The ultrastructure of the bloodstream form of Cryptobia salmositica in rainbow trout was examined during the acute phase of experimental infection. The arrangement of the major groupings of cytoplasmic microtubules originating near the basal bodies is similar to that in other bodonids. The cytostome is reinforced both by pellicular microtubules and an electron-dense plaque. Certain microtubules associated with the flagellar pocket serve as nucleating sites for pellicular microtubules. A flagellar rootlet, consisting of two parallel fibers which are bound together intermittently by electron-dense plaques, curves posteriorly from the basal body of the recurrent flagellum towards the kinetoplast. The basal body associated plaque on the kinetoplast membranes is duplicated at the same time as the basal bodies. Cytoplasmic microtubules are found in association with the plaque and the outer kinetoplastic membrane. A pulsatile vacuole, described for the first time in a hemoparasitic cryptobiid, lies adjacent to the post-flagellar pit. Smaller, interconnected vesicles of the spongiome are continuous with the pulsatile vacuole. Since a pulsatile vacuole occurs not only in free-living and ectoparasitic cryptobiids but in the hemoparasitic (=trypanoplasm) forms as well, this is no longer a character by which the genus Trypanoplasma may be separated from the genus Cryptobia. Possession of this osmoregulatory complex may allow the organism to survive outside of a host and fulfill a monoxenous life cycle, in addition to the usual heteroxenous cycle involving a leech as vector.  相似文献

Isometamidium chloride (Samorin) is therapeutic in rainbow trout (Oncorhynchus mykiss) during preclinical and chronic cryptobiosis. However, the toxic mechanism of isometamidium on Cryptobia salmositica has not been elucidated. The objective of the present study was to examine the in vitro effects of isometamidium on C. salmositica. Under in vitro conditions, isometamidium chloride reduced the infectivity of C. salmositica suspended in whole fish blood. It accumulated rapidly in the kinetoplast (within 1 min) and caused disruption and decantenation of kinetoplast DNA. The in vitro cryptobiacidal activity of isometamidium was reduced when parasites were incubated in medium containing serum supplement, suggesting that isometamidium also binds to plasma proteins. Isometamidium altered glycoprotein receptors (epitopes) for antibodies on the surface of C. salmositica and thus protected some of the parasites from lysis by complement-fixing antibodies. In vitro oxygen consumption and carbon dioxide production decreased in drug-exposed C. salmositica, with increased products of glycolysis, i.e., lactate and pyruvate, after exposure to isometamidium. This suggests that some C. salmositica switched from aerobic respiration to glycolysis when the mitochondrion was damaged by isometamidium.  相似文献

Cryptobia salmositica (pathogenic and vaccine strains), Cryptobia bullocki (pathogenic), and Cryptobia catostomi (nonpathogenic) have similar oxygen consumption rates (0.17 +/- 0.01 nm O2/10(6) parasites). Incubation with sodium azide (5 microliters of a 1-M solution to 1 ml of parasite suspension, i.e., a 5-mM final concentration) reduced the oxygen consumption by approximately 4.5-fold. Motility of the parasites was also greatly reduced in sodium azide. The oxygen consumption and motility of the parasites returned to preazide treatment levels when the azide was removed even after 24 hr of incubation in sodium azide. The activities of hexokinase, pyruvate kinase, and cytochrome C oxidase were not detected in the 3 species of Cryptobia.  相似文献

Whole cell lysates of pathogenic and nonpathogenic strains of Cryptobia salmositica were subjected to subcellular fractionation using differential and isopycnic centrifugation in sucrose. The glycolytic enzymes hexokinase, fructose-1,6-biphosphate aldolase, triosephosphate isomerase, glucosephosphate isomerase and glyceraldehyde-3-phosphate-dehydrogenase and the peroxisomal enzyme catalase were associated with a microbody that had a buoyant density in sucrose of 1.21 g cm-3. Lactate dehydrogenase was detected in whole cell lysates, but not in purified organelles. A microbody with a positive reaction for catalase was detected in electron microscope sections of the pathogenic and nonpathogenic strains. These catalase-containing microbodies fused with lipid bodies and vacuoles, arose by division from pre-existing microbodies and expelled their contents into the cytoplasm of the cell. Both strains also modified the catalase content in their microbodies. Under aerobic conditions, they metabolized glucose to pyruvate and lactate. We conclude that part of the glycolytic pathway in C. salmositica is compartmentalized in a microbody called the glycosome.  相似文献

A serodiagnostic technique, microscopic immuno-substrate-enzyme technique (MISET), is described using the Cryptobia-Oncorhynchus mykiss system. The reactions of specific antibodies, phosphatase-labeled antibody, and substrate with subsequent color development on the parasite (cell membrane, flagella, kinetoplast, and nucleus) are observed using light microscopy. Using this technique, humoral response to Cryptobia salmositica was detected in 2 of 10 O. mykiss 7 days after infection. Subsequently, antibodies were detected in all 10 infected fish. Parasites cultured in minimum essential medium or from experimentally infected pink salmon, Oncorhynchus gobuscha, worked equally well. Antibodies eluted from antisera or whole blood dried on filter paper (stored at -20 C) gave similar reactions as those from antisera stored in containers. MISET is at least as sensitive as the indirect fluorescent antibody technique. When MISET is modified and adapted for other parasitic diseases (e.g., for those of medical and or of economic importance), it may be useful especially in smaller centers or in developing countries where equipment cost, maintenance of equipment, operating costs, and well trained personnel are important considerations. Because MISET worked with dried whole blood or serum on filter paper it may also be a useful field technique for large scale seroepidemiological surveys.  相似文献

Studies were conducted primarily to ascertain the mode of transmission of Cryptobia dahli parasitizing the digestive tract of lumpfish (Cyclopterus lumpus). Another flagellate, morphologically similar to C. dahli, was also observed in the gut of a deepsea fish (Macrourus berglax). Several invertebrates, which are food for lumpfish, were examined for flagellates, but were neither infected nor showed evidence of cystic stages. Parasites were more abundant in the stomach, especially at about pH 5, than in other areas of the digestive tract. Transmission was achieved by pipetting the parasites into the stomach of uninfected fish, by feeding food contaminated with flagellates, and also by holding infected and uninfected fish in the same aquarium. In nature, lumpfish probably acquire parasites during winter when they aggregate and regurgitate into seawater because parasites can survive for short periods outside their host.  相似文献

The incidence of skeletal anomalies could be used as an indicator of the “quality” of rearing conditions as these anomalies are thought to result from the inability of homeostatic mechanisms to compensate for environmentally-induced stress and/or altered genetic factors. Identification of rearing conditions that lower the rate of anomalies can be an important step toward profitable aquaculture as malformed market-size fish have to be discarded, thus reducing fish farmers’ profits. In this study, the occurrence of skeletal anomalies in adult rainbow trout grown under intensive and organic conditions was monitored. As organic aquaculture animal production is in its early stages, organic broodstock is not available in sufficient quantities. Non-organic juveniles could, therefore, be used for on-growing purposes in organic aquaculture production cycle. Thus, the adult fish analysed in this study experienced intensive conditions during juvenile rearing. Significant differences in the pattern of anomalies were detected between organically and intensively-ongrown specimens, although the occurrence of severe, commercially important anomalies, affecting 2–12.5% of individuals, was comparable in the two systems. Thus, organic aquaculture needs to be improved in order to significantly reduce the incidence of severe anomalies in rainbow trout.  相似文献

The mating behavior of Mahanarva spectabilis (Distant) was studied under laboratory conditions. Virgin adults were individually maintained in transparent cages for observation of the mating sequence. Mating behavior of 23 couples was monitored during the photophase portion of the day. Copulation was observed in 17 of the 23 pairs. The mating sequence includes male approaching the female, mounting of the female, and copulation. Of the 17 mating pairs, 70.6 % mated on the second day after emergence. Most of the copulations began between 8 and 9 h after the onset of photophase. The average duration of copulation was 268?±?24.9 min, and most pairs mated once or twice during their lifetimes. The mating behavior of M. spectabilis is important to understand because it determines the age and ideal time for further behavioral testing, which is essential for determining the cues involved in the communication system of the species.  相似文献

We examined the generation time and the house renewal rate ofOikopleura dioica under various conditions. Animals were fedtwo flagellates, Isochrysis galbana and Tetraselmis sp., withconcurrent determination of the carbon contents of body andhouse to estimate house production. The generation time was6 days at 15°C, 4 days at 20°C and 3 days at 25°Cat both 25 and 30 p.s.u. with a food concentration of 4 x 104cells ml–1. The carbon content of newly secreted housesranged from 0.5 to 0.8 µg, corresponding to 15.3 ±4.8% of body carbon. The house renewal rates increased withincreasing temperature and decreasing salinity. Food concentrationsranging from 100 to 16 x 104 cells ml–1, body size andlight condition had no effect on house renewal rate. Cloggingof the inlet filter by adding the large diatom Ditylum sol causedan increase in house renewal rates. The total number and carboncontent of houses during an animal's lifetime ranged from 46to 53 houses and from 6.5 to 10.4 µg, respectively. Sincedaily house production calculated for the O. dioica populationcorresponded to 130–290% of its biomass and daily discardedhouse materials corresponded to 490–1100% of the biomass,this organism must play an important role as a producer of macroscopicaggregates.  相似文献

We studied the genomic organization of Hox genes in Atlantic salmon (Salmo salar) and made comparisons to that in rainbow trout (Oncorhynchus mykiss), another member of the family Salmonidae. We used these two species to test the hypothesis that the Hox genes would provide evidence for a fourth round of duplication (4R) of this gene family given the recent polyploid ancestry of the salmonid fish. Thirteen putative Hox clusters were identified and 10 of these complexes were localized to the current Atlantic salmon genetic map. Syntenic regions with the rainbow trout linkage map were detected and further homologies and homeologies are suggested. We propose that the common ancestor of Atlantic salmon and rainbow trout possessed at least 14 clusters of Hox genes, and additional clusters cannot be ruled out. Salmonid Hox cluster complements seem to be more similar to those of zebrafish (Danio rerio) than medaka (Oryzias latipes) or pufferfish (Sphoeroides nephelus and Takifugu rubripes), as both Atlantic salmon and rainbow trout have retained HoxCb ortholog, which has been lost in medaka and pufferfish but not in zebrafish. However, our data suggest that phylogenetically, the homologous genes within each cluster express mosaic relationships among the teleosts tested and, thus, leave unresolved the interfamilial relationships among these taxa. Phospholipid which is able to induce the differentiation of some undifferentiated tumour cells was isolated from the rainbow trout (Salmo gairdneri) embryos. As HPLC gave only one peak, the phospholipid seemed to be completely purified. However, the spectra of SIMS and FD-MS have shown that it was a mixture of two molecular species. By the chemical and enzymic studies their structures were determined as C16:0/C22:6-and C18:1/C22:6-phosphatidyl cholines. The isolated and structurally elucidated phosphatidyl cholines induce haemoglobin synthesis in murine erythroleukemia cells and a rapid decrease in alkaline phosphatase activity in teratocarcinoma cells. Some phosphatidyl cholines which are commercially available have shown no or conspicuously low activities. Recently we isolated another differentiation-inducing substance(s), which proved to be diglyceride(s) containing the completely same fatty acids as those in the above mentioned phosphatidyl cholines, that is, C16:0/C22:6-and C18:1/C22:6. These results suggest that the lipids containing C22:6 fatty acid or this fatty acid itself might play an important part in the differentiation and development.  相似文献

The life cycles of two separate populations (colonies A and B) of the rabbit tick, Haemaphysalis leporis-palustris, were studied under laboratory conditions. Domestic New Zealand rabbits, Oryctolagus cuniculus, and wild rabbits, Sylvilagus brasiliensis, were used as hosts for ticks from colony B and only O. cuniculus rabbits were used as hosts for ticks from colony A. Developmental periods were observed in an incubator at 27±1°C and RH 90±5%. Larvae from colonies A and B fed for 8.0±3.7 days and 8.5±1.3 days, respectively, on O. cuniculus. On S. brasiliensis larvae from colony B fed for 7.2±1.3 days. Nymphs from colony A fed for 8.1±1.4 days on O. cuniculus and nymphs from colony B fed for 8.1±1.0 days on S. brasiliensis. Only one engorged nymph from colony B was recovered from O. cuniculus. Females from colony A fed for 20.9±5.9 days on O. cuniculus and females from colony B fed for 18.6±2.4 days on O. cuniculus and 18.7±3.7 days on S. brasiliensis. Engorged larvae from colony A required 13.7±3.7 days to molt while engorged larvae from colony B required 11.8±3.0 and 11.5±1.8 days to molt, after having fed on O. cuniculus and S. brasiliensis, respectively. Engorged nymphs from colonies A and B required 16.3±1.9 days and 14.7±1.4 days to molt, respectively. Engorged females from colonies A and B required 4–7 and 3–5 days, respectively, to start oviposition. Mean egg incubation periods lasted for 33–34 days. For ticks from colony B, host species accounted for significant differences (p<0.05) in larval and nymphal feeding periods, oviposition weights and CEIs. Significant differences (p<0.05) between the two colonies when ticks fed on O. cuniculus were observed for larval and nymphal feeding and premolt periods, engorged female and oviposition weights and conversion efficiency indexes (CEI). S. brasiliensis were always a more suitable host for H. leporis-palustris than O. cuniculus. Significantly more larvae and nymphs engorged and molted when fed on S. brasiliensis (p<0.001). Females fed S. brasiliensis were more successful to lay fertile eggs and showed the highest engorged and egg mass weights, and the highest CEIs. Data of H. leporis-palustris fed on wild rabbits (one of its natural host species) are reported for the first time.  相似文献

Three new species of Rhabdochona Railliet, 1916 are described and illustrated from Salmo gairdneri Richardson (rainbow trout) in freshwater streams in California: Rhabdochona californiemis n. sp., R. paxmani n. sp., and R. satmonis n. sp. Rhabdochona californiensis n. sp. is characterized by 14 anteriorly directed teeth in the prostome, egg devoid of filaments or floats, male and female tail terminus with a single mucro, left (long) spicule slender with a moderate distended podoid terminal end, spicular ratio 1:3.8. Rhabdochona paxmani n. sp. is characterized by 10 teeth in the prostome, eggs with polar floats, left (long) spicule slender with podoid terminus distended and having a minute subterminal spine; right spicule with prominent gorgeret (barb), spicular ratio 1:4.3, male and female tail terminus with a cuticular conical rounded short projection. Rhabdochona salmoni, n. sp. is characterized by 10 teeth anteriorly directed in the prostome, eggs with polar floats, left spicule slender with a distended podoid terminus; right spicule with a sharply indented gorgeret, spicular ratio 1:4.3, male and female tail terminus with a conical or rounded tip.  相似文献

Morphologic and biometric data on bloodstream stages of Trypanosoma melophagium are presented. An increasing parasitemia with 111 trypomastigote stages of T. melophagium were found in Giemsa-stained thin blood smears taken from a splenectomized, cortisone-treated sheep recently infested with Melophagus ovinus infected with T. melophagium. The arithmetic mean and standard deviation in μm of the distances between posterior end and kinetoplast were 14.7 and 2.9, from the kinetoplast to the center of the nucleus 5.1 and 1.1, and from there to the anterior end 19.5 and 1.9. The free flagellum measured 6.0 μm ± 1.6 μm. The median and the range of the central 70% of values (median ± 35%) of the nuclear index were 1.1 and 0.9–1.2 and of the kinetoplastic index 3.8 and 3.3–4.9. The same data in μm for the maximal width were 3.1 and 2.1–4.6, and for the width at the level of the nucleus 2.9 and 2.2–4.6. The larger and smaller diameters of the nucleus measured 2.6 (2.2–3.7) μm and 1.7 (1.3–1.7) μm, respectively. The corresponding kinetoplast diameters were 1.1 (0.9–1.3) μm and 0.9 (0.6–0.9) μm, respectively.  相似文献

The leech Calliobdella vivida (Verrill) is the vector of Trypanoplasma bullocki. At 10°C, infective-stage flagellates were first present in the leech's proboscis sheath five days after feeding. At 5°C, infective-stage flagellates were not present in the leech's proboscis sheath until 10 days after feeding, but at 20°C, flagellates were located there as early as 24 h after feeding. Infected leeches retained flagellates through three subsequent feeds on uninfected fish. When flagellates were first observed in hogchoker, Trinectes maculatus (Bloch & Schneider), they were much larger than infective stages from the leech. Average flagellate length then decreased during early acute phase, but gradually increased thereafter. Peak parasitemia was greater in a hogchoker inoculated by only one leech but held at colder temperature than in a hogchoker inoculated by 45 leeches, suggesting that temperature may be more important than inoculum in determining peak parasitemia. Cell division in the fish host is described. SEM studies of fish blood flagellates revealed a pre-oral ridge and a cytostome.  相似文献

We examined the genomic organization of Hox genes in rainbow trout (Oncorhynchus mykiss), a tetraploid teleost derivative species, in order to test models of presumptive genomic duplications during vertebrate evolution. Thirteen putative clusters were localized in the current rainbow trout genetic map; however, analysis of the sequence data suggests the presence of at least 14 Hox clusters. Many duplicated genes appear to have been retained in the genome and share a high percentage of amino acid similarity with one another. We characterized two Hox genes located within the HoxCb cluster that may have been lost independently in other teleost species studied to date. Finally, we identified conserved syntenic blocks between salmonids and human, and provide data supporting two new linkage group homeologies (i.e., RT-3/16, RT-12/29) and three previously described homeologies (RT-2/9, RT-17/22, and RT-27/31) in rainbow trout.
