Male Sprague-Dawley rats were treated ip with beta-naphthoflavone (40 mg/kg/day) in corn oil or in DMSO for three days. Diphenaldehyde (90 mg/kg in DMSO) was injected ip 24 hr after pretreatment. The increase in the levels of aspartate aminotransferase, alanine aminotransferase, sorbitol dehydrogenase, gamma glutamyl transpeptidase, and lactate dehydrogenase was significantly lower in rats pretreated with BNF. This suggests that the BNF-induced P-450 isozyme systems have a protective effect against the acute hepatotoxicity of diphenaldehyde.  相似文献

Cultured epithelial cell lines from normal rat livers were shown to undergo gradual transformation and malignancy which increased with time. Morphological changes appeared both before and after cells had attained a malignant state, as detected by agar tests. The progression of the degree of malignancy was determined by the morphological appearance of the cells, the increase in the number and size of cell colonies in soft agar, the expression of gamma glutamyl transferase (GT) and the shortening of the latency period necessary for tumor formation after transplantation to syngeneic rats of cells from sequential passages.  相似文献

Anthglutin, a new inhibitor of γ-glutamyl transpeptidase, has been isolated from the cultured medium of Penicillium oxalicum and its structure established as l-γ-l-glutamyl-2-(2-carboxyphenyl)hydrazine. The isolation of anthglutin was achieved by ion-exchange chromatography. Anthglutin inhibited γ-glutamyl transpeptidase specifically and the kinetic analysis of the inhibition showed that anthglutin inhibited the enzyme competitively with regard to the glutamyl donor, γ-glutamyl-p-nitroanilide, and noncompetitively with regard to the glutamyl acceptor, glycylglycine. K1 values were 5.7 μm for the hog kidney enzyme, 18.3 μm for the human kidney enzyme, 13.6 μm for the human liver soluble enzyme, and 10.2 μm for the bound enzyme. After oral administration of [14C]methionine and anthglutin to rats, no effect of anthglutin was observed on the absorption of methionine in the intestine.  相似文献

The histopathological response and cell culture characteristics of liver cells from the R16 (grc ) strain of rats, which carries an MHC-linked deletion, were examined one week after a single intraperitoneal injection of 200 mg/ kg body weight diethylnitrosamine (DEN) and were compared with the response of liver cells from wild type (grc+) rats. The DEN exposure induced hydropicl vacuolar changes in the parenchymal cells and a limited proliferation of oval cells in the periportal areas of the livers of both grc+ and grc rats. Primary culture of collagenase-digested livers consisted of parenchymal, bile ductular and oval-related cells as determined by cell-specific immunohistochemistry. Subpassaged cells from grc+ rats exhibited oval cell ultrastructural morphology, inducible histochemical staining for gammaglutamyl transpeptidase (GGT), and DEN-associated onset of anchorage-independent growth. Spontaneous cholelithiasis was found in seven owl monkeys (Aotus spp.) at necropsy. There were four male and three female animals. Antemortem clinicopathologic findings included weight loss, anemia, increased alanine aminotransferase and gamma glutamyl transpeptidase, and hyperbilirubinemia in several animals. Choleliths ranged in size from sand-like particles to 5 mm in diameter. Gallstones from five animals were analyzed by accepted analytical methods. Results showed the gallstones to be composed primarily of cholesterol (89%). The gallbladder was histologically normal in all cases examined. The etiopathogenesis of cholelithiasis in the owl monkey is unknown.  相似文献

Anti-hepatotoxic activity of methanol extract of Coscinium fenestratum stem (MEC) was investigated against carbon tetrachloride-induced hepatopathy in rats. Hepatotoxic rats were treated with MEC for a period of 90 days (60mg/kg body weight, daily, orally by intubation). Anti-hepatotoxic effect was studied by assaying the activities of serum marker enzymes like aspartate aminotransferase, alanine aminotransferase, alkaline phosphatase, gamma glutamyl transpeptidase, lactate dehydrogenase etc. and glucose (6) phosphate dehydrogenase in liver. We also estimated the concentrations of total proteins, total lipids, triglycerides, phospholipids and cholesterol in serum, liver and kidney. The activities of all the marker enzymes registered a significant elevation in carbon tetrachloride-treated rats, which were significantly recovered towards an almost normal level in animals co-administered with MEC. Other biochemical changes induced by carbon tetrachloride too showed reliable signs of retrieving towards the normalcy. Histopathological analysis confirmed the biochemical investigations. This study unravels the anti-hepatotoxic activity of MEC.  相似文献

The effects of garlic and neem leaf extracts on lipid peroxidation and antioxidant status during administration of N-methyl-N'-nitro-N-nitrosoguanidine (MNNG), a carcinogenic nitrosamine were evaluated in male Wistar rats. Extracts of garlic and neem leaf were administered orally for five consecutive days before intraperitoneal injection of MNNG. Enhanced lipid peroxidation in the stomach, liver and circulation of MNNG-treated rats was accompanied by a significant decrease in glutathione (GSH) and the activities of glutathione peroxidase (GPx), glutathione-S-transferase (GST) and gamma glutamyl transpeptidase (GGT). Administration of garlic and neem leaf extracts significantly decreased the formation of lipid peroxides and enhanced the levels of antioxidants and detoxifying enzymes in stomach, the primary target organ for MNNG, as well as in the liver and circulation. The results of the present study suggest that garlic and neem may exert their protective effects by modulating lipid peroxidation and enhancing the levels of GSH and GSH-dependent enzymes.  相似文献

Zinc nutritional status appears to decline with age in humans and rodents. Since germ-free rats outlive their conventional counterparts in better health, serum Zn levels were determined in male germ-free and conventional Lobund Wistar rats in samples originating from the Lobund Aging Study. Starting at 5 months of age, germ-free rats showed significantly higher serum Zn levels than did their conventional counterparts. In conventional rats sacrificed up to 30 months of age in apparently good health, serum Zn levels showed no effect of age, while a slight but significant increase with age was observed in the germ-free rats. In healthy germ-free adults (6-24 months of age), serum Zn concentrations were approximately 25% higher than those in comparable conventional animals. In conventional rats 18-30 months of age (average, 24.5 months), sacrificed because of an obvious moribund condition, serum Zn levels were significantly lower than those in rats of the same age range (average, 24.9 months) that were obviously healthy. Results suggest that the often observed higher absorptive capacity of the germ-free gut might have contributed to higher serum Zn levels, and that a decline in serum Zn concentration with age may be a consequence, rather than a causative factor, of declining health.  相似文献

Pretreatment of rats with colchiceine (10 micrograms/day/rat) for seven days protected against CCl4-induced liver damage. CCl4 intoxication was demonstrated histologically and by increased serum activities of alanine amino transferase (ALT), alkaline phosphatase (Alk. Phosph.) gamma glutamyl transpeptidase (GGTP), bilirubins and decreased activity of glucose-6-phosphatase (G-6Pase). Furthermore, an increase in liver lipid peroxidation and a decrease in plasma membrane GGTP and Alk. Phosph. activities were found. Colchiceine increased 1.5-fold the LD50 of CCl4 and prevented the release of intracellular enzymes as well as the decrease in GGTP and Alk. Phosph. activities in plasma membranes. It also completely prevented the lipid peroxidation induced by CCl4 and limited the extent of the histological changes.  相似文献

A retrospective study evaluated the influence of sex and age on plasma biochemistry and haematology parameters in a captive-bred colony of baboons. Over 1,140 ETDA and heparin blood samples were obtained from 160 clinically normal baboons between the ages of 11 months and 11 years. Data for these blood tests were analysed for the effects of sex, age and sex age interactions. Sex, age and sex age interactions were detected for many plasma biochemistry and haematological parameters. The reference range values for platelets, white-blood cells and mean corpuscular volume and plasma chloride, glucose, total protein and iron were higher (P < 0.01) and red blood cell, plasma sodium, potassium, total CO2, creatinine, urea, total bilirubin, albumin, alkaline phosphate, gamma glutamyl transpeptidase and phosphate were lower (P < 0.01) in the female compared to the male population. Sex age interactions (P < 0.05) were seen with haemoglobin, white blood cells, haematocrit, mean corpuscular volume, sodium, creatinine, urea, calcium, phosphate, total bilirubin, total protein alkaline phosphatase, the liver enzymes and triglycerides. Plasma alkaline phosphatase was highest ( > 800 micro/l) in young juveniles of both sexes; creatinine was higher in older ( > 4 years) compared to younger baboons of the same sex (P < 0.05). Plasma cholesterol and triglycerides were greater (P < 0.01) in young baboons compared to older animals.  相似文献

Unrestricted access of vervet monkeys to a palatable solution of ethanol resulted in chronic intake or abstinence. Self-selected dose averaged 4.42 g/kg body mass/day. Growth of juveniles was not grossly affected. In plasma, chronic ethanol intake was associated with lower average K and Cl values, increased gamma glutamyl transpeptidase in some adults, and higher average total cholesterol. Neither hepatic histology nor haematology were affected by chronic ethanol intake.  相似文献

Effect of treatment of female rats with an oral contraceptive agent (OCA), Ovulen-50, for 7 weeks on agglutination of hepatocytes with concanavalin A (con A) and activities of certain tumor marker enzymes were examined to find out if OCA treatment is related to preneoplastic or neoplastic processes. Hepatocytes from regenerating and nonregenerating livers of control female rats showed negligible agglutination with Con A, whereas hepatocytes from non regenerating but not from the regenerating livers of female rats treated with a combination of 5 micrograms ethinyl estradiol and 100 micrograms ethynodiol diacetate showed agglutination. Of the tumor marker enzymes such as hepatic glucose 6-phosphatase, gamma-glutamyl transpeptidase (gamma-GT), and arginase examined in the liver, only gamma-glutamyl transpeptidase showed a significant increase in activity in the steroid-treated rats. Plasma alkaline phosphatase activity was also higher in the treated animals. However, the magnitude of the changes observed was relatively small and perhaps unrelated to the neoplastic process.  相似文献

CCl4 alone treatment (0.lml of liquid paraffin/100g body weight, ip) for 7 days followed by 0.l ml of CCl4 (in liquid parafiin/100g body weight, ip) from day 8 till day 14, caused a 16 fold increase in lipid peroxidation and a 50% reduction in catalase and glutathione reductase in liver tissue of rats accompanied by an increase in the activities of transaminases. alkaline phosphatase, lactate dehydrogenase and gamma - glutamyl transpeptidase in serum as compared to liquid paraffin treated control. Pretreatment of ethanolic leaf extract of C. fistula (500mg/kg body weight/day for 7 days) followed by CCl4 treatment (0.1 ml/100g body weight from day 8 till day 14) completely reversed back lipid peroxidation and the activities of catalase and glutathione reductase in the liver tissue towards normalcy. This treatment also reversed the elevated levels of the enzymes in the serum. Ethanolic leaf extract alone treatment did not produce any change in all the parameters studied. The results suggest antioxidant and hepatoprotective properties of C. fistula during its pretreatment against CCl4 induced hepatotoxicity.  相似文献

Silkworm, Bombyx mori, contains beneficial components such as protein, minerals, amino acids and omega-3. We previously reported a technique to produce silkworms in an easy to eat Hongjam. In this study, the inhibitory effect of Hongjam (50, 100, and 300 mg/kg body weight) on ethanol liver damage was investigated. Normal diet, ethanol, ethanol with Hongjam were administered to male Sprague-Dawley rats for 4 weeks. Administration of Hongjam reduced triglyceride levels in plasma and liver. In addition, plasma concentrations of alanine transaminase, aspartate aminotransferase and gamma glutamyl transpeptidase were significantly decreased in the Hongjam administered group. Moreover, Hongjam administration effectively reduced the plasma level of interleukin-1 beta, a pro-inflammatory cytokine. These results suggest that dietary intake of Hongjam can prevent alcoholic liver disease.  相似文献

Activity of gamma glutamyl transpeptidase (GGT) in the testes of mature and prepuberal rats was investigated histochemically and biochemically. Histochemically, the enzyme activity was localized predominantly in the arterial and arteriolar endothelium and was absent from the capillaries and the seminiferous tubules. The activity in the arterial endothelium extended to the testicular artery on the surface of the testis and in the spermatic cord, but the veins in both the pampiniform plexus and on the testicular surface were negative. The endothelium of the testicular artery was already faintly positive at birth, and the activity increased during the second and third postnatal week during the branching and remodeling of the intratesticular arteries and arterioles. Activity of GGT was estimated quantitatively after dissection of the testis into tubular and interstitial tissue. The enzyme activity was very low in the tubules. It was fivefold stronger in the interstitium, and this activity was further enhanced by pretreatment of the dissected tissue with collagenase to remove the Leydig cells.  相似文献

A comparative study on the biochemical parameters in hydatid cyst fluids of sheep, goats, camels, cattle and human cystic forms of Echinococcus granulosus has been made in Libya. Quantitative variations in the levels of sodium, potassium, calcium, cholesterol, glucose, urea, creatinine and gamma glutamyl transpeptidase (gamma GT) were found in the cystic fluids of different host origins although these differences were statistically insignificant compared with the hydatid fluids of sheep. However, the concentration of triglycerides and proteins were significantly elevated in the cyst fluids of sheep compared with the other fluids studied. Similarities in the biochemical composition of different hydatid cyst fluids suggest the existence of sheep strains of E. granulosus in human and other domestic animal intermediate hosts in Libya.  相似文献

The exorbital lacrimal glands (ELG) of rats were examined for both sexes to determine what degree of harderianization occurred as a function of age and after castration, and to investigate its time course and origin in ELG. Light microscopically, very small Harderian foci were seen in the ELG of both sexes at 3 weeks of age. As the male rats became older, the relative volume of the Harderian gland (HG) cells in the ELG increased. At age 6 months, the value was 1.25 +/- 0.31% in males and 0.13 +/- 0.05% in females (p less than 0.05). After castration, a significant decrease (0.21 +/- 0.01%, p less than 0.05) was observed in that of male ELG. In contrast, in female ELG, HG cells were inconspicuous and the relative volume of those did not vary during this experimental period or after castration. It appeared that the HG cells had developed from undifferentiated basal cells of the acini and the intercalated ducts in the ELG at age 2-6 months. Then, at age 22 months, they also probably developed from those of the excretory ducts of the ELG.  相似文献

Initiation of RNA synthesis was studied in an attempt to determine a possible molecular mechanism for age-related biochemical and physiological changes. Initiation of RNA synthesis was determined by incorporation of [gamma-32 P]ATP and of [gamma-32P]GTP into an acid-insoluble product by intact nuclei isolated from livers of Sprague-Dawley CD-strain rats of various ages. When the rats were grouped into young (0.75-9 months) and old (12-30 months) rats, a significant decrease (P less than or equal to 0.001) in incorporation of initiating nucleotides was observed. The rat population was divided into five age groups (0.75-3 months, 4-9 months, 12-18 months, 19-23 months and 30 months) for further analysis of the effect of age on the initiation of RNA synthesis. Analysis of data from these groups indicated a significant trend for an age-related decrease in RNA-synthesis initiation (correlation coefficient = 0.94). Long-term hypophysectomy coupled with minimal hormone-replacement therapy was shown to have a significant effect on the reversal of the age-related decrease in initiation of RNA synthesis. It was observed that initiation of RNA synthesis in nuclei from 19-month-old rats, hypophysectomized at 12 months of age, was closest to that in 3-month-old intact rats and was not significantly different from that in liver nuclei of 0.75-9-month-old intact rats.  相似文献

Free radicals cause cell injury, when they are generated in excess or when the antioxidant defense is impaired. Carbon tetrachloride (CCl4) is used as a model for liver injury. In this study antioxidant activity of ethanol extract of A. fertilisima (EEA) was investigated using CCl4 intoxicated rat liver as the experimental model. Oral administration of EEA at a dose of 100 mg/kg body weight, for 14 consecutive days, the rate of the production of antioxidant enzymes like super oxide dismutase, catalase, glutathione peroxidase and glutathione transferase in rats compared to the CCl4 treated group without any supporting treatment. Liver damage is detected by the measurement of the activities of serum enzymes like aspartate aminotransferase, alanine aminotransferase, gamma glutamyl transpeptidase and alkaline phosphatase which were released in to the blood from damaged cells. The normalization of these enzymes levels was observed in rats treated with EEA (100 mg/kg body weight) by reducing the leakage of the above enzymes in to the blood. The findings provide a rationale for further studies on isolation of active principles and its pharmacological evaluation. Protection offered by silymarin (standard reference drug) seemed relatively greater.  相似文献
