In this study, potential banker plant systems against the cabbage whitefly Aleyrodes proletella Linnaeus (Hemiptera: Aleyrodidae) were developed under controlled conditions. The two most promising banker plant systems, that is, the parasitoid Encarsia tricolor Förster (Hymenoptera: Aphelinidae) either with Aleyrodes lonicerae Walker on European columbine (columbine system) or with Trialeurodes vaporariorum Westwood (Hemiptera: Aleyrodidae) on Hokkaido squash (pumpkin system), were further evaluated in the field. Although the pumpkin system produced three times more parasitoids than the columbine system, both banker plants led to an 1.5-fold increase in A. proletella parasitism rates. However, only the pumpkin system increased the abundance of syrphid larvae on cabbage by 61.5% and reduced A. proletella populations on average by 4.4%–25.8% depending on the respective assessment date. In conclusion, the pumpkin system revealed to be a promising (supplementary) control measure against A. proletella. Options for further improvement and standardization of the pumpkin system as well as a potential implementation in cabbage production are discussed.  相似文献

A transgene stacking system is a prerequisite for the introduction of multiple genes and for the modification of complex metabolic pathways in plants. We demonstrate here that the MAT-vector system previously used for generating marker-free transgenic plants is also an efficient and reliable transformation system for the repeated introduction of multiple transgenes independent of sexual crossing. We previously reported that the GST-MAT vector system, in which excision of the yeast site-specific recombination R/RS system is regulated by the maize GST-II-27 promoter, could generate marker-free transgenic plants containing a single transgene with high frequency. Here we show that the GST-MAT vector can be used successfully to introduce a second transgene (GFP) into a marker-free transgenic tobacco line containing single copies of the first transgenes (nptII and uidA genes). The transgene-stacked marker-free transgenic tobacco plants were generated from ca. 20% of excision-positive ipt-shooty explants within 5 months of Agrobacterium infection. The presence of uidA, nptII, GFP genes and the absence of the ipt gene were verified by PCR analyses. Furthermore, Southern blot analysis showed that no chromosomal rearrangements were introduced between the first and second transformations.  相似文献

Summary Neurospora crassa can utilize various purine bases such as xanthine or uric acid and their catabolic products as a nitrogen source. The early purine catabolic enzymes in this organism are regulated by induction and by ammonium repression. Studies were undertaken to investigate purine base transport and its regulation in Neurospora. The results of competition experiments with uric acid and xanthine transport strongly suggest that uric acid and xanthine share a common transport system. It was also shown that the common transport system for uric acid and xanthine is distinct from a second transport system shared by hypoxanthine, adenine and guanine, and apparently also distinct from the transport system(s) for adenosine, cytosine and uracil. Regulation of the uric acid-xanthine transport system and the hypoxanthine-adenine-guanine transport system was studied. The results reveal that the uric acid-xanthine transport system is regulated by ammonium repression, but does not require uric acid induction. Neither ammonium repression nor uric acid induction controls the hypoxanthine-adenine-guanine transport system. A gene, designated amr, which is believed to be a positive regulatory gene for nitrogen metabolism of Neurospora crassa, was found to dramatically affect both the uric acid-xanthine transport system and the hypoxanthine-adenine-guanine transport system. A model for the action of the amr locus as a positive regulatory gene and for the interaction between the amr gene product and its recognition sites will be discussed.  相似文献

Sporobolomyces ruberrimus is insensitive to antimycin A which is a respiratory inhibitor of the cytochrome system, as cyanide is. When this red yeast was cultured in the presence of antimycin A, the growth curve showed the same pattern as that of the normal culture in the absence of it, but the growth mass was only about 70% of that of the normal culture. The antimycin A-insensitive and cyanide-insensitive respiration of Sp. ruberrimus was inhibited by pyrocatechol and salicylhydroxamic acid. Sporobolomyces red yeasts have two characteristic terminal oxidase systems; one is a cytochrome oxidase system and the other is a cyanide- and antimycin A-insensitive oxidase system. The proportions of the two respiratory systems differed among the species and strains of Sporobolomyces red yeasts examined.  相似文献

It takes about 2 months for the molding process of Katsuobushi (dried bonito) production. A model of a short-term system for the molding process was desirable for the efficient selection of useful fungi for Katsuobushi production by evaluating the flavor change in the model system.

A liquid culture system using Nijiru (waste fluid of Katsuobushi sterilization), which is obtained from the Kezuribushi (sliced Katsuobushi) manufacturing process and contains abundant phenolic compounds of smoke tar, was found to be suitable for the short-term evaluation of a 10 day culture.  相似文献

Bacillus subtilis 168 was developed as a genome vector to manipulate large DNA fragments. The system is based on the inherent natural transformation (TF) activity. However, DNA size transferred by TF is limited up to approximately 100 kb. A conjugal transfer system capable of transferring DNA fragments considerably larger than those transferred by TF was developed. A well-defined oriT110 sequence and a cognate relaxase gene from the pUB110 plasmid were inserted into the xkdE gene of the B. subtilis genome. Transfer of antibiotic resistance markers distant from the oriT110 locus to the recipient B. subtilis occurred only in the presence of pLS20, a helper plasmid that provides a type IV secretion system. Marker transmission was consistent with the orientation of oriT110 and required a recA-proficient recipient. The first conjugal transfer system of genomic DNA should provide a valuable alternative genetic tool for editing the B. subtilis genome.  相似文献

Summary The locomotion ofAmoeba proteus has been investigated by algorithms evaluating correlation dimension and Lyapunov spectrum developed in the field of nonlinear science. It is presumed by these parameters whether the random behavior of the system is stochastic or deterministic. For the analysis of the nonlinear parameters, n-dimensional time-delayed vectors have been reconstructed from a time series of periphery and area ofA. proteus images captured with a charge-coupled-device camera, which characterize its random motion. The correlation dimension analyzed has shown the random motion ofA. proteus is subjected only to 3–4 macrovariables, though the system is a complex system composed of many degrees of freedom. Furthermore, the analysis of the Lyapunov spectrum has shown its largest exponent takes positive values. These results indicate the random behavior ofA. proteus is chaotic and deterministic motion on an attractor with low dimension. It may be important for the elucidation of the cell locomotion to take account of nonlinear interactions among a small number of dynamics such as the sol-gel transformation, the cytoplasmic streaming, and the relating chemical reaction occurring in the cell.  相似文献

China remains by far the largest aquaculture producer in the world. However, biofilms formed by pathogenic Vibrio strains pose serious problems to marine aquaculture. To provide a strategy for biofilm prevention, control, and eradication, extracts from 88 marine actinomycetes were screened. Thirty-five inhibited the biofilm formation of Vibrio harveyi, Vibrio vulnificus, and Vibrio anguillarum at a concentration of 2.5% (v/v). Thirty-three of the actinomycete extracts dispersed the mature biofilm. Six extracts inhibited the quorum-sensing system of V. harveyi by attenuating the signal molecules N-acylated homoserine lactones’ activity. Strain A66, which was identified as Steptomyces albus, both attenuated the biofilms and inhibited their quorum-sensing system. It is suggested that strain A66 is a promising candidate to be used in future marine aquaculture.  相似文献

Areas with ancient clearance cairns have been studied in Hamneda, Småland Uplands, southern Sweden, by archaeological and palaeoecological methods. Based on numerous radiocarbon dates and stratigraphical analyses, the local introduction of stone clearance is dated to the first century A.D. The clearance cairns reflect a system of semi-mobile cultivation that lasted until c. A.D. 900. Pollen and macrofossil analyses provide information on cereal growing and pastures in these clearance cairn areas, while charcoal analyses reveal details on the agrarian expansion dynamics and the use of fire in vegetation clearance. In the expansion phase, Quercus (oak) woodlands were cleared and transformed to open pastures and arable land, partly by the use of fire. A secondary succession of Betula (birch) and Corylus (hazel) was dealt with by fire clearances to keep pastures open and to prepare new arable plots. In the long run, Betula in particular was favoured by the land-use system. The mobility of the cultivation system is discussed together with the causes behind the introduction of stone clearance. A possible causal relationship with the introduction of hay mowing is also discussed.  相似文献

为了解鸳鸯茉莉(Brunfelsiaacuminata)花色变化的机理,采用高效液相色谱(HPLC)体系检测其开花过程中花青素组成的变化。结果表明,优化的HPLC体系为:流速为0.8 mL min–1,流动相A为7.5%甲酸乙腈,流动相B为7.5%甲酸水,洗脱程序为0 min,8%A;15 min,18%A;25 min,23%A;45 min,40%A;50 min,8%A。利用优化体系检测到鸳鸯茉莉花瓣中含有锦葵色素-3-O-葡萄糖苷、矮牵牛素葡萄糖苷和飞燕草素葡萄糖苷3种花青苷,其中锦葵色素-3-O-葡萄糖苷的含量最高,飞燕草素葡萄糖苷含量最低,且在花色由深变浅的过程中3种花青苷的含量均降低。因此,鸳鸯茉莉的呈色与这3种花青苷有关,且锦葵色素-3-O-葡萄糖苷起主导作用。  相似文献

A biallelic, unifactorial, sexual incompatibility system, consisting of two mating types, is characteristic of the ascomycetous yeasts, such asSaccharomyces cerevisiae. In addition to the biallelic system, the basidiomycetous yeasts,Rhodosporidium andLeucosporidium have a multiple allelic bifactorial system with 2 A and 2B factors for a total of four mating types. This report presents evidence for an increase in this number of mating types inLeucosporidium scottii due to the presence of a minimum of 5 A and 3 B specificities with linked loci at each factor. These results indicate that sexual incompatibility system in basidiomycetous yeasts is similar to that found in other basidiomycetes such asSchizophyllum commune.  相似文献

Summary A large number of motility mutants of the gliding bacterium Myxococcus xanthus have been isolated and analyzed by transduction. Almost all nonmotile mutants are found to be double mutants. This is explained by the existence of two parallel and almost independent multi-gene systems controlling motility, in which case at least one mutation in each system would be required eliminate motility. Only one locus, called mgl, has been found to be shared by both systems. Wild type cells move singly and in groups. Single cells move if they carry a complete gene system A, the genes of which are described in the preceding paper. Groups of cells can move if they carry a complete gene system S, but single AS+ cells do not move. Linkage analysis reveals at least 9 different loci in system S. One class of S mutants, those mutated in the locus tgl, are conditional mutants which, after contact with tgl + cells, become temporarily motile as cell groups. Most system A mutations have little effect on fruiting but many system S mutations block it, suggesting that system S plays a role in the fruiting process.  相似文献

The chromosome of E. coli is maintained in a negatively supercoiled state, and supercoiling levels are affected by growth phase and a variety of environmental stimuli. Regulation of DNA supercoiling yields a complex spectrum of effects on the E. coli recA system. Previous studies indicated that inhibition of DNA gyrase by antibiotics that act on the DNA gyrase A subunit results in turning on the recA system. Here we show that antisense ribozymes that act on the DNA gyrase A subunit can also induce recA. We used real time PCR and immunoblot to analyze the impact of DNA gyrase A inhibition by antisense ribozymes on recA expression. When gyrase A was inhibited by the RNase P mediated antisense ribozymes the expression of recA was induced around 130-fold as seen by real time PCR analysis. This suggests that repair pathway is induced by antisense ribozymes against DNA gyrase A and the damage produced by these ribozymes may be similar to that produced by fluroquinolones.  相似文献

Acacia sciophanes is an extremely rare and Critically Endangered species known from two small populations separated by less than 7 km. Specifically we aimed to investigate whether rarity in A. sciophanes is associated with decreased levels of genetic variation and increased levels of selfing by comparing patterns of genetic variation and mating system parameters with its widespread and common sister species A. anfractuosa. Fourteen polymorphic allozyme loci were used to assess genetic diversity with four of these used in the estimation of mating system parameters. At the species level A. sciophanes has lower allelic richness, polymorphism, observed heterozygosity and gene diversity than A. anfractuosa and significantly lower levels of gene diversity at the population level. Both species have a mixed mating system but the largest population of A. sciophanes has lower levels of outcrossing, higher correlated paternity and increased bi-parental inbreeding compared with two A. anfractuosa populations. However, both correlated paternity and bi-parental inbreeding appear to be at least partly influenced by population size regardless of the species. We suggest that A. sciophanes is likely to be an intrinsically rare species and that in particular the lower levels of genetic diversity and increased selfing are a feature of a species that has the ability to persist in a few localised small populations. Despite recent extensive habitat destruction our comparative study provided no clear evidence that such events have contributed to the lower genetic diversity and increased selfing in A. sciophanes and we believe its ability to exist in small populations may not only be an important factor in its survival as a rare species but also indicates that it may be less susceptible to the impacts of habitat loss and fragmentation. The key to this species conservation will be the maintenance of suitable habitat, particularly through improved fire regimes and control of invasive weeds, that will allow the two small populations to continue to persist in extremely restricted areas of remnant vegetation.  相似文献

We studied the effect of the common ascidian Styela clava on the growth of small holothurians of the species Apostichopus japonicus (Selenka, 1867). In a traditional aquaculture system, the oxygen consumption rates, ammonia excretion rates, and ingestion rates of small A. japonicus were reduced significantly, which suggested that this group was stressed by the presence of large A. japonicus, and that this stress grew stronger with time. Oxygen consumption rates, ammonia excretion rates, and ingestion rates of small A. japonicus in an integrated aquaculture (IA) system with S. clava, microalgae, and A. japonicus were higher than those observed in the traditional culture system. Metabolic and digestive enzymes were more active in small A. japonicus in the IA system than in those in the traditional aquaculture system. These results suggest that the IA model did not affect the growth of large A. japonicas, but produced a marked positive impact on the growth of small individuals.  相似文献

Summary Following UV irradiation of Bacillus subtilis there is a coordinate induction of: 1) a new protein, 2) a W-reactivation system, 3) a DNA modification system, and 4) prophages. These functions are induced following UV irradiation of repair proficient bacteria and mutants deficient in excision repair (uvr-1) and DNA polymerase I activity (polA5). However, they are not induced, or are impaired in their ability to be induced in bacteria containing the recA1 and the recG13 mutations. This inducible system is compared to the SOS system observed in E. coli.  相似文献

Summary We have cloned the seven genes that are responsible for biosynthesis of the antibiotic fortimicin A (FTM A) using a recently developed self-cloning system that employs the plasmid vector pMO116 for Micromonospora olivasterospora. Five chimeric plasmids that restored FTM A production in M. olivasterospora mutants blocked at different biosynthetic steps were isolated by shotgun cloning. Secondary transformation using other non-producing mutants showed that two additional FTM A biosynthetic genes were included on these plasmids, and that at least four of the genes were clustered. Interestingly AN38-1, a non-producing mutant that had a defect in dehydroxylation of a precursor of FTM A, was complemented by the DNA fragment containing a neomycin resistance gene that had been cloned from a neomycin-producing strain (Micromonospora sp. FTM A non-producing strain) in the course of constructing the plasmid vector pM0116. These results clearly show that this novel gene cloning system in Micromonospora is of practical use.  相似文献

Since the lacZα-based blue/white screening system was introduced to molecular biology, several different visual reporter systems were developed and used for various purposes in Escherichia coli. A common limit to the existent visual reporter systems is that an extracellular chromogenic substrate has to be added for the visible pigment production. In this study, we developed a new blue/white screening system based on a non-ribosomal peptide synthetase encoded by idgS from Streptomyces and a phosphopantetheinyl transferase encoded by sfp from Bacillus. When IdgS is activated from an apo-form to a holo-form via a posttranslational modification catalyzed by Sfp, it can synthesize a blue pigment indigoidine using L-glutamine, the amino acid abundant in cells, as a substrate. The new blue/white screening system contains a recipient E. coli strain with an optimized idgS gene cassette and a cloning vector harboring an sfp gene with an in-frame insertion of a multiple cloning site close to its N-terminal. We demonstrated that the IdgS/Sfp-based blue/white screening system is a powerful alternative to the lacZα-based screening system, which does not require any external substrate addition.
