The objectives of the study were to evaluate the effectiveness of phenolic resin in protecting oil palm stem (OPS) plywood against both subterranean termites (Coptotermes curvignathus) and white rot fungi (Pycnoporous sanguineus). Specially cooked, Low molecular weight phenol formaldehyde (LMW PF) resin was used to treat the OPS veneer whilst commercial urea formaldehyde (UF) resin was used to bond the phenolic-treated veneer. OPS plywood were produced using two types of lay-up (100% outer veneer type and 100% inner veneer type) with adhesive spread rate of 200 g/m2. The results show that treatment of OPS veneer with LMW PF has significantly enhanced the resistance of OPS plywood against both termites and white rot fungi. In the termites resistance test, the percentage of weight loss for untreated samples were 19.2% (outer veneer) and 23.9% (inner veneer), while for phenolic treated samples were only 10.7% and 15.8%, respectively. The phenolic treatment was able to enhance the resistance towards termites by 38% and towards white rot fungi by 62%. The study has shown LMW PF resin can be used to protect OPS plywood from termites and white rot fungi.

Fire is an important part of forest ecosystems, as is the insect fauna. Changes in wood brought about by fire may alter the ability of termites to use the wood, interrupting the decay cycle of woody debris. The ability of termites to find, infest, and feed upon wood after it had been charred was evaluated in the laboratory and field. Eastern subterranean termites, Reticulitermes flavipes (Kollar) (Isoptera: Rhinotermitidae), fed on char from burned wood had significantly reduced numbers of protozoa compared to termites fed on pine shavings, but significantly more than starved termites. The ability of termites to find and infest wood was not affected by surface charring. In a laboratory choice test, there were no significant differences in the onset of feeding by termites between charred and non‐charred wood boards. Likewise in the field, no differences were observed in the time to initial attack by termites on charred and non‐charred wood boards or bolts. Because termites will likely survive fires of low to moderate intensity, in most cases, there should be no disruption of the termite contribution to forest nutrient and carbon cycles.

The natural occurrence of 6-benzylaminopurine, 6-(2-hydroxybenzylamino)purine (ortho-topolin), 6-(3-hydroxybenzylamino)purine (meta-topolin), their ribosides and 9-glucosides is reported using specific antibodies to these groups of compounds in high performance liquid chromatography/enzyme-linked immunosorbent assay (HPLC/ELISA). Compounds were identified by their retention times and differential cross-reactivities with six antisera in analyses carried out in two laboratories using different HPLC gradient systems. Identities were confirmed by immunoaffinity purification followed by HPLC with on-line UV spectrum analysis. Further confirmation of the occurrence of ortho-topolin riboside and isopentenyladenine-9-glucoside was obtained from gas chromatography-mass spectrometry analysis of permethylated HPLC fractions of an extract of oil palm tissues. The aromatic cytokinins, and in particular, ortho-topolin riboside, were found in a variety of oil palm tissues at concentrations exceeding those of the isoprenoid cytokinins, zeatin, isopentenyladenine, dihydrozeatin, and their ribosides. The 9-glucosides of isopentenyladenine and zeatin were more abundant than those of the aromatic types. The cross-reactivity of benzyladenine compounds with antibodies to isopentenyladosine is discussed in relation to the interpretation of ELISA data.Abbreviations BA N 6-benzyladenine - HPLC high performance liquid chromatography - ELISA enzyme-linked immunosorbent assay - GC-MS gas-chromatography-mass spectrometry - mT meta-topolin - oT ortho-topolin - TEAA triethylammonium acetate - IAC immunoaffinity chromatography - SPD spectral photodiode  相似文献

Thousands of tonnes of dry oil palm trunks will be produced annually in Malaysia after about 1990. A project was initiated to study the feasibility of converting palm trunks into charcoal. Carbonisation was done at terminal temperatures of 400–550°C with holding times of 1–3 h and at two heating rates. From laboratory-scale pyrolysis studies, it was found that holding time does not affect the quantity and quality of the charcoal produced, while heating rate has a minor influence. However, as terminal temperature increases, both yield and volatile content decrease while the fixed carbon content increases. The calorific value and ash contents are independent of the parameters studied and their respective values are 4032 kcal/kg and 37.2%. Since the calorific value is low and the ash content high, it is concluded that oil palm trunks are not suitable for the production of charcoal fuel.

The technique of mRNA fingerprinting was used to isolate flower-specific cDNAs in the oil palm. Differences in the RNA populations between vegetative tissue (leaf) and inflorescences at various stages of flower development were examined using 18 primer combinations. A total of 16 flower-specific cDNAs were identified, of which 15 were successfully re-amplified. Reverse Northern analysis confirmed that 8 of the 15 cDNAs appeared to truly represent differentially expressed mRNAs in flowering tissues. Northern blot analysis subsequently showed that 5 of the clones are preferentially or exclusively expressed in the flowering tissues of oil palm. The use of stone-tools to open palm nuts (Elaeis guineensis) was studied in a group of 16 chimpanzees (Pan troglodytes) released from captivity to a natural island setting in Liberia. The behaviour was started by one female of the group; it then spread to 12 others over periods varying from a few seconds to a few weeks. Nut-cracking soon spread to three other sites, both spontaneously and with human encouragement. Both nuts and tools were carried distances of several hundred meters. Social interaction at cracking sites was rich and varied, ranging from fights over possession of tools to unsolicited sharing of nuts. Nut-crackers showed selectivity in schoosing “good” nuts and varied their methods according to the qualities of the nuts. Individual differences in technique emerged. These are the first detailed behavioural data on palm-nut-cracking, and they show many parallels with nut-cracking of other species by chimpanzees of the Tai Forest, Ivory Coast. This is another example of hammer-stone use from a limited region of West Africa: southeastern Guinea, eastern Liberia, and western Ivory Coast. This suggests limited cultural diffusion of the custom.

We compared the feeding excavations on wood blocks of three species of subterranean termites, Coptotermes formosanus Shiraki, Reticulitermes flavipes (Kollar), and R. virginicus (Banks). Feeding rate followed the order C. formosanus > R. flavipes > R. virginicus. Wood surface area (mm2) exposed per unit feeding was higher for C. formosanus and R. flavipes than for R. virginicus. This was caused by the tendency of C. formosanus and R. flavipes to make internally penetrating tunnels, thereby increasing surface area, whereas R. virginicus made trough- and bowl-like depressions on the outside of blocks, sometimes decreasing the size of blocks outwardly without a corresponding high increase in surface area typical with the tunnels of the other species. Consequently, wood surface area was sometimes reduced, rather than increased as a result of feeding by R. virginicus. Different patterns of wood excavation suggest that these termites have divergent roles in wood decay processes.

Oil palm trunks are a possible lignocellulosic source for ethanol production. Low enzymatic digestibility of this type of material (11.9% of the theoretical glucose yield) makes pretreatment necessary. An enzymatic digestibility of 95.4% with insoluble solids recovery of 49.8% was achieved after soaking shredded oil palm trunks in ammonia under optimum conditions (80 °C, 1:12 solid-to-liquid ratio, 8 h and 7% (w/w) ammonia solution). Treatment with 60 FPU of commercial cellulase (Accellerase 1000) per gram of glucan and fermentation with Saccharomyces cerevisiae D5A resulted in an ethanol concentration of 13.3 g/L and an ethanol yield of 78.3% (based on the theoretical maximum) after 96 h. These results indicate that oil palm trunks are a biomass feedstock that can be used for bioethanol production.

The genetic variation among different accessions of oil-palm germplasm collected from Africa was estimated using random primers and the polymerase chain reaction. The present study revealed high levels of genetic variation in these accessions. Electrophoresis of the amplification products indicated that nine out of 20 primers were able to generate polymorphic products ranging in length from 0.2 kb to 2.3 kb. No individual palm or population-specific products were observed. Greatest diversity was seen in Zaire population 5 and the least in Zaire population 2.

Abstract. A study was undertaken to assess the impact of the strepsipteran endoparasite Stichotrema dallatorreanum Hofeneder on the performance of its host Segestidea novaeguineae (Brancsik), a tettigoniid pest of oil palm in Papua New Guinea. This involved analyses of morphology, gut contents, body composition and reproductive variables of field-collected adults of stylopized and unstylopized hosts. The morphological consequences of stylopization by S. dallatorreanum included reduced wing, ovipositor and digestive tract lengths, whereas antennal length was largely unaffected. Feeding activity by the host, as estimated indirectly through weighing gut contents, was significantly reduced in parasitised female hosts, whereas smaller males showed no effect. Parasitized hosts had a reduced carcass dry weight associated with reduced nitrogen content and a substantial decline in carcass lipid content. Host reproductive variables were profoundly affected by stylopization. In females this included impaired ovarian development, reduced egg number and gonadal weight, the last of which was also reduced in males. There was an association between the number of S. dallatorreanum present (maximally six in males and nine in females) within a host and the extent of the impact of stylopization on most of the host characters investigated. The ability of S. dallatorreanum to reduce feeding and reproduction in the host suggests it may have potential use as a bio-control agent in the management of tettigoniid pests in Papua New Guinea.

The palm oil industry constantly attempts to increase the sustainability along the entire palm oil value chain. One important strategy is to utilize all co‐products. Oil palm trunks, which become available upon replanting of existing plantations, represent an important and increasing flow of underexploited biomass. In recent years, innovative technologies are emerging to use them for producing furniture or plywood or providing bioenergy. We assessed the life cycle greenhouse gas emissions of such products and the greenhouse gas emission savings due to replaced alternative products. Although challenging material properties result in a relatively high energy demand and related greenhouse gas emissions in the oil palm wood processing, substantial reductions in greenhouse gas emissions can arise from producing furniture or bioenergy from oil palm trunks, especially if the process energy demand is met by the energy recovery from oil palm wood‐processing residues.

Oil palm empty fruit bunches (EFB) were pretreated by aqueous ammonia soaking for ethanol production. Pretreated EFB, which were pretreated at the optimal conditions of 60 °C, 12 h, and 21% (w/w) aqueous ammonia, showed 19.5% and 41.4% glucose yields during an enzymatic digestibility test for 96 h when using 15 and 60 FPU of cellulase, respectively. Using the pretreated EFB, simultaneous saccharification and fermentation for 168 h with 5% (w/v) glucan loading and 60 FPU of cellulase and 30 CBU of β-glucosidase per gram glucan resulted in ethanol production of 18.6 g/L titer, 65.6% of theoretical maximum yield, and 0.11 g/L/h of productivity.

The oil palm industry is one of the main economic drivers in Southeast Asia. The industry has caused tropical deforestation on a massive scale in producing countries, and this forest conversion to oil palm agriculture has decimated the habitat of numerous native species. Monoculture and polyculture practices are two distinctive oil palm production systems. We hypothesize that polyculture farming hosts a greater diversity of species than monoculture farming. Habitat complexity in smallholdings is influenced by multiple farming practices (i.e. polyculture and monoculture). However, little is known about the effects of such farming practices in smallholdings on mammalian biodiversity, and particularly frugivorous bats. Our study aimed to find the best farming practice to reconcile oil palm production with biodiversity conservation. Mist-nets were used to trap frugivorous bats at 120 smallholdings in Peninsular Malaysia. We compared species richness and the abundance of frugivorous bats between monoculture and polyculture smallholdings. We investigated their relationships with vegetation structure characteristics. Our results revealed that species richness and abundance of frugivorous bats were significantly greater in polyculture smallholdings than monoculture smallholdings. We also found that 28.21% of the variation in species richness was explained by in situ habitat characteristics, including the number of dead standing oil palms and immature oil palms, non-grass cover, height of non-grass cover, and farming practices. The in situ habitat quality was closely associated with oil palm farming management. Commercial growers should implement polyculture rather than monoculture farming because polyculture farming has positive effects on the abundance and species richness of bats in oil palm production landscapes.

A comparative study has been conducted on seven white rot fungi to investigate their abilities to produce laccase and selectively degrade lignin. Laccase was produced constitutively on the different media tested. Of the different lignins, phenolic compounds and sugars involved, the highest laccase yield was obtained on indulin AT. Salicylic acid inhibited enzyme activity. A temperature of 20°C and 0.2% of indulin AT were found to be optimum for enzyme activity. No correlation was found between the amount of enzyme and fungal mass produced. During semisolid degradation of angiospermic wood sawdust, Daedalea flavida caused a total weight loss of 11%, with a lignin loss of 15.77% during two months of decay. Lignin removal was comparatively selective during the first month, during which time laccase production was also higher, indicating its probable role in lignin degradation.

The objective of the research was to investigate the suitability of the white-rot fungus Trametes versicolor for biopulping using oil palm biomass as the substrate. Fungi are grown on solid-state cultures Kirk's enrichment media to determine the lignocellulolytic activities. Samples subjected to fungal pretreatment for periods of 1, 2, 3, and 4 weeks were investigated and compared to the untreated control. The crude enzyme extracts were assayed using specific substrates and enzyme activities were calculated. The highest level of laccase activity was 218.66 U/L; the peak activity of manganese peroxidase was 162.10 U/L, and lignin peroxidase is 42.56 U/L. The activity levels of cellulase and hemicellulase were insignificant in all extracts (53.30 and 1.50 U/L, respectively). When the chips were pulped mechanically the Kappa number, pulp yield, and screened pulp yield decreased significantly and paper strength increased marginally with the exposure time. Hand sheet properties were also improved significantly by fungal treatment. Weight loss, lignin loss, cellulose, and holocellulose loss were 8.45%, 9.35%, 4.58%, and 7.2%, respectively. Images from SEM seem to indicate a simultaneous type of decay pattern involving cell wall breakdown combined with lignin modification. Considering all its pulping and papermaking properties, the performance of T. versicolor is good and has potential for use in large-scale biotechnological processes.

Exudates were collected from stumps of pre-anthesis inflorescences of oil palm and analysed for cytokinin and gibberellin content using combined HPLC-ELISA techniques. Three antisera, for zeatin-type, dihydrozeatin-type and isopentenyladenine-type cytokinins, were used in ELISAs to identify members of these three groups of cytokinins. Ribotides, 9-glucosides, free bases and ribosides were detected for each of the groups with zeatin riboside the most abundant cytokinin identified in the exudate. Isopentenyladenine-type and dihydrozeatin-type cytokinins were also identified but at lower levels. In addition, two monoclonal antibodies were used in the development of novel ELISAs for members of the 13-hydroxylated and non-13-hydroxylated families of gibberellins. The new ELISAs allow the determination of gibberellins in smaller amounts of tissue than are required for GC-MS. The most abundant gibberellins identified in exudates were GA19 and GA44, as well as other members of the early 13-hydroxylation pathway. Gibberellins were confirmed by GC-MS. The presence of these types of growth regulators in exudate supplying immature inflorescences suggest they have a role in growth and development of these structures.

Strain Trichoderma koningii D-64 was improved for enhanced cellulase production. A potential mutant MF6 was obtained and its enzymes contained filter paper cellulase (FPase), carboxymethylcellulase (CMCase), β-glucosidase and xylanase with respective activities of 2.0, 1.3, 2.0 and 3.0 folds of those for the parental strain. MF6 cellulases showed enhanced hydrolysis performance for the treated lignocellulosic biomass. Hydrolysis of treated oil palm empty fruit bunch (OPEFB), horticulture wastes (HW) and wood chips (WC) resulted in cellulose to glucose conversion of 96.3 ± 2.2%, 98.2 ± 3.0% and 81.9 ± 1.4%, respectively. The corresponding conversions of xylan to xylose were 96.9 ± 1.5%, 95.0 ± 2.2% and 76.1 ± 3.1%. Consistently, high sugar yield of 770–844 mg/g biomass was obtained for high-loading (10–16%, w/v) of OPEFB hydrolysis and sugar titer of 135.1 g/L was obtained for 16% (w/v) OPEFB loading at 96 h. In addition, MF6 enzymes alone performed equally well for high-loading OPEFB hydrolysis compared to the enzyme mixture of β-glucosidase from Aspergillus niger and cellulase from T. reesei Rut C30.

Industrial oil palm expansion has led to dramatic landscape changes that have negatively affected forest biodiversity in the tropics. In contrast to large-scale plantations, oil palm smallholdings may support greater levels of biodiversity through the implementation of multi-cropping system or polyculture. We examined bird species richness, together with community structure, conservation status, and feeding guild of existing smallholdings in Peninsular Malaysia. Based on point transect sampling, we sampled birds in 100 smallholdings that practiced either monoculture or polyculture farming. Our results revealed that bird species richness was significantly greater in monoculture smallholdings than in polyculture smallholdings, but the opposite was true for bird abundance. Non-forest birds constituted the major species of bird communities in oil palm smallholdings. However, we found that the abundances of insectivores and frugivores were greater in polyculture smallholdings than in monoculture smallholdings. In the monoculture models, predictor variables explained 11.31–19.98% of the variation in bird species richness. When polyculture was being practiced, bird species richness increased significantly with the height of ground vegetation cover, distance to major roads, and distance to rice fields. In the polyculture models, predictor variables accounted for 11.71–24.85% of the variation in bird species richness. We also found that bird species richness increased significantly with height of ground vegetation, but it decreased with ground vegetation cover and distance to rivers. The evidence from this study suggests that monoculture and polyculture farming were able to maintain farmland biodiversity in smallholdings, at least for birds, but differed in richness, population, and feeding guild.
