Superficial (0 to 2 cm) sediments were sampled from 62 sites in Kattegat and Skagerrak during autumn 1989 and spring 1990, tested for toxicity to Daphnia magna and Nitocra spinipes (Crustacea) and analyzed for heavy metals (Cd, Cr, Cu, Hg, N, Pb, Zn), nutrients (N and P) and organic carbon. Whole sediment toxicity to Nitocra spinipes, expressed as 96-h LC50, ranged from 1.8 to > > 32 percent sediment (wet wt), which is equivalent to 0.63 to 53 percent dry wt. Sediment total metal concentrations (mg kg-1 dry wt) ranged from 0.01 to 0.32 for Cd, 8 to 57 for Cr, 3 to 40 for Cu, 0.03 to 0.86 for Hg, 3 to 43 for Ni, 6 to 37 for Pb and 21 to 156 for Zn. Analyzed concentrations of heavy metals were tested for correlation with whole sediment toxicity normalized to dry wt, and significant correlations (Spearman p<0.05) were found for Cd, Cr, Cu, Hg, and Ni. However, the analyzed concentrations of these metals were below the spiked sediment toxicity of these heavy metals to N. spinipes, except for Cr and Zn for which analyzed maximum concentrations approached the 96-h spiked sediment LC50s. There was no improvement in correlation between the sum of heavy metal concentrations normalized to their spiked toxic concentrations (Toxic Unit approach) and the whole sediment toxicity. Calculated heavy-metal-derived toxicity based on toxic units and whole sediment toxicity ranged from 0.1 to 24 (mean value 2.3 and SD 4.2). Theoretically, a value of 1.0 would explain whole sediment toxicity from measured metal concentrations using this approach. Thus, in spite of the fact that the total concentrations of the heavy metals were sufficient to cause toxicity based on an additive model for most of these sediments, the observed toxicity of the sediments from Kattegat and Skagerrak could not exclusively be explained by the concentrations of heavy metals, except for Cr and Zn at their maximum concentrations. Zinc, lead and mercury accumulation in the amphipod Hyalella azteca increases with increasing exposure to metals. During 10 week chronic toxicity tests, metal accumulated at the highest non-toxic/lowest toxic concentration was 126/136 µg Zn g–1, 7.1/16 µg Pb g–1 and 56/90 µg Hg g–1 dry weight. Concentrations of lead and mercyry in control animals were substantially lower (1.3 µg Pb g–1 and 0.4 µg Hg g–1), but concentrations of zinc in controls (74 µg g–1) were about one half those of the lowest toxic concentration. Copper was completely regulated. Accumulated copper concentrations after 10 weeks exposure to all waterborne copper concentrations resulting in less than 100% mortality were not significantly different from controls (79 µg g–1). Lead and mercury concentrations in wild H. azteca should be useful indicators of potential toxicity. Zinc accumulation may also be a useful indicator of zinc toxicity, but careful comparison with control or reference animals is necessary because of the small differences between toxic and control concentrations. Copper is not accumulated by H. azteca under chronic exposure conditions and body burdens of field animals cannot be used as an indicator of exposure or potential toxic effects. Short term exposures to copper, however, result in elevated copper concentrations in H. azteca, even at concentrations below those causing chronic toxicity. Short term bioaccumulation studies might, therefore, provide a useful indication of potential chronic copper toxicity.  相似文献

Specimens of the prosobranch Littorina littorea (L., 1758) collected along the East Frisian North Sea coast in summer 1993 exhibited alterations of the pallial oviduct termed as intersex in response to tributyltin (TBT) pollution. The range of TBT body burden was between 150.9 and 1289.5 μg as Sn kg−1 (dry wt.). Five stages of intersex development (0–4) could be distinguished and are documented with scanning electron micrographs. In stages 2–4, which can be found in the direct vicinity of harbours and marinas, the morphological malformations of the oviduct inhibit successful copulation and capsule formation, resulting in sterilization. The intersex index (ISI, calculated as the average intersex stage of a population) and the average prostate length of females were used as parameters for the determination of intersex intensities in the populations. Both indices show significant and positive correlations to the TBT body burden of L. littorea and are promising parameters for TBT biomonitoring. A comparison of TBT bioconcentration factors with populations from England and France indicates that the threshold concentration for intersex development is in the range of 15 ng TBT as Sn/l. Morphometric analyses of the midgut gland revealed no significant differences between sampling stations. In the ovary a retardation and blockage of maturation (atresia) was observed in populations close to harbours. Lytic processes in ovary follicles were observed not only at TBT exposed sites but also at reference stations.  相似文献

A new standardized bioassay procedure for testing the chronic toxicity of sediments to Hyalella azteca was developed. Tests were initiated with 0–1 wk old amphipods exposed to sediments from Hamilton Harbour, Toronto Harbour, and Lake Ontario for 4 to 8 weeks. Both survival and growth were significantly reduced in the Hamilton Harbour sediments relative to those from the lake after 4 weeks. Exposures of 8 weeks resulted in greater variability; survival of amphipods in sediments from one of the harbour stations, and growth in sediments from both harbour stations with surviving young were not statistically different from survival and growth in lake sediments. Growth and survival in lake sediments were comparable to cultures grown with cotton gauze and no sediment after 4 weeks, but survival was poorer by week 8. Replication was good in 12 out of 13 tests done in duplicate; the difference in survival between replicates averaged 2.2 animals (20 amphipods/replicate, 4 week exposure). We propose that 4 week exposures of young (0–1 wk old) Hyalella would provide a suitable standardized chronic toxicity test for sediments. Sediments were sampled from 62 sites in the Kattegat and Skagerrak, which are located between the Baltic and the North Sea in the Western Atlantic, during autumn 1989 and spring 1990. From each site 5 to 6 samples were taken wit ha box-corer. After mixing to composite samples on board, transport and storage (at 4 °C for 2 to 4 weeks), the samples were tested for toxicity to Daphnia magna and Nitocra spinipes. Immobility in Daphnia after exposure to 16 percent sediment (wet wt) in reconstituted standardized water (ISO, 1982) ranged from 0 to 88 percent after 24 h and from 3 to 95 percent after 48 h. For Nitocra the toxicity, determined as the 96-h LC50 (% wet wt) at 7 salinity, ranged from > > 32 percent (nontoxic) to 1.8 percent (most toxic). All exposures were made in duplicates and the effects obtained in the duplicates with the same sediment were correlated to each other. However, sediment toxicity to Daphnia and Nitocra was not. The test with Nitocra, which was made at several concentrations of sediment, was considered to give the most reliable picture of sediment toxicity in the Kattegat and Skagerrak. This ambient toxicity assessment identified three areas with toxic sediment, (1) the Göta älv estuary (outside the city of Göteborg) and its surroundings, (2) the Bay of Laholm in southern Kattegat, which is an area with periodic oxygen depletion and where repeated mussel kills have occurred during the last decade, and (3) an area in the open Skagerrak northwest of Skagen (the tip of the Jutland peninsula). Sediments, which had been stored at 4 °C, were tested again after 6 to 13 mos with the Nitocra test. Stored sediment toxicity was poorly correlated with fresh sediment toxicity. The average detoxification during storage was 5 times, but the range was 3 orders of magnitude, from 17 times more toxic to 73 times less toxic. The reasons for the observed areal and storage differences in sediment toxicity are so far not understood.  相似文献

To study environmental risk assessment, echinoderms provide a useful model for ecotoxicological testing. However, limited knowledge of the life history of field collected heart urchins is a problem and the use of cultured urchins has been investigated here. The present study describes a culture method for the heart urchin Echinocardium cordatum under controlled laboratory conditions, providing organisms with a low biological variation. Based on our optimized growth protocol both larvae and juveniles have a growth rate comparable to E. cordatum in the wild. The toxicological response of cultured and field-collected E. cordatum was compared in standard saltwater toxicity bioassays. Using ammonium chloride as a water-soluble reference toxicant, mean 96 h LC50 values for cultured heart urchins versus field collected animals were 37.4 ± 7.6 mg NH4+/l (n = 5) versus 22.5 ± 4.9 mg NH4+/l (n = 19), respectively. Additional toxicity experiments with tributyl tin (TBT) spiked sediments revealed 14d LC50 values of 1,242 (95% confidence interval 986–1,564) and 964 (95% confidence interval 843–1,102) µg Sn/kg dw respectively in cultured and field collected E. cordatum. From this it was concluded that cultured heart urchins are less sensitive to TBT than field collected E. cordatum. Furthermore in whole sediment toxicity tests, survival of cultured sea urchins was higher or at least similar to that of field collected E. cordatum. The increased sensitivity of field urchins compared to cultured urchins in various toxicity tests may be due to multiple environmental stressors reducing their overall performance. Overall it was demonstrated that the use of cultured E. cordatum provides a significant advance for urchin-based bioassays for marine environmental toxicity testing, resulting in a more homogeneous, vital population with experimental data displaying reduced variability.  相似文献

Food concentration supplied during subchronic and chronic toxicity tests, is one of the most important factors that might influence the response of test organisms to toxicants. The green microalga Scenedesmus incrassatulus was used as food for the cladoceran Daphnia magna, in a chronic toxicity test with the toxicant sodium dodecyl sulphate (SDS). Test concentrations were 0.625, 1.25, and 2.5 mg l-1 of SDS, equivalent to 1/40, 1/20, and 1/10 of the average 48-h LC50 previously determined. Food concentrations were 9.5, 19, and 38 mg l-1 (dry wt.). Survival and reproduction were recorded, and the data were analyzed using a life-table approach. After 55 days, the main findings were as follows:
1.  Average clutch size decreased as food concentration increased and there was a negative interaction between the toxicant and the algal concentration.
2.  The Net Reproductive Rate (R 0) of the controls fed the lowest food concentration was approximately 10 times higher than the controls fed the highest food concentration.
3.  R 0's for the treatments with SDS fed the lowest food concentration, were 2.5 to 5 times higher than those fed the highest food concentration.
4.  Reproduction in controls and treatments with the same SDS concentrations, became similar as food level increased.
According to these results, the food concentration to be supplied in chronic toxicity tests should be accurate, as an incorrect food concentration might negatively affect the survival and reproduction of test organisms, thus masking or confounding the effects of the toxicant being examined. A concentration around 10 mg l-1 (dry wt.) of S. incrassatulus seems to be appropriate for D. magna economic toxicity tests.  相似文献

The purpose of the presented paper was to study the effect of high concentrations of tributyltin (TBT) on the potassium retention and fatty acid (FA) composition of the fungus Cunninghamella elegans recognized as a very efficient TBT degrader. An increase in TBT had a strong influence on the potassium concentration in the fungus. In growth medium without TBT, the potassium content of the fungal cells was 5.8 mg K+ g dry weight−1. The maximum concentration of K+ was 15.06 mg g−1 dry weight at 30 mg l−1 of TBT. The major FAs that characterized the tested strain were C16:0, C18:1, C18:2, C18:3 and C18:0. TBT in the concentration range 5–30 mg l−1 strongly influenced the FA composition. In the presence of the organotin, the degree of saturation increased. It suggests that the observed changes promote an increase in the lipid ordering of the membrane by reducing its permeability and inhibiting potassium ion efflux.  相似文献

The effects of tributyltin (TBT) compounds on gill morphology were examined in the mummichog, Fundulus heteroclitus , in 96-h LC50 and 6-week sublethal exposures. Morphometry was used for the identification and quantification of effects with the light microscope. A 96-h LC50 of 17.2 μg 1 1 was determined. Morphometric analysis of gill tissues revealed hypertrophy of the lamellar epithelium in fish exposed to 17.2 μg 1–1. Relative diffusing capacity was significantly decreased (−41 %); ( P <0.05, ANOVA, Bonferroni t -test). At 35.6 μg 1−1, TBT exposure resulted in a significant reduction (− 40%) in the volume of the lamellar blood channels. Both of these observations occurred in fish that showed signs of acute poisoning including loss of equilibrium. In fish exposed to sublethal concentrations of 0.105–2.000 μg TBT 1−1 for 6 weeks, there were no pathological changes in the gill. There were no treatment-related changes in the surface morphology of the gills of fish from both experiments upon scanning electron microscopic examination. Although gill pathology was observed in acutely toxic exposures, it does not appear to be a major mechanism of TBT toxicity.  相似文献

The objective of this article is to summarize and map bifenthrin sediment measurements from California waterbodies from 2001 to 2010 to show sites where bifenthrin has been measured in sediment (including both detected and non-detected concentrations) and where corresponding toxicity or its lack has been reported or predicted. Bifenthrin measurements were available from depositional areas at 359 sites with concurrent total organic carbon (TOC) measurements in California waterbodies and values from approximately 37% of these sites were below the level of detection. Sediment toxicity data based on Hyalella azteca single species ambient toxicity tests were available for 268 sites with concurrent bifenthrin measurements and 62% of these sites showed no significant toxicity. Sixteen percent of the 140 California sites with non-detected bifenthrin concentrations had some significant sediment toxicity thus suggesting that toxicity at these sites is due to factors other than bifenthrin. One percent TOC normalized bifenthrin measurements reported from 268 sites showed no predicted significant toxicity at 83% of the sites based on a comparison with a Hyalella azteca acute value of 6.1 ng/g while 99.3% of the sites showed no predicted significant toxicity based on a Chironomus tentans acute toxicity value of 177.5 ng/g. The test species toxicity data (i.e., Hyalella or Chironomus) used to predict bifenthrin toxicity in the field is therefore critical as the use of Hyalella toxicity data (a highly sensitive species to bifenthrin) may be overprotective and this species is less representative of most California waterbodies when compared with Chironomids.  相似文献

The freshwater amphipod Gammarus pulex was sampled from several sites in Northumberland, England and analysed for total body zinc and cadmium concentrations.The toxicity of cadmium to animals from the different sites was determined and a positive relationship was found between the initial total body zinc concentration and the ability to survive in an artificial medium containing 0.5 mg 1–1 cadmium as cadmium sulphate.Exposure in the laboratory to a low concentration of zinc (0.01 mg 1–1) as zinc sulphate for two weeks resulted in a decreased susceptibility to cadmium poisoning.  相似文献

Tributyltin (TBT) contamination remains a major problem worldwide. Many laboratories are committed to the development of remediation methodologies that could help reduce the negative impact of this compound in the environment. Furthermore, it is important to have at hand simple methodologies for evaluating TBT toxicity in the laboratory, besides the use of complex and costly analytical instrumentation. With that purpose, a method was adapted that is based on the inhibition of growth of an indicator strain, Micrococcus luteus ATCC 9341, under TBT. Different types of matrices, of TBT concentrations and sample treatments were tested. The results herein reported show that the bioassay method can be applied for both aqueous and soil samples and also for a high range of TBT concentrations (at least up to 500 μmol/L). Besides being cheap and easy to perform, it can be performed in any laboratory. Additionally, one possible application of the method to monitor TBT degradation is presented as an example.  相似文献

Summary Acute toxicity tests using a static-with-replacement testing procedure were carried out with the four larval instars of the freshwater detritivore Chironomus riparius (Meigen). Median lethal concentrations (10, 24, 48 and 96 h LC50's) indicated great differences in sensitivity to cadmium between instars. Larvae became more tolerant with increasing age, the most resistant stage (fourth instar) having a 24 h LC50 of 2,400 mg Cd l-1, approximately 950 times greater than the corresponding value of 2.1 mg Cd l-1 recorded for the most sensitive (first instar) stage. These observations are discussed in relation to the establishment of water quality standards.  相似文献

The effect of Si(OH)4 on Cr toxicity and elemental concentrations in ryegrass were investigated in a growth chamber using an acid and a neutral mineral soil. Each soil was treated with 50 mg Cr, as CrO3, kg−1 soil dry weight, singly, or in combination with 25 mg Si as Si(OH)4. Plants growing in unamended soils were used as controls. Chromium toxicity, expressed as decrease in shoot or root dry weight, was increased by the Si. This increase was accompanied by a higher Cr uptake particularly on the acid soil. The shoot and root dry weights were significantly correlated (P=1%) with the concentration of Cr, where r=−0.80 and −0.65, respectively. Uptake of Al, Cu, Fe, P and Zn did not show any consistent relationship to the magnitude of Cr toxicity.  相似文献

The toxicity and binding of aluminium to Escherichia coli has been studied. Inhibition of growth by aluminium nitrate was markedly dependent on pH; growth in medium buffered to pH 5.4 was more sensitive to 0.9 mM or 2.25 mM aluminium than was growth at pH 6.6–6.8. In medium buffered with 2-(N-morpholino)ethanesulphonic acid (MES), aluminium toxicity was enhanced by omission of iron from the medium or by use of exponential phase starter cultures. Analysis of bound aluminium by atomic absorption spectroscopy showed that aluminium was bound intracellularly at one type of site with a K m of 0.4 mM and a capacity of 0.13 mol (g dry wt)-1. In contrast, binding of aluminium at the cell surface occurred at two or more sites with evidence of cooperativity. Addition of aluminium nitrate to a weakly buffered cell suspension caused acidification of the medium attributable to displacement of protons from cell surfaces by metal cations. It is concluded that aluminium toxicity is related to pH-dependent speciation [with Al(H2O) 6 3+ probably being the active species] and chelation of aluminium in the medium. Aluminium transport to intracellular binding sites may involve Fe(III) transport pathways.  相似文献

The marine gastropod Buccinanops globulosus is known to have high imposex incidence in areas moderately polluted by tributyltin (TBT). Acute toxicity was previously studied in adults but no information is known about embryonic intracapsular development. To estimate the potential effects of organotin pollution on the progeny of B. globulosus, acute toxicity tests were conducted on encapsulated and excapsulated pre-hatching embryos. The lethal median concentration estimated for 96?h (LC50 96?h) in B. globulosus excapsulated embryos was 196.70?µg?TBTCl?L?1, while in encapsulated embryos it was 2951.28?µg?TBTCl?L?1. The LC50 96?h was 15-fold higher for encapsulated embryos compared to excapsulated embryos, denoting egg capsule protection against pollutants from the external environment. Our results show that TBT pollution can have significant effects in molluscs other than the chronic effect of imposex.  相似文献

Experiments were performed with the mesogastropodLittorina littorea on Helgoland, in Roscoff, and in the laboratory in order to evaluate the reaction of the female genital system to TBT, an environmental toxicant. The snails were either injected with 50 or 100 ng tributyltin (TBT) soluted in ethanol or exposed to artificial sea water treated with 5, 50, 100, and 200 ng TBT/l, and 33 ng testosterone/l. The duration of the experiments was either four or eight weeks. None of the analysed femaleL. littorea showed signs of imposex. Compared to results for the control groups, the size of the female glandular complex was significantly reduced if the pre-experimental toxication was already high, as is the case in snails collected around Helgoland. TBT-related gland complex reduction occurs also in femaleL. littorea from other sampling sites. In addition, injection of ethanol also causes a decrease in gland size. The experimental results demonstrate that the distal female genital system responds with significantly lower sensitivity to TBT than that of other prosobranchs. This behaviour is ascribed to the lack of an androgen receptor at the ovipositor. The results further strengthen the case of the extreme rarity of imposex described for femaleL. littorea in natural habitats. However, strong TBT-toxication may affectL. littorea populations significantly because of increasing masculinization of the females, which reduces reproduction ability.  相似文献

Summary Sorghum plants were grown in the greenhouse in modified Steinberg nutrient solution containing ten Al rates (0 to 297 μM) and harvested 28 days after transplanting. Top and root dry weight were not affected by added Al up to 74 μM; but decreased sharply at concentration of 148 μM and greater. Aluminum concentrations in blade 1 (recently matured blade) and plants remained constant from 0 to 297 μM added Al. Root Al concentration increased as added Al increased. No correlation existed between top dry weight and Al concentration in blade 1 or in plant. Root Al concentration was related to top dry weight and root dry weight to estimate the Al critical toxicity level. The Al critical toxicity levle in the root was 54 mmol kg−1 root dry weight basis for either top or root dry weight. In blade 1 Cu concentration negatively correlated with Al while Fe and P were positively correlated. In roots Ca, Mg, Mn and Fe concentrations were negatively correlated with Al while Zn, Cu, P, and K were positively correlated with Al concentration.  相似文献

The midge Chironomus tentans Fabricius is a commonly used freshwater invertebrate in sediment toxicity tests. Rigorous laboratory culturing techniques are needed to provide organisms of uniform quality and known age for use in testing and for the continuation of the culture itself. This study was conducted to determine the effect of initial culture stocking density on: (1) post-hatch (larval) dry weight, body length and head-capsule width at 10 and 20 days; (2) time to emergence; (3) number and sex of emergent adults; (4) number of larvae and pupae at test termination (day 42 post hatch); and (5) adult dry weight. Three egg stocking densities were used 690 (1.1 eggs cm–2), 1043 (1.7 eggs cm–2) and 1463 (2.4 eggs cm–2). Mean weight of larvae at 10 days in high density tanks (0.13 mg/organism) was significantly higher (P=0.003) than both the medium and low density tanks (0.10 and 0.09 mg/organism, respectively). No significant differences between the three stocking densities were observed for the body length or head-capsule width at either 10 or 20 days post-hatch. Although not statistically significant, larval dry weight decreased with increased stocking density at day 20. A significantly (P=0.02) greater number of females (173±28) emerged from the low stocking density compared to both the medium and high stocking densities (123±45 and 118±54, respectively). Peak adult emergence for the low and medium stocking densities occurred between days 22 and 25 post-hatch, whereas peak adult emergence occurred between days 30 and 33 for the high stocking density. Survival relative to the initial number of eggs stocked was significantly greater (P=0.007) in the low density treatment compared to that in either the medium or the high density treatments. Mean adult weight exhibited an inverse relationship with initial stocking densities. At test end, there was not a significant difference in the mean number of organisms surviving and emerging in the three density levels. The central tendency for number of organisms surviving for all three treatments was 504 organisms per tank (0.82 organisms cm–2). The results of this experiment suggest that an optimal egg stocking density of 1.0 egg cm–2 (600 eggs/tank) be used with the feeding rate identified. This would ensure uniform larvae at the appropriate developmental stage (2nd–3rd instar) needed for toxicological research/testing (e.g. 10 days post-hatch), as well as producing sufficient emergence of males and females for future culture establishment.  相似文献

BACKGROUND: Ephedra was commonly used in herbal products marketed for weight loss until safety concerns forced its removal from products. Even before the ban, manufacturers had begun to replace ephedra with other compounds, including Citrus aurantium, or bitter orange. The major component in the bitter orange extract is synephrine which is chemically similar to ephedrine. The purpose of this study was to determine if relatively pure synephrine or synephrine present as a constituent of a bitter orange extract produced developmental toxicity in rats. METHOD: Sprague‐Dawley rats were dosed daily by gavage with one of several different doses of synephrine from one of two different extracts. Caffeine was added to some doses. Animals were sacrificed on GD 21, and fetuses were examined for the presence of various developmental toxic endpoints. RESULTS AND CONCLUSION: At doses up to 100 mg synephrine/kg body weight, there were no adverse effects on embryolethality, fetal weight, or incidences of gross, visceral, or skeletal abnormalities. There was a decrease in maternal weight at 50 mg synephrine/kg body weight when given as the 6% synephrine extract with 25 mg caffeine/kg body weight; there was also a decrease in maternal weight in the caffeine only group. This decrease in body weight may have been due to decreased food consumption which was also observed in these two groups.
