Sucrose (2,5–1000 mmol l–1), labeled with [14C]sucrose, was taken up by the xylem when supplied to one end of a 30-cm-long leaf strip of Zea mays L. cv. Prior. The sugar was loaded into the phloem and transported to the opposite end, which was immersed in diluted Hoagland's nutrient solution. When the Hoagland's solution at the opposite end was replaced by unlabeled sucrose solution of the same molarity as the labeled one, the two solutions met near the middle of the leaf strip, as indicated by radioautographs. In the dark, translocation of 14C-labeled assimilates was always directed away from the site of sucrose application, its distance depending on sugar concentration and translocation time. When sucrose was applied to both ends of the leaf strip, translocation of 14C-labeled assimilates was directed toward the lower sugar concentration. In the light, transport of 14-C-labeled assimilates can be directed (1) toward the morphological base of the leaf strip only (light effect), (2) toward the base and away from the site of sucrose application (light and sucrose effect), or (3) away from the site of sucrose application independent of the (basipetal or acropetal) direction (sucrose effect). The strength of a sink, represented by the darkened half of a leaf strip, can be reduced by applying sucrose (at least 25 mmol l–1) to the darkened end of the leaf strip. However, equimolar sucrose solutions applied to both ends do not affect the strength of the dark sink. Only above 75 mmol l–1 sucrose was the sink effect of the darnened part of the leaf strip reduced. Presumably, increasing the sucrose concentration replenishes the leaf tissue more rapidly, and photosynthates from the illuminated part of the leaf strip are imported to a lesser extent by the dark sink.Supported by Deutsche Forschungsgemeinschaft  相似文献

Summary Exudate can be obtained from incisions made in the bark of the stem of actively growing Ricinus plants. 14C-labelled assimilates from a fed leaf are rapidly detected in the exudate. This movement was both acropetal and basipetal from the fed leaf, at rates of over 100 cm h-1. Estimated rates within intact plants were 80–84 cm h-1.In contrast with xylem sap obtained from the same plant, the exudate obtained had an alkaline pH (8.2), a high dry matter content (10–12.5%), high sugar content (8–10%) which was predominantly sucrose; high potassium content (60–80 mM) and low calcium content (0.5–1.0 mM).It is concluded, on the basis of the present evidence, that the exudate is a true sample of the sieve tube sap undergoing translocation.  相似文献

We tested the possible cytokinin effect on the functioning of the active transport system involved in the assimilate loading into the phloem as a cause for the cytokinin sink and retention effect. This effect is manifested in the deceleration of substance export from and the stimulation of substance import to the sites of local phytohormone application to the mature detached leaf from untreated leaf areas. To affect the membrane mechanisms of the substance transport, we used leaf treatment with the phytotoxin fusicoccin, an enhancer of plasmalemmal H+-ATPase and a potential stimulator of assimilates export, and with the phytohormone ABA affecting transport, metabolism, and plant growth. However, fusicoccin did not enhance 14C-sucrose export from the leaf blade and did not interfere with the cytokinin-induced export deceleration. ABA reduced substantially 14C export from the leaf but eliminated the cytokinin effect on this process. Similar results were obtained for broad bean (Vicia faba L.) leaves with apoplastic phloem loading, involving H+-ATPase activity, and pumpkin (Cucurbita pepo L.) leaves with symplastic phloem loading, that is, occurring without sucrose transmembrane translocation and without H+-ATPase involvement. The conclusion is that the cytokinin-induced development of sink zones in source leaves is not related to the membrane mechanisms of the substance transport in the mesophyll–phloem system. The data obtained support the idea that the cause for the cytokinin sink and retention effect is the enhancement of elongation growth and total activation of metabolism in the mesophyll cells of the detached leaf.  相似文献

Summary Localized cooling of a 2 cm length of the leaf blade of the grass Lolium temulentum to 0° C, had only a small effect on the mass transfer of assimilates from the distal part of the leaf. There was no evidence of a reduction in the velocity of movement of 14C-labelled assimilates through the low temperature zone, and the retention of assimilates along the pathway of movement was reduced by low temperature. In contrast to the longitudinal movement, lateral movement of 14C, induced by steam killing sections of the leaf, was extremely sensitive to temperature and showed no sign of acclimatization, even after 3 days at low temperature. These results suggest that, although loading sugars into the transport system was sensitive to temperature, the movement of assimilates was not directly controlled by metabolic processes along the pathway.  相似文献

Summary The rate of translocation of 14C assimilates from leaves to seminal roots in wheat seedlings was considerably reduced by lowering root temperature from 20° to 10° or 5° although the total translocation of 14C to the roots after 24 h was little affected by temperature. The lowered root temperatures (particularly 5°) resulted in a more uniform distribution of assimilate along the roots than did a temperature of 20°, the ratios of radioactivity/cm in the apical cm, elongating zone, and basal parts of the root after 24 h being 14.0:9.6:1 in 20° roots by contrast with 2.8:1:1 in 5° roots. Temperature effects on assimilate distribution may help explain the observations that for roots grown below 15° ion uptake is sustained in older parts and that roots grown at a low temperature are thicker than roots grown at a higher one.  相似文献

Distribution of assimilates in cultivars of spring barley with different resistance against powdery mildew (Erysiphe graminis f. sp. hordei) Transport and distribution of radioactive labelled assimilates in spring barley cultivars with different degrees of resistance to powdery mildew were studied after 14CO2-treatment of single leaves. Plants of the cultivars ‘Amsel’ (susceptible), ‘Asse’ (adult plant resistant), and ‘Rupee’ (resistant) were analyzed at the vegetative growth stage (5. leaf unfolded) and the generative growth stage (anthesis). At the vegetative growth stage the assimilate export from the mildew inoculated 5. leaf of ‘Amsel’ and ‘Rupee’ is decreased; in ‘Asse’, there is no considerable change of assimilate distribution due to infection. At the generative growth stage the assimilate export from the infected flag leaf of ‘Amsel’ is reduced when the fungus, is sporulating. In the cultivar ‘Asse’ the assimilates are bound at the infection site until the seventh day after inoculation, then the transport of assimilates to the ear is increased. In ‘Rupee’ mildew inoculation causes an enhanced assimilate transport to the ear. The changes in assimilate distribution due to mildew inoculation are discussed with respect to the different types of host-parasite-interactions and the source-sink-activities in the different cultivars.  相似文献

育苗移栽不仅使陆地棉(Gossypium hirsutum L.)株在苗期、蕾期、花铃期的(14)~CO_2同化强度和同化量显著提高,而且对上述各生育期同化产物向籽棉的再动员具有明显的促进作用。各期标记后3d的测定结果显示,育苗移栽增加了棉苗同化物质向根系中的分配而减少了向顶芽的分配,有利于促根壮苗;使蕾期同化物质向顶芽和边心的分配比例下降,有利于控制棉花的营养生长,防止或减轻茎叶狂长,使棉株健壮稳长,促进花芽分化;在花铃期,促使更高比例的同化物质向成铃中分配,有利于蕾铃发育。  相似文献

There is considerable evidence to indicate that the floral stimulus moves along with photosynthate in the phloem. In the present study the effect of cooling a localized region of the stem of the short-day plant Pharbitis nil Chois. on translocation was investigated. This low-temperature treatment simultaneously inhibited translocation of photosynthate and of the floral stimulus, thus further supporting the idea that the floral stimulus is transported concurrently with assimilates in the phloem.  相似文献

Summary An analysis of the pattern of movement of 14C-labelled flag leaf assimilates in wheat (Triticum aestivum l. c.v. Gabo) during grain development, indicated that the greater the requirement for assimilates by the ear the more rapid was the speed of movement of these through the peduncle to the ear and also the lower their concentration. Experiments with [14C] indoleacetic acid ([14C]IAA) suggested that auxin production by the grains was not responsible for the control of assimilate translocation through the peduncle. Limiting the supply of available assimilates by shading the lower parts of the plant, did not significantly alter the speed of movement of 14C-photosynthate through the peduncle, while severing half of the vascular tissue in the peduncle altered the pattern of movement of 14C to the ear and enhanced the speed of movement of 14C through the remaining functional conducting tissue. These results are discussed in relation to the mechanism of translocation.  相似文献

Partitioning and translocation of 14C-photosynthates were examined during flowering and seed maturation in soybean (Glycine max [L.]Merr.) plants to quantify allocation to sugars, amino acids, organic acids, and starch and to study transport of C and N from leaves to reproductive sinks. The trifoliolate leaf at the eighth node was exposed to steady state levels of 14CO2 for 2 hours, followed by immediate extraction and identification of radioactive assimilates in the fed leaf blade, tissues of the transport path (e.g. petiole and stem), and fruits if they were present. About one-third of the total 14C recovered from the leaf blades was in starch until late pod-filling, after which the proportion dropped to 16%. Sugars comprised 70% to 86% of the recovered 14C from soluble assimilates of the source leaf, with highest proportions occurring during late flowering and early pod-filling. Amino acids accounted for 8% to 17% of the 14C recovered from the soluble fraction, and were most evident during early flowering and mid to late pod-filling. The 14C-organic acids comprised from 3% to 14% of the soluble 14C-assimilates in leaves. Petioles consistently contained a higher percentage of recovered radioactivity in sugars (87-97%) and a lower percentage in amino acids (3-12%) than did leaf blades. 14C-Amino acids in petioles attained their highest levels during mid and late pod-filling, while 14C-organic acids comprised 2% or less of the recovered radioactivity after pod initiation. The distribution of 14C-assimilates in the internode below the source leaf was similar to that found in petioles. A comparison of the above data to calculated C and N requirements for seed development suggests that 14C-amino acids derived from current photosynthesis and translocated from source leaves supply at least 12% to 48% of the seed N depending on the stage of pod-filling.  相似文献

The effects of increased sink-source ratios, induced by elevating night temperatures, on remobilization of 14C-assimilates and N within field-grown soybeans (Glycine max [L.] Merr.) was investigated from preflowering to maturity. Raising the mean minimum night temperature for the entire growing season from 10 (check, uncontrolled) to 16°C increased seed growth without appreciable effect on final leaf area. Increasing this temperature to 24°C increased seed growth and reduced final leaf area. Leaves, stems, petioles, and pods acted as intermediate storage sites for 14C assimilates. Only plants with higher night temperatures remobilized some of the stored assimilates during the period of rapid seed growth. Even the seeds in the 24°C plants with the largest sink-source ratios did not utilize all the C-assimilates potentially available for remobilization. Nitrogen was readily remobilized from petioles, stems, and pods of all treatments as early as the beginning of seed development, but from the leaves only during late seed-filling. However, only plants with elevated night temperatures tended to remobilize all of the available N from vegetative tissues and pods. We concluded that a larger portion of stored assimilates may be remobilized to the seed if a strong seed sink can be sustained. It also appeared that with increasing sink-source ratios, N shortage might limit seed yield before a lack of C-assimilates would. A proposed model for soybean assimilate demand, distribution, partitioning, and remobilization is presented.  相似文献

A well-developed infection of Yellow Rust on a leaf of springwheat (Jufy I) caused the assimilation of 14CO2 by that leafto decrease to 43.5 per cent of that of an uninfected leaf.Over a period of three hours translocation of 14C from an infectedleaf was only 0.87 per cent of that from a control leaf. Whencontrol plants were kept in the light for periods up to 16 hoursafter assimilating 14CO2 translocation continued at a steadyrate, whereas there was only negligible translocation from infectedleaves after the first few hours. The retention of labelledassimilates in the infected leaf could be partly, but not completely,accounted for by a conversion of assimilates to an alcohol-insolubleform. Rust infection had no effect on the distribution patternof 14C to other leaves from one which had assimilated 14CO2.In contrast to the marked retention of assimilate by an infectedleaf, such a leaf was unable to distort the normal distributionby attracting assimilates from the other leaves.  相似文献

Evidence is presented that growth substances have a role indirecting the movement of assimilates towards the developingflowers of carnation. Removal of the terminal flower resultedin decreased upward movement of 14C-labelled assimilates froma source leaf on the flowering shoot. Applications of indole-3yl-aceticacid or gibberellic acid to the stump of the flower stem partiallyrestored the pattern of movement to that found in the intactshoot. Two gibberellin-like substances and one auxin-like substancewere detected in extracts of the floral tissues. Two similar field trials were carried out during 2003 in a hot tropical region of eastern Ethiopia to investigate the effect of leaf and soil applied paclobutrazol on the growth, dry matter production and assimilate partitioning in potato. A month after planting paclobutrazol was applied as a foliar spray or soil drench at rates of 0, 2, 3, and 4 kg a.i. paclobutrazol ha–1. Plants were sampled during treatment application and subsequently 2, 4, 6 and 8 weeks after treatment application. The data was analyzed using standard growth analyses techniques. None of the growth parameters studied was affected by the method of paclobutrazol application. Paclobutrazol decreased leaf area index, crop growth rate, and total biomass production, and increased specific leaf weight, tuber growth rate, net assimilation rate, and partitioning coefficient of potato. At all harvesting stages, paclobutrazol reduced the partitioning of assimilate to the leaves, stems, and roots and stolons and increased allocation to the tubers. Although paclobutrazol decreased the total biomass production it improved tuber yield by partitioning more assimilates to the tubers. Paclobutrazol improved the productivity of potato under tropical conditions by redirecting assimilate allocation to the tubers.  相似文献

Summary Two weeks after anthesis, when the grain is filling rapidly, the rate of photosynthesis by flag leaves of wheat cv. Gabo was between 20 and 30 mg CO2 dm-2 leaf surface hour-1 under the conditions used. About 45% of flag-leaf assimilates were translocated to the ear, and only about 12% to the roots and young shoots.On removing the ear, net photosynthesis by the flag leaves was reduced by about 50% within 3–15 hours, and there was a marked reduction in the outflow of 14C-labelled assimilates from the flag leaves.Subsequent darkening of all other leaves on plants without ears led to recovery of flag-leaf photosynthesis, as measured by gas analysis and 14CO2 fixation, and to increased translocation of assimilates to the roots and young shoots. Minor changes in the rates of dark respiration accompanied these major, reversible changes in photosynthetic rate.After more than a week in continuous, high-intensity light, the rate of photosynthesis by flag leaves of intact plants had fallen considerably, but could be restored again by a period in darkness, or by inhibiting photosynthesis in the ears by spraying them with DCMU. The inhibition of ear photosynthesis increased translocation of labelled assimilates from the flag leaf to the ears, without affecting leaf sugar levels.The application of TIBA to the culm below the ear inhibited auxin movement throught the culm, but had no influence on flag-leaf photosynthesis.These results suggest that, at least in this system, photosynthesis by the flag leaf is regulated directly by the demand for assimilates from the flag leaf and not indirectly through action in the leaf of auxins produced by the sink organs.  相似文献

Summary Aqueous solutions of 2-thiouracil (TU) were applied selectively either to the growing point or to the leaves of the long-day plant Hyoscyamus niger in order to determine whether this antimetabolite has an effect on the synthesis of the floral stimulus in the leaves. Applications to the growing point were made by means of a small glass tube covering the shoot apex; application to the leaves was performed by vacuum infiltration. In all experiments all leaves except the three youngest fully expanded leaves and the 8–10 youngest primordia were removed before application. Plants were recorded as having initiated flowers when flower primordia were visible under a dissection microscope 5 weeks after the experiment.TU was inhibitory to photoperiodic induction by long-days of 16 hours when applied to the growing point during the second 8 hours of the daily photoperiod. A concentration of 5·10-3 M of TU fully suppressed flowering without significant inhibition of leaf primordia increment; however, leaves developing from treated primordia had reduced leaf blades. These results are in agreement with findings already published by other investigators.However, when the leaves were infiltrated by TU, the antimetabolite did not inhibit photoperiodic induction but on the contrary initiated flowering even under short-day conditions. This effect was investigated in more detail by repeated daily infiltrations of TU-solutions in concentrations of 10-5–10-2 M during the second part of an 8 hour photoperiod up to 5 following days. Even after one single infiltration of a 10-4 M solution 18% of the treated plants were flowering; the percentage of flowering plants increased with increasing concentrations of TU and number of days of application up to approximately 80%. In no case was a flower initiation of 100% obtained. Leaves developing from primordia after infiltration of the leaves with TU have reduced and deformed leaf blades, indicating that TU is transported to the shoot apex to some extent.Some possible explanations of this inductive effect of TU were tested experimentally. Oxygen uptake of the leaves was not decreased and the respiratory quotient was not affected by TU. Photoperiodic induction is not stimulated by low concentrations of TU when applied to the growing point. Infiltration of the leaves by solutions of 2,4-dinitrophenol (10-4 M) and sodium azide (10-3 M) had no inductive effect under short-day conditions; a single complete defoliation (except for the 8–10 youngest primordia) is also not inductive. Under short-day conditions additional leaves remaining on the plant that were not infiltrated by TU decreased the percentage of flowering plants but did not fully suppress flower initiation.From these results it is concluded that TU does not act by inhibition of particular metabolic processes concerned in flower initiation or by inhibition of the synthesis of an inhibitor. We suggest that application of TU may lead to synthesis of a floral stimulus in the leaves under short-day conditions also.  相似文献

The effects of infection of root systems by Plasmodiophora brassicae on the translocation of 14C-labelled assimilates from the first and third leaves of cabbage seedlings were investigated. During the early phases of Plasmodium development, there were small differences in the distribution patterns of 14C-labelled assimilate between healthy and infected seedlings. At the end of growth of plasmodia and during resting spore formation, both first and third leaves exported more assimilates than corresponding leaves of healthy seedlings. When the infected roots were dissected into various regions after exposure of the fed leaves to 14CO2, more assimilate accumulated in the club root region than in any other part.  相似文献

The partitioning of14C-assimilates was studied in pigeonpea(Cajanus cajan L.) at vegetative and flowering stages (40 and 70 d after sowing, respectively) exposed to moderate and severe drought induced by withholding the irrigation. At vegetative stage, the ethanol soluble fraction in shoot decreased from 99 to 43.16 % between 0 and 120 h, whereas in underground part it increased from 1% to 56.84 % with maximum amount in nodules (35.51 %). Similar trend was observed in ethanol insoluble fraction. At moderate drought, a significant increase in soluble fraction (11.31 %) in nodules was observed. Stem showed significant reduction of assimilates (13.09 %). After flowering, the assimilates produced in leaves were exported to reproductive parts, especially under drought. In plants recovering from severe drought, 14C in soluble and insoluble fractions in reproductive parts was reduced to 85 % and 43 %, respectively, whereas leaf and nodules showed a significant increase. Thus the assimilate partitioning to different plant parts was dependent on growth stage and affected by drought.  相似文献

The translocation of14C-labelled assimilates from a single leaf in bent and intact apple shoots was studied in varying stages of shoot development. In actively growing shoots14C-labelled assimilates translocated from the treated leaf and accumulated mainly in the shoot apex. In moderately growing apple shoots radioactive assimilates were translocated from the treated leaf in both directions towards and down the shoot. In apple shoots showing only slight growth activity the14C-labelled assimilates were transported from the treated leaf mainly to the base of the shoot, stem and roots. Bending shoots changed the pattern of distribution of radioactive assimilates. Bending actively and moderately growing shoots resulted in higher concentration of 14-carbon in the shoot apex than in controls. In slowly growing and non-growing apple shoots bending caused a higher accumulation of radioactive assimilates in the bent section than in an equal section of control shoots.  相似文献
