The reaction of 6′-chloro-6′-deoxyraffinose deca-acetate with a variety of nucleophilic anions (Br-,I-,N-3) gave the corresponding 6′-substituted raffinoses. The 6′-azide was further converted into 6′-amino-6′-deoxyraffinose, isolated as its N-acetyl derivative, and silver fluoride-induced elimination of hydrogen iodide from the 6′-iodide gave the 6′-deoxy-5′-ene. Treatment of 6′-chloro-6′-deoxyraffinose and 6′,6″-dichloro-6′,6″-dideoxyraffinose with base afforded, respectively, 3′,6′-anhydro-and 3′,6′;3″,6″-dianhydro-raffinose in high yields. In addition, the dichloride was converted into 6′,6″-diazido-6′,6″-dideoxyraffinose.  相似文献

5-Acetyl-2-aryl-6-methyl-4-(2,3,4,6-tetra- O -acetyl-β-D-glucopyranosylmercapto)pyramidines 3a–c were obtained by the reaction of 5-acetyl-2-aryl-6-methyl-pyrimidine thiol 1a–c with 2,3,4,6-tetra- O -acetyl-α-D-glucopyranosyl bromide (2) in aq. KOH/acetone. The reaction of 1a–c with peracetylated galactose 5 and peracetylated ribose 8 under MW irradiation gave 5-acetyl-2-aryl-6-methyl-4-(2,3,4,6-tetra- O -acetyl-β-D-galactopyranosylmercapto)pyrimidine 6a–c and 5-acetyl-2-aryl-6-methyl-4-(2,3,5-tri- O -acetyl-β-D-ribofuranosylmercapto)pyrimidines 9a–c. The deprotection of 3a–c, 6a–c, and 9a–c in the presence of methanol and TEA/H2O yielded the deprotected products 4a–c, 7a–c, and 10a–c. The structures of the compounds were confirmed by using IR, 1H, 13C spectra and microanalysis. Selected members of these compounds were screened for antimicrobial activity.  相似文献

Phenyl, p-tolyl, and p-tert-butylphenyl β-1-thio-N-acetylglucosaminides were synthesized by the treatment of thiophenols with peracetate of α-D-glucosaminyl chloride in the presence of triethylamine or under the conditions of phase-transfer catalysis with quaternary ammonium salts. The compounds synthesized were used for obtaining of glycosides of 4,6-O-isopropylidene-N-acetylmuramic acid, which were coupled with L-Ala-D-Glu(NH2)-OBzl and then deprotected to obtain the target aryl β-thioglycosides of N-acetylmuramyl-L-analyl-D-isoglutamine (MDP). The aryl β-thioglycosides of MDP were found to stimulate an antibacterial resistance toward Staphylococcus aureus in mice. The reliable induction of the spontaneous activity of natural killers in the population of blood mononuclear cells was observed only for phenyl β-thio-MDP at a dose of 200 μg/ml. Arabinosylation of some 4-amino- and 4-arylideneamino-5-(pyridin-3-yl)-2,4-dihydro-[1,2,4]-triazole-3-thiones with 2,3,4-tri-O-acetyl-β-L-arabinopyranosyl bromide led to an efficient synthetic approach to the corresponding N-and S-α-L-arabinopyranosides. Structure assignment of these two regiosiomers was based on chemical and spectroscopic evidences. Antimicrobial activities of two selected regioisomeric N-and S-α-L-arabinopyranosides were compared. The N-α-L-arabinopyranoside showed higher inhibitory effect than its regioisomeric S-α-L-arabinopyranoside against Aspergillus fumigatus, Penicillium italicum, Staphylococcus aureus, and Pseudomonas aeruginosa.  相似文献

Pharmacokinetic studies of optically pure compounds after single enantiomer administration are becoming increasingly important. The process of racemization in vivo can diminish all expected advantages of single enantiomer treatment. Amlodipine, one of the calcium channel blockers, currently used in therapy as a racemate, is one of such drugs under study. In order to administer single enantiomers of amlodipine to healthy volunteers both were chromatographically purified and characterised. The two optical isomers of amlodipine, active S-(−)- and non-active R-(+)-amlodipine, were purified using chromatographic procedure adopted from the analytical separation. Enantiomers were successfully converted to benzenesulphonic salt without any racemization. All semi-preparative purifications were monitored with complementary analytical methods, HPLC and CE, along with the determination of optical activity so that the final product was sufficiently defined for further in vivo studies. The analytical method developed for the determination of plasma concentrations of each enantiomer of amlodipine in these studies is also briefly described.  相似文献

Methyl 6′, 6′-didemethyl abscisate (5) was synthesized and assayed to elucidate the physiological activity of methyl substituents on the cyclohexene ring of abscisic acid (ABA). During this study two new chiral stereoisomeric analogs 6 and 7 were synthesized from l-and d-carvone. The rice seedling assay and germination assay of garden radish showed that 6′-methyl groups of ABA were not important in biological activity and that 5′-isopropenyl analogs 6 and 7 were inactive.  相似文献

The present study, utilizing thioglycolamido as the reactive group, describes the synthesis and pharmacology of a new opioid antagonist affinity ligand, 6-thioglycolamido-6-desoxynaltrexone (TAN) and compares TAN with a related known compound, 6-bromoacetamido-6-desoxynaltrexone (BAN). Both compounds were tested for their reversible and irreversible inhibition of [3H]naloxone binding to calf brain membranes. Reversible binding of BAN and TAN had Ki values of 1×10–9 and 1×10–10 M, respectively as determined by log probit plots. Irreversible binding was determined after extensive washing to remove all non-covalently bound ligand. At a concentration of 5×10–8 and 1×10–8 M for BAN and TAN irreversible binding was inhibited 50% of the maximum value. A study of the time course of irreversible inhibition of [3H]naloxone binding revealed that maximal inhibition occurred within 5 min with a concentration of 1×10–7 M of either agent. TAN but not BAN when administered systematically to mice produced an antinociceptive effect as measured by the writhing test. When administered intracerebraventricularly BAN did not block morphine-induced analgesia for more than 2 hr; whereas, with a single ED50 dose of 20 nmoles of TAN i.c.v. morphine-induced analgesia was almost completely blocked for a period of over 24 hr, as determined by the tail flick test. Although the SH group of TAN were required for the covalent interaction with opioid receptors, the site of TAN's interaction appears to involve other than protein SH groups.  相似文献

2,3,4,6,1′,3′,4′-Hepta-O-benzylsucrose, obtained by acid-catalysed hydrolysis of the 6′-O-trityl derivative, was oxidised with the Pfitzner-Moffatt reagent and the product was alkylated with methylmagnesium iodide. Removal of the protecting groups then gave a mixture of diastereomers, namely 7-deoxy-β-d-altro and -α-l-galacto-hept-2-ulofuranosyl α-d-glucopyranoside. Application of this reaction sequence to 2,3,4,1′,3′,4′,6′-hepta-O-benzylsucrose afforded β-d-fructo-furanosyl 7-deoxy-dl-glycero-α-d-gluco-heptopyranoside.  相似文献

A vesicle is a microscopic particle composed of a lipid bilayer membrane that separates the inner aqueous compartment from the outer aqueous environment. Palmitoleate–palmitoleic acid vesicles were prepared and their physico-chemical properties were investigated. Moreover, mixed vesicles composed of palmitoleic acid and PEGylated lipid and/or a mixture of phospholipids were also prepared. The stabilizing effects of these double-chain lipids on the formation of palmitoleate–palmitoleic acid vesicles were studied. Stability of the vesicle suspension was examined using particle size and zeta potential at 30?°C. The magnitude of the zeta potential was relatively lower in the vesicle suspension with the presence of phospholipid. Although some of the mixed vesicles that were formed were not very stable, they displayed potential for encapsulating the active ingredient calcein and the encapsulation efficiencies of calcein were encouraging. The palmitoleate–palmitoleic acid–DPPE-PEG2000 vesicle showed the most promising stability and encapsulation efficiency.  相似文献

S-腺苷甲硫氨酸(SAM)是甲硫氨酸和三磷酸腺苷相结合的代谢物,广泛存在于动植物和微生物体内,参与40多种生化反应,主要作为三种代谢途径(转甲基、转硫基、转氨丙基)的前体,临床上被广泛用于治疗肝病、抑郁症、关节炎等。SAM的制备方法主要有化学合成法、酶促合成法、发酵法三种。化学合成的SAM是消旋体,需进行光学拆分,且存在产率低、原料L-高半胱氨酸价格昂贵和环境污染等问题。酶促合成法合成的SAM纯度高,但原料ATP成本太高。发酵法已成为目前生产SAM最常用的方法,欧洲利用发酵法生产SAM已实现了产业化,但国内的起步较晚,目前还处于实验室研究阶段。因此,应加强发酵法生产SAM的产业化关键技术研究。  相似文献

A region-specific regulation of inflammation on the expression hepcidin in the brain has been demonstrated, however, it remains unknown whether there is also a cell-specific regulation of inflammation on hepcidin in the brain. Here, we investigated the effects of lipopolysaccharides (LPS) on the expression of hepcidin mRNA and also the expression of IL-6 mRNA, the phosphorylation of STAT3 and the expression of ferroportin 1 (Fpn1) and ferritin light chain (Ft-L) proteins in neurons and astrocytes obtained from wild type (IL-6+/+) and IL-6 knockout (IL-6?/?) mice. We demonstrated that the responses of the expression of hepcidin and IL-6 mRNAs, the phosphorylation of STAT3, and the expression of Fpn1 protein to LPS in IL-6+/+ astrocytes and also the responses of the expression of hepcidin mRNA, the phosphorylation of STAT3 and the expression of Fpn1 protein to IL-6 in IL-6?/? astrocytes were much stronger than those in IL-6+/+ and IL-6?/? neurons. A significant increase in Ft-L was found in LPS-treated IL-6+/+ and IL-6-treated IL-6?/? astrocytes, but not in LPS-treated IL-6+/+ and IL-6-treated IL-6?/? neurons. Our findings provide in vitro evidence for the existence of a cell-specific regulation of LPS on the expression of hepcidin and also Ft-L in the brain.  相似文献

Fluorescent in situ hybridization (FISH) was used to investigate the chromosomal location of 18S-5.8S-26S rDNA loci in Thinopyrum intermedium (Host) Barkworth et Dewey (2n=6x=42). In all accessions and individuals studied, 3 or 4 pairs of major loci were detected. Subsequent genomic in situhybddization (GISH) analyses revealed that one pair was located on the ends of the short arms of one pair of homologous chromosomes of the St genome, while the other 2 or 3 pairs of major loci were located in the E genomes (including the E^o and E^b). It is suggested that 2 to 3 pairs of major loci were probably lost during the evolution of this hexaploid species. The variation in rDNA positions and copy numbers between the diploid donors and Th. interrnedium, as well as the diversity among the accessions of Th. intermedium confirmed that the rDNA gene family conveyed the characters of DNA mobile elements. The internal transcribed spacer (ITS) regions of the rDNA in Th. intermedium were also investigated. Sequence data of seven positive clones from one individual suggested high degree of individual heterogeneity exists among ITS repeats. Phylogenetic analyses showed that there were two distinct types of ITS sequences in Th. intermedium, one with homology to that of Pseudoroegneria species (St genome) and the other to that of the E genome diploid species. This showed that the ITS paralogues in Th. intermedium have not been uniformly homogenized by concerted evolution. The limitation of using the chromosomal location of rDNA loci for phylogenetic analysis is discussed.  相似文献

A high-performance capillary electrophoresis (HPCE) assay method for the quantitation of S-(+)- and R-(−)-ondansetron in human serum was developed. Resolution was achieved using 15 mM heptakis-(2, 6-di-O-methyl)-β-cyclodextrin (DM-β-CD) in 100 mM phosphate buffer (pH 2.5). A 72-cm untreated fused-silica capillary, at a constant voltage of 20 kv, was used for the analysis. A 0.03-mM cationic detergent was used as a buffer additive to decrease the adsorption of endogenous substances onto the silica wall. The analytes of interest were isolated from endogenous substances using a solid-phase extraction procedure. The cyanopropyl cartridge gave good recoveries in excess of 85% for both S-(+)- and R-(−)-ondansetron, without any interferences. To decrease the limits of detection of the analytes, an on-capillary sample concentration technique was employed. The detection limit was 10 ng/ml using 2 ml of serum and the limit of quantitation was 15 ng/ml. The calibration curve was linear over a range of 15–250 ng/ml, with procainamide as the internal standard, and the coefficients of determination obtained were greater than 0.999 (n=3). Precision and accuracy of the method were 2.76–5.80 and 2.10–5.00%, respectively, for S-(+)-ondansetron, and 3.10–6.57 and 2.50–4.35%, respectively, for R-(−)-ondansetron. The HPCE method is a useful alternative to existing chiral high-performance liquid chromatographic methods.  相似文献

【目的】克隆一种梨小食心虫Grapholita molesta触角中谷胱甘肽S-转移酶(glutathione Stransferase,GST)基因并分析其结构特征,明确其表达特点及该基因编码蛋白的酶学特征,以期为该基因的功能研究提供理论依据。【方法】根据梨小食心虫雌虫触角转录组数据,利用RT-PCR克隆梨小食心虫GST基因;通过实时荧光定量PCR测定该基因在成虫触角、头(去触角)、胸、腹、足和翅中的转录水平;利用原核表达系统和Ni离子亲和层析技术表达和纯化梨小食心虫GST重组蛋白,并对重组蛋白的稳定性和酶促动力学参数进行分析。【结果】获得了梨小食心虫谷胱甘肽S-转移酶基因GmolGST6(GenBank登录号:MF503496)的完整编码序列,开放阅读框为645 bp,编码214个氨基酸,分子量为24.21 kD,理论等电点为5.10;进化树分析和三维结构模拟表明GmolGST6属于Delta亚家族。qPCR测定结果表明,GmolGST6基因在雌雄成虫触角中高丰度表达,且在雄虫触角中的表达量显著高于雌虫。重组蛋白GmolGST6具有催化通用底物CDNB的活性,并且在pH7.5,40℃时具有最高的酶活力。重组蛋白GmolGST6的米氏常数K_m为0.21±0.06 mmol/L,最大反应速度V_(max)为14.02±1.40μmol/min·mg。【结论】根据GmolGST6的表达特点及其重组酶对模式底物CDNB的催化活性,推测在触角中高丰度表达的GmolGST6具有参与降解外源物质、保护嗅觉系统免受外源性物质毒害的功能。  相似文献
