Puccinia striiformis f. sp. tritici (PST), a clonal basidiomycete causing yellow rust disease on wheat, has a long record of 'overcoming' cultivar resistance introduced by breeders. Despite the long dispersal capacity of its spores, the French population of PST presents a strong geographical structure, with the presence of a specific pathotype (array of avirulence genes) at high frequencies in the south of France. The genetic diversity underlying this differentiation was analysed by microsatellite and AFLP markers. A total of 213 French isolates belonging to 10 pathotypes collected over a 15-year period were investigated. For each of the 12 microsatellites used, polymorphism resulted from a unique allelic variant associated to the south-specific pathotype. This pathotype was characterized by 40 specific markers over the total of 63 polymorphins detected using 15 AFLP primer combinations. Phylogeographical analysis indicated a strictly clonal structure of the population, and a strong genomic divergence between the northern population and a south-specific clone. Both virulence and molecular data show that the northern French population belongs to the northwestern European population, whereas the southern clone is most likely related to a Mediterranean population, the two subpopulations resulting from the ancient divergence of two clonal lineages. While the virulence complexity in the northern population may be explained by the successive introduction of corresponding resistance genes in cultivars, the maintenance of a simple virulence type in southern France, despite gene flow between the two populations, may be explained in terms of host cultivars repartition and local adaptation to specific host or climatic conditions.  相似文献

The effects of evolutionary processes in fungal pathogen populations may occur more rapidly and display larger effects in agricultural systems than in wild ecosystems because of human involvement by plant breeding and crop management. In this study, we analysed the rate of evolution in three lineages of a northwest European population of a biotrophic and asexual reproduced fungal pathogen, Puccinia striiformis f. sp. tritici, causing yellow rust on wheat. Pathogen samples were collected between 1975 and 2002 in the UK and Denmark, and assayed for 14 individual avirulence/virulence alleles and up to 234 amplified fragment length polymorphism (AFLP) primer pairs producing approximately 17,000 AFLP fragments. The large number of fragments and a targeted sampling of isolates allowed a reconstruction of phylogenies in great detail, i.e. no homoplasy and a representation of sequential, evolutionary steps by pathogen samples. A recent, phenotypic loss of avirulence was observed at least once for loci corresponding to P. striiformis f. sp. tritici resistance Yr2, Yr3, Yr4, Yr7, Yr9, and Yr15, whereas Avr6 and Avr17 were lost independently in all three lineages, corresponding to 16 events of loss of avirulence (emergence of virulence). The opposite process, restoration of avirulence, was observed for Yr9 and Yr32. An interpretation of phenotypic changes within lineages as independent mutation events resulted in mutation frequencies from 1.4x10(-6) to 4.1x10(-6) per AFLP fragment (locus) per generation, whereas the effective rate by which a mutation from avirulence to virulence was established in the pathogen population, when subject to selection by host resistance genes, was approximately three orders of magnitude faster.  相似文献

Most plant pathogens present complex life cycles, in which the clonal reproduction may impede the delimitation of population entities. By studying the genetic structure of the wheat yellow rust caused by Puccinia striiformis f.sp. tritici (PST), we highlighted difficulties impeding population delimitation in highly clonal species. Despite the high dispersal potential of PST, southern France isolates were shown to be divergent from a northwestern European population. A 2-year survey was performed in the Mediterranean region to assess the geographic distribution of southern isolates: 453 isolates collected in 11 countries were genotyped using 15 simple sequence repeat markers. A subsample was analysed for virulence against 23 resistance genes. The dominant strain in the western Mediterranean region was further studied with amplified fragment length polymorphism markers to test for a geographic substructure. Both 'individual'- and 'population'-centred analyses of polymorphism markers revealed two south-specific groups: a predominant group, with a broad variability and a wide distribution in both western and eastern Mediterranean countries, and a minor group in the western Mediterranean. The east–west gradient of genetic diversity suggested gene flow from the Middle East with subsequent founder effects and genetic divergence, and demonstrated the local survival of a western Mediterranean population. The high frequency of the resistance gene Yr8 observed in cultivars from Tunisia and Algeria may contribute to maintain the north/south structure observed in France. In addition to migration and local adaptation, the dynamics of clonal lineage diversification and replacement should be considered to define population entities in strongly clonal species.  相似文献

利用TP-M13-SSR自动荧光检测技术,对四川省小麦条锈菌群体遗传多样性水平进行了分析。研究结果表明,四川省小麦条锈菌群体遗传多样性比较丰富,地区之间存在明显的差异,川西北和四川盆地的种群遗传多样性相对较高,而四川西南部和四川东南部的种群遗传多样性较低。四川小麦条锈菌群体存在一定的遗传分化,地区间的遗传变异仅占14.92%,群体间的遗传变异占总变异的23.06%,群体内遗传变异占60.02%,遗传变异主要存在于群体内部。基因流和共享基因型从分子水平证实了四川小麦条锈菌在地区间的传播,且川西北和四川盆地之间的菌源交流最为广泛。  相似文献

Stripe rust (yellow rust) of wheat, caused by Puccinia striiformis f. sp. tritici, is one of the most important diseases in both China and the United States. The Chinese and US populations of the stripe rust fungus were compared for their virulence phenotypes on wheat cultivars used to differentiate races of the pathogen in China and the US and molecular genotypes using simple sequence repeat (SSR) markers. From 86 Chinese isolates, 54 races were identified based on reactions on the 17 Chinese differentials and 52 races were identified based on the 20 US differentials. The selected 51 US isolates, representing 50 races based on the US differentials, were identified as 41 races using the Chinese differentials. A total of 132 virulence phenotypes were identified from the 137 isolates based on reactions on both Chinese and US differentials. None of the isolates from the two countries had identical virulence phenotypes on both sets of differentials. From the 137 isolates, SSR markers identified 102 genotypes, of which 71 from China and 31 from the US. The virulence data clustered the 137 isolates into 20 virulence groups (VGs) and the marker data clustered the isolates into seven molecular groups (MGs). Virulence and SSR data had a low (r = 0.34), but significant (P = 0.01) correlation. Principal component analyses using either the virulence data or the SSR data separated the isolates into three groups: group a consisting of only Chinese isolates, group b consisting of both Chinese and US isolates and group c consisting of mostly US isolates. A neighbour-joining tree generated using the molecular data suggested that the P. striiformis f. sp. tritici populations of China and the US in general evolved independently.  相似文献

小麦条锈病是世界范围内小麦上最重要的流行性病害之一,可造成严重的产量损失。陇南地区是我国小麦条锈菌主要越夏易变区和新小种发源地,了解该地区不同海拔高度区域内条锈菌遗传多样性有重要意义。本研究采用TP-M13-SSR荧光标记技术对11个种群330个小麦条锈菌分离株基因组DNA进行了SSR标记分析。不同海拔区域的条锈菌遗传多样性有明显的差异,高山区的遗传多样性比较丰富,半山区次之,川道区相对比较低。不同生态区域内,小麦条锈菌群体遗传分化程度不同,高山区和半山区遗传分化程度大,基因流小,川道区群体遗传分化程度比较小,基因流大。来自不同海拔区域的菌系具有相同的基因型,这一结果从分子水平证明了在陇南地区小麦条锈菌在山区与川地之间存在广泛的菌源交流,可就地完成周年循环。  相似文献

甘肃天水地区小麦条锈菌自然群体DNA指纹分析   总被引:1,自引:0,他引:1  
对1997—1999连续三年采自甘肃省天水地区16个地点的244个小麦条锈菌分离系进行了PSR331S3/Bg1Ⅱ指纹分析,共鉴定出表现型185个,遗传多样性Shannon指数M=4.467, 加权修正值M*=0.9106,显现出高度的遗传多样性水平;其中三个主要采样点凤凰、甘谷和麦积山病菌群体M值分别为3.5720、3.0268和3.4186,加权修正值M*分别为0.9330、0.8513和0.8610。三个群体之间的遗传分化系数GST=0.03167,显示出较低的遗传分化水平。本文以多拷贝的RFLP标记为手段,揭示了小麦条锈菌作为一种活体寄生的远程气传病原真菌,其越夏栖息地关键区(天水地区)自然群体存在丰富的遗传多样性,不同海拔生境对病菌群体结构有显著的影响。但是,地区内亚群体间分化水平很低,表明区域内菌源交流十分频繁。  相似文献

Rapid global spread of two aggressive strains of a wheat rust fungus   总被引:2,自引:1,他引:1  
Rust fungi can overcome the effect of host resistance genes rapidly, and spores can disperse long distance by wind. Here we demonstrate a foreign incursion of similar strains of the wheat yellow rust fungus, Puccinia striiformis f. sp. tritici , in North America, Australia and Europe in less than 3 years. One strain defined by identity at 15 virulence loci and 130 amplified fragment length polymorphism (AFLP) fragments was exclusive to North America (present since 2000) and Australia (since 2002). Another strain of the same virulence phenotype, but differing in two AFLP fragments, was exclusive to Europe (present since 2000–2001) as well as Western and Central Asia and the Red Sea Area (first appearance unknown). This may be the most rapid spread of an important crop pathogen on the global scale. The limited divergence between the two strains and their derivatives, and the temporal–spatial occurrence pattern confirmed a recent spread. The data gave evidence for additional intercontinental dispersal events in the past, that is, many isolates sampled before 2000 in Europe, North America and Australia had similar AFLP fingerprints, and isolates from South Africa, which showed no divergence in AFLP, differed by only two fragments from particular isolates from Central Asia, West Asia and South Europe, respectively. Previous research has demonstrated that isolates of the two new strains produced up to two to three times more spores per day than strains found in USA and Europe before 2000, suggesting that increased aggressiveness at this level may accelerate global spread of crop pathogens.  相似文献

The fungus Puccinia striiformis f. sp. tritici, the causal agent of wheat stripe rust, is an obligate biotrophic basidiomycete. Urediniospores are the most common spore type involved in the epidemiology of this disease. Tip growth of germ tubes of germinated urediniospores is a key step during infection of wheat, but few studies have investigated it so far. Recent research has found that actin is closely associated with hyphal tip growth. In this study, we have cloned and obtained the full-length actin cDNA from P. striiformis f. sp. tritici and characterized its expression. Furthermore, actin filament (F-actin) patterns were visualized microscopically during germ tube formation. The most conspicuous actin-containing structures were actin patches. They were mainly concentrated near the hyphal tip and scattered throughout the cortex. By using cytochalasin B, we observed that depolymerization of F-actin greatly reduced the germination rate of urediniospores and disrupted the transport of vesicles to the germ tube tip, indicating that F-actin played a key role in the tip growth of P. striiformis f. sp. tritici. This work helps us to understand the tip growth mechanism of P. striiformis f. sp. tritici, and may provide a theoretical framework for designing novel pesticides.  相似文献

用萌发的小麦条锈菌冬孢子接种采自陕西省境内的陕西小檗、少齿小檗和长穗小檗,3种小檗均产生了性孢子器和锈孢子器。用人工接种小麦条锈菌冬孢子在陕西小檗上产生的锈孢子器接种小麦铭贤169产生了典型的条锈菌夏孢子堆症状。特异性PCR和DNA序列分析表明,人工接种产生于小檗上的锈孢子、接种锈孢子于小麦上产生的夏孢子堆与小麦条锈菌DNA的ITS区序列完全一致。更为重要的是,用采自田间受锈菌侵染的小檗叶片产生的锈孢子接种小麦铭贤169,经培养在小麦铭贤169叶片上产生了典型的条锈病症状。从而证实,在自然条件下,在中国,小檗不仅可作为小麦条锈菌的转主寄主,而且小麦条锈菌可在小檗上完成其有性繁殖过程。这一发现对进一步揭示我国小麦条锈菌高度的群体遗传多样性与毒性变异机理、完善小麦条锈病的防治策略具有十分重要的理论和实际意义。  相似文献

为了解陇南地区越夏自生麦苗上条形柄锈菌的毒性组成及群体遗传结构,将采自天水、陇南及定西的自生麦苗上的39个单孢子堆菌系采用中、美鉴别寄主毒性分析法和TP-M13-SSR荧光标记技术,进行毒性鉴定并对其基因组DNA进行SSR标记分析。结果显示:在中国鉴别寄主上供试菌系被区分为9个致病类型,在美国鉴别寄主上则得到24个毒性类型,在中美鉴别寄主上共得到30个毒性表型。通过中国鉴别寄主鉴定的CYR32、CYR33为优势小种,毒性比率分别达到35.9%和30.8%。SSR标记将这些菌系划分为36个基因型,毒性分析与  相似文献

Pyrenophora teres f. sp. teres mating-type genes (MAT-1: 1190 bp; MAT-2: 1055 bp) have been identified. Their predicted proteins, measuring 379 and 333 amino acids, respectively, are similar to those of other Pleosporales, such as Pleospora sp., Cochliobolus sp., Alternaria alternata, Leptosphaeria maculans, and Phaeosphaeria nodorum. The structure of the MAT locus is discussed in comparison with those of other fungi. A mating-type PCR assay has also been developed; with this assay we have analyzed 150 isolates that were collected from 6 Sardinian barley landrace populations. Of these, 68 were P. teres f. sp. teres (net form; NF) and 82 were P. teres f. sp. maculata (spot form; SF). Within each mating type, the NF and SF amplification products were of the same length and were highly similar in sequence. The 2 mating types were present in both the NF and the SF populations at the field level, indicating that they have all maintained the potential for sexual reproduction. Despite the 2 forms being sympatric in 5 fields, no intermediate isolates were detected with amplified fragment length polymorphism (AFLP) analysis. These results suggest that the 2 forms are genetically isolated under the field conditions. In all of the samples of P. teres, the ratio of the 2 mating types was consistently in accord with the 1:1 null hypothesis. This ratio is expected when segregation distortion and clonal selection among mating types are absent or asexual reproduction is rare. Overall, sexual reproduction appears to be the major process that equalizes the frequencies of the 2 mating types within populations.  相似文献

研究夏孢子萌发过程的分子机制对于从分子水平上理解条锈菌的侵染过程及其与寄主互作的关系具有重要的理论意义。本研究以小麦条锈菌Pucciniastriiformisf.sptritici32号生理小种(CY32)为材料,研究了用水培方法萌发夏孢子的适宜条件。结果显示,CY32夏孢子萌发的最适温度为9℃,最适宜的孢子量是6mg/200mL水,适宜的溶液是无菌蒸馏水。水化能促进夏孢子的萌发,新鲜夏孢子和干燥容器中放置2d的夏孢子经水化15h后,萌发率显著提高。此方法获得的萌发夏孢子提取的RNA可以满足cDNA文库构建和基因表达分析等分子生物学研究的要求,并为小麦条锈菌的分子生物学研究奠定了物质基础。  相似文献

Tan spot, a foliar disease of wheat, is caused by the fungus Pyrenophora tritici‐repentis. On susceptible wheat cultivars, P. tritici‐repentis induces two distinct symptoms: tan necrosis and extensive chlorosis. Presently isolates of P. tritici‐repentis are classified into 11 races based on their virulence on a set of wheat differential genotypes. In nature, this pathogen reproduces both sexually and asexually, but the extent of genetic variability in the P. tritici‐repentis population of western Canada is unknown. This study was conducted to assess the genetic variability among different isolates of P. tritici‐repentis and to determine if similarities among isolates are correlated with race classification or geographic origin of the isolates. Thirty‐three isolates of P. tritici‐repentis and one isolate each of P. teres f. sp. teres, P. teres f. sp. maculata, P. graminea, Helminthosporium sativum and an uncharacterized isolate were studied with 30 random amplified polymorphic DNA (RAPD) primers. Cluster analysis showed that all isolates had unique banding patterns and that clustering of isolates was independent of their race designation or geographic origin. Analysis of molecular variation (amova ) showed that 96.8% of variability occurred among isolates and among race variability accounted for only 3.2% of the total variability.  相似文献

选用2套遗传背景不同的抗条锈病近等基因系作为供试寄主材料,研究了不同抗条锈病近等基因系丁布的含量及其在感病过程中丁布含量的动态变化.结果表明,在未受病菌侵染情况下,2套分别含有Yr2、Yr9和YrSpP基因的近等基因系(抗病系)与其轮回亲本Taichung 29、铭贤169(感病系)间丁布含量没有显著差异(P>0.05).接种条锈病菌后,感病系在病菌侵染初期丁布含量下降,而抗病系在病菌侵染初期丁布含量迅速大幅度上升.感染条锈病最终导致感病植株丁布含量比未接种的植株明显减少, 感病系的减少幅度明显高于抗病系.在整个病程中,抗病系丁布的含量始终高于感病系,表明接种条件下小麦植株体内丁布含量变化与小麦抗条锈近等基因系的抗性有关.  相似文献

Vast amounts of effort have been devoted to investigate patterns of genetic diversity and structuring in plants and animals, but similar information is scarce for organisms of other kingdoms. The study of the genetic structure of natural populations of wild yeasts can provide insights into the ecological and genetic correlates of clonality, and into the generality of recent hypotheses postulating that microbial populations lack the potential for genetic divergence and allopatric speciation. Ninety‐one isolates of the flower‐living yeast Metschnikowia gruessii from southeastern Spain were DNA fingerprinted using amplified fragment length polymorphism (AFLP) markers. Genetic diversity and structuring was investigated with band‐based methods and model‐ and nonmodel‐based clustering. Linkage disequilibrium tests were used to assess reproduction mode. Microsite‐dependent, diversifying selection was tested by comparing genetic characteristics of isolates from bumble bee vectors and different floral microsites. AFLP polymorphism (91%) and genotypic diversity were very high. Genetic diversity was spatially structured, as shown by amova (Φst = 0.155) and clustering. The null hypothesis of random mating was rejected, clonality seeming the prevailing reproductive mode in the populations studied. Genetic diversity of isolates declined from bumble bee mouthparts to floral microsites, and frequency of five AFLP markers varied significantly across floral microsites, thus supporting the hypothesis of diversifying selection on clonal lineages. Wild populations of clonal fungal microbes can exhibit levels of genetic diversity and spatial structuring that are not singularly different from those shown by sexually reproducing plants or animals. Microsite‐dependent, divergent selection can maintain high local and regional genetic diversity in microbial populations despite extensive clonality.  相似文献
