Three cases of spontaneous cataracts were investigated in a colony of African green monkeys (Chlorocebus aethiops). Clinical, pathological and microbiological investigations were conducted in two cases of juvenile cataracts and in one case of a mature cataract. These investigations revealed no indication of an infection as the cause of cataract development. A genetic correlation existed between the affected individuals. Clinical chemistry gave a hint that calcium might be a "key factor" in the development of these cataracts: in both cases of the juvenile cataracts, the calcium content in the serum and in the aqueous humor was clearly decreased in the affected babies. The calcium uptake in the affected baby itself was not altered but the calcium content in the mother's milk was low. 相似文献

The increasingly limited availability and high cost of the hitherto most commonly used monkey species in dengue vaccine research has augmented the importance of identifying alternative suitable models for these studies. In this study we examined the capacity of green monkeys ( Chlorocebus aethiops sabaeus ) to develop dengue viremia, and thus provide a potential model for dengue vaccine testing. Monkeys were inoculated with two different doses of dengue virus type 2. All animals in both groups became viremic after inoculation of the virus. In the lower dose group, mean viremia duration of 5.66 days was detected, whereas in the group that received the 106 PFU dose, viremia had a mean duration of only 1.66 days. Antibody titers were similar to those obtained in previous experiments with rhesus and cynomolgus macaques. We conclude that green monkeys develop viremia and antibody responses and therefore provide a potential model for the preclinical evaluation of novel candidates for dengue vaccines. 相似文献

This study was designed to evaluate the timecourse of ovarian and pituitary endocrine events throughout the menstrual cycle in the vervet monkey, and whether circulating luteinizing hormone (LH) or the uterus regulates the functional lifespan of the vervet corpus luteum. Daily saphenous blood samples were collected from adult females (1) during spontaneous menstrual cycles (n = 7), and (2) during cycles in which a gonadotropin-releasing hormone antagonist (acyline) was administered for 3 days at midluteal phase (n = 3), and (3) for 30 days following recovery from hysterectomy (n = 4). Estradiol (E) and progesterone (P) levels were assayed using electrochemoluminescent assays. Gonadotropin levels were measured by radioimmunoassay using reagents developed for the assay of follicle-stimulating hormone and LH in macaques. Spontaneous cycles exhibited a midcycle E rise (476+/-49 pg/ml), engendering an LH surge, 12+/-1 days after onset of menses, followed by a luteal phase with peak P levels of 4.7+/-0.9 ng/ml. Histologic evaluation of the ovaries at late follicular phase or early luteal phase revealed the presence of a single, large Graafian follicle or developing corpus luteum, respectively. Acyline treatment caused a significant (P<0.05) decline in P levels (2.9+/-0.5 vs 0.5+/-0.3 ng/ml, 0 vs 48 h post-treatment) and premature menstruation compared with untreated controls (P<0.05). Hysterectomy had no apparent effect on the monthly pattern or levels of circulating E or P. Thus, the characteristics and regulation of the ovarian cycle in vervets appear similar to those in women and macaques, with cyclicity dependent on pituitary gonadotropin hormones and independent of a uterine luteolytic factor. 相似文献

Studies of blackfly vectors of Onchocerca dewittei japonica Uni, Bain & Takaoka (Spirurida: Onchocercidae), a parasite of wild boar implicated in the aetiology of zoonotic onchocerciasis in Japan, and six other zoonotic Onchocerca species of this country are reviewed. Molecular identification of infective larvae found in wild‐caught female blackflies showed that Simulium bidentatum (Shiraki) (Diptera: Simuliidae) is a natural vector of O. dewittei japonica, and also Onchocerca sp. sensu Fukuda et al., another parasite of wild boar. Inoculation experiments demonstrated that Simulium arakawae Matsumura and four other Simulium species are putative vectors. Similarly, S. arakawae, S. bidentatum and Simulium oitanum (Shiraki) are putative vectors of Onchocerca eberhardi Uni & Bain and Onchocerca skrjabini Rukhlyadev, parasites of sika deer. Morphometric studies of infective larvae indicated that Onchocerca lienalis Stiles, a bovine species, is transmitted by S. arakawae, Simulium daisense (Takahasi) and Simulium kyushuense Takaoka, and that Onchocerca sp. sensu Takaoka & Bain, another bovine species, is transmitted by S. arakawae, S. bidentatum, S. daisense and S. oitanum. Prosimulium sp. (Diptera: Simuliidae) and Simulium japonicum Matsumura are suspected vectors of Onchocerca suzukii Yagi, Bain & Shoho and O. skrjabini [Twinnia japonensis Rubtsov (Diptera: Simuliidae) may also transmit the latter], parasites of Japanese serow, following detection of the parasites' DNA genes in wild‐caught blackflies. 相似文献

In poplar (Populus x euramericana cv. Robusta) infected with poplar mosaic virus, rod-shaped, virus-like particles (c. 13 nm in diameter) occurred singly or in loose bundles in the cytoplasm of cells in palisade and mesophyll tissue and in vascular parenchyma. The cell walls had local overgrowths consisting of membranous elements (vesicles and/or tubules) in a loosely fibrillar matrix. Accumulations of endoplasmic reticulum with enlarged cisternae occurred near cell nuclei. We are indebted to Mr A. G. Infection of bean pods with Colletotrichum lindemuthianum leads to the appearance in diffusates of a range of fluorescent and phenolic compounds and of at least two inhibitory compounds. These compounds were found to be absent or in reduced concentration in control diffusates, although the inhibitors were frequently observed to appear at low concentrations without infection. Although sugars and amino acids were found to be released into diffusates, variation in the concentration of these stimulants was found to have little effect on spore germination. Evidence from solvent partition, spectrophotometry and chromatography suggests that the two inhibitors can be tentatively identified with inhibitors previously described from French bean, and both appear to be phenolic substances. It is suggested that inhibitor production may be regarded as part of a general change in aromatic biosynthesis following infection. 相似文献

Forty-five, weanling goats of mixed-sex, were randomly allotted to five treatment groups: no dust (n = 16), raw organic dust exposure for 15 min (n = 6), raw organic dust exposure for 1 h (n = 7), raw organic dust exposure for 3 h (n = 7), and raw organic dust exposure for 4 h (n = 9). Inhalation exposures were performed in a closed tent for the allotted times. The amount of endotoxin calculated to be in the dust was shown to be 26.9 μg/g. The amounts of dust introduced into the tent for each time period were 4.58 g/m3 of air for 15 min; 7.46 g/m3 of air for 1 h; 14.82 g/m3 of air for 3 h; and 40.60 g/m3 of air for 4 h. There was a significant increase in the white blood cell count in the animals dusted for 4 h, at 8 and 12 h following exposure. There was a significant decrease in the peripheral lymphocyte cell count following the 15 min exposure at 12 h post exposure, and there was a significant increase in the peripheral lymphocyte cell count following the 4 h exposure, 24 h after the exposure. There was a significant increase in the neutrophil cell count 8 and 12 h following the 4 h exposure, while there was a significant decrease in the neutrophil cell count 48 h following the 4 h exposure. There was a significant increase in the rectal temperatures of all goats receiving the 4 h dust exposure at all time periods (0 time, 4, 12, 24, and 48 h). There was a significant increase in the rectal temperatures of goats following the 1 h exposure, 8 h later. These results indicate that the larger the dose of inhaled endotoxin, the higher the resultant fever and leukocytosis. 相似文献

The density of eosinophils in the cerebrospinal fluid (CSF) and blood of male ddY mice infected with Angiostrongylus cantonensis was examined on days 14, 20 and 27 post-infection (p.i.) with discontinuous Percoll gradient centrifugation. Normal blood eosinophils had a density of between 1.070 and 1.080 g ml-1. No significant changes in density in blood eosinophils were noted during the course of the observations. CSF eosinophils began to become hypodense (defined as density less than 1.070 g ml-1) on day 20 p.i., and 88% of eosinophils were hypodense on day 27. Our results suggest therefore that eosinophils probably become hypodense in the CSF and brain tissues, but not in the blood. 相似文献

Three bulls selected for high faecal worm egg counts and three bulls selected for low faecal worm egg counts were mated to Africander-Hereford cross cows. Faecal worm egg counts were taken on four occasions from the 132 offspring. Also, each animal was typed for 32 class I antigens of the bovine major histocompatibility system (BoLA). Least squares analysis of variance showed that line, sex and some of the antigens were associated with differences in worm egg output in the faeces. After adjusting for the effects of line and sex, cattle with antigen W9 had about twice as many worm eggs in their faeces as cattle without W9; cattle with antigen CA45 had about half the concentration of faecal worm eggs as cattle without CA45. However, the antigen associations were of borderline significance at the 5% level and more work in additional populations is necessary to confirm these associations. 相似文献

AIMS: The identification of the culturable and nonculturable bacterial population in ground water of a municipal water supply in Mainz (Germany) during the year 2002. METHODS AND RESULTS: Total counts varied between 3.5 x 103 and 2.2 x 104 cells ml-1, viable counts were approximately between 8.1 x 102 and 3.3 x 103 cells ml-1. After cultivation on different nutrient media (R2A, DEV, PCA, Endo, Standard) <1% appeared to be culturable on the media used. After denaturating gradient gel electrophoreses, up to 24 different bacterial species were detected in the ground water. With the aid of 16S rDNA isolation, amplification and sequencing, the isolated organisms and clones could be identified. CONCLUSIONS: The isolated and cultured organisms mainly belonged to the Proteobacteria (alpha, beta and gamma), Flavobacteria or Actinobacteria. However, most of the noncultured micro-organisms were beta-Proteobacteria. SIGNIFICANCE AND IMPACT OF THE STUDY: This is the first study in which the identification of all culturable and nonculturable bacteria in a ground water has been attempted. 相似文献

Eosinophils in the cerebrospinal fluid of mice infected with Angiostrongylus cantonensis are resistant to apoptosis. 总被引:1,自引:0,他引:1
Interleukin-5 (IL-5) transgenic mice were used to assess the immunological features of CSF eosinophils from mice infected with Angiostrongylus cantonensis. CSF eosinophils were hypodense by day 14 post infection (p.i.). CSF eosinophils survived longer in vitro than peritoneal eosinophils collected from cadmium sulphate (CdSO4) -treated normal IL-5 transgenic mice. Apoptosis was measured by Annexin V binding and the presence of a distinct laddering pattern of DNA fragmentation on agarose electrophoresis. Regardless of the presence or absence of Actinomycin D, CSF eosinophils collected from IL-5 transgenic mice from days 15–36 p.i. exhibited less apoptosis than peritoneal eosinophils collected from uninfected IL-5 transgenic mice. CSF eosinophils collected from A. cantonensis infected C57BL/6 mice at days 15–34 p.i. showed elongation of survival time and less apoptosis during in vitro cultivation. Reduced apoptosis was noted only in CSF eosinophils, but not in peritoneal eosinophils recovered from the same infected IL-5 transgenic mice. CPP32/Caspase 3 activity of cultured peritoneal eosinophils from both infected and uninfected IL-5 transgenic mice was higher than that of cultured CSF eosinophils. Stimulation with A23187 readily induced apoptosis of peritoneal eosinophils, but not CSF eosinophils or peritoneal eosinophils cultured with mouse recombinant IL-5. The latter cells were morphologically identical to hypodense eosinophils. RT-PCR analysis indicated that bcl-2 and bcl-xL mRNA expression was higher in CSF eosinophils compared with peritoneal eosinophils and this expression in the latter cells was upregulated after culture with mouse recombinant IL-5. These results suggest that CSF eosinophils, shifting to hypodense status through an accumulation from peripheral blood, are resistant to apoptosis. These changes may explain the long-lasting, helminthotoxic and neurotoxic actions of CSF eosinophils in A. cantonensis infection. 相似文献

目的:了解小鼠肝炎病毒( MHV)污染的设施内,ICR小鼠自然感染后MHV抗原抗体存在情况。方法选择50只ICR小鼠,通过更换“脏垫料”的方式进行饲养,分别在实验第2,4,8,14,21,28,35,42,56和84天各剖杀动物5只,采集血清、盲肠内容物、粪便、肝脏以及肺脏检测抗原抗体分布情况。结果实验开始第2天,肺脏中检出小鼠肝炎病毒,检出率为20%(1/5),第4天开始,肝脏、肺部、盲肠以及粪便中均可检出小鼠肝炎病毒;84天后,肺脏、盲肠、粪便和肝脏阳性检出率降为0。实验第8天,血清中抗体检出阳性,阳性率为100%(5/5),直至第84天实验结束,抗体阳性率仍维持在100%(5/5)。结论血清学方法可作为日常监督主要诊断方法,而抗原检测方法只能应用于早期诊断。 相似文献

This study investigated the molecular prevalence of oral trichomonads in household dogs. Of the 144 dogs, 21 (14.6%, 21/144) tested positive for oral trichomonads. The prevalence was significantly higher in dogs with severe gingivitis (gingival index 3: 30.0%, 8/26) than that in normal dogs (gingival index 0: 2.7%, 1/37). Therefore, an interaction between oral trichomonads and the development of periodontal disease is suggested. Of the 21 positive samples, 16 isolates were T. brixi, four isolates were T. tenax, and one was Tetratrichomonas sp. Considering T. tenax is recognized as a zoonotic agent, transmission between dogs and humans cannot be neglected. 相似文献
