Plasma transferrin (Tf) types in dogs were studied by a method of horizontal polyacrylamide gel electrophoresis with a 10 % separation gel and a discontinuous buffer system (Tris-citrate-borate, pH 9.0). Samples were analysed from 1127 dogs belonging to 60 different breeds. TfB and Tfc alleles, each in considerably high frequency, were observed in most of the breeds. TfA with relatively lower frequency was observed in Beagle, Cocker Spaniel, Doberman Pinscher, German Shepherd, German Shorthaired Pointer, Old Danish Pointer and Poodle. Two rare Tf alleles, TfD and the other designated as TfE, were observed only in Cocker Spaniel and in Poodle respectively. Basenji (44 samples) and Small Münsterlander (17 samples) showed the highest frequency (0.97) for TfC allele while Carelian Bear Dog (19 samples) showed the highest frequency (0.97) for TfB allele. In the total material, the frequencies of Tf alleles A, B, C, D and E were found to be 0.0182, 0.5075, 0.4716, 0.0009 and 0.0018 respectively. By using the observed and expected numbers of Tf heterozygotes, the average inbreeding coefficient (F) within breeds was estimated to be 0.14. Six samples of wolf (Canis lupus L.) studied seemed to be of Tf B type. The present study, along with earlier reports, showed that some of the plasma proteins and enzymes (albumin, transferrin, eserine resistant esterase) exhibit considerably more polymorphism than that reported for hemoglobin and some of the red cell and tissue enzymes in the domestic dog.  相似文献

The frequencies of three co-dominant alleles Tf A, Tf B and Tf c controlling the serum transferrin locus in native and exotic breeds of pig in the United Kingdom are shown. Matings between different transferrin genotypes and segregation of the transferrin alleles in piglets from 131 litters are also recorded.
In eleven litters from matings within a closely related group of animals heterozygous for the Tf c allele, no offspring of the Tf c/ Tf c genotype were born. Matings between heterozygous related boars believed to be carrying a lethal factor linked to the Tf locus and heterozygous unrelated sows of the Saddleback breed, resulted in offspring of the Tf c/ Tf c genotype being born.
An excess of heterozygote genotypes was found in all litters from matings between animals heterozygous for the Tf c allele. In all these litters the average litter size was 0.5 piglets less per litter than the average for all other litters studied. For the eleven litters from matings involving closely related animals heterozygous for the Tf c allele, the average litter size was 1.4 piglets less per litter than the average for all other litters.
The possibility of an early lethal factor being linked to the Tf locus in this family of pigs is discussed.  相似文献

Serum samples from 412 Spanish wild rabbits were analysed by starch and polyacrylamide gel electrophoresis. Three different transferrin (Tf) phenotypes (A, AB and B) were observed by both methods. The occurrence of two codominant alleles (TfA and TfB with frequencies of 0.89 and 0.11 respectively) at an autosomal locus (Tf) was supported by the population data on genetic equilibrium. Electrophoretic mobility differences between the Tf variants A and B could not be explained by differences in sialic acid or iron contents. Each of the two Tf variants were shown to have two sialic acid residues by neuraminidase treatment. These variants had similar affinities for iron, and iron binding did not lead to the conversion of one variant into the other.  相似文献

对我国11个地方鸡种1087羽鸡群进行了血型和血浆蛋白质多态7个位点22个等位基因的群体遗传学分析。结果表明,血型A、C位点,基因频率各鸡种存在显著差异,B位点则没有显著差异,血型和血浆蛋白质多态位点基因纯合系数普遍较低,血型因子分布在群体中较为分散,表明这些地方品种存在较大的选择潜力。血浆蛋白质多态位点,中国与外国的某些鸡种比较,存在着较大的相同或相异,可能意味着我国和外国鸡种起源上的同缘或品种起源、进化上的多样性。聚类分析表明,这11个地方鸡种可系统聚类成4个类群,其结果与目前人们对这些鸡种的表征分类不尽一致。  相似文献

In starch gel electrophoresis of horse sera each transferrin variant is formed by a strong anodal band and a weaker cathodal band. An 'atypical' variant, Tf C, has two zones of about equal intensity. Family data show that Tf C is genetically controlled by an allele Tf C at the Tf locus. Frequencies of transferrin alleles in various horse breeds are also presented.
After isolation and fractionation of individual transferrin variants (Tf O, Tf D, Tf C) on DEAE-Sephad Summary ex, additional weak bands were detected. The two main zones of each variant were isolated in a pure state and treated with neuraminidase. In all three variants studied the electrophoretic mobility of the slower band (2a) was decreased in two steps, and the faster band (4b) in four steps. The mobilities of bands derived from the fast zone (4b) were slower than mobilities of corresponding bands derived from the slow zone (2a). These results suggest the presence of two sialic acid residues in the slow zone, and of four residues in the fast zone. Residual heterogeneity was independent of sialic acid.  相似文献

Transferrin types in the Japanese quail Coturnix coturnix japonica are controlled by a single autosomal locus Tf with at least two codominant alleles TfB and Tfc. The frequencies of TfB and Tfc in a commercial population of the domestic quail were 1.00 and 0.00, respectively.
Previous studies demonstrated that in the domestic populations of the Japanese quail three electrophoretic patterns AB, B and BC existed in egg white conalbumin and that the electrophoretic variation of conalbumin occurred in parallel with that of serum transferrin. Furthermore, from the preliminary mating experiments the transferrin-conalbumin variation was proposed to be under the control of at least two codominant alleles 77s and Tfc at an autosomal locus (Kimura et al., 1977, 1978).
The present study was designed (1) to report a large amount of family data from three generations in order to support the previously published hypothesis on genetic control of electrophoretic patterns of transferrin, and (2) to survey the gene constitution of transferrin in a commercial population of the domestic quail.
Sera were added with iron, heated for 5 min at 65°C (Stratil, 1967), then were investigated by means of horizontal starch gel electrophoresis. Hydrolysed starch from Connaught Medical Laboratories, Toronto, was used. A discontinuous citrate/Tris/LiOH/borate buffer system (pH 8.0) of Ferguson & Wallace (1961) was employed. Under the name of Creole goats, individuals of various origins, morphological and productivity characters are grouped. To characterize the genetic pool of goats from different sources in central Argentina, protein polymorphism revealed by electrophoresis was analyzed. A total of 109 adult animals from Santa Marı́a, Colón and Ischilı́n Districts, Córdoba Province, were studied. The red cells lysates were subjected to starch gel electrophoresis and specific staining procedures to reveal the activity of different enzymes and hemoglobin. Separation of plasmatic proteins was carried out by electrophoresis in polyacrylamide gel and stained with Coomassie Blue. From a total of 14 loci analyzed, eight were polymorphic in at least one flock. Four codominant alleles were detected in the locus βHemoglobin (βHb); three alleles in malic enzyme (Me), glucose 6 phosphate dehydrogenase (G6pdh) and lactate dehydrogenase (Ldh) and two alleles in catalase (Cat), transferrin (Tf), esterase-2 (Es-2) and phosphoglucomutase (Pgm). In all the cases, the observed genotype frequencies were not significantly different from those expected from Hardy–Weinberg equilibrium. The proportion of polymorphic loci (P%) varied between 21.4% and 42.9% and mean heterozygosity (H), between 0.061 and 0.117. Alleles at loci Cat, Me and Es-2 showed significantly different frequencies according to the sample origin. The flock from Colón District presented the highest levels of polymorphism. This is the only stock which includes hybrids of varying grade between Creole stocks and male specimens from Saanen and Nubian breeds.  相似文献

Allozyme electrophoresis (horizontal starch gel and PAGE) and histochemical staining techniques were used to study the genetic composition of an endemic southern African domestic dogCanis familiaris Linnaeus, 1758, the Africanis breed. Genetic differentiation was analysed at 21 protein-coding loci. The results were compared to those for three other populations/breeds: representatives of established Western breeds, crossbred dogs of Western descent from rural areas in South Africa, and indigenous Saluki dogs from the Middle East. Nine polymorphic loci were found (Ak-1,-2, Ck, Per, Hb, Po-A-1 to-3 andPo-Tf). Two unique alleles at theCk andPo-A-2 loci separated the Africanis breed from the other groups. There were also significant differences between Africanis and the other breeds in pair-wise comparisons of allelic frequencies at polymorphic loci. An assignment test, fixation index values, gene flow and genetic distance values indicated a closer genetic association between the Africanis and Saluki breeds than with dogs of Western origin. This finding supports archaeological evidence that the endemic Africanis breed was introduced from the Middle East into Africa thousands of years ago, and not through later western influences. The average heterozygosity ranged from 0.106–0.15, with least heterozygosity in the Africanis and most in the rural crossbred group. The percentage of polymorphic loci, the mean number of alleles per locus (biologically more significant than heterozygosity), and conformation of genotypes to Hardy-Weinberg proportions showed no evidence of recent loss of genetic diversity in Africanis. Genetic differentiation and support of archaeological evidence by genetics indicate that the endemic southern African domestic dog breed is unique.  相似文献

The gene pool formation of the modern domestic breeds of pig Sus scrofa domestica and their genesis based on hybridization of wild ancestral forms of the European and Asian origin were studied using molecular immunogenetic methods. Males of the European and Central Asian S. scrofa subspecies (S. s. scrofa and S. s. nigripes were hybridized with domestic pigs of the Swedish Landrace and Vietnamese Black Masked breeds. In addition, we examined the genotypic structure of 65 wild, aboriginal, and local populations as well as cultured breeds, including the stock breeds with different levels of selection. Frequencies of alleles and suballelles of the chromosome 4 locus controlling antigens of the L blood group system were analyzed. The origin of marker suballeles of the European and Asian origin was estimated in the most widespread world pig breeds. Unexpectedly, a strikingly high frequency of the Asian elements was found in the most productive European and American breeds, as well as in the best breeds of Russia and other CIS countries. Only one form of heterozygosity (bcgi/bdfi) was found in a population of wild European ancestors, whereas domestic pig breeds displayed heterozygosity for far more numerous suballeles of the locus studied. On the basis of the examination of random samples of seven populations of reindeer inhabiting the Montane-Taiga Zone and of nine population inhabiting the Tundra Zone of the Magadan Region 8 alleles of transferrin locus described earlier (TfA, TfC, TfE, TfG, TfH, TfJ, TfK and TfM) were identified and the ninth hitherto unknown allele (TfN) was discovered. Gene concentrations of all these alleles in populations studies were determined. The reindeer of the North-East of Siberia differ in frequencies of Tf alleles from those of Norway and, apparently, also from the reindeer inhabiting the northern regions of the European part of the USSR and from the American caribou. The allele TfK is possibly associated with the resistance of reindeer to necrobacteriosis.  相似文献

Abstract

Samples from the feral and domestic sheep on Pitt Island were examined for parasites. The feral sheep had never been treated against parasites, and most were due for extermination. The domestic sheep were examined by way of comparison. The sheep ked (Melophagus ovinus) and the chorioptic mange mite (Chorioptes bovis) were found on feral but not domestic sheep. The sheep body louse (Damalinia ovis) was found on domestic but not feral sheep. Lung nodules and liver lesions, presumed to be caused by parasites, were significantly more prevalent in domestic than in feral sheep. The prevalence of Cysticercus tenuicollis cysts and Sarcocystis spp. macrocysts was similar in feral and domestic sheep. The species of intestinal nematode and their relative abundance in feral sheep were similar to those in mainland sheep. Fewer feral sheep were found to have antibodies to Toxoplasma gondii when compared with other breeds; the proportion with antibodies to Sarcocystis ovicanis differed little from that in the mainland flocks, however. The data are compared with information from the Campbell Island feral sheep flock.  相似文献

Transferrin phenotypes were determined in six breeds of sheep by starch gel electrophoresis. Two new variants, Tf HCzech and Tf KCzech, were found and some evidence of their genetic control was obtained. Tf HCzeeh was detected only in Sumava sheep; it has an intermediate mobility between Tf A and Tf B. Tf KCzech was found only in Tsigais; it was localized between Tf B and Tf C. The frequencies of corresponding alleles were very low.
Individual transferrin variants (I, A, HCzech, B, Kczech, C, D, E, and P) were treated with neuraminidase. Electrophoretic mobility of the strong band was decreased by two steps in each case. It suggests that in the strong Tf band two sialic acid residues are accessible to the enzyme.  相似文献

Blood plasma of 253 specimens from eight population samples of the sciaenid fish, pescada (Plagioscion squamosissimus), caught from four sites in the Central Amazon, was tested for molecular variants of transferrin. Starch gel electrophoresis was used to distinguish six species of transferrin molecules; 12 of the 21 theoretically possible genotypes were found. There were highly significant departures from genetic equilibrium in seven of the eight population samples (chi-square (chi(2)) test for Hardy-Weinberg expectations) due to an excess of homozygotes and a corresponding deficiency of heterozygotes. A dendrogram based on UPGMA cluster analysis of genetic distances at the transferrin gene locus, estimated among the population samples and statistical analyses of the distribution of Tf allele frequencies, indicated three genetically discreet sub-populations of P. squamosissimus. The three sub-populations, "Careiro/Iranduba", "Coari" and "Tefe", were found to have high frequencies of alleles Tf(2), Tf(4) and Tf(3), respectively. This genetic instability may be attributed to genetically discreet "allopatric stocklets", which diverged during past isolation.  相似文献

Five alleles with eight electrophoretic phenotypes of 6-phosphogluconate dehydrogenase were found in 1,195 blood samples from fourteen populations of nine macaque species.Macaca fascicularis from Malaya showed the most polymorphism, with three Pgd alleles resulting in five phenotypes.Macaca mulatta, M. speciosa, M. nemestrina, andM. cyclopis had two alleles each (although the last two species showed a high percentage of homozygosity). The remaining four species (M. fuscata, M. radiata, M. maura, andM. nigra) were homozygous for the Pgda allele. The predominance of Pgda was observed in all macaque species, exceptM. speciosa which showed a high (57%) frequency of Pgdd. The distinctive position ofM. speciosa with regard to 6PGD variants parallels observations that indicate that this species carries transferrin and carbonic anhydrase I alleles in different frequencies from those of the other macaque species. Other similarities between the patterns of transferrin and 6PGD variations include a tendency toward homozygosity at the Pgd locus in the insular macaque forms. The domestic horse (Equus caballus) was re-introduced to the Americas by Spanish explorers. Although horses from other parts of Europe were subsequently introduced, some New World populations maintain characteristics ascribed to their Spanish heritage. The southeastern United States has a history of Spanish invasion and settlement, and this influence on local feral horse populations includes two feral-recaptured breeds: the Florida Cracker and the Marsh Tacky, both of which are classified as Colonial Spanish horses. The feral Banker horses found on islands off the coast of North Carolina, which include, among others, the Shackleford Banks, the Corolla and the Ocracoke, are also Colonial Spanish horses. Herein we analyse 15 microsatellite loci from 532 feral and 2583 domestic horses in order to compare the genetic variation of these five Colonial Spanish Horse populations to 40 modern horse breeds. We find that the Corolla horse has very low heterozygosity and that both the Corolla and Ocracoke populations have a low mean number of alleles. We also find that the Florida Cracker population has a heterozygosity deficit. In addition, we find evidence of similarity of the Shackleford Banks, Marsh Tacky and Florida Cracker populations to New World Iberian horse breeds, while the origins of the other two populations are less clear.  相似文献

Invasive alien species are a significant threat to both economic and ecological systems. Identifying the processes that give rise to invasive populations is essential for implementing effective control strategies. We conducted an ancestry analysis of invasive feral swine (Sus scrofa, Linnaeus, 1758), a highly destructive ungulate that is widely distributed throughout the contiguous United States, to describe introduction pathways, sources of newly emergent populations and processes contributing to an ongoing invasion. Comparisons of high‐density single nucleotide polymorphism genotypes for 6,566 invasive feral swine to a comprehensive reference set of S. scrofa revealed that the vast majority of feral swine were of mixed ancestry, with dominant genetic associations to Western heritage breeds of domestic pig and European populations of wild boar. Further, the rapid expansion of invasive feral swine over the past 30 years was attributable to secondary introductions from established populations of admixed ancestry as opposed to direct introductions of domestic breeds or wild boar. Spatially widespread genetic associations of invasive feral swine to European wild boar deviated strongly from historical S. scrofa introduction pressure, which was largely restricted to domestic pigs with infrequent, localized wild boar releases. The deviation between historical introduction pressure and contemporary genetic ancestry suggests wild boar‐hybridization may contribute to differential fitness in the environment and heightened invasive potential for individuals of admixed domestic pig–wild boar ancestry.  相似文献

Summary. Transferrin variants were examined in population samples of fish, locally known as jaraquis, Semaprochilodus taeniurus and 5. insignis. Both species were sampled on two occasions from two geographical areas in the Amazon region. The two species showed no common transferrin type. The 122 specimens of S. taeniurus showed three genotypes encoded by two codominant alleles at the transferrin gene locus ( Tf ). The 69 specimens of S. insignis showed another two transferrin alleles. Hence the different Tf genotypes support their taxonomic differences, and are predictably capable of identifying any suspected hybrid species. No hybrid was found.  相似文献

Gene frequencies of coat colour and horn types were assessed in 22 Nordic cattle breeds in a project aimed at establishing genetic profiles of the breeds under study. The coat colour loci yielding information on genetic variation were: extension, agouti, spotting, brindle, dun dilution and colour sided. The polled locus was assessed for two alleles. A profound variation between breeds was observed in the frequencies of both colour and horn alleles, with the older breeds generally showing greater variation in observed colour, horn types and segregating alleles than the modern breeds. The correspondence between the present genetic distance matrix and previous molecular marker distance matrices was low (r = 0.08 – 0.12). The branching pattern of a neighbour-joining tree disagreed to some extent with the molecular data structure. The current data indicates that 70% of the total genetic variation could be explained by differences between the breeds, suggesting a much greater breed differentiation than typically found at protein and microsatellite loci. The marked differentiation of the cattle breeds and observed disagreements with the results from the previous molecular data in the topology of the phylogenetic trees are most likely a result of selection on phenotypic characters analysed in this study.  相似文献

Starch gel electrophoresis was performed to study polymorphism of hordeins encoded by the Hrd A, Hrd B, and Hrd F loci in 366 local old barley accessions from Iran and Central Asian countries, including Turkmenistan, Uzbekistan, Tajikistan (Mountain Badahsan), and Kirgizia. In total, 60 alleles with frequencies of 0.0003–0.2818 were observed for the Hrd A locus, 106 alleles with frequencies of 0.0003–0.1603 were observed for the Hrd B locus, and five alleles with frequencies of 0.0164–0.4131 were observed for the Hrd F locus. The alleles and allele frequencies displayed irregular distributions in barley populations of the above countries. Cluster analysis of the matrix of allele frequencies in populations from known collection sites revealed a cluster structure of local barley populations within each country. Local populations formed five differently sized clusters in Iran, six in Turkmenistan, three in Uzbekistan, and three in Kirgizia. The variation and allele frequency distribution of the hordein-coding loci in Iran and Central Asian countries were assumed to result from the introduction and spreading of barley forms via migrations of husbandmen.  相似文献
