Several factors affecting erythritol production from glycerol by Yarrowia lipolytica Wratislavia K1 strain were examined in batch fermentations. Ammonium sulfate, monopotassium phosphate, and sodium chloride were identified as critical medium components that determine the ratio of polyols produced. The central composite rotatable experimental design was used to optimize medium composition for erythritol production. The concentrations of ammonium sulfate, monopotassium phosphate, and sodium chloride in the optimized medium were 2.25, 0.22, and 26.4 g L?1, respectively. The C:N ratio was found as 81:1. In the optimized medium with 100 g L?1 of glycerol the Wratislavia K1 strain produced 46.9 g L?1 of erythritol, which corresponded to a 0.47 g g?1 yield and a productivity of 0.85 g L?1 hr?1. In the fed-batch mode and medium with the total concentration of glycerol at 300 g L?1 and C:N ratio at 81:1, 132 g L?1 of erythritol was produced with 0.44 g g?1 yield and a productivity of 1.01 g L?1 hr?1.  相似文献

The possibilities of parallel lactic acid and biomass production in batch and fed-batch fermentation on distillery stillage from bioethanol production were studied. The highest lactic acid yield and productivity of 92.3 % and 1.49 g L?1 h?1 were achieved in batch fermentation with initial sugar concentration of 55 g L?1. A significant improvement of the process was achieved in fed-batch fermentation where the concentration of lactic acid was increased to 47.6 % and volumetric productivity for 21 % over the batch process. A high number of Lactobacillus rhamnosus ATCC 7469 viable cells of 109 CFU ml?1 was attained at the end of fed-batch fermentation. The survival of 92.9 % of L. rhamnosus cells after 3 h of incubation at pH 2.5 validated that the fermentation media remained after lactic acid removal could be used as a biomass-enriched animal feed thus making an additional value to the process.  相似文献

A newly-isolated strain of Serratia marcescens, G12, was characterized for 2,3-butanediol (2,3-BD) production. In shake-flask and batch fermentations, 2,3-BD reached 48.5 and 51 g l?1, respectively. Low amounts of (~8 g l?1) of acetoin were also formed. In fed-batch fermentations, strain G12 produced 72.8 g 2,3-BD l?1 with glucose initially at 130 g l?1. When aeration rate was increased to 2.5 vvm for the fermentation process, 2,3-BD reached 87.8 g l?1 and the highest productivity was 1.6 g l?1 h?1. Acetoin was at 6.2 g l?1. G12 therefore may be a suitable candidate strain for large-scale production of 2,3-BD.  相似文献

Clostridium beijerinckii optinoii is a Clostridium species that produces butanol, isopropanol and small amounts of ethanol. This study compared the performances of batch and continuous immobilized cell fermentations, investigating how media flow rates and nutritional modification affected solvent yields and productivity. In 96-h batch cultures, with 80 % of the 30 g L?1 glucose consumed in synthetic media, solvent concentration was 9.45 g L?1 with 66.0 % as butanol. In a continuous fermentation using immobilized C. beijerinckii optinoii cells, also with 80 % of 30 g L?1 glucose utilization, solvent productivity increased to 1.03 g L?1 h?1. Solvent concentration reached 12.14 g L?1 with 63.0 % as butanol. Adjusting the dilution rate from 0.085 to 0.050 h?1 to allow extended residence time in column was required when glucose concentration in fresh media was increased from 30 to 50 g L?1. When acetate was used to improve the buffer capacity in media, the solvent concentration reached 12.70 on 50 g L?1 glucose. This continuous fermentation using immobilized cells showed technical feasibility for solvent production.  相似文献

A new xylose fermenting yeast was isolated from over-ripe banana by enrichment in xylose-containing medium. The phylogenetic analysis of ITS1-5.8S-ITS2 region sequences of ribosomal RNA of isolate BY2 revealed that it shows affiliation to genus Pichia and clades with Pichia caribbica. In batch fermentation, Pichia strain BY2 fermented xylose, producing 15 g l?1 ethanol from 30 g l?1 xylose under shaking conditions at 28°C, with ethanol yield of 0.5 g g?1 and volumetric productivity of 0.31 g l?1 h?1. The optimum pH range for ethanol production from xylose by Pichia strain BY2 was 5–7. Pichia strain BY2 also produced 6.08 g l?1 ethanol from 30 g l?1 arabinose. Pichia strain BY2 can utilize sugarcane bagasse hemicellulose acid hydrolysate for alcohol production, efficiency of fermentation was improved by neutralization, and sequential use of activated charcoal adsorption method. Percent total sugar utilized and ethanol yield for the untreated hydrolysate was 17.14% w/v and 0.33 g g?1, respectively, compared with 66.79% w/v and 0.45 g g?1, respectively, for treated hemicellulose acid hydrolysate. This new yeast isolate showed ethanol yield of 0.45 g g?1 and volumetric productivity of 0.33 g l?1 h?1 from sugarcane bagasse hemicellulose hydrolysate detoxified by neutralization and activated charcoal treatment, and has potential application in practical process of ethanol production from lignocellulosic hydrolysate.  相似文献

Microalgal triglycerides (TAGs) represent a sustainable feedstock for food, chemical and biofuel industries. The operational strategy (batch, semi-continuous, continuous cultivations) has an impact on the TAG productivity. In this study, semi-continuous (i.e. with fixed harvesting frequency) and batch cultivations were compared on TAG production both at lab-scale and in outdoor cultivations. At lab-scale, the semi-continuous TAG productivity was highest for a cycle time of 2 days (SC1; 0.21 g L?1 day?1) and similar to the maximum obtained with the batch (optimal harvest time; 0.23 g L?1 day?1). Although TAG content was lower for SC1 (22 %) than for the batch (35 %), higher biomass productivities were obtained with SC1. Outdoors, semi-continuous cultivations were subjected to a lower degree of stress (i.e. higher amount of nitrogen present in the system relative to the given irradiance) compared to lab-scale. This yielded low and similar TAG contents (10–13 %) in the different semi-continuous runs that were outdone by the batch on both TAG content (15–25 %) and productivity (batch, 0.97–2.46 g m?2 day?1; semi-continuous, 0.35–0.85 g m?2 day?1). The lab-scale experiments showed that semi-continuous strategies, besides leading to similar TAG productivities compared to the batch, could make TAG production cost effective by valorising also non-TAG compounds. However, optimization of outdoor semi-continuous cultivations is still required. For instance, the nitrogen supply and the harvest frequency should be adjusted on the total irradiance. Additionally, future research should focus on recovery metabolism upon nitrogen resupply.  相似文献

Among several fatty acids tested, oleic acid was selected as the most efficient inducer for the production of 4-hydroxydodecanoic acid, a metabolite of β-oxidation, by Waltomyces lipofer. Cells were induced by incubation for 12 h in a medium containing 10 g l?1 yeast extract, 10 g l?1 peptone, 5 g l?1 oleic acid, 1 g l?1 glucose, and 0.05 % (w/v) Tween 80. The optimal reaction conditions for the production of γ-lactones by induced cells were pH 6.5, 35 °C, 200 rpm, 0.71 M Tris, 60 g l?1 hydroxy fatty acid, and 20 g l?1 cells. Non-induced cells produced 38 g l?1 γ-dodecalactone from 60 g l?1 10-hydroxystearic acid after 30 h, with a conversion yield of 63 % (w/w) and a productivity of 1.3 g l?1 h?1 under the optimized conditions, whereas induced cells produced 51 g l?1 γ-dodecalactone from 60 g l?1 10-hydroxystearic acid after 30 h, with a conversion yield of 85 % (w/w) and a productivity of 1.7 g l?1 h?1. The conversion yield and productivity of induced cells were 22 % and 1.3-fold higher, respectively, than those of non-induced cells. Induced cells also produced 28 g l?1 γ-decalactone and 12 g l?1 γ-butyrolactone from 60 g l?1 12-hydroxystearic acid and 60 g l?1 10-hydroxydecanoic acid, respectively, after 30 h. The concentration, conversion yield, and productivity of γ-dodecalactone and γ-decalactone are the highest reported thus far. This is the first study on the biotechnological production of γ-butyrolactone.  相似文献

Bio-fixation of carbon dioxide (CO2) by microalgae has been recognised as an attractive approach to offset anthropogenic emissions. Biological carbon mitigation is the process whereby autotrophic organisms, such as microalgae, convert CO2 into organic carbon and O2 through photosynthesis; this process through respiration produces biomass. In this study Dunaliella tertiolecta was cultivated in a semicontinuous culture to investigate the carbon mitigation rate of the system. The algae were produced in 1.2-L Roux bottles with a working volume of 1 L while semicontinuous production commenced on day 4 of cultivation when the carbon mitigation rate was found to be at a maximum for D. tertiolecta. The reduction in CO2 between input and output gases was monitored to predict carbon fixation rates while biomass production and microalgal carbon content are used to calculate the actual carbon mitigation potential of D. tertiolecta. A renewal rate of 45 % of flask volume was utilised to maintain the culture in exponential growth with an average daily productivity of 0.07 g L?1 day?1. The results showed that 0.74 g L?1 of biomass could be achieved after 7 days of semicontinuous production while a total carbon mitigation of 0.37 g L?1 was achieved. This represented an increase of 0.18 g L?1 in carbon mitigation rate compared to batch production of D. tertiolecta over the same cultivation period.  相似文献

Studies were performed on the effect of CaCO3 and CaCl2 supplementation to fermentation medium for ethanol production from xylose, glucose, or their mixtures using Scheffersomyces (Pichia) stipitis. Both of these chemicals were found to improve maximum ethanol concentration and ethanol productivity. Use of xylose alone resulted in the production of 20.68 ± 0.44 g L?1 ethanol with a productivity of 0.17 ± 0.00 g L?1 h?1, while xylose plus 3 g L?1 CaCO3 resulted in the production of 24.68 ± 0.75 g L?1 ethanol with a productivity of 0.21 ± 0.01 g L?1 h?1. Use of xylose plus glucose in combination with 3 g L?1 CaCO3 resulted in the production of 47.37 ± 0.55 g L?1 ethanol (aerobic culture), thus resulting in an ethanol productivity of 0.39 ± 0.00 g L?1 h?1. These values are 229 % of that achieved in xylose medium. Supplementation of xylose and glucose medium with 0.40 g L?1 CaCl2 resulted in the production of 44.84 ± 0.28 g L?1 ethanol with a productivity of 0.37 ± 0.02 g L?1 h?1. Use of glucose plus 3 g L?1 CaCO3 resulted in the production of 57.39 ± 1.41 g L?1 ethanol under micro-aerophilic conditions. These results indicate that supplementation of cellulosic sugars in the fermentation medium with CaCO3 and CaCl2 would improve economics of ethanol production from agricultural residues.  相似文献

Carbon distribution and kinetics of Candida shehatae were studied in fed-batch fermentation with xylose or glucose (separately) as the carbon source in mineral medium. The fermentations were carried out in two phases, an aerobic phase dedicated to growth followed by an oxygen limitation phase dedicated to ethanol production. Oxygen limitation was quantified with an average specific oxygen uptake rate (OUR) varying between 0.30 and 2.48 mmolO2 g dry cell weight (DCW)?1 h?1, the maximum value before the aerobic shift. The relations among respiration, growth, ethanol production and polyol production were investigated. It appeared that ethanol was produced to provide energy, and polyols (arabitol, ribitol, glycerol and xylitol) were produced to reoxidize NADH from assimilatory reactions and from the co-factor imbalance of the two-first enzymatic steps of xylose uptake. Hence, to manage carbon flux to ethanol production, oxygen limitation was a major controlled parameter; an oxygen limitation corresponding to an average specific OUR of 1.19 mmolO2 g DCW?1 h?1 allowed maximization of the ethanol yield over xylose (0.327 g g?1), the average productivity (2.2 g l?1 h?1) and the ethanol final titer (48.81 g l?1). For glucose fermentation, the ethanol yield over glucose was the highest (0.411 g g?1) when the specific OUR was low, corresponding to an average specific OUR of 0.30 mmolO2 g DCW?1 h?1, whereas the average ethanol productivity and ethanol final titer reached the maximum values of 1.81 g l?1 h?1 and 54.19 g l?1 when the specific OUR was the highest.  相似文献

Mutants of Candida magnoliae NCIM 3470 were generated by treatment of ultra-violet radiations, ethyl methyl sulphonate and N-methyl-N′-nitro-N-nitrosoguanidine. Mutants with higher reductase activity were screened by means of 2,3,5-triphenyl tetrazolium chloride agar plate assay. Among the screened mutants, the mutant R9 produced maximum mannitol (i.e. 46 g l?1) in liquid fermentation medium containing 250 g l?1 glucose and hence was selected for further experiments. Preliminary optimization studies were carried out on shake-flask level which increased the mannitol production to 60 g l?1 in liquid fermentation medium containing 300 g l?1 glucose. A two-stage fermentation process comprising of growth phase and production phase was employed. During the growth phase, glucose was supplemented and aerobic conditions were maintained. Thereafter, the production phase was initiated by supplementing fructose and switching to anaerobic conditions by discontinuing aeration and decreasing the speed of agitation. The strategy of two-stage fermentation significantly enhanced the production of mannitol up to 240 g l?1, which is the highest among all fermentative production processes and corresponds to 81 % yield and 4 g l?1 h?1 productivity without formation of any by-product.  相似文献

For the design of a large field of vertical flat plate photobioreactors (PBRs), the effect of four design parameters—initial biomass concentration, optical path length, spacing, and orientation of PBRs—on the biochemical composition and productivity of Chlorella zofingiensis was investigated. A two-stage batch process was assumed in which inoculum is generated under nitrogen-sufficient conditions, followed by accumulation of lipids and carbohydrates in nitrogen-deplete conditions. For nitrogen-deplete conditions, productivity was the most sensitive to initial biomass concentration, as it affects the light availability to individual cells in the culture. An initial areal cell concentration of 50 g m?2 inoculated into 3.8-cm optical path PBR resulted in the maximum production of lipids (2.42?±?0.02 g m?2 day?1) and carbohydrates (3.23?±?0.21 g m?2 day?1). Productivity was less sensitive to optical path length. Optical path lengths of 4.8 and 8.4 cm resulted in similar areal productivities (biomass, carbohydrate, and lipid) that were 20 % higher than a 2.4-cm optical path length. Under nitrogen-sufficient conditions, biomass productivity was 48 % higher in PBRs facing north–south during the winter compared to east–west, but orientation had little influence on biomass productivity during the spring and summer despite large differences in insolation. An optimal spacing could not be determined based on growth alone because a tradeoff was observed in which volumetric and PBR productivity increased as space between PBRs increased, but land productivity decreased.  相似文献

The effects of redox potential used as a control parameter on the process of succinic acid production in batch cultures of Actinobacillus succinogenes NJ113 have been investigated. In batch fermentation, cell growth and metabolite distribution were changed with redox potential levels in the range of ?100 to ?450 mV. From the results, the ORP level of ?350 mV was preferable, which resulted in high succinic acid yield (1.28 mol mol?1), high succinic acid productivity (1.18 g L?1 h?1) and high mole ratio of succinic acid to acetic acid (2.02). The mechanism of redox potential regulation was discussed by metabolic flux analysis and the ratio of NADH/NAD+. We expected that redox potential can be used as a valuable parameter to monitor and control much more anaerobic fermentation production.  相似文献

A wild type strain A-101 of Y. lipolytica and its three acetate-negative mutants (Wratislavia 1.31, Wratislavia AWG7, and Wratislavia K1) were compared for the production of citric acid from glucose and glycerol (pure and crude) in batch cultures. The substrates were used either as single carbon sources or as mixtures of glucose and pure or crude glycerol. The kinetic parameters, i.e., the volumetric citric acid production rate and yield obtained in the study show that the Wratislavia 1.31 and Wratislavia AWG7 strains produced the highest amount of citric acid from glycerol, with a yield from 0.40 to 0.53 g g−1. This substrate was found to be a better carbon source for the biosynthesis of citric acid than glucose. The results obtained with the same strains have shown low content of isocitric acid and polyols, such as erythritol and mannitol. Y. lipolytica A-101 strain produced the highest amount of isocitric acid, from 13.8 to 21% isocitric acid in the sum of citric acids. However, the highest concentrations of erythritol were found in cultures with Y. lipolytica Wratislavia K1, from 18.1 to 30 g l−1, for glucose and pure glycerol, respectively.  相似文献

In these studies, butanol (acetone butanol ethanol or ABE) was produced from concentrated lactose/whey permeate containing 211 g L?1 lactose. Fermentation of such a highly concentrated lactose solution was possible due to simultaneous product removal using a pervaporation membrane. In this system, a productivity of 0.43 g L?1 h?1 was obtained which is 307 % of that achieved in a non-product removal batch reactor (0.14 g L?1 h?1) where approximately 60 g L?1 whey permeate lactose was fermented. The productivity obtained in this system is much higher than that achieved in other product removal systems (perstraction 0.21 g L?1 h?1 and gas stripping 0.32 g L?1 h?1). This membrane was also used to concentrate butanol from approximately 2.50 g L?1 in the reactor to 755 g L?1. Using this membrane, ABE selectivities and fluxes of 24.4–44.3 and 0.57–4.05 g m?2 h?1 were obtained, respectively. Pervaporation restricts removal of water from the reaction mixture thus requiring significantly less energy for product recovery when compared to gas stripping.  相似文献

Microalgal starch is a potential feedstock for biofuel production. Nutrient stress is widely used to stimulate starch accumulation in microalgae. Cell growth and starch accumulation in the marine green microalga Tetraselmis subcordiformis were evaluated under extracellular phosphorus deprivation with initial cell densities (ICD) of 1.5, 3.0, 6.0, and 9.0?×?106 cells mL?1. The intracellular stored phosphorus supported cell growth when extracellular phosphorus was absent. The maximum starch content of 44.1 % was achieved in the lowest ICD culture, while the maximum biomass productivity of 0.71 g L?1 day?1, starch concentration of 1.6 g L?1, and starch productivity of 0.30 g L?1 day?1 were all obtained in the culture with the ICD of 3.0?×?106 cells mL?1. Appropriate ICD could be used to regulate the intracellular phosphorus concentration and maintain adequate photosynthetic activity to achieve the highest starch productivity, along with biomass and starch concentration. The recovery of phosphorus-deprived T. subcordiformis in medium containing 0.5, 1.0, or 6.0 mM KH2PO4 was also tested. Cell growth and starch accumulation ability could be recovered completely. A phosphorus pool in T. subcordiformis was shown to manipulate its metabolic activity under different environmental phosphorus availability. Though lower starch productivity and starch content were achieved under phosphorus deprivation compared with nitrogen- or sulfur-deprived conditions, the higher biomass and starch concentration make T. subcordiformis a good candidate for biomass and starch production under extracellular phosphorus deprivation.  相似文献

Photosynthetic bacteria are known to utilize volatile fatty acids as a carbon source for growth and product formation. In this study, a new isolate, Rubrivivax benzoatilyticus PS-5, possessing self-flocculation properties, was cultivated in modified glutamate-malate (GM) medium containing glutamate and malate as carbon sources. The effect of acetic acid, propionic acid and butyric acid (at 1–4 g L?1) as co-substrates and 7.5 mM glycine, 10 mM succinic acid as precursors for 5-aminolevulinic acid (ALA) production from R. benzoatilyticus PS-5 was investigated. Among the volatile fatty acids tested, acetic acid was preferred to butyric acid and propionic acid, with the optimum concentrations of 3 g L?1, 1 g L?1 and 3 g L?1, respectively. The highest ALA production was 169.71 μM, 162.16 μM and 46.18 μM, respectively, while the highest productivity was 2.57 μM h?1, 2.25 μM h?1 and 0.96 μM h?1, respectively. The precursor was consumed completely (100 %) while the assimilation of the acetic acid and butyric acid was 62.50 % and 48.65 %, respectively. Supplementation of propionic acid (at 1–4 g l?1) had a negative effect on growth and ALA production. To increase production efficiency, the pH-control strategy (at pH 6.0–8.0) during fermentation was tested. The optimum pH was 7.0, giving the maximum ALA production of 286.18 μM and a productivity of 3.97 μM h?1. These values were 1.68-fold and 1.54-fold higher, respectively, than those under uncontrolled pH conditions.  相似文献

The fermentation of both glucose and xylose is important to maximize ethanol yield from renewable biomass feedstocks. In this article, we analyze growth, sugar consumption, and ethanol formation by the yeast Kluyveromyces marxianus UFV-3 using various glucose and xylose concentrations and also under conditions of reduced respiratory activity. In almost all the conditions analyzed, glucose repressed xylose assimilation and xylose consumption began after glucose had been exhausted. A remarkable difference was observed when mixtures of 5 g L?1 glucose/20 g L?1 xylose and 20 g L?1 glucose/20 g L?1 xylose were used. In the former, the xylose consumption began immediately after the glucose depletion. Indeed, there was no striking diauxic phase, as observed in the latter condition, in which there was an interval of 30 h between glucose depletion and the beginning of xylose consumption. Ethanol production was always higher in a mixture of glucose and xylose than in glucose alone. The highest ethanol concentration (8.65 g L?1) and cell mass concentration (4.42 g L?1) were achieved after 8 and 74 h, respectively, in a mixture of 20 g L?1 glucose/20 g L?1 xylose. When inhibitors of respiration were added to the medium, glucose repression of xylose consumption was alleviated completely and K. marxianus was able to consume xylose and glucose simultaneously.  相似文献

Arabic date is overproduced in Arabic countries such as Saudi Arabia and Iraq and is mostly composed of sugars (70–80 wt%). Here we developed a fed-batch fermentation process by using a kinetic model for the efficient production of lactic acid to a high concentration from Arabic date juice. First, a kinetic model of Lactobacillus rhamnosus grown on date juice in batch fermentation was constructed in EXCEL so that the estimation of parameters and simulation of the model can be easily performed. Then, several fed-batch fermentations were conducted by employing different feeding strategies including pulsed feeding, exponential feeding, and modified exponential feeding. Based on the results of fed-batch fermentations, the kinetic model for fed-batch fermentation was also developed. This new model was used to perform feed-forward controlled fed-batch fermentation, which resulted in the production of 171.79 g l?1 of lactic acid with the productivity and yield of 1.58 and 0.87 g l?1 h?1, respectively.  相似文献

An annular internally illuminated photobioreatcor (IIPBR) configuration based on the airlift/bubble column principles was developed and validated at an 18 l prototype scale using Scenedemus sp. and Nannochloropsis salina in batch and semi-continuous modes, at constant light supply and constant gas-to-culture volume ratio, but at varying CO2-to-air ratios. Highest biomass production was recorded at CO2-to-air ratio of 4% with Scenedesmus sp. and at 1% with Nannochloropsis salina. The energetic performance of this IIPBR was quantified in terms of biomass productivity per unit energy input, P/E (g W?1 day?1), considering energy input for illumination and for pneumatic mixing and circulation. Under optimal conditions, the IIPBR evaluated in this study achieved P/E of 1.42 g W?1 day?1 for Scenedesmus sp. and P/E of 0.34 g W?1 day?1 for Nannochloropsis salina. These P/E values are better than those estimated for airlift and bubble column photobioreactor configurations reported in the literature.  相似文献
