Fitness of interspecific hybrids is sometimes high relative to their parents, despite the conventional belief that they are mostly unfit. F(1) hybrids between oilseed rape (Brassica napus) and weedy B. rapa can be significantly more fit than their weedy parents under some conditions; however, under other conditions they are less fit. To understand the reasons, we measured the seed production of B. napus, B. rapa, and different generations of hybrid plants at three different densities and in mixtures of different frequencies (including pure stands). Brassica napus, B. rapa, and backcross plants (F(1) ♀ × B. rapa) produced many more seeds per plant in pure plots than in mixtures and more seeds in plots when each was present at high frequency. The opposite was true for F(1) plants that produced many more seeds than B. rapa in mixtures, but fewer in pure stands. Both vegetative and reproductive interactions may be responsible for these effects. Our results show that the fitness of both parents and hybrids is strongly frequency-dependent and that the likelihood of introgression of genes between the species thus may depend on the numbers and densities of parents and their various hybrid offspring in the population.  相似文献

The likelihood that two species hybridise and backcross may depend strongly on environmental conditions, and possibly on competitive interactions between parents and hybrids. We studied the paternity of seeds produced by weedy Brassica rapa growing in mixtures with oilseed rape (B. napus) and their F(1) hybrids at different frequencies and densities. Paternity was determined by the presence of a transgene, morphology, and AFLP markers. In addition, observations of flower and pollen production, and published data on pollen fertilisation success, zygote survival, and seed germination, allowed us to estimate an expected paternity. The frequency and density of B. napus, B. rapa, and F(1) plants had a strong influence on flower, pollen, and seed production, and on the paternity of B. rapa seeds. Hybridisation and backcrossing mostly occurred at low densities and at high frequencies of B. napus and F(1), respectively. F(1) and backcross offspring were produced mainly by a few B. rapa mother plants. The observed hybridisation and backcrossing frequencies were much lower than expected from our compilation of fitness components. Our results show that the male fitness of B. rapa, B. napus, and F(1) hybrids is strongly influenced by their local frequencies, and that male fitness of F(1)hybrids, when pollinating B. rapa seeds, is low even when their female fitness (seed set) is high.  相似文献

Pollen-mediated movement of transgenes from transplastomic oilseed rape (Brassica napus) into wild relatives will be avoided if chloroplasts are maternally transmitted. We assess the probability of chloroplast exchange between conventional oilseed rape and wild Brassica rapa to model the future behavior of transplastomic cultivars. Primers specific to cpDNA were used to demonstrate maternal inheritance of chloroplasts in 47 natural hybrids between cultivated B. napus and wild B. rapa. We conclude that there will be no or negligible pollen-mediated chloroplast dispersal from oilseed rape. Transgene introgression could still occur in mixed populations, however, if B. napus acted as the recurrent female parent. Rate of transfer would then depend on the abundance of mixed populations, their persistence as mixtures, and hybridization frequency within stands. A low incidence of sympatry (0.6-0.7%) between wild B. rapa and cultivated B. napus along the river Thames, UK, in 1997 and 1998, suggests mixed stands will form only rarely. Eighteen feral populations of B. napus also showed a strong tendency toward rapid decline in plant number, seed return, and ultimately, extinction within 3 years. Conversely, hybrid production is significant in mixed stands, and the absence of control practices means that oilseed rape will have slightly greater persistence. We infer that some introgression from transplastomic B. napus into B. rapa is inevitable in mixed populations even though such populations will occur infrequently and will tend to lose B. napus plants relatively quickly. Chloroplast exchange will be extremely rare and scattered.  相似文献

Frequencies of spontaneous hybridization between oilseed rape (Brassica napus L.) and weedy Brassica campesíris ssp. campestris L. were measured in agricultural fields. Hybrids were identified by enzyme electrophoresis, random amplified polymorphic DNA (RAPD) analysis, chromosome counting, morphology, and pollen fertility. When the two species were mixed 1:1, B. campestris produced 13% hybrid seeds and oilseed rape 9%. In two experiments with single plants of the self-incompatible B. campestris widely spaced within fields of oilseed rape, 56% and 93% hybrid seeds were produced. Analysis of a weedy population of B. campestris in oilseed rape revealed 60% hybrid seeds. Backcrossing of the hybrids to the weedy species seems to occur as supported by the finding in a natural population of two B. campestris-like plants with a marker specific to oilseed rape. The results suggest that transgenes could be dispersed from oilseed rape to B. campestris.  相似文献

We used flow cytometry, chromosome counting and AFLP markers to investigate gene flow from the crop plant oilseed rape, Brassica napus (AACC) to wild B. rapa (AA) in the Netherlands. From 89 B. napus source populations investigated, all near cropping fields or at transhipment sites, only 19 contained a B. rapa population within a 2.5‐km radius. During our survey we found only three populations with F1 hybrids (AAC), as recognized by their nine extra chromosomes and by flow cytometry. These hybrids were all collected in mixed populations where the two species grew in close proximity. Populations with F1 hybrids were not close to crops, but instead were located on road verges with highly disturbed soils, in which both species were probably recruited from the soil seed bank. Many plants in the F2, BC1 or higher backcrosses are expected to carry one to eight C chromosomes. However, these plants were not observed among the hybrids. We further investigated introgression with molecular markers (AFLP) and compared sympatric B. rapa populations (near populations of B. napus) with control populations of B. rapa (no B. napus within at least 7 km). We found no difference between sympatric and control populations in the number of C markers in B. rapa, nor did we find that these sympatric populations closely resembled B. napus. Our data show that hybrids occur but also suggest no recent introgression of alleles from the crop plant B. napus into wild B. rapa in the Dutch populations studied.  相似文献

Turnip mosaic virus (TuMV) and cauliflower mosaic virus (CaMV) have been found infecting field crops of winter oilseed rape (Brassica napus ssp. oleifera) in South Warwickshire. Other viruses found include broccoli necrotic yellows virus (BNYV) and a member of the beet western yellows virus group. Systemic leaf symptoms caused by TuMV varied within and between cultivars; the three predominant reaction types were classified as necrotic, mosaic and immune. Some recently introduced cultivars of oilseed rape were more severely affected by TuMV infection than older cultivars. Reactions to CaMV were less varied and immunity was not found. The seed yield from TuMV and CaMV-infected plants was less than that of healthy control plants. This effect was due to infected plants producing either fewer seeds, smaller seeds or both. Germination of seeds from infected plants was unaffected if sown soon after harvest. After storage for one year the germination of seed from a virus infected plant was significantly less than that of seed from a virus-free plant. All commercial cultivars tested were experimentally susceptible to turnip yellow mosaic virus (TYMV) and some American strains of cucumber mosaic virus (CMV).  相似文献

Twenty five genotypes of oilseed rape (canola and mustard) were tested under varied supply of Zn (+Zn: 2 mg kg–1 soil, -Zn: no Zn added) in two pot experiments in soil culture to determine the genotypic variation in tolerance to the Zn-deficient conditions, that is, to identify the Zn-efficient genotypes. On the basis of performance of genotypes in pot experiments, ten genotypes were tested in 1995 for their performance under varied supply of Zn (+Zn: 3.5 kg ha–1, -Zn: no Zn added) on a Zn-deficient field in South Australia.Zn efficiency (ratio of shoot dry matter in -Zn to shoot dry matter in +Zn treatment and expressed in percentage) in pot Experiment 1 varied from 35% for 92-13 to 74% for Siren. Narendra, Dunkeld, Barossa, Oscar and Xinza 2 performed well under -Zn treatment. Zn efficiency in Experiment 2 varied from 32% for Wuyou 1 to 62% for Pusa Bold. Pusa Bold and CSIRO-1(mustard genotypes) were the most efficient in terms of dry matter production among all the oilseed rape genotypes tested. Root dry matter accumulation was significantly higher in Zn-efficient genotypes. Zn efficiency (ratio of seed yield in -Zn to seed yield in +Zn and expressed in percentage) in field experiment varied from 62% for Huashang 2 to 76% for Dunkeld. With few exceptions, the ranking of genotypes in pot and field experiments indicates similarity in their response to Zn deficiency. There looks to be genetic control over Zn concentration in tissues. Zn-efficient genotypes had lower Zn concentration in roots and higher Zn concentration in youngest fully opened leaf blades, indicating a better transport of Zn. This, together with a higher Zn uptake, appears to be the basis of expression of Zn efficiency.  相似文献

In most experimental hybridizations between oilseed rape (Brassica napus) and weedy B. campestris, either intra- or interspecific pollen has been applied to individual flowers. Under field conditions, however, stigmas will often receive a mixture of the two types of pollen, thereby allowing for competition between male gametophytes and/or seeds within pods. To test whether competition influences the success of hybridization, pollen from the two species was mixed in different proportions and applied to stigmas of both species. The resulting seeds were scored for paternity by isozyme and randomly amplified polymorphic DNA analysis. Using data on the proportion of fully developed seeds and the proportion of these seeds that were hybrids, a statistical model was constructed to estimate the fitness of conspecific and heterospecific pollen and the survival of conspecific and heterospecific zygotes to seeds. B. campestris pollen in B. napus styles had a significantly lower fitness than the conspecific pollen, whereas no difference between pollen types was found in B. campestris styles. Hybrid zygotes survived to significantly lower proportions than conspecific zygotes in both species, with the lowest survival of hybrid zygotes in B. napus pods. This is in contrast to the higher survival of hybrid seeds in B. napus than in B. campestris pods when pollinations are made with pure pollen. Altogether, the likelihood of a foreign pollen grain producing a seed was much lower on B. napus than on B. campestris. In addition, pods on B. napus developed to a lower extent the more heterospecific pollen was in the mix, whereas this had no effect on B. campestris.  相似文献

There is increasing interest in the use of trap crops as components of integrated pest management (IPM) strategies. Understanding the mechanisms underlying host plant preferences of herbivorous pests can lead to improved effectiveness and reliability of the trap crop. We investigated the behavioural and chemical ecology underlying the success of turnip rape, Brassica rapa, trap crops in protecting oilseed rape, Brassica napus, from the pollen beetle, Meligethes aeneus, which feeds in the flowers and lays its eggs in the buds causing yield loss. Using a semi-field arena bioassay, plant growth stage was found to be a major factor in the preference of this pest for B. rapa over B. napus. Plants at early-flowering growth stages were preferred over plants in the bud stage, irrespective of species. No preference was found when both species were flowering. As B. rapa develops faster than B. napus in the field, this could explain part of the mechanism of its success as a trap crop. However, B. rapa was preferred over B. napus when both species were in the bud stage, indicating some inherent preferences for B. rapa. Responses of M. aeneus in olfactometer tests to the odours of B. napus and B. rapa at the bud and flowering growth stages, reflected those of the semi-field arena bioassay. These behavioural responses can be explained by volatile compounds associated with the flowering stage. Phenylacetaldehyde, indole and (E,E)-α-farnesene were found to be present in air entrainment samples of both plant species at the flowering growth stage, but only in those of B. rapa at the bud stage. The former two compounds were behaviourally-active in olfactometer tests. These compounds are likely to be involved in host location by M. aeneus, and, at least partially, responsible for the attractiveness of B. rapa and its success as a trap crop to protect B. napus from this pest.  相似文献

Summary The response of oilseed rape cultivars to infection with Agrobacterium tumefaciens and A. rhizogenes and the possibility of regenerating genetically transformed oilseed rape plants were examined. The frequency at which Agrobacterium induced galls or hairy-roots on in vitro cultured plants ranged from 10% to 70%, depending on the cultivar. From galls induced by the tumorigenic strain T37, known to be strongly shoot inducing on tobacco, roots developed frequently. Occasionally, shoots formed and some of these produced tumour cell specific nopaline. Attempts to grow the transformed shoots into plants have so far been unsuccessful. Whole plants transformed with Ri-T-DNA, however, were regenerated. These had crinkled leaves and abundant, frequently branching roots that showed reduced geotropism, similar to previously isolated Ri T-DNA transformed tobacco and potato plants. The transformed oilseed rape plants flowered, but failed to form seeds.  相似文献

Feral oilseed rape populations on roadsides and along other semi-natural habitats contribute to the uncertainty regarding the potential risk of transgene escape from genetically modified (GM) oilseed rape to wild or weedy populations. Knowledge about the underlying mechanism of establishment, persistence, and the evolutionary potential of feral populations is a prerequisite for a successful risk assessment and of high policy relevance. We analysed the origin, persistence, and genetic variation of feral oilseed rape populations in northwest Germany based on a field survey (2004–2007) and nuclear microsatellite markers. A maximum-likelihood approach was used to allocate individuals of feral populations to their source varieties and to hybrids between these varieties. The majority (72%) of the analysed sites was inhabited by oilseed rape for at least 2 years. The proportion of feral populations setting seeds varied between years (30–48%) and was higher than in comparable studies. Within-population genetic diversity of feral oilseed rape populations was higher than that of the common grown varieties studied. Repeated escapes of different varieties and hybridisation between these varieties were identified as the most important sources of genetic variation in feral oilseed rape populations. Up to four different source varieties were identified in feral populations. The detection of three hybrids between different varieties indicates that feral oilseed rape populations may persist via self-recruitment. Our results highlight the evolutionary potential of feral oilseed rape populations and have consequences for approaches to regulate the coexistence of genetically modified and non-GM oilseed rape varieties in the same region. Feral populations may serve as stepping stones for intraspecific and interspecific gene flow and can promote persistence of transgenes outside cultivation. The likelihood of introgressive interspecific hybridisation depends on the abundance of crossing partners, which may vary strongly from region to region.  相似文献

The objective of this study was to evaluate pollen dispersal inBrassica napus (oilseed rape). The selectable marker, used to follow pollen movement, was a dominant transgene (bar) conferring resistance to the herbicide glufosinate-ammonium. Transgenic and non-transgenic plants of the cultivar Westar were planted in a 1.1 ha field trial, with the transgenic plants in a 9 m diameter circle at the centre, surrounded by non-transgenic plants to a distance of at least 47 m in all directions. A 1 m circle of non-transgenic plants was sown in the centre of the transgenic area to allow estimation of the level of pollen dispersal when plants were in close contact. Honeybee hives were placed at the trial site to optimize the opportunity for cross-pollination. During the flowering period, regular observations were made of the number of plants flowering and the number and type of insects present in 60 1 m2 areas. These areas were located uniformly around the plot at distances of 1, 3, 6, 12, 24, 36 and 47 m from the edge of the 9 m circle of transgenic plants. Seed samples were harvested from each of the 7 distances so that approximately 20% of the circumference of the plot was sampled at each distance. The centre non-transgenic circle was also sampled. Plants were grown from the seed samples and sprayed with glufosinate to estimate the frequency of pollen dispersal at each distance. In order to screen enough samples to detect low frequency cross-pollination events, seed samples were tested in the greenhouse and on a larger scale in the field. Results were confirmed by testing progeny for glufosinate resistance and by Southern blot analysis. The estimated percentage of pollen dispersal in the non-transgenic centre circle was 4.8%. The frequency was estimated to be 1.5% at a distance of 1 m and 0.4% at 3 m. The frequency decreased sharply to 0.02% at 12 m and was only 0.00033% at 47 m. No obvious directional effects were detected that could be ascribed to wind or insect activity.  相似文献

综合评述了转基因油菜的基因流及其生态风险.油菜作为最早的转基因作物之一目前已在加拿大和澳大利亚大面积商业化应用.(常)异花授粉作物油菜的天然异交率可达30%左右,也易与其它芸苔属作物杂交,因此转基因油菜的生态风险已引起各国科学家的高度重视.转基因油菜主要通过与其野生近缘种的花粉交换和与非转基因油菜的花粉交换两种方式进行花粉的输出.基因可能逃逸到相关野生近缘种,但在大田环境下能够得到杂种的可能性很小;由于基因的漂流在油菜田块间确实存在,因此在种植转基因油菜的过程中必须考虑其间隔距离.  相似文献

Few data is available on the nematodes found in Brassicaceae, except for the most important plant parasite. However, studying the structure of nematofauna could be an important database for the soil quality and in order to assess the effects of future disturbance. This is particularly important considering that the diffusion of the canola crop in the world is increasing because of its use as a bio-diesel. Very diffused is also the Bt variety of oil seed rape, and, in this case, the study of the impact on the soil health and on bio-diversity is essential. In this research we have analyzed the nematode community, used as a bio-indicator of the soil condition. The nematofauna found in canola (Brassica napus var. oleifera) fields located in Southern Italy (Metaponto - MT) was investigated. The nematode community was studied considering its abundance, genus composition and trophic structure. Maturity and biodiversity indices were also calculated. A total of 5286 nematodes were extracted. They belong to 14 families and 24 genera. Bacterial and fungal feeders, 50.18% and 42.90% of the total respectively, dominated the trophic structure. Aphelencus is the most abundant genus (23.71%) followed by Acrobeloides (20.49%) and Aphelencoides (19.18%). Among plant feeders (6.59%), Pratylenchus is the dominant genus (2.20%) and Tylenchidae the main family (3.54%). No infestation of Meloidogyne, Heterodera or Naboccus, important plant-parasitic nematodes of canola crops, was recorded. Other important phytophagous were Helycotylenchus (0.5%), Trichotylenchus (0.5%) and Filenchus (0.9%). All of them had an abundance level below injury level. The indices of biodiversity are rather low (H'=0.93, J'=0.67), as is typical for agro ecosystems. However, the nematofauna community is quite well structured (N2=6.31, D=0.16) and the maturity index rather high (EMI=1.94). These values demonstrate that oilseed rape has a lower impact on the soil compared to other crop systems and that it could be taken into consideration for crop rotation programs. Canola could follow other more stressful crops, trying to maintain soil equilibrium.  相似文献

Brassica rapa (AA) has been used to widen the genetic basis of B. napus (AACC), which is a new but important oilseed crop worldwide. In the present study, we have proposed a strategy to develop new type B. napus carrying genomic components of B. rapa by crossing B. rapa with hexaploid (AACCCC) derived from B. napus and B. oleracea (CC). The hexaploid exhibited large flowers and high frequency of normal chromosome segregation, resulting in good seed set (average of 4.48 and 12.53 seeds per pod by self and open pollination, respectively) and high pollen fertility (average of 87.05 %). It was easy to develop new type B. napus by crossing the hexaploid with 142 lines of B. rapa from three ecotype groups, with the average crossability of 9.24 seeds per pod. The genetic variation of new type B. napus was diverse from that of current B. napus, especially in the A subgenome, revealed by genome-specific simple sequence repeat markers. Our data suggest that the strategy proposed here is a large-scale and highly efficient method to introgress genomic components of B. rapa into B. napus.  相似文献

When late torpedo- to early cotyledonary-stage zygotic embryos of Brassica napus were cultured on a solidified B5 basal medium with or without 0.05 mg l−1 6-benzylaminopurine (BAP), they proliferated by direct formation on the hypocotyl surface of structures resembling accessory cotyledons and root poles. When embryos bearing these structures were transferred to slowly rotating liquid culture on the same media, proliferation continued. As the underlying tissues senesced, meristematic structures became detached which produced cotyledonary and root tissues. A random sample of plants raised from these structures by direct transfer to sterilized potting mix all showed normal morphology.  相似文献

Oxylipins are products of oxygenase-catalyzed reactions of fatty acids. Oxylipins have been found or implied to participate in a variety of different functions in or between organisms. In this report we investigated the potential of various naturally occurring oxylipins found in plants for their effects as fungicides on a number of fungal pathogens interfering with Brassica cultivation. The fungi investigated were Alternaria brassicae, Leptosphaeria maculans, Sclerotinia sclerotiorum and Verticillium longisporum. An in vitro growth inhibition assay was used, where the relative growth rate of the fungi were determined in the presence of various concentrations of oxylipins. While no universal fungicidic effect was found for the 10 compounds investigated there were examples of oxylipins having inhibitory effects. In certain cases the inhibitory effects was overcome by time, however. Since several of the oxylipins tested were found to be stable in the absence of the fungus this effect could be explained by induction of the degrading capacity of the fungus or increased tolerance. Several of the oxylipins also inhibited germination of L. maculans spores but the relative potency differed compared to the effects on hyphae. The study suggests that selected oxylipins may be used for disease control on Brassica plants.  相似文献

Oviposition of phytophagous insects is determined either by adaptive behaviours allowing evaluation and response to host plant quality and/or by nutritional constraints occurring during oogenesis. Besides differences found among host plant species, plant intraspecific diversity can also affect insect oviposition. However, to date few studies have extensively investigated the factors accounting for the effect of this intraspecific variation. We addressed this question using oilseed rape (Brassica napus) and the pollen beetle (Meligethes aeneus), a phytophagous insect that uses the same plants and plant organs both for feeding and laying eggs. Our objectives were to test for a genotypic effect of oilseed rape on pollen beetle oviposition and identify the origin of the possible intergenotypic differences. We tested three hypotheses: oviposition is directly linked to (1) the amount of food eaten; (2) the nutritional quality of the food eaten; (3) a preference of females for certain plant genotypes. Results showed intergenotypic differences in both the number and the size of eggs laid. The factor that best accounted for most of these differences was the amount of food eaten. Nutritional quality of the pollen was of minor importance and females exhibited no preference among genotypes. These results reveal the importance of adult feeding on subsequent oviposition in phytophagous insects, an often neglected factor which partly determines the amount of energy available for oogenesis. Taking into account this factor may be of crucial importance in studies conducted on synovogenic insect species feeding on the same plant on which they lay eggs.  相似文献

An integrated linkage map between B. napus and B. rapa was constructed based on a total of 44 common markers comprising 41 SSR (33 BRMS, 6 Saskatoon, and 2 BBSRC) and 3 SNP/indel markers. Between 3 and 7 common markers were mapped onto each of the linkage groups A1 to A10. The position and order of most common markers revealed a high level of colinearity between species, although two small regions on A4, A5, and A10 revealed apparent local inversions between them. These results indicate that the A genome of Brassica has retained a high degree of colinearity between species, despite each species having evolved independently after the integration of the A and C genomes in the amphidiploid state. Our results provide a genetic integration of the Brassica A genome between B. napus and B. rapa. As the analysis employed sequence-based molecular markers, the information will accelerate the exploitation of the B. rapa genome sequence for the improvement of oilseed rape.  相似文献
