The neurotoxin 1-methy-4-phenylpyridinium (MPP+) is used for its’ capacity to induce Parkinsonism through its inhibitory effects on mitochondrial complex I. This inhibition disrupts cellular energy formation and aerobic glycolysis. The objective of this study was to demonstrate that the toxic effect of mitochondrial aerobic pathway inhibition with MPP+ can be reduced by stimulating anaerobic glycolysis using glucose supplementation. In this study, C6 Glioma cell viability was examined in the presence of different concentrations of MPP alone and with the addition of glucose. The results obtained indicate that there was a significant increase (P < 0.001) in cell viability in cells treated with glucose and MPP+ verses cells treated with MPP+ alone. Fluorometric analysis using 100 uM Rhodamine 123 indicated mitochondrial membrane potential was not restored in MPP+ treated cells with glucose; however, normal cell viability was confirmed using 2 ug/ml Fluorescein diacetate. This dual fluorescence indicated mitochondrial damage from MPP+ while glucose augmented cell survival. Further confirmation of cell survival upon damage to the mitochondria was evident in TUNEL staining. Positive staining was prominent only in MPP+ treatment groups alone, while control and co-treated groups exhibited little to no TUNEL staining. ATP measurements of all MPP+ treated groups exhibited a significant (P < 0.001) decrease verses control. Groups co-treated with MPP+ and glucose revealed a significant increase (250 μM group: P < 0.001) in ATP. It was concluded from this study that glucose supplementation was able to sustain cellular viability and ATP production through anaerobic glycolysis despite the inhibitory effect of MPP+ on aerobic glycolysis.  相似文献

Immortal cell lines are used to investigate various aspects of neurodegeneration. These cells display high glycolytic turnover rate and produce an abundant amounts of lactate. Our previous studies indicate that these cells survive the loss of mitochondrial oxidative phosphorylation (OXPHOS) with ample glucose supply. In the current study, we investigate if cell type (w/variation in basal metabolic rate (MR)), can alter glucose utilization patterns which in turn may affect LC50 for the mitochondrial toxin 1-methyl-4-phenylpyridinium (MPP+) in various cell lines. The data obtained indicate that cell lines MRs examined were generally consistent with the average of species adult body weight where mouse N-2A > rat-PC-12 > human SH-SY5Y. A higher MR was associated with accelerated utilization of glucose and earlier cell death with MPP+: LC50 mouse = 294 μM, rat = 695 μM, and human = 5.25 mM at 24 h. Cell death appears to be a function of the velocity by which glucose disappears, leading to the failure of glycolysis and subsequent halt of energy production. Similar effects were also observed at higher plating densities where the demand for glucose is amplified. A time-lapse study of MPP+ toxicity (0–36 h) in N-2A cells indicates that an anaerobic shift occurs as early as 2 h (evidenced by a rise in lactate), followed by a descent in glucose concentrations at 4 h and exhaustion of glucose supplies at 22 h which was associated with the first detectable sign of cell death. It was also noted that MPP+ toxicity was not associated with the generation of reactive oxygen species (O2, H202, and NO2) and was not attenuated by adding catalase or superoxide dismutase to the media. On the other hand, MPP+ toxicity was reversed by providing additional supply of glucose, pyruvate ± mitochondrial monocarboxylate transporter blocker (α-cyano-4-HCA), or pyruvate ± pyruvate dehydrogenase inhibitor (octanoyl-CoA), suggesting that the exclusive anaerobic survival compensates for the loss of OXPHOS by MPP+. To examine if neuroblastoma were capable of surviving the deprivation of O2 for 24 h, a range of hypoxia to anoxia was established with various concentrations of dithionite. The data suggest that cell lines examined continue to thrive when incubated with high-glucose media (25 mM). In summary, vulnerability of immortal neuroblastoma cell lines to MPP+ toxicity is dependent upon glucose concentrations within the media and cell MR, which indirectly dominates the velocity of glucose use and its end point disappearance, leading to cell death by ergogenic failure.  相似文献

Efflux of glutathione (GSH) from astrocytes has been suggested as a key factor for neuroprotection by astrocytes. Here we evaluated if the Nrf2 activator curcumin affects basal and stimulated (Ca2+ omission) GSH efflux from cultures of astroglial cells. Stimulated efflux of GSH was observed at medium concentration of 0, 0.1 mM Ca2+, but not at 0.2 or 0.3 mM Ca2+. Astroglia treated with 30 μM curcumin increased the cellular content of GSH in parallel with elevated basal and stimulated efflux. Conversely treatment with buthionine sulfoximine lowered efflux of GSH. The efflux stimulated by Ca2+- omission was not affected by the P2X7-receptor antagonist Blue Brilliant G (100 nM) or the pannexin mimetic/blocking peptide 10Panx1 but inhibited by the gap junction blocker carbenoxolone (100 μM) and a hemichannel blocker Gap26 (300 μM). RNAi directed against Nrf2 partly inhibited the effect of curcumin. The results show that elevated cellular GSH by curcumin treatment enhance efflux from astroglial cells, a process which appear to be a prerequisite for astroglial mediated neuroprotection.  相似文献

In the present study, we investigated the protective mechanism of paeoniflorin (PF), a monoterpene glycoside extracted from Radix Paeoniae alba roots, on MPP+-induced neurotoxicity in cultured rat pheochromocytoma cells (PC12). Our work included examination of cell viability assessment, amounts of released lactic dehydrogenase (LDH), intracellular Ca2+ concentration, cell apoptosis, mitochondrial membrane potential, caspase-3 activity, and expression profiling of two apoptosis-related genes (Bcl-2 and Bax). It was shown that, PF functioned as an MPP+ antagonist, being able to suppress apoptosis, decrease LDH release and Ca2+ concentration, attenuate membrane potential collapse and, inhibit caspase-3 activation, decrease in Bax/Bcl-2 ratio. These observations suggest that PF could protect PC12 cells against MPP+-induced injury and the mechanism PF’s neuroprotective effect was closely associated with Bcl-2 up-regulation and Bax down-regulation. PF has neuroprotective effects on MPP+-induced apoptosis in PC12 cells via regulating mitochondrial membrane potential and Bcl-2/Bax/caspase-3 signaling pathways, and this new insight will help develop a PF-based therapeutic strategy for treatmenting neurodegenerative diseases and injury.  相似文献

It is known that permeability of the inner mitochondrial membrane is low to most univalent cations (K+, Na+, H+) but high to Tl+. Swelling, state 4, state 3, and 2,4-dinitrophenol (DNP)-stimulated respiration as well as the membrane potential (ΔΨmito) of rat liver mitochondria were studied in media containing 0–75 mM TlNO3 either with 250 mM sucrose or with 125 mM nitrate salts of other monovalent cations (KNO3, or NaNO3, or NH4NO3). Tl+ increased permeability of the inner mitochondrial membrane to K+, Na+, and H+, that was manifested as stimulation of the swelling of nonenergized and energized mitochondria as well as via an increase of state 4 and dissipation of ΔΨmito. These effects of Tl+ increased in the order of sucrose <K+ <Na+ ≤ NH4+. They were stimulated by inorganic phosphate and decreased by ADP, Mg2+, and cyclosporine A. Contraction of energized mitochondria, swollen in the nitrate media, was markedly inhibited by quinine. It suggests participation of the mitochondrial K+/H+ exchanger in extruding of Tl+-induced excess of univalent cations from the mitochondrial matrix. It is discussed that Tl+ (like Cd2+ and other heavy metals) increases the ion permeability of the inner membrane of mitochondria regardless of their energization and stimulates the mitochondrial permeability transition pore in low conductance state. The observed decrease of state 3 and DNP-stimulated respiration in the nitrate media resulted from the mitochondrial swelling rather than from an inhibition of respiratory enzymes as is the case with the bivalent heavy metals.  相似文献

Sodium concentrations as low as 2 mM exerted a significant protective effect on the high-pressure inactivation (160–210 MPa) of Rhodotorula rubra at pH 6.5, but not on two other yeasts tested (Shizosaccharomyces pombe and Saccharomyces cerevisiae). A piezoprotective effect of similar magnitude was observed with Li+ (2 and 10 mM), and at elevated pH (8.0–9.0), but no effect was seen with K+, Ca2+, Mg2+, Mn2+, or NH4 +. Intracellular Na+ levels in cells exposed to low concentrations of Na+ or to pH 8.0–9.0 provided evidence for the involvement of a plasma membrane Na+/H+ antiporter and a correlation between intracellular Na+ levels and pressure resistance. The results support the hypothesis that moderate high pressure causes indirect cell death in R. rubra by inducing cytosolic acidification.Communicated by K. Horikoshi  相似文献

The activity of Na+/H+ exchanger to remove toxic Na+ is important for growth of organisms under high salinity. In this study, the halotolerant cyanobacterium Aphanothece halophytica was shown to possess Na+/H+ exchange activity since exogenously added Na+ could dissipate a pre-formed pH gradient, and decrease extracellular pH. Kinetic analysis yielded apparent K m (Na+) and V max of 20.7 ± 3.1 mM and 3,333 ± 370 nmol H+ min−1 mg−1, respectively. For cells grown under salt-stress condition, the apparent K m (Na+) and V max was 18.3 ± 3.5 mM and 3,703 ± 350 nmol H+ min−1 mg−1, respectively. Three cations with decreasing efficiency namely Li+, Ca2+, and K+ were also able to dissipate pH gradient. Only marginal exchange activity was observed for Mg2+. The exchange activity was strongly inhibited by Na+-gradient dissipators, monensin, and sodium ionophore as well as by CCCP, a protonophore. A. halophytica showed high Na+/H+ exchange activity at neutral and alkaline pH up to pH 10. Cells grown at pH 7.6 under high salinity exhibited higher Na+/H+ exchange activity than those grown under low salinity during 15 days of growth suggesting a role of Na+/H+ exchanger for salt tolerance in A. halophytica. Cells grown at alkaline pH of 9.0 also exhibited a progressive increase of Na+/H+ exchange activity during 15 days of growth.  相似文献

Exposing bovine chromaffin cells to a single 5 ns, high-voltage (5 MV/m) electric pulse stimulates Ca2+ entry into the cells via L-type voltage-gated Ca2+ channels (VGCC), resulting in the release of catecholamine. In this study, fluorescence imaging was used to monitor nanosecond pulse-induced effects on intracellular Ca2+ level ([Ca2+]i) to investigate the contribution of other types of VGCCs expressed in these cells in mediating Ca2+ entry. ω-Conotoxin GVIA and ω-agatoxin IVA, antagonists of N-type and P/Q-type VGCCs, respectively, reduced the magnitude of the rise in [Ca2+]i elicited by a 5 ns pulse. ω-conotoxin MVIIC, which blocks N- and P/Q-type VGCCs, had a similar effect. Blocking L-, N-, and P\Q-type channels simultaneously with a cocktail of VGCC inhibitors abolished the pulse-induced [Ca2+]i response of the cells, suggesting Ca2+ influx occurs only via VGCCs. Lowering extracellular K+ concentration from 5 to 2 mM or pulsing cells in Na+-free medium suppressed the pulse-induced rise in [Ca2+]i in the majority of cells. Thus, both membrane potential and Na+ entry appear to play a role in the mechanism by which nanoelectropulses evoke Ca2+ influx. However, activation of voltage-gated Na+ channels (VGSC) is not involved since tetrodotoxin (TTX) failed to block the pulse-induced rise in [Ca2+]i. These findings demonstrate that a single electric pulse of only 5 ns duration serves as a novel stimulus to open multiple types of VGCCs in chromaffin cells in a manner involving Na+ transport across the plasma membrane. Whether Na+ transport occurs via non-selective cation channels and/or through lipid nanopores remains to be determined.  相似文献

The Ca2+-conducting pathway of myocytes isolated from the cricket lateral oviduct was investigated by means of the whole-cell patch clamp technique. In voltage-clamp configuration, two types of whole cell inward currents were identified. One was voltage-dependent, initially activated at –40 mV and reaching a maximum at 10 mV with the use of 140 mM Cs2+-aspartate in the patch pipette and normal saline in the bath solution. Replacement of the external Ca2+ with Ba2+ slowed the current decay. Increasing the external Ca2+ or Ba2+ concentration increased the amplitude of the inward current and the current–voltage (I–V) relationship was shifted as expected from a screening effect on negative surface charges. The inward current could be carried by Na+ in the absence of extracellular Ca2+. Current carried by Na+ (I Na) was almost completely blocked by the dihydropyridine Ca2+ channel antagonist, nifedipine, suggesting that the I Na is through voltage-dependent L-type Ca2+ channels. The other inward current is voltage-independent and its I–V relationship was linear between –100 mV to 0 mV with a slight inward rectification at more hyperpolarizing membrane potentials when 140 mM Cs+-aspartate and 140 mM Na+-gluconate were used in the patch pipette and in the bath solution, respectively. A similar current was observed even when the external Na+ was replaced with an equimolar amount of K+ or Cs+, or 50 mM Ca2+ or Ba2+. When the osmolarity of the bath solution was reduced by removing mannitol from the bath solution, the inward current became larger at negative potentials. The I–V relationship for the current evoked by the hypotonic solution also showed a linear relationship between –100 mV to 0 mV. Bath application of Gd3+ (10 M) decreased the inward current activated by membrane hyperpolarization. These results clearly indicate that the majority of current activated by a membrane hyperpolarization is through a stretch-activated Ca2+-permeable nonselective cation channel (NSCC). Hippocampal slices have been widely used to investigate electrophysiological and metabolic neuronal parameters, as well as parameters of astroglial activity including protein phosphorylation and glutamate uptake. S100B is an astroglial-derived protein, which extracellularly plays a neurotrophic activity during development and excitotoxic insult. Herein, we characterized S100B secretion in acute hippocampal slices exposed to different concentrations of K+ and Ca2+ in the extracellular medium. Absence of Ca2+ and/or low K+ (0.2 mM KCl) caused an increase in S100B secretion, possibly by mobilization of internal stores of Ca2+. In contrast, high K+ (30 mM KCl) or calcium channel blockers caused a decrease in S100B secretion. This study suggests that exposure of acute hippocampal slices to low- and high-K+ could be used as an assay to evaluate astrocyte activity by S100B secretion: positively regulated by low K+ (possibly involving mobilization of internal stores of Ca2+) and negatively regulated by high-K+ (likely secondary to influx of K+).  相似文献

CD4+CD25+Foxp3+ regulatory T (Treg) cells are believed to play an important role in suppressing autoimmunity and maintaining peripheral tolerance. How their survival is regulated in the periphery is less clear. Here we show that Treg cells express receptors for gamma chain cytokines and are dependent on an exogenous supply of these cytokines to overcome cytokine withdrawal apoptosis in vitro. This result was validated in vivo by the accumulation of Treg cells in Bim-/- and Bcl-2 tg mice which have arrested cytokine deprivation apoptosis. We also found that CD25 and Foxp3 expression were down-regulated in the absence of these cytokines. CD25+ cells from Scurfy mice do not depend on cytokines for survival demonstrating that Foxp3 increases their dependence on cytokines by suppressing cytokine production in Treg cells. Our study reveals that the survival of Treg cells is strictly dependent on cytokines and cytokine producing cells because they do not produce cytokines. Our study thus, demonstrates that different gamma chain cytokines regulate Treg homeostasis in the periphery by differentially regulating survival and proliferation. These findings may shed light on ways to manipulate Treg cells that could be utilized for their therapeutic applications.  相似文献

Of various metal ions (Ca2+, Cr3+, Cu2+, Fe2+, Mg2+, Mn2+, Ni2+ and Zn2+) added to the culture medium of Agrobacterium tumefaciens at 1 mM, only Ca2+ increased Coenzyme Q10 (CoQ10) content in cells without the inhibition of cell growth. In a pH-stat fed-batch culture, supplementation with 40 mM of CaCO3 increased the specific CoQ10 content and oxidative stress by 22.4 and 48%, respectively. Also, the effect of Ca2+ on the increase of CoQ10 content was successfully verified in a pilot-scale (300 L) fermentor. In this study, the increased oxidative stress in A. tumefaciens culture by the supplementation of Ca2+ is hypothesized to stimulate the increase of specific CoQ10 content in order to protect the membrane against lipid peroxidation. Our results improve the understanding of Ca2+ effect on CoQ10 biosynthesis in A. tumefaciens and should contribute to better industrial production of CoQ10 by biological processes.  相似文献

The identification and separation of small intestinal epithelial stem cells are still on the preliminary stage. In this study, we planned to utilize immunohistochemistry, fluorescence-activated cell sorting (FACS) and RT-PCR to investigate the possibility of CD133 and CD44 as markers of human small intestinal epithelial stem cells. The expressions of CD133, CD44 and Lgr5 were studied by immunohistochemistry. Four subgroups of CD133+CD44+, CD133+CD44, CD133CD44+, CD133CD44 were sorted out through FACS and the expression level of Lgr5 gene was measured by RT-PCR and polyacrylamide gel electropheresis (PAGE) with sliver stained. Ten cases of samples were available for analyzing. By immunohistochemical staining, few cells with positive expressions of CD133, CD44 and Lgr5 were distributed in the bottom of crypts with the expression locations somewhat overlapped. The average percentage of CD133+CD44+ cells was 0.0580 ± 0.0403%, while the corresponding contents of CD133+CD44 cells, CD133CD44+ cells and CD133CD44 cells were 0.4000 ± 0.1225%, 0.7000 ± 0.2646% and 76.5600 ± 3.5529% respectively. Ten times of positive expressions of Lgr5 were detected in the CD133+CD44+ groups, while 9/10, 8/10 and 4/10 times for CD133+CD44, CD133CD44+ and CD133CD44 subgroups respectively. With the help of Quantityone 4.62 software, the densities of corresponding place to Lgr5 and reference gene were obtained. The density ratios of corresponding place to Lgr5 to reference gene were significant difference between subgroups (P < 0.001). By means of LSD method, the density ratios in CD133+CD44+ subgroups had statistical differences from the other subgroups (P < 0.05). We concluded CD133+CD44+ cells may be human small intestinal epithelial stem cells, which need further researches to confirm.  相似文献

Adult frog skin transports Na+ from the apical to the basolateral side across the skin. Antidiuretic hormone (ADH) is involved in the regulation of Na+ transport in both mammals and amphibians. We investigated the effect of arginine vasotocin (AVT), the ADH of amphibians, on the short-circuit current (SCC) across intact skin and on the basolateral Na+/K+-pump current across apically nystatin-permeabilized skin of the tree frog, Hyla japonica, in which the V2-type ADH receptor is expressed in vitro. In intact skin, 1 pM AVT had no effect on the SCC, but 10 nM AVT was sufficient to stimulate the SCC since 10 nM and 1 μM of AVT increased the SCC 3.2- and 3.4-fold, respectively (> 0.9). However, in permeabilized skin, AVT (1 μM) decreased the Na+/K+-pump current to 0.79 times vehicle control. Similarly, 500 μM of 8Br-cAMP increased the SCC 3.2-fold, yet 1 mM of 8Br-cAMP decreased the Na+/K+-pump current to 0.76 times vehicle control. Arachidonic acid (10−5 M) tended to decrease the Na+/K+-pump current. To judge from these in vitro experiments, AVT has the potential to inhibit the basolateral Na+/K+-pump current via the V2-type receptor/cAMP pathway in the skin of the tree frog.  相似文献

The synergistic benefits of the dual inoculation of legumes with nodule bacteria and arbuscular mycorrhizae (AM) are well established, but the effect of an external NH4+ supply on this tripartite relationship is less clear. This effect of NH4+ supply was investigated with regards to the growth and function of the legume host and both symbionts. Nodulated Phaseolus vulgaris seedlings with and without AM, were grown in a sand medium with either 0 N, 1 mM or 3 mM NH4+. Plants were harvested at 30 days after emergence and measurements were taken for biomass, N2 fixation, photosynthesis, asparagine concentration, construction costs and N nutrition. The addition of NH4+ led to a decline in the percentage AM colonization and nodule dry weights, although AM colonization was affected to a lesser extent. NH4+ supply also resulted in a decrease in the reliance on biological nitrogen fixation (BNF); however, the AM roots maintained higher levels of NH4+ uptake than their non-AM counterparts. Furthermore, the non-AM plants had a higher production of asparagine than the AM plants. The inhibitory effects of NH4+ on nodule function can be reduced by the presence of AM at moderate levels of NH4+ (1 mM), via improving nodule growth or relieving the asparagine-induced inhibition of BNF.  相似文献

The expression of Na+, K+-ATPase α3 subunit and synaptosomal membrane Na+, K+-ATPase activity were analyzed after administration of ouabain and endobain E, respectively commercial and endogenous Na+, K+-ATPase inhibitors. Wistar rats received intracerebroventricularly ouabain or endobain E dissolved in saline solution or Tris–HCl, respectively or the vehicles (controls). Two days later, animals were decapitated, cerebral cortex and hippocampus removed and crude and synaptosomal membrane fractions were isolated. Western blot analysis showed that Na+, K+-ATPase α3 subunit expression increased roughly 40% after administration of 10 or 100 nmoles ouabain in cerebral cortex but remained unaltered in hippocampus. After administration of 10 μl endobain E (1 μl = 28 mg tissue) Na+, K+-ATPase α3 subunit enhanced 130% in cerebral cortex and 103% in hippocampus. The activity of Na+, K+-ATPase in cortical synaptosomal membranes diminished or increased after administration of ouabain or endobain E, respectively. It is concluded that Na+, K+-ATPase inhibitors modify differentially the expression of Na+, K+-ATPase α3 subunit and enzyme activity, most likely involving compensatory mechanisms.  相似文献

Others have shown that exposing oocytes to high levels of (10–20 mM) causes a paradoxical fall in intracellular pH (pHi), whereas low levels (e.g., 0.5 mM) cause little pHi change. Here we monitored pHi and extracellular surface pH (pHS) while exposing oocytes to 5 or 0.5 mM NH3/NH4 +. We confirm that 5 mM causes a paradoxical pHi fall (−ΔpHi ≅ 0.2), but also observe an abrupt pHS fall (−ΔpHS ≅ 0.2)—indicative of NH3 influx—followed by a slow decay. Reducing [NH3/NH4 +] to 0.5 mM minimizes pHi changes but maintains pHS changes at a reduced magnitude. Expressing AmtB (bacterial Rh homologue) exaggerates −ΔpHS at both levels. During removal of 0.5 or 5 mM NH3/NH4 +, failure of pHS to markedly overshoot bulk extracellular pH implies little NH3 efflux and, thus, little free cytosolic NH3/NH4 +. A new analysis of the effects of NH3 vs. NH4 + fluxes on pHS and pHi indicates that (a) NH3 rather than NH4 + fluxes dominate pHi and pHS changes and (b) oocytes dispose of most incoming NH3. NMR studies of oocytes exposed to 15N-labeled show no significant formation of glutamine but substantial accumulation in what is likely an acid intracellular compartment. In conclusion, parallel measurements of pHi and pHS demonstrate that NH3 flows across the plasma membrane and provide new insights into how a protein molecule in the plasma membrane—AmtB—enhances the flux of a gas across a biological membrane.

Na+, K+-ATPase is inhibited by neurotensin, an effect which involves the peptide high affinity receptor (NTS1). Neurotensin effect on cerebral cortex synaptosomal membrane Na+, K+-ATPase activity of rats injected i.p. with antipsychotic clozapine was studied. Whereas 3.5 × 10−6 M neurotensin decreased 44% Na+, K+-ATPase activity in the controls, the peptide failed to modify enzyme activity 30 min after a single 3.0, 10.0 and 30.0 mg/kg clozapine dose. Neurotensin decreased Na+, K+-ATPase activity 40 or 20% 18 h after 3.0 or 5.6 mg/kg clozapine administration, respectively, and lacked inhibitory effect 18 h after 17.8 and 30.0 mg/kg clozapine doses. Results indicated that the clozapine treatment differentially modifies the further effect of neurotensin on synaptosomal membrane Na+, K+-ATPase activity according to time and dose conditions employed. Subtypes of purinergic receptors involved in modulation of cytoplasmic calcium ion concentration ([Ca2+]i) and insulin release in mouse pancreatic β-cells were examined in two systems, pancreatic islets in primary culture and beta-TC6 insulinoma cells. Both systems exhibited some physiological responses such as acetylcholine-stimulated [Ca2+]i rise via cytoplasmic Ca2+ mobilization. Addition of ATP, ADP, and 2-MeSADP (each 100 μM) transiently increased [Ca2+]i in single islets cultured in the presence of 5.5 mM (normal) glucose. The potent P2Y1 receptor agonist 2-MeSADP reduced insulin secretion significantly in islets cultured in the presence of high glucose (16.7 mM), whereas a slight stimulation occurred at 5.5 mM glucose. The selective P2Y6 receptor agonist UDP (200 μM) transiently increased [Ca2+]i and reduced insulin secretion at high glucose, whereas the P2Y2/4 receptor agonist UTP and adenosine receptor agonist NECA were inactive. [Ca2+]i transients induced by 2-MeSADP and UDP were antagonized by suramin (100 μM), U73122 (2 μM, PLC inhibitor), and 2-APB (10 or 30 μM, IP3 receptor antagonist), but neither by staurosporine (1 μM, PKC inhibitor) nor depletion of extracellular Ca2+. The effect of 2-MeSADP on [Ca2+]i was also significantly inhibited by MRS2500, a P2Y1 receptor antagonist. These results suggested that P2Y1 and P2Y6 receptor subtypes are involved in Ca2+ mobilization from intracellular stores and insulin release in mouse islets. In beta-TC6 cells, ATP, ADP, 2-MeSADP, and UDP transiently elevated [Ca2+]i and slightly decreased insulin secretion at normal glucose, while UTP and NECA were inactive. RT-PCR analysis detected mRNAs of P2Y1 and P2Y6, but not P2Y2 and P2Y4 receptors.  相似文献

Powered by the mitochondrial membrane potential, Ca2+ permeates the mitochondria via a Ca2+ channel termed Ca2+ uniporter and is pumped out by a Na+/Ca2+ exchanger, both of which are located on the inner mitochondrial membrane. Mitochondrial Ca2+ transients are critical for metabolic activity and regulating global Ca2+ responses. On the other hand, failure to control mitochondrial Ca2+ is a hallmark of ischemic and neurodegenerative diseases. Despite their importance, identifying the uniporter and exchanger remains elusive and their inhibitors are non-specific. This review will focus on the mitochondrial exchanger, initially describing how it was molecularly identified and linked to a novel member of the Na+/Ca2+ exchanger superfamily termed NCLX. Molecular control of NCLX expression provides a selective tool to determine its physiological role in a variety of cell types. In lymphocytes, NCLX is essential for refilling the endoplasmic reticulum Ca2+ stores required for antigendependent signaling. Communication of NCLX with the store-operated channel in astroglia controls Ca2+ influx and thereby neuro-transmitter release and cell proliferation. The refilling of the Ca2+ stores in the sarcoplasmic reticulum, which is controlled by NCLX, determines the frequency of action potential and Ca2+ transients in cardiomyocytes. NCLX is emerging as a hub for integrating glucose-dependent Na+ and Ca2+ signaling in pancreatic β cells, and the specific molecular control of NCLX expression resolved the controversy regarding its role in neurons and β cells. Future studies on an NCLX knockdown mouse model and identification of human NCLX mutations are expected to determine the role of mitochondrial Ca2+ efflux in organ activity and whether NCLX inactivation is linked to ischemic and/or neurodegenerative syndromes. Structure-function analysis and protein analysis will identify the NCLX mode of regulation and its partners in the inner membrane of the mitochondria.  相似文献
