Genetic diversity has been investigated by the application of molecular markers in, for the first time, all the taxa recognised in recent treatises of the genusHippophae. RAPD (random amplified polymorphic DNA) analyses were conducted with 9 decamer primers, which together yielded 219 polymorphic markers. We found 16 fixed RAPD markers, i.e. markers that either occurred in all plants of a population or were absent from all plants. Several of these markers were useful for analysis of interspecific relationships, whereas others can be considered as taxon-specific markers. Clustering of taxa and populations in our neighbour-joining based dendrogram was in good agreement with some recently suggested taxonomic treatises ofHippophae. Amount and distribution of genetic variability varied considerably between species. Partitioning of molecular variance withinH. rhamnoides supported earlier findings that a considerable part of the total variance resides among subspecies (59.6%) Within-population variability also differed considerably. Percentage polymorphic RAPD loci and Lynch and Milligan within-population gene diversity estimates showed relatively high values for some species close to the geographic centre of origin in Central Asia, e.g.H. tibetana and the putatively hybridogenousH. goniocarpa. Spatial autocorrelation analyses performed on 12 populations ofH. rhamnoides revealed positive autocorrelation of allele frequencies when geographic distances ranged from 0 to 700 km, and no or negative autocorrelation at higher distances. At distances between 700 and 1900 km, we observed deviations from the expected values with strongly negative autocorrelation of allele frequencies. A corresponding relationship between geographic and genetic distances could not be found when the analysis instead was based on one population from each of 8 species.  相似文献

用31个可重复性好的随机引物对中华绒螯蟹(Eriocheir sinensis)的辽河、瓯江和长江种群进行了RAPD分析。121个扩增片段中有27个多态片段。遗传距离指数(D)表明中华绒螯蟹种内遗传变异较低(D=0.004~0.063)。3个种群中,辽河种群(D=0.018)和瓯江种群(D=0.025)遗传变异较高,而长江种群遗传变异较低(D=0.013);辽河种群和瓯江种群间遗传距离(D=0.023)小于它们与长江种群间的遗传距离(D值分别为0.028和0.032)。提示人类经济活动可能已使这2个种群发生了基因交流。  相似文献

Genetic variation is a key component for improving a stock through selective breeding programs. Randomly amplified polymorphic DNA (RAPD) markers were used to assess genetic variation in three wild population of the catla carp (Catla catla Hamilton 1822) in the Halda, Jamuna and Padma rivers and one hatchery population in Bangladesh. Five decamer random primers were used to amplify RAPD markers from 30 fish from each population. Thirty of the 55 scorable bands were polymorphic, indicating some degree of genetic variation in all the populations. The proportion of polymorphic loci and gene diversity values reflected a relatively higher level of genetic variation in the Halda population. Sixteen of the 30 polymorphic loci showed a significant (p < 0.05, p < 0.01, p < 0.001) departure from homogeneity and the F(ST) values in the different populations indicated some degree of genetic differentiation in the population pairs. Estimated genetic distances between populations were directly correlated with geographical distances. The unweighted pair group method with averages (UPGMA) dendrogram showed two clusters, the Halda population forming one cluster and the other populations the second cluster. Genetic variation of C. catla is a useful trait for developing a good management strategy for maintaining genetic quality of the species.  相似文献

We examined genetic variation in sympatric diploid and polyploid brine shrimp Artemia parthenogenetica from each of three populations (China, Italy and Spain). Italian and Spanish tetraploids are closely related (I=0.964). Diploids and tetraploids within each of the two European populations are also closely related (mean I=0.905). Most alleles found in diploids also exist in sympatric polyploids. In contrast, the asexual Artemia (2N, 4N and 5N) in our study share few alleles with their close sexual relative, A. tunisiana (mean I=0.002). These results, as well as the work of other authors, strongly suggest that at least the tetraploid Artemia in our study have an autopolyploid origin.Clonal diversity of polyploid Artemia can be very high at least in some population. Both diploids and polyploids had low clonal diversities in the populations dominated by polyploids and high clonal diversities in the population dominated by diploids.The most common genotypes of sympatric diploid and polyploid Artemia frequently differed. Some alleles occurred only in diploids, while others were restricted to polyploids. These results suggest that polyploidy in Artemia has led to genetic divergence from diploid progenitors, and that ploidy-level variation must also be considered in developing an understanding of spatial and temporal allozyme polymorphism in asexual populations.  相似文献

Random amplified polymorphic DNA (RAPD) markers were used to measure genetic diversity of Coelonema draboides (Brassicaceae), a genus endemic to the Qilian Mountains of the Qinghai-Tibet Plateau. We sampled 90 individuals in 30 populations of Coelonema draboides from Datong and Huzhu counties of Qinghai Province in P.R. China. A total of 186 amplified bands were scored from the 14 RAPD primers, with a mean of 13.3 amplified bands per primer, and 87% (161 bands) polymorphic bands (PPB) was found. Analysis of molecular variance (AMOVA) shows that a large proportion of genetic variation (84.2%) resides among individuals within populations, while only 15.8% resides among populations. The species shows higher genetic diversity between individuals than other endemic and endangered plants. The RAPDs provide a useful tool for assessing genetic diversity of rare, endemic species and for resolving relationships among populations. The results show that the genetic diversity of this species is high, possibly allowing it to adapt more easily to environmental variations. The main factor responsible for the high level of differentiation within populations and the low level of diversity among populations is probably the outcrossing and long-lived nature of this species. Some long-distance dispersal, even among far separated populations, is also a crucial determinant for the pattern of genetic variation in the species. This distributive pattern of genetic variation of C. draboides populations provides important baseline data for conservation and collection strategies for the species. It is suggested that only populations in different habitats should be studied and protected, not all populations, so as to retain as much genetic diversity as possible.  相似文献

Bosmina (Eubosmina) longicornis kessleri and B. (Bosmina) longirostris are minor components among the crustacean zooplankton in the pelagial of Bukovec Reservoir. B. (E.) l. kessleri is a recent colonist in the Carpathian region and was first found in the Bukovec reservoir in 1986. The present study gives some information about the niche occupied by B. (E.) l. kessleri in the zooplankton community; the seasonal abundance of zooplankton and phenotypic variability (cyclomorphosis) of B. (E.) l. kessleri are also examined.  相似文献

Turkey is one of the most important mulberry fruit producers in the world. In particular, in Eastern and Inner part of Turkey, mulberry fruits are processed into several products such as `Mulberry Pekmez', `Mulberry Pestil', `Mulberry Kome' etc. Therefore, mulberry fruits give higher economic returns thus have greater employment potentials in Turkey. In order to improve the yield and fruit quality through breeding, the genetic variations and relationships among 15 selected white mulberry genotypes (Morus alba L.) were evaluated using Random Amplified Polymorphic DNA (RAPD). A total of 101 DNA markers were generated by 16 random primers, with an average of 6.3 easily detectable markers per primer. Several RAPD markers showed unique patterns of mean frequency that differed among the white mulberry genotypes. The distance matrix showed that the highest genetic distance (0.902) was between EMS13 and EMS8 and the least (0.413) was between EMS12 and EMS15. According to the results, RAPD analysis can be used for the characterization and grouping of mulberry genotypes. The genetically divergent genotypes identified from this study would be of much use in the future breeding program.  相似文献

Nine Swedish populations, 1–5 individuals/population, and one cultivated individual of the rare speciesVicia pisiformis were investigated for genetic variation. In hybridizations with two rDNA probes using 8 restriction enzymes, only two individuals belonging to one population were polymorphic. A map of the rDNA gene cluster was constructed for four of the restriction enzymes used. Two of the polymorphic sites were mapped and were found to be located outside regions coding for rRNA, presumably caused by single point mutations or small deletions. The repeat length of the rDNA region was c. 10,000 bp, which corresponds well with the size found for other species belonging toFabaceae. No length polymorphism was found in the intergenic spacer, contrary to the situation found for most other plant species investigated for rDNA variation. The haplotype diversity for the species (Hsp Shannon) was very low (0.055). Within-population values (Hpop) was 0 for all populations except the variable one, which had 0.301. PCR amplification with 6 random primers also revealed very low levels of genetic diversity. A polymorphism was observed in a limited number of individuals for four populations. Hsp was 0.065 and was 0.050. The average D value (Wetton) for the PCR haplotypes was 0.99.  相似文献

We compared genetic variation of sexualAmelanchier bartramiana and facultatively agamospermous (asexually seed-producing)A. laevis at one site where the two species are sympatric. We analyzed 77 random amplified polymorphic DNA (RAPD) markers in 29A. bartramiana individuals and 76 RAPD markers in 31A. laevis individuals. The two species do not differ significantly in mean genetic variation. However, 22.4% of genetic similarity values betweenA. laevis individuals exceed the highest value ofA. bartramiana and may represent the effect of agamospermy. Variation withinA. laevis may be the result of sexuality, hybridization, polyploidy, and other factors.  相似文献

The genetic polymorphism of Malagasy prosimian populations is studied by using RAPD markers. The analysis includes twoLepilemur septentrionalis populations of the area of Analamera separated by a deforested hill crest and one population ofL. dorsalis from the island of Nosy-Be. The genetic diversity is higher in the two populations of Analamera than in that of Nosy-Be and the level of genetic differentiation is higher between the population ofL. dorsalis and the two populations ofL. septentrionalis than between the two populations ofL. septentrionalis themselves. Despite the hill crest separating the two populations ofL. septentrionalis, our results demonstrate that they belong to one population. The respective roles of the geographical barriers and the reproductive barrier between the two species, are discussed.  相似文献

应用RAPD标记对19份国内不同生态区的谷子品种的遗传变异进行了研究。结果表明:分子水平上,不同生态区的谷子品种间存在一定的遗传差异,但遗传差异程度并不高。11个随机引物共扩增出54条多态性带,不同引物扩增的带数差异较大,每个引物可扩增2—8条多态性带,平均每个引物扩增出4.91条多态性带。引物1050扩增的多态性带最多(8条)。聚类结果表明,基于RAPD标记分析的遗传聚类群与生态类型有很大的一致性。  相似文献

中华结缕草 (ZoysiasinicaHance.)在我国是分布于东部沿海地区的一个受威胁禾草 ,形态上还有一个分布在海边的变种 ,长花中华结缕草 (Z .sinicavar.nipponicaOhwri)。采用 1 0条随机引物对采自我国不同地区 7个居群的 1 0 5个个体进行了RAPD扩增 ,对结果的AMOVA分析表明 ,中华结缕草组间遗传分化不显著 ,遗传变异只占总变异的 4.84% ,居群间遗传变异占总遗传变异的 2 4.71 % ,出现了显著的遗传分化 ,大部分变异存在于居群内部 ,占总变异的 70 .44%。不同数据处理方法得到了类似的结果 ,都支持哈迪 -温伯格平衡的假设。其中宁国汪溪居群和射阳海边居群变异最大 ,歙县新安江居群遗传变异最小。考虑到中华结缕草遗传结构和变异的这些特性 ,在取样策略、保护和育种方法上 ,都应该以群居为主 ,同时兼顾主要的异地居群  相似文献

Psathyrostachys huashanica Keng is a perennial grass and belongs to genus Psathyrostachys under Triticeae. sathyrostachys is found in the center of Middle Asia and the Caucasus Mountain, while P. huashanica, a species endemic to China, is only located in Mt. Hua in the Shaanxi province, China. At present, the population of this species is decreasing, and reaching the edge of extinction. Due to the limitation in distribution and the importance as breeding material for germplasm storage, it has been considered as first class among the national protected rare plants. For this reason, the present study is significant in probing plant flora, origin and evolution of Triticeae, and crop breeding. Randomly amplified polymorphic DNA (RAPD) markers were used to analyze the genetic structure and differentiation of P. huashanica populations sampled in three valleys (Huangpu, Xian, and Huashan Valleys) in Mt. Hua. One hundred and twenty-two RAPD fragments were obtained in all 266 individuals with 20 primers with a mean of 6.1 (2–10) fragments per primer. The percentage of polymorphic loci (PPB) was 60.66% in Huangpu Valley, 90.98% in Xian Valley, 95.08% in Huashan Valley, and the total PPB was 95.08%, which indicated a highly genetic variability of P. huashanica. The Shannon’s Information index and G ST were 0.3306 and 0.3263, respectively, indicating that there were more genetic variations within the subpopulations than those among the subpopulations. The gene flow among the subpopulations of P. huashanica (Nm = 1.0322) was much less than that of the common anemophytes (Nm = 5.24). Mean genetic distance is 0.1 571 (range: 0.0022–0.2901). The highest value of genetic distance was found between the subpopulation (hp1) of Huangpu Valley and the highest altitude subpopulation (h8) of Huashan Valley. Correlation analysis detected significant correlation between genetic distance and vertical distance of altitude. Clustering analysis and principal coordinate analysis revealed the genetic differentiation among the populations of P. huashanica. Differentiation mainly occurred between the higher altitude subpopulations and the lower altitude subpopulations, suggesting that altitude might be the major factor that restricted the gene flow between different altitude subpopulations and resulted in differentiation of subpopulations. The chukar (Alectoris chukar, Galliformes) is one of the most important game birds as it is widely distributed and hunted over the whole of its range. The aim of this work was to assess the genetic differentiation as well as the possible presence of hybrid specimens in A. chukar populations from Italy, Greece and Cyprus. To provide phylogenetic context, conspecific, allopatric specimens from Israel, Georgia, Armenia, Kazakhstan, Afghanistan, Pakistan, Mongolia, China and USA were compared. Sequencing of the mitochondrial DNA (mtDNA) Control Region supplied information on the ancestry of A. chukar populations, whereas Random Amplified Polymorphic DNA (RAPD) fingerprinting was used to assess whether hybridization had occurred. The Italian population was found to be an inter-specific mixture of A. chukar and A. rufa (i.e., the red-legged partridge) mtDNA lineages, whereas the representatives from Greece and Cyprus showed only the A. chukar maternal line. RAPD markers revealed introgression with A. rufa genes in the Italian population, whereas no A. chukar × A. rufa hybrid specimens were detected in the eastern Mediterranean populations. The genetic data obtained from the Italian A. chukar population as well as from a few Greek specimens pointed against their Mediterranean kinship, suggesting relationships with A. chukar subspecies from the easternmost part of the Asian continent.  相似文献

华山新麦草特产于我国陕西华山,为国家一级重点保护植物。应用RAPD技术,用2 0条随机引物对黄埔峪、仙峪、华山峪3个居群不同海拔的13个亚居群共2 6 6个华山新麦草取样个体进行扩增,共得12 2个扩增片段。平均每个引物得到的扩增片段数为6 .1(2~10 )。实验数据的统计分析表明:黄埔峪居群的多态性位点比率为6 0 .6 6 % ,仙峪居群的多态性位点比率为90 .98% ,华山峪居群的多态性位点比率为95 .0 8% ,总的多态性位点比率为95 .0 8% ,说明华山新麦草具有较高的遗传变异性。Shannon多样性指数(0 .330 6 )和基因分化系数(GST=0 .32 6 3)揭示了华山新麦草居群遗传变异多存在于亚居群内。华山新麦草亚居群间的基因流N m=1.0 32 2 ,低于一般风媒传粉植物(N m=5 .2 4 )的基因流水平。亚居群间平均的遗传距离为0 .15 71(变化范围:0 .0 0 2 2~0 .2 90 1) ,最大的遗传距离值出现在黄埔峪亚居群(hp1)和华山峪高海拔亚居群(h8)之间,仙峪亚居群与华山峪高海拔亚居群之间以及华山峪高海拔亚居群与低海拔亚居群之间的遗传距离值也较大。遗传距离与海拔垂直距离之间的相关性分析表明二者有显著的相关性(p<0 .0 1)。聚类分析和主成分分析也显示华山新麦草自然居群已发生明显分化,主要表现为黄埔峪居群、仙峪居群与华山峪居群  相似文献

Ten snap bean (Phaseolus vulgaris) genotypes were screened for polymorphism with 400 RAPD (random amplified polymorphic DNA) primers. Polymorphic RAPDs were scored and classified into three categories based on ethidium bromide staining intensity. An average of 5.19 RAPD bands were scored per primer for the 364 primers that gave scorable amplification products. An average of 2.15 polymorphic RAPDs were detected per primer. The results show that primer screening may reduce the number of RAPD reactions required for the analysis of genetic relationships among snap-bean genotypes by over 60%. Based on the analysis of the distribution of RAPD amplification, the same number of polymorphic RAPDs were amplified from different genotypes for all RAPD band intensity levels. A comparison of RAPD band amplification frequency among genotypes for the three categories of bands classified by amplification strength revealed a measurable difference in the frequencies of RAPDs classified as faint (weakly amplifying) compared to RAPD bands classified as bold (strongly amplifying) indicating a possible scoring error due to the underscoring of faint bands. Correlation analysis showed that RAPD bands amplified by the same primer are not more closely correlated then RAPD bands amplified by different primers but are more highly correlated then expected by chance. Pairwise comparisons of RAPD bands indicate that the distribution of RAPD amplification among genotypes will be a useful criterion for establishing RAPD band identity. For the average pairwise comparison of genotypes, 50% of primers tested and 15.8% of all scored RAPDs detected polymorphism. Based on RAPD data Nei's average gene diversity at a locus was 0.158 based on all scorable RAPD bands and 0.388 if only polymorphic RAPD loci were considered. RAPD-derived 1 relationships among genotypes are reported for the ten genotypes included in this study. The data presented here demonstrate that many informative, polymorphic RAPDs can be found among snap bean cultivars. These RAPDs may be useful for the unique identification of bean varieties, the organization of bean germplasm, and applications of molecular markers to bean breeding.  相似文献

Somaclonal variants resistant to sugarcane mosaic virus (SCMV) were obtained from susceptible sugarcane cv PR62258 through somatic embryogenesis by increasing the number of subcultures of the embryogenic callus tissue in MS medium with 3 mg/L 2,4-dichlorophenoxyacetic acid. Transfers were made at 30-day intervals for 1, 2 or 3 subcultures. Two somaclones, namely AT626 and BT627, were selected by their resistance to SCMV. These subclones have maintained the resistance trait over seven years of testing in the field. In this report we identified the somaclonal SCMV resistant variants from the maternal line and the nonresistant somaclones, using the RAPD technique.  相似文献

RAPD markers were used to measure the genetic diversity of 119 individuals of Ixodes ricinus collected from Lithuania and Norway. The samples were analysed within and also between the populations. We analysed 74 loci in each of 6 populations. Our results show high levels of diversity within the populations. The percentage of polymorphic loci of the six analysed populations: Birzai, Vilnius, Kretinga, Tjore, Kjosvik and Odderoya were 68.9%, 58.1%, 78.38%, 62.2%, 44.6% and 68.9%, respectively. The percentage of polymorphic loci in the Lithuanian populations was 93.2%, and in the Norwegian populations 81.08%. The genetic distance ranged from 0.019 to 0.079 within Norwegian populations and from 0.005 to 0.0967 within Lithuanian populations and between the countries from 0.022 to 0.146. The genetic variation of I. ricinus among Norwegian populations was lower than among Lithuanian populations. The highest part of genetic variation in I. ricinus ticks depends on variation within Kretinga (Lithuania) and Odderoya (Norway) populations situated in coastal areas where many migratory and sea birds are aggregated.  相似文献

Hippophae rhamnoides ssp. sinensis is endemic to China, and it is a dioecious, outcrossing plant. Although many studies have been undertaken mainly on its agricultural, nutritional, medical, and ornamental value, little is known about its population genetics. This study uses random amplified polymorphic DNA to investigate the genetic diversity and population genetic structure of 13 natural populations of the subspecies sinensis. Fifteen primers amplified 107 reproducible bands, with 95 (88.79%) being polymorphic. The gene diversity within population was 0.168, considerably lower than that of tree species and most perennial, outcrossing species, but higher than that of annual or short-lived, selfing species. The Gst value showed that 18.3% of the total genetic variation resided among populations, a little lower than that of outcrossing species. The present results are quite similar to those previously reported in another subspecies, H ssp. . rhamnoides rhamnoides. The low genetic differentiation among populations in ssp. sinensis may be attributed to the long-distance dispersal of seeds facilitated by birds, in addition to its characteristics of outcrossing, wind pollination, and widespread distribution. No association between genetic distance and geographical distribution was found. The population relationships revealed by the UPGMA dendrogram parallel this result, in that genetic distance did not increase with geographic separation. This pattern of population differentiation may imply the adaptation of ssp. s populations to the local environment, given that its habitats vary greatly across its distribution.  相似文献

Salsola komarovi lljin is a herbaceous annual native to the sand dunes and beaches of Japan, northern China, Sakhalln and Korea. Starch-gel electrophoresis was conducted on leaves and stems collected from 300 plants in eight Korean populations. The mean number of alleles per locus (A p=1.51), mean expected heterozygosity (He p=0.116), and total genetic diversity (H T=0.279) were comparable with those for species with similar life history and ecological traits. A general conformance of genotype frequencies to Hardy-Weinberg expectations (meanF IS=−0.030) indicates thatS. komarovi is an outcrossing species. Slightly more than 20% of the genetic variation was found among populations (F ST=0.204). In addition, significant differences in allele frequency were detected between populations at all 11 polymorphic loci (P<0.001). Nei's genetic identities range from 0.885 to 0.985 with a mean of 0.942. However, indirect estimates of the number of migrant per generation (0.97, calculated fromF ST and 0.31, calculated from seven private alleles) indicate that the levels of gene flow is low among Korean populations. Although the species maintains a moderate level of genetic variation within populations, the small, isolated natural populations of the species have been severely destructed by human activities, particularly in summer season. If this is true, conservation efforts should be focused on those populations that currently maintain the most genetic diversity (e.g., populations of Cheju Island and coast of the southwestern Korean Peninsula).  相似文献
