五爪金龙Ipomoeacairica和圆叶牵牛Pharbitispurpurea是华南地区重要的草质藤本杂草。 6 5× 10 - 6以上浓度的 2甲 4氯和 36 0× 10 - 6以上的 2 ,4 -D丁酯完全能使圆叶牵牛种子不能萌发 ,幼苗致死。叶片喷洒 6 50× 10 - 6以上浓度的 2甲 4氯及 36 0 0× 10 - 6以上浓度的 2 ,4 -D丁酯能杀灭五爪金龙和圆叶牵牛成熟植株。分别在茎干基部注射 1ml的 2 ,4 -D丁酯、“恶草灵”和“毒莠定”能较彻底地灭除五爪金龙。  相似文献

选用10种含不同浓度2,4-D、Dicamba及2,4-D与Dicamba组合的诱导培养基和3种含0.01mg.L-1 2,4-D和不同浓度ZT的分化培养基,研究Dicamba、2,4-D、ZT对愈伤组织诱导和分化的影响。结果表明:2,4-D与Dicamba组合的诱导激素在再生分化上显著优于2,4-D和Dicamba。以添加1.5mg.L-1 2,4-D和0.5mg.L-1Dicamba为诱导培养基,分化培养基添加1mg.L-1 ZT和0.01mg.L-1 2,4-D对愈伤组织分化效果为最佳,绿苗分化率最高可达90%。分化培养基中1mg.L-1 ZT有利于根的分化。该研究结果为建立高效、稳定的新疆主栽小麦品种的遗传转化系统奠定了基础。  相似文献

对五爪金龙(Ipomoea cairica (Linn.) Sweet)进行切割试验,对其在不同光环境及不同季节的萌芽率、成活率、主茎长度和生物量等进行研究。结果表明:五爪金龙切条越短其萌芽越迟,其成活率、主茎长度和生物量下降;五爪金龙在光下和林下的成活率无显著差异,但林下生物量较低;切条越短,林下条件越不利于主茎的伸长;冬季五爪金龙的主茎长度和生物量最低;夏季在光下和林下不能再生的最长切条长度均为4 cm;秋季均为1 cm;而冬季光下为1 cm,林下为3 cm;春季光下则为4 cm,林下为5 cm。春季时将五爪金龙切割为5 cm并置于林下是最经济的防除方法;如果不考虑季节性及光条件,应将其切割至1 cm以下的片段,可以有效防止五爪金龙再生。  相似文献

不同除草剂的田间杂草防效及对糜子生长发育的影响   总被引:1,自引:0,他引:1  
筛选糜子适宜除草剂来防范农田药害,是糜子高效安全生产和改善生态环境亟待解决的关键问题。本研究以粳性糜子品种‘榆糜2号'为材料,探究22种除草剂对糜子田间杂草的防除效果及对糜子生长发育的影响。结果表明: 1)喷施土壤封闭型除草剂谷友、丁草胺、莠去津、苄嘧·丙草胺和茎叶型除草剂苯唑·二甲钠、阔世玛、藤净、陶氏·优先、阔菲后,基本无药害作用,糜子幼苗生长正常,其余除草剂均对糜子有不同程度的药害影响;2)参试的22种除草剂在糜子田中对杂草均表现出一定的防除效果,总体而言,土壤封闭型除草剂的杂草防效相对优于茎叶型除草剂,但所有参试除草剂对糜子株高、功能叶片叶绿素含量、单株穗重均造成不同程度的影响;3)与人工除草相比,参试除草剂均导致糜子产量有不同程度的下降;但与不除草对照相比,部分除草剂有明显的增产增效作用。土壤封闭型除草剂中,谷友、丁草胺、莠去津、苄嘧·丙草胺的杂草防效较好,较不除草对照增产60%以上;茎叶型除草剂中,阔世玛、苯唑·二甲钠的杂草防效较好,较不除草对照增产50%以上。因此,在糜子出苗前可用38%莠去津或44%单嘧磺隆进行土壤封闭处理,或在出苗后喷施茎叶型除草剂3.6%二磺·甲碘隆或55%苯唑·二甲钠,农田杂草防效较好,且对糜子生长发育的负面影响较小。  相似文献

以杜衡(Asarum forbesii Maxim.)叶柄为外植体,采用L9(34)正交实验设计分别研究了培养基中外源激素的种类及质量浓度对杜衡叶柄愈伤组织诱导和分化的影响,并据此筛选出适宜的诱导及分化培养基.结果表明:在杜衡叶柄愈伤组织的诱导过程中,培养基中细胞分裂素的种类(1.00 mg·L-16-BA、1.00 mg·L-1KT和1.00 mg·L-1ZT)和NAA质量浓度(0.00、0.10和0.30 mg·L-1)的影响效应均不显著,而2,4-D质量浓度(0.10、0.50和1.00 mg·L-1)则有显著影响(P<0.05);在愈伤组织的分化培养过程中,NAA(0.10、0.30和0.50 mg·L-1)的影响效应大于6-BA(1.00、3.00和5.00 mg·L-1)和IBA(0.01、0.05和0.10 mg·L-1).综合比较结果显示,适宜于杜衡叶柄愈伤组织诱导的培养基为添加1.00 mg·L-16-BA、0.30 mg·L-1NAA和1.00 mg·L-12,4-D的MS培养基(含6.5 g·L-1琼脂和30 g·L-1蔗糖,pH 5.8~pH 6.0),在此培养基上愈伤组织诱导率达到83.33%,且愈伤组织生长速度快、颗粒紧密;适宜于杜衡愈伤组织分化和不定芽增殖的培养基为添加3.00 mg·L-16-BA、0.10 mg·L-1IBA和0.30 mg·L-1NAA的MS培养基(含6.5 g·L-1琼脂和30 g·L-1蔗糖,pH 5.8~pH 6.0),在此培养基上愈伤组织的分化率最高(达到53.33%),增殖系数也最高(3.13).  相似文献

在不同季节采集并测定了入侵植物五爪金龙不同器官中伞形花内酯(Umbelliferone(UMB))和东莨菪亭内酯(Scopoletin(SCO))的含量、同时室内分析了这两种物质的杀螺效果及其对水稻和稗草生长的影响、并研究了这两种物质在土壤中的降解情况。结果表明:五爪金龙植株中SCO含量高于UMB含量,两物质均是夏季含量最高,春季次之,秋冬季低,SCO夏季在茎中含量最大,秋冬季幼茎和幼叶中含量较高;UMB对福寿螺的毒杀力强于SCO,UMB浓度为C2(200μg/mL)时处理24 h对福寿螺的致死率达100%,而SCO浓度为C2时处理24 h对福寿螺的致死率仅为8%;SCO对水稻和稗草苗生长的影响较UMB强,在C0(50μg/mL),C1(100μg/mL)、C2浓度下均促进水稻及稗草苗的生长,而C3(400μg/mL)浓度下则抑制水稻和稗草苗的生长。UMB和SCO混合溶液比单一物质在相同浓度的杀螺效果及对水稻和稗草的生长影响要强;UMB和SCO在土壤中10 d内完全降解。入侵植物五爪金龙中的次生物质UMB和SCO在稻田中的生态效应值得进一步研究。  相似文献

在田间条件下,选用3个小麦材料与不同玉米品种进行杂交,用含不同体积分数二甲基亚砜(DMSO)的100 mg/L 2,4-D溶液和3%DMSO与2,4-D配比处理两种方法诱导小麦×玉米产生单倍体.结果表明,在不同DMSO处理中,2%体积分数的DMSO最佳处理平均得胚率最高为15.8%,3%处理最低(7.1%),二者差异显著;在激素配比试验中,以授粉后48 h采用小花滴注100 mg/L 2,4-D溶液、授粉后72 h再次采用小花滴注并穗下茎节注射3%DMSO的100 mg/L 2,4-D溶液的处理得胚率最高(14.1%),但与3%DMSO的2,4-D 1次处理(13.4%)2、,4-D 1次处理(12.7%)、2,4-D 2次处理(10.6%)和3%DMSO的2,4-D 2次处理(12.6%)之间无显著差异,而这5种处理的得胚率均与对照和未授粉只进行3%DMSO的2,4-D 1次处理存在极显著差异.  相似文献

在深圳市内伶仃岛薇甘菊危害的不同群落生境中,设立9块样地81个小样方,用森草净(即70%嘧碘降水溶性粉剂)杀灭样地中的薇甘菊施量为0.0001~0.02g.m-2,结果表明:各浓度的森草净杀灭效果均较好,杀灭率随着用药量的增加而提高;在坡地和溪谷生境中,森草净用药量分别为0.05~0.1g.m-2、>0.2g.m-2能较彻底地杀灭薇甘菊。应用HPLC法检测样地土壤中嘧磺隆残留量,溪谷土壤中嘧磺隆半衰期C=C0.e-0.083T,T1/2=8.4,施药后37d消解95.9%,坡地高浓度级半衰期C=C0.e-0.046T,T1/2=15.1d,施药后37d消解85.0%,坡地低浓度级半衰期C=C0.e-0.090T,T1/2=7.7d,施药后15d消解74.2%。不同浓度森草净处理样地,施药后7、15、37d均可检测到嘧磺隆,并且含量越来越小,但施药后68d的土样,均未检测到嘧磺隆的存在。  相似文献

干旱胁迫对五爪金龙种子萌发与幼苗生长的影响   总被引:7,自引:3,他引:4  
以粤东地区危害严重的入侵杂草五爪金龙为材料,采用聚乙二醇(PEG-6000)模拟不同程度的干旱胁迫,测定了干旱胁迫下五爪金龙种子萌发率、萌发指数及其幼苗的叶绿素含量、细胞水势、生物量、抗氧化酶活性等指标,以探讨五爪金龙的抗旱机理.结果表明:随着干旱胁迫程度的增加,(1)五爪金龙种子的初始萌发时间延迟,萌发率、萌发指数显著降低,在重度干旱胁迫(30%)时,种子完全不能萌发且伴随腐烂现象;(2)五爪金龙幼苗叶绿素含量、细胞水势、生物量及苗高显著降低,根茎比显著增大;(3)幼苗叶片的超氧化物歧化酶(SOD)活性和丙二醛(MDA)含量持续上升,过氧化氢酶(CAT)活性持续下降,过氧化物酶(POD)活性在轻度胁迫时无明显变化,但中度胁迫时显著上升,重度胁迫时显著下降(P<0.05).研究发现,干旱胁迫对五爪金龙种子萌发和幼苗生长均有显著抑制作用,但同时也能通过自身一系列生物学、生理学指标的改变主动适应干旱逆境.  相似文献

Plants were regenerated from mesophyll protoplasts of Ipomoea cairica L., a wild relative of sweetpotato (Ipomoea batatas (L.) Lam.), and somatic hybrids between I. cairica L. and sweetpotato cv. Xushu 18 were obtained by PEG-mediated method. I. cairica L. protoplasts were isolated from the leaves of in vitro grown plants and cultured in a modified MS medium containing 0.05 mg l−1 2,4-D and 0.5 mg l−1 kinetin. Nine weeks after plating, the obtained small calluses up to about 2 mm in diameter were transferred to solid MS medium supplemented with 0.05 mg l−1 2,4-D and 0.5 mg l−1 kinetin for callus proliferation. Three weeks after transfer, the calluses were transferred to MS medium supplemented with 0–1.0 mg l−1 IAA and 1.0–3.0 mg l−1 BAP and further to hormone-free MS medium for plant regeneration. The frequencies of calluses forming plants ranged from 6.0% to 41.3% based on the different concentrations of IAA and BAP, and 2.0 mg l−1 BAP gave the highest regeneration frequency of protoplast-derived calluses in I. cairica L.. The regenerated plants, when transferred to soil, showed 100% survival. No morphological variations were observed. Mesophyll protoplasts of I. cairica L. were fused with protoplasts isolated from embryogenic suspension cultures of Xushu 18 by PEG-mediated method. The fused products were cultured with the best protoplast culture system of I. cairica L.. Finally, 114 plants were produced from 63 of the 182 calluses derived from the fused protoplasts, and 46 plants of them were confirmed to be somatic hybrids through peroxidase isozyme, RAPD, morphological and cytological analyses.  相似文献

Calli were induced from mature caryopses of timothy grass (Phleum pratense L.) on MS medium (Murashige and Skoog 1962) supplemented with 500 mg·dm−3 casein hydrolysate and 5 mg·dm−3 2,4-D (2,4-dicholorophenoxyacetic acid) or 2 mg·dm−3 dicamba (3,6-dichloro-o-anisic acid). Twelve-week-old calli were passaged on media with reduced levels of auxins (2 mg·dm−3 2,4-D or 1 mg·dm−3 dicamba). Tissues induced on medium with 2,4-D were transferred on medium with 2,4-D and on medium with dicamba; parallely calli initiated on medium with dicamba were passaged on medium with 2,4-D or dicamba. Calli from various media sequences were used to establish cell suspension cultures in media containing 2 mg·dm−3 2,4-D or 1 mg·dm−3 dicamba. An assessment of regeneration ability of calli was made on MS medium containing 0.2 mg·dm−3 kinetin. Callus tissue induced and/or subcultured on any of the media with 2,4-D did not regenerate plants while dicamba added to the media was the effective stimulator of regenerability. In the presence of 2,4-D calli and suspensions produced a jelly-like extracellular matrix. In cell suspension this phenomenon was observed 4–5 days after each passage. The measurements of electric potential of calli, growing on MS medium with kinetin were performed. Non-regenerating callus areas had an electric potential close to 0 mV while parts of tissue with meristematic centres were characterized by lower values of electric potential.  相似文献

为建立紫茉莉(Mirabilis jalapa L.)悬浮细胞培养体系,以紫茉莉无菌苗叶片诱导的愈伤组织为材料,筛选紫茉莉悬浮细胞的适宜培养体系。结果表明,紫茉莉愈伤组织在MS+2,4-D 1 mg L-1+KT 0.5 mg L-1的液体培养基中悬浮继代培养3~4次,能得到稳定的悬浮细胞系。培养基的pH值为5.5~5.9,蔗糖浓度为30 g L-1更适合悬浮细胞的生长。紫茉莉悬浮细胞的生长曲线大致呈S型。最佳继代培养时间是10 d,培养液的体积为40 mL时,接种量为7.5 mL,可以较好地保持悬浮细胞系。1 L培养液中可提取分泌蛋白(0.42±0.15) g。这些有助于对悬浮细胞提取分泌蛋白的研究。  相似文献

Summary This study was conducted to establish and optimize a regeneration system for adapted U.S. rice genotypes including three commercial rice cultivars (LaGrue, Katy, and Alan) and two Arkansas breeding lines. Factors evaluated in the study were genotype, sugar type, and phytohormone concentration. The system consisted of two phases, callus induction and plant regeneration. In the callus induction phase, mature caryopses were cultured on MS medium containing either 1% sucrose combined with 3% sorbitol or 4% sucrose alone, and 0.5 to 4 mg·L−1 (2.26 to 18.10 μM) 2,4-D with or without 0.5mg·L−1) (2.32 μM) kinetin. In the plant regeneration phase, callus was transferred to 2,4-D-free MS medium containing 0 or 2 mg·L−1 (9.29 μM) kinetin combined with 0 or 0.1 mg·L−1 (0.54 μM) NAA. Callus induction commenced within a week, independent of the treatments. Callus growth and plant regeneration, however, were significantly influenced by interactions among experimental factors. Generally, the greatest callus growth and plant regeneration were obtained with 0.5 mg·L−1 (2.26 μM) 2,4-D and decreased with increasing 2,4-D concentrations. Kinetin enhanced callus growth only when combined with 0.5 mg·L−1 (2.26 μM) 2,4-D, and 4% sucrose. Inducing callus on kinetin-containing medium generally enhanced regeneration capacity in the presence of sucrose but not with a sucrose/sorbitol combination. Media containing sucrose alone generally supported more callus proliferation, but the sucrose/sorbitol combination improved regeneration of some cultivars. An efficient and rapid plant regeneration system through somatic embryogenesis was developed using 13-week-old zygotic embryos of oil palm (Elaeis guineensis Jacq.) cv. ‘Tenera’. Zygotic embryos were cultured on MS and N6 media supplemented with 2.0 mg L−1 picloram, 2,4-D and dicamba. The highest embryogenic callus formation (32%) was observed on N6 medium with 2,4-D after 3 month culture on callus induction medium. Somatic embryos were continuously formed from nodular calli on embryo maturation medium [N6 + 0.1 mg L−1 2,4-D, 0.16 g L−1 putrescine, 0.5 g L−1 casein amino acids and 2.0 g L−1 activated charcoal(AC)] for 3–5 months. Histological analysis confirmed that embryo development occurred via somatic embryogenesis. For plant regeneration, modified N6 medium (MN6) with AC (0.5 g L−1) without growth regulators, induced both shoot and root formation simultaneously with the highest regeneration rate of 56%. This combined shoot and root induction protocol shortened the culture time to 9–12 months. Furthermore, after acclimatization, more than 85% of transferred plants from our protocol developed successfully in the soil.  相似文献

Background

Invasive plants are often confronted with heterogeneous environments and various stress factors during their secondary phase of invasion into more stressful habitats. A high tolerance to stress factors may allow exotics to successfully invade stressful environments. Ipomoea cairica, a vigorous invader in South China, has recently been expanding into salt marshes.

Methodology/Principal Findings

To examine why this liana species is able to invade a stressful saline environment, we utilized I. cairica and 3 non-invasive species for a greenhouse experiment. The plants were subjected to three levels of salinity (i.e., watered with 0, 4 and 8 g L−1 NaCl solutions) and simulated herbivory (0, 25 and 50% of the leaf area excised) treatments. The relative growth rate (RGR) of I. cairica was significantly higher than the RGR of non-invasive species under both stress treatments. The growth performance of I. cairica was not significantly affected by either stress factor, while that of the non-invasive species was significantly inhibited. The leaf condensed tannin content was generally lower in I. cairica than in the non-invasive I. triloba and Paederia foetida. Ipomoea cairica exhibited a relatively low resistance to herbivory, however, its tolerance to stress factors was significantly higher than either of the non-invasive species.

Conclusions/Significance

This is the first study examining the expansion of I. cairica to salt marshes in its introduced range. Our results suggest that the high tolerance of I. cairica to key stress factors (e.g., salinity and herbivory) contributes to its invasion into salt marshes. For I. cairica, a trade-off in resource reallocation may allow increased resources to be allocated to tolerance and growth. This may contribute to a secondary invasion into stressful habitats. Finally, we suggest that I. cairica could spread further and successfully occupy salt marshes, and countermeasures based on herbivory could be ineffective for controlling this invasion.  相似文献

为探究华南地区严重入侵植物五爪金龙(Ipomoea cairica)生物入侵的分子机制,对五爪金龙及其近缘种七爪金龙(I. digitata)和裂叶牵牛(I. nil)进行de novo转录组测序和组装,得到56551条unigenes,其中56522条得到注释,7815条GO注释,15615条COG注释,180201条KEGG数据库注释。转录组分析结果表明,五爪金龙氮代谢通路关键酶基因的表达高于对照。次生代谢关键酶(PAL、4CL、CAD、查耳酮合酶、苯基丙乙烯酮异构酶、槲皮黄3-O-甲基转移酶等)基因在五爪金龙与七爪金龙及裂叶牵牛中均得到协同性的差异表达,而这些代谢通路指导的产物合成对五爪金龙的抗逆境能力、生长、化感作用等均起关键作用。关键基因的RT-qPCR验证结果与转录组结果具有一致性。因此,这从分子生物学层面上对解释五爪金龙在华南地区的入侵机制提供了新的证据。  相似文献

Mature embryos of einkorn (Triticum monococcum ssp. monococcum) and bread (Triticum aestivum L.) wheat were used for callus induction on media containing four different doses (0, 1, 2 and 4 mg L?1) of 2,4-D and dicamba supplemented with five different boron concentrations (0, 6.2, 12.4, 24.8, and 37.2 mg L?1). The obtained callus was transferred to culture media with three (0, 0.5, and 2 mg L?1) different BAP doses with five boron concentrations for further regeneration. The maximum callus weight in einkorn wheat was in culture media with 1 mg L?1 dicamba and 6.2 mg L?1 (3.71?±?0.13 g). Bread wheat had the maximum callus weight on culture media with 4 mg L?1 dicamba and 12.4 mg L?1 (3.46?±?0.40 g). The highest plantlet numbers were in only 2 mg L?1 BAP (2.92?±?0.88) for einkorn wheat and 0.5 mg L?1 BAP supplemented with 6.2 mg L?1 boron (3.71?±?1.12) for bread wheat. This indirect regeneration protocol using mature embryos of einkorn and bread wheat under boron stresses expected to be useful for future wheat breeding studies.

采用正交试验设计法,研究了NAA、KT、2,4-D、蔗糖浓度和不同外植体类型等因素对银杏愈伤组织诱导的影响。结果表明:不同外植体类型对银杏愈伤组织诱导率影响最大,KT和NAA其次,2,4-D和蔗糖浓度最小。银杏愈伤组织诱导最佳培养基为MS+NAA 0.5 mg·L-1+KT 0.5 mg·L-1+蔗糖40 g·L-1,最佳外植体为茎段,其愈伤组织诱导率可达100%。  相似文献
