentrations in both shoot and roots and total uptake of P per plant. The percentages of dry matter in both shoot and roots were lower in mycorrhizal plants.
In the second experiment different P treatments were applied to both mycorrhizal and non-mycorrhizal P. major plants to separate any effects of mycorrhizal infection from increased uptake of P. In addition to the effects found in the first experiment, mycorrhizal, P, and mycorrhizal x P interaction effects were found on root respiration rate and the concentration of soluble sugars in the roots. No clear effects on total dry weight, N and starch concentrations in shoot and roots and sugar concentraion in the shoot were found. Irrespective of the mycorrhizal treatment, increased P concentration in the shoot correlated with an increased shoot to root ratio and root respiration rate, and a decreased percentage dry matter and sugar concentration in the roots. However, the root respiration rate and the P concentration in the roots of mycorrhizal plants were enhanced more than expected from the increased P concentrations in the shoots of these plants.  相似文献

The level of vesicular-arbuscular mycorrhizal (VAM) infection in the roots of Leucaena grown in a sand-soil mixture in the greenhouse increased rapidly with time and reached a peak value of 84% at 30 days from planting. The pattern of immobile nutrient uptake and accumulation closely paralleled that of the development of infection, particularly during the first 10–30 days after planting. Significant changes in dry matter yield were also observed only after a significant portion of the root length was colonized byGlomus aggregatum. The development of VAM infection was not accompanied by growth depression at any of the sampling periods. However, VAM roots had very high levels of Cu which was not translocated to shoots. It is hypothesized that such a diversion of Cu by the endophyte from the host could cause growth depression under conditions where the soil volume is supplied with sub-optimal levels of Cu.
