Carbohydrate and abscisic acid (ABA) metabolism were investigated in normal and phenotypically small 'Hass' avocado ( Persea americana Mill.) fruit in an attempt to link alterations in sugar and ABA content with changes in 3-hydroxy-3-methylglutaryl coenzyme A reductase (HMGR, EC 1.1.1.34) activity and fruit size. The small-fruit phenotype was characterized by reduced seed HMGR activity, increased seed insoluble acid invertase ( β - d -fructofuranosidase, EC 3.2.1.26), decreased sucrose synthase (SS; UDP- d -glucose: d -fructose-2- α -glucosyl-transferase, EC 2.4.1.13) activity, decreased sucrose content, and increased glucose as a proportion of the total soluble sugar. Sucrose phosphate synthase (SPS; UDP- d -glucose: d -fructose 6-phosphate 2- α - d -glucosyltransferase, EC 2.4.1.14) activity was unaffected in seed but reduced in mesocarp of the small fruit. In addition, the small-fruit variant displayed enhanced respiration and both seed and mesocarp tissue showed increased ABA metabolism. Applied ABA caused an increase in insoluble acid invertase activity in seed tissue of normal fruit while mevastatin reduced HMGR activity in this tissue, caused sucrose depletion and increased the proportion of glucose from 5 to 57% of total soluble sugars. Exogenous glucose suppressed HMGR activity in seed tissue whereas in mesocarp tissue, HMGR activity was reduced to 38% of the control after 6 h but enhanced by 46% by 48 h. Glucose increased ABA biosynthesis and turnover in competent tissues. These results suggest that ABA turnover is mediated, in part, by carbohydrate content and composition which also affects HMGR activity. It is proposed that sugar and ABA signals act in concert to modulate expression and/or activity of HMGR in the control of 'Hass' avocado fruit growth and final fruit size.  相似文献

Renal hypertrophy was induced in mice by injection of testosterone. Associated with an increase in renal tissue mass were increases in the concentrations of spermine and spermidine and in the activity of ornithine decarboxylase (ODC), the rate limiting enzyme in their synthesis. The increased activity of ODC was shown to be due to an increase in tissue ODC content. Total tissue actin was not altered by testosterone treatment although the proportion of unpolymerized (soluble) actin was increased significantly. These data are discussed in relation to the postulated mechanism of polyamine stimulation of actin polymerization.  相似文献

Production of extracellular polysaccharidases by Irpex lacteus Fr. was studied in different culture conditions.

The presence of fatty acids such as linolic, linolenic, erucic, palmitic acids etc. caused remarkably to increase the production of ceîlulase (filter paper disintegrating activity, FD), laminarinase and xylanase. On the contrary, fatty acids had not any special effect on the production of cellulolytic enzymes such as Avicelase and CMCase and of plant tissue macerating enzymes (MA).

When two kinds of carbon sources, e.g., cellulose powder and potato pulp were mixed together and used as an inducer, polysaccharidase production investigated, with the exception of CMCase, increased higher than when the two substances were used separately.  相似文献

This research compares effects of the compound 5-chloro-3-methyl-4-nitro-1 H -pyrazole (CMNP), a plant growth regulator that selectively promotes abscission in mature citrus fruit ( Citrus sinensis ), and the ethylene-releasing agent ethephon (2-chloroethylphosphonic acid). Application of CMNP and ethephon to mature citrus fruit reduced fruit detachment force and changed peel color from green to orange. More total chlorophyll was extracted from flavedo in early season (November) than late season (January), and both compounds caused a similar reduction in chlorophyll. In contrast, total carotenoid content was similar in November and January. Both abscission compounds increased total carotenoids, but induction was greater in January, and CMNP was more effective in both months. Phospholipase A2 (PLA2) activity increased after CMNP but not ethephon application. Electrolyte leakage increased 2 h after CMNP treatment, and total protein content was reduced by 50% after 72 h. Ethephon caused only minor changes in electrolyte leakage and total protein content. Inhibition of PLA2 activity with aristolochic acid did not reduce leakage but inhibited total protein loss and reduced visual peel damage associated with CMNP. Ultrastructural observations indicated decreased number, and length of starch grains 3 h after CMNP treatment. A transient increase in soluble sugars was measured 3 h after CMNP application. Ethephon had little effect on soluble sugar content and changes in starch grains. Collectively, the results indicate that CMNP and ethephon induced color change in peel and advanced mature fruit abscission. However, CMNP but not ethephon promoted other physiological changes associated with senescence.  相似文献

The systemic insecticides namely phorate (Thimet 10 G) oxydemeton methyl (metasystox 25 EC) and dimethoate (Rogor 30 EC) decreased oil content in the developing seeds of Indian mustard (Brassica juncea L.) but showed an increase in the mature seeds. The inhibitory effect in the developing seed was accompanied by an increase in soluble sugars and a corresponding decrease in malate dehydrogenase and G6P dehydrogenase activity. The relative proportion of triacylglycerols and glycolipids decreased significantly while that of phospholipids and free fatty acids increased in the developing seeds. In the mature seeds, the proportion of triacylglycerols did not change appreciably from that in control. The erucic acid synthesis which was less at 10 and 20 days after fertilization (DAF) increased at 30 DAF with oxydemeton methyl and dimethoate; phorate was ineffective. In mature seeds, the proportion of erucic acid increased at the cost of linoleic and linolenic acids. All the insecticides appreciably decreased the rate of [1-14C]acetate incorporation into lipids both in vivo and in vitro experiments. In the in vivo experiment, the synthesis of polar lipids was enhanced at 10 and 20 DAF, and the higher doses of oxydemeton methyl and dimethoate at 30 DAF. On the other hand, the 14C-incorporation into triacylglycerols showed an opposite trend to that of polar lipids. In the in vitro experiment, oxydemeton methyl and dimethoate enhanced the synthesis of polar lipids at 10 and 20 DAF while these inhibited it at 30 DAF. The synthesis of triacylglycerols was inhibited by the use of these insecticides.  相似文献

Two experiments were undertaken to investigate the influence of (1) pig bodyweight and (2) dietary fibre content of the diet on the in vitro gas production of sugar beet pulp fibre using faecal inoculum.In the first experiment, inocula prepared from young pigs (Y; 16–50 kg), growing pigs (G; 62–93 kg) and sows (S; 216–240 kg) were compared. Sugar beet pulp, hydrolysed in vitro with pepsin and then pancreatin, was used as the fermentation substrate. The cumulated gas productions over 144 h were modelled and the kinetics parameters compared. Lag times (Y: 4.6 h; G: 6.4 h; S: 9.2 h) and half-times to asymptote (Y: 14.7 h; G: 15.9 h; S: 20.8 h) increased with pig bodyweight (P<0.001) and the fractional degradation rates of the substrate differed between the pig categories (Y: 0.110 h−1; G: 0.115 h−1; S: 0.100 h−1; P<0.001). The final gas production was not affected (P=0.10) by the inoculum source.In the second experiment hydrolysed sugar beet pulp was fermented with four inocula prepared from pigs fed diets differing in their total and soluble dietary fibre contents, i.e. low fibre diet rich in soluble fibre (LOW-S) or in insoluble fibre (LOW-I) or high fibre diet rich in soluble fibre (HIGH-S) or in insoluble fibre (HIGH-I). The total and the soluble dietary fibres influenced the kinetics of gas production. The presence of soluble fibres decreased the lag times, whatever the total dietary fibre content (2.7 h for LOW-S versus 3.5 h for LOW-I, 4.0 h for HIGH-S versus 4.4 h for HIGH-I; P<0.001). The half-times to asymptote were higher with the low fibre diets (P<0.001) and, for similar total dietary fibre contents, they were lower when the proportion of soluble fibres increased (LOW-S: 9.9 h; LOW-I: 11.4 h; HIGH-S: 8.9 h; HIGH-I: 10.1 h; P<0.001). The fractional degradation rates of the substrate were the highest with the fibre-rich diet containing a high proportion of soluble fibres (0.158 h−1; P<0.001).In conclusion, the bodyweight of the faeces donors and the dietary fibre composition of the pig diet influence the in vitro fermentation kinetics of hydrolysed sugar beet pulp, but not the final gas production.  相似文献

Lipids were extracted from alfalfa samples collected at intervals over the growing season and were fractionated to yield pure sulfolipid. In the sulfolipid and in a phospholipid fraction the major fatty acids were palmitic, linolenic, and linoleic, of which the palmitic acid increased in proportion during the season while the proportion of linolenic acid dropped. The sulfolipid contained more linolenic acid and less palmitic and linoleic acids than the phospholipids, and had a greater rate of change of fatty acid composition.  相似文献

Studies performed on winter rape plants(Brassica nnpus var.oleifera, cv. ‘Gór-czański’) revealed that cold treatment affected the cell membranes and led to the temporary increase in electrolytic leakage from a tissue. This was followed by the marked decrease of the electrolytic leakage in the course of hardening. Changes in membrane properties were accompanied by the promotion of soluble protein accumulation. Inhibition of protein accumulation by the cycloheximide treatment brought about wilting of plants under cold conditions. Possible role of soluble protein in protection of cells against secondary water stress caused by the coldinduced changes in membrane properties is suggested. Cold-induced changes in the electrophoretic pattern of soluble protein are described and discussed.  相似文献

Formation of linolenic acid in vivo from various precursors [1-14C]-2:0, -12:0, -16: 0, -18:0, -18:1, 18:2 in the cold resistant wheat cultivar Miranovskaja 808 and cold sensitive wheat cultivar Penjamo 62 was investigated at three different temperatures (+25, +5, and ?6 °C). Both cultivars converted the offered precursors to linolenic acid only very slowly. Decreasing the experimental temperature brought about an increase formation of linolenic acid, however, Miranovskaja 808 being more successful than Penjamo 62. Comparison of the specific activities of linolenic acid at the “time of equal level of tissue labeling” revealed that Miranovskaja 808 formed 2 to 10 times faster linolenic acid from various precursors upon exposure to cold than Penjamo 62. Considering the low rate of formation of linolenic acid in leaves it appears probable that even the cold resistant cultivars are unable to increase the proportion of linolenic acid in their membranes fast enough to prevent the thermotropic phase transition from liquid crystalline to solid gel state at beginning of the onset of cold. It is suggested that rapid accumulation of hitherto unknown cryoprotective substance (s) of lipidic nature precedes the accumulation of linolenic acid upon exposure of the seedlings to chilling temperatures.  相似文献

Second and third leaves of 25-day old wheat (Triticum aestivum L.) planta, cultivar WL-711, were inoculated with three races of the brown rust pathogen,Puccinia recondita f. sp.tritici. The host reflected a differential interaction response towards these races, viz., susceptible reaction to race 77 (reaction type 4) intermediate reaction to race 104 (reaction type 1 - 3) and resistant reaction to race 63 (reaction type 0). Post-inoculation mutual interaction brought about observable changes in peroxidase activity along with leakage of electrolytes and phenolic constituents indicating altered permeability of host tissue. Differential increase in leakage of electrolytes and phenols occurred with increase of peroxidase activity during 12 h and 24 h post-inoculation, however, during 36 h, 48 h and 72 h post-inoculation, electrolyte leakage increased continuously with corresponding increases in peroxidase activity in all three interactions, while leakage of phenols showed a differential pattern. The differential changes in isoenzymic pattern of peroxidase activity were also noted during the progressive infection process which may also be an indication of altered permeability of the host issue at the very onset of brown rust infection.  相似文献

Linolenic acid contents of glycolipids increased in irradiated potatoes during storage, accompanied by a decrease of linoleic acid. The puncturing of a potato tuber with a needle of a microsyringe caused the similar changes; the elevation of linolenic acid level and decline of linoleic acid were observed within 24 h after puncturing. Irradiation before the puncturing reduced the degree of the increase of linolenic acid in response to the mechanical injury. The rate of [13C]acetate incorporation into lipid fractions of irradiated tubers was smaller than that of unirradiated tubers, and polyunsaturated fatty acids (linoleic and linolenic acids) of lipid fractions were weakly labeled in irradiated tubers as compared with unirradiated ones. The results in this study indicate that irradiation retards lipid metabolism in response to mechanical injury.  相似文献

The effects of hot water treatment and storage temperature (4°C, 13°C or 22°C) on the quality and impedance of outer and inner mesocarp of mango were assessed in two experiments during storage, impedance being a potential non‐destructive measure of tissue damage following heat treatment. Fruits were subjected to equivalent heat units at 36.5°C for 60 min plus 46.5°C for 43 min or 46.5°C for 90 min by hot water treatments (hwt) on the assumption of cumulative heat effects and a base temperature of 12–13°C. Fruit reflectance decreased whereas chroma and hue angle increased over storage time and also with increase in storage temperature. The yellow colour increased with a rise in storage temperature in hot water treated mangoes. Soluble solids content of mangoes held at 22°C was highest at 5 days of storage but decreased subsequently over storage time. Impedance of all fruits decreased with increase in frequency, storage temperature and time in store. The impedance of hwt mangoes was lower than that of non‐hwt fruits 8 h after immersion, but recovered almost to control levels on day 5 at 4°C or 13°C, but decreased gradually after 5 days at 13°C. Impedance of all mangoes stored at 22°C decreased continuously during storage. Impedance was higher in the inner mesocarp than outer pulp. Impedance of hwt fruits was poorly correlated with soluble solid content and chroma but well correlated with reflectance of fruit pulp at 22°C. Changes in impedance of mangoes are discussed in relation to physiological and biochemical changes that occur during heat treatment and storage.  相似文献

以结球甘蓝品种‘冬升’种子为材料,研究高温(40℃)高湿(相对湿度100%)人工老化处理过程中种子的萌发特性、种子浸出液相对电导率和丙二醛、可溶性蛋白、可溶性糖含量以及抗氧化酶活性的变化,以揭示种子劣变的机理。结果表明:(1)人工老化处理甘蓝种子的含水量和不正常苗率均随着老化时间的延长逐渐增加,而种子发芽势、发芽率、发芽指数和活力指数的增加均逐渐降低。(2)随着处理天数的延长,老化处理甘蓝种子的浸出液电导率显著增大,浸出液可溶性糖含量逐渐升高,可溶性蛋白含量表现出显著下降趋势,而种子MDA含量呈先升高后逐渐下降的趋势。(3)在结球甘蓝种子老化进程中,其种子中SOD、POD、CAT活性变化的趋势相似,均随老化程度的加深而逐渐降低;而APX活性在老化处理的最初2d显著增加,第6天显著降低。研究发现,在结球甘蓝种子老化进程中,种子活力和萌发率显著降低,其种子浸出液电导率、可溶性蛋白含量、可溶性糖含量、保护酶活性变化与种子老化及劣变程度密切相关,膜脂过氧化作用可能是引起或加剧种子老化劣变的重要原因之一。  相似文献

The effect of a single intraperitoneal injection of retinoic acid on liver transglutaminase (EC 2.3.2.13) activity and total putrescine, spermidine and spermine was studied. The results demonstrate that: (1) transglutaminase activity is increased over control values as early as 4-6 h after treatment, reaching a maximum (2-fold increase) at 12 h and returning to control values at 36 h; (2) the retinoic acid-induced form of enzyme is the soluble tissue transglutaminase; (3) actinomycin D treatment does not completely inhibit the early (6 h) increase of activity, while suppressing that at 12 h; (4) the immunoassay of the soluble transglutaminase shows that, 6 h after treatment, there is no increase in the protein, whereas at 12 and 24 h a significant increase is observed; (5) putrescine, but not spermidine and spermine, increases (5-7-fold) 6 and 18 h after the retinoic acid treatment. The possibility also that the expression of soluble transglutaminase is modulated in vivo by retinoic acid and the relationship to polyamine levels are discussed.  相似文献

The hypothesis for this study was that a higher dietary proportion of soluble fibre would result in stable and constant plasma metabolite and regulatory hormone concentrations. The study was a 4×4 Latin Square design with a sequence of 17 days adaptation to the ration followed by 8 sampling days. The feed rations consisted of only timothy hay (H), hay plus molassed sugar beet pulp combined with either whole oats (OB) or barley (BB) and hay plus a loose chaff-based concentrate (M). Four horses were fitted with permanent caecal cannulas and liquid caecal content was withdrawn manually and blood was drawn from the jugular vein at 0, 3 and 9 h postprandial. The horses were exercised daily at medium level for about 1 h. Samples were analysed for short-chain fatty acids (SCFA) and metabolic traits. Caecal SCFA and propionic acid concentrations increased with increased dietary starch and soluble fibre. The diet highest in soluble fibre (M) resulted in the highest plasma glucose and insulin concentrations in the morning, which then remained stable and constant throughout the day. A strong interaction (P<0.01) between time and diet was measured for plasma urea, glucose, insulin and leptin. The greatest variations in plasma glycaemic and insulinaemic responses were associated with the cereal grain diets (OB and BB). There were indications of a negative energy balance, which was reflected in a significantly higher plasma β-hydroxybutyrate concentration and a numerically higher non-esterified fatty acid concentration. In conclusion, this study found that inclusion of soluble fibre resulted in increased total caecal SCFA and propionic acid concentrations. This consequently resulted in stable and constant plasma glycaemic and insulinaemic responses. Diets with a high content of soluble fibre provided enough energy for horses at medium work level.  相似文献

Low and high concentrations of iodoacetate stimulated and inhibitedthe CO2 production, respectively. Soluble sugars in tissue slicesdecreased sharply in high iodoacetate and showed little decreasein low concentrations of the inhibitor. The same pattern ofchanges was observed with soluble nitrogen fractions. Polysaccharidesand proteins were subjected to slight, if any changes. Leakage of different metabolites from tissue slices was operativein low and high concentrations of iodoacetate; the magnitudeof leakage being increased with an increase in concentrationof the inhibitor. That low concentrations of iodoacetate may have increased CO2output through increased accessibility of substrates to enzymesin the cytoplasm, was suggested. The proportion of 26 fatty acids (FA) in the lipids of the cerebral cortex, subcortical formations (the thalamus, hypothalamus and basal ganglia) and the medulla oblongata was studied in rats aged 5, 10, 14 and 90 days. Very marked developmental changes in the proportion of the various FA were demonstrated in the different parts of the brain. In the cerebral cortex the proportion of 17:1 rose by 285%, 18:3 n-3 by 1820% and 22:6 n-3 by 80%, while the proportion of 14:0, 16:0 and 16:1 fell significantly. In the tissue of subcortical formations we found an increase in the proportion of FA with 18 carbons (18:0 by 40%, 18:1 by 100%, 18:3 n-3 by over 5000%) and a decrease in the proportion of 14:0, 16:0, 16:1 and 20:4 n-6. The situation in medulla oblongata tissue was similar to the one in subcortical formations. On comparing the proportion of FA in individual parts of the CNS in the same age category, we found the smallest number of statistically significant differences in 5-day-old rats. In adult rats we found significant differences chiefly in the proportion of palmitic acid (16:0), oleic acid (18:1), linolenic acid (18:3 n-3) and acids with 20-22 carbons.  相似文献

Studies on mediators of manganese peroxidase for bleaching of wood pulps   总被引:3,自引:0,他引:3  
In order to enhance the bleaching effect of manganese peroxidase (MnP), unsaturated fatty acids, thiol-containing compounds and various other organic compounds were applied in pulp bleaching experiments with MnP. Thiol-containing compounds did not improve the pulp bleaching effect by MnP. Some unsaturated fatty acids, linoleic acid and linolenic acid provided a better pulp bleaching effect than Tween 80. The correlation between the number of C=C bonds in a fatty acid and its pulp bleaching effect was also investigated. The MnP pulp bleaching capability was shown to depend on the carboxylic acid used. A combination of Tween 80 and a carboxylic acid resulted in higher pulp brightness than that obtained with Tween 80 alone. A laccase mediator, 3-hydroxy-1,2,3-benzotriazin-4(3H)-one, could also enhance the MnP pulp bleaching effect.  相似文献

大花三色堇性喜冷凉、忌酷热。该研究以大花三色堇3个自交系08H 、HAR 和 E01为材料,分别测定了40℃高温处理4、8和12 h 时不同基因型大花三色堇幼苗的生理指标,以及不同浓度(0.1、1、2 mmol?L-1)水杨酸预处理对热胁迫下大花三色堇幼苗耐热性的影响。结果表明:在高温胁迫下大花三色堇电解质外渗量增加,随着处理时间的延长,电解质外渗更多,可溶性糖含量先增加后降低,POD 酶活性先提高后降低;与其他2个自交系相比,HAR 表现出较好的耐热性,其可溶性糖含量、POD 酶活性的增加均较高,而电解质外渗率偏低;与对照相比,3种浓度 SA 预处理均显著降低了大花三色堇幼苗的电解质外渗率,增加了幼苗体内可溶性糖含量,提高了大花三色堇的幼苗体内脯氨酸含量和 POD 酶活性;其中1 mmol?L-1的 SA 预处理对高温胁迫下大花三色堇幼苗体内可溶性糖的含量增加最高,最大程度减缓幼苗体内的电解质外渗量,08H 和 HAR 的脯氨酸含量和 POD 酶活性达到最大值。而对 E01而言,0.1 mmol?L-1的水杨酸预处理的脯氨酸含量和 POD 酶活性最高。该研究结果探讨了高温胁迫下不同基因型大花三色堇幼苗的生理表现,以及外施水杨酸对增强大花三色堇幼苗耐热性的效果,为大花三色堇抗热栽培提供重要的基础资料。  相似文献
