Silkworm hemolymph is an important defense tissue to resist bacteria and virus infections. To study the response of silkworm hemolymph in the resistance of Bombyx mori L. nucleopolyhedrovirus (BmNPV), we constructed a near-isogenic silkworm line with BmNPV resistance using highly resistant and highly susceptible parental strains. In this paper, two-dimensional gel electrophoresis (2-DE) and Matrix-Assisted Laser Desorption/Ionization (MALDI)-mass spectrometry were employed to investigate the differences of protein patterns in the hemolymph of the highly resistant, highly susceptible and near-isogenic silkworm strains after BmNPV was administrated to the larvae. A comparison between the proteomes of these three silkworm strains led us to identify two differentially expressed proteins, beta-N-acetylglucosaminidase 2 and aminoacylase. The expression levels of these proteins were higher in the BmNPV resistant strains.  相似文献

家蚕中肠上皮是病毒经口侵入遇到的第一个组织。昆虫幼虫抵御杆状病毒的感染,可通过选择性的使感染的中肠上皮细胞发生调亡并在释放病毒粒子进入血淋巴之前使感染的细胞从中肠脱落。为研究家蚕抗核型多角体病毒(Bombyx mori nucleopolyhedrovirus, BmNPV)病的机制,通过对BmNPV高度抗性和高度敏感性的家蚕品系杂交和回交构建了近等基因系。本文对家蚕高抗,敏感及近等基因系5龄起蚕中肠组织的蛋白质表达谱进行了二维电泳 (two-dimensional gel electrophoresis,2-DE) 分析,并利用基质辅助激光解吸电离飞行时间 (matrix-assisted laser desorption/ionization-time of flight, MALDI-TOF) 质谱对差异蛋白进行鉴定。结果发现了5个差异表达的蛋白。推测这些蛋白可能与家蚕中肠对BmNPV的抗性或感性有关。  相似文献

We investigated variations in the gene expression of Bombyx mori following infection with a densonucleosis virus (BmDNV-Z). Two B. mori near-isogenic lines, Jingsong and Jingsong.nsd-Z.NIL, which are highly susceptible and completely resistant to BmDNV-Z, respectively, were used in this study. The infection profiles of BmDNV-Z in the midguts of the B. mori Jingsong and Jingsong.nsd-Z.NIL larvae revealed that the virus invaded the midguts of both of these strains. However, its proliferation was notably inhibited in the midgut of the resistant strain. By using the suppression subtractive hybridization method, three cDNA libraries were constructed to compare BmDNV-Z responsive gene expression between the two silkworm lines. In total, 151 differentially expressed genes were obtained. Real-time qPCR analysis confirmed that 11 genes were significantly up-regulated in the midgut of the Jingsong.nsd-Z.NIL strain following BmDNV-Z infection. Our results imply that these up-regulated genes might be involved in B. mori immune responses against BmDNV infection.  相似文献

【目的】家蚕核型多角体病毒(Bombyx mori nucleopolyhedrovirus,BmNPV)促使的血液型脓病是一种产业上非常严重的家蚕疾病,目前有效的防控方法较少。本研究以大造和CVDAR家蚕品系(对BmNPV有较强抗性的品系)为试验材料,通过分析CVDAR对BmNPV抗性特征,以期确定CVDAR对BmNPV的抗性机制。【方法】本研究通过半致死剂量分析,发现CVDAR品系比大造品系对BmNPV感染的半致死剂量提高10倍以上;进一步HE染色分析大造与CVDAR品系病毒感染前后的中肠组织的变化,具体解析抗性品系CVDAR的抗BmNPV机制。【结果】感染BmNPV 72 h后,大造中肠细胞细胞核明显膨大,着色变浅,到96h后,细胞核持续增大有脱落趋势;而CVDAR抗性品系只在感染96 h后有中肠部分细胞核膨大,但排列整齐;同时通过荧光定量分析大造与CVDAR品系病毒感染后的增殖情况,结合各个时期代表病毒基因的转录水平分析比较发现,感染BmNPV后0–12h也没有发现抗性品系CVDAR和大造之间的病毒拷贝数以及病毒基因转录水平的不同,但感染24h后发现抗性品系CVDAR无论是病毒拷贝数还是病毒基因的转录表达水平都明显低于对照大造。【结论】证明CVDAR口服添毒后中肠中病毒基因的转录在第一轮复制期间不受影响,之后转录水平降低。鉴定CVDAR品系抑制BmNPV增殖的关键时期是在感染BmNPV后的24 h,为解析抗性机制奠定基础。  相似文献

Baculovirus demonstrates specific infection spectrums and thus one certain host exhibits particular response to single baculovirus isolate. Autographa californica multiple nucleopolyhedrovirus (AcMNPV) is considered to be not an innate pathogen to Bombyx mori, but some silkworm strains have been identified to be permissive to AcMNPV, indicating the positive or negative involvement of certain host factors in baculovirus replications in vivo. To provide a fundamental knowledge of this process, we performed large-scale screening to investigate the responses of 448 silkworm strains against recombinant AcMNPV inoculation. By genetic analysis between permissive and resistant strains identified, we further confirmed that a potential corresponding locus on chromosome 3 regulates host responses to AcMNPV in silkworm. Additionally, we found that it is available for AcMNPV–silkworm baculovirus expression vector system to produce proteins of interest.  相似文献

家蚕浓核病毒 (Bombyx mori densonucleosis virus,BmDNV)是蚕业生产上危害比较严重的一类病毒。用完全抗浓核病中国镇江株(BmDNV-Z)的家蚕品系秋丰、感性品系华八及以华八为轮回亲本回交8代和自交8代构建的近等基因系BC8为材料,采用mRNA荧光差显技术首次分离克隆了家蚕羧酸酯酶(B. mori carboxylesterase,BmCarE)基因全长cDNA,并用实时荧光定量PCR检测了添毒后12 h、36 h、72 h BmCarE在感、抗BmDNV-Z家蚕品系中肠内的表达差异。结果表明: (1)添毒后12 h不同品系家蚕中肠BmCarE表达差异最大,抗性品系BC8和秋丰分别是感性品系华八的17.714倍和3.602倍,三者彼此间的差异达到极显著水平;(2)同一品系添毒后12 h与添清水后12 h BmCarE表达也有较大差异, BC8添毒是BC8添清水的15.08倍, 秋丰添毒是秋丰添清水的3.39倍, 差异达到极显著水平,而华八添毒和添清水的BmCarE表达量均低,二者差异不显著;(3)同一品系添毒后不同时间BmCarE表达也有较大差异, BC8和秋丰添毒后12 h BmCarE表达量最高,显著高于各自添毒后36 h和72 h表达水平,而添毒后36 h与72 h表达无显著差异;华八添毒后12 h、36 h和72 h,BmCarE表达无显著差异。上述结果提示羧酸酯酶基因可能与家蚕抗浓核病毒有一定关系。  相似文献

利用荧光差异显示技术分离的家蚕抗NPV相关基因s3a   总被引:6,自引:0,他引:6  
通过荧光差异显示技术,分析了家蚕Bombyx mori对BmNPV抗性品系NB、感性品系306和近等基因系306NNZZ添毒和未添毒处理区的基因表达的差异。根据差异显示的结果克隆了一条702 bp长度的cDNA片段,并用Northern blot进行了验证。该序列经过NCBI EST库的同源性比较获得了电子延伸。延伸后的序列用特异引物进行RT-PCR扩增获得了一条782 bp的序列,拼接后基因cDNA序列全长为827 bp,推导的氨基酸序列与草地夜蛾Spodoptera frugiperda S3A同源性最高达97.7%;其次是烟芽夜蛾Heliothis virescens S3A,同源性为94.0%;与黑腹果蝇Drosophila melanogaster S3A同源性为75.3%。比较结果显示这是一个新的家蚕基因,定名为家蚕s3a基因。本实验获得的s3a基因在家蚕感性和抗性品系以及添毒处理和未添毒处理中都具有差异表达,其中在抗性品系和近等基因系中的表达高于感性品系,在添毒处理中的表达高于未添毒组。因此推测它是一个与家蚕抗BmNPV相关的新基因。  相似文献

To gain new insights into the mechanism of soybean (Glycine max) resistance to the soybean cyst nematode (Heterodera glycines), we compared gene expression profiles of developing syncytia in soybean near-isogenic lines differing at Rhg1 (for resistance to Heterodera glycines), a major quantitative trait locus for resistance, by coupling laser capture microdissection with microarray analysis. Gene expression profiling revealed that 1,447 genes were differentially expressed between the two lines. Of these, 241 (16.8%) were stress- and defense-related genes. Several stress-related genes were up-regulated in the resistant line, including those encoding homologs of enzymes that lead to increased levels of reactive oxygen species and proteins associated with the unfolded protein response. These results indicate that syncytia induced in the resistant line are undergoing severe oxidative stress and imbalanced endoplasmic reticulum homeostasis, both of which likely contribute to the resistance reaction. Defense-related genes up-regulated within syncytia of the resistant line included those predominantly involved in apoptotic cell death, the plant hypersensitive response, and salicylic acid-mediated defense signaling; many of these genes were either partially suppressed or not induced to the same level by a virulent soybean cyst nematode population for successful nematode reproduction and development on the resistant line. Our study demonstrates that a network of molecular events take place during Rhg1-mediated resistance, leading to a highly complex defense response against a root pathogen.  相似文献

【目的】家蚕Bombyx mori核型多角体病毒(BmNPV)核衣壳蛋白VP39为病毒装配所必需。本研究旨在初探VP39在病毒侵染家蚕细胞过程中的功能及特征,以期为家蚕抗病毒研究提供研究基础。【方法】本研究通过构建原核表达载体,诱导原核表达得到多克隆抗体,以Western blot验证VP39表达时相;构建真核表达载体,转染细胞后以免疫荧光手段观测VP39表达定位及影响病毒增殖现象。【结果】制备了VP39多克隆抗体。VP39在病毒感染后大量定位于家蚕细胞核,部分定位于胞质,而过表达的VP39定位于家蚕细胞胞质;过表达VP39后抑制BmNPV感染家蚕细胞。【结论】在BmN-SWU1细胞中过表达VP39会影响BmNPV的扩散,导致BmNPV感染细胞数目大量减少。该结果为VP39调控宿主与病毒的相互作用提供了新的思路。  相似文献

Bombyx mori nucleopolyhedrovirus (BmNPV) is a DNA virus that causes huge losses to the silkworm industry but the piRNA responses during BmNPV infection in the silkworm remain uninvestigated. Here, silkworm piRNA profiles of uninfected and BmNPV-infected fat body and midgut were determined by high-through sequencing in the early stages of BmNPV infection. A total of 2675 and 3396 genome-derived piRNAs were identified from fat body and midgut, respectively. These genome-derived piRNAs mainly originated from unannotated instead of transposon regions in the silkworm genome. In total, 572 piRNAs were associated with 280 putative target genes in fat body and 805 piRNAs with 380 target genes in midgut. Compared to uninfected tissues, 322 and 129 piRNAs were significantly upregulated in BmNPV-infected fat body and midgut, respectively. In addition, 276 and 117 piRNAs were significantly downregulated. Moreover, differentially expressed (DE) piRNAs during BmNPV infection differed significantly between fat body and midgut. Putative DE piRNA–targeted genes were associated with “response to stimulus” and “environmental information processing” in fat body after infection with BmNPV, which may indicate an active piRNA response to BmNPV infection in fat body. This study may lay the foundation for future research of the potential roles of the piRNA pathway and specific piRNAs in BmNPV pathogenesis.  相似文献

To investigate the molecular mechanism of silkworm resistance to BmNPV infection, we constructed a near-isogenic line (BC8) with BmNPV resistance using highly resistant (NB) and highly susceptible parental strains (306). We investigated variations in the gene expression in the midguts of BmNPV-infected BC8 and 306 at 12 h pi using the microarray. 92 differentially expressed genes were identified. Real-time qPCR analysis confirmed that 10 genes were significantly up-regulated or down-regulated in the midguts of BC8 and NB compared to 306. To our knowledge, we first defined the role of the amino acid transporter and 26S proteasome in insect antiviral. However, serine protease was not completely consistent with data of reported previously in insect antiviral. The role of the 5 genes (Bm123, Bm122, COP β′, aquaporin, glycoside hydrolases) was also demonstrated in insect antiviral. Our results provided new insights into the molecular mechanism of the Bombyx mori immune response against BmNPV infection.  相似文献

Bombyx mori nucleopolyhedrovirus (BmNPV) that infects the silkworm, B. mori, accounts for >50% of silk cocoon crop losses globally. We speculated that simultaneous targeting of several BmNPV essential genes in transgenic silkworm would elicit a stable defense against the virus. We introduced into the silkworm germline the vectors carrying short sequences of four essential BmNPV genes in tandem, either in sense or antisense or in inverted-repeat arrangement. The transgenic silkworms carrying the inverted repeat-containing transgene showed stable protection against high doses of baculovirus infection. Further, the antiviral trait was incorporated to a commercially productive silkworm strain highly susceptible to BmNPV. This led to combining the high-yielding cocoon and silk traits of the parental commercial strain and a very high level of refractoriness (>75% survival rate as compared to <15% in nontransgenic lines) to baculovirus infection conferred by the transgene. We also observed impaired infectivity of the occlusion bodies derived from the transgenic lines as compared to the wild-type ones. Currently, large-scale exploitation of these transgenic lines is underway to bring about economic transformation of sericulture.  相似文献

The recombinant protein expression by Bombyx mori nucleopolyhedrovirus (BmNPV) infecting silkworm larvae or pupae may endow us with a potent system for the production of large eukaryotic proteins. However, the screening of silkworm strains ideally suited to this method has scarcely been conducted. In the present study, we injected recombinant BmNPV containing a reporter gene, luciferase or DsRed, into hemocoel of fifth instar larvae of selected 12 silkworm strains. Among them, the strain d17 is found to be the highest in reporter expression from the intrinsic polyhedrin promoter of Autographa californica NPV or the silkworm actin A3 promoter. These results suggest that the d17 strain is highly permissive for BmNPV replication and is the most likely candidate of a “factory” for large-scale expression using the BmNPV bacmid system.  相似文献

The Bombyx mandarina nucleopolyhedrovirus (BomaNPV) S1 strain can infect the silkworm, Bombyx mori, but is significantly less virulent than B. mori nucleopolyhedrovirus (BmNPV) T3 strain. The complete nucleotide sequence of the S1 strain of BomaNPV was determined and compared with the BmNPV T3 strain. The circular, double stranded DNA genome of the S1 strain was 126,770 nucleotides long (GenBank accession no. FJ882854), with a G+C content of 40.23%. The genome contained 133 potential ORFs. Most of the putative proteins were more than 96% identical to homologs in the BmNPV T3 strain, except for bro-a, lef-12, bro-c, and bro-d. Compared with the BmNPV T3 strain, however, this genome did not encode the bro-b and bro-e genes. In addition, hr1 lacked two repeat units, while hr2L, hr2R, hr3, hr4L, hr4R, and hr5 were similar to the corresponding hrs in the T3 strain. The sequence strongly suggested that BomaNPV and BmNPV are variants with each other, and supported the idea that baculovirus strain heterogeneity may often be caused by variation in the hrs and bro genes.  相似文献
