Ten different oligonucleotide primers of arbitrary sequence were used in Capillary and Air Thermocycler to amplify genomic total DNA of Paeonia suffruticosa subsp. spontanea and P. rockii isolated from several local populations in Shanxi, Shaanxi and Gansu provinces. Under the strictly standardized amplification condition for all the primers, these primers yielded clear and reproducible bands corresponding to amplified products and separable by agarose gel electrophoresis. Among a total of 71 bands amplified, 16(22. 5%) were polymorphic in a single individual of P. suffruticosa subsp. spontanea, while among a total of 76 bands 21(27.6%)were polymophic in a single individual of P. rockii. On an average,the pairwise marker difference between band profiles of conspecific individuals (different populations) was 7.9 for P. suffruticosa subsp. spontanea and 8.7 for P. rockii respectively. The average marker difference between P. suffruticosa subsp. spontanea and P. rockii was 10.3. Obviously, greater number of plants and primers will be required to detect satisfactorily level of genetic diversity. These preliminary results showed that intraspecific genetic diversity was low for the two endangered species. RAPD as a molecular marker was useful and feasible for detecting the genetic variation within species of wild moutans. And it was also potential for studying evolution and relationships between species.  相似文献

矮牡丹与紫斑牡丹RAPD分析初报   总被引:55,自引:0,他引:55  
用10个任意序列的寡核苷酸片段作为引物,将采自陕西、山西、甘肃等地矮牡丹与紫斑牡丹基因组DNA,在毛细管气浴式PCR热循环仪上随机扩增。在对所有引物扩增条件严格标准化的条件下,这些引物可产生清楚的、可重复的与扩增产物相应的琼脂糖凝胶电泳区带。这10个有效扩增的引物在矮牡丹平均每一个体中扩增出71条带,其中多态的带16条(22.5%),在紫斑牡丹平均每一个体中扩增出76条带,其中多态的带2l条(27.6%)。对每两个个体,每条多态带进行成对比较,累加后求分子标记差异的平均值。在矮牡丹3个居群间的平均差异是7.9,在紫斑牡丹4个居群间的平均差异是8.7,在矮牡丹与紫斑牡丹2个种间的平均差异是10.3。显然,若用来扩增的植物个体数目和引物数目增加,将会得到更满意的结果。初步结果表明濒危植物矮牡丹与紫斑牡丹种内低水平的DNA多态性。RAPD技术用于检测野生牡丹居群内与居群间的遗传变异是有用的与可行的;用于研究种间的进化和亲缘关系也有潜力。  相似文献

自我们近年发表一系列牡丹组分类文章以来, 国内外基本上赞同我们8个种的分类系统,但对一些问题仍有不同见解。本文进一步申述牡丹Paeonia suffruticosa Andrews 是一个独立的种而不是人工杂种, 以及银屏牡丹P. suffruticosa ssp. yinpingmudan是牡丹P. suffruticosa的野生类型而不是逸生的P. ostii的理由。上述论点也得到了分子树的支持。P. jishanensis T. Hong &; W. Z. Zhao是一个合法名称, 而P. spontanea (Rehder) T. Hong &; W. Z. Zhao则确实是一个多余名。太白山紫斑牡丹的学名应是P. rockii ssp. atava (Brühl) D. Y. Hong &; K. Y. Pan, 因此P. moutan Sims ssp. atava Brühl不应是可疑的分类群。本文还对Halda的6个组合和两个杂交种名作了处理。结果, 本文包括了5个新异名和一个新组合。  相似文献

对芍药属牡丹组Paeonia L.sect.Moutan DC.(全部野生种)40个居群进行了基于形态学证据的系统学分析,试图建立组内种间的系统发育关系。利用PAUP (4.0)计算机程序分别构建了建立在25个形态学性状基础上的所有研究类群的距离树(UPGMA、NJ)和最大简约树(MP)。所得树的拓扑结构基本一致,差异只发生在距离树和简约树之间,在由形态和细胞学关系都很近的5个种(牡丹P.suffruticosa、矮牡丹P.jishanensis、卵叶牡丹P.qiui、紫斑牡丹P.rockii和凤丹P.o  相似文献

Paeonia suffruticosa Andrews subsp. spontanea (Rehder) S. G. Haw et L. A. Lauener ( =P. spontanea = P. jishanensis) has been considered as the wild form of widely cultivated P. suffruticosa. However, the subspecies differs from the cultivated one in shape, division and indumentum of leaves and reproductive biology. As a result of extensive field work, a form with two individuals in Yinping Shan, Caohu, Anhui, and Songxian, Henan, are found to be extremely similar to P. suffruticosa, but apparently different from P. jishanensis. The form is considered to be the wild one of P. suffruticosa and described as new: P. suffruticosa subsp, yinpingmudan Hong, K. Y. Pan et Z. W. Xie.  相似文献

Tree peony, being crowned the title “King of Flowers” in China, is of great medicinal, ornamental, and economic values. In the present study, the phylogeny of the wild tree peony species (section Moutan, Paeonia, Paeoniaceae), represented by twelve accessions collected from all eight species in the section, was investigated based on the DNA sequence in five DNA fragments from both nuclear (Adh1A, Adh2 and GPAT) and chloroplast (trnS-trnG and rps16-trnQ) genomes, as well as morphological characters. Both maximum parsimony (MP) and Bayesian inference of phylogeny (BI) trees were reconstructed based on the combined data of the DNA sequences and morphological data, respectively. The MP and BI trees have the similar topology, and the sect. Moutan clearly branched into two clades. One clade consists of two species, P. delavayi and P. ludlowii, corresponding to the subsect. Delavayanae, and another clade is composed of other six species. Within the second clade, the six species can be divided into three subclades consisting of P. rockii and P. decomposita, P. jishanensis and P. qiui, P. suffruticosa and P. ostii, respectively. Among the three subclades, P. jishanensis/P. qiui is most closely related to P. suffruticosa/P. ostii. These results provide up to date the clearest picture of the phylogeny of wild tree peony species in the sect. Moutan.  相似文献

牡丹被认为是中国的国花,具有很高的医学、观赏和经济价值.野生牡丹被认为是栽培牡丹的野生祖先,因此弄清牡丹组的种间亲缘关系具有重要的理论和实践意义.由于受到信息量的限制,根据单基凼数据或形态数据往往无法对牡丹组的种间关系得到明确的结果.本研究用12份样品代表野生牡丹组(Paeonia section Moutan DC.,Paeoniaceae)8个种,利用包括核基因(Adh1A、Adh2和GPAT)和叶绿体基因(trnS-trnG和rps16-trnQ)的DNA序列以及形态性状的多套数据来探讨野牛牡丹的种间关系.合并分析得到具高支持率的牡丹组物种间的系统发育关系.结果表明,芍药属牡丹组8个野生种分为两个亚组,即肉质花盘亚组subseet.Delavayanae和革质花盘亚组subsect.Vaginatae.肉质花盘亚组包括滇牡丹P delavayi和大花黄牡丹P.ludlowii;革质花盘亚组包括其余6个种.革质花盘亚组中,四川牡丹P.decomposita ssp.decomposita和紫斑牡丹P. rockii ssp. rockii关系密切;卵叶牡丹P.qiui和矮牡丹P. jishanenMs关系密切;银屏牡丹P. suffruticosa ssp.yinpingmudan与风丹P. ostii关系 密切,并且后两个分支为姊妹群.  相似文献

In this paper, the paeonol, paeoniflorin and their analogs were analyzed in the roots of 14 species and 2 subspecies of Paeonia L. The existence and content of these compounds were discussed in three sections, sect. Moutan, sect. Paeonia and sect. Onaepia. In sect. Moutan, paeonol and its analogs were high in content in all species. In sect. Paeonia, low content of paeonol and its analogs were found in plants of four taxa, P. lactiflora, P. anomala ssp. veitchii, P. mairei and P. intermedia. None of these compounds was found in sect. Onaepia. Paeonol has a simple structure and is distributed widely in plant; its decrease and loss may be the result of evolution. Therefore, it is deduced that the relationship among the three sections of Paeonia might be that woody sect. Moutan is the more primitive and derived from the ancestor of Paeonia first. For the herbaceous sections, sect. Paeonia is more closely related to sect. Moutan than to sect. Onaepia. In sect. Moutan, there are less paeonol and its analogs in the species of subsect. Vaginata than in those of subsect. Delavayanae. Thus, the former may be considered more advanced. In sect. Paeonia, the taxa with minor content of paeonol and its analogs are diploid except P. mairei. Among them, P. lactiflora and P. anomala ssp. veitchii are relatively primitive by morphology. None of paeonol and its analogs was detected in the species with specialized form.  相似文献

? Premise of the study: Microsatellite primers were developed for Paeonia delavayi and P. ludlowii (Paeoniaceae) to study their population genetics and phytogeography. ? Methods and Results: Nine polymorphic nuclear microsatellite loci were isolated from an enriched library of P. delavayi and primers were designed. The number of alleles per locus ranged from two to 16; the observed and expected heterozygosities ranged from 0.014 to 0.687 and 0.042 to 0.875, respectively. Six polymorphic chloroplast microsatellite loci were identified in P. delavayi and primers were provided. The number of alleles per locus ranged from two to six and the polymorphic information content ranged from 0.08 to 0.716. Both nuclear and chloroplast primers were successfully applicable to P. ludlowii. ? Conclusions: The markers developed here will facilitate analyses of genetic diversity, population genetic structure, phytogeographical patterns, and conservation for P. delavayi and P. ludlowii.  相似文献

淹水胁迫下江南牡丹生长及光合特性研究   总被引:1,自引:0,他引:1  
以3年生江南牡丹品种‘凤丹白’为材料,利用盆栽淹水法,设置正常管理、轻度胁迫和重度胁迫3个水平,研究不同淹水胁迫水平对牡丹生长和光合特性的影响。结果表明:经过30 d胁迫后,正常管理、轻度胁迫和重度胁迫下的江南牡丹苗高生长量分别为3.6、1.1和0.73 cm,地径生长量分别为0.21、0.11和0.06 cm,植株总生物量增加量分别为7.0、3.0和2.75 g,淹水胁迫和正常生长差异显著,淹水胁迫严重影响了江南牡丹的生长。同时,在正常管理时,牡丹总叶绿素含量升高,而在淹水胁迫下呈下降趋势。淹水胁迫不同时间根系活力均呈下降趋势且随着胁迫程度的增加下降越大。正常管理下光合速率逐渐增加而胁迫条件下光合速率逐渐降低。同时胁迫条件下,牡丹蒸腾速率、气孔导度均明显下降;轻度淹水胁迫下胞间CO2浓度先升高后降低;而重度胁迫下胞间CO2浓度呈现逐渐升高的变化趋势。淹水胁迫对牡丹根系活力、茎段生长和叶片光合特性影响较大。该研究结果为江南牡丹耐涝胁迫机理研究奠定了理论基础。  相似文献

The taxonomical concept of the Paeonia suffruticosa complex i.e. Sect. Moutan Subsect. Vaginatae, has changed greatly since 1990. Six species and four subspecies have been described as new and two subspecies raised to specific level. Five species and two subspecies are recognized in the present revision, viz. P. suffruticosa subsp. suffruticosa and subsp. yinpingmudan, P. jishanensis, P. qiui, P. ostii, P. rockii subsp. rockii and subsp. taibaishanica. P. yananensis, P. ridleyi, P. spontanea, P. moutan subsp. atava, P. suffruticosa subsp. atava, P. rockii subsp. linyanshanii and P. ostii var. lishizhenii are treated as synonyms. P. papaveracea and P. baokangensis are proposed to be interspecific hybrids. A key to the recognized species and subspecies is provided. Biological features of the species are described and their distributions are mapped. The relationships between species are inferred and the origins of commonly cultivated tree peonies ( P. suffruticosa and P. ostii ) are discussed.  相似文献

滇牡丹花色类群性状变异分析   总被引:6,自引:0,他引:6  
对滇牡丹(Paeonia delavayi)10个花色类群的13个数量性状和5个质量性状进行了调查及统计分析,结果表明:(1)未发现所有花部性状均为同色的花色类群;(2)从花色与其它质量性状的连锁看,绿色类群均具绿色柱头,且是唯一存在直立花朵、绿色花丝、绿色花药的类群;(3)10个花色类群13个数量性状的平均变异系数范围为32.80%~50.56%,说明不同类群内个体问的性状稳定性差异较大;(4)10个花色类群平均表型分化系数为42.25%,表明滇牡丹10个花色类群表型变异主要来源于类群内,类群间的表型变异也很突出;(5)据花色演化趋势,初步推断云南省迪庆州中甸、德钦一带可能为滇牡丹的起源中心。  相似文献

为了比较分析芍药组不同类群光合生理差异以及它们对各自不同光照环境的适应性, 测定了芍药组(Paeonia sect. Paeonia) 4种2变种的光合日变化、光响应曲线、CO2响应曲线, 以及叶绿素荧光特性。结果显示, 芍药组中不同类群之间光合速率差异明显, 各个种类的“午休”程度不同, 芍药(Paeonia lactiflora )和毛果芍药(P. lactiflora var. trichocarpa)的强光抑制现象没有川赤芍(P. veitchii)、美丽芍药(P. mairei)和窄叶芍药(P. anomala)明显。叶绿素荧光特征能够反映芍药组不同类群光合生理的差异。芍药组内不同类群地理分布的差异能部分从光合生理特征的适应性方面得到解释。  相似文献

Premise of the study: Microsatellite primers were developed in the South African sclerophyllous shrub Protea punctata to investigate the degree of population differentiation within and between P. punctata and closely related species. ? Methods and Results: 10 primer pairs were identified from three individuals of Protea punctata. The primers amplified di- and tri-nucleotide repeats. Across all P. punctata samples, the loci have 8-49 alleles. All primers also amplified in Protea section Exsertae (P. aurea, P. aurea subsp. potbergensis, P. mundii, P. venusta, P. lacticolor, and P. subvestita). The loci had 14-69 alleles across the subgenus. ? Conclusions: These results show the broad utility of microsatellite loci for future studies of population genetics in the white proteas and their potential utility across the entire genus.  相似文献

丛枝菌根真菌对牡丹生长及相关生理指标的影响   总被引:7,自引:1,他引:6  
以采自山东省菏泽牡丹园的牡丹'凤丹'种子为试材,采用盆栽的方法研究了接种丛枝菌根(Arbuscular mycorrhizal,AM)真菌Glomus mosseae、Glomus versiforme和Glomus intraradices对牡丹幼苗生长及生理生化特性的影响.结果表明:(1)不同AM真菌的侵染率不同,并以G.mosseae的侵染率最高,为48.3%;(2)AM真菌能够促进牡丹幼苗生长,接种G.mosseae的植株干重比对照显著提高了69.5%;(3)AM真菌能够提高牡丹的叶绿素、可溶性糖、可溶性蛋白、矿质元素(N、P、K)含量和硝酸还原酶活性,并对可溶性蛋白含量和硝酸还原酶活性的影响达显著水平.(4)植株总干重与菌根侵染率、硝酸还原酶活性、叶绿素含量呈极显著正相关,与可溶性糖、可溶性蛋白含量呈显著正相关.研究发现,G.mosseae是最适宜牡丹接种的优良菌种.  相似文献
