The mitochondrial DNA (mtDNA) of individuals from 79 colonies of Apis mellifera from five Canary Islands was studied using the Dra I test based on the restriction of PCR products of the tRNAleu–COII intergenic region. Five haplotypes of the African (A) lineage and one of the west European (C) lineage were found. The haplotypes A14 and A15 are described for the first time. These haplotypes have a new P sequence named P1 . The wide distribution and high frequency of haplotype A15 suggest that it is characteristic of the Canarian Archipelago. Sources of haplotype variability of honeybee mtDNA in the Canary Islands (waves of colonization from Africa, queen importations, habitat diversification) are discussed. 相似文献

The hypopharyngeal gland (HG) of the honeybee (Apis mellifera L.) produces royal jelly (RJ) that is essential to feed and raise broods and queens. A strain of bees (high royal jelly producing bee, RJb) has been selected for its high RJ production, but the mechanisms of its higher yield are not understood. In this study, we compared HG acini size, RJ production, and protein differential expressions between the RJb and nonselected honeybee (Italian bee, ITb) using proteomics in combination with an electron microscopy, Western blot, and quantitative real-time PCR (qRT-PCR). Generally, the HG of both bees showed age-dependent changes in acini sizes and protein expression as worker behaviors changed from brood nursing to nectar ripening, foraging, and storage activities. The electron microscopic analysis revealed that the HG acini diameter of the RJb strain was large and produced 5 times more RJ than the ITb, demonstrating a positive correlation between the yield and HG acini size. In addition, the proteomic analysis showed that RJb significantly upregulated a large group of proteins involved in carbohydrate metabolism and energy production, those involved in protein biosynthesis, development, amino acid metabolism, nucleotide and fatty acid, transporter, protein folding, cytoskeleton, and antioxidation, which coincides with the fact that the HGs of the RJb strain produce more RJ than the ITb strain that is owing to selection pressure. We also observed age-dependent major royal jelly proteins (MRJPs) changing both in form and expressional intensity concurrent with task-switching. In addition to MRJPs, the RJb overexpressed proteins such as enolase and transitional endoplasmic reticulum ATPase, protein biosynthesis, and development proteins compared to the ITb strain to support its large HG growth and RJ secretion. Because of selection pressure, RJb pursued a different strategy of increased RJ production by involving additional proteins compared to its original counterpart ITb. To our knowledge, this morphological and proteomic comparison study on the HG of the two strains of worker honeybees associated with their age-dependent division of labor is the first of its kind. The study provided not only the quantity and quality differences in the HG from the RJb and the ITb, but also addressed the cellular and behavioral biology development question of how the RJb strain can produce RJ more efficiently than its wild type strain (ITb). 相似文献

In honeybees, worker policing via egg eating enforces functionalworker sterility in colonies with a queen and brood. It is thoughtthat queens mark their eggs with a chemical signal, indicatingthat their eggs are queen-laid. Worker-laid eggs lack this signaland are, therefore, eaten by policing workers. Anarchistic workerhoneybees have been hypothesized to circumvent worker policingby mimicking the queen egg-marking signal. We investigated thisphenomenon by relating chemical profiles of workers and theireggs to egg acceptability. We found that the ability of someworkers (anarchistic workers in queenright colonies and deviantworkers from a queenless colony) to lay more acceptable eggsis due to them producing significant amounts of queen-like estersfrom their Dufour's gland. These esters appear to be transferredto eggs during laying and increase egg survival. However, theseesters cannot be the normal queen egg-marking signal, as theyare generally absent from queen-laid eggs and only increasethe short-term persistence of worker-laid eggs, because only730% of anarchistic worker-laid eggs persisted to hatchingversus 9192% of queen-laid eggs. All workers can producesome esters, but only workers that greatly increase their esterproduction lay more acceptable eggs. Abstract. 1. Nectar-foraging honeybees ( Apis mellifera ) on lavender ( Lavandula stoechas ) appear to forage so as to maximise net energy return from foraging bouts; however, evidence from other studies suggests that foraging has a detrimental effect on survival, due at least in part to physiological deterioration of the flight mechanism. But foragers also acquire wing damage during foraging, which may increase foraging effort and reduce foraging lifespan.
2. The accumulation of damage over time and its effects on foraging flight and flower choice were studied in the field using a system in which the criteria for flower preference by foragers was known from previous work. Wing damage accumulated exponentially over time and resulted in foragers becoming less choosy about the flowers they visited.
3. Damage added experimentally contributed independently to the effect on choosiness. Effects of wing damage (natural and added experimentally) were also independent of those of a relative measure of age, which related in an inconsistent way to changes in foraging preferences. 相似文献

Deformed wing virus (DWV) infected semen was used for artificial insemination of DWV-free virgin queens. High titres of DWV could subsequently be detected not only in the spermatheca, but also in the ovaries, demonstrating venereal transmission of DWV in honey bees. Subsequent vertical transmission of the virus to the progeny of DWV infected queens was also demonstrated. Neither transmission route is 100% effective. Whether venereal transmission of DWV occurs during natural mating remains to be determined. The implications for the use, sale and transport of semen samples for artificial insemination are discussed. 相似文献

Hemocytes and the (prophenol-) phenoloxidase system constitute the immediate innate immune system in insects. These components of insect immunity are present at any post-embryonic life stage without previous infection. Differences between individuals and species in these immune parameters can reflect differences in infection risk, life expectancy, and biological function. In honeybees which show an age-related division of labor within the worker caste, previous studies demonstrated that foragers show a strongly reduced number of hemoctyes compared to the younger nurse bees. This loss of immune competence has been regarded advantageous with respect to an already high mortality rate due to foraging and to redistribution of energy costs at the colony level. Based on the idea that abandoning hemocytes in all adults would be a reasonably direct regulatory mechanism, we posed the hypothesis that abandoning hemocytic immunity is not restricted to worker honeybees. We tested our hypotheses by performing a comprehensive analysis of hemocyte number and phenoloxidase (PO)-activity levels in immunologically naive workers, queens, and drones. We found that in all three adult phenotypes hemocyte number is dramatically reduced in early adult life. In contrast, we found that the dynamics of PO-activity levels have sex and caste-specific characteristics. In workers, PO activity reached a plateau within the first week of adult life, and in queens enzyme levels continuously increased with age and reached levels twice as high as those found in workers. PO-activity levels slightly declined with age in drones. These data support our hypothesis, from which we infer that the previously reported reduction of hemocyte in foragers is not worker specific but represents a general phenomenon occurring in all honeybee adult phenotypes. 相似文献

Deformed wing virus (DWV) of honeybees (Apis mellifera) is closely associated with characteristic wing deformities, abdominal bloating, paralysis, and rapid mortality of emerging adult bees. The virus was purified from diseased insects, and its genome was cloned and sequenced. The genomic RNA of DWV is 10,140 nucleotides in length and contains a single large open reading frame encoding a 328-kDa polyprotein. The coding sequence is flanked by a 1,144-nucleotide 5' nontranslated leader sequence and a 317-nucleotide 3' nontranslated region, followed by a poly(A) tail. The three major structural proteins, VP1 (44 kDa), VP2 (32 kDa), and VP3 (28 kDa), were identified, and their genes were mapped to the N-terminal section of the polyprotein. The C-terminal part of the polyprotein contains sequence motifs typical of well-characterized picornavirus nonstructural proteins: an RNA helicase, a chymotrypsin-like 3C protease, and an RNA-dependent RNA polymerase. The genome organization, capsid morphology, and sequence comparison data indicate that DWV is a member of the recently established genus Iflavirus. 相似文献

Honeybees (Apis mellifera) undergo iron biomineralization, providing the basis for magnetoreception. We showed earlier the presence of superparamagnetic magnetite in iron granules formed in honeybees, and subscribed to the notion that external magnetic fields may cause expansion or contraction of the superparamagnetic particles in an orientation-specific manner, relaying the signal via cytoskeleton (Hsu and Li 1994). In this study, we established a size-density purification procedure, with which quantitative amount of iron granules was obtained from honey bee trophocytes and characterized; the density of iron granules was determined to be 1.25 g/cm(3). While we confirmed the presence of superparamagnetic magnetite in the iron granules, we observed changes in the size of the magnetic granules in the trophycytes upon applying additional magnetic field to the cells. A concomitant release of calcium ion was observed by confocal microscope. This size fluctuation triggered the increase of intracellular Ca(+2) , which was inhibited by colchicines and latrunculin B, known to be blockers for microtubule and microfilament syntheses, respectively. The associated cytoskeleton may thus relay the magnetosignal, initiating a neural response. A model for the mechanism of magnetoreception in honeybees is proposed, which may be applicable to most, if not all, magnetotactic organisms. 相似文献

Summary. Mechanical stresses by a narrow glass capillary were applied to unfertilized eggs of honeybees to determine whether the removal of meiotic blocks of the eggs could be caused by simple mechanical stimuli. The treated eggs developed into the anaphase of the first meiotic division at 15 min after treatment, whereas the untreated eggs remained arrested at the metaphase of the first meiotic division. The results of histological examination of the common oviduct showed that its inner widths were sufficiently narrow to cause the distortion of eggs passing through it. The distorted eggs could be fertilized and develop into diploid embryos if they were exposed to the semen immediately (within 30 sec) after egg distortion. However, this would not happen if the distorted eggs were exposed to semen later (30 min). The eggs exposed to the semen but not given mechanical stimuli could initiate the embryonic development with diploid chromosomes. The interval between mechanical distortion and sperm acceptance by eggs in vitro is compatible with that of natural oviposition of fertilized eggs by honeybee queens. These results suggest that egg activation by mechanical stresses in the common oviduct is valid for the natural oviposition in honeybees. 相似文献

Abstract. To explore the role of brain tyramine in reproductive worker honeybees, its effects after injection and oral treatment on brain dopamine levels and ovarian development in queenless worker honeybees are determined. Both tyramine injection and oral treatment in 10-day-old queenless bees leads to tyramine transportation into the brain and significantly elevates brain dopamine levels as a function of the tyramine concentration. Ovarian diameters are significantly larger in 10-day-old queenless bees treated with tyramine compared with queenless bees of the same age without tyramine treatment. Results on yolk formation in the ovary support the finding of increased ovarian diameter, suggesting that oral tyramine treatment accelerates ovarian development through dopamine effects and/or direct effects of tyramine on the ovary in queenless bees. Thus, tyramine has potential effects on the enhancement of brain dopamine levels and the acceleration of ovarian development for the transition of normal workers to reproductive worker honeybees. 相似文献

A population genetical model is used to investigate the effects of queen and drone fitness and swarming ability on nuclear and mitochondrial (mt) DNA variation in honeybees. The analysis of both types of DNA is particularly useful for a genetic study of the Africanized bee problem in the Americas. Both an analytical model and a Monte Carlo simulation show that even if mtDNA proves to be selectively neutral, Africanized mitotypes are expected at high frequencies because of the more frequent swarming of Africanized honeybees. Since the fitness of Africanized drones is higher than that of European drones, European and African mitotypes are expected to be polymorphic and consequently unreliable as diagnostic tools to discriminate between the two types. Samples of Africanized honeybees in Brazil reveal high mtDNA polymorphisms as predicted by the theoretical models. 相似文献

The honeybee pupae development influences its future adult condition as well as honey and royal jelly productions. However, the molecular mechanism that regulates honeybee pupae head metamorphosis is still poorly understood. To further our understand of the associated molecular mechanism, we investigated the protein change of the honeybee pupae head at 5 time-points using 2-D electrophoresis, mass spectrometry, bioinformatics, quantitative real-time polymerase chain reaction and Western blot analysis. Accordingly, 58 protein spots altered their expression across the 5 time points (13-20 days), of which 36 proteins involved in the head organogenesis were upregulated during early stages (13-17 days). However, 22 proteins involved in regulating the pupae head neuron and gland development were upregulated at later developmental stages (19-20 days). Also, the functional enrichment analysis further suggests that proteins related to carbohydrate metabolism and energy production, development, cytoskeleton and protein folding were highly involved in the generation of organs and development of honeybee pupal head. Furthermore, the constructed protein interaction network predicted 33 proteins acting as key nodes of honeybee pupae head growth of which 9 and 4 proteins were validated at gene and protein levels, respectively. In this study, we uncovered potential protein species involved in the formation of honeybee pupae head development along with their specific temporal requirements. This first proteomic result allows deeper understanding of the proteome profile changes during honeybee pupae head development and provides important potential candidate proteins for future reverse genetic research on honeybee pupae head development to improve the performance of related organs. 相似文献

Summary A new approach is presented to estimate the genetic variance of social behaviour of groups. Honeybees (Apis mellifera L.) are used as an example for highly social organisms. Most characters of economic importance strongly rely on collective group characters of honeybee colonies. The average relatedness between small groups of workers of one honeybee colony can be estimated using a discrete multinomial distribution. The genetic variance of a social behaviour (alarm behaviour) of groups of honeybee workers is estimated with the intraclass correlation between groups within a colony. In two populations tested, the coefficient of genetic determination was high (0.96–0.98) indicating that the metabolic bio-assay used was only weakly affected by environmental effects. 相似文献
