This study describes morphology and fine structure of the Persian sturgeon (Acipenser persicus) (Acipenseridae, Chondrostei) spermatozoon. The results show that the spermatozoon of A. persicus is differentiated into an elongated head (length: mean±SD: 7.1±0.5μm) with an acrosome (length: 1.2±0.2μm), a cylindrical midpiece (length: 1.8±0.5μm), a flagellum (length: 50.3±5.9μm) and a total length of 59.2±6.2μm. Ten posterolateral projections (PLPs) arise from the posterior edge of the acrosome and there were 3 endonuclear canals that traversed the nucleus from the acrosomal end to the basal nuclear fossa region. Three to six mitochondria were in peripheral midpiece and the proximal and distal centrioles were located near to "implantation fossa" and basement of the flagellum. The axoneme has a typical eukaryotic structure composed of 9 peripheral microtubules and a central pair of single microtubule surrounded by the plasma membrane. Lateral fins were observed along the flagellum. The fins started and ended at 0.5-1μm from midpiece and at 4-6μm from the end of flagellum. There were significant differences in the size of almost all measured morphological parameters between males and flagellar, midpiece and nucleus characters were more isolated parameters that can be considered for detecting inter-individual variations. This study showed that sperm morphology and fine structure are similar among sturgeon species, but the dimensions of the parameters may differ.  相似文献

BACKGROUND INFORMATION: Available data concerning the sperm morphology of teleost fishes demonstrate wide variation. In the present study, the spermatozoa of Siberian sturgeon (Acipenser baerii Brandt, 1869), a chondrostean fish, was investigated. In contrast with teleost fish, chondrostean spermatozoa have a head with a distinct acrosome, whereas other structures, such as a midpiece and a single flagellum, are present in spermatozoa of most species. RESULTS: The average length of the head including the acrosome and the midpiece was 7.01+/-0.83 microm. Ten posterolateral projections derived from the acrosome were present on a subacrosomal region, with mean lengths of 0.94+/-0.15 microm and widths of 0.93+/-0.11 microm. The nucleus consisted of electrodense homogeneous nuclear chromatin. Three intertwining endonuclear canals, bound by membranes, traversed the nucleus longitudinally from the acrosomal end to the basal nuclear fossa region. There were between three and six mitochondria, two types of centrioles (proximal and distal) in the midpiece and two vacuoles composed of lipid droplets. The flagellum (44.75+/-4.93 microm in length), originating from the centriolar apparatus, had a typical 9+2 eukaryotic flagellar organization. In addition, there was an extracellular cytoplasm canal between the cytoplasmic sheath and the flagellum. CONCLUSIONS: A principal components analysis explained the individual morphological variation fairly well. Of the total accumulated variance, 41.45% was accounted for by parameters related to the head and midpiece of the sperm and the length of the flagellum. Comparing the present study with previous studies of morphology of sturgeon spermatozoa, there were large inter- or intra-specific differences that could be valuable taxonomically.  相似文献

Ultrastructure of sterlet Acipenser ruthenus L. 1758 sperm was examined by scanning and transmission electron microscopy, which allowed us to use various methods for visualizations of different parts of sterlet spermatozoa. Sperm cells possess a head with a distinct acrosome, a midpiece and a single flagellum surrounded by the flagellar plasma membrane. The average length of the head including the acrosome and the midpiece was estimated as 5.14+/-0.42 microm. Nine to 10 posterolateral projections were derived from the acrosome. Three inter-twining endonuclear canals bounded by membranes traversed the nucleus in its whole length from the acrosome to the implantation fossa. Acrosin was located in all the three parts (acrosome, endonuclear canals and implantation fossa). The proximal and distal centrioles located in the midpiece compacted of nine peripheral triplets of microtubules. One cut of the midpiece contained from two to six mitochondria with area of 215+/-85 nm(2) in average. The flagellum was 42.47+/-1.89 microm in length with typical eukaryotic organization of one central pair and nine peripheral pairs of microtubules. It passed through a cytoplasmic channel in the midpiece, which was formed by an invagination at the plasmalemma. The flagellum gradually developed two lateral extensions of its plasma membrane, so-called "fins". Detected morphological variation can be described by four principal component axes corresponding to groups of individual morphometric characters defined on the sperm structures. Correlations among the characters indicate that the sperms are variable in their shape rather than size. Significant variation among examined fish individuals was found only in flagellum and nucleus length. Comparison between the present and previous studies of morphology of sturgeon spermatozoa confirmed large inter- and/or intra-specific differences that could be of substantial taxonomic value.  相似文献

Lates niloticus is a valuable commercial fish species with good potential for aquaculture. However, there is limited information on the type and structure of the Nile perch spermatozoon, which could potentially aid in culture of this species. Here, we describe the spermatozoon ultrastructure in L. niloticus using transmission and scanning electron microscopy. The spermatozoon had a round head-shape, medio-laterally flat, no acrosome, a short midpiece located laterally to the nucleus, uniflagella with one wing. The head of the spermatozoon contained the nucleus, centriolar system, proximal part of the flagellum, and cytoplasmic channel. Centrioles were arranged at an angle of 90° to each other, forming a T-shape, parallel to the nucleus. The midpiece was cylindrical, loaded with cytoplasm, five to seven spherical mitochondria; and the flagellum’s plasma membrane extended to form one lateral wing. The spermatozoa were classified as type II spermatozoa. L. niloticus spermatozoon differed from that of its Australian congener L. calcarifer, especially in the centriole arrangement and nuclear shape, length of the midpiece and the number of mitochondria and lateral wings.  相似文献

分别以扫描电镜和透射电镜方法研究了圆斑星鲽 (Veraspervariegates)精子的超微结构。头部呈圆形、中段不发达 ,具有多个未分化的圆形小线粒体、鞭毛结构极简单 ,具有 9 2型的轴丝等特征 ,均表示圆斑星鲽的精子属于简单的原始类型 ,与多数其它高等鱼类精子结构相似。但是 ,圆斑星鲽精子核前部的凹陷 ,即不含染色质的电子透明区 ,以及核前区的囊泡结构却未见在其它新鳍鱼类精子中报道。圆斑星鲽精子核前凹陷开口于核前端稍偏处 ,凹陷呈不规则状 ,可深入到核的中央。凹陷处与染色质区无界膜分隔。凹陷区的前部及其开口处常有多个小的具单层界膜的囊泡 ,有时可见在凹陷区的开口处汇集成一个大泡。在某些处理过程中 ,例如在冷冻 -解冻过程中 ,囊泡可能丢失 ,而在精子顶端留下一个凹坑。尽管顶体在现存最早的新鳍鱼类 ,如雀鳝鱼及弓鳍鱼中已经完全消失 ,但考虑甚至在某些纯真骨鱼 ,如鲑形目中的螈鱼精子上 ,顶体仍然存在 ,而在其它一些纯真骨鱼精子或精子细胞中也存在顶体或顶体遗迹等事实。我们推测在圆斑星鲽精子中的上述特殊结构也是顶体的一种遗迹 ,但也不排除其是向外释放核内物质的一种途径的可能性  相似文献

The fine structure of the spermatozoa of two spionids is described. The spermatozoon of Prionospio cf. queenslandica is typical of an animal utilizing external fertilization, in having a subspheroidal nucleus, a midpiece composed of unmodified rounded mitochondria surrounding two centrioles and a free flagellum. The acrosome is unusual in showing bilateral symmetry. The spermatozoon of Tripolydora sp. resembles that of spionids utilizing spermatophores, in possessing an extremely elongate nucleus and midpiece. The nucleus is penetrated by the 9+2 axoneme for its entire length, linking with a single centriole at the anterior end. Platelets surround the nucleus and intermingle with the mitochondria of the midpiece, which terminates with an annulus. The acrosome shows some internal vesiculation and substructuring. Sperm structure in relation to reproductive methods is discussed and the view of external fertilization as primitive is questioned.  相似文献

透射和扫描电镜研究显示玫瑰无须售巴(Puntius conchonius)的精子由头、中片和尾三部分组成。头部无顶体,呈球形或卵圆形,主要由细胞核组成,核内染色质致密。核前端几乎无细胞质存在,核膜紧密靠近细胞质膜,而在核的后端有少量细胞质存在。在核后端偏于一侧处有一个浅的核后凹,中心粒复合体部分地镶嵌于其中,中心粒复合体由近端中心粒和远端中心粒组成,二者呈钝角形排列,鞭毛从远端中心粒的末端发出。中片由前边的主要部分——领和后边细薄的袖套构成。领内含有数个不规则分布的线粒体包埋于细胞质中,袖套的长短、粗细差别较大,有的精子没有袖套。由于与鞭毛的不对称连接,使得头部及中片均呈不对称型。尾是一根细长的鞭毛,尾丝具有典型的“9+2”微管结构,尾部两侧均无侧鳍。与鲤科其它鱼精子相比,该鱼精子的主要特征是具有长短不一的袖套,领内有不同数量的液泡,且有些空泡向外界开口呈孔状。袖套的长短与领内液泡化水平似有某种相互联系,这也许与精子的老化程度有关[动物学报51(5):892—897,2005]。  相似文献

Unlike the primitive type of spermatozoon found in most polychaetes, the spermatozoon of Autolytus has a bilateral symmetry with elongated nucleus, and the mitochondria surround the posterior part of the nucleus. A rather large disk-shaped acrosome is situated along one side of the anterior part of the nucleus. From the anterior margin of the distal centriole emerge long striated rootlets, which run along the nuclear envelope to the anterior part of the nucleus. The spermatozoon of Chitinopoma serrula has an elongated, slightly bent nucleus, a thimble-like acrosome apically on the anterior surface of the nucleus, and an elongated middle piece containing 4 rod-like mitochondria developed from spherical mitochondria surrounding the basal part of the tail flagellum. In the spermatozoon of Capitella capitata, both nucleus and middle piece are elongated compared to the primitive type. The large and conical acrosome is placed asymmetrically at the nucleus and consists of an acrosomal vesicle and subacrosomal substance. The greater part of the middle piece forms a collar around the initial part of the tail flagellum. The cytoplasm of the collar contains granular material. One or two small mitochondria lie around the 2 centrioles at the base of the nucleus.

These types of spermatozoa represent early steps in the evolution of modified spermatozoa combined with changed biology of reproduction. The modified spermatozoa are larger than the primitive ones.  相似文献

This paper investigates by scanning and transmission electron microscopy spermiogenesis and spermatozoon morphology of the gonochoristic eutardigrade Xerobiotus pseudohufelandi (Macrobiotidae). During spermiogenesis clusters of spermatids are connected by cytoplasmic bridges that persist up to an advanced stage of maturation. Spermiogenesis is characterized by distinctive modifications of the nucleus and by the differentiation of an acrosome, tail and substantial midpiece. Testicular spermatozoa are folded with the hinge located between the head and midpiece, thus resembling a nut-cracker. The head includes a rod-shaped, bilayered acrosome and an elongated, helicoidal nucleus with condensed chromatin. The large kidney-shaped midpiece has hemispherical swellings/ovoid elements surrounding the centriole and an incomplete mitochondrial sleeve. The flagellum contains a ‘9+2’ axoneme and terminates in a tuft of microtubules. Spermathecal spermatozoa always have linear profiles. The acrosome and nucleus have the same morphological pattern as in testicular spermatozoa, whereas the midpiece is thin and lacks the hemispherical swellings, and the tail is reduced to a short stub. Functional considerations are presented, based upon this different morphology. The present study was carried out with two sturgeon species, Siberian sturgeon (Acipenser baerii) and sterlet (A. ruthenus) to compare spermatological parameters to better understand inter‐species differences. Significant differences between morphometric features were observed such as acrosome length, acrosome width, head length, midpiece width and flagellar length, while midpiece length did not reveal such differences. The sterlet has a shorter spermatozoon than the Siberian sturgeon. Ultrastructural parameters vary significantly in terms of length of the nucleus, diameter of the endonuclear canals (EC), size of posterolateral projections (PP) and diameter of flagellum. Mean values for density of spermatozoa in the semen, seminal plasma pH, osmolality (mOsmol kg?1), along with Ca2+, Na+, K+, Cl? ions concentrations (mm ) were determined to be 0.61 ± 0.37 × 109, 8.16 ± 0.18, 77.20 ± 52.28, 0.24 ± 0.06, 31.39 ± 10.21, 3.51 ± 1.10, 14.00 ± 4.30 in A. baerii and 0.41 ± 0.32 × 109, 8.13 ± 0.19, 50.74 ± 6.27, 0.16 ± 0.11, 20.11 ± 3.78, 1.26 ± 0.54, 6.11 ± 0.60 in A. ruthenus, respectively. Significant differences were observed in Na+, K+ and Cl? concentrations in the seminal plasma as well as in sperm velocity. The percentage of motile spermatozoa did not show any significant difference between the two species. Comparing the results of this study with published literature data on sturgeon spermatozoa reveals that morphological and ultrastructural parameters of spermatozoa together with some parameters of the seminal fluid and spermatozoa velocity can be used in comparative spermatology to better understand inter‐species differences. The observed biochemical and physiological differences should be also considered for the development of methods for controlled reproduction and for sperm cryopreservation techniques.  相似文献

Summary

Light and electron microscopy were used to analyze the morphology and ultrastructure of the spermatozoon of Fissurella nigra and Fissurella picta, two sympatric species of keyhole limpets from the southern Chilean coast. The spermatozoa of both species are of the aquasperm type, typical of species with external fertilization. Each species had its own distinctive spermatic morphology, particularly in relation to size. Proceeding from anterior to posterior, the spermatozoa of both species each consists of an elongated head with a conical acrosome having a deep subacrosomal space and truncated conical nucleus, followed by a midpiece containing five mitochondria associated in a compact ring around the proximal and distal centrioles, and, at the end, a flagellum. The spermatozoon head of F. nigra is almost twice as long as that of F. picta. This difference constitutes the first morphological evidence that the size of the spermatozoon could represent a candidate for the maintenance of reproductive isolation between these two sympatric species.  相似文献

Spermatozoa of the echiurans Bonellia viridis and Hamingia arctica show a similar ultrastructure. They are of a modified type. The head consists of a roughly cylindrical nucleus, which has a cover of electron-dense material. The acrosome is very large and consists of an acrosomal vesicle and a rod-shaped perforatorium or acrosomal rod. In close association with the nucleus, one or two mitochondria are found forming an irregular ring around the posterior tip of the nucleus and the centriolar apparatus. There are two centrioles, the proximal one with the conventional triplet microtubular structure. The tail flagellum is about 50 μm long and has the 9+2 axonemal structure. The oblique attachment of the acrosome to the anterior part of the nucleus gives the spermatozoon a bilateral symmetry. However, in the nuclear morphology, the arrangement of electron-dense material around the nucleus, in the mitochondria, and in the attachment of the tail flagellum, the spermatozoon shows asymmetric organization. The sperm structure in bonelliids is unique but its genesis and the morphology of the mitochondrial midpiece support the theory that the echiurans are related to the annelids. Spermatogenesis in Sparganophilus tamesis results in a filiform spermatozoon of the advanced type. Structures in proximal-distal sequence are the slender acrosome, the extremely elongate nucleus, the short cylindrical midpiece and the long flagellum. The acrosome is unique in those Lumbricina so far studied in persistence of the subvesicular space in the anterior (proximal) bulb formed by the primary acrosome vesicle, external to the acrosome tube and in extension of the acrosome rod into this proximal subvesicular space. A further unusual feature is the very long basal chamber in the acrosome tube, distal to the rod, as in the Phreodrilidae. The acrosome tube has a distal limen and its junction with the nucleus is domed. There is little similarity with the acrosome of the Lumbricidae, a family with which the Sparganophilidae have recently been grouped. The midpiece has the highest number of mitochondria yet recorded for oligochaetes. The number is deduced to be eight, of which seven are normally visible in cross section because one of the mitochondrial loci consists, uniquely for the Oligochaeta, of two mitochondria aligned longitudinally in tandem. The structure of the flagellum, including the arrangement of its peripheral glycogen granules, is typical for oligochaetes. The spermatozoon is straight, lacking a spiral coil or keel. The main characteristic features of spermiogenesis in Chilodus punctatus (Characiformes) are rotation of the nucleus, development of a nuclear fossa, which extends as a narrow invagination deep into the nucleus and the way in which flagellum is formed. The chromatin condensation proceeds during the spermiogenesis from heterogeneous through homogenous and granular to a highly compact one present in the mature spermatozoon. Mature Ch. punctatus spermatozoon shows a spherical nucleus, short midpiece and flagellum with lateral fins. The centrioles are in perpendicular arrangement and are located in the deep nuclear fossa, which extends towards the anterior pole of the nucleus. The midpiece contains a few mitochondria, which are separated from the anterior fragment of flagellum by the cytoplasmic channel. Spermiogenesis and spermatozoon ultrastructure conform to the pattern observed in other ostariophysans, but for the first time the presence of lateral fins along flagellum has been documented in a representative of Characiformes.  相似文献

Paddlefish, Polyodon spathula, spermatozoa were examined by transmission electron microscopy. Their structure has the same characteristic architectural features as sturgeon spermatozoa. Paddlefish spermatozoa are of the primitive type and consist of a rod-shaped head, a midpiece and a long flagellum. The head is about 5.15 mm in length and contains the nucleus and an apical acrosomal complex. Inside the nucleus there are three nuclear channels that begin in the subacrosomal area and have a triple helical arrangement. An nuclear fossa is present centrally, at the posterior end of the nucleus. The midpiece contains a pair of centrioles in a perpendicular arrangement, mitochondria and a narrow cytoplasmic sleeve. The flagellum has a central axoneme with a 9 + 2 pattern and two lateral projections or fins.  相似文献

In the present review, sperm morphology, acrosome reaction, motility, short-term storage and cryopreservation are summarized and discussed in sturgeon (Chondrostei, Acipenseriformes). The elongated head of spermatozoon comprises an acrosome with 8?C12 posterolateral projections. Usually three endonuclear canals are observed in the nucleus. Proximal and distal centrioles and 3?C6 mitochondria are located in the midpiece region. The flagellum consists of an axoneme with a typical ??9?+?2?? structure of microtubules and presents a ribon-like structure due to two lateral membranous fins. Egg water, Ca2+ and Mg2+ can trigger acrosome reaction. Trypsin- and chymotrypsin-like activities are reported in sturgeon sperm. These physiological properties of sturgeon sperm are identified as serine activity with 33?kDa molecular mass and can be inhibited by their respective inhibitors. The K+ prevents sperm activation in seminal plasma, and hypo-osmolality or decrease of extracellular K+ triggers sperm activation. Extracellular Ca2+ is involved in flagellar beating pattern and sperm velocity. After activation, sperm motility, velocity, and flagellar beating frequency, wavelength and amplitude decrease, while number of waves and curvature increase. Sturgeon sperm can be stored for several days at 4?°C; however it is better to add K+ into the immobilizing medium because it prevents sperm activation during incubation. Regarding sperm cryopreservation, methanol is a better cryoprotectant than DMSO. Either short-term storage or cryopreservation of sperm generates damage to spermatozoa that lead to reduction of sperm motility performance. Some studies suggest using an activation medium containing Ca2+ for enhancing sperm motility performance of incubated or frozen-thawed sperm.  相似文献

A fully differentiated spermatozoon of both Ascidia zara and Ascidia gemmata is approximately 35 microM long. It contains a head and a tail lacking a midpiece. The head (approximately 4 microM long for A. zara and 5 microM long for A. gemmata) contains an elongated nucleus and a single mitochondrion that flanks the nucleus. Multiple acrosomal vesicles (three or four in number) are present at the apex of the sperm head in both species. Each vesicle is approximately 50 x 50 x 60 nm, and contains moderately electron-dense material. During spermiogenesis of A. zara, three or four vesicles appear in a blister of an early stage spermatid. These vesicles transform into multiple acrosomal vesicles without fusing with each other. Spermiogenesis and acrosome differentiation are similar in A. gemmata and A. zara. Three types of acrosome differentiation in ascidians are described.  相似文献
