Three hundred and sixteen (35%) of the American-born Japanese children whose height, weight, and skeletal age were recorded in 1956–57 were reexamined as young adults between 1968 and 1974, when they were found to be taller, heavier, and shorter legged than men and women in Japan who were born in the same years as they. These differences between the American-born and the native Japanese adults were relatively smaller than they had been during childhood, due to both an acceleration in the growth rate of the native Japanese and a concomitant decline in that of the American-born Japanese during the intervening years. A comparison of our 1956–57 data with Kondo and Eto's findings in Los Angeles in 1971 shows that there has been very little increase in the size of California-Japanese children since 1956. The curves of average heights of native Japanese children 6 to 20 years of age, at 10-year intervals from 1900 to 1970, disclosed the changing rate at which they grew during different decades of that period. Those curves and other data discussed in this paper provide additional evidence of the biological superiority of the human female as compared with the male. 相似文献

Many of the deaths caused by tuberculosis (TB) in the world are due to wrong or late diagnosis. The World Health Organization (WHO) calls for better and cheaper TB tests method for this reason. In this paper, a new multi-channel series piezoelectric quartz crystal (MSPQC) sensor system was developed for rapid growth and detection of Mycobacterium tuberculosis. It is an automatic continuous monitoring system. The system was used to detect TB based on the volatile metabolic products NH(3) and CO(2) during the growth of M. tuberculosis. The metabolic products, diffusing from the medium into the KOH absorbing solution, resulted in the conductance change of the absorbing solution detected by the MSPQC sensitively. The frequency shift versus time response curves were recorded by self-developed software. Frequency detection time (FDT) corresponding to -100Hz in frequency shift value was used as a parameter to quantitatively determine M. tuberculosis H37Ra (an avirulent strain). H37Ra and 40 strains clinic positive samples were detected by the proposed system successfully. As for H37Ra, the FDT had a linear relationship with the logarithm of its initial concentration in the range of 10(2)-10(7) colony forming units (cfu)ml(-1) (R=-0.998) and the detection limit was low to 10cfuml(-1). 4% NaOH solution that can kill contaminating microorganisms and make M. tuberculosis alive was used as pretreatment reagent to provide selectivity to this method. Comparative tests were also carried out by using BACTECtrade mark MGITtrade mark 960 and conventional Lowenstein-Jensen (L-J) slants. The results showed that the proposed system was quicker than BACTECtrade mark MGITtrade mark 960 and it is also cheaper and will be widely used in TB tests in the world. 相似文献

A simultaneous model for analysis of net energy intake and growth curves is presented, viewing the animal's responses as a two dimensional outcome. The model is derived from four assumptions: (1) the intake is a quadratic function of metabolic weight; (2) the rate of body energy accretion represents the difference between intake and maintenance; (3) the relationship between body weight and body energy is allometric and (4) animal intrinsic variability affects the outcomes so the intake and growth trajectories are realizations of a stochastic process. Data on cumulated net energy intake and body weight measurements registered from weaning to maturity were available for 13 pigs. The model was fitted separately to 13 datasets. Furthermore, slaughter data obtained from 170 littermates was available for validation of the model. The parameters of the model were estimated by maximum likelihood within a stochastic state space model framework where a transform-both-sides approach was adopted to obtain constant variance. A suitable autocorrelation structure was generated by the stochastic process formulation. The pigs’ capacity for intake and growth were quantified by eight parameters: body weight at maximum rate of intake (149-281 kg); maximum rate of intake (25.7-35.7 MJ/day); metabolic body size exponent (fixed: 0.75); the daily maintenance requirement per kg metabolic body size (0.232-0.303 MJ/(day×kg0.75)); reciprocal scaled energy density ; a dimensional exponent, θ6 (0.730-0.867); coefficient for animal intrinsic variability in intake (0.120-0.248 MJ0.5) and coefficient for animal intrinsic variability in growth (0.029-0.065 kg0.5). Model parameter values for maintenance requirements and body energy gains were in good agreement with those obtained from slaughter data. In conclusion, the model provides biologically relevant parameter values, which cannot be derived by traditional analysis of growth and energy intake data. 相似文献

The prenatal growth of the macaque craniofacial skeleton is described using lateral radiographs of 82 fetal and 25 neonatal Macaca nemestrina whose known gestational ages range from 50 to 186 days. The ossification sequence of the craniofacial bones resembles that in the human fetus. During gestation, the macaque neurocranium loses its round, globular shape, becoming flattened and elongated in an anteroposterior direction. In contrast, the morphologic pattern of the face is established early in fetal life, and little change takes place during the remaining prenatal period. The macaque craniofacial dimensions develop along the general skeletal growth pattern, unlike the human craniofacial dimensions, which follow an intermediate pattern between the neural and general skeletal patterns. However, despite minor differences, the macaque and human fetal faces follow the same basic patterns of growth. 相似文献

Glucose-limited, continuous cultures (dilution rate 0.1 h-1) of Streptococcus bovis JB1 fermented glucose at a rate of 3.9 mol mg protein-1 h-1 and produced acctate, formate and ethanol. Based on a maximum ATP yield of 32 cells/mol ATP (Stouthamer 1973) and 3 ATP/glucose, the theoretical glucose consumption for growth would have been 2.1 mol mg protein-1 h-1. Because the maintenance energy requirement was 1.7 mol/mg protein/h (Russell and Baldwin 1979), virtually all of the glucose consumption could be explained by growth and maintenance and the YATP was 30. Glucose-limited, continuous cultures produced heat at a rate of 0.29 mW/mg protein, and this value was similar to the enthalpy change of the fermentation (0.32 mW/mg protein). Batch cultures (specific growth rate 2.0 h-1) fermented glucose at a rate of 81 mol mg protein-1 h-1, and produced only lactate. The heat production was in close agreement with the theoretical enthalpy change (1.72 versus 1.70 mW/mg protein), but only 80% of the glucose consumption could be accounted by growth and maintenance. The YATP of the batch cultures was 25. Nitrogen-limited, glucose-excess, non-growing cultures fermented glucose at a rate of 6.9 mol mg protein-1 h-1, and virtually all of the enthalpy for this homolactic fermentation could be accounted as heat (0.17 mW/mg protein). The nitrogenlimited cultures had a membrane potential of 150 mV, and nearly all of the heat production could be explained by a futile cycle of protons through the cell membrane (watts = amperes x voltage where H+/ATP was 3). The membrane voltage of the nitrogen-limited cells was higher than the glucose-limited continuous cultures (150 versus 80 mV), and this difference in voltage explained why nitrogen-limited cultures consumed glucose faster than the maintenance rate. Batch cultures had a membrane potential of 100 mV, and this voltage could not account for increased glucose consumption (more than growth plus maintenance). Canonical correlation analysis was used to test an hypothesized morphological relationship between vault form and cranial capacity relative to length of the chondrocranium. Ninety-five adult male Czech skulls were measured for vault form expressed as length, width and height of the brain case; the chondrocranium was represented by nasion-basion and basion-opisthion lengths. In terms of explained variation, the first and most important dimension of covariation between vault and chondrocranial variables was size. The second most significant dimension of covariation expressed the hypothesized shape relationships—i.e., overall size being equal, the shorter the chondrocranial base relative to cranial capacity, the shorter and wider the vault. Furthermore, the competing hypothesis that vault form is determined by facial length proved untenable since facial length was predictive of vault shape only when measured as prosthion-basion, a measure that incorporates basal length. When corrected for basal length, facial length is unrelated to vault form. The results are consistent with the assumption that phylogenetic and microevolutionary trends toward brachycephaly in man stem from changes in the relationship between two components of skull growth, the chondrocranial base and the brain. 相似文献

We introduce a new method for assessing the extent of residue exposure in proteins. For each atom of every residue a Gaussian-weighted atomic surroundings value (the G-neighborhood) is calculated. A normalized sum of G-neighborhood values over all the atoms of a residue is complementary to conventional surface accessibility characteristics. The G-0neighborhood value of a residue is a sensitive indicator of its location, strongly dependent on the 3D structure of a the protein. Correlations between secondary structures and patterns of G-neighborhood values for six different protein molecules are discussed. Comparison of the distribution of hydrophobic and charged residues in the 3D structure for the alcohol-soluble protein crambin and that of five water-soluble proteins (cytochrome c, flavodoxin, myoglobin, rhodanese, and Bence–Jones protein) shows striking differences in their G-neighborhood patterns. Contacts between the prosthetic group and the peptide portion of a protein as well as protein interdomain contacts and monomer–monomer contacts are characterized. © 1993 Wiley-Liss, Inc. 相似文献

Lactobacillus helveticus was cultivated batchwise on whey permeate supplemented with 2–30g yeast extract l-1. For low supplementation levels, growth was slow, and a long maintenance phase in the absence of any cell autolysis was observed. When the yeast extract concentration was increased, growth time increased slightly; under these circumstances, a large increase of maximum biomass concentration was recorded, indicating a clear nitrogen source starvation at low supplement concentration. At the same time the length of the maintenance phase decreased; for a 30gl-1 supplementation, it vanished and a death phase was observed instead: this behaviour highlighted a shift from nitrogen to carbon source starvation. For more and more supplemented media, the final lactic acid concentration was constant, while fermentation time decreased significantly; a growth-associated production phase was followed by a maintenance production phase. The part played by the former mecha nism was under tight control of yeast extract concentration: for 30gl-1 supplementation, 83% of total lactic acid was provided by a growth-linked production process. 相似文献

Aminoacyl tRNA synthetases are enzymes that specifically attach amino acids to cognate tRNAs for use in the ribosomal stage of translation. For many aminoacyl tRNA synthetases, the required level of amino acid specificity is achieved either by specific hydrolysis of misactivated aminoacyl-adenylate intermediate (pre-transfer editing) or by hydrolysis of the mischarged aminoacyl-tRNA (post-transfer editing). To investigate the mechanism of post-transfer editing of alanine by prolyl-tRNA synthetase from the pathogenic bacteria Enterococcus faecalis, we used molecular modeling, molecular dynamic simulations, quantum mechanical (QM) calculations, site-directed mutagenesis of the enzyme, and tRNA modification. The results support a new tRNA-assisted mechanism of hydrolysis of misacylated Ala-tRNAPro. The most important functional element of this catalytic mechanism is the 2′-OH group of the terminal adenosine 76 of Ala-tRNAPro, which forms an intramolecular hydrogen bond with the carbonyl group of the alanine residue, strongly facilitating hydrolysis. Hydrolysis was shown by QM methods to proceed via a general acid-base catalysis mechanism involving two functionally distinct water molecules. The transition state of the reaction was identified. Amino acid residues of the editing active site participate in the coordination of substrate and both attacking and assisting water molecules, performing the proton transfer to the 3′-O atom of A76. 相似文献

Angiogenesis, a key step in many physiological and pathological processes, involves proteolysis of the extracellular matrix. To study the role of two enzymatic families, serine-proteases and matrix metalloproteases in angiogenesis, we have adapted to the mouse, the aortic ring assay initially developed in the rat. The use of deficient mice allowed us to demonstrate that PAI-1 is essential for angiogenesis while the absence of an MMP, MMP-11, did not affect vessel sprouting. We report here that this model is attractive to elucidate the cellular and molecular mechanisms of angiogenesis, to identify, characterise or screen “pro- or anti-angiogenic agents that could be used for the treatment of angiogenesis-dependent diseases. A new potentially tetradentate (N4) Schiff base ligand (L), 1,9,12,20-tetraazatetracyclo[18.2.2.02,7.014,19]tetracosa-2(7),3,5,8,12,14(19),15,17-octaene containing a piperazine moiety is described. Macrocyclic Schiff base complexes, [NiL](ClO4)2 (1) and [CuL](ClO4)2 (2) have been obtained from equimolar amounts of ligand (L) with nickel(II) and copper(II) metal ions. While the equilibrium reaction in the presence of cobalt(II) and zinc(II) metal ions with ligand L in a 1:1 molar ratio yielded the open-chain Schiff base complexes, [CoL′](ClO4)2 (3) and [ZnL′](ClO4)2 (4) containing two terminal primary amino groups. The ligand L′ is 1,4-bis(2-(2-aminoethyliminomethyl)phenyl)piperazine. The crystal structures of (1) and (4) have been also determined by X-ray diffraction. It was shown that the Ni(II) is coordinated to the ligand L by two nitrogen atoms of piperazine group and two nitrogen atoms of the imine groups, in a slightly distorted square-planar geometry. Also single crystal X-ray analysis of (4) confirmed a distorted octahedral arrangement in the vicinity of Zn atom with N6 donor set. The spectroscopic characterization of all complexes is consistent with their crystal structures. 相似文献

ATP, AMP and β-GP on in vitro MV-calcification was studied. O-cresolpthalein complexone assay with non-radioactive calcium demonstrated that AMP or β-GP were more effective in promoting
calcium deposition by isolated MVs than ATP. The application of this nonradioactive technique, which is highly sensitive and
simple, would offer a useful alternative approach to the routinely used radiometric biomineralization assay which employs
radioactive 45Ca.

Many species of marine animals have larval stages whose rates of growth in the plankton are regulated by complex combinations of biological and environmental factors. In this study, we focus on the physiological bases that underlie endogenous variation in growth potential of larvae. Our approach was based on experimental crosses of gravid adults from pedigreed families of the Pacific oyster, Crassostrea gigas. This produced large numbers of larvae with different growth rates when reared under similar environmental conditions of food and temperature. A total of 35 larval families were reared to test hypotheses regarding the physiological bases of growth variation. Growth rate of these larval families varied over a five-fold range, from 3.4 (± 0.5, S.E.) to 17.6 (± 0.6) μm day− 1. The suite of integrated measurements applied to study growth variation included size, biochemical compositions, rates of particulate and dissolved nutrient acquisition, absorption efficiencies, respiration rates and enzyme activities. We show that a complex set of physiological processes regulated differences in genetically determined growth rates of larvae. One-half of the energy required for faster growth came from an enhanced, size-specific feeding ability. Differences in absorption rates were not significant for slow- and fast-growing larvae, nor were differences in size-specific respiration rates. Metabolic processes accounted for the additional 50% of the energy “savings” required to explain enhanced growth rates. We propose that different protein depositional efficiencies could account for this energy saving. Quantitative analyses of the endogenous physiological factors that cause variation in growth rate will allow for a more sophisticated understanding of growth, survival and recruitment potential of larvae. 相似文献

After 12 and 18 months of daily wastewater discharge into mangrove plots in Mayotte Island, SW Indian Ocean, leaf pigment content, photosynthesis rate and growth of Rhizophora mucronata and Ceriops tagal mangrove trees were evaluated and compared with similar individuals from control plots. Chlorophyll and carotenoid contents, measured using an HPLC analyser, were significantly higher in leaves of mangrove trees receiving wastewater discharges. Photosynthesis and transpiration rates, analysed using an LCi portable system, increased significantly for mangrove trees in impacted plots. Measurements of leaf areas, young branch length and propagule length showed significant increases in plots receiving wastewater. These results suggest a beneficial effect of domestic wastewater on R. mucronata and C. tagal mangrove tree functioning. Analyses and observations on mangrove ecosystems as a whole - taking into account water and sediment compartments, crab populations and nitrogen and phosphorus cycles - are nevertheless necessary for evaluation of bioremediation capacities of mangrove ecosystems. 相似文献

Parsimony and neighbor-joining analyses of 16S rDNA nucleotide sequences of 68 species and strains of cyanobacteria and prochlorophytes supported a monophyletic Nostocales, a monophyletic Stigonematales, three independent lineages of prochlorophytes within the cyanobacteria, and a paraphyletic Chroococcales (p<0.0001) and Oscillatoriales (p = 0.0147). Within the Oscillatoriales, the genus Oscillatoria formed an unnatural taxon (p<0.0001) and needs major revisions. Using constraint analysis, the genus Microcystis was found to be monophyletic and the paraphyletic positions of Microcystis elabens and M. holsatica are probably due to long-branch attraction. Further, a separation of Chroococcales based on varying levels of polyunsaturated fatty acids is more consistent with nucleotide-based phylogenies than with existing morphological groupings. It is proposed that Chroococcales be redefined to exclude the genus Microcystis, and that a new order be erected for Microcystis. Finally, it is more parsimonious to assume a common cyanobacterial/prochlorophyte ancestor, than to evoke de novo synthesis of chlb in each prochlorophyte lineage plus in the lineage leading to green chloroplasts. This common ancestor is proposed to have contained both chlorophyll a and b plus phycobilins. Subsequent multiple losses of chlb in cyanobacteria and the loss of chla and phycobilins in prochlorophytes, led to the currently observed pigment distribution. It is therefore, recommended that Prochlorales be reclassified as cyanobacteria. 相似文献

Aims: To investigate the appropriateness of the extended Lambert–Pearson model (ELPM) to model the effect of pH (as hydrogen and hydroxyl ions) over the whole biokinetic pH range in comparison with other available models. Methods and Results: Data for the effect of pH on microbial growth were obtained from the literature or in‐house. Data were examined using several models for pH. Models were compared using the residual mean of squares. Using the ELPM, pH was modelled as hydrogen ions and hydroxyl ions; hence, the model was monotonic in each. The ELPM was able to model data more successfully than the cardinal pH model (CPM) and other models in the majority of cases. Conclusions: Examining the effect of pH as hydrogen and hydroxyl ions has the advantage that the basic form of the ELPM can be retained as each is treated as a distinct antimicrobial effect. With the ELPM, each inhibitor is described by two parameters; from these parameters, the pHmin, pHopt and pHmax can be obtained. Furthermore, the idea of a dose response, absent from other models, becomes important. Significance and Impact of the Study: The CPM is an excellent model for certain situations – where there is a high degree of symmetry between the suboptimal pH and superoptimal pH response and where there are few data points available. The ELPM is more amenable to highly asymmetric behaviour, especially where plateaus of effect around the pH optimum are observed and where the number of data points is not restrictive. 相似文献

Two new amides, aurantiamide and aurantiamide acetate, were isolated from Piper aurantiacum. Their structures were determined as N-(N′-benzoyl-S-phenylalaninyl)-S-phenylalaninol and its acetate, respectively, from chemical and spectroscopic studies. The structures and stereochemistry were confirmed by synthesis. The corresponding diastereoisomers were also synthesized and their spectroscopic properties compared with those of the natural compounds. 相似文献
