Polyamines are new plant growth regulators that participate in various physiological processes modulating cell division and differentiation, stimulating secondary metabolite production and in stress responsiveness. In the present study, we evaluated the effect of polyamine application on CYCB1-GUS reporter line in Arabidopsis, in order to monitor changes in cell division. We observed that polyamines modulate the expression of CYCB1-GUS, most likely in an amine-specific manner. In particular, spermidine and spermine induced significant increases in CYCB1-GUS expression in shoot apex and root meristems. According of this view, mainly the higher polyamines stimulate the lateral root formation in Arabidopsis. Furthermore, the application of d-arginine and methylglyoxal bis-(guanylhydrazone) polyamine inhibitors drastically reduced Arabidopsis CYCB1-GUS root growth and plant fresh weight, as well as CYCB1-GUS expression. Another key point on this study was to analyze the effect of polyamines on CYCB1-GUS expression under salt stress. Salt stress treatments repressed CYCB1-GUS expression in a concentration dependent manner; this negative effect was ameliorated by polyamine application, in particular by spermidine and spermine, even at 125 mM NaCl, allowing the maintenance of CYCB1-GUS levels under salt stress. This work is one more contribution on the role of polyamines in cell cycle modulation and abiotic stress protection.  相似文献

Differential effects of polyamines on rat thyroid protein kinase activities   总被引:1,自引:0,他引:1  
Ornithine decarboxylase, the rate-limiting enzyme in polyamine biosynthesis, has been shown to be regulated in thyroid by thyrotropin both in vivo and in vitro. Little, however, is known of the role of polyamines in thyroid cell function. Since studies in other tissues suggest that polyamines may influence protein phosphorylation, we studied the effect of the polyamines on various protein kinase activities in rat thyroid. Putrescine, spermidine, and spermine inhibit cyclic-AMP-dependent histone H1 kinase activity when measured in the cytosol fraction of rat thyroid; this effect is largely reproduced by NaCl concentrations of equivalent ionic strength. Both spermidine and spermine effect a 1.6-2.4-fold increase in cytosolic cyclic-AMP-independent (messenger-independent) casein kinase activity; stimulation by both polyamines is maximal at 5mM. A similar profile of stimulation is observed for messenger-independent casein kinase activity in crude nuclear preparations. Sodium chloride fails to stimulate both cytosolic and nuclear messenger-independent casein kinase activities at ionic strength equivalent to the spermine concentrations used. Spermine, but not putrescine, spermidine, or sodium chloride, inhibits calcium/phospholipid-dependent protein kinase C activity in cytosol extracts partially purified by DEAE chromatography. These findings suggest that regulation of protein kinase(s) by polyamines may represent a proximal locus (i) of action of thyrotropin-regulated ornithine decarboxylase activity in thyroid.  相似文献

To gain a better understanding of the mechanism of rice (Oryza sativa L.) in response to salt stress, we performed a proteomics analysis of rice in response to 250 mM NaCl treatment using shoots of 3-day-old nascent seedlings. The changes of protein patterns were monitored with two-dimensional gel electrophoresis. Of 57 protein spots showing changes in abundance in response to salt stress, 52 were identified by matrix-assisted laser desorption/ionization time-of-flight mass spectrometry. The identified proteins were classified into eight functional categories. Several novel salt stress-responsive proteins, including protein synthesis inhibitor I, photosystem II stability/assembly factor HCF136, trigger factor-like protein and cycloartenol-C24-methyltransferase are upregulated upon salt stress. In order to figure out the different and similar molecular mechanism among salt and other stresses, regulation of some salt responsive proteins under other abiotic stress (cold and dehydration) and abscisic acid application was also analyzed. The possible molecular mechanism of rice seedlings in response to salinity and other stresses were discussed.  相似文献

Most of commercially important crops, including maize and common bean, are sensitive to water deficit and salinity. Polyamines are considered to be osmotic and salt tolerance modulators and biochemical indicators of these stresses. In the present study, we measured organ-specific changes in levels of free, microsome- and thylakoid-associated polyamines in leaves and roots of maize and common bean plants exposed for 24 h to osmotic and saline stresses. Putrescine levels were generally higher in the studied organs of both species and under both stresses; only in the roots of salt-treated bean it considerably decreased. In both species, salt stress (200 mM NaCl) induced a significant decrease in free spermidine in roots. We observed a significant decrease in the contents of all polyamines associated with the microsomes isolated from the roots of maize and bean growing in sorbitol and salt conditions. Also the microsomes isolated from the leaves of stressed plants were characterized by the lower contents of polyamines. Our data showed a reduction of putrescine content, with significantly decreased spermidine and spermine levels in thylakoids isolated from the chloroplasts of maize and bean plants growing under both stresses. The results indicate that the studied maize and bean cultivars are rather drought-sensitive. Additionally, microsome- and thylakoid-associated polyamines seem to be good markers of plant stress tolerance.  相似文献

以西伯利亚白刺试管苗为材料,研究其在0、50和200 mmol·L-1NaCl胁迫及外源环己胺调控下,生物量和体内多胺含量的改变,以探讨多胺对盐生植物白刺在盐胁迫下的生理调节功能.结果显示:50 mmol·L-1NaCl更有利于西伯利亚白刺的生长,处理后30 d白刺根系和地上部干重分别比对照增加75%和95%,处理15 d生根率比对照提高44.42%.而0(对照)和200 mmol·L-1NaCl都显著抑制白刺试管苗的生根与生长;200 mmol·L-1NaCl处理45 d时地上部干重比对照显著降低11.71%.白刺叶片中的多胺(精胺和亚精胺)含量随着盐浓度的增加呈降低趋势,茎和根中的多胺含量在200 mmol·L-1NaCl处理后才显著受到抑制;同时施环己胺和200 mmol·L-1NaCl处理使白刺叶片中的亚精胺含量显著低于200 mmol·L-1NaCl处理组,且叶绿素合成减少,细胞质膜透性增高.研究表明,西伯利亚白刺叶片内保持合适的多胺含量可能是其适应盐渍环境的重要机制之一.  相似文献

The effect of NaCl on the endogenous levels of diamine, putrescine and polyamines, spermidine and spermine, was studied in the shoot system of salt-tolerant and salt-sensitive lines of rice (Oryza sativa L.) cultivars during three growth stages. Salt stress increased the levels of diamine and polyamine in varying degrees among nine rice cultivars investigated. Salt tolerant AU1, Co43, and CSC1 were effective in maintaining high concentrations of spermidine and spermine, while the content of putrescine was not significantly altered in all the growth stages when plants were exposed to salinity. The salt sensitivity in rice was associated with excessive accumulation of putrescine and with low levels of spermidine and spermine in the shoot system of salt-sensitive cultivars Co36, CSC2, GR3, IR20, TKM4, and TKM9 under saline condition. One of the possible mechanisms of saline resistance was observed to be due to the highly increased polyamines against the low increase in diamines. Alternatively, the salt sensitivity could be due to high increase of diamines and an incapacity to maintain high levels of polyamines.  相似文献

Effects of salinity (0, 50, 100 and 250 mM NaCl) on growth, root:shoot dry mass ratio, osmotic potential (ψx), electrolyte leakage and contents of Na+ and K+, polyamines and abscisic acid (ABA) were studied in the grape rootstocks Dogridge, 1613, St. George and Salt Creek. In control rootstocks, the root length was highest in Dogridge and contents of K+ and ABA in Salt Creek. Salinity treatments increased root Na+ and decreased K+ content and St. George exhibited highest Na+ content and Na+:K+ ratio. The root:shoot dry mass ratio in all rootstocks increased upto 100 mM NaCl. With increasing NaCl concentration, putrescine, spermine and spermidine contents showed consistent increase and putrescine increase was highest in St. George and spermidine and spermine in the Dogridge and Salt Creek. Under salinity, the ABA content increased in all the rootstocks but more in Salt Creek and Dogridge than in St. George.  相似文献

The accumulation of conjugated and free polyamines in plants is very important for their protection against oxidative stress induced by abiotic factors. In the present study, the species halophytic plant Mesembryanthemum crystallinum L. was used as a model system in which the process of Crassulacean Acid Metabolism induction is linked with oxidative stress, especially under salinity conditions. A comparative analysis of the content of free polyamines, perchloric (PCA)-soluble and PCA-insoluble conjugated polyamines in mature leaves and roots was carried out with plants exposed to salinity. It was found that adult leaves and roots under normal conditions or salinity (400 mM NaCl) contained all types of free polyamines (putrescine, spermidine, spermine, and cadaverine). In leaves only PCA-insoluble conjugates were found, which showed a tendency to grow with increased duration of salt action (1.5–48 h). In contrast to leaves, in roots all forms of polyamine conjugates (PCA-soluble and -insoluble) were detected. However, the formation of all conjugates, especially PCA-soluble forms in roots, was sharply inhibited by salt shock (400 mM NaCl, 1.5 h) or exogenous cadaverine (1 mM) treatment. PCA-soluble conjugates of cadaverine in roots were found only when the treatment was carried out in combination with aminoguanidine (1 mM), as a result of diamine oxidase inhibition and consequently a decreasing of H2O2 production in plant cells. The activation of diamine oxidase and guaiacol peroxidase by NaCl or exogenous cadaverine was observed in leaves and roots. Thus, the activation of oxidative degradation of polyamines combined with H2O2–peroxidase reaction in cells are involved in the regulation of free and conjugated polyamines titers under salinity.  相似文献

Two rice varieties, viz. Nonabokra and Pokkali, have been evaluated for their responses to salinity in terms of some physiological and biochemical attributes. During the exposure to salinity (200 mM concentration of sodium chloride for 24, 48, and 72 h), a significant increase in sodium was recorded which was also concomitant with the changes of other metabolic profiles like proline, phenol, polyamine, etc. The protein oxidation was significantly increased and also varied between the two cultivars. The changes in activities of anti-oxidative enzymes under stress were significantly different to the control. The detrimental effects of salinity were also evident in terms of lipid peroxidation, chlorophyll content, protein profiles, and generation of free radicals; and these were more pronounced in Pokkali than in Nonabokra. The assessment and analysis of these physiological characters under salinity could unravel the mechanism of salt responses revealed in this present study and thus might be useful for selection of tolerant plant types under the above conditions of salinity.  相似文献

Salt stress perturbs a multitude of physiological processes such as photosynthesis and growth. To understand the biochemical changes associated with physiological and cellular adaptations to salinity, two lettuce varieties (Verte and Romaine) were grown in a hydroponics culture system supplemented with 0, 100 or 200 mM NaCl. Verte displayed better growth under 100 mM NaCl compared to Romaine, but both genotypes registered relatively similar reductions in growth under 200 mM NaCl treatment. Both varieties showed differences in net photosynthetic activity in the absence of salt and 8 days after salt treatment. These differences diminished subsequently under prolonged salt stress (14 days). Verte showed enhanced leaf proline and restricted total cations especially Na+, lesser malondialdehyde (MDA) formation and lignification in the roots under 100 mM NaCl salinity. Membrane damage estimated by electrolyte leakage increased with elevated salt concentrations in roots of both varieties, but Verte had significantly lower electrolyte leakage relative to Romaine under 100 mM NaCl. Moreover, Verte also accumulated greater levels of carotenoids under increasing NaCl concentrations compared to Romaine. Taken together, these findings suggest that the greater tolerance of Verte to 100 mM NaCl is related to the more restricted accumulation of total cations and toxic Na+ in the roots and enhanced levels of antioxidative metabolites in root and leaf tissue.  相似文献

Modulation of differential gene expression and change of polyamine content by salt stress are analyzed for the first time in a well-known medicinal plant, Panax ginseng C.A. Meyer. Three ginseng genes (PgSPD, PgSAMDC, and PgADC) involved in polyamine biosynthesis showed differential up-regulation patterns after 1 and 7 days of salt treatments. The modulation of gene expression resulted in the elevation of total polyamine content with relatively high levels of spermidine and spermine, while putrescine level diminished depending on the salt concentration. Conversely, salt stress led to a significant increase in diamine oxidase and subsequent decline in polyamine oxidase. The proline content caused by salinity follows a similar pattern as the total polyamine content and exogenous spermidine also resulted in the alleviation of proline content under salinity. Further, polyamine biosynthesis inhibitors, such as cyclohexylamine and methylglyoxal bis-(guanylhydrazone) mediated down-regulation of PgSPD and PgSAMDC, and affected cellular polyamine levels. Thus, polyamines may enhance the ginseng plant tolerance in response to the salt stress by increasing the levels of endogenous polyamines.  相似文献

To examine the response of rice to salt stress, changes in protein expression were analyzed using a proteomic approach. To investigate dose- and time-dependent responses, rice seedlings were exposed to 50, 100 and 150 mM NaCl for 6 to 48 h. Proteins were extracted from leaf sheath and separated by two-dimensional polyacrylamide gel electrophoresis. Eight proteins showed 1- to 3-fold up-regulation in leaf sheath, in response to 50 mM NaCl for 24 h. Among these, three proteins were unidentified (LSY081, LSY262 and LSY363) while five proteins were identified as fructose bisphosphate aldolases, photosystem II (PSII) oxygen evolving complex protein, oxygen evolving enhancer protein 2 (OEE2) and superoxide dismutase (SOD). The maximum expression levels of seven proteins were at 24 h. Their expression declined after 48 h of 50 mM NaCl treatment. In contrast, SOD maintained its elevated expression throughout these conditions. The increased expression of proteins seen in the 50 mM NaCl treatment group was less pronounced in the groups receiving 100 or 150 mM NaCl for 24 h. The expression of SOD was a common response to cold, drought, salt and abscisic acid (ABA) stresses while the expression of LSY081, LSY363 and OEE2 was enhanced by salt and ABA stresses. LSY262 was expressed in leaf sheath and root, while fructose bisphosphate aldolases, PSII oxygen evolving complex protein and OEE2 were expressed in leaf sheath and leaf blade. LSY363 was expressed in leaf sheath but was below the level of detection in leaf blade and root. These results indicate that specific proteins expressed in specific regions of rice show a coordinated response to salt stress.  相似文献

Polyamines(mainly putrescine(Put),spermidine(Spd),and spermine(Spm))have been widely found in a range of physiological processes and in almost all diverse environmental stresses.In various plant species,abiotic stresses modulated the accumulation of polyamines and related gene expression.Studies using loss-of-function mutants and transgenic overexpression plants modulating polyamine metabolic pathways confirmed protective roles of polyamines during plant abiotic stress responses,and indicated the possibility to improve plant tolerance through genetic manipulation of the polyamine pathway.Additionally,putative mechanisms of polyamines involved in plant abiotic stress tolerance were thoroughly discussed and crosstalks among polyamine,abscisic acid,and nitric oxide in plant responses to abiotic stress were emphasized.Special attention was paid to the interaction between polyamine and reactive oxygen species,ion channels,amino acid and carbon metabolism,and other adaptive responses.Further studies are needed to elucidate the polyamine signaling pathway,especially polyamine-regulated downstream targets and the connections between polyamines and other stress responsive molecules.  相似文献

Aims

The study aims (1) to evaluate the effect of Mesorhizobium tianshanense on plant proline and polyamine levels of Lotus tenuis and its modulatory effect during plant response to short-term salt stress and (2) to compare these effects with those caused by mycorrhizal symbiosis.

Methods

Experiments consisted of a randomized factorial design of two factors: salinity (two levels, 0 and 150 mM NaCl) and symbiosis (three levels, uninoculated, Glomus intraradices, and M. tianshanense).

Results

Salinization led to increased proline levels regardless of plant organ and symbiotic status, excepting mycorrhizal L. tenuis roots. Salinity diminished the total polyamine level of control and rhizobial plants but not in mycorrhizal ones. Variations in the pattern response of the three individual polyamines (putrescine, spermidine, and spermine) differed in accordance with the symbiotic status of the plant, highlighting a divergence on proline and polyamine metabolisms between rhizobial and mycorrhizal symbiosis.

Conclusions

Spermidine and spermine contributed the most with the salt-induced root polyamine increment observed upon salinization in roots of nodulated plants, suggesting that these polyamines might mediate an adaptive role of the plant–M. tianshanense symbiosis in L. tenuis plants growing in a saline environment.  相似文献

Responses of sunflower tissues to NaCl stress were studied in control (C), salt-stressed (S) and salt-adapted (T) calluses in terms of proline, polyamines and ethylene content for a period of 21 days. Salt-adapted calluses showed their adaptation to salinity by growing in the medium with 175 mM NaCl, at a similar rate than C calluses on medium without salt. Proline concentration was 27 times higher in salt-adapted calluses compared to control calluses at time 0, but salt stressed calluses (S calluses) were able to increase proline by day 21, demonstrating that proline was not just an osmoregulator but might be involved in other responses in sunflower salt-stressed calluses. Putrescine (Put) was the most abundant polyamine in C calluses at time 0, while spermidine (Spd) was the main polyamine in salt tolerant (T) calluses. Ethylene increased in C calluses until day 14, decreasing thereafter. In salt-adapted calluses, ethylene increased significantly over the concentration in C and S calluses by the end of the experiment. In control calluses, the highest level of total polyamines and the lowest of ethylene was found on day 21, while T calluses synthesized the highest ethylene level and had the lower polyamines level by this time. It seems that in salt-adapted calluses ethylene was related to stress tolerance and in salt sensitive tissues (S calluses), ethylene formation was related to senescence. The present data suggests a close relationship between proline, polyamines, ethylene and salt-stress tolerance in sunflower dedifferentiated tissues.  相似文献
