获得大量、高质量的全基因组DNA是在DNA水平上研究许多生物的分子机制的基本前提。微小昆虫由于其体型微小,单头提取DNA浓度低,无法满足部分试验所需。为寻找一种方便、快捷、高效的全基因组提取方法,参考国内外单头微小昆虫基因组DNA提取常用方法,以烟粉虱为研究材料,选取醋酸钾(KAc)法、盐析法、氯仿-异戊醇法和苯酚提取法四种方法进行多头烟粉虱的全基因组DNA提取。通过微量核酸蛋白分析仪、基因组DNA直接琼脂糖凝胶电泳、甲基化敏感扩增多态性(Methylation sensitive amplification polymorphism,MSAP)引物扩增产物的琼脂糖凝胶电泳对DNA样品进行检测,比较提取DNA的产量、质量,并综合分析各提取方法所需时间及所用试剂的毒性大小。结果表明,盐析法提取的DNA平均浓度为521 ng/μL,纯度能满足分子检测要求,用MSAP引物能得到较好的扩增结果,相比其它方法,盐析法更简便、快速且无毒。因此盐析法是多头烟粉虱全基因组DNA提取的最佳方法。  相似文献

参考国内外昆虫基因组DNA提取的常用方法,选取KAc法、氯仿-异戊醇法和盐析法3种方法对苹果绵蚜(Eriosoma lanigerum)基因组DNA进行提取.通过基因组DNA直接琼脂糖凝胶电泳、SSR引物扩增产物的琼脂糖凝胶电泳和聚丙烯酰胺凝胶电泳检测,对3种方法所提基因组DNA的质量进行了比较,并综合分析了提取所需时间及所用试剂的毒性大小等.结果表明,盐析法操作程序较简便快捷,节时省工,可得较好质量的DNA样本,且对试验操作人员无伤害,是一种值得推广应用的基因组DNA提取方法.  相似文献

目的:外周血DNA的提取是研究乙型肝炎病毒相关临床疾病的基础,所提取DNA的质与量直接关乎下游研究的成败,经济、高效、便捷的外周血DNA提取方法对于疾病分子水平的研究尤为重要,本实验旨在比较两种外周血DNA提取方法,从而为临床研究提供有力的参考。方法:以外周抗凝血为试验样本,分别采用改良盐析法和DNA提取试剂盒法(硅胶柱纯化)进行基因组DNA的提取,通过分光光度仪测量DNA浓度和纯度,并进行PCR扩增及电泳实验。比较改良盐析法与试剂盒提取法(硅胶柱纯化)的效果。结果:试剂盒提取法(硅胶柱纯化)标本用量甚微,省时,提取DNA纯度高,步骤繁琐,PCR条带单一、亮度差;改良盐析法操作步骤少,提取DNA浓度高,PCR条带亮度佳、杂带多,耗时长。结论:两组方法各有优缺点,试剂盒提取法(硅胶柱纯化)可靠、快速,但所获DNA量少、极易降解,改良盐析法耗时,但所获DNA浓度高、量多,可根据实验时间与经费,实验所需的DNA纯度与浓度,提供的样本体积等不同的临床研究需求及条件来综合选择适宜的提取方法。  相似文献

分别采用醋酸钾(KAc)法、十二烷基硫酸钠-蛋白酶K(SDS-PK)消化法、十六烷基三乙基溴化铵(CTAB)法及DNA提取试剂盒(吸附柱型)对单只松突圆蚧的基因组DNA进行提取。同时针对盾蚧科昆虫的特点,在提取前利用氯仿和解剖针去除样品表面的介壳。结果表明,用同种提取方法提取的经氯仿处理与未经处理样本的提取效果无明显差异,而采用解剖针去除介壳的样本提取效果较好。所采用的4种提取方法均能从新鲜标本中提取出DNA,其中CTAB法提取的DNA量较多,而SDS-PK法提取的DNA质量较好。  相似文献

【目的】股带针蓟马,又称温室条带蓟马,是一种多食性农业害虫,寄主超过50种,在一些国家被列为检疫性生物。2020年9月,在北京发现其成虫和幼虫危害海芋叶片,本文介绍了其形态特征、分子鉴定数据、分布地区及寄主植物等数据,以期为其准确鉴定和防治提供参考。【方法】采用拍打法收集标本;采用非破坏性方法提取单头蓟马DNA,扩增线粒体细胞色素c氧化酶Ⅰ基因(COⅠ)的部分序列,进行分子鉴定和系统发育分析;制作永久玻片进行形态鉴定。【结果】结合形态和分子数据鉴定其为股带针蓟马,是中国新记录属、种,海芋是该种新记录的寄主植物。股带针蓟马与该属其他种的主要区别在于该种前翅的亚中部浅色带比端部暗带短。COⅠ分子序列分析表明,北京种群与国外种群聚集成一个分支,序列相似性高达100%。【结论】应采取进一步调查、监测和管理措施,防止该种外来有害蓟马的传播。  相似文献

介绍一种可从单头麦红吸浆虫Sitodiplosis mosellana(Gehin)成虫、幼虫和蛹成功地提取基因组DNA的方法。经检测提取DNA样品浓度为100~200ng/μL,纯度在1.4~1.6范围之间,以此为模板,可顺利地进行RAPD引物扩增。该提取方法简单、安全、经济,可为小型昆虫基因组DNA的提取提供参考。  相似文献

目的:比较改良酚一氯仿抽提法、盐析法、试剂盒法从人全血中提取基因组DNA的效果,以期建立一种快速、经济的提取高质量基因组DNA的方法。方法:分别用上述三种方法从人全血中提取基因组DNA,通过紫外分光光度计、琼脂糖凝胶电泳、聚合酶链式反应(PCR)、限制性内切酶酶切检测提取的基因组DNA的产量、纯度和质量。结果:改良酚一氯仿抽提法与试剂盒法提取的基因组DNA相比,DNA的产量有统计学差异,DNA的纯度无统计学差异,但试剂盒法提取的基因组DNA有较明显的降解现象:盐析法与改良酚.氯仿抽提法、试剂盒法相比,基因组DNA的产量和纯度都存在统计学差异,并且基因组DNA聚合酶链式反应(PCR)扩增的稳定性也明显劣于另外两种方法;三种方法提取基因组DNA均能进行限制性内切核酸消化。结论:改良酚一氯仿抽据取法是一种经济、快速、高效、稳定提取人全血基因组DNA的方法,适用于批量临床标本处理。  相似文献

实验以日本龟蜡蚧CeroplastesjaponicusGreen,白蜡绵粉蚧PhenacoccusfraxinusTang ,朝鲜球蚧DidesmococcuskoreanusBorchseniush和瘤大球坚蚧EulecaniumgiganteaShinji等 4种蚧虫为材料 ,分别用十二烷基硫酸钠 (SDS)法、十六烷基三乙基溴化铵 (CTAB)法、醋酸钾 (KAc)法和氯化钠 (NaCl)法等 4种方法 ,对单只蚧虫进行基因组DNA提取 ,用 0 8%琼脂糖凝胶电泳检测所提DNA。结果表明 ,4种方法都可以提取到基因组DNA ,但是比较而言 ,CTAB法和NaCl法所提取的DNA质量明显优于SDS法和KAc法 ,并适用于PCR。因此认为 ,CTAB法和NaCl法是实验室提取单只蚧虫基因组DNA更有效而实用的方法。  相似文献

Associations between insects and gut bacteria are ubiquitous. It is possible to make a distinction between permanent associations (called symbiosis), in which the same type of bacteria is present in more than one generation of the insect, and transient associations. Transient bacteria are ingested together with food but do not settle in the insect gut in such a way that they will be passed on to the next generation. In this study, we describe the permanent association between Western flower thrips (Frankliniella occidentalis), a polyphagous insect species that is a major pest worldwide, and one type of gut bacteria. On the basis of direct microscopic observations and counts of bacteria, it was found that thrips from the populations studied contained large numbers of bacteria in their hindgut. Bacteria were isolated from their host and grown on 10 different agar media. The number of bacteria isolated on agar media equaled the number of direct counts. All isolates had the same colony morphology. On the basis of their 16S rDNA sequence these bacteria were identified as Enterobacteriaceae, closely related to Escherichia coli. Isolates tested with API 20E biochemical tests were Erwinia species. This was the only type of bacteria found in all thrips individuals on any of the 10 different agar media. Universal primers, which would potentially pick up DNA from any bacterium present in the insect, were applied on crude DNA extracts from thrips with bacteria. We only found 16S rDNA sequences similar to those of the isolated thrips gut bacteria. The same type of bacteria was present in all life stages of the thrips and was found to persist in the thrips populations for at least 2 years (more than 50 generations).  相似文献

2. A correlation is demonstrated between type of host-plant of thrips and the relative numbers caught by traps of different colours.

The literature is reviewed and some general relationships with the effectiveness of different trap colours are hypothesized for: non-grass foliage insects and their predators and parasites; grass foliage insects; flower-dwelling insects; predators and parasites not associated with foliage; biting insects; and wood-borers.

In this study, we investigated the relationships among insect orders with a main focus on Polyneoptera (lower Neoptera: roaches, mantids, earwigs, grasshoppers, etc.), and Paraneoptera (thrips, lice, bugs in the wide sense). The relationships between and within these groups of insects are difficult to resolve because only few informative molecular and morphological characters are available. Here, we provide the first phylogenomic expressed sequence tags data ('EST': short sub-sequences from a c(opy) DNA sequence encoding for proteins) for stick insects (Phasmatodea) and webspinners (Embioptera) to complete published EST data. As recent EST datasets are characterized by a heterogeneous distribution of available genes across taxa, we use different rationales to optimize the data matrix composition. Our results suggest a monophyletic origin of Polyneoptera and Eumetabola (Paraneoptera + Holometabola). However, we identified artefacts of tree reconstruction (human louse Pediculus humanus assigned to Odonata (damselflies and dragonflies) or Holometabola (insects with a complete metamorphosis); mayfly genus Baetis nested within Neoptera), which were most probably rooted in a data matrix composition bias due to the inclusion of sequence data of entire proteomes. Until entire proteomes are available for each species in phylogenomic analyses, this potential pitfall should be carefully considered.  相似文献

We have developed a novel strategy for the introduction of durable insect resistance in crops. This strategy was based on intervention in the natural relationship between plants and insects. For many insects, including pests such as thrips (Frankliniella occidentalis), the flower is an important factor in their life cycle, serving either as a food source or as a place for mating. The insects are attracted to the flower by scent, which is mainly produced by the petals, and by the bright colour of these floral organs. We therefore anticipated that removal or changing the identity of the petals would significantly reduce the attractiveness of the flower to thrips. To test this hypothesis, we used cucumber as a model species because most modern varieties are parthenocarpic, in which the fruit develops without fertilization. The cucumber mutant green petals, in which the petals are homeotically transformed into green sepals, was particularly useful for this study. The susceptibility of the cucumber plants to damage by thrips was determined by recording thrip numbers and by measuring leaf damage. Large differences were observed when greenhouse compartments with either wild-type or green petal mutant plants were compared. The rate of population growth of the insects on the mutant plants was significantly reduced and the leaves were almost undamaged. These results demonstrate that alterations in the structure of flowers may interfere with the life cycle of insects, providing the means for a novel and natural strategy for obtaining insect resistance.  相似文献

While the distribution of herbivorous insects over leaves along the stem often shows a peak at some distance from the apex this does not necessarily reflect an innate preference as alternative explanations can be provided such as impact of predators and inter- or intraspecific competitors. It is of interest to determine which factors shape the distribution of insects over the leaves of a plant. Do leaves from different positions differ in suitability for insects and is that reflected in the insect's preference, or are other factors involved? In this paper we assess how the herbivorous insect western flower thrips, Frankliniella occidentalis (Pergande) (Thysanoptera: Thripidae), chooses among leaves from different positions relative to the apex of cucumber, Cucumis sativus (L.) plants. On leaf discs of a susceptible and three partially resistant cucumber accessions, thrips reproduction was highest on apical leaves and lowest on basal leaves. In dual-choice essays thrips females preferred younger leaves over older leaves for oviposition in all cucumber accessions tested, as was predicted from the no-choice assay. This indicates that differences in leaf suitability are an important factor in determining thrips distribution on cucumber plants.  相似文献

Entomophthora thripidum is an obligate biotrophic insect pathogenic fungus that grows as protoplasts within the hemocoel of thrips. Prior to penetration through the insect cuticle and spore formation at the insect surface the protoplasts switch to hyphal growth. In vitro, the differentiation to hyphal growth was a prerequisite for the subsequent formation of infectious spores and was detected 10-20 days after inoculation. E. thripidum secreted a factor that autoinduced the differentiation to hyphal growth. The discovery of this activity inducing hyphal growth made possible the reliable production of spores, the infection of host insects and the consecutive re-isolation of the fungus from the infected insects.  相似文献

RNA interference (RNAi) methods for insects are often limited by problems with double-stranded (ds) RNA delivery, which restricts reverse genetics studies and the development of RNAi-based biocides. We therefore delegated to insect symbiotic bacteria the task of: (i) constitutive dsRNA synthesis and (ii) trauma-free delivery. RNaseIII-deficient, dsRNA-expressing bacterial strains were created from the symbionts of two very diverse pest species: a long-lived blood-sucking bug, Rhodnius prolixus, and a short-lived globally invasive polyphagous agricultural pest, western flower thrips (Frankliniella occidentalis). When ingested, the manipulated bacteria colonized the insects, successfully competed with the wild-type microflora, and sustainably mediated systemic knockdown phenotypes that were horizontally transmissible. This represents a significant advance in the ability to deliver RNAi, potentially to a large range of non-model insects.  相似文献

Scanning electron microscopy was used to elucidate the morphology and sequential movement of thrips feeding structures in the 2 species, Frankliniella occidentalis and F. schultzei, (Thysanoptera : Thripidae). The mouthcone consists of paired paraglossae, and fringed labral pad, a single, apically fused mandible and a pair of interlocking maxillae. The maxillae are open apically and form a feeding tube. Ten pairs of sensory pegs of 3 distinct morphological types (sensilla basiconica with a cuticular collar, sensilla basiconica without a cuticular collar, and sensilla trichoidea) were found on the paraglossae. The possible function of these sensory structures in host finding and choice are discussed. No structure for rasping the leaf's surface was found on the mouthcone or the ventral surface of the insect. Live specimens were observed feeding through Parafilm on artificial media (sucrose solution) and lettuce leaf tissue. These observations support earlier findings that thrips feed by piercing leaf cells with the mandible and ingesting cell contents through the feeding tube formed by the maxillary stylets. Based on these findings, we suggest that thrips be classified as piercing-sucking rather than rasping-sucking insects.  相似文献

Modeling the effect of temperature on the sustainability of insect-plant interactions requires assessment of both insect and plant performance. We examined the effect of temperature on western flower thrips, Frankliniella occidentalis (Pergande), a generalist herbivore with a high reproductive rate, and chrysanthemum inflorescences, a high quality but relatively fixed, ephemeral resource for thrips population growth. We hypothesized that different thrips versus plant responses to temperature result in significant statistical interaction of temperature with thrips abundance and flower damage attributes over time. Experiments were conducted at five temperatures between 20.7 and 35.3 degrees C, with thrips infestation and time after infestation as main effects. Only minor, uncontrolled variations in relative humidity and light intensity may otherwise have influenced the results. High temperatures lead to an initially rapid increase in density of thrips followed by abrupt declines in abundance. The rate of floral senescence increased with temperature and thrips infestation, as indicated by a reduced fresh biomass and greater leaching of yellow pigments. Multiple regression indicated that indices of plant damage responded more directly to thrips density at low than high temperature, supporting the conclusion that temperature affected the outcome beyond what was predictable simply from differential plant and insect optima. The relative intensity of damage caused by individual thrips decreased with increasing temperature, likely caused by thrips competition and reduced survival, growth, and fecundity on depleted inflorescences. Reduced per capita damage at high temperature may be common in insects exploiting fixed plant resources that exhibit an accelerated rate of deterioration at high temperatures.  相似文献
