Several investigators have reported on the detection of enteric viruses in marine sediments, but none determined the efficiency of their methods and only limited volumes of sediment were sampled. The purpose of this investigation was to develop a quantitative method for detecting enteroviruses in marine sediments so that their relative proportion to viruses freely suspended in estuarine water could be more accurately determined. Poliovirus was found to adsorb readily to natural marine sediments collected along the Texas Gulf coast. A number of substances were evaluated for their ability to elute adsorbed viruses. A solution of 10% fetal calf serum adjusted to pH 10.5 and 0.05M ethylenediaminetetraacetate (pH 11.0) were found to be the best eluents. Using ethylenediaminetetraacetate as an eluent, it was possible to elute virus from large volumes of sediment and reconcentrate the sediment eluate into an economically assayable volume (30 to 50 ml). Poliovirus could be recovered from the sediment with an overall efficiency of 50%. This method was found to be satisfactory for the recovery of naturally occurring animal viruses in estuarine sediments from the upper Texas Gulf coast.  相似文献

This study was designed to determine the degree of adsorption of enteric viruses to marine sediment and factors controlling this association. Adsorption and elution characteristics of several enteroviruses and one rotavirus to estuarine sediments were studied under varying conditions of pH, salinity, and presence of soluble organics. Greater than 99% of the added poliovirus type 1 (LSc), coxsackievirus type B3 (Nancy), echovirus type 7 (Wallace), and rotavirus (SA-11) adsorbed to sediment. Echovirus 1 (Farouk) and a recent isolate typed as coxsackievirus B4 adsorbed significantly less than poliovirus 1 under similar conditions of varying salinity and pH. The presence of soluble organic matter, in the form of secondary sewage effluent or humic acid, did not affect these patterns of adsorption. Only echovirus 1 (Farouk) desorbed when the pH or salinity was altered and then only to a small extent. Three recent isolates of echovirus 1 and echovirus 29 (strain JV-10) also demonstrated varying amounts of adsorption to sediment. These data indicate that enteric viruses can become readily associated with sediment in the estuarine environment and that this association may play a major role in their hydrotransportation and survival.  相似文献

Current standards for evaluation of the public health safety of recreational and shellfish-harvesting waters are based upon bacteriological analysis, but do not include an evaluation of the number of viruses. The objective of this study was to determine the occurrence of enteric viruses in estuarine sediments and to find a relationship, if any, between the presence of viruses in seawater or sediment or both and various biological and physicochemical characteristics of the environment. Viruses were found in greater numbers in sediment than in overlying seawater on a volume basis. Several types of enteroviruses were isolated: coxsackievirus types A16, B1, and B5, echovirus type 1, and poliovirus type 2. On several occasions, viruses were isolated from sediments when overlying seawaters met bacteriological water quality standards for recreational use. Statistical analysis of the relationship between viruses in seawater or in sediment and other variables measured yielded only one significant association: the number of viruses in sediment was found to be positively correlated with the number of fecal coliforms in sediment. No other physical, chemical, or biological characteristic of seawater or sediment that was measured showed statistically significant association with viral numbers. No correlation was found between bacterial indicators and virus in the overlying waters. The data indicated that evaluation of the presence of bacteria and viruses in sediment may provide additional insight into long-term water quality conditions and that indicator bacteria in water are not reflective of the concentration of enteric viruses in marine waters.  相似文献

Endospores of thermophilic bacteria are found in cold and temperate sediments where they persist in a dormant state. As inactive endospores that cannot grow at the low ambient temperatures, they are akin to tracer particles in cold sediments, unaffected by factors normally governing microbial biogeography (e.g., selection, drift, mutation). This makes thermophilic endospores ideal model organisms for studying microbial biogeography since their spatial distribution can be directly related to their dispersal history. To assess dispersal histories of estuarine bacteria, thermophilic endospores were enriched from sediments along a freshwater‐to‐marine transect of the River Tyne in high temperature incubations (50°C). Dispersal histories for 75 different taxa indicated that the majority of estuarine endospores were of terrestrial origin; most closely related to bacteria from warm habitats associated with industrial activity. A subset of the taxa detected were marine derived, with close relatives from hot deep marine biosphere habitats. These patterns are consistent with the sources of sediment in the River Tyne being predominantly terrestrial in origin. The results point to microbial communities in estuarine and marine sediments being structured by bi‐directional currents, terrestrial run‐off and industrial effluent as vectors of passive dispersal and immigration.  相似文献

While whole sediment toxicity tests with macrofaunal polychaetes are well developed and standardized, they are oftentimes not very sensitive to environmental contaminants. Meiofaunal polychaetes, however, are sensitive to contaminants, easy to culture, and representative of the interstitial habitat. These are desirable attributes for ecotoxicological assessments of marine and estuarine sediments. The meiofaunal polychaete, Dinophilus gyrociliatus Schmidt, 1857, is a cosmopolitan species that has become a useful tool for ecotoxicological assessments, particularly for its use in toxicity testing with sediment pore waters. Due to its short life cycle it is suitable for sublethal toxicity tests, with egg production by the females as a sensitive endpoint, which can be assessed in a 7-day exposure period. Toxicity assessments of pore waters from an industrialized bay in Texas, U.S.A., and of pore waters from sediments spiked with nitroaromatic explosives, demonstrated that the D. gyrociliatus reproduction endpoint was consistently among the most sensitive, when compared to early-life stage tests with several other marine species, including macro-algae, sea urchins and fish. There was also excellent agreement among the results of porewater tests with D. gyrociliatus, nauplii of the meiofaunal copepod, Longipedia americana, and embryos of the sea urchin, Arbacia punctulata in a survey with pore waters extracted from sediments collected in the vicinity of an oil platform in the Gulf of Mexico, where metals were the primary contaminants of concern.  相似文献

The Gulf of Paria is bordered by both Trinidad and Venezuela, from which various metallic pollutants and other contaminants can originate. The Gulf is still a significant source of fish, crabs and shellfish for human consumption to both countries, where concerns over the quality of this marine environment have been long expressed but never properly addressed. In addition, the circulatory current patterns in the Gulf ensure that contaminants originating from either country are likely to affect both countries eventually. Heavy metals were determined in oysters (Crassostrea rhizophorae and C. virginica), green mussels (Perna viridis) and sediments from the Gulf of Paria. Samples were obtained at four sites in Trinidad and three sites in Venezuela in the Gulf of Paria, in addition to comparative samples collected from three sites on the north coast of Venezuela. Edible tissues of twelve shellfish from each location were blended and aliquots digested with concentrated nitric acid, for extraction of cadmium, chromium, copper, lead, nickel and zinc. The solutions were analysed by flame atomic absorption spectroscopy. Mercury was extracted with a mixture of nitric, hydrochloric and sulphuric acids and determined by cold vapour atomic absorption. Sediments were oven-dried at 60'C, before being similarly extracted. Results showed that mercury in sediments at all sites in Trinidad and Venezuela exceeded NOAA and Canadian sediment quality guidelines, while cadmium, copper, nickel, lead and zinc also exceeded these guidelines at several sites. Heavy metal levels in oysters and green mussels varied widely with location. However, oysters from the Gulf of Paria contained significantly higher mean levels of cadmium, copper, nickel and zinc than those from the north coast of Venezuela, but this difference was not apparent in mussels. Cadmium, mercury and zinc in sediments were significantly correlated with those of mussels, but not of oysters, in which copper and zinc at several sites in the Gulf of Paria exceeded local maximum permissible levels (Cu = 20 microg g(-1) wet wt; Zn = 50 microg g(-1) wet wt) for human consumption. These findings indicate that while mussels may be better biological indicators of heavy metal pollution in sediments than oysters, the latter may provide copper and zinc contamination. Further research is needed to determine the most appropriate biological indicators of heavy metal and other pollutants in the local marine environment and to develop protocols for their use.  相似文献

Hurricanes Katrina, Rita, Gustav, and Ike deposited large quantities of sediment on coastal wetlands after making landfall in the northern Gulf of Mexico. We sampled sediments deposited on the wetland surface throughout the entire Louisiana and Texas depositional surfaces of Hurricanes Katrina, Rita, Gustav, and the Louisiana portion of Hurricane Ike. We used spatial interpolation to model the total amount and spatial distribution of inorganic sediment deposition from each storm. The sediment deposition on coastal wetlands was an estimated 68, 48, and 21 million metric tons from Hurricanes Katrina, Rita, and Gustav, respectively. The spatial distribution decreased in a similar manner with distance from the coast for all hurricanes, but the relationship with distance from the storm track was more variable between events. The southeast-facing Breton Sound estuary had significant storm-derived sediment deposition west of the storm track, whereas sediment deposition along the south-facing coastline occurred primarily east of the storm track. Sediment organic content, bulk density, and grain size also decreased significantly with distance from the coast, but were also more variable with respect to distance from the track. On average, eighty percent of the mineral deposition occurred within 20 km from the coast, and 58% was within 50 km of the track. These results highlight an important link between tropical cyclone events and coastal wetland sedimentation, and are useful in identifying a more complete sediment budget for coastal wetland soils.  相似文献

Methods were developed for detecting and concentrating enteric viruses in municipal solid waste landfill leachates. Poliovirus added to a leachate was not readily detectable, possibly because the virus was adsorbed to the leachate particulates. The masking effects associated with suspended solids in the leachate were overcome by adding a final 0.1 M sodium (tetra)ethylenediaminetetraacetate concentration to the leachate. A sodium (tetra)ethylenediaminetetraacetate-treated leachate could be clarified by filtration at pH 8.0 without a loss of virus. The clarified and sodium (tetra)ethylenediaminetetraacetate-treated leachate contained interfering materials of an anionic nature which prevented virus adsorption to epoxy-fiber glass filters. This interfering effect was overcome by treating the leachate with an anion-exchange resin. Viruses in the resin-treated leachate were concentrated by adjusting the leachate to pH 3.5, adding AlCl(3) to a final 0.005 M concentration, adsorbing the viruses to an epoxy-fiber glass virus adsorbent, and eluting the adsorbed viruses in a small volume. When this method was used to concentrate poliovirus 100-fold in a variety of leachates, the average virus recovery efficiency was 37%. With the methods described in this study, it should be possible to efficiently monitor solid waste disposal site leachates for enteric viruses.  相似文献

AIMS: This paper evaluates the presence of human pathogen micro-organisms in marine sediments in a coastal area suitable for bathing. In addition, the grain size analysis was performed in order to correlate the characteristics of the sediments and the microbial content. METHODS AND RESULTS: The samples were collected in two small bays along the central Adriatic coast, where breakwaters had been built for the purpose of halting marine erosion. Faecal contamination indicators, Salmonella and Vibrio species, enteric viruses were investigated using standard techniques for isolation and identification. The grain size was determined using calibrated sieves and 'Sedigraph' device. In some samples, the faecal contamination indices overstepped legislative limits. Salmonella was never found. Vibrio and enteric viruses were isolated: the micro-organisms were preferentially abundant in fine sediments. CONCLUSIONs: Marine sediments can represent an important reservoir of allochthonous and marine micro-organisms and the microbial charge correlates with the characteristics of the sediments. SIGNIFICANCE AND IMPACT OF THE STUDY: This study indicates that it is important to analyse marine sediments before defining the quality of coastal areas.  相似文献

Bacteria are the most abundant and active organisms in marine sediments and are critical for nutrient cycling and as a food source to many benthic and pelagic organisms. Bacteria are found both as free-living cells and as particle-associated cells, which can make investigations of these communities difficult. We found that common procedures for extracting bacteria from sediments leave the bacteria clay particle-associated and the clay particles clump, which reduce the reproducibility of direct counts. We optimized a sonication/surfactant method that produces a homogeneous suspension of bacterial cells against a uniform background of clay particles, which results in reproducible samples for epifluorescence microscopy. We developed a method to estimate CTC-positive cells and cell-specific CTC content in intact cores of surficial sediment communities from riverine, estuarine and coastal sites. Benthic bacterial abundances averaged 4.9x10(8) cells/g dry wt sediments in Apalachicola River, Florida sediments, 4.9-13.8x10(9) cells/g dry wt sediments in a variety of Apalachicola Bay sediments and 3.6x10(8) cells/g dry weight in shallow, anoxic Gulf of Mexico sediments. Percent CTC-positive cells ranged from low values of 9-10% CTC-positive cells in Apalachicola River and Apalachicola Bay sediments to high values of 25% CTC-positive cells in anoxic Gulf of Mexico sediments. After correction for abiotic CTC reduction and chlorophyll interference, estimates of cell-specific CTC reduction ranged from 0.15 to 0.55 fmol CTC(red)/active cell in the Apalachicola Bay sediments to 1.6 to 3.8 fmol CTC(red)/active cell in anoxic Gulf of Mexico sediments.  相似文献

Samples were collected sequentially from water, suspended solids, fluffy sediments (uppermost layer of bottom sediments), and compact sediment. A total of 103 samples were examined of which 27 (26%) were positive for virus. Polioviruses were recovered most often, followed by coxsackie B viruses and echoviruses 7 and 29. Virus was found most often attached to suspended solids: 72% of these samples were positive, whereas only 14% of water samples without solids yielded virus. Fluffy sediments yielded virus in 47% of the samples, whereas only 5% of compact bottom-sediment samples were positive. When associated with solids, poliovirus and rotavirus retained their infectious quality for 19 days. The same viruses remained infectious for only 9 days when freely suspended in seawater. Collection of suspended solids at ambient water pH appears to be very useful for the detection of virus; it has advantages over collecting and processing large volumes of water, with accompanying pH adjustment and salt addition for processing.  相似文献

Capitella capitata was traditionally used as a biological indicator species due to its ubiquitous distribution and high densities in disturbed and polluted marine and estuarine sediments. Based on allozyme and developmental studies, it is now clear that C. capitata is a species complex consisting of multiple distinct lineages worldwide, including the recently described C. teleta, a model species for spiralian development. The coast of the northern Gulf of Mexico, with its numerous bays and estuaries and frequently occurring natural and anthropogenic disturbances, provides an appropriate region for such studies. We sequenced a fragment of the mitochondrial cytochrome c oxidase subunit I gene for individuals of C. cf. capitata and C. cf. aciculata (distinguished by acicular spines on the first two chaetigers) collected from Texas and Florida coasts and analyzed them in conjunction with data available in GenBank. Our results indicate the presence of a Gulf of Mexico clade that is distinct from populations in Canada and the Indo‐Pacific. Populations in the northern Gulf of Mexico are structured geographically, with support for Texas and Florida clades, and there do not seem to be clear boundaries between C. cf. capitata and C. cf. aciculata. This is corroborated by the fact that multiple specimens were morphologically intermediate between the two species. In future studies, we aim to clarify whether the intermediate morphologies represent ontogenetic stages, neutral morphological variation, phenotypic plasticity, or sexual dimorphism in a single species or whether several lineages with incomplete reproductive barriers are present.  相似文献
