The insulin-producing pancreatic islet -cell, characterized by low proliferative potential, is normally not responsive to the polypeptide epidermal growth factor (EGF) or its homolog transforming growth factor (TGF-). Since EGF receptors in other tissues can be up-regulated by other growth factors and by cytokines, we have in this paper investigated whether such a -cell responsiveness to TGF-, or EGF, can be conferred by co-culture with interferon (IFN-), tumor necrosis factor (TNF-) or transforming growth factor (TGF-) in various combinations. To this end, fetal rat pancreatic islets enriched in -cells were isolated and cultured for 3 days with or without 200 pM or 20 nM TGF-. It was found that neither of these TGF- concentrations affected -cell mitogenesis, insulin content or insulin secretion. However, IFN- (1000 U/ml) evoked a modest stimulation of -cell replication, while suppressing insulin secretion and leaving the islet insulin content unaltered. TNF- (1000 U/ml), on the other hand, affected none of these parameters either alone or in any combination with TGF- or IFN-. However, when TNF- or IFN-, either alone or in combination, were combined with the cytokine interleukin-1, this resulted in islet disintegration, whereas the latter cytokine alone did not exert any gross necrotic changes evident by light microscopy. TGF- (500 pM) stimulated insulin secretion but did not influence islet insulin content or -cell mitogenesis either alone or in combination with TGF- (200 pM or 20 nM). In no instance could any mitogenic or secretory response to low or high concentrations of TGF- be conferred by IFN-, TNF- or TGF- whether used alone or in combinations. Summary A fungus identified as Cunninghamella blakesleeana (Lendner) can carry out 15-hydroxylation of lithocholic acid to a new bile acid (3,15-dihydroxy-5-cholanic acid). By optimizing the fermentation conditions, the amount of the product increased from 0.17 g/l to 1.2 g/l. Summary Extracts of hamster-human and mouse-human hybrids, some with translocations involving chromosome 19, have been assayed for both human spleen ferritin (rich in L subunits) and human heart ferritin (rich in H subunits). Hybrid lines retaining part of the long arm of chromosome 19 including the region 19q13.319qter produced human L type ferritin. This confirms the previous assignment of the ferritin gene to chromosome 19 (Caskey et al. 1983). However, lines retaining chromosome 11 were found to contain human H type ferritin suggesting that the gene for the H subunit is on this chromosome. The presence of chromosome 6 was not necessary for the expression of either H or L type human ferritin. It thus seems unlikely that the gene for idiopathic haemochromatosis is a ferritin gene.  相似文献

The present study determined whether putative phosphorylation sites within the M3/M4 cytoplasmic domain of the human 4 subunit of 42 neuronal nicotinic receptors are substrates for cAMP-dependent protein kinase (PKA) or protein kinase C (PKC). Five peptides corresponding to predicted phosphorylation sequences were synthesized, and phosphorylation was compared with standard peptide substrates for each kinase, that is, Kemptide for PKA and glycogen synthase (GS) 1-8 for PKC. VRCRSRSI had the highest affinity for PKA, with a Km of 44.5 M; Kemptide had a Km of 7.7 M. LMKRPSVVK and KARSLSVQH were also phosphorylated by PKA, but had lower affinities of 593 M and 2896 M, respectively. LMKRPSVVK had the highest affinity for PKC with a Km of 182 M; GS 1–8 had a Km of 2.1 M. VRCRSRSI had a comparative affinity for PKC with a Km of 327 M. PCKCTCKK was not phosphorylated by PKA, but was a substrate for PKC with a Km of 1392 M, whereas PGPSCKSP was not phosphorylated by either kinase. Based on these findings, results suggest that Ser-362 and Ser-486 on the human 4 subunit may be phosphorylated by either PKA or PKC, Ser-467 is a putative PKA site, and Thr-532 represents a likely PKC substrate; Ser-421 does not appear to be phosphorylated by either kinase.  相似文献

Zusammenfassung Die Tentakel der Edelkoralle vermögen sich an den Spitzen bis über die dreifache Länge der Normtentakel fadenartig zu verlängern (Fadententakel). Diese Tentakelfäden wickeln sich bei Berührung eines Nahrungsobjekts korkzieherartig auf und nähern dieses durch Kontraktion der Mundöffnung.Damit koordiniert führen die zugehörigen Basalabschnitte (Normtentakel) durch S-förmige Biegungen und eventuelle Verkürzungen die Stelle der gereizten Tentakelfadens präzise zum Mund.Werden die Normtentakel allein gereizt, so antworten sie mit raschem Einschlag zur Mundöffnung. Bei gleichzeitiger Reaktion aller Tentakel wird dadurch eine Tentakel-Faust oder ein Fangkorb gebildet. Bei schwachen Reizen antwortet der Normtentakel anstelle der schlagenden Reaktionen mit langsamen Krümmungen, die das Objekt zur Mundöffnung dirigieren. Unabhängig von der Reaktionsart der Tentakeln wird die Beute vor der Aufnahme stets geprüft und häufig abgelehnt. Ist der Polyp in Freßstimmung, so wird die Nahrung rasch durch das glockenartig erweiterte Schlundrohr geschleust und verschwindet nach wenigen Minuten im Coelenteron.
The tentacle apparatus of the red coral (Corallium rubrum L.) and its role in feeding behaviour
Summary The tentacles of the octocoral Corallium rubrum may become extremely elongated (threadlike-tentacle) the apical part of the norm-tentacle becoming stretched more than three times as much as the base.The threadlike parts of the tentacles contract corcscrew-like when touched by food particles.The stimulated part of the tentacle is brought close to the mouth by coordinated movements of its basal portion. Very often both threadlike-tentacle and norm-tentacle shorten in order to carry the particle to the pharynx.When the norm-tentacles only are touched, they react with quick strokes towards the oral disc. This behaviour, when shown by all tentacles at the same time results in the formation of either a tentacle-fist or a basketlike structure. At slight irritations the norm-tentacle will respond with slow bending movements, which lead the particle towards the mouth. Independent of the type of tentacle reactions the prey is always checked before entering the pharynx, and very often refused.If the polyp decides to accept, food is transported quickly through the elongated pharynx and within a few minutes will have disappeared in the coelenteron.

-Crystallin, a major protein of the eye lens, is known to have chaperone activity in preventing heat-induced aggregation of enzymes and other crystallins. In this study, we investigate the ability of -crystallin to inhibit UV-light-induced aggregation of other lens proteins and the effect of exposure of -crystallin to UV irradiation on its chaperone activity. The chaperone activities of -crystallin preincubated at different temperatures were found to be different and could be correlated with its change in quaternary structure as determined by the fluorescence probe ANS (8-anilo-1-naphthalene sulfonate). -Crystallin can inhibit the aggregation of -crystallin from UV irradiation at room temperature, and the preheated -crystallins provide more protection than the native one. Upon irradiation by UV light, -crystallin gradually lost its ability to protect -crystallin against thermal aggregation. The loss of the chaperone efficacy of -crystallin to protect other lens proteins may shed light on human cataract formation induced by long-term exposure to UV irradiation.  相似文献

Summary Adult female mice of the sensitive NMRI/Han strain ovulate diploid oocytes after gonadotropin treatment. Other mouse strains are non-sensitive with respect to the ovulation of such diploid oocytes. In this study we combined the impaired ovarian situation in the XO karyotype with the trait diploidy, which is determined genetically, by mating Ta/O (Ta=Tabby) females of C3Hx101 background to males of the NMRI/Han strain. The adult female F1 hybrids were stimulated to ovulation by gonadotropins and identified by their karyotype (XX or XO). The cytogenetic analysis of ovulated oocytes revealed a low level of diploidy in the XX littermates (1.0%), but a very high level in females with the XO karyotype (24.6%). All of the XO females ovulated at least one diploid oocyte. We suggest that it is the XO status which drastically impairs meiosis I in our gonadotropin-sensitive F1 females due to (1) alterations of the developmental program within the oocyte, (2) a disturbed communication between oocyte and follicle, (3) a preferential maturation and ovulation of follicles at risk, or (4) an exceptional recruitment of many such follicles, by, e.g., a premature responsiveness to gonadotropins in our XO females. A novel mono-sulfated glycosphingolipid based on the gangliotriaose core structure was isolated from rat kidney. The isolation procedure involved extraction of lipids with chloroform/methanol, mild alkaline methanolysis, column chromatographies with anion exchangers and silica beads. The structure was characterized by compositional analysis, FTIR spectroscopy, methylation analysis,1H-NMR spectroscopy and negative-ion liquid secondary ion mass spectrometry (LSIMS) using the intact glycolipid and its desulfation product. The two dimensional chemical shift correlated spectroscopy provided information on the sugar sequence as well as anomeric configurations, and indicated the presence of a 3-O-sulfatedN-acetylgalactosamine within the molecule. Negative-ion LSIMS with high- and low-energy collision-induced dissociation defined the sugar sequence and ceramide composition, confirming the presence of a sulfatedN-acetylgalactosamine at the non-reducing terminus. From these results, the complete structure was proposed to be HSO3-3GalNAc1-4Gal1-4Glc1-1Cer (Gg3Cer III3-sulfate, SM2b). The ontogeny of the antennal glands was studied during the embryonic and post-embryonic development of Astacus leptodactylus. The future glands arising from undifferentiated columnar cells were detectable at the metanauplius stage EI 150 m (EI: eye index; approximately 440 m at hatching). The tubule and labyrinth differentiated in embryos at EI 190 m, and the bladder and coelomosac at EI 250 m. At EI 350 m, the tubule lengthened and divided into proximal and distal sub-regions. In later stages, the gland retained the same morpho-anatomy but the differentiation and size of each part increased. The cells of the coelomosac displayed the cytological features of podocytes in late embryonic development at EI 440 m. Only small apical microvilli and a few mitochondria were observable in the labyrinth cells at EI 250 m; by EI 440 m, these cells presented well-shaped apical microvilli, formed bodies, basal infoldings and mitochondria. In the cells of the tubules and bladder, mitochondria and basal infoldings occurred at EI 440 m and EI 250 m, respectively. The differentiation of the tubules and bladder cells suggested that they were involved in active transport at EI 440 m. Following hatching, the differentiation of the cells and the size of the glands increased. The ontogeny of the antennal glands thus starts in early embryos, the specific cellular functional features being differentiated in the various parts of the glands by EI 440 m. Zusammenfassung Der hier beschriebene neue Vertreter der Chytridiaceen lebt als Parasit auf der Oospore von Eudorina elegans. Der Thallus ist eukarpisch, monocentrisch und entwickelt sich intramatrikal und extracellular. Das Sporangium entwickelt ein verhältnismäßig langes Entleerungsröhrchen, was das charakteristischste Gattungsmerkmal dieses Pilzes darstellt. Die geschlechtliche Fortpflanzung verläuft in einem sehr frühen Entwicklungsstadium beider Gametangien. An dem Zoosporenrest des ganz jungen weiblichen Thallus befestigt sich eine männliche Zoospore, die sich dann mit einer Zellhülle überzieht. Nachher bildet sich zwischen den Hüllen der beiden Zoosporenreste ein echter Kopulationsschlauch, worauf die Plasmogamie und danach die Entwicklung einer Dauerspore folgt.  相似文献

SUMMARY 1. We previously reported that angiotensin III modulates noradrenergic neurotransmission in the hypothalamus of the rat. In the present work we studied the effects of angiotensin III on norepinephrine release and tyrosine hydroxylase activity. We also investigated the receptors and intracellular pathways involved in angiotensin III modulation of noradrenergic transmission.2. In rat hypothalamic tissue labeled with [3H]norepinephrine 1, 10, and 100 nM and 1 M losartan (AT1 receptor antagonist) had no effect on basal neuronal norepinephrine release, whereas 10 and 100 nM and 1 M losartan partially diminished norepinephrine secretion evoked by 25 mM KCl. The AT2 receptor antagonist PD 123319 showed no effect either on basal or evoked norepinephrine release. The increase in both basal and evoked norepinephrine output induced by 1 M angiotensin III was blocked by 1 M losartan, but not by 1 M PD 123319.3. The phospholipase C inhibitor 5 M neomicin inhibited the increase in basal and evoked norepinephrine release produced by 1 M angiotensin III.4. Tyrosine hydroxylase activity was increased by 1 M angiotensin III and this effect was blocked by 1 M LST and 5 M neomicin, but not by PD 123319. On the other hand, 1 M angiotensin III enhanced phosphatidyl inositol hydrolysis that was blocked by 1 M losartan and 5 M neomicin. PD 123319 (1 M) did not affect ANG III-induced phosphatidyl inositol hydrolysis enhancement.5. Our results confirm that angiotensin III acts as a modulator of noradrenergic transmission at the hypothalamic level through the AT1-phospholipase C pathway. This enhancement of hypothalamic noradrenergic activity suggests that angiotensin III may act as a central modulator of several biological processes regulated at this level by catecholamines, such as cardiovascular, endocrine, and autonomic functions as well as water and saline homeostasis.  相似文献

This review describes a method by which the human natural anti-Gal antibody can be exploited as an endogenous adjuvant for targeting autologous tumor vaccines to antigen-presenting cells (APCs). Tumor cells remaining in the patient after completion of surgery, radiation, and chemotherapy are the cause of tumor relapse. These residual tumor cells can not be detected by imaging, but their destruction may be feasible by active immunotherapy. Since specific tumor-associated antigens (TAAs) have not been identified for the majority of cancers, irradiated autologous tumor vaccines have been considered as an immunotherapy treatment that may elicit an immune response against the residual tumor cells expressing TAAs. However, tumor cells evolve in cancer patients in a stealthy way, i.e., they are not detected by APCs, even in the form of vaccine. Effective targeting of tumor vaccines for uptake by APCs is a prerequisite for eliciting an effective immune response which requires transport of the vaccine by APCs from the vaccination site to the draining lymph nodes. In the lymph nodes, the APCs transporting the vaccine process and present peptides, including the autologous TAA peptides for activation of the tumor-specific T cells. The required targeting of vaccines to APCs is feasible in humans by the use of anti-Gal. This antibody interacts specifically with the -gal epitope (Gal1-3Gal1-4GlcNAc-R) and is the only known natural IgG antibody to be present in large amounts in all humans who are not severely immunocompromised. The -gal epitope can be synthesized on any type of human tumor cell by the use of recombinant 1,3galactosyltransferase (1,3GT). Solid tumors obtained from surgery are homogenized and their membranes subjected to -gal epitope synthesis. Similarly, -gal epitopes can be synthesized on intact tumor cells from hematological malignancies. Administration of irradiated autologous tumor vaccines processed to express -gal epitopes results in in situ opsonization of the vaccinating cells or cell membranes due to anti-Gal binding to these epitopes. The bound antibody serves to target the autologous tumor vaccine to APCs because the Fc portion of the antibody interacts with Fc receptors on APCs. Since patients receive their own TAAs, the vaccine is customized for autologous TAAs in the individual patient. The repeated vaccination with such autologous tumor vaccines provides the immune system of each patient with an additional opportunity to be effectively activated by the autologous TAAs. This study examines various optimization criteria as potential sources of constraints that eliminate (or at least reduce the degree of) mechanical redundancy in prehension. A model of nonvertical grasping mimicking the experimental conditions of Pataky et al. (current issue) was developed and numerically optimized. Several cost functions compared well with experimental data including energylike functions, entropylike functions, and a motor command function. A tissue deformation function failed to predict finger forces. In the prehension literature, the safety margin (SM) measure has been used to describe grasp quality. We demonstrate here that the SM is an inappropriate measure for nonvertical grasps. We introduce a new measure, the generalized safety margin (GSM), which reduces to the SM for vertical and two-digit grasps. It was found that a close-to-constant GSM accounts for many of the finger force patterns that are observed when grasping an object oriented arbitrarily with respect to the gravity field. It was hypothesized that, when determining finger forces, the CNS assumes that a grasped object is more slippery than it actually is. An operative friction coefficient of approximately 30\% of the actual coefficient accounted for the offset between experimental and optimized data. The data suggest that the CNS utilizes an optimization strategy when coordinating finger forces during grasping.  相似文献

The synthesis of thioglycoside glycosyl donors with a disaccharide -D-Gal-(1 3)-D-GalNAc backbone was studied using the glycosylation of a series of suitably protected 3-monohydroxy- and 3,4-dihydroxyderivatives of phenyl 2-azido-2-deoxy-1-thio-- and 1-thio--D-galactopyranosides by galactosyl bromide, fluoride, and trichloroacetimidate. In the reaction with the monohydroxylated glycosyl acceptor, the process of intermolecular transfer of thiophenyl group from the glycosyl acceptor onto the cation formed from the molecule of glycosyl donor dominated. When glycosylating 3,4-diol under the same conditions, the product of the thiophenyl group transfer dominated or the undesired (1 4), rather than (1 3)-linked, disaccharide product formed. The aglycon transfer was excluded when 4-nitrophenylthio group was substituted for phenylthio group in the galactosyl acceptor molecule. This led to the target disaccharide, 4-nitrophenyl 2-azido-4,6-O-benzylidene-2-deoxy-3-O-(2,3,4,6-tetra-O-acetyl--D-galactopyranosyl)-1-thio--D-galactopyranoside, in 57% yield. This disaccharide product bears nonparticipating azido group in position 2 of galactosamine and can hence be used to form -glycoside bond. Azido group and the aglycon nitro group were simultaneously reduced in this product and then trichloroacetylated, which led to the -glycosyl donor, 4-trichloroacetamidophenyl 4,6-di-O-acetyl-2-deoxy-3-O-(2,3,4,6-tetra-O-acetyl--D-galactopyranosyl)-1-thio-2-trichloroacetamido--D-galactopyranoside, in 62% yield. The resulting glycosyl donor was used in the synthesis of tetrasaccharide asialo-GM1.  相似文献

Conclusion In conclusion, a number of theoretical points need to be made. The first relates to the question of the primacy of the economic infrastructure in determining the shape of social institutions and the direction of social change in society. We accept this theoretical position, but still one may ask, why then in the case of Kuwait is it the state which is now dominant in affecting sociocultural life? While appreciating the analytical value of the Marxist thesis which sees that it is the material economic base which in the final instance determines the evolution of society, at the same time we see that this does not mean that the material infrastructural base will also by necessity dominate society. For our purposes here Godelier's analytical distinction between what he terms infrastructural determination and superstructural domination is quite useful, and provides an adequate explanation as to why the state has come to be dominant in Gulf societies. Godelier argues that in both historical and contemporary comparative cases where superstructures (kinship, religion, the state, etc.) appeared to be dominant in society, such superstructures always functioned as a relation of production. For a social institution and/or an activity to play a dominant role in the functioning and evolution of society, it must necessarily, in addition to its own ostensible purpose and explicit functions, function directly and internally as a relation of production. Godelier's proposed hypothesis turns the analytical focus not so much on what the social relations or institutions are, but rather on what they do, or better, make people do. It is evident in the discussion above that the state structure in Kuwait, due to the peculiarities of the oil economy and other social factors, has come to play (in addition to its explicit political functions) a very important and again explicit economic role. The oil state controls not only the means of production and allocation of wealth, but simultaneously functions as a controller of the relations of production in society.The second point relates to an earlier reference made about George Foster's conceptualization of the image of limited good. Unlike the cognitive conceptual formulation made by Foster two decades ago about peasant society, our argument in this paper emphasizes that the image of the unlimited good is a derivative social and psychocultural phenomenon resulting from the impact of broader economic and historical transformations which have been taking place in recent years in the oil-rich Arab Gulf countries. The major shortcoming in Foster's analysis of the peasants' image of limited good is that he relies only on cognitive formulation and does not go beyond the limitations of derivative analysis.The third point ties in with our note on Foster's formulation. It emphasizes the fact that it is the broader impersonal socioeconomic conditions that in the final analysis produce certain images in a given society at a given time. These conditions also determine, but may not necessarily dominate, the forms and other cultural peculiarities these images may take. In fact, it has recently become noticeable that the image of the unlimited good has begun to shrink in people's minds, especially after the 1982–83 Al-Manakh stock market crash in Kuwait and the recent dramatic downfall in oil prices.Since we are using a Marxist socioeconomic perspective in our analysis one may ask whether there are no contradictions in this Kuwaiti image of the unlimited good. I believe that the absence of discussion on contradictions does not create an analytical gap. We have shown in the preceding discussion that the emergence of the oil welfare state with its tremendous capacities to dominate society as a result of its lavish wealthfarism and its role as a controller of wealth in society has enabled it to accomodate for the rising needs and expectations of a small society. In addition, the existence of other socioeconomic and political conditions and variables—such as smallness, expatriate labor force, national and ethnic loyalties still overshadowing real class loyalties, the short historical period for this social experiment of oil wealth, and so on — has helped in repressing the rise of contradictions along class lines.Moreover, a discourse which shows how it is possible for new socioeconomic conditions to arrest, at least for a given time period, the development of contradictions can competently follow a Marxist mode of analysis. I see, therefore, no analytical disjunction arising from the fact that a Marxist perspective has been used while at the same time we have tried to elucidate the nature of the forces and conditions which have brought about harmony epitomized by the image of the unlimited good. Such an elucidation has also aided us in seeing why it is satisfaction and acceptance of the economicopolitical system, and not contradictions, that have come to prevail in modern Kuwait.We also note at the end of our discussion how change in the larger impersonal socioeconomic conditions since 1984 has begun generating not only new perceptions away from the image of the umlimited good but also immature class contradictions and consciousness expressing themselves in mystified forms of envy of the super-rich or rivalry with expatriates.The last important fact is that perceptions, images and worldviews do not have a functional autonomy of their own. Isolated intact eyespot apparatuses, the photoreceptive organelles involved in blue-light-mediated photoresponses of flagellate green algae, were analyzed regarding their carotenoid composition. Carotenoids from the eyespot apparatuses of Spermatozopsis similis were identified by high-performance liquid chromatography, visible-light absorption spectra, mass spectroscopy and by 1H-nuclear magnetic resonance spectroscopy (carotenes), and compared with those of whole-cell extracts. Both extracts contained ,-carotene, ,-carotene (formerly -carotene), lycopene, lutein, zeaxanthin, violaxanthin and all-E-and 9-Z-neoxanthin. The relative carotenoid compositions, however, differed significantly. A twofold relative increase in the total carotene level was evident in the fraction enriched in eyespot apparatuses. This was mainly due to an increase in the monocyclic ,-carotene and the aliphatic lycopene, whereas the relative content of ,-carotene remained unchanged. On the other hand a relative decrease in the total xanthophyll content, especially of lutein and the epoxidic carotenoid neoxanthin, was observed in the eyespot apparatuses compared with the whole-cell extracts. The decrease of the latter resulted almost solely from a reduction of the 9-Z-rather than the all-E-isomer. The bulk of the carotenes is thought to be localized in the highly organized eyespot lipid globules, which act as a combined quarter-wave interference reflector and absorption screen for the photoreceptor in green algae. The enrichment of ,-carotene and lycopene in the eyespot apparatuses, extending the range of visible light absorption to longer wavelengths, represents an adaptation of the screen to the retinal-based photoreceptor of flagellate green algae and is one of the prerequisites for maximal directional sensitivity of the eyespot apparatus.Abbreviations 1H-NMR nuclear magnetic resonance - IUPAC International Union of Pure and Applied Chemistry - VIS visible absorption spectra This work was supported by the Deutsche Forschungsgemeinschaft (G.K. and M.M.). As a result of earlier breeding efforts, portions of the genome of Basmati 370 have been introgressed into a rice breeding line, B8462T3-710. Cooked-kernel elongation was increased in this breeding line to a level equal to that of Basmati 370. The objective of this study was to identify and locate quantitative trait loci (QTLs) associated with cooked-kernel elongation in an F3 population derived from a cross between B8462T3710 and the reduced-elongation recurrent parent variety, Dellmont. DNA from the parental lines and Basmati 370 as a control, were screened for RFLPs using 170 clones chosen to cover the rice genome at intervals of 8 cM on average. Eighteen markers identified RFLPs common to Basmati 370 and B8462T3-710, but different from Dellmont, suggesting possible associations with kernel elongation. The B8462T3-710/Dellmont F3 population was analyzed for segregation of those RFLPs and for kernel elongation. Analysis of variance of the kernel elongation ratio revealed that two markers, 14.6 cM apart on chromosome 8, are significantly associated with this trait (RZ323 P 0.005, RZ562 P 0.05). Interval mapping suggests a single QTL with a close proximity to RZ323. This QTL was tested in F6 lines derived from the same cross and the presence of the B8462T3-710 segment detected by RZ323 caused a highly significant increase of the kernel elongation ratio (P 0.04). In addition, the QTL for kernel elongation and a gene for aroma, which are major components of the grain quality characteristics of Basmati-type rices, showed linkage. The availability of linked markers to the QTL may facilitate early selection for kernel elongation in rice breeding programs.  相似文献

Summary The intercellular junctions between the pinealocytes of male rats were investigated by freeze-fracture and conventional electron microscopy.Our findings reveal that the intercellular contacts between pineal cells, formerly described as zonulae adhaerentes or zonulae occludentes, are in fact gap junctions which are difficult to characterize in thin sections due to their peculiar geometrical arrangement, which is in the form of fenestrated communicating zonules.The arrangement of these communicating zonules around rudimentary lumina of pineal clusters and rare transitions between tight and gap junctions may point to phylogenetic transformations of occluding into communicating zonules, corresponding with the change of the pineal gland from a sensory to a secretory organ. Zusammenfassung Das olfaktorische Epithel von erwachsenen Katzen wurde nach Perfusionsfixierung mit Glutaraldehyd und Nachfixierung mit Osmiumtetroxyd stereomikroskopisch im Totalpräparat und elektronenmikroskopisch in Schnittserien untersucht. Im Auflichtverfahren zeigt die Regio respiratoria eine blaugrüne und die Regio olfactoria eine seidighelle graublaue Interferenzfarbe. Bei stärkerer lichtmikroskopischer Vergrößerung sind die ausgerichteten Bündel der Sinnesgeißeln zu erkennen. Besondere Aufmerksamkeit wurde dem olfaktorischen Saum gewidmet, der sich in drei Schichten gliedert: 1. In eine Innenzone, die die olfaktorischen Vesikel und die Mikrozotten der Stützzellen beherbergt. 2. In eine Außenzone, in der die distalen Segmente der Sinnesgeißeln liegen und 3. in den strukturdichteren Terminalfilm. Die Geißelenden tauchen von unten in den Film ein und tragen dann auf ihrer Membranoberfläche entweder Filamenteverdichtungen oder sie sind von einem optisch leeren Hof umgeben. Die distalen Geißelsegmente können wahrscheinlich bis 80 lang sein, sie enthalten nur zwei mikrotubuläre Geißelfibrillen. Das proximale Geißelsegment trägt unmittelbar über dem Ursprung im olfaktorischen Vesikel eine Ringmanschette. Unterschiedliche Formen der Sinneszellendkolben sprechen dafür, daß die Perikarya ihr olfaktorisches Bläschen im Rahmen einer stetigen Mauserung ersetzen können. Der vonOkano et al. (1967) als jugendliche Stützzelle angesehene vierte Zelltyp des Riechepithels ist auch bei der Katze vorhanden. Ein weiterer fünfter Zelltyp hat basal im Epithel ein Perikaryon und wie die Sinneszellen einen dünnen peripheren Fortsatz, der mit steifen Mikrozotten in die Innenzone des olfaktorischen Saumes hineinreicht.Es wird diskutiert, wieweit Innenstrukturen des Geißelapparates an den Vorgängen der Aufnahme von Geruchsreizen und der Erregungsbildung beteiligt sein können.
On the olfactory epithelium of the cat
Summary An examination of the olfactory epithelium of adult cats was carried out after perfusion with glutaraldehyde and postfixation with osmium tetroxide. Unsectioned preparations were studied with the stereomicroscope, and serial sections were examined under the electron microscope. With incident illumination light the regio respiratoria shows a blue-green interference colour while the regio olfactoria exhibits a distinctively silky bluegrey shade. A higher lightmicroscopic magnification reveals bundles of olfactory cilia arranged in parallel rows. Particular attention was paid to the fact that the olfactory border is composed of three layers, namely: 1. an inner layer, containing olfactory vesicles and the microvilli of supporting cells; 2. an outer layer with the distal segments of the olfactory cilia; and 3. a dense terminal film. The distal segments of the olfactory cilia are probably up to 80 microns long and usually contain two microtubules. They extend into the dense terminal film, having on their membrane a concentration of filamentous material or being surrounded by an electron opaque halo. The proximal segment of the cilium looks like a kinocilium bearing a circular cuff just above the olfactory vesicle. Different shapes of the olfactory vesicles indicate that the perikarya are capable of replacing their olfactory vesicles from time to time. The juvenile (fourth) cell type described byOkano et al. (1967) is also observed in the cat. A fifth cell type was found in the regio olfactoria of the cat; its cell body lies deep in the olfactory mucosa among the perikarya of the olfactory cells. A thin distal process reaches the inner layer of the olfactory border with straight stiff microvilli.It is also discussed to which extent the internal structure of the cilia is involved in olfactory perception, or in the initiation of a generator potential.

To appraise the usefulness of the enzyme-linked immunosorbent assay (ELISA) technique for examining the serological diversity of slow-growing rhizobia, twelve diverse strains from three countries were examined with four antisera. Eleven of the strains were from the cowpea miscellany, and the twelfth was a Rhizobium japonicum strain. Some cowpea strains showed no antigenic relatedness with each other while others were closely related, and some showed a greater affinity with the R. japonicum strain than with other cowpea strains. All of the strains showed antigenic homology to an isolate from a wild Arachis sp., while two strains isolated from adjacent plants of the same cultivar had little homology. These patterns ofrelatedness and diversity clearly demonstrated the utility of the ELISA method, and so it was used to examine 53 strains isolated from cowpeas grown at three West African locations, Maradi (Niger), Ibadan and Onne (Nigeria). Broad ranges of serological diversity were found in the rhizobia at each location, moreover each population had its own general characteristics. Maradi strains were highly reactive with the five antisera used, Onne strains less so, and Ibadan strains even less so. ELISA reactivity correlated with colony morphology but not with nodulation potential.  相似文献
