Pulmonary hypertension (PH) is characterized by increased pulmonary vascular remodeling, resistance, and pressures. Reactive oxygen species (ROS) contribute to PH-associated vascular dysfunction. NADPH oxidases (Nox) and mitochondria are major sources of superoxide (O2•−) and hydrogen peroxide (H2O2) in pulmonary vascular cells. Hypoxia, a common stimulus of PH, increases Nox expression and mitochondrial ROS (mtROS) production. The interactions between these two sources of ROS generation continue to be defined. We hypothesized that mitochondria-derived O2•− (mtO2•−) and H2O2 (mtH2O2) increase Nox expression to promote PH pathogenesis and that mitochondria-targeted antioxidants can reduce mtROS, Nox expression, and hypoxia-induced PH. Exposure of human pulmonary artery endothelial cells to hypoxia for 72 h increased mtO2•− and mtH2O2. To assess the contribution of mtO2•− and mtH2O2 to hypoxia-induced PH, mice that overexpress superoxide dismutase 2 (TghSOD2) or mitochondria-targeted catalase (MCAT) were exposed to normoxia (21% O2) or hypoxia (10% O2) for three weeks. Compared with hypoxic control mice, MCAT mice developed smaller hypoxia-induced increases in RVSP, α-SMA staining, extracellular H2O2 (Amplex Red), Nox2 and Nox4 (qRT-PCR and Western blot), or cyclinD1 and PCNA (Western blot). In contrast, TghSOD2 mice experienced exacerbated responses to hypoxia. These studies demonstrate that hypoxia increases mtO2•− and mtH2O2. Targeting mtH2O2 attenuates PH pathogenesis, whereas targeting mtO2•− exacerbates PH. These differences in PH pathogenesis were mirrored by RVSP, vessel muscularization, levels of Nox2 and Nox4, proliferation, and H2O2 release. These studies suggest that targeted reductions in mtH2O2 generation may be particularly effective in preventing hypoxia-induced PH.  相似文献

Pharmacological ascorbate (AscH) selectively induces cytotoxicity in pancreatic cancer cells vs normal cells via the generation of extracellular hydrogen peroxide (H2O2), producing double-stranded DNA breaks and ultimately cell death. Catalytic manganoporphyrins (MnPs) can enhance ascorbate-induced cytotoxicity by increasing the rate of AscH oxidation and therefore the rate of generation of H2O2. We hypothesized that combining MnPs and AscH with the chemotherapeutic agent gemcitabine would further enhance pancreatic cancer cell cytotoxicity without increasing toxicity in normal pancreatic cells or other organs. Redox-active MnPs were combined with AscH and administered with or without gemcitabine to human pancreatic cancer cell lines, as well as immortalized normal pancreatic ductal epithelial cells. The MnPs MnT2EPyP (Mn(III)meso-tetrakis(N-ethylpyridinium-2-yl) porphyrin pentachloride) and MnT4MPyP (Mn(III)tetrakis(N-methylpyridinium-4-yl) porphyrin pentachloride) were investigated. Clonogenic survival was significantly decreased in all pancreatic cancer cell lines studied when treated with MnP + AscH + gemcitabine, whereas nontumorigenic cells were resistant. The concentration of ascorbate radical (Asc•−, an indicator of oxidative flux) was significantly increased in treatment groups containing MnP and AscH. Furthermore, MnP + AscH increased double-stranded DNA breaks in gemcitabine-treated cells. These results were abrogated by extracellular catalase, further supporting the role of the flux of H2O2. In vivo growth was inhibited and survival increased in mice treated with MnT2EPyP, AscH, and gemcitabine without a concomitant increase in systemic oxidative stress. These data suggest a promising role for the use of MnPs in combination with pharmacologic AscH and chemotherapeutics in pancreatic cancer.  相似文献

Oxidants derived from myeloperoxidase (MPO) contribute to inflammatory diseases. In vivo MPO activity is commonly assessed by the accumulation of 3-chlorotyrosine (3-Cl-Tyr), although 3-Cl-Tyr is formed at low yield and is subject to metabolism. Here we show that MPO activity can be assessed using hydroethidine (HE), a probe commonly employed for the detection of superoxide. Using LC/MS/MS, 1H NMR, and two-dimensional NOESY, we identified 2-chloroethidium (2-Cl-E+) as a specific product when HE was exposed to hypochlorous acid (HOCl), chloramines, MPO/H2O2/chloride, and activated human neutrophils. The rate constant for HOCl-mediated conversion of HE to 2-Cl-E+ was estimated to be 1.5 × 105 m−1s−1. To investigate the utility of 2-Cl-E+ to assess MPO activity in vivo, HE was injected into wild-type and MPO-deficient (Mpo−/−) mice with established peritonitis or localized arterial inflammation, and tissue levels of 2-Cl-E+ and 3-Cl-Tyr were then determined by LC/MS/MS. In wild-type mice, 2-Cl-E+ and 3-Cl-Tyr were detected readily in the peritonitis model, whereas in the arterial inflammation model 2-Cl-E+ was present at comparatively lower concentrations (17 versus 0.3 pmol/mg of protein), and 3-Cl-Tyr could not be detected. Similar to the situation with 3-Cl-Tyr, tissue levels of 2-Cl-E+ were decreased substantially in Mpo−/− mice, indicative of the specificity of the assay. In the arterial inflammation model, 2-Cl-E+ was absent from non-inflamed arteries and blood, suggesting that HE oxidation occurred locally in the inflamed artery. Our data suggest that the conversion of exogenous HE to 2-Cl-E+ may be a useful selective and sensitive marker for MPO activity in addition to 3-Cl-Tyr.  相似文献

High environmental salt elicits an increase in cytosolic Ca2+ ([Ca2+]cyt) in plants, which is generated by extracellular Ca2+ influx and Ca2+ release from intracellular stores, such as vacuole and endoplasmic reticulum. This study aimed to determine the physiological mechanisms underlying Ca2+ release from vacuoles and its role in ionic homeostasis in Populus euphratica. In vivo Ca2+ imaging showed that NaCl treatment induced a rapid elevation in [Ca2+]cyt, which was accompanied by a subsequent release of vacuolar Ca2+. In cell cultures, NaCl-altered intracellular Ca2+ mobilization was abolished by antagonists of inositol (1, 4, 5) trisphosphate (IP3) and cyclic adenosine diphosphate ribose (cADPR) signaling pathways, but not by slow vacuolar (SV) channel blockers. Furthermore, the NaCl-induced vacuolar Ca2+ release was dependent on extracellular ATP, extracellular Ca2+ influx, H2O2, and NO. In vitro Ca2+ flux recordings confirmed that IP3, cADPR, and Ca2+ induced substantial Ca2+ efflux from intact vacuoles, but this vacuolar Ca2+ flux did not directly respond to ATP, H2O2, or NO. Moreover, the IP3/cADPR-mediated vacuolar Ca2+ release enhanced the expression of salt-responsive genes that regulated a wide range of cellular processes required for ion homeostasis, including cytosolic K+ maintenance, Na+ and Cl exclusion across the plasma membrane, and Na+/H+ and Cl/H+ exchanges across the vacuolar membrane.  相似文献

Nitration of arachidonic acid (AA) to nitroarachidonic acid (AANO2) leads to anti-inflammatory intracellular activities during macrophage activation. However, less is known about the capacity of AANO2 to regulate the production of reactive oxygen species under proinflammatory conditions. One of the immediate responses upon macrophage activation involves the production of superoxide radical (O2•−) due to the NADPH-dependent univalent reduction of oxygen to O2•− by the phagocytic NADPH oxidase isoform (NOX2), the activity of NOX2 being the main source of O2•− in monocytes/macrophages. Because the NOX2 and AA pathways are connected, we propose that AANO2 can modulate macrophage activation by inhibiting O2•− formation by NOX2. When macrophages were activated in the presence of AANO2, a significant inhibition of NOX2 activity was observed as evaluated by cytochrome c reduction, luminol chemiluminescence, Amplex red fluorescence, and flow cytometry; this process also occurs under physiological mimic conditions within the phagosomes. AANO2 decreased O2•− production in a dose- (IC50=4.1±1.8 μM AANO2) and time-dependent manner. The observed inhibition was not due to a decreased phosphorylation of the cytosolic subunits (e.g., p40phox and p47phox), as analyzed by immunoprecipitation and Western blot. However, a reduction in the migration to the membrane of p47phox was obtained, suggesting that the protective actions involve the prevention of the correct assembly of the active enzyme in the membrane. Finally, the observed in vitro effects were confirmed in an in vivo inflammatory model, in which subcutaneous injection of AANO2 was able to decrease NOX2 activity in macrophages from thioglycolate-treated mice.  相似文献

It has been known for more than three decades that outward Kir currents (IK1) increase with increasing extracellular K+ concentration ([K+]o). Although this increase in IK1 can have significant impacts under pathophysiological cardiac conditions, where [K+]o can be as high as 18 mm and thus predispose the heart to re-entrant ventricular arrhythmias, the underlying mechanism has remained unclear. Here, we show that the steep [K+]o dependence of Kir2.1-mediated outward IK1 was due to [K+]o-dependent inhibition of outward IK1 by extracellular Na+ and Ca2+. This could be accounted for by Na+/Ca2+ inhibition of IK1 through screening of local negative surface charges. Consistent with this, extracellular Na+ and Ca2+ reduced the outward single-channel current and did not increase open-state noise or decrease the mean open time. In addition, neutralizing negative surface charges with a carboxylate esterifying agent inhibited outward IK1 in a similar [K+]o-dependent manner as Na+/Ca2+. Site-directed mutagenesis studies identified Asp114 and Glu153 as the source of surface charges. Reducing K+ activation and surface electrostatic effects in an R148Y mutant mimicked the action of extracellular Na+ and Ca2+, suggesting that in addition to exerting a surface electrostatic effect, Na+ and Ca2+ might inhibit outward IK1 by inhibiting K+ activation. This study identified interactions of K+ with Na+ and Ca2+ that are important for the [K+]o dependence of Kir2.1-mediated outward IK1.  相似文献

Reactive oxygen species (ROS) play an important role in immune responses; however, their excessive production and accumulation increases the risk of inflammation-related diseases. Although irradiation is known to accelerate immunological aging, the underlying mechanism is still unclear. To determine the possible involvement of ROS in this mechanism, we examined 10,023 samples obtained from 3752 atomic-bomb survivors in Hiroshima and Nagasaki, who participated in repeated biennial examinations from 2008 to 2016, for the effects of aging and radiation exposure on intracellular ROS (H2O2 and O2•−) levels, percentages of T-cell subsets, and the effects of radiation exposure on the relationship between cell percentages and intracellular ROS levels in T-cell subsets. The cell percentages and intracellular ROS levels in T-cell subsets were measured using flow cytometry, with both fluorescently labeled antibodies and the fluorescent reagents, carboxy-DCFDA and hydroethidine. The percentages of naïve CD4+ and CD8+ T cells decreased with increasing age and radiation dose, while the intracellular O2•− levels in central and effector memory CD8+ T cells increased. Additionally, when divided into three groups based on the percentages of naïve CD4+ T cells, intracellular O2•− levels of central and effector memory CD8+ T cells were significantly elevated with the lowest radiation dose group in the naïve CD4+ T cells. Thus, the radiation exposure-induced decrease in the naïve CD4+ T cell pool size may reflect decreased immune function, resulting in increased intracellular ROS levels in central and effector memory CD8+ T cells, and increased intracellular oxidative stress.  相似文献

We hypothesized that NO is generated in isolated cardiac mitochondria as the source for ONOO production during oxidative stress. We monitored generation of ONOO from guinea pig isolated cardiac mitochondria subjected to excess Ca2+ uptake before adding succinate and determined if ONOO production was dependent on a nitric oxide synthase (NOS) located in cardiac mitochondria (mtNOS). Mitochondria were suspended in experimental buffer at pH 7.15, and treated with CaCl2 and then the complex II substrate Na-succinate, followed by menadione, a quinone redox cycler, to generate O2•−. L-tyrosine was added to the mitochondrial suspension where it is oxidized by ONOO to form dityrosine (diTyr) in proportion to the ONOO present. We found that exposing mitochondria to excess CaCl2 before succinate resulted in an increase in diTyr and amplex red fluorescence (H2O2) signals, indicating that mitochondrial oxidant stress, induced by elevated mtCa2+ and succinate, increased mitochondrial ONOO production via NO and O2•−. Changes in mitochondrial ONOO production dependent on NOS were evidenced by using NOS inhibitors L-NAME/L-NNA, TEMPOL, a superoxide dismutase (SOD) mimetic, and PTIO, a potent global NO scavenger. L-NAME and L-NNA decreased succinate and menadione-mediated ONOO production, PTIO decreased production of ONOO, and TEMPOL decreased ONOO levels by converting more O2•− to H2O2. Electron microscopy showed immuno-gold labeled iNOS and nNOS in mitochondria isolated from cardiomyocytes and heart tissue. Western blots demonstrated iNOS and nNOS bands in total heart tissue, bands for both iNOS and nNOS in β-tubulin-free non-purified (crude) mitochondrial preparations, and a prominent iNOS band, but no nNOS band, in purified (Golgi and ER-free) mitochondria. Prior treatment of guinea pigs with lipopolysacharride (LPS) enhanced expression of iNOS in liver mitochondria but not in heart mitochondria. Our results indicate that release of ONOO into the buffer is dependent both on O2•− released from mitochondria and NO derived from a mtCa2+-inducible nNOS isoform, possibly attached to mitochondria, and a mtNOS isoform like iNOS that is non-inducible.  相似文献

Methods for the detection and quantitation of hydroethidine (HE) and its oxidation products by HPLC analysis are described. Synthetic methods for preparation of authentic standards (2-hydroxyethidium and diethidium) are provided. Potential applications of the HPLC methods to chemical and biological systems are discussed. Specific examples of chromatograms obtained using UV–Vis absorption, fluorescence, electrochemical, and mass spectrometry detectors are provided. The development of a dual electrochemical and fluorescence detection methodology and its applications are described. The HPLC-based method enables analyses of HE and its oxidation products such as ethidium and the dimeric products of HE. The ramifications of HPLC measurement of HE and its oxidation products in the detection and quantitation of 2-hydroxyethidium, the diagnostic marker product of superoxide and HE, in the intracellular milieu are discussed. Similarly, mitochondria-targeted HE conjugated to a triphenylphosphonium group (Mito-HE or Mito-SOX) also forms oxidation products (dimers of Mito-HE and Mito-E+) that can affect the detection and quantitation of 2-hydroxy-mito-ethidium, the diagnostic marker product of Mito-HE and superoxide in mitochondria.  相似文献

Lutoids represent a lysosomal microvacuolar compartment of rubber-tree (Hevea brasiliensis) latex. We observed acidification of isolated vesicles after imposing an outward Mg2+ diffusion gradient and dissipation of a preformed pH gradient in the presence of exogenous Mg2+. These results suggest the presence of a Mg2+/H+ antiporter. The maximum Mg2+/H+ exchange rate was observed at pH 8.5. The Km values for Mg2+ (2.6 mm) were identical for both influx and efflux experiments. When membrane potential was clamped at zero with K+ and valinomycin, the response of the membrane potential probe oxonol VI showed that the Mg2+/H+ exchange was electroneutral. Mg2+/H+ exchange was inhibited by amiloride and imipramine. Both the inhibiting concentration range and the Km for Mg2+ are similar to those reported for the Mg2+/2Na+ antiporter in animals cell. These data are consistent with the existence of a Mg2+/2H+ antiporter in a plant tonoplast.  相似文献

Summary Using intracellular microelectrode technique, we investigated the changes in membrane voltage (V) of cultured bovine pigmented ciliary epithelial cells induced by different extracellular solutions. (1)V in 213 cells under steady-state conditions averaged –46.1±0.6 mV (sem). (2) Increasing extracellular K+ concentration ([K+] o ) depolarizedV. Addition of Ba2+ could diminish this response. (3) Depolarization on doubling [K+] o was increased at higher [K+] o (or low voltage). (4) Removing extracellular Ca2+ decreasedV and reduced theV amplitude on increasing [K+] o . (5)V was pH sensitive. Extra-and intracellular acidification depolarizedV; alkalinization induced a hyperpolarization.V responses to high [K+] o were reduced at acidic extracellular pH. (6) Removing K o + depolarized, K o + readdition after K+ depletion transiently hyperpolarizedV. These responses were insensitive to Ba2+ but were abolished in the presence of ouabain or in Na+-free medium. (7) Na+ readdition after Na+ depletion transiently hyperpolarizedV. This reaction was markedly reduced in the presence of ouabain or in K+-free solution but unchanged by Ba2+. It is concluded that in cultured bovine pigmented ciliary epithelial cells K+ conductance depends on Ca2+, pH and [K+] o (or voltage). An electrogenic Na+/K+-transport is present, which is stimulated during recovery from K+ or Na+ depletion. This transport is inhibited by ouabain and in K+-or Na+-free medium.  相似文献

Mitochondria are a major source of reactive oxygen species (ROS). Recent studies have estimated that mitochondrial DNA mutations inducing the overproduction of ROS are associated with human cancer. However, a substantial challenge in elucidating their diverse roles in regulating tumorigenesis is the lack of methods for probing ROS in living systems with molecular specificity. In this study, we reported the application of two fluorescent probes, 2‐chloro‐1,3‐dibenzothiazolinecyclohexene and naphthofluorescein disulfonate, which showed high selectivity for superoxide (O2•−) and hydrogen peroxide (H2O2). They were capable of detecting and visualizing O2•− and H2O2 overproduction caused by a mutation in the gene encoding nicotinamide adenine dinucleotide dehydrogenase subunit 6 (ND6) in HepG2 cells. The levels of O2•− and H2O2 in mitochondria isolated from HepG2 cells were found to be 0·63 ± 0·07 and 1·13 ± 0·05 μM, respectively. Using assays of tumorigenesis in mouse models, we found that treatment of the mice with different ROS scavengers suppressed tumour growth. These findings suggested that ROS generated by ND6 gene mutation do play an important role in regulating tumorigenesis and H2O2 may be a key modulator. We investigated epidermal peels from the leaves of pea (Pisum sativum L.) consisting of a monolayer of the cells of two types: stomatal guard cells (GC) with chloroplasts and mitochondria and basic epidermal cells (EC) containing only mitochondria. As inducers of programmed cell death, we used KCN destroying the nuclei in GC and EC and chitosan that destroys nuclei only in EC. AgNO3 (10 μM) stimulated the destruction of nuclei in GC and EC induced by CN? and suppressed chitosan-induced destruction of nuclei in EC. The destruction of nuclei in GC induced by CN? occurred under aerobic conditions and was prevented under anaerobiosis. The destruction of nuclei in GC induced by (CN? + Ag+) occurred both under aerobic and anaerobic conditions and was not suppressed by antioxidants. Among the tested cations of metals (Ag+, Hg2+, Fe2+, Fe3+, Cu2+, and Mn2+), Ag+ turned out to be the most efficient in respect to the stimulation of cyanide-induced destruction of nuclei in GC. Half-maximum concentrations of Ag+ and Hg2+ were equal to 4–5 μM. In epidermal peels treated with chitosan, GC were permeable to propidium iodide; however, the nuclei in GC (in contrast to EC) were not destructed in the presence of chitosan. It was concluded that Ag+, acting as an electron acceptor during photosynthetic electron transfer in the chloroplasts from pea leaves, impeded the O2 evolution by the chloroplasts treated with ferricyanide or silicomolybdate as electron acceptors, impeded the consumption of O2 in the course of electron transfer from the (ascorbate + N,N,N′,N′-tetramethyl-p-phenylenediamine) to methylviologen and suppressed the production of reactive oxygen species (ROS) in GC and EC.  相似文献

Charge compensation is an effective way to eliminate charge defects and improve the luminescent intensity of phosphors. In this paper, a new green‐emitting phosphor ZnB2O4:Tb3+ was prepared by solid‐state reaction at 750°C. The effects of Tb3+ doping content and charge compensators (Li+, Na+ or K+) on photoluminescence properties of ZnB2O4:Tb3+ were investigated. X‐ray powder diffraction analysis confirms the sample has cubic structure of ZnB2O4. The excitation and emission spectra indicate that this phosphor can be excited by near ultraviolet light at 378 nm, and exhibits bright green emission with the highest peak at 544 nm corresponding to the 5D47F5 transition of Tb3+. The critical quenching concentration of Tb3+ in ZnB2O4 host is 8 mol%. The results of charge compensation show that the emission intensity can be improved by Na+ and K+. Specifically, K+ is the optimal one for ZnB2O4:Tb3+. Copyright © 2014 John Wiley & Sons, Ltd.  相似文献

The present study examined the antioxidant effect of histidine, a singlet oxygen (1O2) scavenger, on para-nonylphenol (an environmental estrogen-like chemical)-enhanced hydroxyl radical (·OH) generation induced by 1-methyl-4-phenylpyridinium ion (MPP+) in extracellular fluid of rat striatum. Rats were anesthetized, and sodium salicylate in Ringer’s solution (0.5 nmol/μl/min) was infused through a microdialysis probe to detect the generation of ·OH as reflected by the non-enzymatic formation of 2,3-dihydroxybenzoic acid (DHBA) in the striatum. Introduction of para-nonylphenol (10 μM) significantly enhanced MPP+-induced ·OH generation. Histidine (25 mM) decreased the para-nonylphenol-enhanced ·OH formation. Although the level of MPP+-induced ·OH formation trapped as DHBA after para-nonylphenol treatment increased, para-nonylphenol failed to increase either the level of dopamine and DHBA formation in the reserpinized animals. These results indicate that para-nonylphenol and MPP+-enhanced ·OH generation was based on 1O2 production, and histidine may have a preventive effect on para-nonylphenol and MPP+-induced ·OH generation in rat striatum.  相似文献

Abstract

New techniques and probes are routinely emerging for detecting short-lived free radicals such as superoxide radical anion (O2?–), nitric oxide (?NO), and transient oxidants derived from peroxynitrite (ONOO/ONOOH). Recently, we reported the profiles of oxidation products (2-hydroxyethidium, ethidium, and various dimeric products) of the fluorogenic probe hydroethidine (HE) in the ?NO/O2?– system (Zielonka et al. 2012). In this study, we used HPLC analyses of HE oxidation products in combination with continuous wave electron paramagnetic resonance (CW-EPR) spin trapping with 5-tert-butoxycarbonyl-5-methyl-1-pyrroline N-oxide (BMPO) to define the identity of the oxidizing species formed in the ?NO/O2?– system. EPR spin-trapping technique is still considered as the gold standard for characterization of free radicals and their intermediates. We monitored formation of BMPO-superoxide (BMPO-?OOH) and BMPO-hydroxyl (BMPO-?OH) radical adducts. Simultaneous analyses of results from EPR spin-trapping and HPLC measurements are helpful in the interpretation of the mechanism of formation of products of HE oxidation.  相似文献

We investigated the effects of changing extracellular K+ concentrations on block of the weak inward-rectifier K+ channel Kir1.1b (ROMK2) by the three intracellular cations Mg2+, Na+, and TEA+. Single-channel currents were monitored in inside-out patches made from Xenopus laevis oocytes expressing the channels. With 110 mM K+ in the inside (cytoplasmic) solution and 11 mM K+ in the outside (extracellular) solution, these three cations blocked K+ currents with a range of apparent affinities (Ki (0) = 1.6 mM for Mg2+, 160 mM for Na+, and 1.8 mM for TEA+) but with similar voltage dependence (zδ = 0.58 for Mg2+, 0.71 for Na+, and 0.61 for TEA+) despite having different valences. When external K+ was increased to 110 mM, the apparent affinity of all three blockers was decreased approximately threefold with no significant change in the voltage dependence of block. The possibility that the transmembrane cavity is the site of block was explored by making mutations at the N152 residue, a position previously shown to affect rectification in Kir channels. N152D increased the affinity for block by Mg2+ but not for Na+ or TEA+. In contrast, the N152Y mutation increased the affinity for block by TEA+ but not for Na+ or Mg2+. Replacing the C terminus of the channel with that of the strong inward-rectifier Kir2.1 increased the affinity of block by Mg2+ but had a small effect on that by Na+. TEA+ block was enhanced and had a larger voltage dependence. We used an eight-state kinetic model to simulate these results.  One- and 2-year-old Pinus sylvestris saplings were exposed to chronic doses of ozone (O3) and sulphur dioxide (SO2) in short-term (3 months) and long-term (18 months) experiments. Microsomal and plasma membrane fractions were purified by phase partitioning from current-year needles. The following membrane enzyme activities were determined in the microsomal and/or purified plasma membrane fractions: K+, Mg2+-ATPase (EC 3.6.1.3), NADH ferricyanide oxidoreductase (EC 1.6.99.3), NADH-duroquinone reductase (EC 1.6.5.1), NADH oxidase type I (EC 1.6.99.2), NADH oxidase type II or peroxidase-like enzyme (EC 1.11.1.7) and pyrophosphatase (EC 3.6.1.1). NADH oxidase type I was slightly stimulated in the microsomal fraction after a short-term exposure to O3 whereas NADH-dependent duroquinone reductase was not affected by this pollutant. However, in the long term experiment, NADH oxidase type II measured in the plasma membrane fraction was more than 2-fold stimulated in the SO2 treated pines and more than 4-fold when O3 was added to SO2. However, pyrophosphatase was decreased by 50% in trees treated with SO2+O3 and remained unchanged in the SO2 treatment, indicating that this enzyme is probably sensitive to oxidation. K+, Mg2+-ATPase showed a trend towards an enhancement of activity when exposed to chronic concentrations of air pollutants, this enhancement was more important in the long-term experiment after the combined effect of SO2 and O3. However, the K+-stimulated component was inhibited by the combination of both pollutants. Finally, NADH ferricyanide reductase was significantly enhanced after O3 and SO2+O3 exposures appearing as the most sensitive oxidoreductase to these air pollutants. The stimulation of ATPase and membrane oxidoreductases could facilitate the adaptation and defense of trees by maintaining an adequate redox potential in the plasma membrane region and perhaps stimulating the reduction of extracellular electron acceptors generated by the exposure to air pollutants. Recently, D.J. Hall et al. reported that ethidium (E+) is formed as a major product of hydroethidine (HE) or dihydroethidium reaction with superoxide (O2) in intact animals with low tissue oxygen levels (J. Cereb. Blood Flow Metab. 32:23–32, 2012). The authors concluded that measurement of E+ is an indicator of O2 formation in intact brains of animals. This finding is in stark contrast to previous reports using in vitro systems showing that 2-hydroxyethidium, not ethidium, is formed from the reaction between O2 and HE. Published in vivo results support the in vitro findings. In this study, we performed additional experiments in which HE oxidation products were monitored under different fluxes of O2. Results from these experiments further reaffirm our earlier findings (H. Zhao et al., Free Radic. Biol. Med. 34:1359, 2003). We conclude that whether in vitro or in vivo, E+ measured by HPLC or by fluorescence lifetime imaging is not a diagnostic marker product for O2 reaction with HE.  相似文献

Rat melanotrophs express several types of voltage-gated and ligand-gated calcium channels, although mechanisms involved in the maintenance of the resting intracellular Ca2+ concentration ([Ca2+]i) remain unknown. We analyzed mechanisms regulating resting [Ca2+]i in dissociated rat melanotrophs by Ca2+-imaging and patch-clamp techniques. Treatment with antagonists of L-type, but not N- or P/Q-type voltage-gated Ca2+ channels (VGCCs) as well as removal of extracellular Ca2+ resulted in a rapid and reversible decrease in [Ca2+]i, indicating constitutive Ca2+ influx through L-type VGCCs. Reduction of extracellular Na+ concentration (replacement with NMDG+) similarly decreased resting [Ca2+]i. When cells were champed at –80 mV, decrease in the extracellular Na+ resulted in a positive shift of the holding current. In cell-attached voltage-clamp and whole-cell current-clamp configurations, the reduction of extracellular Na+ caused hyperpolarisation. The holding current shifted in negative direction when extracellular K+ concentration was increased from 5 mM to 50 mM in the presence of K+ channel blockers, Ba2+ and TEA, indicating cation nature of persistent conductance. RT-PCR analyses of pars intermedia tissues detected mRNAs of TRPV1, TRPV4, TRPC6, and TRPM3-5. The TRPV channel blocker, ruthenium red, shifted the holding current in positive direction, and significantly decreased the resting [Ca2+]i. These results indicate operation of a constitutive cation conductance sensitive to ruthenium red, which regulates resting membrane potential and [Ca2+]i in rat melanotrophs.  相似文献
