Abstract

Cholecalciferol (vitamin D3) is widely used as a vertebrate pesticide in New Zealand. However, cholecalciferol also occurs naturally in animals. Therefore, when trying to determine whether a non-target animal has been exposed to cholecalciferol baits, knowledge of the baseline cholecalciferol concentrations in the animal's plasma and tissue is required. We analysed cattle, sheep, pig, deer, dog and cat plasma and liver samples for the vitamin D3 metabolite 25-hydroxycholecalciferol (25-OHD), a sensitive biomarker for cholecalciferol. Based on these data and a literature search we present 25-OHD reference ranges. We also examined the literature for 25-OHD concentrations in poisoned animals and compared these to the reference ranges. Where plasma and liver samples have 25-OHD concentrations at least four times higher than our reference ranges it is likely that the animal has been exposed to cholecalciferol baits. 25-OHD concentrations 10 times higher than the reference range indicate ingestion of abnormally high amounts of cholecalciferol.  相似文献

Abstract Two Australian cladocerans, Moina australiensis Sars and a species of Ceriodaphnia, were evaluated as possible biological indicator organisms to assess the toxicity of irrigation supply and drainage water of the Murrumbidgee and Coleambally Irrigation Areas. M. australiensis, being large (~2000 μm) and orange, was initially chosen to overcome visibility problems in highly turbid Australian inland waters. However, the organism responded erratically in culture. Mortality was high and neonate production was unpredictable when cultured under recommended United States Environmental Protection Agency protocols. Attempts to improve culture performance by optimizing food (quality and quantity), water (control source, hardness, volume) and temperature were only marginally successful. Similar difficulties were not evident when Ceriodaphnia sp. was used as the test organism. Although Ceriodaphnia sp. is small (~1000 μm), grey and more difficult to see in turbid water its responses were more predictable and reliable than those of M. australiensis. Results of initial trials comparing the two organisms suggest that Ceriodaphnia sp. was a better test organism and more suited to local requirements.  相似文献

Estuarine sediments frequently are repositories and therefore potential sources of anthropogenic contaminants. Many organic and metallic chemical compounds released into aquatic systems bind to particulates and so accumulate in the sediments, thus, sediments become repositories of contaminants in estuaries. These may also cause contamination through diffusion of porewater, resuspension of particulates and dispersal of benthic fauna. There is a need to assess the biological affects of these anthropogenic contaminants because they may be toxic to infauna and bottomfish. Sediment toxicity bioassays are a means for carrying out such an assessment and primarily provide data on toxicity by measuring the effects on the test organism. Existing sediment toxicity bioassays rely on a battery of aquatic toxicity tests, which are based on the extraction of pore water, and elutriate from sediments and then subjecting these sediment phases to toxicity testing regimes. Two estuarine bivalve molluscs, Scrobicularia plana and Tapes semidecussatus were used to assess the ecotoxicity of field-collected sediments from estuarine and coastal areas around the Irish and English Coast over a 3-year study period. A variety of endpoints were measured during the study including survival in air, behaviour, animal condition, biochemistry, soft tissue metal concentrations, lysosomal membrane integrity and histopathology. Of these endpoints, the most sensitive were survival, survival in air, lysosomal membrane integrity, behaviour and histopathology.  相似文献

In July, 1986 the Johns Hopkins University Applied Physics Laboratory's Aquatic Ecology Section established a bioassay facility for conducting an effluent biomonitoring program for the State of Maryland. Acute toxicity test procedures were developed and implemented for testing freshwater (Pimephales promelas, Daphnia magna and Ceriodaphnia sp.) and estuarine (Cyprinodon variegatus and Mysidopsis bahia) invertebrates and fish. Procedures and test species are similar to those used by the U.S. Environmental Protection Agency (EPA) except that low salinity testing ( 15) is conducted with the sheepshead minnow and an estuarine mysid found in the Chesapeake Bay.Results from the first year of the program involving acute screening bioassays of major industrial and municipal dischargers are presented. Over 90 dischargers were tested during the first full year of operation. The frequency of toxicity was 36% and 14% for industrial and municipal dischargers, respectively. Reference toxicity test results are also summarized. Results from these tests are presented by outfall type. A number of examples are presented concerning the use of these data for regulating toxic discharges by the State of Maryland's Department of the Environment. These results indicate the importance of implementing a biomonitoring program with proven, easily managed methods that can be readily understood by both regulators and permit holders.  相似文献

  • 1 To contribute to the development of IPM strategies in olive groves, the selectivity of several insecticides to Pullus mediterraneus Fabr. (Coleoptera: coccinellidae) was investigated in the laboratory. The study assessed the toxicity of seven chemical pesticides and one bacteriological insecticide to adult P. mediterraneus.
  • 2 The LC50 was estimated by applying the pesticides on the dorsal side of adults. Lambda‐cyhalothrin, methomyl and cypermethrin resulted in the highest toxicity followed, in decreasing order of toxicity, by malathion, fenthion and dimethoate. Parathion had the least effect on the coccinellid.
  • 3 Consumption of Bacillus thuringiensis bacteria (applied as a commercial formulation), over a period of 10 consecutive days, resulted in low mortality of adult P. mediterraneus.
  • 4 The results of these laboratory experiments indicate that the most widely used chemical pesticides were toxic to P. mediterraneus. Their use in olive groves must take account of the activity period of this predator. In contrast, B. thuringiensis seemed to be a suitable candidate to be included in pest management systems.

The midge Chironomus tentans Fabricius is a commonly used freshwater invertebrate in sediment toxicity tests. Rigorous laboratory culturing techniques are needed to provide organisms of uniform quality and known age for use in testing and for the continuation of the culture itself. This study was conducted to determine the effect of initial culture stocking density on: (1) post-hatch (larval) dry weight, body length and head-capsule width at 10 and 20 days; (2) time to emergence; (3) number and sex of emergent adults; (4) number of larvae and pupae at test termination (day 42 post hatch); and (5) adult dry weight. Three egg stocking densities were used 690 (1.1 eggs cm–2), 1043 (1.7 eggs cm–2) and 1463 (2.4 eggs cm–2). Mean weight of larvae at 10 days in high density tanks (0.13 mg/organism) was significantly higher (P=0.003) than both the medium and low density tanks (0.10 and 0.09 mg/organism, respectively). No significant differences between the three stocking densities were observed for the body length or head-capsule width at either 10 or 20 days post-hatch. Although not statistically significant, larval dry weight decreased with increased stocking density at day 20. A significantly (P=0.02) greater number of females (173±28) emerged from the low stocking density compared to both the medium and high stocking densities (123±45 and 118±54, respectively). Peak adult emergence for the low and medium stocking densities occurred between days 22 and 25 post-hatch, whereas peak adult emergence occurred between days 30 and 33 for the high stocking density. Survival relative to the initial number of eggs stocked was significantly greater (P=0.007) in the low density treatment compared to that in either the medium or the high density treatments. Mean adult weight exhibited an inverse relationship with initial stocking densities. At test end, there was not a significant difference in the mean number of organisms surviving and emerging in the three density levels. The central tendency for number of organisms surviving for all three treatments was 504 organisms per tank (0.82 organisms cm–2). The results of this experiment suggest that an optimal egg stocking density of 1.0 egg cm–2 (600 eggs/tank) be used with the feeding rate identified. This would ensure uniform larvae at the appropriate developmental stage (2nd–3rd instar) needed for toxicological research/testing (e.g. 10 days post-hatch), as well as producing sufficient emergence of males and females for future culture establishment.  相似文献

镉是对生物毒性最强的污染物之一。过量的镉能够抑制植物的生长和光合作用,干扰矿质代谢并诱发氧化胁迫。硅作为一种有益元素,主要以Si(OH)_4的形态通过主动或被动方式被植物体吸收并转运到地上部分。硅对植物镉毒害具有缓解作用,但其缓解机制在不同物种、品种或生态型之间存在显著差异,并表现出一定的硅/镉浓度依赖性。总体上可概括为避性机制和耐性机制。避性机制包括:(1)在器官水平,减少植物根系对镉的吸收及其向地上部的转运;(2)在细胞水平,增强细胞壁对镉的吸附能力,减少共质体中镉的含量。耐性机制包括:(1)诱导细胞产生小分子螯合剂,增强对镉的螯合作用,减少细胞中游离态镉的含量;(2)增强抗氧化机制,减轻氧化胁迫;(3)改善光合作用和无机营养,促进植物生长。从植物对硅的吸收和转运、镉对植物的毒害作用以及硅对缓解植物镉毒害的生理生态机制3个方面进行了综述,并基于目前的研究现状和薄弱之处,对今后的研究重点进行了展望。  相似文献

Smith ABC3 Part I: a review of the guidelines for terminology, classification and management of cervical cytology in England The provision of guidance on cytology reporting and evaluation, first outlined in 1995 with the publication of Achievable Standards, Benchmarks for Reporting, and Criteria for Evaluating Cervical Cytopathology (ABC), and subsequently revised and expanded in a second edition in 2000, has been pivotal to the success of the National Health Service Cervical Screening Programme (NHSCSP), ensuring that standards are upheld, and that rigorous evaluation and quality assurance take place. In the last decade, major changes to the NHSCSP, notably the adoption of revised age ranges and screening intervals for all women in England, implementation of liquid‐based cytology and, most recently, the decision to introduce high‐risk human papillomavirus (HR‐HPV) testing for triage of low‐grade and borderline (equivalent to 'atypical') cytological abnormalities and test of cure after treatment of cervical intraepithelial neoplasia (CIN) determined that an updated version of ABC was required. The third edition of ABC recommends adoption, with minor modification, of the revised British Society for Clinical Cytology terminology and provides guidance on the management of abnormal cytology results linked to this terminology taking account of HR‐HPV testing. To accommodate these changes, expanded result codes, which are electronic codes used to transfer management information to central computers for follow‐up, call and recall of individual women, have been developed. Further guidance on specimen adequacy is also provided. Revised performance indicators are described and explained in a separate article by R. Blanks in this issue of Cytopathology. All the changes in ABC3 are designed to support the mission statement of the NHSCSP that ‘the objective of cervical screening is to reduce cervical cancer incidence and mortality by screening with a high sensitivity for the detection of CIN2 or worse, whilst maintaining a high specificity’.  相似文献

尽管不同纳米材料 (<100 nm) 在污染环境修复研究中的应用越来越受到重视,但纳米技术在给污染环境修复带来重要突破的同时给环境和人类健康带来的风险也值得关注. 有研究表明,在污染环境修复研究中,不同的纳米颗粒对生物体的健康会造成一定的危害. 目前,在纳米材料的环境修复研究中,针对不同纳米材料的修复效果、修复过程与机理研究较多,而对纳米材料的环境安全性研究才刚刚起步. 本文对近年来国内外关于纳米材料环境毒性的研究进行综述,并对该研究领域的前景和重点进行了展望,以期为该领域的深入研究提供借鉴并拓展新的思路.  相似文献

Specific pathogen free (SPF) macaques provide valuable animal models for biomedical research. In 1989, the National Center for Research Resources [now Office of Research Infrastructure Programs (ORIP)] of the National Institutes of Health initiated experimental research contracts to establish and maintain SPF colonies. The derivation and maintenance of SPF macaque colonies is a complex undertaking requiring knowledge of the biology of the agents for exclusion and normal physiology and behavior of macaques, application of the latest diagnostic technology, facilitiy management, and animal husbandry. This review provides information on the biology of the four viral agents targeted for exclusion in ORIP SPF macaque colonies, describes current state‐of‐the‐art viral diagnostic algorithms, presents data from proficiency testing of diagnostic assays between laboratories at institutions participating in the ORIP SPF program, and outlines management strategies for maintaining the integrity of SPF colonies using results of diagnostic testing as a guide to decision making.  相似文献

Currently there is a surge of interest in exploiting toxicogenomics to screen the toxicity of chemicals, enabling rapid and accurate categorisation into classes of defined mode-of-action (MOA), and prioritising chemicals for further testing. Direct infusion FT-ICR mass spectrometry-based metabolomics can provide a sensitive and unbiased analysis of metabolites in only 15 mins and therefore has considerable potential for chemical screening. The water flea, Daphnia magna, is an OECD test species and is utilised internationally for toxicity testing. However, no metabolomics studies of this species have been reported. Here we optimised and evaluated the effectiveness of FT-ICR mass spectrometry metabolomics for toxicity testing in D. magna. We confirmed that high-quality mass spectra can be recorded from as few as 30 neonates (<24 h old; 224 μg dry mass) or a single adult daphnid (301 μg dry mass). An OECD 24 h acute toxicity test was conducted with neonates at copper concentrations of 0, 5, 10, 25, 50 μg l−1. A total of 5447 unique peaks were detected reproducibly, of which 4768 were assigned at least one empirical formula and 1017 were putatively identified based upon accurate mass measurements. Significant copper-induced changes to the daphnid metabolome, consistent with the documented MOA of copper, were detected thereby validating the approach. In addition, N-acetylspermidine was putatively identified as a novel biomarker of copper toxicity. Collectively, our results highlight the excellent sensitivity, reproducibility and mass accuracy of FT-ICR mass spectrometry, and provide strong evidence for its applicability to high-throughput screening of chemical toxicity in D. magna.
