Ion-pairing has little effect on ionic activity calculations for weakly mineralized natural water (ionic strength < 2 mM). However, for more strongly mineralized freshwaters, corrections for ion-pairing are necessary if highly accurate ionic activities are required.  相似文献

A considerable proportion of biology teachers in schools and technical colleges are at present using or contemplating using radioactive isotopes for certain experiments in their courses. These experiments (a general list appears at the end of the article) are easy to perform and usually biologically instructive.

For the teacher the experiment naturally does not stop with performing it; solutions have to be prepared, supervision and suggestions have to be given to students concerning data handling, half-life and self-absorption corrections, etc. An awareness of the pitfalls in both technique and result interpretation has to be gained and this is usually not possible even in an intensive one week course that most teachers have attended.

For this reason this article covers the minimum necessary nuclear fundamentals such as levels of activity, specific activity and the use of carrier materials. The way the isotope is assayed and the corrections that need to be made or taken into account to obtain a valid result is also described in detail. A minimum of statistics necessary for such a valid result is similarly covered. From experience gained in running short radio-isotope courses for teachers some common misconceptions that often arise due to lack of course time have also been included.  相似文献

Using the statistical theory of nonequilibrium thermodynamics we explore the nature of nonequilibrium corrections to chemical potentials in simple enzyme-catalyzed reactions. The statistical definition of the chemical potential, which pertains to systems that are at stable steady states, is applied to the Michaelis-Menten reaction scheme in a cellular-sized compartment that communicates with out-side reservoirs. Calculations based on the kinetic parameters for hexokinase and triose phosphate isomerase show that substantial corrections to the chemical potential of product (the order of 25 mV) are possible if the reaction is sufficiently far from equilibrium. The dependence of the corrections to the chemical potentials on the size of the cellular compartment are explored, and the relevance of the corrections for understanding the thermodynamics of metabolites is discussed.  相似文献

As an alternative to the conventional ZAF correction for microprobe analysis, a combined absorption and atomic number correction can be obtained from ø(ρz) curves. This can be easily applied to the analysis of bulk frozen-hydrated biological samples using inorganic standards. It is shown that the method is accurate by analysis of a number of organic and inorganic compounds and gelatine models. Because the corrections vary little with changes in protein concentration over the normal cellular range, iterative ø(ρz) computations are not necessary and the correction procedure is therefore fast and simple.  相似文献

The sensitive specific radioenzymatic procedure for determination of catecholamines originally described from our laboratory by Coyle and Henry (1) has been optimized for use in assay of human plasma levels of dopamine, norepinephrine and epinephrine. Dopamine and the total of norepinephrine and epinephrine are assayed by 0-methylation while norepinephrine is determined by N-methylation. Epinephrine is calculated from the difference between the 0-methylation and N-methylation procedures. In a group of 13 normal subjects, plasma levels of epinephrine were found to be 67 ± 9.2 pg/ml, norepinephrine 208 ± 16.9 pg/ml and dopamine 33 ± 8.1 pg/ml. Dopamine determinations are of low reliability because of relatively high blanks and necessary corrections.  相似文献

The degree of deacetylation (DDA) of chitosan determines the biopolymer's physico-chemical properties and technological applications. pH-Potentiometric titration seems to offer a simple and convenient means of determining DDA. However, to obtain accurate pH-potentiometric DDA values, several factors have to be taken into consideration. We found that the moisture content of the air-dry chitosan samples can be as high as 15%, and a reasonable fraction of this humidity cannot be removed by ordinary drying. Corrections have to be made for the ash content, as in some samples it can be as high as 1% by weight. The method of equivalence point determination was also found to cause systematic variations in the results and in some samples extra acid as high as 1 mol% of the free amino content was also identified. To compensate for the latter effect, the second equivalence point of the titration has to be determined separately and the analytical concentration of the acid be corrected for it. All the corrections listed here are necessary to obtain DDA values that are in reasonable agreement with those obtained from (1)H NMR and IR spectroscopic measurements. The need for these corrections severely limits the usefulness of pH-metry for determining accurate DDA values and thus potentiometry is hardly able to compete with other standard spectroscopic procedures, that is, (1)H NMR spectroscopy.  相似文献

A hot-air sterilizer capable of exposing airborne microorganisms to elevated temperatures with an almost instantaneous heating time was developed and evaluated. With this apparatus, aerosolized Bacillus subtilis var. niger spores were killed in about 0.02 sec when exposed to temperatures above 260 C. This is about 500 times faster than killing times reported by others. Extrapolation and comparison of data on the time and temperature required to klll B. subtilis var. niger spores on surfaces show that approximately the same killing time is required as is necessary for spores in air, if corrections are made for the heating time of the surface.  相似文献

Four different luminescent lanthanide complexes have been studied with respect to multiphoton excitation using near-infrared femtosecond pulses. The method for measuring action cross sections of two-photon excited fluorescence in solution relative to a known standard is reviewed. Two refractive index-related corrections are necessary in this method: one for the multiphoton excitation process, the other for the collection of the emitted light. It has been found that (2,4,6-trimethoxyphenyl)dipicolinic acid and Michler's ketone are reasonable sensitisers of two-photon excited lanthanide luminescence in solution, whereas dipicolinic acid and carbostyril-124 do not give rise to any detectable two-photon excited lanthanide luminescence using modest excitation powers (<20 mW focused at the sample) in the 700-1000 nm range.  相似文献

By assuming that the electric charge of proteins is primarily determined by the hydrogen ion activity of the medium, and by making corrections when necessary for the effect of salt, it is possible to derive a simple relationship between the electric mobility of proteins and the effective reciprocal thickness of the electric double layer. The decrease in electric mobility of proteins in solutions is readily predicted for gelatin in acetate buffers and for egg albumin in the presence of the alkali halides, of acetate buffers, and of hydrochloric acid on the basis of the assumptions made.  相似文献

In this paper previous binding studies of Mg2+ and Mn2+ ions to tRNA's are reconsidered. Binding data of some re- presentative examples are interpreted including interactions between charges located on the macroion. Both curved and bell-shaped Scatchard plots can be accounted for quantitatively if corrections are made for electrostatic interactions and, if necessary, for the effect of conformational changes on these interactions. It appears that there is no need to invoke more than one class of binding sites on tRNA's, meaning that the experimental binding data can be described using the same in- trinsic pK value for all phosphate groups.  相似文献

To fulfill the purpose as a sink for trace elements, soils must not be overloaded with As and Cd. Therefore, it is necessary to get knowledge of the sorption capacities of soils on a regional scale. The determination of these sorption capacities for large areas is, however, impeded by the great expenditure of laboratory work involved. With data presented here retention capacities for cadmium and arsenic from routinely determined soil parameters are estimated. In batch experiments the sorption behaviour of 40 soils from the area of Freiberg/Saxony in Germany was examined. The obtained sorption isotherms from the laboratory were fitted to the Freundlich equation (S = k*Cm). The two constants (k, m) of this equation were used for multiple linear regression to correlate the sorption capacity and the soil parameters, namely clay content, pH value, total organic carbon, and dithionite extractable Fe contents. Due to long lasting ore mining of Freiberg there exist high background levels in that area for the two surveyed elements As and Cd. Therefore, this study offers two different mathematical procedures to take these contaminations into account. Thus the experimental data were corrected before they were fitted to Freundlich and pedotransfer equations were determined. Using the transfer equation, parameter k and m for cadmium sorption could be estimated with statistical certainties of 91% and 61% (adjusted R2), respectively, whereas the predictability for the arsenic sorption is not practicable because achieved R2 values are very low (17% and 7%). When the assay of maize leaf phosphoenolpyruvate carboxylase (EC 4.1.1.31) activity is started with phosphoenolpyruvate, much lower reaction rates are obtained as compared to the enzyme-initiated reaction. The difference is due to the lability of the dilute enzyme in the absence of its substrate and is increased with incubation time in the absence of substrate or stabilizers. The activation of the enzyme by glucose-6-phosphate is overestimated with the substrate-initiated assay since a part of the apparent activation is due to stabilization of the enzymic activity by this effector during the minus-substrate preincubation. In contrast, the inhibitory effect of malate is underestimated when the reaction is started with the substrate. Using gas chromatography/mass spectrometry we have developed a method for the simultaneous determination of six plasma steroids: testosterone, 4-androstenedione, 17 alpha-hydroxyprogesterone, 5 alpha-androstane-3 alpha,17 beta-diol, 5 alpha-dihydrotestosterone, and dehydroepiandrosterone. For each analyte, a deuterium-labeled internal standard was used for quantification. Due to the high isotopic purity of our standards, no complex corrections for isotope contributions were necessary. The procedure provides a sensitive and specific technique with good accuracy and precision.  相似文献

The surgical correction of gynecomastia presents particular problems in the body builder who takes medical treatment of anabolic steroids. In 38 corrections, no fat suction was necessary and only an extensive surgical extirpation of the gland was performed. Superior cosmetic results were obtained with this treatment, with the only problem being increased bleeding in these patients who have very large chest-wall muscles and an enlarged circulation.  相似文献

Since 1989, superior pedicle vertical scar mammaplasty as described by Lejour has been used in the authors' department as the only technique for breast reduction. From 1991 through 1994, a series of 170 consecutive patients (330 breasts) underwent an operation. In these patients, minor complications were observed in 30 percent of the patients and major complications in 15 percent. Surgical revision for scar or volume corrections was necessary in 28 percent of the breasts, which seemed unacceptable. Therefore, the original technique was modified by decreasing the skin undermining and avoiding liposuction in the breast. Primary skin excision was performed in the submammary fold at the end of the operation if the skin could not be puckered adequately. This modified technique was used from 1996 through 1999 in 138 consecutive patients (227 breasts). In the second series, minor complications were observed in 15 percent of the patients and major complications in 5 percent. However, the technical modifications did not significantly change the rate of secondary scar and volume corrections, which were still necessary in 22 percent of the breasts. In large breasts, the addition of a horizontal scar at the end of the operation did not change the rate of secondary revision, which however compares favorably with the figures obtained with the inverted T, superior pedicle mammaplasty.  相似文献

From December of 1980 to May of 1998, 390 male-to-female transsexuals underwent vaginoplasty by inversion of the penile skin and a triangular perineoscrotal flap. Although minor modifications were made throughout the years, the basic surgical technique remained the same over this 17.5-year period. In 86 of the 390 patients (22 percent), secondary corrections of the vulva were deemed necessary. A total of 130 corrections were performed in these 86 patients. In the same 17.5-year period, the authors performed 26 secondary corrective procedures in 19 patients in whom the initial vaginoplasty had been done elsewhere. Bilateral Z-plasties were performed 69 times to center the labia in instances when the ventral part of the labia majora remained too far apart. This is not advisable, primarily because it will reduce the vascular supply of the penile skin flap. Introital widening by five-flap advancement was performed in 40 cases in which a dorsal skin fold obstructed the introitis. The use of the triangular perineoscrotal flap favors the vaginal and introital width, but its base should be close to the anal ring to prevent such a skin fold. Secondary construction of the labia minora was performed 27 times, and a skin reduction of the labia majora was performed 20 times. So far, the authors have been unable to develop a satisfactory method for primary construction of the labia minora. Because the appearance of the vulva may charge gradually during the first postoperative year, secondary vulvar corrections should not be performed in that period.  相似文献

Samadi and Barberousse attempt to provide a precise, formal definition of the species category that is faithful to the internodal species concept (Samadi S, Barberousse A. 2006. The tree, the network, and the species. Biological Journal of the Linnean Society 89: 509–521). Here, it is argued that their study is technically flawed. Most of the necessary corrections to their definitions are provided in order to accurately portray the internodal concept. It is then argued that the internodal concept is fundamentally flawed; it does not solve the 'classical problems' that Samadi and Barberousse claim it does. In particular, it does not allow for the possibility of interspecies hybridization. A number of small-sample corrections have been proposed for the conditional maximum-likelihood estimator of the odds ratio for matched pairs with a dichotomous exposure. I here contrast the rationale and performance of several corrections, specifically those that generalize easily to multiple conditional logistic regression. These corrections or Bayesian analyses with informative priors may serve as diagnostics for small-sample problems. Points are illustrated with a small exact performance comparison and with an example from a study of electrical wiring and childhood leukemia. The former comparison suggests that small-sample bias may be more prevalent than commonly realized.  相似文献
