The prevalence and intensity of oocysts of coccidia, particularly Eimeria species, and eggs of gastrointestinal nematodes in kids (2–4 months), young goats (5–12 months) and adult goats (>1 year) were determined in five large herds managed extensively in five villages in the dry zone. Of the representative samples examined, oocysts were found in 88% of kids, 91% of young goats and 83% of adults. Seven species of Eimeria were identified in faecal samples by salt flotation. Eimeria ninakohlyakimovae (31%), E. alijevi (29%) and E. arloingi (21%) were the most common species in all age categories. Other species encountered were E. christenseni (7%), E. jolchijevi (6%), E. hirchi (3%) and E. aspheronica (1%). Gastrointestinal nematode eggs were found in 89% of the kids, 94% of the young goats and 84% of the adult goats. Identification of gastrointestinal nematodes revealed Haemonchus contortus (90%) followed by Oesophogostomum spp. (8.5%) and Trichostrongylus spp. (1%) in all three age categories examined. Both Eimeria and gastrointestinal nematodes were found in 77% of the kids, 86% of the young goats and 71% of adult goats. The mean oocyst counts per gram of faeces in kids, young goats and adult goats were 9728, 1946, and 2667, respectively. The oocyst counts were not different significantly between age groups (P>0.05). The mean egg counts per gram of faeces in kids, young goats and adult goats were 1217, 1641 and 1092, respectively. The egg counts in kids were significantly lower than that in young goats (P<0.05). The intensity of Eimeria and gastrointestinal nematode infections between herds were significantly different (P<0.01).  相似文献

Interspecific competition between red squirrels and grey squirrels was investigated by comparing the population demography, spacing behavior and habitat use of red squirrels in two large conifer plantations in northern England: one site had only red squirrels (the red-only site), in the other both red and grey squirrels occurred (the red–grey site). Despite more abundant food at the red–grey site, red squirrel densities (0.26 ha–1 at the red–grey site, 0.29 ha–1 at the red-only site), adult survival rates and the breeding rates of females were similar at both study sites. Grey squirrels at the red–grey site occurred at higher densities (0.92–1.1 ha–1) than did the reds and tended to have higher breeding rates. In the presence of grey squirrels, the recruitment pattern of red squirrels changed and there was little recruitment of subadults. The juvenile recruitment rate in the red–grey site (13%) was much lower than in the red-only site (50%). Grey squirrels, in contrast, had higher juvenile recruitment rates at the red–grey site (41%). The core areas of the home ranges of red squirrels in the red–grey site were more strongly overlapped by grey squirrels than by conspecifics. Red squirrels did not select the habitat with the best tree seed crop (Scots pine) but preferred dense Sitka spruce plantations; they appeared to avoid the Scots pine area with its high grey squirrel density. Data on foot length and body condition indicated decreased body growth in young red squirrels when grey squirrels were present. Our data suggest that adult red squirrels suffered little from interspecific competition with grey squirrels and that the key factor is decreased juvenile recruitment in red squirrels.  相似文献

The frequent co-occurrence of two or more genotypes of the same parasite species in the same individual hosts has often been predicted to select for higher levels of virulence. Thus, if parasites can adjust their level of host exploitation in response to competition for resources, mixed-clone infections should have more profound impacts on the host. Trematode parasites are known to induce a wide range of modifications in the morphology (size, shell shape or ornamentation) of their snail intermediate host. Still, whether mixed-clone trematode infections have additive effects on the phenotypic alterations of the host remains to be tested. Here, we used the snail Potamopyrgus antipodarum-infected by the trematode Coitocaecum parvum to test for both the general effect of the parasite on host phenotype and possible increased host exploitation in multi-clone infections. Significant differences in size, shell shape and spinosity were found between infected and uninfected snails, and we determined that one quarter of naturally infected snails supported mixed-clone infections of C. parvum. From the parasite perspective, this meant that almost half of the clones identified in this study shared their snail host with at least one other clone. Intra-host competition may be intense, with each clone in a mixed-clone infection experiencing major reductions in volume and number of sporocysts (and consequently multiplication rate and cercarial production) compared with single-clone infections. However, there was no significant difference in the intensity of host phenotype modifications between single and multiple-clone infections. These results demonstrate that competition between parasite genotypes may be strong, and suggest that the frequency of mixed-clone infections in this system may have selected for an increased level of host exploitation in the parasite population, such that a single-clone is associated with a high degree of host phenotypic alteration.  相似文献

Knowledge of genetic relationships among wildlife populations is fundamental to their conservation, particularly where translocations are concerned. This study involved a survey of mitochondrial DNA variation in the Irish red squirrel population. Our main aims were: (1) to determine whether the Irish red squirrel population is distinct from that found in Britain, given known translocations that took place from Britain in the 1800’s; and (2) whether inclusion of Irish data into a reanalysis of European red squirrel data could reveal patterns of postglacial spread in Ireland. We found evidence that the current Irish red squirrel population may be a mixture of native and translocated stock, and relationships between Irish and European haplotypes supported a number of colonisation events of the island. Although only one haplotype was common to both Ireland and Britain, it is probable that the most common haplotypes in Ireland are British introductions that have since become extinct in Britain. There was a significant regional genetic structure in Ireland (P < 0.001), as well as between all Irish and British regions. Although it is likely that the red squirrel will not be fundamental in tracing the colonisation of Ireland by mammals, the data demonstrated that individual regions within Ireland, as well as the Irish population as a whole, are distinct both from the British population and from each other and, therefore, these populations should be treated as separate Management Units (MU) in conservation strategies.  相似文献

Complete development of Cryptosporidium parvum in host cell-free culture   总被引:2,自引:0,他引:2  
The present study describes the complete in vitro development of Cryptosporidium parvum (cattle genotype) in RPMI-1640 maintenance medium devoid of host cells. This represents the first report in which Cryptosporidium is shown to multiply, develop and complete its life cycle without the need for host cells. Furthermore, cultivation of Cryptosporidium in diphasic medium consisting of a coagulated new born calf serum base overlaid with maintenance medium greatly increased the total number of Cryptosporidium stages. Type I and II meronts were detected giving rise to two morphologically different merozoites. Type I meronts, which appear as grape-like clusters as early as 48 h post culture inoculation, release merozoites, which are actively motile, and circular to oval in shape. Type II meronts group in a rosette-like pattern and could not be detected until day 3 of culturing. Most of the merozoites released from type II meronts are generally spindle-shaped with pointed ends, while others are rounded or pleomorphic. In contrast to type I, merozoites from type II meronts are less active and larger in size. Sexual stages (micro and macrogamonts) were observed within 6-7 days of culturing. Microgamonts were darker than macrogamonts, with developing microgametes, which could be seen accumulating at the periphery. Macrogamonts have a characteristic peripheral nucleus and smooth outer surface. Oocysts at different levels of sporulation were seen 8 days post culture inoculation. Cultures were terminated after 4 months when the C. parvum life cycle was still being perpetuated with the presence of large numbers of excysting and intact oocysts. Culture-derived oocysts obtained after 46 days p.i. were infective to 7- to 8-day-old ARC/Swiss mice. The impact of C. parvum developing in cell-free culture is very significant and will facilitate many aspects of Cryptosporidium research.  相似文献

Across the Pacific the invading gecko species Hemidactylus frenatus has competitively displaced the resident gecko species Lepidodactylus lugubris in urban/surburban habitats. Do parasites enhance, inhibit, orhave no effect on this invasion? Parasites can confer an advantage to an invading species when the invader (1) introduces a new parasite to a resident species that has a greater detrimental effect on the resident than the invader, (2) is less susceptible to endemic parasites than the resident, and/or (3) increases the susceptibility of the resident to parasites. Conversely, parasites may protect a resident against invasion when endemic parasites have a greater impact on the invader than the resident. We screened more than one thousand H. frenatus and L. lugubris in areas of sympatry and allopatry from 28 islands and 5 sites on mainland Asia for a broad array of blood parasites, coccidia and helminths in order to evaluate the potential for parasites to affect their interaction. We found that 1) There were no parasites which appear to protect L. lugubris against invasion by H. frenatus. 2) H. frenatus does not introduce the same parasite to L. lugubris in every location where the two come in to contact, but probably has introduced different parasites in different locations. L. lugubris also seems to have introduced at least one parasite to H. frenatus. 3) The prevalence of parasite species shared by the two hosts is generally higher in H. frenatus; however, prevalence is determined by many factors and cannot be directly translated as susceptibility. We discuss the implications of this difference in prevalence for the Red Queen hypothesis. 4) The prevalence of the cestode Cylindrotaenia sp. is significantly higher in L. lugubris that are sympatric with H. frenatus than those which are allopatric.  相似文献

British S. vulgaris are classified as aseparate subspecies, S. v. leucourus, tomainland Europe. While S. vulgaris is notunder threat across most of its Eurasian range,in Britain, Ireland and Italy populations aredeclining, mainly due to the introduction ofthe American grey squirrel (S.carolinensis). In this study, we conducted anextensive survey of mitochondrial DNA variationin British S. vulgaris populations and apreliminary survey of continental Europeanpopulations. Our main aims were to determinethe extent to which any populations of S.vulgaris in Britain are partially or whollythe product of artificial translocation of redsquirrels from continental Europe, and whethercontinental population variation will provideinformation on post-glacial reafforestationpatterns in Europe. We found that the majorityof extant populations of British S.vulgaris are of continental ancestry, manywith a very recent (last 40 years) Scandinavianancestry. The Scandinavian haplotype hasrapidly become the most dominant innortheastern Britain, despite not appearing innorthern English populations until 1966. Thissuggests that these squirrels may have anadaptive advantage in the non-native sprucedominated conifer plantations of northernEngland. Our preliminary examination ofcontinental populations demonstrated that theyare sufficiently differentiated to allow aphylogeographic study of this species.  相似文献

The present study was undertaken to investigate the infection status of Cryptosporidium parvum in the villagers and the reservoir hosts in several rural areas in Korea. A total 5,262 fecal samples were collected from the inhabitants residing at Gangwon-do, Chungcheongbuk-do, Jeollanam-do, and Gyeongsangnam-do between the dates of September, 2001 to June, 2002. In addition, 1,453 fecal samples were collected from livestock reared in Gokseong-gun, Jeollanam-do and Chungju-si, Chungcheongbuk-do. All the fecal smears were prepared by formalin-ether sedimentation, and examined by light microscopy after modified acid-fast staining. The overall positive rate of human cryptosporidiosis was 3.3%. Gokseong-gun, Jeollanam-do showed a 8.2% positive rate and appeared as the highest endemic area among the surveyed areas. Haman-gun, Gyeongsangnam-do showed a 0.4% positive rate and was the lowest endemic area. The positive rate of livestock infection in Gokseong-gun, Jeollanam-do was 94%, which was more than ten times higher than that of Chungju-si, Chungcheongbuk-do (9.3%). From these results, it was revealed that cryptosporidiosis was an endemic disease in some rural areas of Korea, and the livestock could be an important source of human infection.  相似文献

Cryptosporidium parvum is a waterborne protozoan parasite that is found intracellularly in host animals, including humans, and causes severe diarrhea, which can lead to the death of an immunocompromised individual. Previously, we found that this organism is highly radioresistant as it can productively infect mice after exposure to a 10-kGy dose of γ-radiation.To understand how C. parvum avoids radiation damage, we characterized its protein expression patterns 6, 24, and 48 h after a 10-kGy dose of γ-radiation using two-dimensional PAGE. The gels showed 10 silver-stained spots that increased or decreased in size following γ-irradiation. Five proteins contained in these spots were identified using MALDI-TOF MS peptide fingerprinting, and two of these showed an increase in expression after γ-irradiation. These proteins were identified by LC–MS/MS as proteasome subunit alpha type 4 (NTN hydrolase fold) and thioredoxin peroxidase-like protein. The roles of these two upregulated proteins as related to the radioresistance of C. parvum remain to be evaluated.  相似文献

Cryptosporidium parvum is a parasitic protozoan that causes the diarrheal disease cryptosporidiosis, for which no satisfactory chemotherapy is currently available. Although the presence of mitochondria in this parasite has been suggested, its respiratory system is poorly understood due to difficulties in performing biochemical analyses. In order to better understand the respiratory chain of C. parvum, we surveyed its genomic DNA database in GenBank and identified a partial sequence encoding cyanide-insensitive alternative oxidase (AOX). Based on this sequence, we cloned C. parvum AOX (CpAOX) cDNA from the phylum apicomplexa for the first time. The deduced amino acid sequence (335 a.a.) of CpAOX contains diiron coordination motifs (-E-, -EXXH-) that are conserved among AOXs. Phylogenetic analysis suggested that CpAOX is a mitochondrial-type AOX, possibly derived from mitochondrial endosymbiont gene transfer. The recombinant enzyme expressed in Escherichia coli showed quinol oxidase activity. This activity was insensitive to cyanide and highly sensitive to ascofuranone, a specific inhibitor of trypanosome AOX.  相似文献

The intracellular stages of apicomplexan parasites are known to extensively modify their host cells to ensure their own survival. Recently, considerable progress has been made in understanding the molecular details of these parasite-dependent effects for Plasmodium-, Toxoplasma- and Theileria-infected cells. We have begun to understand how Plasmodium liver stage parasites protect their host hepatocytes from apoptosis during parasite development and how they induce an ordered cell death at the end of the liver stage. Toxoplasma parasites are also known to regulate host cell survival pathways and it has been convincingly demonstrated that they block host cell major histocompatibility complex (MHC)-dependent antigen presentation of parasite epitopes to avoid cell-mediated immune responses. Theileria parasites are the masters of host cell modulation because their presence immortalises the infected cell. It is now accepted that multiple pathways are activated to induce Theileria-dependent host cell transformation. Although it is now known that similar host cell pathways are affected by the different parasites, the outcome for the infected cell varies considerably. Improved imaging techniques and new methods to control expression of parasite and host cell proteins will help us to analyse the molecular details of parasite-dependent host cell modifications.  相似文献

Microsporidiosis is an emerging and opportunistic infection associated with wide range of clinical syndromes in humans. Confirmation of the presence of microsporidia in different samples is laborious, costly and often difficult. The present study was designed to evaluate the utility of the Co-agglutination test (Co-A test) for detection of urinary, fecal and circulating microsporidial antigens in experimentally infected mice. One hundred and twenty male Swiss albino mice were divided into non infected control and infected experimental groups which were further subdivided into two equal subgroups; immunosuppressed and immunocompetent. Microsporidial spores were isolated from human stools and identified to be Encephalitozoon intestinalis by the molecular methods. They were used to infect each subgroup of mice, then their urine, stools and sera were collected at the 1st, 3rd, 5th, 7th and 9th days post-infection (PI). Co-A test, using prepared hyperimmune serum, was used to detect antigens in all samples collected. The cross reactivity of microsporidial hyperimmune sera with antigens of Cyclospora cyatenensis and Cryptosporidium parvum was investigated by Co-A test. The results showed that Co-A test was effective in detecting microsporidial antigen in stool of immunosuppressed infected mice from the 1st day PI, and in urine and serum from the 3rd day PI till the end of the study. In the immunocompetent subgroup, Co-A test detected microsporidial antigens in stool, serum and urine of mice from the 1st day, 3rd day and the 5th day PI, respectively till the end of the study, without cross reactivity with C. cyatenensis or C. parvum in both subgroups. Co-A test proved to be simple and suitable tool for detecting microsporidial antigen in different specimens and did not need sophisticated equipment. It is very practical under field or rural conditions and in poorly equipped clinical laboratories.  相似文献

Summary. Symbiotic ant species vary in their effects on the same host plant, and some ant species are exploiters. These can destabilise the interaction. The behavioural repertoire of ant associates should be important in determining their effects on the host. We examined several traits of foraging behaviour of two ant species associated with the African understorey tree Leonardoxa africana africana – the protective mutualist Petalomyrmex phylax and the parasite Cataulacus mckeyi – to examine the mechanistic basis of the difference in their effects on the plant. These two ant species displayed significant differences not only in activity rhythms, P. phylax patrolling leaves day and night while C. mckeyi was strictly diurnal, but also in their predatory behaviour (P. phylax killed more herbivores) and recruitment dynamics (C. mckeyi recruited at a lower level). For all traits examined, C. mckeyi expressed behaviours reflecting an opportunistic exploitation of the nectar produced by the host plant, whereas those displayed by P. phylax contribute to conferring effective protection of the host plant. Our results suggest that prey capture may be important as a source of food for Petalomyrmex, in addition to its function in increasing plant-provided resources through protection.Received 27 August 2004; revised 11 November 2004; accepted 16 November 2004.  相似文献

We compared phloem characteristics of individual Pinus ponderosa attacked by the dwarf-mistletoe Arceuthobium vaginatum (Viscaceae) or by the beetle Dendroctonus ponderosae (Scolytidae) or by neither species. We quantified total nonstructural carbohydrates and a broad range of chemical elements for these three categories of trees. There were significant differences between trees parasitized by Arceuthobium, trees parasitized by Dendroctonus, and non-infected trees. Discriminant function analysis of trees attacked by either Arceuthobium or Dendroctonus correctly predicted group membership for 59 of 60 trees tested. Some of the differences detected may be induced, but many probably are not. Given that the accumulation of certain elements and compounds is under genetic control, and that both parasites often cause severe reductions in fitness, including death of their host, our results suggest that the two parasites may generate diversifying selection in ponderosa pine populations.  相似文献

Cone-cores discarded by Eurasian red squirrels were used to study the habitat selection of Korean pine-seeds hoarding, in forest patch Nos. 16 and 19 in Liangshui Nature Reserve, China. Ten transects with a total length of 15 km were uniformly set, and data from 343 valid samples were collected in a 369 hm2 area. One hundred and eighty four were hoarding samples which were determined according to the cluster analysis based on the number of the cone-cores, while the other 159 were control samples. The principal component analysis, using 11 habitat factors, suggested that the distance from Korean pine forest, forest type, number of Korean pine seedlings, density and type of bush significantly influenced the habitat selection of hoarding by Eurasian red squirrels. The results of Bailey’s method indicated that the squirrels showed (1) preference for natural coniferous forest, natural fir and spruce forest and planted spruce forest; (2) avoidance of planted Korean pine forest and planted larch forest; and (3) random use of natural Korean pine forest. Moreover the distance from the Korean pines in the range of 150–600 m showed no effect on the habitat selection of hoarding by the Eurasian red squirrels. More than 50% of the cone-cores were discarded in either fringe or gap of the Korean pine forest with more cone-cores found at <300 m than at 300 m away (One-Way ANOVA; df = 3, 183, F = 5.76, p = 0.0009). This demonstrated that the Eurasian red squirrels could take the cone-cores out of the Korean pine forest. The density of bushes in samples of hoarding area was significantly lower than that in control samples (Kruskal–Wallis test; df = 1, χ2 = 83.99, p < 0.0001). The number of the Korean pine seedlings in samples of hoarding area was significantly higher than that in the control samples (Kruskal–Wallis test; df = 1, χ2 = 104.13, p < 0.0001). This illustrated that the hoarding habitat favoured the germination of the Korean pine seedlings. In conclusion the behavior of hoarding Korean pine seeds by the Eurasian red squirrels can promote the regeneration and dispersal of the Korean pines.  相似文献
